用高效液相色谱法和分光光度法测定了四种加工方法所得山蓝加工提取物中有效成分含量 ,结果表明 ,经阴干 ,晒干 ,乙醇浸泡几种传统加工方法加工的山蓝中紫蓝素含量分别为 31 1、4 5 6、4 72 mg/1 0 0 g(干重 ) ,而采用避氧热处理加工的山蓝中 ,紫蓝素含量达 1 .73g/1 0 0 g。用传统加工方法对山蓝进行加工 ,有效成分含量低 ,制约了山蓝的利用价值 ,而避氧热处理能大大提高有效成分含量 ,提升山蓝的品质。研究结果对山蓝的加工、炮制和开发利用有重大意义。  相似文献

A method is described for the accurate estimation of protein samples ranging in size from 1 to 100 μg. It is based on fluorescence quenching of eosin Y by protein and modifies a method originally published by Hiraoka and Glick. The modified method is convenient to use. It is carried out with a stabilized reagent in cuvettes of standard size and permits the estimation of 5 μg or less of protein with precision of ± 9%. The measurements are shown to be sensitive to slight variations in ambient temperature and these contribute at least 3% to the error term shown. A graphical manner of presenting the data has been devised that provides a linear relationship over nearly 50-fold range of protein concentrations. Quenching is unaffected by presence of glucose or urea and partially inhibited by glycine or mercaptoethanol only if these are present in better than 100-fold excess over protein. Different proteins show different degrees of quenching and the method is thus most suitable for comparing quantitatively protein mixtures of similar composition. The modified method should be useful not only where microgram quantities of protein must be measured, but also with automated equipment or in cases where the Folin-Ciocalteau method cannot be employed due to presence of interfering substances.  相似文献

Coumaphos is a common organophosphorus pesticide used in agricultural products. It is harmful to human health and has a strictly stipulated maximum residue limit (MRL) on fruits and vegetables. Currently existing methods for detection are complex in execution, require expensive tools and are time consuming and labor intensive. The surface plasmon resonance method has been widely used in biomedicine and many other fields. This study discusses a detection method based on surface plasmon resonance in organophosphorus pesticide residues. As an alternative solution, this study proposes a method to detect Coumaphos. The method, which is based on surface plasmon resonance (SPR) and immune reaction, belongs to the suppression method. A group of samples of Coumaphos was detected by this method. The concentrations of Coumaphos in the samples were 0 µg/L, 50 µg/L, 100 µg/L, 300 µg/L, 500 µg/L, 1000 µg/L, 3000 µg/L and 5000 µg/L, respectively. Through detecting a group of samples, the process of kinetic reactions was analyzed and the corresponding standard curve was obtained. The sensibility is less than 25 µg/L, conforming to the standard of the MRL of Coumaphos stipulated by China. This method is label-free, using an unpurified single antibody only and can continuously test at least 80 groups of samples continuously. It has high sensitivity and specificity. The required equipments are simple, environmental friendly and easy to control. So this method is promised for a large number of samples quick detection on spot and for application prospects.  相似文献

目的:建立液-液萃取、GC/μECD气相色谱测定生物检材中佐匹克隆的方法。方法:以家兔为模型动物,500 mg佐匹克隆标准品灌胃家兔,6.5小时后处死,取其胃、肝、血、脑、心脏、尿,采用乙醚液-液萃取提取,Rtx-5毛细管柱分离,电子捕获检测器检测,标准曲线法定量。结果:该方法灵敏度达0.02μg/mL,在0.05-50μg/mL范围内线性关系良好;低、中、高3种浓度的加样回收率分别为91.7%、99.5%、94.4%,相对标准偏差(RSD)均小于5%;佐匹克隆在家兔脏器中的浓度分别为7.31μg/mL(血液)、7.63μg/g(肝脏)、7.96μg/g(脑)、14.67μg/g(心脏)、32.18μg/g(胃组织)、42.65μg/mL(尿液),在各脏器中呈特异性分布。结论:该法操作简单、灵敏度高,结果可靠,完全满足临床检测及公安机关在办理佐匹克隆麻醉案件中的需要。  相似文献

A new chemiluminescence (CL) method is described for the determination of synephrine. It is based on the reaction between synephrine and Ce(IV) in a nitric acid medium and measurement of the CL intensity produced by rhodamine B used as a luminophore, similar to luminol or lucigenin in basic media, instead of as a sensitizer. In the optimum conditions, the increase of CL intensity was correlated with synephrine concentration over the range 5.0 x 10(-9)-1.0 x 10(-6) g/mL with a detection limit of 1.0 x 10(-9) g/mL. The relative standard deviation (RSD) was 2.9% for 1.0 x 10(-7) g/mL synephrine (n = 11). The method was applied to the determination of a drug in herbal products, citrus fruit and biological samples, with satisfactory results. The results given by the proposed method are in good agreement with those given by HPLC-UV and UV spectrophotometry.  相似文献

为了初步研究蛇足石杉内生真菌g5发酵产物的乙酰胆碱酯酶(ACHE)抑制动力学以及对该株真菌做初步形态学鉴定,采用DTNB显色法检测g5发酵液醇提物抑制AChE活性的效果,并采用玻片培养法进行真菌鉴定.结果发现g5菌株属于半知菌目丛梗孢科青霉属;其发酵液醇提物对AChE的抑制作用表现为混合竞争型可逆抑制,其对游离酶的抑制常数KI>与对酶底物络合物的抑制常数KIS>分别为0.0789mL和1.1352mL,因此g5菌株代谢产物可作为开发AChE抑制剂类药物的潜在资源.  相似文献

Abstract— —A method to measure the rate of acetylcholine turnover in mouse brain in vivo has been developed. It is based on the formation of labelled acetylcholine from intravenously injected labelled choline. The isotopic dilution of choline in the brain has been measured by assaying endogenous choline in the brain by an enzymatic method using tritium-labelled acetyl-CoA and purified choline acetyltransferase.
The rate of acetylcholine turnover in the brain could be calculated at 50 n-moles acetylcholine/g/min in conscious mice. In anaesthetized mice and in mice treated with oxotremorine, a decrease of acetylcholine turnover to about 10 n-moles/g/min was found.  相似文献

A new flow injection chemiluminescent method has been developed for the determination of procaine hydrochloride, based on the inhibition of the chemiluminescence reaction of luminol–hydrogen peroxide by procaine hydrochloride. The influence of several surfactants and β‐cyclodextrin on the chemiluminescence intensity were studied. It was found that β‐cyclodextrin enhanced the decrease in chemiluminescence intensity. The method is simple, convenient and sensitive, with a detection limit (3 σ) of 0.08 µg/mL. The decreased chemiluminescence intensity is linear, with the concentration of procaine hydrochloride in the range 0.2–100.0 µg/mL and 100.0–400.0 µg/mL. The relative standard deviation for 10 repeated measurements were 4.5% and 3.4% for 1.0 and 20.0 µg/mL procaine hydrochloride, respectively. A sensitive and rapid method for detection and quantitative determination of pyrrolizidine alkaloids is presented. It is based on stoichiometric reaction of protonated alkaloids with methyl orange, followed by a release of the latter from the complex. The color intensity of the dye is assessed visually or spectrophotometrically. The method easily detects alkaloids at a concentration of 0.5 μg/ml. The sensitivity of the quantitative assay ranges from 0.006 to 0.045 μmol/ml (from about 2 to 15μg/ml). The method proved to be especially useful during extraction, purification, and separation of minor pyrrolizidine alkaloids.  相似文献

A method to quantitate collagen in solution is described. It is based on binding of the dye Sirius Supra red F3BA by collagen followed by elution and estimation of the bound dye in a spectrophotometer. Free iron content has been estimated in autotrophic and heterotrophic bacteria. It constituted 40-50 micrograms/g dry weight as compared to 15 micrograms/g dry weight in animal cells. A method for estimation of free iron has been proposed. It is based on formation of paramagnetic dinitrosyl iron complexes by free iron and protein thiol groups or low molecular weight thiol ligands. The reasons for high iron content in bacteria have been discussed.  相似文献

An improved iterative method for computing association constants from sedimentation equilibrium results obtained with self-interacting protein systems is presented which accounts for the composition-dependence of the activity coefficients of all oligomeric species. The method is based on the calculation of virial coefficients from covolume and charge considerations, the statistical mechanical basis of which is discussed in relation to the DLVO theory. The method is applied to results obtained with lysozyme in diethylbarbiturate buffer of pH 8.0 and ionic strength 0.15 at 15°C. It is shown that these results, encompassing a range of total solute concentration up to 19.7 g/liter are consistent with self-association patterns comprising either a monomer-dimer-trimer system or an isodesmic indefinite self-association of the monomer, the latter being favored. A firmer distinction between these possibilities is sought on the basis of the dependence of the weight-average partition coefficient, determined by frontal gel chromatography, on total solute concentration (up to 56.6 g/liter). This analysis accounts for the composition-dependence of the ratio of the activity coefficients of partitioning monomer in mobile and stationary phases. It is concluded that all results are consistent with an indefinite self-association of lysozyme governed by a single association constant of 4.61 × 102 liter/mole.  相似文献

An alternative method to determine the sn-2 monopalmitin in infant formulas was developed and validated. This method offers many advantages over the traditional methods. It follows the official method in the first steps, purification of the fat or oil through an alumina column, and subsequently the triglycerides are incubated with pancreatic lipase in order to obtain the sn-2 monoglycerides. In traditional methods the sn-2 monoglycerides are separated by preparative thin-layer chromatography and then, the 2-monoglycerides are converted into the corresponding fatty acid methyl esters and analysed by gas chromatography. In our method, separation, quantification and identification of the sn-2 monoglycerides were achieved by high-performance liquid chromatography with evaporative light-scattering detection. The detection limit (0.19 μg), quantification limit (0.38 μg), linearity range (r=0.999, range 1–200 μg) and precision (SD=1.10) show the suitability of the proposed method. This method is faster, cheaper and simple and does not consume large quantities of reagents and materials.  相似文献

A method for the simultaneous quantitation of 7 estrogens in pregnancy urine is described. It involves enzymatic hydrolysis, extraction of free steroids, ion-exchange column chromatography and quantitation of the trimethylsilyl derivatives by gas chromatography on OV 1. Data obtained from normal and twin pregnancies and from women with anencephalic fetus or intra uterine fetal death are analysed. The sensibility of the method is about 40 μg of each estrogen by liter of urine.  相似文献

A new flow injection chemiluminescence method is described for the determination of captopril. It is based on the enhancing effect of captopril on the chemiluminescence reaction of luminol with potassium ferricyanide in alkaline solution in the presence of potassium ferrocyanide. The method allows the determination of captopril over 0.1–40 µg/mL range, with a relative standard deviation (SD) of 1.0% for the determination of 0.5 µg/mL captopril solution in 11 repeated measurements. The method was satisfactorily applied to the determination of captopril in commercial captopril tablets. The possible reaction mechanism is also discussed briefly. 利用温和的SDS-PAGE法分离纯化了菠菜光系统Ⅰ的反应中心复合物(CP1),并进行了低温(77K)和室温下的电子自旋共振谱(ESR)测定,获得了自由基P700~+的ESR吸收信号.其一级微分谱的g值为2.0024,谱线近以于高斯型,与从菠菜叶绿体中发现的P700~+的ESR信号相符.根据这些结果,我们认为以温和SDS-PAGE法从菠菜得到的CP1组份保存了大部分P700的光氧化活性.  相似文献

We describe a quick and simple method for the quantitative detection of Listeria monocytogenes in meat products. This method is based on filtration, Chelex-100-based DNA purification, and real-time PCR. It can detect as few as 100 CFU/g and quantify as few as 1,000 CFU/g, with excellent accuracy compared to that of the plate count method. Therefore, it is a promising alternative for the detection of L. monocytogenes in meat products.  相似文献

Treatment of intact liver and liver homogenate with sodium nitrite, or desferal, brings about the appearance of g = 2.03 and g = 4.3 electron paramagnetic resonance spectroscopy (EPR) signals, respectively. The g = 2.03 signal is conditioned by the formation of dinitrosyl complexes of Fe(II); the g = 4.3 signal is related to the appearance of paramagnetic desferal-Fe(III) complexes. Desferal and sodium nitrite were administered successively into liver homogenate, resulting in only a g = 4.3 EPR signal. And, vice versa, if desferal was administered after sodium nitrite, there appeared only the signal with g = 2.03. These data testify to the fact that one and the same endogenous free iron is included in both paramagnetic centers. The concentration of iron ions was measured in intact tissue according to the formation of dinitrosyl-iron complexes and desferal-iron complexes. It was 33.2 +/- 4.6 and 20.3 +/- 4.0 nmol/g of tissue weight, respectively. The data obtained testify to the fact that free endogenous iron is present in intact tissue. Possibilities of the EPR method for estimation of the content of intracellular free iron are discussed.  相似文献

研究了用固相萃取技术结合氢火焰气相色谱法测定水和废水中四种氯苯类有机污染物。通过实验比较Sdex C18,Sep-Park Vac Silica,Bond Elut CARBON,Bond Elut SI和Bond Elut PLEXA五种SPE小柱对四种氯苯类的萃取效率,发现Dionex的Sdex C18柱有很好的回收率。系统研究了最佳萃取条件,甲醇洗脱体积为4.0 mL,且洗脱液不可通过氮吹浓缩;四种氯苯类化合物除氯苯外穿透体积都在1.0 L以上,而氯苯的穿透体积为300 mL,表明Sdex C18柱对二氯苯和三氯苯有很强的吸附性。在最佳萃取和测定条件下,方法线性范围为20.0~400μg/L,检出限为0.383~0.635μg/L,完全满足日常环境监测分析要求。  相似文献

The interest in the studies of the intestines using the method of tissue organ culture has considerably grown in recent years. It can be explained by the great possibilities of obtaining unique data about the state of intestines in normal and pathological condition, e.g. malabsorption syndrome. The paper deals with the method modified by the authors to obtain long-living (24 hours) intestinal tissue organ culture. The investigations used bioptic sections which were obtained by jejunoscopy with spot biopsy of children suffering from intestinal malabsorption. The viability of the explants was proved by histological and histochemical tests. The promise held by the methods is emphasized.  相似文献
