Some of the most productive plants on the planet use a variant of photosynthesis known as the C(4) pathway. This photosynthetic mechanism uses a biochemical pump to concentrate CO(2) to levels up to 10-fold atmospheric in specialized cells of the leaf where Rubisco, the primary enzyme of C(3) photosynthesis, is located. The basic biochemical pathways underlying this process, discovered more than 40 years ago, have been extensively studied and, based on these pathways, C(4) plants have been subdivided into two broad groups according to the species of C(4) acid produced in the mesophyll cells and into three groups according to the enzyme used to decarboxylate C(4) acids in the bundle sheath to release CO(2). Recent molecular, biochemical, and physiological data indicate that these three decarboxylation types may not be rigidly genetically determined, that the possibility of flexibility between the pathways exists and that this may potentially be both developmentally and environmentally controlled. This evidence is synthesized here and the implications for C(4) engineering discussed.  相似文献

Summary The -cells of the pancreatic islets have been shown to contain -aminobutyric acid (GABA) together with insulin. Autoradiographic analysis indicated that high affinity GABA binding sites (GABA receptors) are not present in the pancreas. High affinity GABA uptake sites are present, not in -cells, but in a few cells on the periphery of the islets. These observations cast doubt on the suggestion that GABA has a paracrine role in the pancreas.  相似文献

Using an immunoblotting technique and goat antihuman C4, we observed five distinct electrophoretic variants of C4 in a panel of 60 random dogs. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of immunoprecipitated C4 showed that dog C4 is composed of three polypeptide subunit chains (, , and ) and that structural variability occurs within the - and -chain regions. Two distinct molecular weight forms of both the C4- ( A and B) and C4-( A and B) chain were detected. The variant forms of C4 and C4 were found in association with particular C4 allotypes.  相似文献

Background: The COBAS AMPLICOR (CA) instrument for the amplification and detection steps of the AMPLICOR molecular diagnostic assays has recently been introduced. The system contains a single thermal cycler with two independently controlled heating/cooling blocks, a pipettor, a magnetic particle washer, a photometer and an incubator.Objective: The performance of the CA instrument was evaluated in a routine diagnostic laboratory for the detection of hepatitis C virus (HCV) RNA. The new system was compared with the corresponding microwell plate assay (AMPLICOR HCV Test).Study design: Routine clinical sera (350) from hemodialysis patients and patients with chronic HCV infection and interferon therapy were studied. If discrepant results were obtained, both assays were repeated (specimen preparation, amplification and detection); in addition, the HCV copy number was determined with the AMPLICOR HCV MONITOR Test.Results: There was a correlation between the CA HCV Test and the AMPLICOR HCV Test in 341 of 350 specimens (97%). After resolution of 9 discrepant results, the CA HCV Test gave a sensitivity of 97.8% and a specificity of 99.4%. The most common reason for discrepant results was a low HCV RNA copy number.Conclusion: The CA system was found to be a labor-saving, fast and reliable instrument for the amplification and detection steps of a RT-PCR molecular assay for detection of HCV RNA.  相似文献

A previous study on the genetics of mouse C4-binding protein (C4-bp) indicated the existence of a genetic polymorphism. Two genetic variants were reported and their segregation used to determine the mapping position of the C4BP locus to the H-2D-Qa interval of the mouse H-2 system. We show here, however, that purified C4-bp does not display the previously reported polymorphism. The mapping position of C4BP in the mouse therefore remains undetermined.  相似文献

An anatomical, histological and ultrastructural study was made of the reproductive system and spermatogenesis of Iheringichthys labrosus. The testis are digitiform and consist of a sperm atogenic cranial region, a spermatogenic/secretory medial (transition) region, and a strictly secretory caudal region. The cranial region represents 66% of the total length of the maturing testis and its fringes or lobes have a length of 5.59 + 0.73 mm. The medial and caudal regions represent each 17% of the testicular length and their fringes have a length of 5.37 +/- 0.69 mm and 3.12 +/- 0.38 mm, respectively. Histologically, the cranial region of the testis is made up of seminiferous tubules with spermatogenic cells contained in cysts. These cells undergo synchronous development, inside the cysts where spermatogenesis is completed. The secretory caudal region does not constitute an individualized gland. Ultrastructurally, its secretory cells have a vesiculous nucleus and a cytoplasm with abundant dilated cisternae of rough endoplasmic reticulum. The caudal region produces a glycoproteic secretion and exhibits variable electron density during maturation. During the resting period, these cells are poor in synthesis organelles. The spermatozoa are of the primitive type, with a round head (1.56 +/- 0.11 microm), a rudimentary middle piece, and a long flagellum with a 9 + 2 axonemal arrangement.  相似文献

The modularity and versatility of an engineered functional reversal of the β-oxidation cycle make it a promising platform for the synthesis of longer-chain (C≥4) products. While the pathway has recently been exploited for the production of n-alcohols and carboxylic acids, fully capitalizing on its potential for the synthesis of a diverse set of product families requires a system-level assessment of its biosynthetic capabilities. To this end, we utilized a genome scale model of Escherichia coli, in combination with Flux Balance Analysis and Flux Variability Analysis, to determine the key characteristics and constraints of this pathway for the production of a variety of product families under fermentative conditions. This analysis revealed that the production of n-alcohols, alkanes, and fatty acids of lengths C3–C18 could be coupled to cell growth in a strain lacking native fermentative pathways, a characteristic enabling product synthesis at maximum rates, titers, and yields. While energetic and redox constraints limit the production of target compounds from alternative platforms such as the fatty acid biosynthesis and α-ketoacid pathways, the metabolic efficiency of a β-oxidation reversal allows the production of a wide range of products of varying length and functionality. The versatility of this platform was investigated through the simulation of various termination pathways for product synthesis along with the use of different priming molecules, demonstrating its potential for the efficient synthesis of a wide variety of functionalized compounds. Overall, specific metabolic manipulations suggested by this systems-level analysis include deletion of native fermentation pathways, the choice of priming molecules and specific routes for their synthesis, proper choice of termination enzymes, control of flux partitioning at the pyruvate node and the pentose phosphate pathway, and the use of an NADH-dependent trans-enoyl-CoA reductase instead of a ferredoxin-dependent enzyme.  相似文献

Summary The nervous system (NS) of Bothriomolus balticus (Proseriata) was studied by the immunocytochemical (ICC) method with antisera to RFamide, SALMFamide and serotonin and with the histochemical GAIF method. The use of the ICC technique provided a much more precise morphological account of the nervous system than had previously been possible. The obtained data are discussed in connection with the comparative morphology of the nerve cords of the Plathelminthes. A similar position does not grant direct correspondence between nerve cords in the taxon Seriata. Marginal cords had probably an independent origin in the Monocelididae and Otoplanidae. The ventral (main) cords of B. balticus seem to correspond to the lateral (main) cords of the Monocelididae. It can be hypothesized that both: (1) a shift of the main cords accompanied by formation of new cords from the plexus and fusion of other cords and (2) a redistribution of nerve processes and perikarya between the cords, take part in the evolution of cords in the Plathelminthes. The first hypothesis seems to explain the difference in the position of main cords in proseriates, though, the second hypothesis might dominate, for example, in the Neorhabdocoela and the Neodermata. The correctness of the evolutionary analysis of the nerve cords in plathelminths can only be provided by neurons or neuron groups marking these structures.  相似文献

Morphological details of the testis, seminal vesicles and vas deferens of Orchestia platensis are described. The follicular lumen of the mature testis contains spermatogonia, spermatocytes, spermatids and spermatozoa. The histochemical nature of the testis and the vas deferens is elucidated. The spermatozoa and vas deferens contain acid sulphated mucopolysaccharides and neutral mucopolysaccharides. In addition, they contain basic proteins, disulphide groups, lipids, phospholipids, RNA and DNA.  相似文献

Summary The cellular localization of biogenic monoamines in crustaceans was studied by means of a highly specific and sensitive fluorescence method devised by Falck and Hillarp. It was found that neurons displaying specific fluorescence in the central nervous system were confined to the protocerebrum, the medulla externa and interna and the ventral nerve cord. The method allows a distinction between the fluorophores of 5-hydroxytryptamine (and 5-hydroxytryptophan), which emit the yellow light, and the fluorophores deriving from the catecholamines (and DOPA), which emit the green light. Green-fluorescent neurons occurred abundantly in the aforementioned parts of the central nervous system while yellow-fluorescent neurons were sparsely present in the same parts.The present work has been carried out at the departments of Histology and Zoology at the University of Lund. Summary The intergeneric amphiploid Hordeum chilense × Aegilops squarrosa has been synthesized. The amphiploid plants have the expected chromosome number of 28. The average meiotic chromosome pairing was 12.48 bivalents + 3.04 univalents. The morphology of the amphiploid resembles that of the Aegilops parent. Nucleoli from both H. chilense and A. squarrosa are expressed in the amphiploid. Neither chromosome instability nor homoeologous pairing was found. The amphiploid is fertile and vigorous.  相似文献

At two sites, one with a 4-year-old (4-Y) secondary vegetation and the other with a 20-year-old (20-Y) vegetation, the influence of burning slashed vegetation on crop performance was studied during three seasons. In the first season after clearing, also the influence on weed growth was studied. At both sites, burning significantly decreased the number of weed seedlings. The lowest number of seedlings was found on the burnt plots of the 20-Y site. Burning increased yield and nutrient uptake significantly in the first and second season after clearing. In the third season after burning, only at the 4-Y site a significantly higher yield and nutrient uptake were found. At the 20-Y site the effect had disappeared. Calculations of efficiency of utilization of absorbed N, P and K indicated that P was the least available nutrient, also after burning. At both sites three consecutive crops absorbed approximately 40% of P applied in ash, while the cumulative recovery of K was at least 36% at the 4-Y site and at least 59% at the 20-Y site. On non-burnt plots, yields were not lower in the third season than in the first season after clearing, thus indicating that the inherent soil fertility did not decrease. Hence, yield decline on the burnt plots could be ascribed to ash depletion. It was concluded that in the local shifting cultivation system, the combination of ash depletion and infestation of weeds are the main reasons for abandoning the fields.  相似文献

One of the characteristics of all somatic cells is a finite life span. Cells may proliferate until they reach a point after which, although they are metabolically active, they can no longer produce daughter cells. This observation is central to the clonal exhaustion hypothesis, a mechanism cited to explain age-associated immune dysfunction. In this hypothesis, repeated division of lymphocytes leads to a replicative limit, after which they enter the senescent phase but are not lost from the pool of T cells. Advancing age would then be associated with an increase in the number of T cells that are unable to proliferate to a stimulus which induces a proliferative response in T cells from younger individuals. This hypothesis seems both logical and reasonable and is supported by data from both humans and mice with the demonstration of an age-related accumulation of senescent T cells in both species. However, there is an apparent paradox. The paradox arises because the onset of immunosenescence appears to be more closely linked to the life span of the animal rather than the life span of the lymphocyte. Immunohistochemical tests were applied to sections of intestine of uninfected and Pomphorhynchus laevis Muller-infected chub, Leuciscus cephalus (L.) using 15 different antisera. Nerve cell bodies and fibres immunoreactive (IR) to the anti-bombesin, -Cholecystokinin-8 (CCK-8), -galanin, -Gastrin-Releasing Peptide (-GRP), -Nitric Oxide Synthase (-NOS), -Substance P (-SP), and -Vasoactive Intestinal Peptide (-VIP) sera were observed in the myenteric plexus of uninfected chub. The density of nerve components immunoreactive to these antisera was high in the intestine of the infected fish, especially near the site of attachment. Moreover, numerous nerve fibres, immunoreactive to anti-bombesin, -GRP, -galanin, -SP, and -VIP sera, were encountered in the connective tissue capsule surrounding the bulb and proboscis of P. laevis. The occurrence of P. laevis in the chub gut significantly increased the number of endocrine cells per intestinal fold immunoreactive to galanin, met-enkephalin and leu-enkephalin antisera. CCK-8, Neuropeptide Y and glucagon-like immunoreactive cells were less numerous in the intestine of infected chub. A large number of cells in the tunica propria-submucosa of L. cephalus infected with P. laevis were immunoreactive to anti-serotonin and -leu-enkephalin sera.  相似文献
