In glasshouse pot experiments, uptake of benomyl, thiophanate-methyl and carbendazim from equivalent soil applications (in the range 0–003– 0–035 %a-i- atarateof 2&4 ml/plant) gave equal levels of control of Botrytis lesions developing from inoculations of freshly exposed leaf scars on tomato stems. Spray applications of benomyl to exposed leaf scars controlled infection at concentrations down to 0025 % a.i. The effect of lower concentrations of the stem spray could be markedly enhanced either by the addition of a mineral oil (2 % Actipron) or by a prior soil application of benomyl at a low rate which on its own had little effect on lesion development. Protectant spray applications of glycophene and vinclozolin gave levels of control quite comparable to that of benomyl at equivalent concentrations. Evidence was obtained that the lesions formed at the artificially-inoculated leaf scars at the top of the stems of young pot-grown tomato plants were larger than those lower down. In spite of this, the level of disease control with soil applications of fungicides containing or generating carbendazim (MBC) was greater at the top than at the bottom, probably because of the normal migration of the fungicides and their accumulation at the extremities of the plant. In an observation trial in a commercial crop of tomatoes, benomyl applied either as five soil drenches at approximately monthly intervals, or as two drenches followed by five sprays at three-weekly intervals, or as five sprays alone gave marked reductions in plant loss and number of Botrytis stem lesions in both cvs Eurocross BB and Cudlow Cross. Those stem lesions which did develop, however, were generally as large as those on untreated control plants. Five sprays of dichlofiuanid gave similar levels of disease control. All the treatments gave apparently higher yields (statistically untested) in Eurocross BB, but less consistent responses were recorded in Cudlow Cross.  相似文献

Chestnut blight cankers, caused by the fungus Cryphonectria parasitica, are prone to invasion by other microorganisms as the canker ages. This microbial community has the potential to alter canker expansion, which may influence the probability that the canker girdles the infected stem. Hypoviruses infect the pathogen mycelium directly and are known to decrease pathogen virulence (i.e. hypovirulent). These viral infections can slow pathogen growth, decreasing the rate of canker expansion and lowering the probability of girdling. Saprophytic fungi also invade the expanding canker and may antagonize C. parasitica leading to reduced pathogen growth. The combined effects of fungal antagonism and a hypovirulent pathogen could work in combination to reduce the probability of girdling the infected stem. We assessed the ability of different fungal taxa, isolated from low severity cankers, to inhibit the growth of virulent and hypovirulent forms of C. parasitica in dual culture tests on two cultural media. Percent growth inhibition of virulent C. parasitica by potentially antagonistic fungi ranged from 2 % to 34 %, while inhibition of hypovirulent C. parasitica ranged from 18 % to 54 %. Only one isolate, identified as Umbelopsis isabellina (UmbelopsisWS) inhibited the virulent form of the pathogen more than the hypovirulent form. All three Trichoderma isolates caused the greatest growth inhibition of virulent C. parasitica, but they, like all other fungal isolates tested, inhibited the hypovirulent form of the pathogen more than the virulent form. These results suggest that commonly occurring fungi in chestnut blight cankers, including Trichoderma, may inhibit the hypovirulent C. parasitica more than virulent C. parasitica. Thus, the presence of other fungi in cankers may not enhance the effect of hypovirulent C. parasitica to delay cankers from girdling a stem but instead intensify canker development.  相似文献

Seeding parsnip plants on four commercial holdings in Essex showed not only the root cankers and leaf spots associated with infection by Itersonilia pastinacae, but also extensive lesions on the petioles and necrosis of the inflorescences. These last-named symptoms were proved to be caused by I. pastinacae which could also be found on some of the seeds. Seed infection, though largely superficial, was sometimes more deep-seated. It could be eliminated by soaking the seed in an aqueous suspension of 0·2% thiram at 30 °C for 24 h. Heavily infected seed gave a low percentage of seedlings bearing cotyledon lesions: it is uncertain whether hypocotyl lesions, which also occurred, were caused directly by the fungus. An outdoor test failed to show that infected seed gave rise ultimately to roots bearing Itersonilia cankers: the significance of this and other possible sources of infection is discussed.  相似文献

The atypical symptoms first described by Bryan (1932) of the angular leaf spot disease of cotton caused by Xanthomonas malvacearum (E. F. Sm.) Dowson were reproduced by inoculation into seeds, stem apices or buds. The lesions that developed on the veins of the newly produced leaves were elongated and water-soaked, becoming dark brown. The leaf tissue dependent upon infected veins became yellow, flaccid and withered. The development of these symptoms was enhanced when inoculations were made into opening buds or germinating seeds as compared with inoculations into closed buds or dormant seeds.
In other bacterial diseases caused by Xanthomonas spp., somewhat atypical symptoms could also be produced by bud inoculation into the appropriate host. Those produced by X. ricini (Yoshi & Takimato) Dowson on castor, closely resembled the vein lesions described above on cotton but resulted only from bud inoculations; inoculations into stem apices and seeds failed to produce them. In dolichos bean inoculated with X. phaseoli (Smith) Dowson, atypical symptoms were produced only by seed inoculations and were confined to the first simple leaves (prophylls).
The differences in the production of atypical symptoms on the three hosts are correlated with differences in host structure and with the degree of virulence of the pathogen. The leaf parasite X. ricini , for example, which cannot infect castor bean stems, does not produce atypical symptoms when inoculated below the stem apex.
From the data discussed below, the incidence of atypical symptoms is attributed to infection either of an actively growing tissue or of a telescoped structure which subsequently completes its development.
The atypical symptoms of the cotton disease are not caused by a special strain of X. malvacearum. Further, they are not a peculiarity of this disease but may also develop in other necrotic diseases under similar conditions.  相似文献

Experiments have shown that pigeons, Columba livia, are able to develop navigational abilities even if reared and kept confined in an aviary, provided that they are exposed to the natural winds. In contrast, pigeons reared in a wind-screened aviary do not learn to navigate. Pigeons kept confined in a screened aviary when young do not learn to navigate even if, once they become adult, the screens are removed and the birds are exposed to natural winds for a period sufficiently long for map learning. In all of these experiments, pigeons were kept confined until the day of test release. In the present study we found that pigeons reared in a wind-screened aviary developed navigational abilities if, once adult, they were allowed to perform spontaneous flights around the loft. We describe a tuberculosis outbreak caused by Mycobacterium bovis in a conditioned colony of rhesus (Macaca mulatta) and cynomolgus (Macaca fascicularis) macaques. Animals in five rooms were exposed, but most (16/27) infections were confined to the room that housed a mixed population of cynomolgus and rhesus macaques. In this room, rhesus (8/8) and cynomolgus (10/11) macaques naturally exposed to M. bovis were infected at nearly identical rates (Fisher exact test, 2-tailed P = 1). The clinical signs of disease and pathologic lesions in infected macaques, however, were moderately different between the two species. Rhesus macaques were more likely (5/8) to exhibit clinical signs of persistent coughing and inappetance, and had more severe pulmonary lesions. By contrast, clinical signs of disease were seen in only 1 of 19 cynomolgus macaques, and overall, the pulmonary lesions were often focal and less severe, although some still had severe involvement of the lungs similar to that seen in rhesus macaques. These differences should be taken into consideration when developing or evaluating a tuberculosis-screening program. On the basis of observations made during this outbreak, we recommend that alternative screening methods such as the PRIMAGAM test and the ESAT-6 ELISA, be incorporated into the screening program to aid in the identification of infected animals.  相似文献

Pathways of infection of Brassica napus roots by Leptosphaeria maculans   总被引:1,自引:0,他引:1  
Infection of Brassica napus cotyledons and leaves by germinating ascospores of Leptosphaeria maculans leads to production of leaf lesions followed by stem cankers (blackleg). Leptosphaeria maculans also causes root rot but the pathway of infection has not been described. An L. maculans isolate expressing green fluorescent protein (GFP) was applied to the petiole of B. napus plants. Hyphal growth was followed by fluorescence microscopy and by culturing of sections of plant tissue on growth media. Leptosphaeria maculans grew within stem and hypocotyl tissue during the vegetative stages of plant growth, and proliferated into the roots within xylem vessels at the onset of flowering. Hyphae grew in all tissues in the stem and hypocotyl, but were restricted mainly to xylem tissue in the root. Leptosphaeria maculans also infected intact roots when inoculum was applied directly to them and hyphae entered at sites of lateral root emergence. Hyphal entry may occur at other sites but the mechanism is uncertain as penetration structures were not observed. Infection of B. napus roots by L. maculans can occur via above- and below-ground sources of inoculum, but the relative importance of the infection pathways under field conditions is unknown.  相似文献

Effect of Interferon on Some Aspects of Transformation by Polyoma Virus   总被引:1,自引:0,他引:1  
WHEN BHK 21 hamster cells are infected with polyoma virus1, there is no vegetative growth of virus, but stably transformed cells appear. These transformed cells are more easily transplanted than BHK 21 cells; they initiate their growth cycle in otherwise restrictive cultural conditions such as the absence of serum, high density and suspension; they grow with random orientation and have exposed on their surfaces receptor sites for certain glycoprotein agglutinins2–5. The proportion of stably transformed cells is low, even after high doses of virus. But a much higher proportion (sometimes all cells) shows abortive transformation—changes characteristic of transformation, but which last only a few days. In suspension cultures, for example, most of the infected cells grow into small colonies of four to thirty-two cells6. In surface cultures deprived of serum DNA synthesis is initiated and the cells may then divide at least once7: they also temporarily lose the normal parallel orientation and develop the typical random appearance of transformed cells. Moreover, the polyoma nuclear T-antigen and also a surface antigen detected by immunofluorescence, appear temporarily in most polyoma infected BHK 21 cells8, while 3T3 cells exposed to SV40 virus show transient exposure of cell surface sites reacting with conconavalin A (ref. 9).  相似文献

Currently accepted conservative treatments of hypertrophic scars are limited to steroid injections, radiation therapy, and silicone occlusive therapy. However, the use of Mederma for these problematic lesions has become quite prevalent in the clinical setting. Little scientific evidence exists to support the efficacy of this product in reducing hypertrophic scars. The aim of this study was to study the effects of Mederma on hypertrophic scars in the rabbit hypertrophic scar model, allowing the histologic quantification of scar elevation, dermal collagen organization, vascularity, and inflammation and the gross examination of scar erythema. Full-thickness wounds down to cartilage, four per ear, were created in four New Zealand White rabbits, for a total of 32 scars. Twenty-eight days after the initial wounding, the hypertrophic scars were photographed, and treatment of half of the scars on each ear was begun with Mederma three times per day for a total of 4 weeks. The untreated scars served as control scars and were left exposed to air. After 4 weeks of treatment, the scars were once again photographed. The rabbits were then killed, and the scars were analyzed histologically. The pretreatment and posttreatment photographs were compared by using computer quantification of magenta, yellow, and cyan expression within the scars.Histologic analysis demonstrated no significant reduction in scar hypertrophy or scar elevation index. However, a significant improvement in dermal collagen organization was noted on comparing Mederma-treated scars with untreated control scars (p < 0.05). No significant difference in dermal vascularity or inflammation was noted. Computer analysis of the scar photographs demonstrated no significant reduction in scar erythema with Mederma treatment. The active product in Mederma, allium cepa, has as its derivative quercetin, a bioflavonoid noted for its antiproliferative effects on both normal and malignant cells, and its antihistamine release effects. These properties could theoretically prove beneficial in reversing the inflammatory and proliferative responses noted in hypertrophic scars. Despite the authors' inability to demonstrate a reduction in scar hypertrophy, the improvement in collagen organization noted in the Mederma-treated scars suggests it may have an effect on the pathophysiology of hypertrophic scar formation.  相似文献

Vermetidae are a small family of warm-water sessile gastropods capable of building upright tubes (feeding-tubes) to take advantage of the water flow. Laboratory and field experiments carried out on some Mediterranean species ( Vermetus triquetrus, Vermetus granulatus and Serpulorbis arenaria ) suggest that these structures function as exploratory tubes built not only to receive a better water flow, but mainly to avoid obstacles. In fact, vermetids experimentally exposed in situ to different hydrodynamic conditions do not produce them, but do so in the presence of an obstacle, such as thalli during the massive spring algal growth. This strategy allows them to compete for a virtual space, not directly occupied by the vermetid itself but necessary to spread its mucous net. This interpretation may improve the inference of paleo-environmental events from shell morphology. When building a feeding-tube, vermetids first cut off a portion of the shell on the side towards which they prepare to turn, using their radula, and then produce a new tube formed by short segments, at different angles, till they have reached the desired direction. This process is confirmed by the presence of scars on the shell, composed by a succession of lamellae. The regular distribution of these scars on fossil Petaloconchus intortus , which lived in soft substrates, may be interpreted as a response to periodical anoxic crises or an increase in the sedimentation rates. Their great morphological plasticity makes vermetids close to colonial or modular animals. Thanks to their capability of expressing more than one growth-form, and of re-moulding their shell, they successfully compete for substrate space and are key-stone species in fringe habitats.  相似文献

Biological control of plant diseases generally requires release of living organisms into the environment. Cryphonectria hypoviruses function as biological control agents for the chestnut blight fungus, Cryphonectria parasitica, and hypovirus-infected C. parasitica strains can be used to treat infected trees. We used naturally occurring molecular marker polymorphisms to examine the persistence and dissemination of the three genomes of a hypovirus-infected C. parasitica strain, namely, the double-stranded RNA genome of Cryphonectria hypovirus 1 (CHV1) and the nuclear and mitochondrial genomes of its fungal host. The hypovirus-infected strain was experimentally introduced into a blight-infested chestnut coppice forest by treating 73 of 246 chestnut blight cankers. Two years after introduction, the hypovirus had disseminated to 36% of the untreated cankers and to 35% of the newly established cankers. Spread of the hypovirus was more frequent within treated sprout clusters than between sprout clusters. Mitochondrial DNA of the introduced fungus also was transferred into the resident C. parasitica population. Concomitant transfer of both the introduced hypovirus and mitochondrial DNA was detected in almost one-half of the treated cankers analyzed. The introduced mitochondrial DNA haplotype also was found in three resident isolates from newly established cankers. The nuclear genome of the introduced strain persisted in the treated cankers but did not spread beyond them.  相似文献

STUDIES IN THE BACTERIAL DIE-BACK AND CANKER DISEASE OF POPLAR   总被引:1,自引:0,他引:1  
The bacterial die-back and canker of poplar is characterized by dark necrotic areas in the leaves, die-back accompanied by cracks in the bark of 1–4-year-old branches, and by cankers on the trunk and branches of different ages. The cankers may be 'closed' or 'open' and when moist exude slime containing bacteria.
The disease is caused by Pseudomonas syringae f. sp. populea , a new forma specialis closely resembling Ps. syringae in bacteriological characters but differing from it in parasitism. The association of the slime with the bacteria is essential for the production of typical lesions.
Inoculations with the pathogen alone, or in association with other organisms produce atypical symptoms.
The bacteria attack the cortical parenchyma and occasionally invade the xylem vessels.  相似文献

Low concentrations of adenine arabinoside inhibited growth of two Epstein-Barr virus producer cell lines in culture, while not significantly affecting a nonproducer cell line and a B-cell-negative line. These observations were extended to include freshly infected cells. Mitogen-stimulated human umbilical cord blood lymphocytes were unaffected by the drug at concentration levels that inhibited [3H]thymidine incorporation into the DNA of Epstein-Barr virus-stimulated cells. DNA synthesis in Epstein-Barr virus-superinfected Raji cells was also adversely affected by adenine arabinoside. However, these same low concentrations of adenine arabinoside in the triphosphate form produced less effect on DNA synthesis in nuclear systems and DNA polymerase assays than on growth or DNA synthesis in whole cells. Therefore the effects reported here of low concentrations of the drug on whole cells may be only in part related to DNA polymerase inhibition. The work reported here suggests that adenine arabinoside has multiple sites of action in infected cells.  相似文献

Plum decline was associated with Pseudomonas syringae pathovars syringae and morsprunorum in Baden‐Württemberg. The trunks of affected plum trees (Prunus domestica) were girdled by bacterial cankers resulting in sudden death of infected trees. Copper compounds that were applied extensively during leaf fall and bud burst, were not effective. A minority of P. syringae strains isolated from cankers on plum trees were moderately resistant, while most strains were sensitive to cupric ions. Invasions through blossoms, leaves and wounds during the vegetation period were limited to the infection sites and plum trees coped effectively with both P. syringae pathovars eliminating them eventually. Infections after dormancy including very rare leaf scar infections did not induce cankers on the trunk. However, infections of dormant trees through frost injuries, (pruning) wounds or non‐injurious ingress by freezing and thawing were serious, because they led to cankers girdling the trunk. Control strategies to manage plum decline have to be adapted to the disease cycle. They should concentrate on the dormant period beginning with early frosts in autumn and ending with bud burst.  相似文献

Black women have not embraced cosmetic and reconstructive surgery of the breast with the same enthusiasm as their Caucasian counterparts because of fear of hypertrophic scars. The authors offer suggestions on how to minimize the scarring associated with breast surgery in black women. They feel that intraareolar incisions should be used whenever circumareolar incisions are indicated in augmentation mammaplasty, because the areola, being a favored area, is less likely to produce hypertrophic scars. The Marchac technique of reduction mammaplasty is recommended because it produces a short horizontal scar of 5 to 8 cm confined to the breast without medial and lateral extension, which may hypertrophy in black women. In the reduction of large breasts, secondary excision of dogears 6 or more weeks after mammaplasty reduces the medial and lateral extents of the scar. The use of liposuction as an adjunct to reduction mammaplasty may also accomplish the same thing. Amputation and free nipple-areola grafting should be used with caution in black patients because of the tendency of the grafted areola to hypopigment. In postmastectomy reconstruction, the authors suggest that the techniques described by Ryan and Radovan should be considered first before the techniques of reconstruction utilizing myocutaneous flaps. In these procedures, no new scars which may hypertrophy are created away from the site of reconstruction. Staples should not be used in skin closure in blacks because they cause cross-hatching of the wound even when removed early.  相似文献

Six antisporulant compounds, selected from 120 chemicals screened against Gloeosporium fructigenum in vitro , were applied to lesions on Cox apple shoots caused by Nectria galligena or G. perennans . In the following 11 months, 42 and 33% fewer spores were trapped from N. galligena cankers treated with 2,4,5,6-tetrachlorophenol or benomyl respectively, and 87 and 71% fewer from G. perennans cankers treated with 8-hydroxyquinoline or benomyl respectively, compared with those caught from untreated cankers. Miscible tar-petroleum oil had an antisporulant effect equal to that of benomyl on both N. galligena and G. perennans .  相似文献

Exposing both surfaces of leaves systemically infected with cabbage black ring spot virus (CBRSV) or henbane mosaic virus to ultra-violet radiation decreases the infectivity of expressed sap to about one-fifth. As irradiation probably inactivates virus mainly in the epidermis, which occupies about one-quarter the volume of the leaves, these viruses seem to occur at much higher concentrations in sap from the epidermis than in sap from other cells. By contrast, tobacco mosaic virus seems not to occur predominantly in the epidermis.
CBRSV and henbane mosaic virus are normally transmitted most frequently by previously fasted aphids that feed for only short periods on infected leaves, but aphids treated like this transmit rarely from leaves that have been exposed to ultraviolet radiation. Irradiation has relatively little effect on the proportion of aphids that transmit after long infection feedings. Fasting seems to increase transmission by increasing the probability that aphids will imbibe sap from the epidermis of leaves they newly colonize. With longer periods on infected leaves, the ability of fasted aphids to transmit probably decreases because they then feed from deeper cells and their stylets contain sap with less virus. Only virus contained in the stylets seems to be transmitted, not virus taken into the stomach. About half the transmissions of henbane mosaic virus by aphids that have colonized tobacco leaves for hours may be caused by insects that temporarily cease feeding on the phloem and newly penetrate the epidermis.
Irradiating infected leaves affected the transmission of sugar-beet mosaic virus in the same way as that of henbane mosaic virus, but had little effect on the transmission of beet yellows virus, whose vectors become more likely to transmit the longer they feed on infected plants.  相似文献

During the first trimester of pregnancy the uterus is massively infiltrated by decidual natural killer cells (dNK). These cells are not killers, but they rather provide a microenvironment that is propitious to healthy placentation. Human cytomegalovirus (HCMV) is the most common cause of intrauterine viral infections and a known cause of severe birth defects or fetal death. The rate of HCMV congenital infection is often low in the first trimester of pregnancy. The mechanisms controlling HCMV spreading during pregnancy are not yet fully revealed, but evidence indicating that the innate immune system plays a role in controlling HCMV infection in healthy adults exists. In this study, we investigated whether dNK cells could be involved in controlling viral spreading and in protecting the fetus against congenital HCMV infection. We found that freshly isolated dNK cells acquire major functional and phenotypic changes when they are exposed to HCMV-infected decidual autologous fibroblasts. Functional studies revealed that dNK cells, which are mainly cytokines and chemokines producers during normal pregnancy, become cytotoxic effectors upon their exposure to HCMV-infected autologous decidual fibroblasts. Both the NKG2D and the CD94/NKG2C or 2E activating receptors are involved in the acquired cytotoxic function. Moreover, we demonstrate that CD56pos dNK cells are able to infiltrate HCMV-infected trophoblast organ culture ex-vivo and to co-localize with infected cells in situ in HCMV-infected placenta. Taken together, our results present the first evidence suggesting the involvement of dNK cells in controlling HCMV intrauterine infection and provide insights into the mechanisms through which these cells may operate to limit the spreading of viral infection to fetal tissues.  相似文献
