In order to study the effects of desiccation on a photosynthetic system, light harvesting and light-induced electron transport processes were examined in pea cotyledons at various moisture levels, using in vivo fluorescence excitation spectra and fluorescence induction kinetics. Water sorption isotherms yielded thermodynamic data that suggested very strong water binding between 4 to 11% water, intermediate sorption between water contents of 13 to 22%, and very weak binding at moisture contents between 24 to 32%. The fluorescence properties of the tissue changed with the moisture contents, and these changes correlated generally with the three regions of water binding. Peak fluorescence and fluorescence yield remained at low levels when water content was limited to the tightly bound regions, below 12%. Several new peaks appeared in the chlorophyll a excitation spectrum and both peak fluorescence and fluorescence yield increased at intermediate water-binding levels (12-22%). At moisture contents where water is weakly bound (>24%), peak fluorescence and fluorescence yield were maximum and the fluorescence excitation spectrum was unchanging with further increases in water content.

The state of water is an important component in the energy transfer and electron transport system. At hydration levels where water is most tightly bound, energy transfer from pigments is limited and electron transport is blocked. At intermediate water binding levels, energy transfer and electron transport increase and, in the region of weak water binding, energy transfer and electron transport are maximized.

In an attempt to correlate freezable water with freezing injury, the thermal behavior of pea (Pisum sativum L.) and soybean (Glycine max L. Merr) seed parts at different moisture contents were compared with survival of the seeds when exposed to low temperatures. Thermal transitions between −150 and 10°C were studied using differential scanning calorimetry. In pea, reduction of germinability, after exposure of seeds to temperatures between − 18 and − 180°C, occurred at a constant moisture content (about 0.33 gram H2O/gram dry weight) regardless of the temperature; this moisture level was above that at which freezable water was first detectable by differential scanning calorimetry (0.26 gram H2O/gram dry weight). In contrast, damage to soybean seeds was observed at progressively lower moisture contents (from 0.33 to 0.20 gram H2O/gram dry weight) when the temperature was decreased from −18°C to −50°C. At −18 and −30°C, moisture contents at which damage to soybean seeds was evident were above that at which freezable water was first detectable (0.23 gram H2O/gram dry weight). However, at −50, −80, and −180°C, damage was evident even when freezable water was not detectable. The data suggest that, while the quantity of water is important in the expression of freezing injury, the presence of freezable water does not account for the damage.  相似文献

In an effort to define the nature of desiccation tolerance, a comparison of the water sorption characteristics was made between tissues that were resistant and tissues that were sensitive to desiccation. Water sorption isotherms were constructed for germinated and ungerminated soybean axes and also for fronds of several species of Polypodium with varying tolerance to dehydration. The strength of water binding was determined by van't Hoff as well as D'Arcy/Watt analyses of the isotherms at 5, 15, and/or 25°C. Tissues which were sensitive to desiccation had a poor capacity to bind water tightly. Tightly bound water can be removed from soybean and pea seeds by equilibration at 35°C over very low relative humidities; this results in a reduction in the viability of the seed. We suggest that region 1 water (i.e. water bound with very negative enthalpy values) is an important component of desiccation tolerance.  相似文献

Application of water stress to isolated spinach (Spinacia oleracea) chloroplasts by redutcion of the osmotic potentials of CO2 fixation media below −6 to −8 bars resulted in decreased rates of fixation regardless of solute composition. A decrease in CO2 fixation rate of isolated chloroplasts was also found when leaves were dehydrated in air prior to chloroplast isolation. An inverse response of CO2 fixation to osmotic potential of the fixation medium was found with chloroplasts isolated from dehydrated leaves—namely, fixation rate was inhibited at −8 bars, compared with −16 or −24 bars.  相似文献

A new molecularly imprinted polymer (MIP) adsorbent for clofibric acid (CA) was prepared by a non-covalent protocol. Characterization of the obtained MIP was achieved by scanning electron microscopy (SEM) and nitrogen sorption. Sorption experimental results showed that the MIP had excellent binding affinity for CA and the adsorption of CA by MIP was well described by pseudo-second-order model. Scatchard plot analysis revealed that two classes of binding sites were formed in the MIP with dissociation constants of 7.52±0.46 mg L−1 and 114±4.2 mg L−1, respectively. The selectivity of MIP demonstrated higher affinity for CA over competitive compound than that of non-imprinted polymers (NIP). The MIP synthesized was used to remove CA from spiked surface water and exhibited significant binding affinity towards CA in the presence of total dissolved solids (TDS). In addition, MIP reusability was demonstrated for at least 12 repeated cycles without significant loss in performance.  相似文献

Antibodies elicited in rabbits by immunization with an N6-benzyladenosine-bovine serum albumin conjugate bound N6-benzyladenosine specifically. The affinity constants and specific binding site concentrations for a number of cytokinins and related compounds were estimated by nonlinear least squares analysis of direct or competitive ultrafiltration data. The antisera contained 230 to 330 nanomoles of cytokinin binding sites per gram protein. Affinity constants were 8.8 × 108 molar−1 for 6-benzylaminopurine, 8.4 × 107 molar−1 for kinetin, 9.1 × 107 molar−1 for 6-(3-methyl-2-butenylamino)purine, 6.6 × 106 molar−1 for 6-(4-hydroxy-3-methyl-trans-2-butenylamino)purine, and 2.0 × 104 molar−1 for 6-methylaminopurine. Affinity constants were below the limit of detectability (<104 molar−1) for benzylamine, adenine, and other adenine derivatives without an N6-side chain. The N6-substituent was thus immunodominant, but the purine moiety was also necessary for binding affinity. The antibodies were immobilized on cyanogen bromide-activated Sepharose with 95% retention of binding capacity and without apparent change in affinity constants. Columns of the immobilized antibody retained 64% of the [3H]6-(3-methyl-2-butenylam-ino)purine from 2 nanomolar solutions and readily trapped [14C]6-benzylaminopurine that had been added to crude extracts of cabbage. Aqueous 10% pyridine adjusted to pH 7.3 with formic acid effectively eluted bound cytokinins from gel columns without loss of binding capacity of the immobilized antibody.  相似文献

Total water potential (ψ), solute potential, and turgor potential of field-grown muskmelon (Cucumis melo L.) fruit tissue (pericarp) and seeds were determined by thermocouple psychrometry at 5-day intervals from 10 to 65 days after anthesis (DAA). Fruit maturity occurred between 44 and 49 DAA, and seed germination ability developed between 35 and 45 DAA. Pericarp ψ was essentially constant at approximately −0.75 megapascal (MPa) from 10 to 25 DAA, then decreased to a minimum value of −1.89 MPa at 50 DAA before increasing to −1.58 MPa at 65 DAA. Seed ψ remained relatively constant at approximately −0.5 MPa from 10 to 30 DAA then decreased to −2.26 MPa at 50 to 60 DAA before increasing to −2.01 MPa at 65 DAA. After a rapid increase to 20 DAA, seed fresh weight declined until 30 DAA due to net water loss, despite continuing dry weight gain. As fruit and seed growth rates decreased, turgor potential initially increased, then declined to small values when growth ceased. A disequilibrium in ψ was measured between seeds and pericarp both early and late in development. From 20 to 40 DAA, the ψ gradient was from the seed to the tissue, coinciding with water loss from the seeds. From 50 to 65 DAA, seed ψ decreased, causing a reversal of the ψ gradient and a slight increase in seed water content. The partitioning of solutes between symplast and apoplast may create and maintain ψ gradients between the pericarp and seed. The low solute potential within the pericarp due to solute accumulation and loss of cellular compartmentation during ripening and sensecence may be involved in prevention of precocious germination of mature seeds.  相似文献

Changes in water status of developing seeds of Soybean (Glycine max L. Merrill.) grown under different moisture stress conditions were characterized by proton nuclear magnetic resonance (NMR)- spin–spin relaxation time (T2). A comparison of the seed development characteristics, composition and physical properties indicated that, characteristics like seed weight, seed number/ear, rate of seed filling increased with development stages but decreased with moisture stress conditions. The NMR- spin–spin relaxation (T2) component like bound water increased with seed maturation (40–50%) but decreased with moisture stress conditions (30–40%). The changes in seed water status to increasing levels of moisture stress and seed maturity indicates that moisture stress resulted in more proportion of water to bound state and intermediate state and less proportion of water in free-state. These changes are further corroborated by significant changes in protein and starch contents in seeds under high moisture stress treatments. Thus seed water status during its development is not only affected by development processes but also by moisture stress conditions. This study strongly indicated a clear moisture stress and development stage dependence of seed tissue water status in developing soybean seeds.  相似文献

Uptake of glutamine by the scutellum of germinating barley grain   总被引:1,自引:1,他引:0  
Scutella separated from germinating grains of barley (Hordeum vulgare L. cv Himalaya) took up [14C]glutamine at an initial rate of about 10 micromoles·gram−1·hour−1 in the standard assay conditions (pH 5, 30°C, 1 millimolar glutamine). Inhibition by unlabeled glutamine and by dinitrophenol indicated that about 95% of the uptake was due to carrier-mediated active transport. The pH optimum of the uptake was 5, and after correction for a nonmediated component the uptake appeared to conform to Michaelis-Menten kinetics with an apparent Km of about 2 millimolar and a Vmax of about 25 micromoles·gram−1·hour−1.

The uptake of glutamine was inhibited by all of the 18 amino acids tested; the mode of inhibition was studied only with proline and was competitive. Eight of the ten amino acids tested at high concentrations appeared to be able to inhibit the mediated uptake of glutamine virtually completely. However, when the inhibitory effect of asparagine was extrapolated to an infinitely high concentration of asparagine, about 24% of the mediated uptake of glutamine remained uninhibited. These results suggest that glutamine is taken up by two (or more) rather unspecific amino acid uptake systems, the minor one having no affinity for asparagine.

Glutamine and alanine could completely inhibit the mediated uptake of 1 millimolar leucine, but about 12% of the mediated uptake appeared to be uninhibitable by asparagine. Furthermore, the ratio of the mediated uptake of glutamine to that of leucine changed from 0.9 to 1.7 between days 1 and 3 of germination. These results give further support for the presence of two unspecific amino acid uptake systems in barley scutella.

Pea (Pisum sativum L.) mutant near-isogenic lines (RRrbrb, rrRbRb, rrrbrb) with lower starch but higher lipid contents, brought about by lesions in the starch biosynthetic pathway, had seed moisture sorption isotherms displaced below that of the wild type (RRRbRb). The negative logarithmic relationship between seed longevity and seed storage moisture content (%, f.wt basis), determined in hermetic storage at 65 degrees C, also differed: longevity in the mutant near-isogenic lines was poorer and less sensitive to moisture content than in the wild type (i.e. C(W) was lower). The low-moisture-content limit (m(c)) to this relation also differed, being lower in the mutant near-isogenic lines (5.4-5.9%) than in the wild type (6.1%). In contrast, all four near-isogenic lines showed no difference (P >0.25) in the negative semi-logarithmic relationship between equilibrium relative humidity (ERH) and seed longevity. It is concluded that the effect of these alleles at the r and rb loci on seed longevity was largely indirect; a consequence of their effect on seed composition and hence on moisture sorption isotherms. However, this explanation could not be invoked at moisture contents below m(c) where differences in longevity remained substantial (RRRbRb double that of rrrbrb). Hence, these mutant alleles affected seed longevity directly at very low moisture contents.  相似文献

Muskmelon (Cucumis melo L.) seeds are germinable 15 to 20 days before fruit maturity and are held at relatively high water content within the fruit, yet little precocious germination is observed. To investigate two possible factors preventing precocious germination, the inhibitory effects of abscisic acid and osmoticum on muskmelon seed germination were determined throughout development. Seeds were harvested at 5-day intervals from 30 to 65 days after anthesis (DAA) and incubated either fresh or after drying on factorial combinations of 0, 1, 3.3, 10, or 33 micromolar abscisic acid (ABA) and 0, −0.2, −0.4, −0.6, or −0.8 megapascals polyethylene glycol 8000 solutions at 30°C. Radicle emergence was scored at 12-hour intervals for 10 days. In the absence of ABA, the water potential (Ψ) required to inhibit fresh seed germination by 50% decreased from −0.3 to −0.8 megapascals between 30 and 60 DAA. The Ψ inside developing fruits was from 0.4 to 1.4 megapascals lower than that required for germination at all stages of development, indicating that the fruit Ψ is sufficiently low to prevent precocious germination. At 0 megapascal, the ABA concentration required to inhibit germination by 50% was approximately 10 micromolar up to 50 DAA and increased to >33 micromolar thereafter. Dehydration improved subsequent germination of immature seeds in ABA or low Ψ. There was a linear additive interaction between ABA and Ψ such that 10 micromolar ABA or −0.5 megapascal osmotic potential resulted in equivalent, and additive, reductions in germination rate and percentage of mature seeds. Abscisic acid had no effect on embryo solute potential or water content, but increased the apparent minimum turgor required for germination. ABA and osmoticum appear to influence germination rates and percentages by reducing the embryo growth potential (turgor in excess of a minimum threshold turgor) but via different mechanisms. Abscisic acid apparently increases the minimum turgor threshold, while low Ψ reduces turgor by reducing seed water content.  相似文献

The occurrence of trichloro- and tetrachloroguaiacols, -catechols, and -veratroles and their transformation was studied in freshwater and brackish water sediments putatively exposed to bleachery discharge. The samples contained both chloroguaiacols and chlorocatechols, of which >90% could not be removed by simple extraction. The bound concentrations varied and ranged from 550 μg kg of organic C−1 for 3,4,5-trichloroguaiacol to 8,250 μg kg of organic C−1 for tetrachlorocatechol. Chlorinated substrates added to the aqueous phase were rapidly bound to the sediment with Kp values between 1.3 and 2.8 ml kg of organic C−1 for the chloroguaiacols and chloroveratroles and 22 to 36 ml kg of organic C−1 for the chlorocatechols. Sediment samples incubated aerobically brought about O-methylation of 4,5,6-trichloroguaiacol to 3,4,5-trichloroveratrole in a yield of ca. 25%. Under anaerobic conditions, however, de-O-methylation of both the chloroguaiacols and chloroveratroles took place with synthesis of the corresponding chlorocatechols. In separate experiments, the chlorocatechols were not completely stable under anaerobic conditions, but their ultimate fate has not yet been resolved. Sediment which had been autoclaved twice at 121°C for 20 min was unable to bring about any of these transformations; we therefore conclude that they were mediated by biological processes. These results emphasize that, in determining the fate of chloroguaiacols and related compounds discharged into the aquatic environment, the cardinal roles of sorption to the sediment phase and of the oxygen tension must be taken into account. We propose a hypothetical guaiacol cycle to accommodate our observations.  相似文献

Effects of 2 °C chilling on the threshold moisture contentsand water potentials for various physiological processes wereestimated forAesculus hippocastanumL. seed. Seed harvested atthe time of maximum seed fall exhibited a dual response to drying:partial drying from near 50% to 32–40% moisture contentprogressively increased germination percentage (at 16 °C)up to various peak values; further desiccation was detrimental,confirming that the seeds are ‘recalcitrant’. Themoisture content for optimum germination was increased by atleast 10% as the chilling period was raised from 0 to 9 weeks.A negative linear relationship was found between log10mean timeto germinate and probit final germination, regardless of pre-treatment,indicating that partial desiccation and chilling are interchangeablein promoting germination of hydrated seed. For nearly fullyhydrated seeds, increasing the chilling period from 6 to 26weeks increased the viability-loss onset point for desiccationinjury from near 40% to about 48% moisture content without alteringthe drying rates of seed tissues. Extending moist chilling invarious seed lots from 0 to 26 weeks decreased subsequent longevityat 16 °C. For 26-week-chilled seeds longevity (the periodto lose one probit of germination) differed above and belowa threshold moisture content of 48%. It remained constant inthe moisture-content range 48–38%, but increased progressivelyas moisture content was raised above 48%. This threshold moisturecontent coincided with the value above which chilled seed pre-germinatedin storage. Recruitment for many arid‐zone plant species is expected to be impacted by the projected increase in soil temperature and prolonged droughts associated with global climate change. As seed dormancy is considered a strategy to avoid unfavorable conditions, understanding the mechanisms underpinning vulnerability to these factors is critical for plant recruitment in intact communities, as well as for restoration efforts in arid ecosystems. This study determined the effects of temperature and water stress on recruitment processes in six grass species in the genus Triodia R.Br. from the Australian arid zone. Experiments in controlled environments were conducted on dormant and less‐dormant seeds at constant temperatures of 25°C, 30°C, 35°C, and 40°C, under well‐watered (Ψsoil = −0.15 MPa) and water‐limited (Ψsoil = −0.35 MPa) conditions. Success at three key recruitment stages—seed germination, emergence, and survival—and final seed viability of ungerminated seeds was assessed. For all species, less‐dormant seeds germinated to higher proportions under all conditions; however, subsequent seedling emergence and survival were higher in the more dormant seed treatment. An increase in temperature (35–40°C) under water‐limited conditions caused 95%–100% recruitment failure, regardless of the dormancy state. Ungerminated seeds maintained viability in dry soil; however, when exposed to warm (30–40°C) and well‐watered conditions, loss of viability was greater from the less‐dormant seeds across all species. This work demonstrates that the transition from seed to established seedling is highly vulnerable to microclimatic constraints and represents a critical filter for plant recruitment in the arid zone. As we demonstrate temperature and water stress‐driven mortality between seeds and established seedlings, understanding how these factors influence recruitment in other arid‐zone species should be a high priority consideration for management actions to mitigate the impacts of global change on ecosystem resilience. The knowledge gained from these outcomes must be actively incorporated into restoration initiatives.  相似文献

Water deficits during seed filling often decrease seed size in soybean (Glycine max L.). The physiological basis for this response is not known but may result from direct effects of low seed water potential (Ψw) on the seed filling process. To determine whether low Ψw occurred in reproductive tissues of soybean, we monitored the water status (Ψw, Ψs, and Ψp) of leaf, pericarp, and seed (embryo and testa) tissue of greenhouse-grown plants subjected to a brief water deficit during the linear period of seed growth. Water deficits were imposed by withholding water and monitored in the reproductive tissues by thermocouple psychrometry. When water was abundant, leaf, pericarp, and seed Ψw were −0.5 to −0.7 megapascal at midday. When water was withheld, leaf Ψw decreased to −2.3 megapascals within 6 days. Pericarp Ψw also decreased to −1.9 megapascal during this time. Pericarp Ψs followed the decline in Ψw, but osmotic adjustment was not evident as the pericarp lost turgor completely by day 6. However, seed Ψw, Ψs, and Ψp were not significantly different from the controls. These results indicate that the water status of the developing seeds of soybean is not altered by short-term water deficits severe enough to inhibit the metabolic activity of the maternal plant. Maintenance of a favorable water status may be important for the conservation of seed growth rate exhibited by soybean under dry conditions.  相似文献

The initiation of radicle growth during seed germination may be driven by solute accumulation and increased turgor pressure, by cell wall relaxation, or by weakening of tissues surrounding the embryo. To investigate these possibilities, imbibition kinetics, water contents, and water (Ψ) and solute (ψs) potentials of intact muskmelon (Cucumis melo L.) seeds, decoated seeds (testa removed, but a thin perisperm/endosperm envelope remains around the embryo), and isolated cotyledons and embryonic axes were measured. Cotyledons and embryonic axes excised and imbibed as isolated tissues attained water contents 25 and 50% greater, respectively, than the same tissues hydrated within intact seeds. The effect of the testa and perisperm on embryo water content was due to mechanical restriction of embryo swelling and not to impermeability to water. The Ψ and ψs of embryo tissues were measured by psychrometry after excision from imbibed intact seeds. For intact or decoated seeds and excised cotyledons, Ψ values were >−0.2 MPa just prior to radicle emergence. The Ψ of excised embryonic axes, however, averaged only −0.6 MPa over the same period. The embryonic axis apparently is mechanically constrained within the testa/perisperm, increasing its total pressure potential until axis Ψ is in equilibrium with cotyledon Ψ, but reducing its water content and resulting in a low Ψ when the constraint is removed. There was no evidence of decreasing ψs or increasing turgor pressure (Ψ-ψs) prior to radicle growth for either intact seeds or excised tissues. Given the low relative water content of the axes within intact seeds, cell wall relaxation would be ineffective in creating a Ψ gradient for water uptake. Rather, axis growth may be initiated by weakening of the perisperm, thus releasing the external pressure and creating a Ψ gradient for water uptake into the axis. The perisperm envelope contains a cap of small, thin-walled endosperm cells adjacent to the radicle tip. We hypothesize that weakening or separation of cells in this region could initiate radicle expansion.  相似文献

The effect of cooling rate on seeds was studied by hydrating pea (Pisum sativum), soybean (Glycine max), and sunflower (Helianthus annuus) seeds to different levels and then cooling them to − 190°C at rates ranging from 1°C/minute to 700°C/minute. When seeds were moist enough to have freezable water (> 0.25 gram H2O/gram dry weight), rapid cooling rates were optimal for maintaining seed vigor. If the seeds were cooled while at intermediate moisture levels (0.12 to 0.20 gram H2O per gram dry weight), there appeared to be no effect of cooling rate on seedling vigor. When seeds were very dry (< 0.08 gram H2O per gram dry weight), cooling rate had no effect on pea, but rapid cooling rates had a marked detrimental effect on soybean and sunflower germination. Glass transitions, detected by differential scanning calorimetry, were observed at all moisture contents in sunflower and soybean cotyledons that were cooled rapidly. In pea, glasses were detectable when cotyledons with high moisture levels were cooled rapidly. The nature of the glasses changed with moisture content. It is suggested that, at high moisture contents, glasses were formed in the aqueous phase, as well as the lipid phase if tissues had high oil contents, and this had beneficial effects on the survival of seeds at low temperatures. At low moisture contents, glasses were observed to form in the lipid phase, and this was associated with detrimental effects on seed viability.  相似文献

Glycoprotein D (gD) of herpes simplex virus (HSV) is essential for virus entry and has four functional regions (I to IV) important for this process. We previously showed that a truncated form of a functional region IV variant, gD1(Δ290-299t), had an enhanced ability to block virus entry and to bind to the herpesvirus entry mediator (HveAt; formerly HVEMt), a cellular receptor for HSV. To explore this phenotype further, we examined other forms of gD, especially ones with mutations in region IV. Variant proteins with deletions of amino acids between 277 and 300 (region IV), as well as truncated forms lacking C-terminal residues up to amino acid 275 of gD, were able to block HSV entry into Vero cells 1 to 2 logs better than wild-type gD1(306t). In contrast, gD truncated at residue 234 did not block virus entry into Vero cells. Using optical biosensor technology, we recently showed that gD1(Δ290-299t) had a 100-fold-higher affinity for HveAt than gD1(306t) (3.3 × 10−8 M versus 3.2 × 10−6 M). Here we found that the affinities of other region IV variants for HveAt were similar to that of gD1(Δ290-299t). Thus, the affinity data follow the same hierarchy as the blocking data. In each case, the higher affinity was due primarily to a faster kon rather than to a slower koff. Therefore, once the gDt-HveAt complex formed, its stability was unaffected by mutations in or near region IV. gD truncated at residue 234 bound to HveAt with a lower affinity (2.0 × 10−5 M) than did gD1(306t) due to a more rapid koff. These data suggest that residues between 234 and 275 are important for maintaining stability of the gDt-HveAt complex and that functional region IV is important for modulating the binding of gD to HveA. The binding properties of any gD1(234t)-receptor complex could account for the inability of this form of gDt to block HSV infection.  相似文献

To study drought stress effects on bound water, adsorption isotherms and pressure-volume curves were constructed for two durum wheat (Triticum durum Desf.) cultivars: Capeiti 8 (drought tolerant) and Creso (drought sensitive). Plants were grown under well-watered and water-stressed conditions in a controlled environment. Differential enthalpy (ΔH) was calculated through van't Hoff analysis of adsorption isotherms at 5 and 20°C, which allowed us to determine the strength of water binding. ΔH reached the most negative values at approximately 0.06 gram H2O/gram dry weight and then increased rapidly for well-watered plants (until 0.10 gram H2O/gram dry weight) or more slowly for drought-stressed plants (until 0.15-0.20 gram H2O/gram dry weight). Bound water values from pressure-volume curves were greater for water-stressed (0.17 gram H2O/gram dry weight) than for well-watered plants (0.09 gram H2O/gram dry weight). They may be estimates of leaf moisture content where ΔH reaches the less negative values and hence some free water appears. With respect to the well-watered plants, tightly bound water tended to be less bound during drought, and more free water was observed in cv Creso compared to cv Capeiti 8 at moisture contents >0.10 gram H2O/gram dry weight.  相似文献
