Among predicted impacts of climate change in the Arctic are greater thaw depth and shifts in vegetation patterns and hydrology that are likely to increase organic carbon and nutrient loading to lakes. We measured substrate limitation of sediment methane (CH4) flux, examined pathways of methanogenesis, and potential CH4 oxidation using stable isotope labeled acetate in intact sediment cores from arctic lake GTH 112 (68°40′20″N, 149°14′57″W). We hypothesized that the acetoclastic pathway would dominate methanogenesis, reflecting dissolved organic carbon supply from the surrounding landscape, and that sediment CH4 flux would be stimulated by addition of acetate. Experiments demonstrated acetate limitation of sediment CH4 flux with short-term CH4 flux response to availability of acetate, high rates of CH4 oxidation, and strong dominance of the acetoclastic over the hydrogenotrophic methanogenic pathway. The experiments also indicated that isotopic fractionation effects during isotope enrichment experiments are large during methanogenesis and can alter the methanogenic pathways being investigated. Under oxic conditions, CH4 oxidation at the sediment–water interface or in the water column is likely to account for much of diffusive CH4 flux, but under anoxic hypolimnetic conditions and increased substrate availability, conditions that are likely to occur with climate change, sediment CH4 flux will likely increase, with oxidation utilizing a smaller portion of sediment CH4 production.  相似文献

Organic matter decomposition regulates rates of carbon loss (CO2 and CH4) in wetlands and has implications for carbon sequestration in the context of changing global temperature. Here we determined the influence of temperature and vegetation type on both aerobic and anaerobic decomposition of organic matter in subtropical wetland soils. As in many other studies, increased temperature resulted in higher rates of respiration and methanogenesis under both aerobic and anaerobic conditions, and the positive effect of temperature depended on vegetation (source of carbon substrate to soil). Under anaerobic incubations, the proportion of gaseous C (CO2 and CH4) lost as CH4 increased with temperature indicating a greater sensitivity of methanogenesis to temperature. This was further supported by a wider range of Q10 values (1.4–3.6) for methane production as compared with anaerobic CO2 (1.3–2.5) or aerobic CO2 (1.4–2.1) production. The increasing strength of positive linear correlation between CO2:CH4 ratio and the soil organic matter ligno-cellulose index at higher temperature indicated that the temperature sensitivity of methanogenesis was likely the result of increased C availability at higher temperature. This information adds to our basic understanding of decomposition in warmer subtropical and tropical wetland systems and has implications for C models in wetlands with different vegetation types.  相似文献

The permafrost on the North Slope of Alaska is densely populated by shallow lakes that result from thermokarst erosion. These lakes release methane (CH4) derived from a combination of ancient thermogenic pools and contemporary biogenic production. Despite the potential importance of CH4 as a greenhouse gas, the contribution of biogenic CH4 production in arctic thermokarst lakes in Alaska is not currently well understood. To further advance our knowledge of CH4 dynamics in these lakes, we focused our study on (i) the potential for microbial CH4 production in lake sediments, (ii) the role of sediment geochemistry in controlling biogenic CH4 production, and (iii) the temperature dependence of this process. Sediment cores were collected from one site in Siqlukaq Lake and two sites in Sukok Lake in late October to early November. Analyses of pore water geochemistry, sedimentary organic matter and lipid biomarkers, stable carbon isotopes, results from CH4 production experiments, and copy number of a methanogenic pathway‐specific gene (mcrA) indicated the existence of different sources of CH4 in each of the lakes chosen for the study. Analysis of this integrated data set revealed that there is biological CH4 production in Siqlukaq at moderate levels, while the very low levels of CH4 detected in Sukok had a mixed origin, with little to no biological CH4 production. Furthermore, methanogenic archaea exhibited temperature‐dependent use of in situ substrates for methanogenesis, and the amount of CH4 produced was directly related to the amount of labile organic matter in the sediments. This study constitutes an important first step in better understanding the actual contribution of biogenic CH4 from thermokarst lakes on the coastal plain of Alaska to the current CH4 budgets.  相似文献

The turnover and exchange rates, as well as the sedimentation rates concerning the input and output of carbon in the anaerobic hypolimnion and the sediment in Lake Vechten were studied. The carbon input data were derived from those on sedimentation. By comparison of C-fixation data, sedimentation rates measurements and microbial reduction rates of observed fluxes of electron acceptors a mineralization of 20–30% of the limnetic C-fixation was found in the hypolimnion. The rate-limiting step of methanogenesis in anaerobic mineralization is the breakdown of algal cell wall components to their main product, namely acetate. This intermediate has a relatively fast turnover rate to the end-products, namely the gases CH4 and CO2. Finally, from the data on the diffusion of low-molecular weight metabolic intermediates and on the rates of formation of CH4 and CO2 the output of carbon and its cycling in Lake Vechten are discussed.  相似文献

Methanogenic degradation of organic matter is an important microbial process in lake sediments. Temperature may affect not only the rate but also the pathway of CH4 production by changing the activity and the abundance of individual microorganisms. Therefore, we studied the function and structure of a methanogenic community in anoxic sediment of Lake Dagow, a eutrophic lake in north-eastern Germany. Incubation of sediment samples (in situ 7.5°C) at increasing temperatures (4, 10, 15, 25, 30°C) resulted in increasing production rates of CH4 and CO2 and in increasing steady-state concentrations of H2. Thermodynamic conditions for H2/CO2 -dependent methanogenesis were only exergonic at 25 and 30°C. Inhibition of methanogenesis with chloroform resulted in the accumulation of methanogenic precursors, i.e., acetate, propionate, and isobutyrate. Mass balance calculations indicated that less CH4 was formed via H2 at 4°C than at 30°C. Conversion of 14CO2 to 14CH4 also showed that H2/CO2 -dependent methanogenesis contributed less to total CH4 production at 4°C than at 30°C. [2–14 C]Acetate turnover rates at 4°C accounted for a higher percentage of total CH4 production than at 30°C. Collectively, these results showed a higher contribution of H2-dependent methanogenesis and a lower contribution of acetate-dependent methanogenesis at high versus low temperature. The archaeal community was characterized by cloning, sequencing, and phylogenetic analysis of the 16S rRNA genes retrieved from the sediment. Sequences were affiliated with Methanosaetaceae, Methanomicrobiaceae, and three deeply branching euryarchaeotal clusters, i.e., group III, Rice cluster V, and a novel euryarchaeotal cluster, the LDS cluster. Terminal restriction fragment length polymorphism (T-RFLP) analysis showed that 16S rRNA genes affiliated to Methanosaetaceae (20–30%), Methanomicrobiaceae (35–55%), and group III (10–25%) contributed most to the archaeal community. Incubation of the sediment at different temperatures (4–30°C) did not result in a systematic change of the archaeal community composition, indicating that change of temperature primarily affected the activity rather than the structure of the methanogenic community.  相似文献

Methanogenic processes can be quantified by stable carbon isotopes, if necessary modeling parameters, especially fractionation factors, are known. Anoxically incubated rice roots are a model system with a dynamic microbial community and thus suitable to investigate principal geochemical processes in anoxic natural systems. Here we applied an inhibitor of acetoclastic methanogenesis (methyl fluoride), calculated the thermodynamics of the involved processes, and analyzed the carbon stable isotope signatures of CO2, CH4, propionate, acetate and the methyl carbon of acetate to characterize the carbon flow during anaerobic degradation of rice roots to the final products CO2 and CH4. Methyl fluoride inhibited acetoclastic methanogenesis and thus allowed to quantify the fractionation factor of CH4 production from H2/CO2. Since our model system was not affected by H2 gradients, the fractionation factor could alternatively be determined from the Gibbs free energies of hydrogenotrophic methanogenesis. The fractionation factor of acetoclastic methanogenesis was also experimentally determined. The data were used for successfully modeling the carbon flow. The model results were in agreement with the measured process data, but were sensitive to even small changes in the fractionation factor of hydrogenotrophic methanogenesis. Our study demonstrates that stable carbon isotope signatures are a proper tool to quantify carbon flow, if fractionation factors are determined precisely.  相似文献

土壤是地球表层最为重要的碳库也是温室气体的源或汇。自工业革命以来,对土壤温室气体的容量、收支平衡和通量等已有较多研究和估算,但对关键过程及其源/汇的研究却十分有限。微生物是土壤碳氮转化的主要驱动者, 在生态系统碳氮循环过程中扮演重要的角色,对全球气候变化有着响应的响应、适应及反馈,然而其个体数量,群落结构和多样性如何与气候扰动相互关联、进而怎样影响生态系统过程的问题仍有待进一步探索。从微生物介导的碳氮循环过程入手,重点讨论微生物对气候变化包括温室气体(CO2,CH4,N2O)增加、全球变暖、大气氮沉降等的响应和反馈,并由此提出削减温室气体排放的可能途径和今后发展的方向。  相似文献

Although cold environments are major contributors to global biogeochemical cycles, comparatively little is known about their microbial community function, structure, and limits of activity. In this study a microcosm based approach was used to investigate the effects of temperature, and methanogenic substrate amendment, (acetate, methanol and H2/CO2) on methanogen activity and methanogen community structure in high Arctic wetlands (Solvatnet and Stuphallet, Svalbard). Methane production was not detected in Stuphallet sediment microcosms (over a 150 day period) and occurred within Solvatnet sediments microcosms (within 24 hours) at temperatures from 5 to 40°C, the maximum temperature being at far higher than in situ maximum temperatures (which range from air temperatures of -1.4 to 14.1°C during summer months). Distinct responses were observed in the Solvatnet methanogen community under different short term incubation conditions. Specifically, different communities were selected at higher and lower temperatures. At lower temperatures (5°C) addition of exogenous substrates (acetate, methanol or H2/CO2) had no stimulatory effect on the rate of methanogenesis or on methanogen community structure. The community in these incubations was dominated by members of the Methanoregulaceae/WCHA2-08 family-level group, which were most similar to the psychrotolerant hydrogenotrophic methanogen Methanosphaerula palustris strain E1-9c. In contrast, at higher temperatures, substrate amendment enhanced methane production in H2/CO2 amended microcosms, and played a clear role in structuring methanogen communities. Specifically, at 30°C members of the Methanoregulaceae/WCHA2-08 predominated following incubation with H2/CO2, and Methanosarcinaceaeand Methanosaetaceae were enriched in response to acetate addition. These results may indicate that in transiently cold environments, methanogen communities can rapidly respond to moderate short term increases in temperature, but not necessarily to the seasonal release of previously frozen organic carbon from thawing permafrost soils. However, as temperatures increase such inputs of carbon will likely have a greater influence on methane production and methanogen community structure. Understanding the action and limitations of anaerobic microorganisms within cold environments may provide information which can be used in defining region-specific differences in the microbial processes; which ultimately control methane flux to the atmosphere.  相似文献

Thermokarst lakes are potentially important sources of methane (CH4) and carbon dioxide (CO2). However, considerable uncertainty exists regarding carbon emissions from thermokarst lakes owing to a limited understanding of their patterns and motivators. In this study, we measured CH4 and CO2 diffusive fluxes in 163 thermokarst lakes in the Qinghai–Tibet Plateau (QTP) over 3 years from May to October. The median carbon emissions from the QTP thermokarst lakes were 1440 mg CO2 m−2 day−1 and 60 mg CH4 m−2 day−1, respectively. The diffusive rates of CO2 and CH4 are related to the catchment land cover type. Sediment microbial abundance and hydrochemistry explain 51.9% and 38.3% of the total variance in CH4 diffusive emissions, respectively, while CO2 emissions show no significant relationship with environmental factors. When upscaling carbon emissions from the QTP thermokarst lakes, the annual average CH4 release per lake area is equal to that of the pan-Arctic region. Our findings highlight the importance of incorporating in situ observation data with different emission pathways for different land cover types in predicting carbon emissions from thermokarst lakes in the future.  相似文献

Boreal peatlands contain approximately 500 Pg carbon (C) in the soil, emit globally significant quantities of methane (CH4), and are highly sensitive to climate change. Warming associated with global climate change is likely to increase the rate of the temperature‐sensitive processes that decompose stored organic carbon and release carbon dioxide (CO2) and CH4. Variation in the temperature sensitivity of CO2 and CH4 production and increased peat aerobicity due to enhanced growing‐season evapotranspiration may alter the nature of peatland trace gas emission. As CH4 is a powerful greenhouse gas with 34 times the warming potential of CO2, it is critical to understand how factors associated with global change will influence surface CO2 and CH4 fluxes. Here, we leverage the Spruce and Peatland Responses Under Changing Environments (SPRUCE) climate change manipulation experiment to understand the impact of a 0–9°C gradient in deep belowground warming (“Deep Peat Heat”, DPH) on peat surface CO2 and CH4 fluxes. We find that DPH treatments increased both CO2 and CH4 emission. Methane production was more sensitive to warming than CO2 production, decreasing the C‐CO2:C‐CH4 of the respired carbon. Methane production is dominated by hydrogenotrophic methanogenesis but deep peat warming increased the δ13C of CH4 suggesting an increasing contribution of acetoclastic methanogenesis to total CH4 production with warming. Although the total quantity of C emitted from the SPRUCE Bog as CH4 is <2%, CH4 represents >50% of seasonal C emissions in the highest‐warming treatments when adjusted for CO2 equivalents on a 100‐year timescale. These results suggest that warming in boreal regions may increase CH4 emissions from peatlands and result in a positive feedback to ongoing warming.  相似文献

Lake methane (CH4) emissions are largely controlled by aerobic methane-oxidizing bacteria (MOB) which mostly belong to the classes Alpha- and Gammaproteobacteria (Alpha- and Gamma-MOB). Despite the known metabolic and ecological differences between the two MOB groups, their main environmental drivers and their relative contribution to CH4 oxidation rates across lakes remain unknown. Here, we quantified the two MOB groups through CARD-FISH along the water column of six temperate lakes and during incubations in which we measured ambient CH4 oxidation rates. We found a clear niche separation of Alpha- and Gamma-MOB across lake water columns, which is mostly driven by oxygen concentration. Gamma-MOB appears to dominate methanotrophy throughout the water column, but Alpha-MOB may also be an important player particularly in well-oxygenated bottom waters. The inclusion of Gamma-MOB cell abundance improved environmental models of CH4 oxidation rate, explaining part of the variation that could not be explained by environmental factors alone. Altogether, our results show that MOB composition is linked to CH4 oxidation rates in lakes and that information on the MOB community can help predict CH4 oxidation rates and thus emissions from lakes.  相似文献

The effects of temperature on rates and pathways of CH4 production and on the abundance and structure of the archaeal community were investigated in acidic peat from a mire in northern Scandinavia (68°N). We monitored the production of CH4 and CO2 over time and measured the turnover of Fe(II), ethanol, and organic acids. All experiments were performed with and without specific inhibitors (2-bromoethanesulfonate [BES] for methanogenesis and CH3F for acetoclastic methanogenesis). The optimum temperature for methanogenesis was 25°C (2.3 μmol CH4 · g [dry weight]−1 · day−1), but the activity was relatively high even at 4°C (0.25 μmol CH4 · g [dry weight]−1 · day−1). The theoretical lower limit for methanogenesis was calculated to be at −5°C. The optimum temperature for growth as revealed by real-time PCR was 25°C for both archaea and bacteria. The population structure of archaea was studied by terminal restriction fragment length polymorphism analysis and remained constant over a wide temperature range. Hydrogenotrophic methanogenesis accounted for about 80% of the total methanogenesis. Most 16S rRNA gene sequences that were affiliated with methanogens and all McrA sequences clustered with the exclusively hydrogenotrophic order Methanobacteriales, correlating with the prevalence of hydrogenotrophic methanogenesis. Fe reduction occurred parallel to methanogenesis and was inhibited by BES, suggesting that methanogens were involved in Fe reduction. Based upon the observed balance of substrates and thermodynamic calculations, we concluded that the ethanol pool was oxidized to acetate by the following two processes: syntrophic oxidation with methanogenesis (i) as an H2 sink and (ii) as a reductant for Fe(III). Acetate accumulated, but a considerable fraction was converted to butyrate, making volatile fatty acids important end products of anaerobic metabolism.  相似文献

We investigated the effects of oxygen (O2) concentration on methane (CH4) production and oxidation in two humid tropical forests that differ in long‐term, time‐averaged soil O2 concentrations. We identified sources and sinks of CH4 through the analysis of soil gas concentrations, surface emissions, and carbon isotope measurements. Isotope mass balance models were used to calculate the fraction of CH4 oxidized in situ. Complementary laboratory experiments were conducted to determine the effects of O2 concentration on gross and net rates of methanogenesis. Field and laboratory experiments indicated that high levels of CH4 production occurred in soils that contained between 9±1.1% and 19±0.2% O2. For example, we observed CH4 concentrations in excess of 3% in soils with 9±1.1% O2. CH4 emissions from the lower O2 sites were high (22–101 nmol CH4 m?2 s?1), and were equal in magnitude to CH4 emissions from natural wetlands. During peak periods of CH4 efflux, carbon dioxide (CO2) emissions became enriched in 13C because of high methanogenic activity. Gross CH4 production was probably greater than flux measurements indicated, as isotope mass balance calculations suggested that 48–78% of the CH4 produced was oxidized prior to atmospheric egress. O2 availability influenced CH4 oxidation more strongly than methanogenesis. Gross CH4 production was relatively insensitive to O2 concentrations in laboratory experiments. In contrast, methanotrophic bacteria oxidized a greater fraction of total CH4 production with increasing O2 concentration, shifting the δ13C composition of CH4 to values that were more positive. Isotopic measurements suggested that CO2 was an important source of carbon for methanogenesis in humid forests. The δ13C value of methanogenesis was between ?84‰ and ?98‰, which is well within the range of CH4 produced from CO2 reduction, and considerably more depleted in 13C than CH4 formed from acetate.  相似文献

The intensity of the microbiological processes of methane formation (MF) and methane oxidation (MO) was determined in the sediments and water of different types of Baltic lakes. The emission of methane from the lake sediments and methane distribution in the water column of the lakes were studied as functions of the lake productivity and hydrologic conditions. During summers, the intensity of MF in the lake sediments and waters varied from 0.001 to 106 ml CH4/(dm3 day) and from 0 to 3.2 ml CH4/(l day), respectively, and the intensity of MO in the sediments and water varied from 0 to 11.2 ml CH4/(dm3 day) and from 0 to 1.1 ml CH4/(l day), respectively. The total methane production (MP) in the lakes varied from 15 to 5000 ml CH4/(m2 day). In anoxic waters, the MP comprised 9–18% of the total PM in the lakes. The consumption of organic carbon for methanogenesis varied from 0.03 to 9.7 g/(m2 day). The role of the methane cycle in the degradation of organic matter in the lakes increased with their productivity.  相似文献

Continued current emissions of carbon dioxide (CO2) and methane (CH4) by human activities will increase global atmospheric CO2 and CH4 concentrations and surface temperature significantly. Fields of paddy rice, the most important form of anthropogenic wetlands, account for about 9% of anthropogenic sources of CH4. Elevated atmospheric CO2 may enhance CH4 production in rice paddies, potentially reinforcing the increase in atmospheric CH4. However, what is not known is whether and how elevated CO2 influences CH4 consumption under anoxic soil conditions in rice paddies, as the net emission of CH4 is a balance of methanogenesis and methanotrophy. In this study, we used a long-term free-air CO2 enrichment experiment to examine the impact of elevated CO2 on the transformation of CH4 in a paddy rice agroecosystem. We demonstrate that elevated CO2 substantially increased anaerobic oxidation of methane (AOM) coupled to manganese and/or iron oxides reduction in the calcareous paddy soil. We further show that elevated CO2 may stimulate the growth and metabolism of Candidatus Methanoperedens nitroreducens, which is actively involved in catalyzing AOM when coupled to metal reduction, mainly through enhancing the availability of soil CH4. These findings suggest that a thorough evaluation of climate-carbon cycle feedbacks may need to consider the coupling of methane and metal cycles in natural and agricultural wetlands under future climate change scenarios.  相似文献

A bacterial consortium capable of sucrose degradation primarily to CH4 and CO2 was constructed, with acetate as the key methanogenic precursor. In addition, the effect of agar immobilization on the activity of the consortium was determined. The primary fermentative organism, Escherichia coli, produced acetate, formate, H2, and CO2 (known substrates for methanogens), as well as ethanol and lactate, compounds that are not substrates for methanogens. Oxidation of the nonmethanogenic substrates, lactate and ethanol, to acetate was mediated by the addition of Acetobacterium woodii and Desulfovibrio vulgaris. The methanogenic stage was accomplished by the addition of the acetophilic methanogen Methanosarcina barkeri and the hydrogenophilic methanogen Methanobacterium formicicum. Results of studies with low substrate concentrations (0.05 to 0.2% [wt/vol]), a growth-limiting medium, and the five-component consortium indicated efficient conversion (40%) of sucrose carbon to CH4. Significant decreases in yields of CH4 and rates of CH4 production were observed if any component of the consortium was omitted. Approximately 70% of the CH4 generated occurred via acetate. Agar-immobilized cells of the consortium exhibited yields of CH4 and rates of CH4 production from sucrose similar to those of nonimmobilized cells. The rate of CH4 production decreased by 25% when cysteine was omitted from reaction conditions and by 40% when the immobilized consortium was stored for 1 week at 4°C.  相似文献

Invasion by the exotic species Spartina alterniflora, which has high net primary productivity and superior reproductive capacity compared with native plants, has led to rapid organic carbon accumulation and increased methane (CH4) emission in the coastal salt marsh of China. To elucidate the mechanisms underlying this effect, the methanogen community structure and CH4 production potential as well as soil organic carbon (SOC), dissolved organic carbon, dissolved organic acids, methylated amines, aboveground biomass, and litter mass were measured during the invasion chronosequence (0–16 years). The CH4 production potential in the S. alterniflora marsh (range, 2.94–3.95 μg kg?1 day?1) was significantly higher than that in the bare tidal mudflat. CH4 production potential correlated significantly with SOC, acetate, and trimethylamine concentrations in the 0–20 cm soil layer. The abundance of methanogenic archaea also correlated significantly with SOC, and the dominant species clearly varied with S. alterniflora-driven SOC accumulation. The acetotrophic Methanosaetaceae family members comprised a substantial proportion of the methanogenic archaea in the bare tidal mudflat while Methanosarcinaceae family members utilized methylated amines as substrates in the S. alterniflora marsh. Ordination analysis indicated that trimethylamine concentration was the primary factor inducing the shift in the methanogenic archaea composition, and regressive analysis indicated that the facultative family Methanosarcinaceae increased linearly with trimethylamine concentration in the increasingly sulfate-rich salt marsh. Our results indicate that increased CH4 production during the S. alterniflora invasion chronosequence was due to increased levels of the non-competitive substrate trimethylamine and a shift in the methanogenic archaea community.  相似文献

Within aquatic ecosystems, periphytic biofilms can be hot spots of denitrification, and previous work has suggested that algal taxa within periphyton can influence the species composition and activity of resident denitrifying bacteria. This study tested the hypothesis that algal species composition within biofilms influences the structure and function of associated denitrifying bacterial communities through the composition of organic exudates. A mixed population of bacteria was incubated with organic carbon isolated from one of seven algal species or from one of two streams that differed in anthropogenic inputs. Pyrolysis-gas chromatography-mass spectrometry (Py-GC/MS) revealed differences in the organic composition of algal exudates and stream waters, which, in turn, selected for distinct bacterial communities. Organic carbon source had a significant effect on potential denitrification rates (DNP) of the communities, with organics isolated from a stream with high anthropogenic inputs resulting in a bacterial community with the highest DNP. There was no correlation between DNP and numbers of denitrifiers (based on nirS copy numbers), but there was a strong relationship between the species composition of denitrifier communities (as indicated by tag pyrosequencing of nosZ genes) and DNP. Specifically, the relative abundance of Pseudomonas stutzeri-like nosZ sequences across treatments correlated significantly with DNP, and bacterial communities incubated with organic carbon from the stream with high anthropogenic inputs had the highest relative abundance of P. stutzeri-like nosZ sequences. These results demonstrate a significant relationship between bacterial community composition and function and provide evidence of the potential impacts of anthropogenic inputs on the structure and function of stream microbial communities.  相似文献
