The nodal explants of in vitro shoots of Holarrhena antidysenterica L. were cultured on Murashige and Skoog's (MS) medium augmented with 15 μM N6-benzyladenine (BA) alone (control) or supplemented with different concentrations (1, 5, 10 and 20 mg dm−3) of CdCl2, CuSO4, Pb(NO3)2 and ZnSO4. The maximum morphogenic response in terms of average shoot number (4.95 ± 0.17) was seen in control. ZnSO4 proved to be less inhibitory in comparison to CuSO4, Pb(NO3)2 and CdCl2. None of the explants cultured on CdCl2 containing medium induced multiple shoots. Maximum protein content [3.80 ± 0.04 mg g−1(f.m.)] was observed in control and slightly less [3.50 ± 0.02 mg g−1(f.m.)] in tissues exposed to 1 mg dm−3 of CuSO4 and minimum [1.00 ± 0.02 mg g−1(f.m.)] in Zn treated (20 mg dm−3) explants. SDS-PAGE analysis of the treated tissues revealed that two new polypeptides (29 and 20 kDa) in response to Cu and Zn treatment, respectively, have been synthesized.  相似文献

The hypothesis that copper (Cu) alters drug metabolizing enzymes and functions as an antioxidant nutrient in doxorubicin cardiotoxicity was tested. Male Sprague-Dawley rats were fed Cu adequate (+Cu; 5 mg Cu/kg of diet), marginally Cu deficient (MCu; 1.2 mg Cu/kg of diet), or severely Cu deficient (Cu; 0.5 mg Cu/kg of diet) diets for 6 wk. Doxorubicin (1, 2, or 4 mg/kg body wt) or saline were administered intraperitoneally 1 time/wk for 4 wk. Compared to control hearts, Cu, Zn superoxide dismutase activity was decreased by 9% in MCu rats and by 21–40% inCu rats. Glutathione peroxidase activity was elevated 5–15% inCu rats. Doxorubicin administration increased heart Cu, Zn superoxide dismutase activity in+Cu andCu rats 18 h after the last of 4 injections, but not 18 h after 1 injection. There was no synergism between doxorubicin and Cu deficiency on lipid peroxidation, plasma creatine phosphokinase, cardiac hypertrophy, electrocardiographic abnormalities, or morphological changes. Heart glutathione S-transferase activity was decreased by Cu deficiency, and like Cu, Zn superoxide dismutase activity, returned to normal inCu rats given doxorubicin. Thus, the Cu deficient rat heart may be able to compensate for doxorubicin-induced oxidant stress by increasing the activity of Cu,Zn superoxide dismutase and glutathione S-transferase.  相似文献

The chronic effect (duration of exposure 218 days) of polychlorinated biphenyls (PCBs) and the prolonged effect of the short-term action of chlorophos or of N-methyl-N′-nitro-N-nitrosoguanidine (MNNG) during embryogenesis upon the sensitivity of intestinal glycosidases to Cu and Zn was studied in roach (Rutilus rutilus (L.) underyearlings. The Cu+2 and Zn+2 ions at concentrations ranging from 0.1 to 25 mg/L in vitro cause a 10–77% decrease in amylolytic activity in the intestinal mucosa of control roach. An elevated level of PCBs (50.8 ng/g wet weight of food and 426 ng/g dry weight of ground) increased the sensitivity of glycosidases to Cu and Zn. The embryotoxic action of chlorophos at concentrations of 1 × 10−6−1 × 10−2 mg/L in most cases increased the inhibitory effect of Cu but decreased that of Zn. As a rule, MNNG (3 × 10−7−3 × 10−1 mg/L) reduced the glycosidase sensitivity to the effect of metal ions. The magnitude and direction of the effect depend on the nature and concentration of toxicants.  相似文献

In order to understand the detailed mechanism of the stereoselective photoinduced electron-transfer (ET) reactions of zinc-substituted myoglobin (ZnMb) with optically active molecules by flash photolysis, we designed and prepared new optically active agents, such as N,N′-dimethylcinchoninium diiodide ([MCN]I2) and N,N′-dimethylcinchonidinium diiodide ([MCD]I2). The photoexcited triplet state of ZnMb, 3(ZnMb)*, was successfully quenched by [MCN]2+ and [MCD]2+ ions to form the radical pair of ZnMb cation (ZnMb·+) and reduced [MCN]·+ and [MCD]·+, followed by a thermal back ET reaction to the ground state. The rate constants (k q) for the ET quenching at 25 °C were obtained as k q(MCN)=(1.9±0.1)×106 M−1 s−1 and k q(MCD)=(3.0±0.2)×106 M−1 s−1, respectively. The ratio of k q(MCD)/k q(MCN)=1.6 indicates that the [MCD]2+ preferentially quenches 3(ZnMb)*. The second-order rate constants (k b) for the thermal back ET reaction from [MCN]·+ and [MCD]·+ to ZnMb·+ at 25 °C were k b(MCN)=(0.79±0.04)×108 M−1 s−1 and k b(MCD)=(1.0±0.1)×108 M−1 s−1, respectively, and the selectivity was k q(MCD)/k q(MCN)=1.3. Both quenching and thermal back ET reactions are controlled by the ET step. In the quenching reaction, the energy differences of ΔΔH (MCD–MCN) and ΔΔS (MCD–MCN) at 25 °C were obtained as −1.1 and 0 kJ mol−1, respectively. On the other hand, ΔΔH (MCD–MCN)=11±2 kJ mol−1 and TΔΔS (MCD–MCN)=−10±2 kJ mol−1 were given in the thermal back ET reaction. The highest stereoselectivity of 1.7 for [MCD]·+ found at low temperature (10 °C) was due to the ΔΔS value obtained in the thermal back ET reaction. Ellagic acid (EA) is a natural polyphenolic compound. Although, modulator effects of EA on copper (Cu) and zinc (Zn) levels in some liver diseases have been reported in experimental animals, its effects in obstructive jaundice (OJ) has not been clarified. We aimed to evaluate potential effects of EA on Cu and Zn levels in liver and serum of cholestatic rats. Forty Wistar albino rats were equally divided into four groups. First group was used as controls. Second group received EA (60 mg−1 kg−1 day−1) for 8 days. Third was OJ group, and fourth group was OJ plus EA group. After 8 days, blood and liver samples were obtained. Higher serum and liver Cu and lower serum and liver Zn levels were found in OJ group (p < 0.05) compared with other groups. However, these differences reached to significant levels for Cu in serum and for Zn in lever. Higher serum copper levels were decreased, and lower liver Zn levels were increased by EA treatment in cholestatic rats (p < 0.05). Also, higher Cu/Zn ratio in OJ group was decreased by EA treatment both in liver (p < 0.05) and in serum (p < 0.05). Significantly higher serum bilirubin, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase values were found in OJ and OJ + EA groups compared with the control and EA groups (p < 0.05). In conclusion, result of the current study indicated that ellagic acid has modulator effects on Cu and Zn levels in liver and serum of cholestatic rats.  相似文献

Metal concentrations have been determined in shoots of 12 species considered as Cu and/or Co hyperaccumulators, collected from five subpopulations in a Cu/Co mine in Katanga. Samples have been subjected to three cleansing protocols (water, alconox, alconox + EDTA). Cu/Co concentrations were significantly higher when demineralised water was used to wash samples compared to more aggressive solutions. Washing effect was largest for species with velvety (Acalypha cupricola) or sticky (Haumaniastrum katangense) indumentum. Element concentrations in shoots varied by two orders of magnitude, i.e. 45–2,891 mg kg−1 Cu (median: 329 mg kg−1) and 21–1,971 mg kg−1 Co (median: 426 mg kg−1) and were generally lower than previously published values. Only 9.3% of data exceeded the hyperaccumulation threshold (1,000 mg kg−1) for Cu and 13% for Co. Shoot concentrations varied by up to two orders of magnitude among subpopulations within species which was partly explained by variation of metal concentrations in the soil. Although the species considered in this paper undisputedly accumulate Co and Cu up to very high concentrations that require specific shoot tolerance mechanisms, the concept of hyperaccumulation, as defined for other metals like Zn and Ni, needs to be critically re-examined for Cu and Co. Our results suggest that many species so far regarded as Co/Cu hyperaccumulators might eventually turn out to be indicators.  相似文献

Purified uricase from a caprine kidney, possessed K m and V max values of 1.1 mg ml−1 and 3512 IU (mg protein)−1 for uric acid hydrolysis, respectively. The optimum temperature and pH for catalytic activity were 40 °C and 8.5, respectively. The activation energy for formation of ES complex was 13.6 kJ mol−1. Enthalpy (ΔH*), entropy of activation (ΔS*) and Gibbs free energy demand of uricase inactivation were 62.8 kJ mol−1, −102 J mol−1 K−1 and 104.3 kJ mol−1, respectively. Gibbs free enrgy demand for substrate binding and transition state stabilization were also determined which were comparable with those for themostable enzymes.  相似文献

The aim of the investigations was the determination of the Cu contents in the liver, kidneys and skeletal muscles of canine females. Material for research was collected post mortem from 45 animals aged 1 to 18 years coming from the Warsaw area. The effect of the health state, age and life conditions on the distribution of copper in the investigated organs was estimated. That element was determined using the method of inductively coupled plasma-atomic emission spectrometry (ICP-AES). In the liver, the average Cu contents amounted to 24.04 mg kg−1 wet weight, in kidneys to 2.90 mg kg−1 wet weight and in muscles to 0.94 mg kg−1 wet weight. The highest values of copper content in particular tissues and organs were noted in the group of animals with neoplastic changes. In respect to the animal age the highest mean values of the copper content were noted in the oldest animals. They amounted to 30.97 mg kg−1 in the liver, 3.34 mg kg−1 in kidneys and 1.18 mg kg−1 wet weight in muscles. Considering life conditions of the dogs it was observed that the higher mean values in all the investigated organs occurred in dogs coming from the urban areas.  相似文献

The coastal marine sediments near Santa Rosalía on the eastern coast of the Baja California Peninsula (Mexico) are heavily polluted by metals due to copper mining and smelting over the past century (up to 1984). The present study compares the accumulation of metals in the brown seaweed Padina durvillaei from the central segment of the coast of Santa Rosalía (polluted by Co, Cu, Mn and Zn) and from two segments north and south of the known “hot spot”. The seaweed samples were collected in May and August 2004 and June 2005 at 13 stations located along the Santa Rosalía mining region. Heavy metal concentrations were measured by instrumental neutron activation analysis (Co and Fe) directly in dried homogenized subsamples or by flame atomic absorption spectrophotometry (Cd, Cu, Mn, Ni, Pb and Zn) after complete strong acid digestion of sub-samples. The means and standard deviations of the concentrations in dry tissues of Padina durvillaei specimens for all the studied metals and stations lie in the following sequence: Cd (3.6 ± 1.6 mg kg−1) < Co (6.5 ± 6.1 mg kg−1) < Pb (7.8 ± 6.2) < Ni (9.96 ± 5.28 mg kg−1) < Cu (53 ± 38 mg kg−1) < Zn (63 ± 43 mg kg−1) < Mn (295 ± 269 mg kg−1) < Fe (2243 ± 2325 mg kg−1). This increase of the average concentrations was statistically significant. Principal Component Analysis showed that Factor 1 (36.46%) displays high positive loadings for Co, Cu, Mn and Zn, reflecting the influence of local anthropogenic pollution on the seaweed composition, while Factor 2 (26.91%) is important for Co, Fe and Ni and probably corresponds to the adsorption or accumulation of terrigenous elements of the regional origin, while Factor 3, with a high positive loading for Pb and a high negative loading for Cd, is probably controlled by local and regional anthropogenic input of Pb and episodic supply of Cd by local upwellings. The results of ANOVA for each element do not show any significant differences between the average concentrations for Cd, Fe, Ni and Pb in the seaweed of the three segments, or between the central and southern segments for Cu, Mn and Zn. Cobalt contents in the seaweed from the northern and central segments are, however, significantly different from the southern segment. This indicates that element concentrations in Padina durvillaei generally do not follow the drastic increases and gradients of Cu, Co, Mn and Zn contents detected in surface sediments. The apparent contradiction could be explained by a low geochemical mobility of these metals in the polluted sediments, composed mainly of stable residues of smelter wastes, with very low content of organic matter usually driving diagenetic mobilization of some metals into interstitial waters and then to the overlying water.  相似文献

The polyphasic patterns of fluorescence induction rise in pea leaves in vivo and after the treatment with ionophores have been studied using a Plant Efficiency Analyzer. To analyze in detail photosystem II (PS II) electron transfer processes, an extended PS II model was applied, which included the sums of exponential functions to specify explicitly the light-driven formation of the transmembrane electric potential (ΔΨ(t)) as well as pH in the lumen (pHL(t)) and stroma (pHS(t)). PS II model parameters and numerical coefficients in ΔΨ(t), pHL(t), and pHS(t) were evaluated to fit fluorescence induction data for different experimental conditions: leaf in vivo or after ionophore treatment at low or high light intensity. The model imitated changes in the pattern of fluorescence induction rise due to the elimination of transmembrane potential in the presence of ionophores, when ΔΨ = 0 and pHL(t), pHS(t) changed to small extent relative to control values in vivo, with maximum ΔΨ(t) ∼ 90 mV and ΔΨ(t) ∼ 40 mV for the stationary state at ΔpH ≅ 1.8. As the light intensity was increased from 300 to 1200 μmol m−2 s−1, the heat dissipation rate constants increased threefold for nonradiative recombination of P680+Phe and by ∼30% for P680+QA. The parameters ΔΨ, pHS and pHL were analyzed as factors of PS II redox state populations and fluorescence yield. The kinetic mechanism of fluorescence quenching is discussed, which is related with light-induced lumen acidification, when +QA and P680+ recombination probability increases to regulate the QA reduction.  相似文献

Genotypic variations were observed among 6 variants and an indigenous one as a control. An efficient genotype OCD(L) of Ocimum sanctum had the greatest content of chlorophylls [2.15 g kg−1(FM)], Chl a/b ratio of 1.86, net photosynthetic rate of 0.72 mg(CO2) m−2 s−1, and 1.56 % oil formation. Methyl chavicol formed 50.31 % of total oil in OSP-6 genotype, which had also maximum peroxidase activity [ΔOD 2.4 mg−1(protein)] and maximum production of eugenol (30.44 % of total oil). We found an oxido-reducible reaction of peroxidase and high bands of peroxidase isoenzymes in this OSP-6 genotype for the formation of monoterpene essential oil(s) and possibly the major constituents of eugenol through the high production of photosynthates.  相似文献

The accumulation of copper, zinc and cadmium inA. yokoscense collected from Ashio (copper-contaminated area), Bandai (zinc- and cadmium-contaminated area) and Tama (non-contaminated area), has been investigated. Copper and zinc were accumulated most highly in the root, whilst cadmium was accumulated more in the leaf. The root ofA. yokoscense growing in areas contaminated with metals contained maximum amounts of Cu (5, 989 mg. kg−1 dry weight) and Zn (6,384 mg.kg−1 dry weight), while in the leaf from the Bandai area 164.8 mg Cd.kg−1 dry weight was accumulated. These amounts are far greater than those found inA yokoscense growing on the non-metalliferous habitat (Tama). Twenty five times more zinc and three times more cadmium were found in the dead leaf than in the living leaf. InA. yokoscense growing on soils containing more than 1,000 mg Cu or Zn.kg−1 dry weight, the uptake of copper by the root increased considerably with increasing copper content in the soils, while the uptake of zinc increased only slightly compared with the increase of zinc in the soils.  相似文献

La Paz Bay and La Paz Lagoon are water bodies of the Gulf of California that are influenced by waste water discharges from the City of La Paz and from activities of the phosphorite mining company “Rofomex”. Because seaweeds concentrate elements from the water and are used as effective indicators of contamination by metals, we investigated their usefulness in this region. Concentrations of certain major elements (Ca, Fe, K and Na) and trace elements (As, Ba, Co, Cr, Cs, Hf, Rb, Sb, Sc, Se, Sr, Ta, Th, U, Zn and Zr) were determined in 12 species of seaweeds from La Paz Bay and La Paz Lagoon using instrumental neutron activation analysis. The contents of trace elements of environmental importance (As, Co, Cr, Fe, Sb, Se and Zn) in all studied samples are within the range of typical levels for a pristine environment not subjected to anthropogenic impact. Somewhat higher concentrations of Cr (81mg kg−1), Hf (4mg kg−1), Rb (48mg kg−1), Sc (6.3mg kg−1), Ta (0.95mg kg−1), Th (6.8mg kg−1), U (33mg kg−1) and Zn (90mg kg−1) were found in the green seaweed species Ulva (formerly Enteromorpha) intestinalis, whereas such elements as As (77mg kg−1), Sb (1.4mg kg−1) and Se (1.8mg kg−1) were mainly concentrated in the species Sargassum sinicola, Codium cuneatum and Padina mexicana respectively. Because of their higher abundance and heterogeneity in elemental composition the seaweeds species Ulva intestinalis and Caulerpa sertularioides seem to be more suitable for further biomonitoring of heavy metal pollution of the coastal waters in this zone.  相似文献

A crown rot disease in wheat caused by the fungusFusarium graminearum Schw. Group 1 is a widespread problem in chronically Zn-deficient Australian soils. A link between crown rot and Zn deficiency was established by Sparrow and Graham (1988). This paper reports a test of a further hypothesis, that wheat genotypes more efficient at extracting zinc from low-zinc soils are more resistant to infection by this pathogen. Three wheat cultivars (Excalibur, Songlen and Durati) of differential Zn efficiency were tested at three zinc levels (0.05, 0.5 and 2.0 mg Zn kg−1 of soil) and three levels ofF. graminearum S. Group 1 inoculum (0.1 g and 0.3 g kg−1 live chaff-inoculum and control having 0.1 g kg−1 dead chaff inoculum). Six weeks after sowing dry matter production of shoots and roots was decreased byFusarium inoculation at 0.05 mg and 0.5 mg kg−1 applied Zn.Fusarium inoculum at 0.1 g was as effective as 0.3 g kg−1 for infection and decreasing dry matter. The infection at the basal part of culm decreased significantly by increasing the rate of Zn application. Excalibur, a Zn-efficient cultivar (tolerant to Zn deficiency) produced significantly more shoot and root dry matter, and showed less disease infection compared with Zn-inefficient cultivars (Durati and Songlen) at low (0.05 mg Zn kg−1 soil) and medium (0.5 mg Zn kg−1 soil) Zn fertilization rates. Higher rate of Zn fertilization (2.0 mg Zn kg−1 soil) reduced the disease level in Durati to the level of Excalibur but the disease level of Songlen was still high, indicating its high Zn requirement and or sensitivity to crown rot. The data on Zn uptake show that Excalibur, being Zn-efficient, was able to scavenge enough Zn from Zn-deficient soil, we suggest that besides sustaining growth Excalibur was able to build and maintain resistance to the pathogen; inefficient cultivars needed extra Zn fertilization to achieve performance comparable to that of Excalibur. The present study indicates that growing Zn-efficient cultivars of wheat along with judicious use of Zn fertilizer in Zn-deficient areas where crown rot is a problem may sustain wheat production by reducing the severity of the disease as well as by increasing the plant vigour through improved Zn nutrition. ei]Section editor: R Rodriques-Kalana  相似文献

Heavy metals are widely recognized as potential toxic agents to zooplankton, yet experiments are usually performed with a continuous exposure to the metal being analyzed. Here we describe experiments that examined the influence of pulsed exposure of the heavy metals copper and cadmium to a parthenogenetic population of the planktonic rotifer Brachionus calyciflorus. Our protocol called for exposure durations of 3, 6, 12, and 24 h to either copper (as CuSO4) at concentrations of 0.0375, 0.075, 0.15 mg l−1 or cadmium (as CdCl2) at concentration of 0.025, 0.05, 0.1 mg l−1. Control animals were treated in similar ways but did not receive exposure to heavy metals. Four end points were used to evaluate the outcome of exposure: population growth (r), body size, egg ratio, and egg hatching percent. Increase in heavy metal concentration and exposure time had an adverse influence on the population growth of B. calyciflorus. However, while the response of B. calyciflorus was similar for both heavy metals, the magnitude of the impact of cadmium was more severe. Population growth varied depending on which heavy metal was tested, as well as its concentration and the duration of exposure (r = 0.11–0.28 day−1). There was a significant reduction in lorica size of B. calyciflorus subjected to different exposure times and concentrations of both Cd and Cu. Egg ratios were inversely related to population density in controls and in treatments involving Cu, but not for Cd. While nearly 100% of eggs hatched in the control treatments, egg hatching in experimental treatments containing Cu, were reduced (range = 16–41%) depending on the exposure time and the concentration. The peptides Asp-Ala-His-Lys (DAHK) and Gly-His-Lys (GHK) are naturally occurring Cu(II)-chelating motifs in human serum and cerebrospinal fluid. Here, the sensitive thermodynamic technique isothermal titration calorimetry was used to study the energetics of Cu(II) binding to DAHK and GHK peptides in the presence of the weaker ligand glycine as a competitor. DAHK and GHK bind Cu(II) predominantly in a 1:1 stoichiometry with conditional dissociation constants [i.e., at pH 7.4, in the absence of the competing chelators glycine and 2-(4-(2-hydroxyethyl)-1-piperazinyl)ethanesulfonic acid buffer] of 2.6 ± 0.4 × 10−14 M and 7.0 ± 1.0 × 10−14 M, respectively. Furthermore, the apparent ΔH values were measured and the number of protons released upon Cu(II) binding was determined by performing experiments in different buffers. This allowed us to determine the conditional ΔG, ΔH, and ΔS, i.e., corrected for the contributions of the weaker ligand glycine and the buffer at pH 7.4. We found that the entropic and enthalpic contributions to the Cu(II) binding to GHK and DAHK are distinct, with a enthalpic contribution for GHK. The thermodynamic parameters obtained correspond well to those in the literature obtained by other techniques, suggesting that the use of the weaker ligand glycine as a competitor in isothermal titration calorimetry provides accurate data for Cu(II) binding to high-affinity peptides, which cannot be accurately determined without the use of a competitor ligand.  相似文献

Three hundred sixty healthy Ross×Ross 1-day-old broilers were used to study the effects of zinc glycine chelate (Zn-Gly) on growth performance, hematological, and immunological characteristics. All broilers were randomly assigned into six treatments. Diets were as follows: (1) control (containing 29.3 mg Zn kg−1 basic diet [0–3 weeks] and 27.8 mg Zn kg−1 [4–6 weeks]); (2) basic diet plus 30 mg Zn kg−1 from Zn-Gly; (3) basic diet plus 60 mg Zn kg−1 from Zn-Gly; (4) basic diet plus 90 mg Zn kg−1 from Zn-Gly; (5) basic diet plus 120 mg Zn kg−1 from Zn-Gly; (6) positive control, basic diet plus 120 mg Zn kg−1 from zinc sulfate (ZnSO4). After the 21- and 42-day feeding trials, the results showed that both of Zn-Gly and ZnSO4 could improve the growth performance of broilers, with the greatest average daily feed intake observed in the broilers fed 90 mg Zn kg−1 from Zn-Gly, but the greatest average daily gain observed with 120 mg Zn kg−1 from Zn-Gly (0–3 weeks) and 90 mg Zn kg−1 from Zn-Gly (4–6 weeks). Adding additional Zn-Gly improved the levels of immunoglobulins (IgA, IgM, and IgG) and the contents of total protein and Ca in serum and increased the immune organs index especially with 90 mg Zn kg−1 as Zn-Gly. However, there were no significant differences in responses to complements (C3 and C4) and albumin in serum among the treatments.  相似文献

This paper gives the results from four-year field experiments on compost application, added at the maximum rate allowed by Italian legislation (30 t/ha/y). The purpose of the experiment was to evaluate any eventual heavy metal accumulation in soil and corn plants. Cadmium in corn plants increased particularly in the roots from 0.22 mg kg−1 to 1.31 mg kg−1, concentration of Zn and Cu increased in grains, from 26.8 to 35.8 and from 2.4 to 4.2 mg kg−1 respectively. Relevant increase in the roots was detected for Zn from 34.6 to 146.8 mg kg−1. Only in the 4th year Ni concentration increases in the root portion while the content of Pb and Cr in corn was generally unaffected by the compost application. Heavy metals in the soil determined by a sequential chemical extraction, indicated that extractability changed with time. A certain increase was found from the beginning to the end of the experiment particularly for Zn, from 23.3 mg kg−1 to 45.1 mg kg−1 in extractable forms. This experiment was conducted to evaluate the effect of zinc, manganese, and copper sources (inorganic vs. organic) in the diet on laying performance and eggshell quality characteristics. One hundred and eighty Hy-Line W-36 layers at 38 weeks of age were allocated to 36-layer cages of five hens each. Each six cages were randomly assigned to one of the six experimental diets fed from 38 to 53 week of age. In three experimental treatments, the basal diet was supplemented with 65–75–7 or 65–75–7 or 40–40–7 mg/kg of Zn, Mn, and Cu, respectively, from their oxide or sulfate sources. Three other groups were fed diets supplemented with 20–20–3.5 or 40–40–7.5 or 60–60–10.5 mg/kg of organic forms of Zn, Mn, and Cu, respectively. Dietary treatments significantly did affect feed intake (P < 0.001), feed conversion ratio (P < 0.001) and percentage of broken eggs (P < 0.05). Substitution of Zn and Mn oxides (65 and 75 mg kg−1, respectively) with equal amounts of their sulfate forms significantly improved feed intake, feed conversion ratio, percentage of broken eggs, and Haugh Unit (P < 0.05). In addition, laying hens maintained their performance when substitution of Zn and Mn oxides and Cu sulfate (65, 75, and 7 mg kg−1, respectively) reduced up to 20, 20, and 3.5 mg kg−1 by amino acid complexes of the microelements. The results showed that a corn–soybean diet supplemented with the organic forms of Zn, Mn, and Cu at a dosage 50% to 75% lower than NRC recommendation is sufficient to maintain laying performance and can improve eggshell and albumen qualities of the egg in laying hens.  相似文献

Two respiratory-deficient nuclear petites, FY23Δpet191 and FY23Δcox5a, of the yeast Saccharomyces cerevisiae were generated using polymerase-chain-reaction-mediated gene disruption, and their respective ethanol tolerance and productivity assessed and compared to those of the parental grande, FY23WT, and a mitochondrial petite, FY23ρ0. Batch culture studies demonstrated that the parental strain was the most tolerant to exogenously added ethanol with an inhibition constant. K i, of 2.3% (w/v) and a specific rate of ethanol production, q p, of 0.90 g ethanol g dry cells−1 h−1. FY23ρ0 was the most sensitive to ethanol, exhibiting a K i of 1.71% (w/v) and q p of 0.87 g ethanol g dry cells−1 h−1. Analyses of the ethanol tolerance of the nuclear petites demonstrate that functional mitochondria are essential for maintaining tolerance to the toxin with the 100% respiratory-deficient nuclear petite, FY23Δpet191, having a K i of 2.14% (w/v) and the 85% respiratory-deficient FY23Δcox5a, having a K i of 1.94% (w/v). The retention of ethanol tolerance in the nuclear petites as compared to that of FY23ρ0 is mirrored by the ethanol productivities of these nuclear mutants, being respectively 43% and 30% higher than that of the respiratory-sufficient parent strain. This demonstrates that, because of their respiratory deficiency, the nuclear petites are not subject to the Pasteur effect and so exhibit higher rates of fermentation.
