Injury to lens epithelial cells (LECs) leads to epithelial–mesenchymal transition (EMT) with resultant fibrosis. The tropomyosin (Tpm) family of cytoskeleton proteins is involved in regulating and stabilizing actin microfilaments. Aberrant expression of Tpms leads to abnormal morphological changes with disintegration of epithelial integrity. The EMT of LECs has been proposed as a major cause of posterior capsule opacification (PCO) after cataract surgery. Using in vivo rodent PCO and human cataractous LECs, we demonstrated that the aberrant expression of rat Tpm and human Tpm1α/2β suggested their association in remodelling of the actin cytoskeleton during EMT of LECs. Expression analysis from abnormally growing LECs after lens extraction revealed elevated expression of α‐smooth muscle actin (α‐SMA), a marker for EMT. Importantly, these cells displayed increased expression of Tpm1α/2β following EMT/PCO formation. Expression of Tpm1α/2β was up‐regulated in LECs isolated from cataractous lenses of Shumiya Cataract Rats (SCRs), compared with non‐cataractous lenses. Also, LECs from human patients with nuclear cataract and anterior subcapsular fibrosis (ASF) displayed significantly increased expression of Tpm2β mRNA, suggesting that similar signalling invokes the expression of these molecules in LECs of cataractous SCR and human lenses. EMT was observed in LECs overexpressed with Tpm1α/2β, as evidenced by increased expression of α‐SMA. These conditions were correlated with remodelling of actin filaments, possibly leading to EMT/PCO and ASF. The present findings may help clarify the condition of the actin cytoskeleton during morphogenetic EMT, and may contribute to development of Tpm‐based inhibitors for postponing PCO and cataractogenesis.  相似文献

Protamine–DNA complexes prepared by the method of direct and slow mixing in 2.5 × 10?4M EDTA, pH 8.0, have been studied by thermal denaturation and circular dichroism. The complexes show biphasic melting with Tm at about 50 °C corresponding to the melting of free DNA regions and Tm′ at about 92 °C corresponding to the melting of protamine-bound regions. In protamine-bound regions there are 1.38 amino acid residues per nucleotide, indicating a nearly completely charge neutralization. Tm is increased but Tm′ is not when the ionic strength of the buffer is raised. This also supports a full charge neutralization in protamine-bound regions. The circular dichroism of the complexes can be decomposed into two components, Δε0 of free DNA regions in B-form conformation and Δεb of protamine-bound regions in a characteristic conformation neither that of B- nor C-form but somewhere between them.  相似文献

Aim To evaluate the relative importance of climate, productivity, environmental heterogeneity, biotic associations and habitat use by cattle to account for the species richness of trees, shrubs and herbs across the Subantarctic–Patagonian transition. Location An area of c. 150 × 150 km, within the transition zone between the Subantarctic and Patagonian subregions on the eastern slope of the Andes (c. 39–42° S, 70–72° W). Methods All vascular plants found at each one of 50 (10 × 10 m) sampling plots were counted to estimate the local tree, shrub and herb species richness. Path analysis was used to evaluate the relationship between the richness of the three life‐forms and plant cover, dried litter biomass, mean annual temperature, annual precipitation, daily temperature range, substrate heterogeneity and number of faecal pats. Principal coordinates of neighbour matrices was used to model the spatial autocorrelation of the data. Results Total plant species richness showed a unimodal pattern of spatial variation across the transition. Richness responded positively to indirect effects of precipitation mediated through plant cover, but there was a negative overall effect of precipitation on richness towards the west of the transition, most strongly for trees. An increase in substrate heterogeneity promoted a local increase in herb and shrub richness; the richness of trees increased in sites with steeper slopes. Canopy closure had a direct negative impact on herb richness; it also increased the local accumulation of litter, which negatively affected shrub and herb richness. The impact of habitat use by cattle negatively affected herb richness in areas to the east of the biogeographical transition. Main conclusions We suggest that the importance of indirect climatic effects mediated by vegetation cover can account for species richness patterns across this transition, most strongly for woody species, which supports the productivity hypothesis. The southern temperate forests towards the west may represent a deviation from the predictions of the water–energy dynamics hypothesis. Dissimilar spatial patterns of variation in the richness of woody and herbaceous species, and their different responses to climatic and heterogeneity variables across the transition, suggest that plant life‐form influences the plant species richness–environment relationships.  相似文献

A general theory of helix–coil transition of irreversibly complexed nucleoproteins is presented. The equations are tested by experimental results in basic polypeptide–DNA complexes, nucleohistone I and pea bud nucleohistones. They show good agreement between theory and experiments. The theory provides direct measurement of a fraction of DNA base pairs covered by proteins, yielding a value of about 75% histone-covered base pairs in pea bud nucleohistone. It also provides a measurement of an average number of amino acid residues per nucleotide in protein-bound regions. This number varies from 1.0 to 1.4 in DNA–polylysine or DNA–polyarginine and from 2.9 to 3.3 in nucleohistone Ia, Ib, f1, and pea bud nucleohistone.  相似文献

A structural transition is reported to occur in aqueous sols of agarose, an electrically uncharged biostructural polysaccharide. The transition has no measurable effect on size dispersity on the shape of the solute polysaccharide as observed by precision photon correlation spectroscopy. It originates a low-angle pattern of scattered light similar to that which monitors phase separations in polymer blends. Thus, it must be due to some extent to spatially modulated polymer clustering, typical of spinodal decomposition. In the interval of temperatures studied, it precedes very distinctly in time the thermoreversible sol–gel transition, which is known to be promoted at higher concentrations. It also anticipates to an appreciable extent the spatial density modulation observed in the gel. Although reported here for the first time, a spinodal decomposition of the sol that precedes and possibly triggers the processes leading to gelation does not come unexpectedly in terms of site-bond correlated-percolation theory. In general, this occurrence raises the question as to whether the spontaneous onset of regions of higher and lower polymer concentration (spinodal separation) may be regarded as a novel path for biomolecular interactions and the self-assembly of order in biomolecular systems.  相似文献

A theoretical study of effects of excluded volume intermolecular interactions on the sharpness of helix–coil transitions in solutions of polyamino acids or simple proteins indicates that the transition width may vary appreciably as a function of polymer concentration. The analysis is based on a second virial approximation for the excess free energy of mixing of a solution of polymers of varying degrees of helicity. The virial coefficients involved are roughly estimated on the basis of gross polymer geometry. For large N (degree of polymerization) the transition is found, typically to sharpen with increasing concentration, becoming second order and then first order at sufficiently high concentrations. The critical polymer concentration is found to be roughly of the order N?1.2 ??0?1 for an “all or none” model and of order σ1/2 N?0.2 ??0?1 for a model with continuously variable degree of helicity (??0 is the volume of a single helical molecule and σ1/2 the normalized statistical weight of a helix–coil interface). In the second case for N ~ 103 and σ ~ 10?2–10?4, the predicted critical concentration is in the range 10?1–10?3 g/cm.3 Comparison is made with experiments on solutions of poly(γ-benzyl-L glutamate).  相似文献

The lattice model of Flory has been extended in order to consider equilibrium between isotropic and nematic phases containing helix–coil type chains. Nearly complete exclusion of coil sequences from the lyotropic nematic phase produces an enhanced cooperativity in the helix–coil transition. Potentiometric titrations and some complementary optical rotation data are presented for solutions of poly(L - glutamic acid) (PGA) in several H2O–ethanol mixtures. The data allow the determination of the intrinsic pK (pK0), slope of the apparent. pK (pKapp), versus degree of ionization curves and of the enthalpy of ionization as a function of ethanol concentration. The variation of the degree of ionization at which the helix–coil transformation occurs with ethanol and temperature is also determined. Finally free energy, enthalpy, and intropy changes associated with the helix–coil transformation for the uncharged conformers are determined from the titration curves. The effect of the ethanol is to increase the stability of the helical conformation of PGA for both the charged and the uncharged forms of the polymer. The stabilization of the uncharged helix is essentially an entropic effect.  相似文献

During pregnancy, up‐regulation of heparin‐binding (HB‐) EGF and cyclooxygenase‐2 (COX‐2) in the uterine epithelium contributes to decidualization, a series of uterine morphological changes required for placental formation and fetal development. Here, we report a key role for the lipid mediator lysophosphatidic acid (LPA) in decidualization, acting through its G‐protein‐coupled receptor LPA3 in the uterine epithelium. Knockout of Lpar3 or inhibition of the LPA‐producing enzyme autotaxin (ATX) in pregnant mice leads to HB‐EGF and COX‐2 down‐regulation near embryos and attenuates decidual reactions. Conversely, selective pharmacological activation of LPA3 induces decidualization via up‐regulation of HB‐EGF and COX‐2. ATX and its substrate lysophosphatidylcholine can be detected in the uterine epithelium and in pre‐implantation‐stage embryos, respectively. Our results indicate that ATX–LPA–LPA3 signaling at the embryo‐epithelial boundary induces decidualization via the canonical HB‐EGF and COX‐2 pathways.  相似文献

Abstract The ecotone zone between epigean and hypogean environments has been delimited for two limestone caves using a new method proposed herein. The richness and the diversity of the ecotone, epigean and hypogean environments and their similarities have also been determined. The ecotones were delimited using a similarity matrix between the inner and outer sectors of each cave. The ecotone of Dona Rita's cave was estimated to be 12 m long and the ecotone of Retiro's cave 16 m. The richness (S) of arthropods in Dona Rita's cave was higher in the ecotone (S = 131), intermediate in the epigean environment (S = 75) and lower in the hypogean system (S = 45). The invertebrate diversity (H′) was lower in the hypogean environment (H′ = 2.89) and not statistically different between the epigean environment and the ecotone (H′ = 3.56 and H′ = 3.76, respectively). The richness in Retiro's cave was higher in the ecotone (S = 86), intermediate in the epigean environment (S = 39) and lower in the hypogean system (S = 12). The invertebrate diversity was lower in the hypogean environment (H′ = 0.48), intermediate in the ecotone (H′ = 3.02) and higher in the epigean region (H′ = 3.29). Species migration patterns, differential environmental barriers and determination of accidental versus trogloxenes/troglophylous species are topics that are primarily approached by establishing ecotone zones in caves. The aim of the present paper is to establish the delimitation of theses zones.  相似文献

The thermal triple helix–coil transition of covalently bridged collagenlike peptides with repeating sequences of (Ala-Gly-Pro)n, n = 5–15, was studied optically. The peptides were soluble in water/acetic acid (99:1) and were found to form triple-helical structures in this solvent system beginning with n = 8. The thermodynamic analysis of the transition equilibrium curves for n = 9–13 yielded the parameters ΔH°s = ?7.0 kJ per tripeptide unit, ΔS°s = ?23.1 J deg?1 mol?1 per tripeptide unit for the coil-to-helix transition, and the apparent nucleation parameter σ ? 5 × 10?2. It was suggested through double-jump temperature experiments that the rate-limiting step during refolding is not only influenced by the difficulties of nucleation, but also by cistrans isomerization of the Gly-Pro peptide bond.  相似文献

A simple way to incorporate the solvent–peptide interaction in any available theory of the helix–coil transition is developed. The competition between the intramolecular hydrogen bonding and the solvent–polymer hydrogen bonding is considered in multi-component solvents where some of the components have hydrogen-bonding capacity. Molecular averages are computed by using the theory of Lifson and Roig. The experimental data of Yang are analyzed, and the range of acceptable values of the equilibrium constants of hydrogen bond formation is deduced. The enthalpy of the transition in multicomponent solvents is calculated.  相似文献

The preparation of Ce3+‐doped Sr6AlP5O20 and Ba6AlP5O20 by a combustion method is described. Formation of compounds was confirmed by X‐ray diffraction (XRD) analysis. The photoluminescence (PL) emission spectra were observed at 355 nm when excited at 307 nm for the various concentrations. The PL emission spectra of phosphors showed strong Ce3+ emission due to the 5 d → 4f transition of Ce3+ ions. The Ce3+ emission intensity in Sr6AlP5O20:Ce phosphor was higher than that in Ba6AlP5O20:Ce and it may be useful for scintillation applications. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献

Proton radiotherapy has gained more favor among oncologists as a treatment option for localized and deep-seated tumors. In addition, protons are a major constituent of the space radiation astronauts receive during space flights. The potential for these exposures to lead to, or enhance cancer risk has not been well studied. Our objective is to study the biological effects of low energy protons on epithelial cells and its propensity to enhance transforming growth factor beta 1 (TGFβ1)-mediated epithelial-mesenchymal transition (EMT), a process occurring during tumor progression and critical for invasion and metastasis. Non-transformed mink lung epithelial cells (Mv1Lu) and hTERT- immortalized human esophageal epithelial cells (EPC) were used in this study. EMT was identified by alterations in cell morphology, EMT-related gene expression changes determined using real-time PCR, and EMT changes in specific cellular markers detected by immunostaining and western blotting. Although TGFβ1 treatment alone is able to induce EMT in both Mv1Lu and EPC cells, low energy protons (5 MeV) at doses as low as 0.1 Gy can enhance TGFβ1 induced EMT. Protons alone can also induce a mild induction of EMT. SD208, a potent TGFβ Receptor 1 (TGFβR1) kinase inhibitor, can efficiently block TGFβ1/Smad signaling and attenuate EMT induction. We suggest a model for EMT after proton irradiation in normal and cancerous tissue based on our results that showed that low and high doses of protons can sensitize normal human epithelial cells to mesenchymal transition, more prominently in the presence of TGFβ1, but also in the absence of TGFβ1.  相似文献

Mouse mammary epithelial cells undergo transdifferentiation via epithelial–mesenchymal transition (EMT) upon treatment with matrix metalloproteinase-3 (MMP3). In rigid microenvironments, MMP3 upregulates expression of Rac1b, which translocates to the cell membrane to promote induction of reactive oxygen species and EMT. Here we examine the role of the extracellular matrix (ECM) in this process. Our data show that the basement membrane protein laminin suppresses the EMT response in MMP3-treated cells, whereas fibronectin promotes EMT. These ECM proteins regulate EMT via interactions with their specific integrin receptors. α6-integrin sequesters Rac1b from the membrane and is required for inhibition of EMT by laminin. In contrast, α5-integrin maintains Rac1b at the membrane and is required for the promotion of EMT by fibronectin. Understanding the regulatory role of the ECM will provide insight into mechanisms underlying normal and pathological development of the mammary gland.  相似文献
