An investigation was conducted into the isolation of plasma membrane vesicles from primary roots of corn (Zea mays L., WF9 × M14) by sucrose density gradient centrifugation. Identification of plasma membranes in cell fractions was by specific staining with the periodic-chromic-phosphotungstic acid procedure. Plasma membrane vesicles were rich in K+-stimulated ATPase activity at pH 6.5, and equilibrated in linear gradients of sucrose at a peak density of about 1.165 g/cc. It was necessary to remove mitochondria (equilibrium density of 1.18 g/cc) from the homogenate before density gradient centrifugation to minimize mitochondrial contamination of the plasma membrane fraction. Endoplasmic reticulum (NADH-cytochrome c reductase) and Golgi apparatus (latent IDPase) had equilibrium densities in sucrose of about 1.10 g/cc and 1.12 to 1.15 g/cc, respectively. A correlation (r = 0.975) was observed between K+-stimulated ATPase activity at pH 6.5 and the content of plasma membranes in various cell fractions. ATPase activity at pH 9 and cytochrome c oxidase activity were also correlated.  相似文献

The effects of two host plant species, immature rearing density, and feeding-induced leaf damage on the offspring sex ratios of individual Banks grass mites (BGM) (Oligonychus pratensis Banks) were examined. Offspring sex ratios did not differ among corn-reared females transferred to corn or two sorghum genotypes. Likewise, the offspring sex ratios did not differ among females from three rearing densities. However, female BGM reared on damaged leaves produced significantly more daughters than females reared on undamaged leaves. This suggests that offspring sex ratios of BGM change in response to a deteriorating food source. Greater daughter production on a deteriorating host results in more potential dispersers and colonizers of new host plants. This may be an important adaptation facilitating the colonization-dispersal episodes that characterize the BGM life history.  相似文献

Ambulatory dispersal behavior ofNeoseiulus fallacis (Garman) was studied in the laboratory to evaluate within-plant movement in relation to temperature and prey density. Adult femaleN. fallacis were confined in 2.5-cm-diameter arenas on the abaxial surface of excised corn leaves. Four temperatures (23, 28, 33, and 39° C) and prey densities ranging from 0 to 55 spider mite eggs per cm2 were used. The walking paths of these mites were traced, digitized and used to calculate turning angles, walking speeds and turning rates. A computer simulation of walking behavior used this information to model mite ambulatory behavior and predict dispersal rates.Neoseiulus fallacis behavior while on whole corn leaves was quantified to verify the results of the simulation. The results showed thatN. fallacis will follow a leaf or arena edge (edge-walking) at all temperatures and prey densities. In addition, this behavior was used to the exclusion of the other types of behavior such as resting, and random-walk type search when prey egg density was less than 4 eggs per cm2. The exclusion of edge-walking behavior from the model caused the model to underestimate substantially the dispersal rates leaves. These data suggest that there are at least two recognizable types of ambulatory search used byN. fallacis—the random-walk type, which is used when prey density is high (searching within prey patches), and the edge-walking behavior, which is used when prey density is low. This behavior allows the mite to travel rapidly from leaf to leaf in search of new prey patches.  相似文献

Summary The uptake of Ca and Sr by three-week old tomato (Lycopersicon esculentum) plants from solutions containing Ca++ and Sr++, and chelated Ca and Sr (CaL and SrL) was measured over a two-day period. The solution was double-labelled with Ca45 and Sr85. Two chelates, EDTA (ethylenediaminetetraacetic acid) and DTPA (diethylenetriaminepentaacetic acid) were used at five chelate-cation ratios. When the Ca and Sr content of the solution was held constant, addition of chelate reduced uptake. The reduction was greater with EDTA than with DTPA.The Ca/Sr ratio of uptake was used to measure the proportion of uptake as the chelated and unchelated species. The Ca++/Sr++ ratio was different from the CaL/SrL ratio in solution because of the different equilibrium reactions of Ca and Sr with L. Direct uptake of the CaL and SrL was indicated. In solutions where Ca++ = CaL, uptake of CaEDTA was 0.47 of uptake of Ca++ and uptake of CaDTPA was 0.95 of uptake of Ca++.Journal Paper No. 4969. Young cabbage plants (Brassica oleracea L. var. capitata) that were exposed to an atmosphere at 50% relative humidity transpired freely and accumulated significant quantities of 45Ca in the leaves. Plants that were enclosed by plastic bags to stop transpiration from all leaves exhibited guttation with the development of root pressure and also accumulated significant quantities of 45Ca in the leaves. 45Ca accumulation increased in the leaves and tended to decrease in roots and stems with increasing quantities of water transpired or guttated by the plant. When plants were only partially enclosed so that some leaves were covered and the remainder exposed, only the exposed leaves that were transpiring accumulated significant quantities of 45Ca. The covered leaves of partially enclosed plants exhibited no guttation and accumulated little 45Ca with no measurable 45Ca at the margins of the leaves. The results demonstrate that root pressure flow is required to transport adequate amounts of Ca to those tissues in plants that are not undergoing transpirational water loss.  相似文献

Calcium ion activity was measured in protein-free and protein-containing Krebs-Ringer bicarbonate buffer. Even in protein-free buffer calcium ion activity was 20% less than the total Ca concentration. In solutions containing albumin the calcium ion activity was dramatically reduced, an effect dependent on the albumin concentration and pH of the solution. Acutely changing calcium ion activity from 0.35 mM to 1.25 mM in a Krebs-Ringer bicarbonate buffer (5.4% albumin) perfusing the fetal side of the guinea pig placenta resulted in an immediate increase in transfer (mother to fetus) of 32P but no change in 45Ca transfer. The ratio of 32P to 45Ca clearance was positively related also to maternal plasma calcium ion activity.  相似文献

Summary Small birch plants (Betula pendula Roth.) were grown in a climate chamber at different, exponentially increasing rates of nitrogen supply and at different photon flux densities. This resulted in treatments with relative growth rate equal to the relative rate of increase in nitrogen supply and with different equilibrium values of plant nitrogen concentration. Nitrogen productivity (rate of dry matter increase per plant nitrogen) was largely independent of nitrogen supply and was greater at higher photon flux density. Leaf weight ratio, average specific leaf area (and thus leaf area ratio) were all greater at better nitrogen supply and at lower values of photon flux density. The dependencies were such that the ratio of total projected leaf area to plant nitrogen at a given photon flux density was similar at all rates of nitrogen supply. The ratio was greater at lower values of photon flux density. At a given value of photon flux density, net assimilation rate and net photosynthetic rate per shoot area (measured at the growth climate) were only slightly greater at better rates of nitrogen supply. Values were greater at higher photon flux densities. Acclimation of the total leaf area to plant nitrogen ratio and of net assimilation rate was such that nitrogen productivity was largely saturated with respect to photon flux density at values greater than 230 mol m-2 s-1. At higher photon flux densities, any potential gain in nitrogen productivity associated with higher net assimilation rates was apparently offset by lower ratios of total leaf area to plant nitrogen.  相似文献

Arboreal arthropod communities were censused by insecticidal knockdown in a plantation of Japanese cedar,Cryptomeria japonica, in central Japan from April 1983 to February 1985 at intervals of two months. All arthropods sampled were allocated to higher taxonomic groups or guilds and the composition of the communities was analyzed with respect to abundance, biomass and their seasonal trends. The total densities fluctuated seasonally from 200 to 3500 m−2; corresponding biomass values ranged from 7 to 600 mg fresh weight m−2. Maximum density and biomass generally occurred in summer and were minimum in winter. The consistently dominant guild in terms of abundance were the detritivores, mostly comprising Collembola and oribatid mites, accounting for 36–93% of all arthropods; phytophages, predators and detritivores were major guilds in terms of biomass, showing average proportions of 27%, 23% and 20%, respectively. Biomass ratios of predators to prey were generally high in some canopy communities, suggesting the importance of predation pressure in regulating the population levels of arthropods in forests. Average individual size of predators was approximately proportional to that of prey, irrespective of tree locality. Soil arthropod communities maintained densities approximately 102–103 times as large as the corresponding canopy communities throughout the year. The seasonal variations in abundance were much greater in canopy than in soil communities.  相似文献

Snapping turtles Elseya dentata (Gray) from Magela Creek, Northern Territory, were exposed under laboratory conditions for up to 30 days to waters resembling the inorganic composition of Magela Creek water during the Wet season, with background and elevated Ca and Mg concentrations, that were labelled with 226Ra and 45Ca. The resulting concentrations of 45Ca in muscle, skin, gut, liver, shell bone and leg bone of E. dentata equilibrated or approached equilibrium by 12–18 days. Among the experiments, the concentrations of 45Ca in all six tissues were inversely related to turtle mass. An increase in the Ca water concentration by a factor of 15 increased the 45Ca concentration in all six tissues. The arithmetic factors of increase in the concentration in each tissue were proportional or more than proportional to the factor of increase in Ca water concentration; this factor was highest for muscle tissue (26.6). An increase in the Mg water concentration by a factor of 48 reduced the 45Ca concentration in all tissues except skin where it increased. The concentration of 226Ra in each tissue (except the gut) was positively related to its 45Ca concentration and inversely related to turtle mass in muscle, skin and liver. With the exception of the skin, the increased Ca water concentration did not reduce the 226Ra in the tissues but increased the 226Ra concentration in bone and muscle. The increased Mg water concentration had an inverse effect on the 226Ra concentrations in all tissues except shell. With the exception of the skin the effects of increased Ca and Mg water concentrations and turtle size on 226Ra concentrations in the tissues of E. dentata were similar to their effects on 45Ca tissue concentrations, indicating the similar metabolic behaviour of 226Ra and 45Ca in E. dentata.Exposures of the species Elseya latisternum (Gray), Emydura signata (Ahl) and Chelodina longicollis (Shaw), which are the same or closely related to species reported to occur in Magela Creek, to 45Ca-labelled Sydney tap water for 7 days demonstrated their ability to also accumulate 45Ca from their aquatic medium. The patterns of 45Ca concentrations in the tissues of these species indicated that they were inversely related to turtle mass, as demonstrated in E. dentata. The concentrations of 45Ca accumulated in the tissues were also comparable to those found in single specimens of E. dentata and E. victoriae (Gray) that were exposed for 7 days to simulated Magela Creek water. The data also indicated the larger long-necked C. longicollis accumulated less 45Ca per gram of tissue than similar-sized, short-necked species E. signata and E. latisternum, suggesting that long-necked turtles from Magela Creek would accumulate less 226Ra from their aquatic medium than similar-sized short-necked species.The capacity of E. dentata to accumulate 226Ra from the aquatic medium is about two orders of magnitude less than that of the tissue of the freshwater mussel Velesunio angasi (Sowerby) exposed under similar experimental conditions.  相似文献

Fungi are the dominant organisms decomposing leaf litter in streams and mediating energy transfer to other trophic levels. However, less is known about their role in decomposing submerged wood. This study provides the first estimates of fungal production on wood and compares the importance of fungi in the decomposition of submerged wood versus that of leaves at the ecosystem scale. We determined fungal biomass (ergosterol) and activity associated with randomly collected small wood (<40 mm diameter) and leaves in two southern Appalachian streams (reference and nutrient enriched) over an annual cycle. Fungal production (from rates of radiolabeled acetate incorporation into ergosterol) and microbial respiration on wood (per gram of detrital C) were about an order of magnitude lower than those on leaves. Microbial activity (per gram of C) was significantly higher in the nutrient-enriched stream. Despite a standing crop of wood two to three times higher than that of leaves in both streams, fungal production on an areal basis was lower on wood than on leaves (4.3 and 15.8 g C m−2 year−1 in the reference stream; 5.5 and 33.1 g C m−2 year−1 in the enriched stream). However, since the annual input of wood was five times lower than that of leaves, the proportion of organic matter input directly assimilated by fungi was comparable for these substrates (15.4 [wood] and 11.3% [leaves] in the reference stream; 20.0 [wood] and 20.2% [leaves] in the enriched stream). Despite a significantly lower fungal activity on wood than on leaves (per gram of detrital C), fungi can be equally important in processing both leaves and wood in streams.  相似文献

Tomato (Lycopersicon esculentum Mill. cv. Moneymaker) and maize (Zea mays L. cv. spec.) plants were supplied with 45Ca-labeled nutrient solutions for a period of 8 or 16 h in the dark, in the light, or in a light-dark régime. Plant parts were analyzed for 45Ca content. The partitioning of 45Ca between mature leaves and meristems was shown to be affected by the presence of light. The transport of 45Ca to meristems was higher in a dark period than in a comparable light period. Experiments with excised tomato shoots yielded similar distribution patterns of 45Ca over leaves and meristems, thus excluding root pressure as the main driving force for the enhanced import of 45Ca into the meristems in the dark. We investigated the linkage between the detrital subsystem and generalist predators of meadow ecosystems by manipulating prey availability in two different ways: we increased resource availability for the decomposer subsystem and thereby decomposer prey by adding mulch materials (detritus enhancement), and we added fruitflies (Drosophila melanogaster, Diptera; prey enhancement) to fenced plots. Both supplemented materials significantly differed in their 13C/12C and 15N/14N ratios from those of the natural litter. We measured density responses of detritivorous, herbivorous and predaceous arthropods to the increased resource supply. We used ratios of natural stable isotopes of N and C in arthropod tissues to trace the flux from the added resources to consumers and to relate density responses of consumers to changes in resource supply. Effects of resource enhancement propagated through at least two trophic levels, resulting in higher densities of major decomposer and predator taxa. Effects of detritus enhancement were much stronger than those of prey enhancement. Signatures of δ13C proved density responses of Collembola taxa to be related to the added mulch materials. Among generalist predators, densities of juvenile wolf spiders (Lycosidae) responded more to detritus-enhancement than to prey-enhancement treatments. In contrast, the density of the web-building linyphiid and the non-web gnaphosid spiders remained unaffected. Each spider taxon, including those which did not respond numerically, was significantly enriched in 13C in detritus-enhancement treatments, suggesting that they gain energy from the decomposer system. Numbers of herbivores—cicadellids and aphids—were similar in each of the treatments, indicating that they were unaffected by changes in predator density. Our results indicate that the lack of a numerical response to resource supplementation is not necessarily due to the absence of a trophic linkage, but may be caused by compensatory changes in mortality factors such as cannibalism and intraguild predation.  相似文献

Flooding effects on membrane permeability, lipid peroxidation and activated oxygen metabolism in corn (Zea mays L.) leaves were investigated to determine if activated oxygens are involved in corn flooding-injury. Potted corn plants were flooded at the 4-leaf stage in a controlled environment. A 7-day flooding treatment resulted in a significant increase in chlorophyll breakdown, lipid peroxidation (malondialdehye content), membrane permeability, and the production of superoxide (O 2 - ) and hydrogen peroxide (H2O2) in corn leaves. The effects were much greater in older leaves than in younger ones. Spraying leaves with 8-hydroxyquinoline (an O 2 - scavenger) and sodium benzoate (an .OH scavenger) reduced the oxidative damage and enhanced superoxide dismutase (SOD) activity. A short duration flooding treatment elevated the activities of SOD, catalase, ascorbate peroxidase (AP), and glutathione reductase (GR), while further flooding significantly reduced the enzyme activities but enhanced the concentrations of ascorbic acid and reduced form glutathione (GSH). It was noted that the decline in SOD activity was greater than that in H2O2 scavengers (AP and GR). The results suggested that O 2 - induced lipid peroxidation and membrane damage, and that excessive accumulation of O 2 - is due to the reduced activity of SOD under flooding stress.  相似文献

Summary Tomato plants were grown in water-culture with a different supply of Ca (10, 100 ppm) and B (0, 0.2 ppm), and the effects of B deficiency on the translocation and subcellular distribution of Ca in tomato plants were studied by using45CaCl2 as a carrier of Ca. Boron deficiency slight increased the total Ca uptake by the plant and inhibited the Ca translocation to the upper leaves. The incorporation of45Ca into the cell wall in the upper leaves was increased by B deficiency at both Ca levels. As Ca supply decreased, the distribution of45Ca in the 1N NaCl fraction of the cell wall increased only at 0.2 ppm B. As B supply decreased, the distribution of45Ca in the 0.6N HCl fraction increased at both Ca levels. These results suggest that B deficiency inhibit the translocation of Ca, and induce the abnormal changes of the Ca metabolism in the cell wall.  相似文献

Conjugal plasmid transfer was examined on the phylloplane of bean (Phaseolus vulgaris) and related to the spatial distribution pattern and metabolic activity of the bacteria. The donor (Pseudomonas putida KT2442) harbored a derivative of the TOL plasmid, which conferred kanamycin resistance and had the gfp gene inserted downstream of a lac promoter. A chromosomal insertion of lacIq prevented expression of the gfp gene. The recipient (P. putida KT2440) had a chromosomal tetracycline resistance marker. Thus, transconjugants could be enumerated by plating and visualized in situ as green fluorescent cells. Sterile bean seedlings were inoculated with donors and recipients at densities of approximately 105 cells per cm2. To manipulate the density and metabolic activity (measured by incorporation of [3H]leucine) of the inoculated bacteria, plants were grown at various relative humidities (RH). At 100% RH, the transconjugants reached a density of 3 × 103 CFU/cm2, corresponding to about one-third of the recipient population. At 25% RH, numbers of transconjugants were below the detection limit. Immediately after inoculation onto the leaves, the per-cell metabolic activity of the inocula increased by up to eight times (100% RH), followed by a decrease to the initial level after 96 h. The metabolic activity of the bacteria was not rate limiting for conjugation, and no correlation between the two parameters was observed. Apparently, leaf exudates insured that the activity of the bacteria was above a threshold value for transfer to occur. Transconjugants were primarily observed in junctures between epidermal cells and in substomatal cavities. The distribution of the transconjugants was similar to the distribution of indigenous bacteria on nonsterile leaves. Compared to polycarbonate filters, with cell densities equal to the overall density on the leaves, transfer ratios on leaves were up to 30 times higher. Thus, aggregation of the bacteria into microhabitats on the phylloplane had a great stimulatory effect on transfer.  相似文献

Activity ratios and carbamylation ratios of ribulose-1,5-bisphosphate carboxylase (RuBPCase) were determined for leaves of Phaseolus vulgaris and Spinacia oleracea exposed to a variety of partial pressures of CO2 and O2 and photon flux densities (PFD). It was found that activity ratios accurately predicted carbamylation ratios except in extracts from leaves held in low PFD. In particular, it was confirmed that the loss of RuBPCase activity in low partial pressure of O2 and high PFD results from reduced carbamylation. Activity ratios of RuBPCase were lower than carbamylation ratios for Phaseolus leaves sampled in low PFD, presumably because of the presence of 2-carboxyarabinitol 1-phosphate. Spinacia leaves sampled in darkness also exhibited lower activity ratios than carbamylation ratios indicating that this species may also have an RuBPCase inhibitor even though carboxyarabinitol 1-phosphate has not been detected in this species in the past.  相似文献

Specialized microbodies have previously been isolated and characterized from fatty seedling tissues (glyoxysomes) and leaves (leaf peroxisomes). We have now examined 11 other plant tissues, including tubers, fruits, roots, shoots, and petals, and find that all contain particulate catalase, a distinctive common enzyme component of microbodies. On linear sucrose gradients the catalase activity peaks sharply at a higher equilibrium density (1.20 to 1.25 gram per cm3 in the various tissues) than the mitochondria (1.17 to 1.20). Only small amounts of protein are recovered in the fractions containing catalase, although a definite band is visible in preparations from some tissues, e.g., potato. As in the preparations from castor bean endosperm and spinach leaves for which comparable data are provided, the distribution of glycolate oxidase and uricase follows closely that of catalase on the gradients. The preparations from potato lack glyoxylate reductase and the transaminases, typical enzymes of leaf peroxisomes, and the distinctive enzymes of glyoxysomes are missing. Nonspecialized microbodies with limited enzyme composition can thus be isolated from a variety of plant tissues.  相似文献

The effects of decreasing Ca2+ concentrations (Na/Ca ratios were 24, 49, 99 and 199) of the saline growth medium (NaCl concentration 200 mM) on three grass speciesCenchrus pennisetiformis Hochst. & Steud,Leptochloa fusca L. Kunth. andPcmicum turgidum Forssk. were assessed after 7 weeks growth in sand culture.L. fusca produced the highest dry biomass of all the species at varying Na/Ca ratios. Number of tillers per plant and number of green leaves per tiller were reduced significantly only inC. pennisetiformis. Leaf water potential ofC. pennisetiformis decreased at all external Na/Ca ratios, whereas inL. fusca it decreased only at an Na/Ca ratio of 99. Leaf osmotic potential ofL. fusca consistently decreased at all Na/Ca ratios, whereas that of the other two species remained unchanged. The shoot and root total sugars of all species remained unaffected at all decreasing Ca2+ concentrations. InP. turgidum chlorophyllb and total chlorophyll decreased consistently at all Na/Ca ratios, but inL. fusca they decreased only at the highest Na/Ca ratios. The leaf soluble proteins of all the species remained unaffected at all Na/Ca ratios. The leaf free amino acids decreased significantly inL. fusca with the increase in Na/Ca ratios. The leaf proline content was only decreased inL. fusca at the highest Na/Ca ratio. The significant correlations between the growth of the three grass species and other variables determined in this study were not found.  相似文献

The aim of the study was to the assess the influence of Ca/Mg ions ratio on the photosynthetic activity of Salix viminalis L. ??Cannabina?? plants cultivated in medium enriched with Cu(NO3)2. The experiment was conducted in controlled conditions in a phytotron for 21 days; hence the early plant response was tested. Plants were cultivated with different Ca/Mg ions ratios, i.e. (4:1)l, (4:1)h, and 1:10. Plants were additionally treated with Cu(NO3)2 at 1, 2, and 3 mM concentration in cultivation medium. Net photosynthetic rate, stomatal conductance and transpiration were measured after the first, second and third week of cultivation. Additionally, chlorophyll content, leaf morphology, root biomass and copper accumulation in leaves and roots were investigated. The investigations revealed differences in plant response to particular treatments ?? differences in Cu accumulation for particular Ca/Mg ions ratios were detected. It seems that plants are adapted to high Cu2+ concentrations, when 1:10 Ca/Mg ions ratio is applied. The highest Cu accumulation in roots was noted for plants fertilized with 1:10 Ca/Mg ions ratio, together with high Cu translocation to above-ground plant organs, which suggests its higher potential in phytoremediation.  相似文献
