Resveratrol exhibited the inhibitory activity against mushroom tyrosinase (EC1.14.18.1) through a k(cat) inhibition. Resveratrol itself did not inhibit tyrosinase but rather was oxidized by tyrosinase. In the enzymatic assays, resveratrol did not inhibit the diphenolase activity of tyrosinase when l-3,4-dihydroxyphenylalanin (L-DOPA) was used as a substrate; however, L-tyrosine oxidation by tyrosinase was suppressed in presence of 100 μM resveratrol. Oxidation of resveratrol and inhibition of L-tyrosine oxidation suggested the inhibitory effects of metabolites of resveratrol on tyrosinase. After the 30 min of preincubation of tyrosinase and resveratrol, both monophenolase and diphenolase activities of tyrosinase were significantly suppressed. This preincubational effect was reduced with the addition of L-cysteine, which indicated k(cat) inhibition or suicide inhibition of resveratrol. Furthermore, investigation was extended to the cellular experiments by using B16-F10 murine melanoma cells. Cellular melanin production was significantly suppressed by resveratrol without any cytotoxicity up to 200 μM. trans-Pinosylvin, cis-pinosylvin, dihydropinosylvin were also tested for a comparison. These results suggest that possible usage of resveratrol as a tyrosinase inhibitor and a melanogenesis inhibitor.  相似文献

探讨驱虫斑鸠菊(VW)注射液治疗白癜风的作用机制。进行以下实验探讨VW对小鼠免疫功能的影响:淋巴细胞转化试验测定小鼠脾T、B细胞增殖活性;脾细胞介导羊红细胞定量溶血分光光度法测定B细胞生成抗体活性,流式细胞法测定B细胞上CD19表达活性;迟发型超敏反应(DTH)试验测定T细胞活性、眼3H演-TdR掺入法测定T细胞分泌IL-2活性等。用酶学方法研究VW对小鼠体内酪氨酸酶的作用。用半定量逆转录聚合酶链反应技术检测酪氨酸酶基因表达活性。结果表明,VW可以明显抑制小鼠体内T、B细胞的增殖反应(P<0.01);对绵羊红细胞(SRBC)诱导的正常小鼠脾脏抗体形成细胞活性、CD19B细胞亚类表达、小鼠DTH反应和T细胞分泌IL-2活性也具有明显的抑制作用,这些抑制作用与药物浓度有一定的剂量效应关系。VW还可以提高小鼠血清酪氨酸酶活性,增强酪氨酸酶基因的表达。以上结果说明,VW可抑制小鼠免疫功能,可以从转录水平增强酪氨酸酶活性,进而促进黑素合成。  相似文献

In studies dealing with melanogenesis in mammalian tissues, ultrastructural localization of enzymes, identification of subcellular organelles, differentiation and lignification in plant tissues, it is important to have means to differentiate between tyrosinase and peroxidase activities. For a variety of reasons, established criteria used for this purpose are not always reliable. We suggest that tropolone can aid in differentiating between tyrosinase and peroxidase activities since: (a) it is a very effective inhibitor of tyrosinase; (b) in the presence of hydrogen peroxide it can serve as a substrate for peroxidase; (c) at concentrations that inhibit tyrosinase, it does not inhibit peroxidase activity; and (d) it inhibits tyrosinase activity even in the presence of hydrogen peroxide and peroxidase. In a system containing a mixture of tyrosinase and peroxidase, tropolone can differentiate reliably between peroxidase and monohydroxyphenolase or o-dihydroxyphenolase activities of tyrosinase. Moreover, tropolone can differentiate reliably between peroxidase and tyrosinase activities using slices or crude dialysed extracts of various plant tissues.  相似文献

Indole and tryptophan inhibit tyrosinase reaction and bovine albumin protects against end-product(s) inhibition or inactivation. Novel 3,4-dihydroquinazoline-2(1H)-thiones (QNTs) 1 were found to be potent inhibitors of α-MSH-induced melanin production. The effect of QNTs to inhibit melanin formation in B16 melanoma cells was screened in the presence of α-MSH. In defining the mechanism of activity, the effects on tyrosinase activity, on tyrosinase synthesis and on the depigmentation of melanin were evaluated. QNTs did not affect the catalytic activity of tyrosinase, but rather acted as an inhibitor of tyrosinase synthesis.  相似文献

The thymidine analog 5-bromodeoxyuridine (BrdU) has been found to inhibit terminal differentiation of a variety of cell types without significantly affecting the growth of these cells. We have compared the effect of BrdU with two other thymidine analogs, 5-iododeoxyuridine and 5-fluorodeoxyuridine, on the growth, tyrosinase activity, and tyrosinase-mRNA accumulation in BL-6 mouse melanoma cells. We show that all three analogs inhibit growth and tyrosinase activity, but only BrdU significantly inhibits the accumulation of tyrosinase mRNA. We consider these results in the light of current understanding of BrdU action.  相似文献

Human tyrosinase (5.5 mg) has been purified from a single human melanotic melanoma metastasis (50.5 g). In the presence of dioxygen, L-tyrosine proved to be a very poor substrate for this enzyme with barely detectable activity compared to L-dopa. However, saturating superoxide anion (i.e., greater than 5 x 10(-3) M) enhanced the oxidation rate of L-tyrosine to dopachrome 40-fold. Hydrogen peroxide was shown to be a competitive inhibitor of tyrosinase when L-tyrosine was the substrate. This reversible inhibition is based on a slow pseudocatalase activity for tyrosinase. Monothiols and dithiols inhibit tyrosinase by different mechanisms. Reduced human thioredoxin and 2,3-dithiopropanol are allosteric inhibitors of tyrosinase yielding bis-cysteinate complexes with one of the copper atoms in the enzyme active site. Bis-cysteinate tyrosinase activity is down-regulated to 30% of native enzyme activity in the L-dopa assay; suggesting a true regulatory role for dithiols. Monothiols such as reduced glutathione and beta-mercaptoethanol are much less reactive with tyrosinase although 10(-3) M monothiol totally inhibits enzyme activity. Reduced thioredoxin inhibits tyrosinase 23-fold more than reduced glutathione under the same experimental conditions.  相似文献

An anti-cancer peptide was purified from the Mercenaria (Meretrix meretrix Linnaeus) by the method of chromatography on Sephadex G-25 and FPLC, and its molecular weight was determined to be 3147 Da by the way of MALDI-TOF mass spectrum. The effects of this peptide on human gastric gland carcinoma cells (BGC-823) and their cytoskeletal morphology were investigated. The results showed that the peptide could inhibit the proliferation of BGC-823 cells and obviously destroy the skeletal structures of the cells. When the concentration of the peptide reached 4.0 microg/ml, the inhibition percentage of the cell growth was about 60%. The effects of this anticancer peptide on the activities of superoxide dismutase (SOD), alkaline phosphatase (ALP) and tyrosinase were studied. The results showed that the peptide activated ALP and SOD, but inhibit the tyrosinase activity. When the concentration of the peptide reached to 0.5 microg/ml, the relative activities of SOD, ALP and tyrosinase were determined to be 188.5%, 122.0% and 27.5%, respectively.  相似文献

Incubation of cultured B-16 melanoma cells with 1-methyl-3-isobutyl xanthine (MIX) produced a sustained rise in intracellular adenosine 3',5'-cyclic monophosphate (cAMP) which preceded an increase in the specific activity of tyrosinase (EC 1.10.3.1). Cultures of two clones of melanoma cells, one having a mean population doubling time twice that of the other, showed density-dependent inhibition of growth. The tyrosinase activity of each line increased progressively during logarithmic growth, reaching maximal values shortly after the cultures achieved confluence. Intracellular cAMP levels fell during logarithmic growth, being minimal in confluent cultures. The stimulatory effects of MIX and confluence on tyrosinase activity were additive. Cells plated at high density had a lower tyrosinase activity than cells allowed to achieve a similar density by successive division from sparsely planted cultures although the intracellular cAMP levels of such cultures were not different. We support the observations of other investigators that agents which increase intracellular cAMP concentrations can both inhibit cell division and stimulate tyrosinase activity. There are, however, mechanisms for increasing tyrosinase activity and inhibiting cell division which are expressed as B-16 melanoma cells approach confluence and which are not mediated by an increase in intracellular cAMP concentrations.  相似文献

以银杏外种皮为材料,采用水、乙醇和乙醇-乙醚三种不同的方法分离提取其中的活性物质(分别命名为1#,2#,3#),研究它们对蘑菇酪氨酸酶催化L-多巴(L-DOPA)氧化活力的影响。结果表明,这3种提取物均对蘑菇酪氨酸酶有抑制作用,1#,2#和3#对酶抑制作用的IC50分别为2.25、1.75和0.32mg/mL。抑制作用动力学结果表明:三种提取物对酶的抑制作用均表现为混合型,相应的抑制常数KI依次为2.11、1.62和0.29mg/mL;KIS依次为2.80、2.33和0.45mg/mL。结果显示,采用乙醇-乙醚提取的银杏外种皮提取物对酪氨酸酶抑制作用最强。  相似文献

Maltol (3-hydroxy-2-methyl-4H-pyran-4-one) appears to inhibit the rate of oxidation of DL-DOPA, dopamine, NADA and epinephrine by tyrosinase when assayed spectropho-tometrically but not when assayed polarographically. Maltol has an effect on the spectrum of product(s) formed when each catecholamine was oxidized by tyrosinase showing that maltol hastens the disappearance of the quinones, possibly by conjugating with them. Indeed, at relatively high concentrations, maltol prevented the conversion of DL-DOPA, dopamine, and norepinephrine to their corresponding melanins via tyrosinase.  相似文献

3,3',5-L-triiodothyronine (T3) can inhibit tyrosinase activity in B16/C3 melanoma cells in culture and block the induction of that enzyme by imidazole (Endocrinol. 119, 2118, 1986). The current study examined the effect of thyroxine (T4) on tyrosinase activity. Proliferating cultures were exposed to either T3 (0-500 nM) or T4 (0-50,000 nM) in addition to imidazole (10 mM). Imidazole induced enzyme activity by approximately 3-fold and T3 could block that induction with a maximal inhibition occurring at 10 nM. Even at the highest concentrations used, T4 had no effect on tyrosinase activity. These studies disclose that T4, the tetraiodinated parent compound from which T3 is derived, is devoid of intrinsic biological activity in this T3 responsive system.  相似文献

Down-regulation of melanin synthesis is required for recovery of pigmentary disorders and it is known that direct inhibitors of tyrosinase, the key enzyme in melanin synthesis, such as hydroquinone with a phenol structure, suppress melanin synthesis. We screened several phenolic derivatives using B16 melanoma cells and found that a biphenyl derivative, 2,2'-dihydroxy-5,5'-dipropyl-biphenyl (DDB), down-regulated melanin synthesis effectively. Although DDB has a phenol structure, it did not inhibit tyrosinase in vitro, thus we examined its mechanism in detail. Western blotting revealed that the amount of tyrosinase was decreased by DDB, and pulse-chase labeling and immunoprecipitation analysis showed a decrease of mature tyrosinase and acceleration of tyrosinase degradation in its presence. These results suggest that DDB down-regulates melanin synthesis by inhibiting the maturation of tyrosinase, leading to acceleration of tyrosinase degradation.  相似文献

Tiliroside was found to inhibit both monophenolase and diphenolase activity of mushroom tyrosinase. The lag time of tyrosine oxidation catalyzed by mushroom tyrosinase was obviously lengthened; 0.337?mM of tiliroside resulted in the lag time extension from 46.7?s to 435.1?s. A kinetic analysis shown that tiliroside was a competitive inhibitor for monophenolase and diphenolase with Ki values of 0.052?mM and 0.26?mM, respectively. Furthermore, tiliroside showed 34.5% (p?<?0.05) inhibition of intracellular tyrosinase activity and 54.1% (p?<?0.05) inhibition of melanin production with low cytotoxicity on B16 mouse melanoma cells at 0.168?mM. In contrast, arbutin displayed 9.1% inhibition of cellular tyrosinase activity and 29.5% inhibition of melanin production at the same concentration. These results suggested that tiliroside was a potent tyrosinase inhibitor and might be used as a skin-whitening agent and pigmentation medicine.  相似文献

Compounds, which inhibit tyrosinase, could be effective as depigmenting agents. We have introduced a group of mono-, di-, tri- and tetra-substituted hydroxychalcones as effective tyrosinase inhibitors, showing that the most important factor determining tyrosinase inhibition efficiency is the position of the hydroxyl group(s) rather their number. The aim of the present study was to investigate the contribution of the different functional groups of the tetrahydroxychalcones to their inhibitory potency, with a view to optimizing the design of whitening agents. Four tetrahydroxychalcones were evaluated, the commercially available Butein and other three were synthesized, and their inhibitory effect on tyrosinase was tested. Results showed that a 2,4-substituted resorcinol subunit on ring B contributed the most to inhibitory potency. Changing the resorcinol substitute to position 3,5- or placing it on ring A significantly diminished the inhibitory effect of the compounds. A catechol subunit on ring A acted as a metal chelator (in the presence of copper ions) and as a competitive inhibitor (in the presence of tyrosinase), while a catechol on ring B oxidized to o-quinone (in the presence of both copper ions and tyrosinase). Three of the compounds also demonstrated antioxidant activity, which may contribute to the prevention of pigmentation. An examination of correlations between inhibitory activity and physical properties of the chalcones tested (such as dissociation energy and molecular planarity) showed positive correlation with the moment dipole value in the Y-axis, which may be used as an indicator of the inhibitory potential of new molecules. The present study revealed two very active tyrosinase inhibitors, 2,4,3',4'-hydroxychalcone and 2,4,2',4'-hydroxychalcone (with IC50 of 0.2 and 0.02 microM, respectively). Structure-related activity studies added some understanding of the role and contribution of different functional groups associated with tyrosinase inhibitors.  相似文献

The rationale for melanoma specific dihydroxybenzene containing antitumor agents is based in part upon the ability of the enzyme tyrosinase to oxidize these pro drugs to toxic intermediates. In situ tyrosinase activity was demonstrated to be affected by both cell density and time from plating in pigmented melanoma cells. Phenylthiourea, which completely inhibited tyrosinase activity with minimal cytotoxicity was found to block the growth inhibitory activity of the antitumor dopamine analog 3,4-dihydroxybenzylamine (3,4-DHBA) (NSC 263475). The antioxidant dithioerythritol was also found to inhibit tyrosinase activity and to block the growth inhibitory effects of 3,4-DHBA in pigmented melanoma cell lines. Buthionine sulfoximine (BSO) was shown to be cytotoxic to melanoma cells and its growth inhibitory effects appears to correlate with tyrosinase levels. Furthermore, BSO was shown to potentiate the growth inhibitory effects of 3,4-DHBA on marginally pigmented human melanoma cell lines.  相似文献

Tyrosinase is an enzyme having two copper atoms at the reactive site occurring in prokaryotic and eukaryotic organisms. In animals tyrosinase is responsible for pigmentation, in plants for protection of injured tissues or, as in fungi, to harden cell walls. Some of us have previously shown that tyrosinase is involved in truffle development and differentiation. Here we present the purification, the molecular properties and the reversible inhibition of Tuber melanosporum tyrosinase by dimethyl-sulfide and bis[methylthio]methane, the main flavour compounds of black and whitish truffles. The MW(r) is 39000. L-3,4-dihydroxyphenylalanine and L-tyrosine stain corresponding bands as expected for a true tyrosinase. Phenylthiourea, diethyldithiocarbamate and mimosine inhibit L-tyrosine and L-3,4-dihydroxyphenylalanine oxidation.  相似文献

Down‐regulation of melanin synthesis is required for recovery of pigmentary disorders and it is known that direct inhibitors of tyrosinase, the key enzyme in melanin synthesis, such as hydroquinone with a phenol structure, suppress melanin synthesis.
