Mice were immunized intravenously with 4 × 107 thymocytes from Thy-1 disparate, eitherH-2-compatible orH- 2-incompatible donors. The magnitude of the anti-Thy-1.1 response was measured by determining the number of PFC in spleens of animals 6 days after immunization. Regardless of the origin of immunizing and target thymocytes, the assay employed detected exclusively PFC-producing antibodies to the Thy-1.1 antigen. In almost all instances,H-2-compatible thymocytes elicited a significantly higher response than didH-2-incompatible thymocytes, although the latter occasionally evoked a high response. TheH-2 incompatibility between donor and recipient appeared to be responsible for the differences in responsiveness of the standard inbred mice and theirH-2 mutants immunized with thymocytes compatible with standard inbred strains. The phenomenon observed appears to have several features in common with antigenic competition. We propose that the requirement forH-2 compatibility in the anti-Thy-1.1 response may be the expression of a general requirement of T cells to recognize an antigen in the context of the H-2 molecule.  相似文献

The primary response to Thy-1.1 antigen was measured by a plaque assay that detected cells producing antibodies lytic for AKR thymocytes (PFC). TheH-2 congenic mice (B10.K and B10.BR) carryingH-2 complexes of high responders (CBA and C57BR) on the low-responder background (B10) were found to produce significantly fewer PFC than the corresponding donor of theH-2 complex. On the other hand, C3H.B10 mice carrying theH-2 complex of a low responder on the high-responder background produced significantly more PFC than the donor of theH-2 complex. These findings were interpreted as evidence that alleles at previously described loci believed to be components of theI region of theH-2 complex and controlling immune response to Thy-1.1 are influenced by alleles at another locus. Studies of segregating populations of theH-2 congenic lines supplied evidence that this locus, tentatively calledIr-5, is in chromosome 17 (linkage group IX).  相似文献

We describe a new murine cell-surface alloantigen, provisionally designated Thy-2, which is expressed primarily on thymocytes and brain tissue. Although Thy-2 is also expressed at lower levels on bone-marrow and spleen cells, this antigen does not appear to be present on lymph-node, liver, or red blood cells. Immunoprecipitation of surface-labeled thymocyte extracts from a variety of inbred strains reveals this antigen to be a single polypeptide of 150 000 daltons. Quantitative membrane immunofluorescence demonstrates that Thy-2 is a minor cell-surface component which is present on the majority of thymocytes. Mice heterozygous at theThy-2 locus express approximately 50 percent as much antigen as positive homozygotes. Expression of the Thy-2 alloantigen is controlled by a single semidominant gene located approximately 3 cM to the right of theH-2K locus on chromosome 17.  相似文献

The Thy-1 glycoprotein is a differentiation antigen which exhibits tissue-specific regulation. A mutant of a Thy-1.1+ T-cell lymphoma has been isolated which does not express Thy-1 glycoprotein on the cell surface and does not accumulate Thy-1 mRNA in the cytoplasm. Hybrids between the mutant and a Thy-1.2+ T-cell lymphoma express 20–30-fold lower levels of Thy-1 glycoprotein on their cell surface compared to wild-type T-cell lymphomas, and they have correspondingly low levels of cytoplasmic Thy-1 mRNA. A revertant of one hybrid was isolated which expressed wild-type levels of both Thy-1 alleles on its surface and contained correspondingly increased levels of Thy-1 mRNA. A Thy-1+ revertant of the Thy-1 mutant was isolated by cell sorting. A second generation Thy-1 mutant could be isolated from this revertant which also did not accumulate Thy-1 mRNA and which behaved in a way similar to the first generation mutant when hybridized to a Thy-1.2+ lymphoma. No changes in the structure or copy number of the Thy-1 structural gene could be detected in this series of mutants and revertants. These properties are consistent with a mutation in one (or more) gene(s) which acts in trans position to regulate Thy-1 glycoprotein expression.  相似文献

Early and late primary IgM antibody responses of mice to Thy-1.1 antigens showed different antigenic and cellular requirements. We studied genetic controls of the early primary responses, which could be induced by subcellular thymocyte antigens independently of host T-cell activity. All Thy-1.2 mouse strains of Igh a(BALB/c and BC8), Igh-V aCb(BAB14), Igh d(AKR/Cum), Igh j(CBA/J, C3H/HeN, C3H.SW, and C3H.JK), and Igh n(NZB) definitely responded early to Thy-1.1 antigens from AKR/J (Igh d), A.Thy-1.1 (Igh e), or B10.Thy-1.1 (Igh b) mice or SD rats, whereas all strains of Igh b(C57BL/6, C57BL/10, B10.D2, B10.BR, B10.A, CB20 and CWB), Igh c(DBA/2), Igh e(A/J), and Igh o(C.AL20) responded poorly to the same antigens. This contrasts with the observation that both strains of Igh j(C3H/HeN) and Igh b(B10.BR) responded well at later times. As was the case for late responses, the matching of H-2 between donor and recipient resulted in early responses of exceptional quality in high-responder strains. It was concluded that under the influence of H-2, whose incompatibility between donor and recipient partially interferes with responses, early but not late primary Thy-1.1-specific antibody responses are selectively controlled by Igh-V or closely linked Ir gene(s) as a new V Hmarker.Abbreviations used in this paper Tl T cell-independent - TD T cell-dependent - PFC plaque-forming cell(s) - Igh immunoglobulin heavy chain - V H variable region of heavy chain - C H constant region of heavy chain  相似文献

We have introduced a mouse Thy-1.1 gene into the germline of Thy-1.2 mice. The introduced gene was shown to be expressed at very high levels in thymocytes when compared with the endogenous gene. Transgenic thymocytes were shown to evoke a higher than normal primary anti-Thy-1.1 antibody response in plaque-forming cell (PFC) assays. This result suggests that a direct quantitative interaction of the Thy-1 antigen activates the B cell response.  相似文献

B10.129(6M) mice carrying theH-2 complex of an intermediate responder (129) on a low-responder (B10) background, were low responders. Studies employing different F1 hybrids and segregating generations of 129/J and 6M mice indicated that differences in responsiveness of those two strains depend on alleles at a single locus, loosely linked to theH-2 complex. These results lend further support to the previously advanced concept that the expression of theIr-Thy-1 allcles controlling the response to the Thy-1.1 antigen is influenced by the alleles at theIr-5 locus. In addition, studies employing F1 hybrids produced through matings of 129/J, 6M, C3H.B10 and C57BL/6J mice to a panel of inbred strains suggested that in regard to the responsiveness to the Thy-1.1 antigen, 129/J and 6M mice are phenotypically, and presumably genotypically, similar to C3H.B10 and C57BL/6J mice, respectively.  相似文献

Cell viability and the production of direct PFC were studied in mouse spleen cell cultures after cortisol treatment in vivo or in vitro at various times relative to primary stimulation with SRBC in vitro.Cortisol treatment in vivo reduced spleen cell numbers by 88% after 48 hr, but cultures of the remaining cells produced as many PFC in vitro as did cultures of equal numbers of normal spleen cells.In normal spleen cell cultures incubated with cortisol for 4 hr prior to the addition of antigen, peak responses of PFC/culture and PFC/106 cells occurred 24 hr later than in controls and averaged, respectively, 27% and 141% of control values. Minimum viable cell numbers were observed in cortisol-treated cultures after 3 days; thereafter cell numbers gradually increased. These results were not significantly altered when cultures were treated simultaneously with cortisol and antigen.The response was not suppressed if the addition of antigen preceded that of cortisol by more than 4 hr. Suppression was also considerably reduced if fetal calf serum was used when preparing cells for culture.  相似文献

Viral-induced interferon inhibition of the primary in vitro plaque-formong cell (PFC) response in the mouse (C57B1/6) involves a dynamic relationship between the nature of the antigen, the concentration of interferon added to antigen-stimulated cultures, and the time of addition of interferon relative to antigen addition. The PFC response to a thymus-dependent antigen (sheep red blood cells) was more easily suppressed by viral-induced interferon than was that to a thymus-independent antigen (E. coli 0127 LPS), both in terms of inhibitory concentrations of interferon and the time at which the interferon could be added to cultures after antigen and still inhibit the PFC response. These differential effects of interferon could be related to the difference in cellular requirements (B and T lymphocytes) of the two antigens. Interferon was effective in inhibiting the in vitro PFC response of antigen-primed spleen cells, indicating that it can block the response of memory lymphocytes. By using interferon inducers as inhibitors of the in vitro PFC response, it was possible to show that at least two antigenically distinct interferons may be involved in suppressing the immune response. It is known that one type of interferon is induced by virus and synthetic double-stranded polyribonucleotides. The other type is stimulated by antigen and T cell mitogens. A model is proposed to explain the nature of these interferon inhibitory effects in terms of mediation of immune suppressor cell effects.  相似文献

Mouse bone marrow is barely capable of plaque-forming cell (PFC) activity during the primary response to sheep red blood cells (SRBC). However, during secondary-type responses, it becomes the major organ, containing IgM, IgG, and IgA PFC. In the present paper, the influence of splenectomy (Sx) upon the secondary bone marrow PFC response to SRBC was investigated. When previously primed mice were splenectomized just before the second intravenous (iv) injection of SRBC, the effect of Sx upon the height of the bone marrow PFC response was dependent on the booster dose. Sx just before a booster of 106 SRBC iv almost completely prevented bone marrow PFC activity, whereas an iv booster dose of 4 × 108 SRBC evoked a normal IgM, IgG, and IgA PFC response in Sx mice. Apparently low doses of iv administered antigen require the spleen in order to evoke antibody formation in the bone marrow. Experiments with parabiotic mice, consisting of Sx and sham-Sx mice, showed that this facilitating influence of the spleen upon bone marrow antibody formation occurs via the blood stream. In a subsequent study, it was investigated whether the spleen is required throughout the bone marrow PFC response or only during the few days of the initiation phase. Therefore, mice were splenectomized at different intervals after a booster injection of 106 SRBC iv. It appeared that Sx 2 days after the booster injection could still prevent the normal bone marrow PFC activity, whereas Sx at Day 4 could no longer do so. Apparently, after an iv booster injection, the spleen is only required for initiation of the bone marrow PFC response and not for the maintenance of this PFC activity thereafter.  相似文献

The conditions for the in vitro generation of primary and secondary immune responses by rabbit spleen cells to sheep red blood cell (SRBC) antigen have been examined. Spleen cells from many normal and all previously immunized rabbits are capable of producing in vitro plaque-forming cell (PFC) responses when cultured as dissociated cell suspensions in the presence of antigen. Primed spleen cells generate approximately 100 times the number of PFCs obtained in normal cultures with a shorter lag period. Both types of cultures demonstrate a period of exponential increase in PFCs during which the doubling time is 12–14 hr. This increase occurs after 1 day of culture of spleen cells from primed rabbits and after 4 days of culture of spleen cells from unprimed rabbits. The PFCs which arise in cultures of primed cells appear not to be the progeny of those generated in vivo but to be derived from an increased number of PFC precursors. Repeated immunization of the spleen cell donor is required to produce significant numbers of indirect (IgG) PFC or indirect precursors; most of the PFC found after a single immunization in vivo or in vitro are direct (IgM). There is no evidence for conversion of IgM to IgG PFC in vitro. This system should provide a means for further identification of the cellular interactions involved in the immune response of the rabbit.  相似文献

Spleen cells of inbred mice strains carrying θ-C3H allele have been cultured in the presence of AKR thymus cells and their in vitro primary PFC response against thymic alloantigen θ-AKR was studied.The responses of a magnitude which was comparable with that obtained in previous in vivo experiments were obtained 4 days after stimulation. The strain-dependent variability of the magitude of anti-θ-AKR responses was observed in vitro. RR and C58/J spleen cells produced much more PFC than C57BL/6J and DBA/2J spleen cells. This was in agreement with previous in vivo studies on the genetic control of the anti-θ AKR responses.In the absence of AKR thymus cells, spleen cells of high responders, RR, developed in vitro PFC which released antibodies lytic to AKR thymus cells. Their number was ten-times lower than in stimulated cultures. Spleen cells of all strains tested produced also small numbers of PFC secreting antibodies against θ-identical allogenic thymus cells and even to syngenic thymus cells.  相似文献

The capacity for humoral immune response was evaluated in C57BL/6 mice fed diets with low (8%) or normal (27%) protein content upon primary and secondary stimulation with allogeneic cells from the DBA/2 strain. Primary antibody response was assessed by titration of serum hemagglutinins and by quantitation of direct plaque forming cells (PFC) in immune spleen suspensions, with lymphoma cells L5178Y of the DBA/2 strain as target. Secondary antibody response was assessed by titration of serum hemagglutinins. The following results were obtained: 1) Significant decrease in the total number of spleen cells was observed in protein deficient animals while the numbers of IgM PFC/spleen did show small reduction. 2) The number of direct alloantibody PFC/107 spleen cells was increased in the protein deficient animals in comparison to the normally fed controls. 3) The above effect was observed even after short periods (1 week) of protein depletion. 4) Titers of serum hemagglutinins in protein deficient mice were similar or higher than in normal mice during the primary response but markedly depressed during the secondary response. 5) The synthesis of IgG hemagglutinins was depressed in protein deficient mice during both the primary and secondary responses. The results indicated that cells producing IgM alloantibodies are not affected by protein deficiency starting during the fourth week of age, while one or more of the cell populations interacting in the IgG response to alloantigens is markedly depressed by similar protein restriction. It was suggested also that protein deficient animals present a failure in the regulatory mechanism(s) of the IgM response to alloantigens.  相似文献

To investigate whether the antibody response and T-B-cell collaboration in vitro can be obtained in the absence of mitogens, a method of obtaining an in vitro primary anti-sheep red blood cell antibody response by rabbit spleen and lymph node cells was developed. We used Marbrook culture vessels and a specially prepared medium containing 10% autologous serum and maintained at pH 7.4–7.6. The system was shown to be devoid of any polyclonal mitogens as assessed by [3H]thymidine incorporation and by direct examination for blast cells in stained smears. The primary response increased continuously over the 5-day cultivation period and only IgM but not IgG plaque-forming cells (PFC) were detected. In over 20 experiments, the response ranged from 357 ± 17 to 4425 ± 110 PFC/107 cultured cells with a median stimulation index of 52. The spleen cells required less antigen than the lymph node cells and 2-mercaptoethanol inhibited the response of the spleen cells but not that of the lymph node cells. Lymphocytes were separated into highly pure T- and B-cell populations by negative selection using antibody-coated human erythrocytes to rosette either T or B cells and Ficoll-Hypaque centrifugation to remove rosetted cells. Upon cultivation, B cells alone gave a low IgM response, whereas B cells reconstituted with T cells gave a response similar to that obtained with unseparated lymphoid cells. We concluded that: (a) optimal conditions for obtaining primary in vitro antibody responses using rabbit spleen and lymph node cells were established, (b) T-B-cell collaboration was demonstrated in the rabbit primary antibody response to sheep erythrocytes, and (c) the primary antibody response in vitro and T-B-cell collaboration may occur in the absence of detectable polyclonal mitogens.  相似文献

The incidence of infection by Actinobacillus actinomycetemcomitans, one of the important pathogens in human periodontal diseases, has been reported to be associated with racial background and genetic factors. We attempted to determine the genetic regulation of immune responses to A. actinomycetemcomitans fimbriae, an attachment factor, using various inbred strains of mice. For this purpose, we synthesized an oligopeptide antigen using the amino acid sequence of the fimbriae and conjugated this antigen to branched lysine polymer resin beads. After immunization with the synthetic A. actinomycetemcomitans fimbrial antigen, serum antibody levels and the delayed-type hypersensitivity (DTH) reaction to the antigen were measured by enzyme-linked immunosorbent assay (ELISA) and footpad swelling responses, respectively. The strains of mice found to be high-IgG responders to the antigen were B10.HTT, B10.RIII, B10A (5R) and B10.S (9R). These results indicate that mice with Eβs: Eαk, Eβr: Eαr and Eβb: Eαk respond strongly to the synthetic peptide. All of the high-IgG responders showed a high DTH response. A cell transfer experiment confirmed that CD4 T cells mediated with a DTH response to the synthetic peptide. Thus, the results of this study demonstrate that the immune responses to A. actinomycetemcomitans fimbriae are genetically controlled.  相似文献

BALBc mice immunized with Type III pneumococcal polysaccharide (SIII) had higher numbers of IgM plaque-forming cells (PFC) in the spleen than similarly immunized C57BL/Ks mice. The F1 hybrids of these two strains had intermediate numbers of SIII-specific PFC. Analysis of the responses of F2 and backcross strains indicated that the observed responses were compatible with results expected for control of the immune response to SIII at a single autosomal locus.  相似文献

The immune responses of allogeneic mixed spleen cell cultures (MLC) to the T-dependent antigen, SRBC, and to the T-independent antigen, DNP-PAA, were investigated. The immune response to DNP-PAA in MLC with certain strain combinations was always suppressed as compared with the expected PFC response calculated from the PFC responses of the individual strains. This suppression was eliminated by treating the spleen cells with RAMB antiserum plus complement before the incubation of the MLC with DNP-PAA. It can be concluded that the suppression in the PFC response to the T-independent antigen DNP-PAA in MLC is due to the generation of suppressor T-cells. The PFC response to the T-dependent antigen, SRBC, in MLC showed either suppression, no change, or rarely augmenation, suggesting that the allogeneic mixed spleen cell cultures can generate both suppressor and helper T cells and that the balance between helper and suppressor activity regulates the PFC response to a T-dependent antigen. Suppressor activity was also generated in a one-way MLC, but the degree of suppression depended upon which of the two strains was responding. Similar amounts of thymidine were incorporated in the one-way MLR irrespective of which strains was responding. Thus, the extent of proliferation in one-way MLR is not related to the degree of suppressor activity generated. The results further indicate that a difference between two strains in the I-C, S, and G regions of the major histocompatibility complex is required to generate suppressor activitiy that can depress the response to a T-independent antigen, MLC between strains differing in K, I-A, I-B, I-J, I-E, and D regions generate little or no suppressor activity in this system.  相似文献

The ontogeny of antibody responses to trinitrophenylated (TNP) thymus-independent (TI) antigens was compared in two partially inbred strains of chicken: the SC strain (B2/B2 genotype) and the FP strain (B15/B22 genotype). In the SC chicken, maturation of both the splenic anti-TNP plaque-forming cell (PFC) response and the 19S hemagglutinating antibody response to TI type 2 (TI-2) antigens, TNP-Ficoll and TNP-dextran, were delayed to a significantly later time in ontogeny (20 wk of age) than in the FP chickens (9 wk of age). Four- to 6-wk-old SC chickens were virtually immunologically unresponsive to stimulation with TI-2 antigens. The TI-1 antigen TNP-Brucella abortus was equally immunogenic in both FP and SC chickens of different age groups tested. Kinetic studies of the primary PFC response to TNP-Ficoll in immunologically mature chickens of the SC and FP strains demonstrated a peak PFC response 4 days after antigen injection, followed by a rapid decline in numbers of splenic PFC/spleen on day 6. The results of these studies are discussed in relation to earlier observations that suggested there may be a delay or a defect in the ontogeny of the thymus in the SC chicken.  相似文献

We have already developed nine B10.MOL congenic strains carrying H-2 haplotypes derived from Japanese wild mice, Mus musculus molossinus, with the C57BL/10 genetic background. To obtain monoclonal antibodies against the H-2 antigen of the Japanese wild mouse, we carried out cell fusion using spleen cells from the animal immunized with one of the B10.MOL strains, B10.MOL-SGR (H-2 wm7). As a result, 19 hybridomas producing monoclonal antibodies were produced. Analysis with the intro-H-2 recombinants derived from B10.MOL-SGR indicated that 8 of them reacted with the class I and II with the class II molecule. The class I antibodies were tested for their cross -reactivities on wild mice and on the panels of standard inbred and B10.MOL strains. Most of the antibodies reacted with both the Japanese wild mice and the other subspecies, including standard inbred, while two antibodies highly specific for the donor H-2K region reacted with only three wild-derived mice, two M. m. molossinus from Anj o and Shizuoka, Japan, and one M. m. domesticus from Pigeon, Canada. In addition, all of the other four antibodies reactive with the K antigen of B10.MOL-SGR also reacted with the same three wild mice. The wild mice belonging to different subspecies might share very similar H-2K antigenic determinants in spite of their genetic and geographical remoteness.  相似文献

In the present series of experiments we have studied the effects of anti-lymphocyte serum (ALS) and concanavlin A (Con A) on the immune response to technetium-99m-labeled sheep erythrocytes (SRBC) and have related this to the localization and persistence of antigen at the site of induction and antibody synthesis. The number of 99mTc-labeled SRBC in the spleen and liver was quantified by gamma scintillation counting and the cellular kinetics of the splenic antibody response was determined by means of the hemolytic plaque technique. After injection of normal rabbit serum (NRS)-treated control mice with 4 × 10899mTc-labeled SRBC, the number of cells localizing in the spleen ranged from a high of 4.2 × 106 on Day 1 to a low of 1.7 × 106 on Day 4, while the number in the liver ranged from a high of 68.8 × 108 on Day 1 to 18.6 × 106 on Day 4. The number of splenic plaque-forming cells (PFC) increased from 321–429 on Day 1 to 93,000–101,000 PFC on Day 4 and this was paralled by a rise in serum hemagglutinin and hemolysin titers. In mice treated with ALS on the other hand, splenic localization initially was increased 10-fold, hepatic localization was unchanged, and the antibody response was markedly suppressed. Splenic PFC ranged from approximately 100 between days 1 and 3 and increased to only 500 on Day 4. Mice which received Con A on Day — 1 had a reduction in splenic PFC which ranged from 150 on Day 1 to 1900 on Day 4. Splenic localization of 99mTc-labeled RBC initially was three- to fourfold greater than that in NRS-treated mice and then decreased to control levels. The increased numbers of SRBC detected in the spleens of immunosuppressed mice at the time of peak response can be attributed to decreased in vivo lysis by reduced numbers of splenic antibody-producing cells.  相似文献
