An improved method is described for separating saturated fatty acids by reversed-phase paper chromatography. The 2,4-dinitrophenylhydrazides prepared from saturated fatty acids from C2 to C22 are run upward on paper impregnated with tetralin, using 90% methanol—acetic acid—tetralin, 80% ethanol—acetic acid—tetralin, or 90% methanol—tetralin as the moving solvent. The simultaneous separations of all even-carbon acids from C6 to C22, all odd-carbon acids from C7 to C19, and also all odd- and even-carbon acids from C7 to C19 can be successfully performed by means of this paper chromatography. The method is useful for the detection of component saturated fatty acids in natural fats.  相似文献

There are three mycolic acid homologues with C22-, C24- and C26-α-units in Mycobacterium. In order to reveal the composition and distribution of these homologues in each subclass and molecular species of mycolic acids and to compare them with the composition of constitutive non-polar fatty acids (free and bound forms), we have separated non-polar fatty acids and each subclass of mycolic acids from 21 mycobacterial species by thin-layer chromatography, and analyzed non-polar fatty acid methyl esters by gas chromatography (GC) and the cleavage products of methyl mycolate by pyrolysis GC. We further performed mass chromatographic analysis of trimethylsilyl (TMS) ether derivatives of mycolic acid methyl esters by monitoring [B-29]+ ions (loss of CHO from the α-branched-chain structure of mycolic acids) of m/z 426, 454 and 482 which are attributed to C22-, C24- and C26-α-units of TMS ether derivatives of methyl mycolates, respectively, (Kaneda, K. et al, J. Clin. Microbiol. 24: 1060-1070, 1986). By pyrolysis GC, C22:0, C24:0 and C26:0 fatty acid methyl esters generated by the C2-C3 cleavage of C22-, C24- and C26-α-unit-containing mycolic acid methyl esters, respectively, were detected. Their proportion was almost the same among subclasses of mycolic acids in every Mycobacterium and also similar to the proportion of constitutive non-polar C22:0, C24:0 and C26:0 fatty acids. By mass chromatography, the composition and distribution of C22- and C24-α-unit-containing homologues were revealed to be similar between α- and α'-mycolic acids in every Mycobacterium. We further analyzed in detail M. vaccae and demonstrated that the mass chromatogram of C22-α-unit-containing homologue was analogous in shape to that of the C24-α-unit-containing one, with the latter mass chromatogram being up-shifted from the former by two carbon numbers, in every subclass of α-, α'-, keto and dicarboxy mycolic acids. The present study suggests that the compositions of three homologues of both mycolic acids and constitutive non-polar fatty acids, which are characteristic to each mycobacterial species, may reflect the proportion of the amount of free C22:0, C24:0 and C26:0 fatty acids synthesized in the cell. It is further demonstrated that intermolecular condensation of two fatty acids which become α- and β-units of mycolic acids will occur independently of the carbon chain length or kinds of polar moieties of fatty acids.  相似文献

Biliary bile salt composition of 677 vertebrate species (103 fish, 130 reptiles, 271 birds, 173 mammals) was determined. Bile salts were of three types: C27 bile alcohols, C27 bile acids, or C24 bile acids, with default hydroxylation at C-3 and C-7. C27 bile alcohols dominated in early evolving fish and amphibians; C27 bile acids, in reptiles and early evolving birds. C24 bile acids were present in all vertebrate classes, often with C27 alcohols or with C27 acids, indicating two evolutionary pathways from C27 bile alcohols to C24 bile acids: a) a ‘direct’ pathway and b) an ‘indirect’ pathway with C27 bile acids as intermediates. Hydroxylation at C-12 occurred in all orders and at C-16 in snakes and birds. Minor hydroxylation sites were C-1, C-2, C-5, C-6, and C-15. Side chain hydroxylation in C27 bile salts occurred at C-22, C-24, C-25, and C-26, and in C24 bile acids, at C-23 (snakes, birds, and pinnipeds). Unexpected was the presence of C27 bile alcohols in four early evolving mammals. Bile salt composition showed significant variation between orders but not between families, genera, or species. Bile salt composition is a biochemical trait providing clues to evolutionary relationships, complementing anatomical and genetic analyses.  相似文献

Monoacylglycerols containing α-branched-β-hydroxylated fatty acids (mycolic acids) ranging from C42 to C50 and from C60 to C66, were isolated from Gordona lentifragmenta and from G. bronchialis, respectively. On the other hand, G. rubropertincta showed only a monoacylglycerol fraction which released non-hydroxylated fatty acids; they were identified as C16:0-, C16:1,- C18:1- and branched C19:0-fatty acids. This last component was identified as 10-methyl octadecanoic acid (tuberculostearic acid).  相似文献

The 13C-NMR spectra of 48 cis alkenoic acids and esters (C8–C20), 18 trans alkenoic acids and esters (C9–C18), and 26 polyenoic acids and esters (C18–C22) are reported and interpreted. Wax esters from the epicuticular wax of juvenile and mature-tree Pinus radiata foliage have been shown by capillary column GC-MS to consist mainly of short chain (C6–C12)alkanols esterified with long chain acids (C24–C32) and long chain alkanols (C24–C32) esterified with short chain acids (C6-C14) in a non-random manner. Mature-tree foliage wax esters also contained nonacosan-10-ol esterified with dodecanoic and tetradecanoic acids.  相似文献

The monocarboxylic fatty acids and hydroxy fatty acids of three species of freshwater microalgae—Vischeria punctata Vischer, Vischeria helvetica (Vischer et Pascher) Taylor, and Eustigmatos vischeri (Hulbert) Taylor, all from the class Eustigmatophyceae— were examined. Each species displayed a very similar distribution of fatty acids, the most abundant of which were 20:5n-3, 16:0, and 16:1n-7; C18 polyunsaturated fatty acids were minor components. These fatty acid distributions closely resemble those found in marine eustigmatophytes but are quite distinct from those found in most other algal classes. These microalgae also contain long-chain saturated and unsaturated monohydroxy fatty acids. Two distinct types of hydroxy fatty acids were found: a series of saturated α-hydroxy acids ranging from C24 to C30 with a shorter series of monounsaturated α-hydroxy acids ranging from C26 to C30 together with a series of saturated β-hydroxy acids ranging from C26 to C30. The latter have not previously been reported in either marine or freshwater microalgae, although C30 to C34 midchain (ω-18)-hydroxy fatty acids have been identified in hydrolyzed extracts from marine eustigmatophytes of the genus Nannochloropsis, and C22 to C26 saturated and monounsaturated α-hydroxy fatty acids have been found in three marine chlorophytes. These findings have provided a more complete picture of the lipid distributions within this little studied group of microalgae as well as a range of unusual compounds that might prove useful chemotaxonomic markers. The functions of the hydroxy fatty acids are not known, but a link to the formation of the lipid precursors of highly aliphatic biopolymers is suggested.  相似文献

In isolated tobacco leaves l-valine-U-14C gave rise to labeled even-numbered isobranched fatty acids containing 16 to 26 carbon atoms and iso C29, iso C31, and iso C33 paraffins. l-Isoleucine-U-14C on the other hand produced labeled odd-numbered anteiso C17 to C27 fatty acids and anteiso C30 and C32 paraffins. Trichloroacetic acid inhibited the incorporation of isobutyrate into C20 and higher fatty acids and paraffins without affecting the synthesis of the C16 and C18 fatty acids. Thus the very long branched fatty acids are biosynthetically related to the paraffins. In Senecio odoris leaves acetate-1-14C was incorporated into the paraffins (mainly n-C31) only in the epidermis although acetate was readily incorporated into fatty acids in the mesophyll tissue. Similarly only the epidermal tissue incorporated acetate into fatty acids longer than C18 suggesting that the epidermis is the site of synthesis of both paraffins and the very long fatty acids. In broccoli leaves n-C12 acid labeled with 14C in the carboxyl carbon and 3H in the methylene carbons was incorporated into C29 paraffin without the loss of 14C relative to 3H. Since n-C18 acid is known to be incorporated into the paraffin without loss of carboxyl carbon these results suggest that the condensation of C12 acid with C18 acid is not responsible for n-C29 paraffin synthesis in this tissue. Thus all the experimental evidence thus far obtained strongly suggests that elongation of fatty acids followed by decarboxylation is the most likely pathway for paraffin biosynthesis in leaves.  相似文献

In the preceding paper on the interrelation between sucrose ester of fatty acid and biotin, the fatty acid being a mixture of C10 to C18 acid, it was described that carbon chain length of fatty acid has a great influence on the accumulation of l-glutamic acid. Fatty acids with C12 to C18 chain length, particularly myristic, palmitic and margaric acids were effective on the accumulation of l-glutamic acid in the culture medium containing sufficient biotin, whereas lower and higher length acids were ineffective. In the form of polyoxyethylene sorbitan or polyethylene glycol ester, C16 and C18 acids were remarkably effective. However, the ester of C12 acid and polyoxyethylene ethers of C12 to C18 alcohols had little or no effect.  相似文献

A detailed chemical analysis of the benzene extract of western white pine bark was conducted. The extract consisted of 13% phlobaphenes, 18% strong acids, 21% polar weak acids, 6.5% fatty acids, 9.5% resin acids, and 32% neutrals. The fatty acids consisted mainly of C20:0, C22:0, and C24:0 acids. The resin acids were identified as: isopimaric, anticopalic, dehydroabietic, sandaracopimaric, abietic, 6,8,11,13-abietatetraen-18-oic and pimaric acids. The neutrals on saponification gave fatty acids, sterols, wax alcohols, nonsaponifiables, and other components. The esterified fatty acids consisted primarily of the C16:0, C18:0, C20:0 and C24:0 acids. The sterols included major amounts of sitosterol, campesterol, and stigmasterol, and traces of cholesterol. Over 70 individual compounds were isolated and identified from the nonsaponifiables. These included borneol, sesquiterpenes, diterpenes, steroidal ketones, as well as lanostane and serratane triterpenes. The characterization of12 new natural products or natural products isolated for the first time from Pinus species is reported.  相似文献

The structures of ten fatty acids, which were obtained by the hydrolysis of tunicamycin complex, were determined. GLC-mass, 1H NMR and IR spectra showed that the major acids were trans-α, β-unsaturated iso acids with the formula C14H28O2, C16H28O2, C16H30O2 and C17H32O2. The minor acids were α, β-unsaturated normal acids and saturated normal and iso acids.  相似文献

Fatty acids in vesicular and leaf monogalactosyl diglycerides (MGDG) of citrus were studied. Vesicular MGDG contained front 94.4 to 97.3% C16, C16:1, C18:1, C18:2, and C18:3; whereas leaf MGDG contained ca 90% C18:3, 3% C16 and 1.8 to 9.5% C18:2. Species varied considerably in their percentages of vesicular C18:2, C18:3 and to a lesser degree, C16:1 and C18:1 fatty acids with lemons being the most distinctive. Branched fatty acids were present to the extent of 5.6% in vesicular and to only 0.1% in leaf MGDG.  相似文献

Fatty acids fromChlorella vulgaris, Scenedesmus obliquus var.acutus and from a mixed culture of the two strains, Melnik, were converted to methyl esters, separated by gas chromatography, and identified by means of standards. The spectrum of fatty acids included both saturated and unsaturated acids (with odd and even numbers of carbon atoms) from C12 to C22. Fatty acids C16:0, C18:0 and C20:3 were the major components in all cultures. Pure strains differed from the mixed culture in the production of C18:1, C12:0 and C19:2 acids; the first of these was present in higher amounts in pure cultures only, the latter two being found in the mixed culture. The level of lipids was lower as compared to the literature data and their extractability was affected by the manner of preparation of algae and extraction conditions.  相似文献

Lycopersicon pennellii Corr. (D'Arcy) an insect-resistant, wild tomato possesses high densities of glandular trichomes which exude a mixture of 2,3,4-tri-O-acylated glucose esters that function as a physical impediment and feeding deterrent to small arthropod pests. The acyl moieties are branched C4 and C5 acids, and branched and straight chain C10, C11, and C12 acids. The structure of the branched acyl constituents suggests that the branched chain amino acid biosynthetic pathway participates in their biosynthesis. [14C]Valine and deuterated branched chain amino acids (and their oxo-acid derivatives) were incorporated into branched C4 and C5 acid groups of glucose esters by a process of transamination, oxidative decarboxylation and subsequent acylation. C4 and C5 branched acids were elongated by two carbon units to produce the branched C10-C12 groups. Norvaline, norleucine, allylglycine, and methionine also were processed into acyl moieties and secreted from the trichomes as glucose esters. Changes in the acyl composition of the glucose esters following sulfonylurea herbicide administration support the participation of acetohydroxyacid synthetase and the other enzymes of branched amino acid biosynthesis in the production of glucose esters.  相似文献

Cold-loving microorganisms developed numerous adaptation mechanisms allowing them to survive in extremely cold habitats, such as adaptation of the cell membrane. The focus of this study was on the membrane fatty acids of Antarctic Flavobacterium spp., and their adaptation response to cold-stress. Fatty acids and cold-response of Antarctic flavobacteria was also compared to mesophilic and thermophilic members of the genus Flavobacterium. The results showed that the psychrophiles produced more types of major fatty acids than meso- and thermophilic members of this genus, namely C15:1 iso G, C15:0 iso, C15:0 anteiso, C15:1 ω6c, C15:0 iso 3OH, C17:1 ω6c, C16:0 iso 3OH and C17:0 iso 3OH, summed features 3 (C16:1 ω7cand/or C16:1 ω6c) and 9 (C16:0 10-methyl and/or C17:1 iso ω9c). It was shown that the cell membrane of psychrophiles was composed mainly of branched and unsaturated fatty acids. The results also implied that Antarctic flavobacteria mainly used two mechanisms of membrane fluidity alteration in their cold-adaptive response. The first mechanism was based on unsaturation of fatty acids, and the second mechanism on de novo synthesis of branched fatty acids. The alteration of the cell membrane was shown to be similar for all thermotypes of members of the genus Flavobacterium.  相似文献

Analyses were made of the fatty-acid composition of Candida utilis NCYC 321 grown in a chemostat at a dilution rate (equal to growth rate) of 0.1 hr−1 and at temperatures in the range of 30 to 15 C and dissolved oxygen tensions between 75 and <1 mm of Hg. Cells grown under glucose limitation or NH4+ limitation contained mainly C16:0, C16:1, C18:0, C18:1, C18:2, and C18:3 acids as detected by gas-liquid chromatography of methyl esters of the acids from lipids extracted with chloroform-methanol. The relative proportions of these acids varied with the growth temperature and the dissolved-oxygen tension in the culture. A decrease in growth temperature from 30 to 20 C led to an increased synthesis of unsaturated acids in cells grown under either limitation at a fixed-oxygen tension in the range of 75 to 5 mm of Hg. In cultures with a dissolved-oxygen tension of 1 and <1 mm of Hg, a further decrease in temperature to 15 C caused an increased synthesis of unsaturated fatty acids. A decrease in dissolved-oxygen tension led to a diminished synthesis of unsaturated fatty acids in cells grown at a fixed temperature under either limitation. Cells grown at a fixed temperature under glucose limitation synthesized a greater proportion of C16 acids at the expense of C18 acids as the dissolved oxygen tension was decreased from 75 to <1 mm of Hg. A preferential synthesis of C16 acids also occurred as the growth temperature was decreased from 30 to 15 C in cells grown under glucose limitation at a fixed-oxygen tension. The same effect was observed in cells grown under NH4+ limitation when the temperature was lowered from 30 to 20 C; but when the temperature was decreased further to 15 C, the cells synthesized a slightly greater proportion of C18 acids. Synthesis of a large proportion of C16 acids was accompanied by an excretion of pyruvate, and occasionally traces of 2-ketoglutarate, and an increased intracellular accumulation of certain amino acids.  相似文献

The degree of C4 photosynthesis was assessed in four hybrids among C4, C4-like, and C3-C4 species in the genus Flaveria using 14C labeling, CO2 exchange, 13C discrimination, and C4 enzyme activities. The hybrids incorporated from 57 to 88% of the 14C assimilated in a 10-s exposure into C4 acids compared with 26% for the C3-C4 species Flaveria linearis, 91% for the C4 species Flaveria trinervia, and 87% for the C4-like Flaveria brownii. Those plants with high percentages of 14C initially fixed into C4 acids also metabolized the C4 acids quickly, and the percentage of 14C in 3-phosphoglyceric acid plus sugar phosphates increased for at least a 30-s exposure to 12CO2. This indicated a high degree of coordination between the carbon accumulation and reduction phases of the C4 and C3 cycles. Synthesis and metabolism of C4 acids by the species and their hybrids were highly and linearly correlated with discrimination against 13C. The relationship of 13C discrimination or 14C metabolism to O2 inhibition of photosynthesis was curvilinear, changing more rapidly at C4-like values of 14C metabolism and 13C discrimination. Incorporation of initial 14C into C4 acids showed a biphasic increase with increased activities of phosphoenolpyruvate carboxylase and NADP-malic enzyme (steep at low activities), but turnover of C4 acids was linearly related to NADP-malic enzyme activity. Several other traits were closely related to the in vitro activity of NADP-malic enzyme but not phosphoenolpyruvate carboxylase. The data indicate that the hybrids have variable degrees of C4 photosynthesis but that the carbon accumulation and reduction portions of the C4 and C3 cycles are well coordinated.  相似文献

The effect of growth medium NaCl concentration on the fatty acid composition of phospholipids of 3 strains of Saccharomyces cerevisiae and 6 osmotolerant yeast strains was examined. The S. cerevisiae strains were characterized by a high content of palmitoleic (C16:1) acid and by having no polyunsaturated C18 acids, whereas the osmotolerant strains had a low content of C16:1 and a high proportion of polyenoic C18 acids. An increase of the NaCl concentration from 0% to 8% resulted in a decrease of the cellular phospholipid content on a dry-weight basis, for all strains but one of the osmotolerant strains. For the S. cerevisiae strains increased salinity produced a slight decrease of the proportion of C16 fatty acids with a concomitant increase of C18 acids, whereas the osmotolerant strains showed an increase of the relative content of oleic acid (C18:1) at the expense of the proportion of polyenoic C18 acids.  相似文献

In this paper, partitioning behaviors of typical neutral (Alanine), acidic (Glutamic acid) and basic (Lysine) amino acids into imidazolium-based ionic liquids [C4mim][PF6], [C6mim][PF6], [C8mim][PF6], [C6mim][BF4] and [C8mim][BF4] as extracting solvents were examined. [C6mim][BF4] showed the best efficiency for partitioning of amino acids. The partition coefficients of amino acids in ionic liquids were found to depend strongly on pH of the aqueous solution, amino acid and ionic liquid chemical structures. Different chemical forms of amino acids in aqueous solutions were pH dependent, so the pH value of the aqueous phase was a determining factor for extraction of amino acids into ionic liquid phase. Both water content of ionic liquids and charge densities of their anionic and cationic parts were important factors for partitioning of cationic and anionic forms of amino acids into ionic liquid phase. Extracted amino acids were back extracted into phosphate buffer solutions adjusted on appropriate pH values. The results showed that ionic liquids could be used as suitable modifiers on the stationary phase of an HPLC column for efficient separation of acidic, basic, and neutral amino acids.  相似文献
