Chromosome numbers of taxa of Festuca L. section Eskia Willk. in the Iberian Peninsula are given. The levels of ploidy for five taxa are confirmed. Idiograms and karyotypic formulae of the five taxa are presented for the first time. Two levels of ploidy occur in this section: diploid and tetraploid. One taxon, Festuca elegans ssp. merinoi is tetraploid and two other taxa have diploid and tetraploid populations. We studied hybridization between the diploid Centaurea pseudophrygia and the tetraploid C. jacea by performing crossing experiments and screening natural populations using flow cytometry. The experiments confirm that the studied species exhibit strong reproductive isolation. Interspecific hybrids were formed at a low frequency, including triploids (originating from reduced gametes) and tetraploids (involving unreduced gametes of the diploids). In contrast, hybrids were almost absent among seeds and adult plants of natural mixed populations and among the offspring from experimental pollinations with a mixture of pollen of both ploidy levels. We found that mixed pollination is an important mechanism for preventing hybridization between plants of different ploidy levels and sustaining the reproduction of the tetraploids. A mentor effect (induced selfing in the presence of pollen of different ploidy levels) was observed in both diploids and tetraploids, reinforcing the reproductive isolation between cytotypes. Higher ploidy levels (pentaploid, hexaploid) involving unreduced gametes of the tetraploid species were identified. Notably, pentaploids were discovered for the first time in Centaurea sect. Evolutionary and ecological factors that explain natural variation in ploidy level remain poorly understood. One intriguing possibility is that nutrient costs associated with higher per‐cell nucleic acid content could differentially influence the fitness of different ploidy levels. Here, we test this hypothesis by determining whether access to phosphorus (P), a main component of nucleic acids, differentially affects growth rate in asexual freshwater snails (Potamopyrgus antipodarum) that differ in ploidy. As expected if larger genomes generate higher dietary P requirements, tetraploid P. antipodarum experienced a more than twofold greater reduction in growth rate in low‐P versus high‐P conditions relative to triploids. Mirroring these results, tetraploid P. antipodarum also had a significant reduction in body P content under low P relative to high P, whereas triploid body P content was unaffected. Taken together, these results set the stage for the possibility that P availability could influence the distribution and relative frequency of P. antipodarum of different ploidy levels. These findings could be applicable to many other animal taxa featuring ploidy‐level variation, which includes many mixed sexual/asexual taxa.  相似文献

The systematic structure and postglacial population history of the freshwater amphipod Gammarus lacustris were explored in an allozyme survey of 65 populations across Northern Europe. A strong multilocus pattern of differentiation discriminated populations of the north‐east (north‐eastern Norway, northern Finland) from those in the west and the south (southern and central Scandinavia, Denmark, Poland). This principal division is attributed to postglacial colonization of the area by two main refugial races or lineages, one from the east (Russia), the other from the south (north‐western European continent). The strongly diverged Eastern and Western races (Nei's D= 0.3, from 22 loci) now meet in a secondary contact zone across a narrow sector of northernmost Norway. Genetic population compositions in this zone vary in a mosaic pattern, and show no evidence of reproductive incompatibility. Similar contacts of eastern and western lineages, far older than the latest glaciation, are now known from a number of taxa and they constitute a general pattern in Fennoscandian phylogeography. Within the Western Gammarus race, the populations through coastal north‐western Norway are further distinguished from those in southern Scandinavia and Denmark by a set of unique alleles at high frequencies (D = 0.12). This suggests an independent early colonization of the coastal region by another distinct stock, either along an early deglaciated coastal corridor from the south‐west, or directly from the ice‐free continental shelf off the Norwegian coast – a hypothesis that has also previously been presented for G. lacustris, and parallels controversial suggestions of local refugia for other taxa in Scandinavia. Genetic studies have revealed a large degree of previously unappreciated diversity in morphologically conserved taxa. To understand the implications of this phenomenon, studies investigating the processes responsible for generating and maintaining functional and neutral diversity within such taxa are needed. With this aim, patterns of differentiation across a contact zone between two catchment‐specific, and genetically and subtly morphologically distinguishable, races of the onychophoran Euperipatoides rowelli Reid, 1996 at the Tallaganda State Forest study system, Australia, were quantified using diagnostic body‐pattern differences, microsatellite markers, and the cytochrome c oxidase subunit I (COI) mitochondrial gene. These data were used to test whether hybridization occurred, and whether the reduced survival of hybrids compared with non‐hybrids and/or assortative mating were potentially important factors in retaining the divergence between these races, which probably arose during isolation in Pleistocene glacial refuges. It was found that hybrids at the focal contact zone do not have reduced embryo‐to‐adult survival compared with non‐hybrids, and that races apparently freely interbreed without substantial assortative mating, consistent with little or no cost to hybridization. Nonetheless, at their respective transect ends the races remain distinct. This phenomenon was attributed to the elevated location of the contact zone, which is likely to make it poor habitat for E. rowelli, and thus may restrict dispersal and gene flow. Simultaneous hybridizations among three or more parental taxa occur in nature, but these have rarely been analysed. The present study investigates the natural hybridization of diploid members of the Ranunculus cantoniensis complex over a microgeographical area in south‐western China. Sequence information from maternally inherited plastid DNA (trnQ‐rps16 and rpL32‐trnL) and biparentally inherited nuclear ribosomal DNA was used to identify hybrids, and these were further confirmed by a dramatic reduction in pollen viability. The populations from the contact zone contained individuals that had more than two nuclear ribosomal internal transcribed spacer (nrITS) types; in addition, a network analysis revealed the presence of hard conflicts between the nuclear and plastid DNA data. These results prove that R. trigonus and R. silerifolius var. dolicanthus have relatively little reproductive isolation from each other, leading to a high gene flow rate in contact populations. Sorbus subgenus Soraria encompasses taxa originating from spontaneous hybridization between members of subgenera Aria and Sorbus disjunctly distributed across Europe and Asia. Using molecular data (amplified fragment length polymorphisms, plastid DNA sequences and nuclear microsatellites), flow cytometry (allowing for the determination of ploidy and mode of reproduction) and morphology, we disentangled the relationships among polyploid cytotypes and explored their relationships with their diploid ancestors. Among others, we focused on a large, geographically isolated hybrid population in Bosnia and Herzegovina. Molecular and morphological analyses confirmed the distinct position of this population in relation to its parental (S. aria and S. aucuparia) and other hybridogenous taxa originating independently from the same parents in different parts of Europe. Aim Newly formed polyploids experience problems of establishment and spread similar to those faced by newly introduced alien species. To understand the significance of polyploidy in biological invasions, we mapped the distribution of ploidy levels in Solidago gigantea Aiton in its native range in North America, and in Europe and East Asia where it is invasive. Location North America, Europe and East Asia. Methods Flow cytometry was used to measure ploidy levels in a total of 834 plants from 149 locations. Together with data from contributors and a literature review, ploidy‐level data were assembled for 336 locations. Cytogeographical maps from North America and Europe were prepared, incorporating new and previously published ploidy‐level data. Results In the native range, diploid, tetraploid and hexaploid plants were found, and also one triploid and one pentaploid plant (2n = 3x and 2n = 5x, respectively, each being new reports for this species). There was a high degree of geographical separation among the ploidy levels, and populations with mixed ploidy were rare. However, four zones were identified where plants of different ploidy could come into contact. In Europe and East Asia, only tetraploid plants were found. Main conclusions The geographical pattern in North America suggests that the ploidy levels are ecologically differentiated, although further investigations are needed to identify the nature of these differences. Alien populations appear to be exclusively tetraploid, but it is not clear whether this is because tetraploids were selectively introduced or because diploids were unsuccessful. In any case, comparisons between native and introduced populations need to account for ploidy level.  相似文献

Approximately 70% of the angiosperm species are polyploid, an important phenomenon in the evolution of those plants. But ploidy estimates have often been hindered because of the small size and large number of chromosomes in many tropical groups. Since polyploidy affects cell size, morphometric analyses of pollen grains and stomata have been used to infer ploidy level. Polyploidy is present in many species of the Cerrado, the Neotropical savanna region in Central Brazil, and has been linked to apomixis in some taxa. Eriotheca gracilipes and Eriotheca pubescens are common tree species in this region, and present cytotypes that form reproductive mosaics. Hexaploid individuals (2n = 6x = 276) are polyembryonic and apomictic, while tetraploid and diploid individuals (2n = 2x = 92, 2n = 4x = 184) are sexual and monoembryonic. We tested whether morphometric analysis can be used to estimate ploidy levels in E. gracilipes and E. pubescens individuals. Pollen material from diploid and hexaploid individuals of E. gracilipes, and tetraploid and hexaploid individuals of E. pubescens, were fixed in 50% FAA, and expanded leaves were dried in silica gel. Pollen grains and stomata of at least five individuals from each population were measured. The results demonstrate that all measures were significantly different among cytotypes. Individuals with higher levels of ploidy (hexaploid) all presented measurements that were higher than those with lower levels (diploid and tetraploid). There was no overlap between ploidy levels in each species at 95% confidence interval. Thus, the size of the pollen grains and stomata are effective parameters for analysis of ploidy levels in E. gracilipes and E. pubescens.  相似文献

Reconstructing evolutionary history for emerging species complexes is notoriously difficult, with newly isolated taxa often morphologically cryptic and the signature of reproductive isolation often restricted to a few genes. Evidence from multiple loci and genomes is highly desirable, but multiple inputs require ‘common currency’ translation. Here we deploy a Shannon information framework, converting into diversity analogue, which provides a common currency analysis for maternally inherited haploid and bi‐parentally inherited diploid nuclear markers, and then extend that analysis to construction of minimum‐spanning networks for both genomes. The new approach is illustrated with a quartet of cryptic congeners from the sexually deceptive Australian orchid genus Chiloglottis, still in the early stages of speciation. Divergence is more rapid for haploid plastids than for nuclear markers, consistent with the effective population size differential (Nep < Nen), but divergence patterns are broadly correlated for the two genomes. There are nevertheless intriguing discrepancies between the emerging plastid and nuclear signals of early phylogenetic radiation of these taxa, and neither pattern is entirely consistent with the available information on the sexual cues used by the orchids to lure the pollinators enforcing reproductive isolation. We describe possible extensions of this methodology to multiple ploidy levels and other types of markers, which should increase the range of application to any taxonomic assemblage in the very early stages of reproductive isolation and speciation.  相似文献

The reproductive biology of the genusDamasonium was studied in cultivation and in nature. A correlation was shown between flowering time and latitude. All species are self-compatible but the N. American member of the genus (D. californicum) is strongly protandrous and probably experiences high levels of outcrossing in nature. In the European taxa the stamens and the stigmas mature simultaneously but the levels of inbreeding appear to be greater in the southern species (D. bourgeai, D. polyspermum) than in the northern (tetraploid) speciesD. alisma. The Australian memberD. minus resembles the southern European species in being strongly autogamous and bud-pollination was shown to occur. Floral morphology, stigma exertion, pollen/ovule (P/O) ratio and seed weight each show a strong correlation with the degree of autogamy. Vegetative reproduction occurs only inD. californicum andD. alisma through the production of corm-buds. This study suggests that autogamy inDamasonium is accompanied by a shift in resource allocation from male (pollen) toward female (seed) function. However, further studies are required to assess the quality of seeds and their survivorship in nature.  相似文献

When reproductive barriers break down, interspecific hybridization can lead to gene flow between evolutionarily distinct species. Studying the fate of these introgressing elements can offer valuable insights into the factors contributing to reproductive isolation. We have identified a population of false map turtles (Graptemys pseudogeographica) that hybridized historically with the common map turtle (Graptemys geographica), but were subsequently isolated from interbreeding for several generations by unique geological events. Although many studies conclude that genic interactions involving sex chromosomes impact the introgression of mitochondrial or nuclear genomes, Graptemys turtles have environmental sex determination, and thus introgression can be explored while controlling for the effects of sex‐specific heterogameity. We identified and sequenced a species‐specific mitochondrial control region marker, as well as two nuclear markers (ODC and HNFAL), in turtles from across the ranges of these species. We found both nuclear and mitochondrial introgression in our study population, and present evidence consistent with the proposed time range of reproductive contact and isolation. We also report an absence of cytonuclear or linkage disequilibrium among markers, indicating that some important pre‐ and postzygotic barriers to gene flow that characterize other systems are absent in Graptemys. Phenotypic differentiation of two tetraploid (2n = 4x = 36, 36+1B, 36+2B) populations of Santolina rosmarinifolia geographically isolated from diploid populations was investigated. The karyotype was relatively homogeneous, meiosis was regular and pollen was fertile in both cytotypes. An autopolyploid or allopolyploid origin for tetraploid cytotypes is discussed. Overall, 80.82% of all variance in achene weight, time t0, t50 and t90 of germination and accumulated germination rate was due to achene age at each ploidy level. Partition of the total phenotypic variance showed that there was extensive variation between ploidy levels. The mean of morphological characters was generally higher in polyploids. For diploid cytotypes, flower number, achene production and fruiting percentage were significantly higher than for tetraploid cytotypes. Cluster analysis indicated that the patterns of seedling morphology and development were similar in three diploid individuals and several tetraploids; the same analysis showed high similarity between diploid individuals of the natural populations, whereas tetraploid individuals showed high dissimilarity among themselves and with diploid individuals. Multiple correspondence analysis and logistic regression analysis indicated that qualitative characters contribute strongly to cytotype differentiation. Portulaca oleracea is a noxious annual weed of worldwide distribution in temperate to tropical climates. Its taxonomy has been treated in contradictory ways in the past. Various microspecies have been described, lumped into a single species by other authors. We re‐examined the importance of seed size and ploidy variation, previously applied as the most important taxonomic characters, for systematic classification based on accessions from Europe, Asia, Africa and South America using flow cytometry, chromosome counting and morphometry. Sixteen microspecies and six transitional forms, covering the ploidy and seed character variation, proposed for the complex, were studied from 178 populations. Portulaca grandiflora was included as a reference species from outside the complex. DNA hyper‐pentaploidy or hexaploidy were inferred for the majority of accessions which exhibited the full range of seed size. It is recommended that the only species of lower ploidy (either diploid based on x = 12 or tetraploid based on x = 12) encountered, P. nicaraguensis, should be separated from the P. oleracea complex as it deviates in base chromosome number and monoploid genome size. The frequency distribution of seed size was continuous and unimodal within the wild taxa of the complex and in pairs of taxa defined by testa sculpture. Seed size of DNA hexaploids was slightly negatively correlated with sample/standard fluorescence intensities. Our results conflict with the current microspecies concept.  From the predominantly aposporous Ranunculus cassubicus group, a subgroup of the R. auricomus complex, two species with both diploid and tetraploid cytodemes (R. cassubicifolius W. Koch and R. carpaticola Soó) were known. Nine population samples of both species have been analyzed for variation of ploidy levels and isozymes. DNA image analysis showed that three populations of R. cassubicifolius from Bavaria and Salzburg are diploid, two from Lower Austria tetraploid. In R. carpaticola, two populations from Central Slovakia and one from Romania are diploid, one from northern Slovakia is hexaploid. The polyploid populations had somewhat smaller C-values than expected from diploids. Ploidy levels are consistent within populations, contradicting previous hypotheses that cycles of diploid-tetra-ploid-dihaploid apomictic individuals occur in natural populations of goldilocks. Isozyme-allozyme analysis of nine polymorphic loci showed that individual variation and genetic measures of di-ploid and tetraploid populations are equivalent to those of sexual taxa. Multiple allelism and unbalanced gene dosages in tetraploid R. cassubicifolius give evidence for autopolyploidy that is most probably of multiple origin. The observed excess of homozygotes and the high diversity between populations in the diploid populations of R. cassubicifolius are most probably due to geographical isolation of population groups, that might have happened during the Würm glaciation and might have promoted the separation of autotetraploids in the easternmost part of the distribution range. Genetic distance values analyzed by UPGMA of all sexual populations separated the two taxa and the cytodemes within R. cassubicifolius. Multidimensional scaling of individuals (including the apomicts) based on a presence/absence matrix of alleles confirmed this differentiation with an overlapping zone between R. cassubicifolius and R. carpaticola. The hexaploid R. carpaticola population showed reduced genotypic diversity, an increased number of heterozygotes and fixed heterozygosity as typical for apomictic mode of reproduction. This population shared alleles of both species without any specific ones, multidimensional scaling placed genotypes among R. cassubicifolius. Thus, the hexa-ploid apomicts might have originated from hybrids of R. cassubicifolius and R. carpaticola. In general, evolution of agamic lineages in the R. cassubicus complex might have been facilitated by autopoly-ploids, because they can provide a bridge between the reproductive systems that are otherwise isolated by ploidy levels, and also a starting point for spontaneous origin of apomixis. We quantify the population divergence processes that shaped population genetic structure in the Trans‐Volcanic bunchgrass lizard (Sceloporus bicanthalis) across the highlands of south‐eastern Mexico. Multilocus genetic data from nine nuclear loci and mitochondrial (mt)DNA were used to estimate the population divergence history for 47 samples of S. bicanthalis. Bayesian clustering methods partitioned S. bicanthalis into three populations: (1) a southern population in Oaxaca and southern Puebla; (2) a population in western Puebla; and (3) a northern population with a broad distribution across Hidalgo, Puebla, and Veracruz. The multilocus nuclear data and mtDNA both supported a Late Pleistocene increase in effective population size, and the nuclear data revealed low levels of unidirectional gene flow from the widespread northern population into the southern and western populations. Populations of S. bicanthalis experienced different demographic histories during the Pleistocene, and phylogeographical patterns were similar to those observed in many co‐distributed highland taxa. Although we recommend continuing to recognize S. bicanthalis as a single species, future research on the evolution of viviparity could gain novel insights by contrasting physiological and genomic patterns among the different populations located across the highlands of south‐eastern Mexico. © 2013 The Linnean Society of London, Biological Journal of the Linnean Society, 2013, 110 , 852–865.  相似文献

Polymorphism analyses of the hordeins, main storage proteins in barley, were conducted on 35 natural populations of Hordeum murinum s.l. from North Africa; this specific complex includes three subspecies with two ploidy levels: H. murinum subsp. glaucum (2n=2x=14), H. murinum subsp. leporinum and subsp. murinum (2n=4x=28). Twenty of these populations belong to the diploid subsp. glaucum, 14 other tetraploid populations belong to the subsp. leporinum. In addition, six populations of the tetraploid murinum were sampled in France: two along the Mediterranean coast and four in Brittany. The polymorphism observed in the electrophoretic patterns highlights strong correlations between bioclimatic features and di- and tetraploid taxa distribution. Moreover, the variation was not randomly distributed within the different ploidy levels, and is correlated with environmental factors. The ecological differentiation of the two main taxa, H. murinum subsp. leporinum and subsp. glaucum is clearly highlighted.  相似文献

Close affinities recognized between taxa in Mexico and the contiguous USA have led to a variety of biogeographical scenarios. One such hypothesis suggests that species that occur in both countries have an origin in central Mexico followed by dispersal into the USA. This study expands upon previous phylogeographical work of the ringneck snake Diadophis punctatus by incorporating new data from previously unsampled areas appropriate to critically assess hypotheses regarding a Mexican origin for this species. Maximum likelihood and maximum parsimony analyses inferred a derived position for the lineage from southern Mexico with constraint tests for alternate evolutionary hypotheses resulting in significantly worse likelihood values. Ancestral area reconstructions inferred an origin for D. punctatus in the south‐eastern USA followed by a south‐east to north‐east then westward directionality of historical migration. The position within the phylogeny and date estimate for the south‐western + Mexico clade suggests a recent invasion into central Mexico with expansion into the Nearctic/Neotropic transition zone. The polyploid species complex of Rosa villosa sensu lato (Rosa section Caninae subsection Vestitae) consists of three morphologically similar polyploid species: R. sherardii, R. mollis and R. villosa. Whereas R. sherardii is distributed in central Europe, R. mollis and R. villosa represent a vicariant species pair occurring in northern Europe and in mountains of central and eastern Europe, respectively. In this study we analysed multiple data sets (morphology, cytology, microsatellites, AFLP and plastid DNA sequences) to re‐evaluate the systematics of these species and to examine whether cytological differences are reflected in the taxonomy and geographical distributions. Furthermore, these data sets were used to evaluate hypotheses explaining the vicariant distribution of R. mollis and R. villosa. None of these data sets revealed a clear‐cut differentiation between the species. Cytological and molecular data argued for a discrete taxonomic position of the predominantly pentaploid R. sherardii, but these data did not support a separation between the mostly tetraploid R. mollis and R. villosa. Population genetics revealed that samples of the latter species were assembled according to ploidy, but not to species affiliation or geographical distribution. The deciphering of the process of genetic differentiation of species with insular distributions is relevant for biogeographical and conservation reasons. Despite their importance as old gondwanic islands and part of the western Indian Ocean biodiversity hotspot, little is known about the genetic structure of taxa from the Seychelles Islands. We have examined the patterns of structure and isolation within Urocotyledon inexpectata (Reptilia: Geckkonidae), an endemic species from this archipelago. Genetic diversity was screened from populations across the archipelago for both mitochondrial and nuclear genes. Gene genealogies and model‐based inference were used to explore patterns and timings of isolation between the main lineages. High levels of genetic diversity were found for the mitochondrial and some of the nuclear markers. This species harbours at least two highly differentiated lineages, exclusively distributed across the northern and southern groups of the islands. The main split between these was dated back to the Miocene–late Pliocene, but isolation events throughout the Pliocene and Pleistocene were also inferred. Migration between groups of islands was apparently nonexistent, except for one case. The low dispersal capabilities of this species, together with the intrinsic fragmented nature of its geographical distribution, seem to have resulted in highly structured populations, despite the cyclic periods of contact between the different island groups. These populations may currently represent more than one species, making U. inexpectata another example of a morphologically cryptic lineage with deep genetic divergence within gekkonids. The observed patterns suggest a hypothetical biogeographic scenario (of a main north–south phylogeographic break) for the Seychelles that can be further tested with the exploration of the phylogeographic structure of other Seychellois taxa. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 104 , 177–191.  相似文献
