This is the first mitochondrial phylogeography of the common dormouse, Muscardinus avellanarius (Linnaeus, 1758), a hibernating rodent strictly protected in Europe (Habitat Directive, annex IV; Bern Convention, annex III). The 84 individuals of M. avellanarius, sampled throughout the distributional range of the species, have been sequenced at the mitochondrial DNA gene (cytochrome b, 704 base pairs). The results revealed two highly divergent lineages, with an ancient separation around 7.7 Mya and a genetic divergence of 7.7%. Lineage 1 occurs in Western Europe (France, Belgium, and Switzerland) and Italy, and lineage 2 occurs in Central–Northern Europe (Poland, Germany, Latvia, and Lithuania), on the Balkan Peninsula, and in Turkey. Furthermore, these two lineages are subdivided into five sublineages genetically isolated with a strong geographical association. Therefore, lineage 1 branches into two further sublineages (Western European and Italian), whereas lineage 2 contained three sublineages (Central–Northern European, Turkish, and Balkan). We observed low genetic diversity within the sublineages, in contrast to the significant level of genetic differentiation between them. The understanding of genetic population structure is essential for identifying units to be conserved. The present article extends our previous work on the phylogenetic history of Microtus (Terricola) thomasi, analysing cytochrome b, 12S rRNA, 16S rRNA and the control region in 65 Greek populations. The analysis revealed three clades: one grouping the populations from Peloponnisos (Southern Greece); the second, the populations from Agios (Ag.) Stefanos and Evvoia island (Central East Greece); and the third, all the remaining populations with no geographical substructure. Genetic distances were low for most populations, with only the populations of Evvoia and Ag. Stefanos being substantially distant. Thus, although this species has a recent colonization history and probably descends from a highly polymorphic ancestor, a monophyletic and highly differentiated lineage is formed in Greece and is distributed in Ag. Stefanos and Evvoia. Molecular differentiation, distinct geographical distribution and restriction of gene flow between this lineage and the rest of the Greek populations provide evidence for its probable subspecific status, Microtus (Tericola) thomasi atticus. A possible mechanism leading the differentiation process of the proposed subspecies is suggested, based on the displacement of this species in central Greece by its congeneric, probably better‐fitted Microtus (Microtus) guentheri and the subsequent separation of Ag. Summary Mustela nivalis and M. erminea, two sympatric species of weasels of superficially similar appearance and habits, have different breeding and foraging strategies associated with the difference in their body size. M. nivalis is more efficient in exploiting small rodent prey, and can breed rapidly to take immediate advantage of rodent peaks, but is vulnerable to local extinction during rodent declines. M. erminea has more generalized food habits, and is the larger and probably the dominant species, but is limited by delayed implantation to producing only one litter a year. M. nivalis is therefore superior in exploitation competition, and erminea in interference competition. We offer the hypothesis that the co-existence of the two species is permitted by a balance of these competitive advantages determined, at a given time or place, by the heterogeneity of the environment and the distribution of the prey fauna. We use this hypothesis to explain cases where co-existence has either broken down or is not recorded (the results of simultaneous introductions to New Zealand and Terschelling Island, and of myxomatosis in Britain, and the distribution of nivalis and Erminea on the offshore islands of Britain). We argue that the diversity and size distribution of the prey fauna of an island (which are both related to its area and isolation) are important in deciding the species and size of mustelids surviving there; for example, we suggest that nivalis was present in Ireland in immediate post-glacial times but became extinct with the lemmings.  相似文献

In 2011, Vachon and Freeland presented the results of a study that illustrated the importance of considering mutation patterns, including the potential for homoplasy, when deciding whether to include or exclude repetitive sequences from phylogeographic inferences. Saltonstall and Lambertini (2012) criticized this study by suggesting that some of the analyses and interpretations were flawed. In this reply, we explain why we disagree with most of their criticisms and identify some inconsistencies in their analyses. Most importantly, we reiterate the need to examine underlying assumptions, particularly with respect to mutation patterns, when using molecular genetic data to untangle evolutionary relationships.  相似文献

Two sibling species of the rodent genus Praomys occur in West African forests: P. tullbergi and P. rostratus. By sampling across their geographical ranges (459 individuals from 77 localities), we test the hypothesis that climatic oscillations during the Quaternary made an impact on the observed pattern of cytochrome b sequence variation. We show that, although these two species have parapatric geographical distributions, their phylogeographical histories are dissimilar, which could be related to their distinct ecological requirements. Since the arid phases of the Pleistocene were characterized by isolated forest patches, and intervening wetter periods by forest expansion, these changes in forest cover may be the common mechanism responsible for the observed phylogeographical patterns in both of these species. For example, in both species, most clades had either allopatric or parapatric geographical distributions; however, genetic diversity was much lower in P. tullbergi than in P. rostratus. The genetic pattern of P. tullbergi fits the refuge hypothesis, indicating that a very small number of populations survived in distinct forest blocks during the arid phases, then expanded again with forest recovery. In contrast, a number of populations of P. rostratus appear to have survived during the dry periods in more fragmented forest habitats, with varying levels of gene flow between these patches depending on climatic conditions and forest extent. In addition, historical variations of the West African hydrographic network could also have contributed to the pattern of genetic differentiation observed in both species.  相似文献

We studied the intraspecific genetic variation of 17 Neomys anomalus (Cabrera, 1907) (eight localities) and 24 Neomys fodiens (Pennant, 1771) (nine localities) in Europe. As molecular marker, we used the sequences of the mtDNA gene for cytochrome b (1140 bp from 24 specimens; 272 bp from 15 specimens). The phylogenetic analysis of N. fodiens shows slight divergence among haplotypes, with only one supported clade including two geographically distant haplotypes from Calabria (southern Italy) and Pyrenees (France); these haplotypes diverge from the others by 2.9% and 2.1%, respectively. In N. anomalus , one haplotype from Spain is very different, diverging from all others by 8–10% of the sequence. This level of divergence is similar to that found between N. anomalus and another species, Neomys teres . There are two other supported clades in N. anomalus , an Italian subclade and an eastern subclade comprising haplotypes from Turkey and Slovenia. Moreover, we found a different genetic structure between the two species with greater divergence in N. anomalus than in N. fodiens. The phylogeographic patterns are discussed in a temporal framework and considering the different ecological characteristics of these two species.  相似文献

Hybrid zones are natural laboratories for investigating the dynamics of gene flow, reproductive isolation, and speciation. A predominant marine hybrid (or suture) zone encompasses Christmas Island (CHR) and Cocos (Keeling) Islands (CKE), where 15 different instances of interbreeding between closely related species from Indian and Pacific Oceans have been documented. Here, we report a case of hybridization between genetically differentiated Pacific and Indian Ocean lineages of the three‐spot dascyllus, Dascyllus trimaculatus (Rüppell, 1829). Field observations indicate there are subtle color differences between Pacific and Indian Ocean lineages. Most importantly, population densities of color morphs and genetic analyses (mitochondrial DNA and SNPs obtained via RADSeq) suggest that the pattern of hybridization within the suture zone is not homogeneous. At CHR, both color morphs were present, mitochondrial haplotypes of both lineages were observed, and SNP analyses revealed both pure and hybrid genotypes. Meanwhile, in CKE, the Indian Ocean color morphs were prevalent, only Indian Ocean mitochondrial haplotypes were observed, and SNP analysis showed hybrid individuals with a large proportion (~80%) of their genotypes assigning to the Indian Ocean lineage. We conclude that CHR populations are currently receiving an influx of individuals from both ocean basins, with a greater influence from the Pacific Ocean. In contrast, geographically isolated CKE populations appear to be self‐recruiting and with more influx of individuals from the Indian Ocean. Our research highlights how patterns of hybridization can be different at scales of hundreds of kilometers, due to geographic isolation and the history of interbreeding between lineages.  相似文献

Mitochondrial genetic variations were used to investigate the relationships between two Japanese wild boars, Japanese wild boar (Sus scrofa leucomystax) and Ryukyu wild boar (S.s. riukiuanus). Nucleotide sequences of the control (27 haplotypes) and cytochrome b (cyt-b) regions (19 haplotypes) were determined from 59 Japanese wild boars, 13 Ryukyu wild boars and 22 other boars and pigs. From phylogenetic analyses, the mtDNA of Ryukyu wild boar has a distinct lineage from that of Japanese wild boar, which was classified into the Asian pig lineage. This result suggests that the Ryukyu wild boar has a separate origin from the Japanese wild boar.  相似文献

Although mitochondrial DNA markers have several properties that make them suitable for phylogeographic studies, they are not free of difficulties. Phylogeographic inferences within and between closely related species can be mislead by introgression and retention of ancestral polymorphism. Here we combine different phylogenetic, phylogeographic, and population genetic methods to extract the maximum information from the Liolaemus darwinii complex. We estimate the phylogeographic structure of L. darwinii across most of its distributional range, and we then estimate relationships between L. darwinii and the syntopic species L. laurenti and L. grosseorum. Our results suggest that range expansion of these lineages brought them into secondary contact in areas where they are presently in syntopy. Here we present the first evidence for introgression in lizards from temperate South America (of L. danwinii mitochondrial DNA into L. laurenti and L. grosseorum), and for incomplete lineage sorting (between L. darwinii and L. laurenti). We show that a combination of methods can provide additional support for inferences derived from any single method and thus provide more robust interpretations and narrow the range of plausible hypotheses about mechanisms and processes of divergence. Additional studies are needed in this group of lizards and in other codistributed groups to determine if Pleistocene climatic changes could be a general factor influencing the evolutionary history of a regional biota.  相似文献

1. The light : nutrient hypothesis (LNH) proposes that herbivore growth rates are maximised at intermediate light‐to‐nutrient ratios. A reduction to light intensity (i.e. decreased light‐to‐nutrient ratio) should lead to reduced food availability for herbivores while excessive light intensity in oligotrophic environments (i.e. increased light‐to‐nutrient ratios) should increase the C : N and C : P ratios of producers. However, this hypothesis has not yet been supported by studies on stream ecosystems. 2. We tested the LNH by experimental application of controlled natural gradients in light intensity to oligotrophic laboratory channels that included periphyton and the freshwater snail Gyraulus chinensis. 3. The results in this oligotrophic environment indicate that light regulated the flow of matter between trophic levels and grazer reproduction by controlling C : P ratios of the producers.  相似文献

The large Myotis complex in continental Europe, Asia Minor, and Transcaucasia comprises two sibling bat species, the greater mouse‐eared bat, Myotis myotis, and the lesser mouse‐eared bat, Myotis blythii, also referred to as Myotis oxygnathus. Here, we investigate the phylogeography of these bats using two mitochondrial markers: the second hypervariable domain of the control region (HVII) and a fragment of the cytochrome b gene (cyt b). The HVII haplotypes formed six distinct haplogroups associated with different geographical regions. Most of the European HVII haplotypes were exclusive to M. myotis, whereas the majority of HVII haplotypes found in Asia Minor were exclusive to M. blythii/M. oxygnathus. The phylogenetic reconstruction based on the concatenated cyt b and HVII fragments recovered two major lineages. The first lineage comprised samples from Europe (western lineage), and the second lineage included samples from Asia Minor, Transcaucasia, Crimea, Western Ukraine, Thrace, the Balkans, and Eastern Europe (eastern lineage). The mitochondrial lineage of M. blythii, reported from Kyrgyzstan, was not present in Asia Minor and Transcaucasia. Therefore, we consider the possibility that the M. blythii/M. oxygnathus found in Europe, Asia Minor, and Transcaucasia are not recent descendants of the Central Asian M. blythii. Instead, we suggest that M. blythii/M. oxygnathus and M. myotis diverged through allopatric speciation in Asia Minor and Europe, and that they are represented by the eastern and western mitochondrial lineages. Repeated Quaternary glaciations have significantly shaped the present distribution and diversity of several European species in aquatic and terrestrial habitats. To study the phylogeography of freshwater invertebrates, patterns of intraspecific variation have been examined primarily using mitochondrial DNA markers that may yield results unrepresentative of the true species history. Here, population genetic parameters were inferred for a montane aquatic caddisfly, Thremma gallicum, by sequencing a 658‐bp fragment of the mitochondrial CO1 gene, and 12,514 nuclear RAD loci. T. gallicum has a highly disjunct distribution in southern and central Europe, with known populations in the Cantabrian Mountains, Pyrenees, Massif Central, and Black Forest. Both datasets represented rangewide sampling of T. gallicum. For the CO1 dataset, this included 352 specimens from 26 populations, and for the RAD dataset, 17 specimens from eight populations. We tested 20 competing phylogeographic scenarios using approximate Bayesian computation (ABC) and estimated genetic diversity patterns. Support for phylogeographic scenarios and diversity estimates differed between datasets with the RAD data favouring a southern origin of extant populations and indicating the Cantabrian Mountains and Massif Central populations to represent highly diverse populations as compared with the Pyrenees and Black Forest populations. The CO1 data supported a vicariance scenario (north–south) and yielded inconsistent diversity estimates. Permutation tests suggest that a few hundred polymorphic RAD SNPs are necessary for reliable parameter estimates. Our results highlight the potential of RAD and ABC‐based hypothesis testing to complement phylogeographic studies on non‐model species.  相似文献

Phylogenetic analysis identified haplochromine cichlids from isolated water bodies in the eastern branch of the East African Rift system as an ancient lineage separated from their western sister group in the course of the South Kenyan-North Tanzanian rift system formation. Within this lineage, the close phylogenetic relatedness among taxa indicates a recent common ancestry and historical connections between now separated water bodies. In connection with a total lack of local genetic diversity attributable to population bottlenecks, the data suggest cycles of extinction and colonization in the unstable habitat provided by the small lakes and rivers in this geologically highly active area.  相似文献

Using genetic data to study the process of population divergence is central to understanding speciation, yet distinguishing between recent divergence and introgressive hybridization is challenging. In a previous study on the phylogeography of the yellow‐rumped warbler complex using mitochondrial (mt)DNA data, we reported limited sequence divergence and a lack of reciprocal monophyly between myrtle and Audubon's warblers (Dendroica coronata and Dendroica auduboni, respectively), suggesting very recent isolation. In the present study, we report the results obtained from a subsequent sampling of Audubon's warbler in Arizona and Utah (‘memorabilis’ race), which shows that, although this taxon is similar to auduboni in plumage colour, most memorabilis individuals sampled (93%) carry haplotypes that belong to the divergent black‐fronted warbler lineage (Dendroica nigrifrons) of Mexico. Furthermore, the auduboni and nigrifrons lineages mix in southern Utah at a narrow, yet apparently ‘cryptic’ contact zone. Newly‐available evidence from nuclear markers indicating marked differentiation between auduboni and coronata has focused attention on the possibility of mtDNA introgression in the absence of nuclear gene flow, and the results of the present study are consistent with the hypothesis that the mtDNA of auduboni was indeed historically introgressed from the coronata lineage. Analysis of morphological traits shows that memorabilis is significantly differentiated from auduboni and nigrifrons in some traits, yet is overall intermediate between the two, which is consistent with a shared common ancestor for the auduboni/memorabilis/nigrifrons group. The striking, unexpected mtDNA pattern reported in the present study reveals a complex evolutionary history of the yellow‐rumped warbler complex, and cautions against the exclusive use of mtDNA to infer evolutionary relationships. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 696–706.  相似文献
