Maize roots (Zea mays, cv. DK 626) growing in aerated solutions showed striking variations in the amount of ethylene produced during different stages of development. As endogenous ethylene increases, root elongation decreases. Exogenous 1-aminocyclopropane-1-carboxylic acid (ACC) supplied to these roots also inhibited their elongation and increased both the fresh weight of the apex and the ethylene produced. The inhibitor of ethylene biosynthesis, 2-aminoethoxyvinyl glycine (AVG), and the inhibitor of ethylene action, silver thiosulfate (STS), also reduced growth and increased swelling. As growth diminishes at reduced ethylene concentrations or with impeded ethylene action, these results support the view that ethylene is necessary for root growth. As ACC treatment also inhibited root elongation, it appears that ethylene was inhibitory at both low and high concentrations. Whereas ACC stimulated ethylene production 4 h after the beginning of treatment, inhibition of root elongation and promotion of fresh weight advanced slowly and needed 24 h to be established. At that time, root elongation reached a maximum response of 60% inhibition and 50% increase in weight. At 48 h, higher doses of ACC were required to provoke the same response as at 24 h. The aim of the present work was to determine the factors limiting growth in mechanically impeded roots. Pea roots were grown in compressed and uncompressed sand cores, and then removed and transferred to hydroponics. Root elongation was slowed in impeded sand cores and did not recover to the unimpeded rates until 60 h after transfer to the hydroponics system. Root diameter was greater in impeded roots, and only after 36 h in hydroponics was new root tissue produced of the same diameter as the unimpeded controls. The turgor pressure of the growing cells was measured with a turgor probe and was the same in both treatments. The slower elongation rate of the previously impeded roots was, therefore, the result of axial tightening of the cell walls. Cell length profiles suggested that axial cell wall tightening persisted in the unrestricted hydroponics system. Production of new cells in unrestricted conditions was required before root elongation returned to the unimpeded state. Osmotic potential was decreased by approximately 0.2 MPa in previously impeded roots compared with the unimpeded ones. This corresponds to a decrease in water potential of 0.2 MPa. These data are discussed in relation to regulation of cell extension, solute unloading and the penetration of compacted soils by roots.  相似文献

【背景】土壤盐渍化已经成为日益严重的世界性问题,盐渍化不仅影响作物的产量,还会影响土壤的理化性质,抑制种子的萌发,阻碍植物正常生长,以及种子对水分和养分的吸收,进而影响作物的产量。【目的】玉米在盐渍土壤上生长受限,探究在中、高盐浓度下田菁种子内生菌与田菁胶混合浸种对玉米发芽的影响,为促进盐渍土玉米生长提供技术支持。【方法】利用LB液体培养基测定田菁种子内生菌贝莱斯芽孢杆菌ZH60的耐盐性;分别利用1%浓度田菁胶、OD600为0.8的ZH60菌悬液及两者混合液对玉米浸种3 h,自然风干后分别置于0、100和200 mmol/L NaCl的0.8%琼脂培养基上培养,测定玉米种子发芽势、发芽率、根长及芽长。将两叶一心期的玉米幼苗移至装有蛭石的花盆中培养,用荧光标记的内生菌ZH60灌根,分别于1、5、11、17、25 d取玉米根系研磨,利用平板菌落计数法测定内生菌在玉米根部的定殖量;利用激光共聚焦显微镜观察第28天ZH60在玉米根部的定殖情况。【结果】菌株ZH60耐11%的NaCl盐浓度,在中、高盐浓度下混合浸种的发芽势较对照组分别提高了28%、22%、30%;芽长提高了158%、163%、1...  相似文献

用黄腐酸和汪平酸浸种后,对于小麦种子萌发无明显影响。用不同浓度的黄腐酸和汪平酸溶液培养小麦幼苗,发现适当浓度时对于幼苗及根系生长与分化有促进作用,高浓度则表现为抑制作用。10ppm黄腐酸和50ppm汪平酸可显著增加去根绿豆下胚轴再生根数,而且在一定的浓度范围内,随着浓度的增高,根数不断增加而根长却逐渐减短。  相似文献

This study investigated the effects of increasing CaCO(3) concentrations (0, 5, 10, 20?mM) on arbuscular mycorrhizal (AM) symbiosis establishment as well as on chicory root growth and mineral nutrient uptake in a monoxenic system. Although CaCO(3) treatments significantly decreased root growth and altered the symbiosis-related development steps of the AM fungus Rhizophagus irregularis (germination, germination hypha elongation, root colonization rate, extraradical hyphal development, sporulation), the fungus was able to completely fulfill its life cycle. Even when root growth decreased more drastically in mycorrhizal roots than in non-mycorrhizal ones in the presence of high CaCO(3) levels, the AM symbiosis was found to be beneficial for root mineral uptake. Significant increases in P, N, Fe, Zn and Cu concentrations were recorded in the mycorrhizal roots. Whereas acid and alkaline phosphatase enzymatic activities remained constant in mycorrhizal roots, they were affected in non-mycorrhizal roots grown in the presence of CaCO(3) when compared with the control.  相似文献

通过室内发芽试验研究了卡那霉素对中华结缕草种子萌发及幼苗生长的影响。结果表明,卡那霉素处理对中华结缕草种子萌发影响不显著,而不同浓度及不同处理时间则对其幼苗生长影响显著。随着卡那霉素处理浓度增高,中华结缕草的株高、根系生长及侧根发生受抑制程度加重,白化率增大;随着处理天数的延长,株高、根系生长量降低。综合各项指标,确定卡那霉素最佳浓度为75 mg/L,最佳处理时间为6~10 d。  相似文献

在种子发芽箱中,以清水为对照,用5%、10%、15%、20%、25%、30%和35%共7个浓度聚乙二醇(PEG-6000)溶液浇灌栽培基质,观察文冠果种子的萌发和幼苗生长情况,以及幼苗叶片脯氨酸、丙二醛的含量和细胞膜相对透性的变化,探讨文冠果种子萌发对土壤水分的要求及其幼苗忍耐干旱的能力。结果显示:(1)在PEG浓度5%~35%范围内,文冠果种子发芽率、成苗率、幼苗根长和细胞膜透性均随着胁迫强度的增加呈明显的下降趋势;细胞膜透性随着胁迫强度的增加先升高后降低,但脯氨酸、丙二醛含量和幼苗根数量则呈现逐渐上升趋势。(2)5%的PEG浓度能够促进文冠果种子萌发,提高成苗率和根数量,降低幼苗死亡率;5%~10%的PEG能够明显促进苗高生长,但对根长影响不大;当PEG浓度高于15%后,种子萌发受到抑制,成苗率明显降低。(3)当PEG浓度高于25%时幼苗死亡率急剧上升,幼苗脯氨酸含量、丙二醛含量和细胞膜透性显著增加,细胞膜结构受到严重伤害。研究表明,低浓度PEG处理有利于文冠果种子萌发和幼苗生长,但过高浓度却对文冠果种子萌发和幼苗生长具有一定的抑制作用;文冠果种子萌发及其幼苗可忍受5%~25%的PEG渗透胁迫,即文冠果种子萌发和幼苗生长可忍受土壤水势为-7.94~-11.05MPa的干旱胁迫。  相似文献

BACKGROUND AND AIMS: Root axes elongate slowly and swell radially under mechanical impedance. However, temporal and spatial changes to impeded root apices have only been described qualitatively. This paper aims (a) to quantify morphological changes to root apices and (b) assess whether these changes pre-dispose young root tissues to hypoxia. METHODS: Lupin (Lupinus angustifolius) seedlings were grown into coarse sand that was pressurized through a diaphragm to generate mechanical impedance on growing root axes. In situ observations yielded growth rates and root response to hypoxia. Roots were then removed to assess morphology, cell lengths and local growth velocities. Oxygen uptake into excised segments was measured. KEY RESULTS: An applied pressure of 15 kPa slowed root extension by 75% after 10-20 h while the same axes thickened by about 50%. The most terminal 2-3 mm of axes did not respond morphologically to impedance, in spite of the slower flux of cells out of this region. The basal boundary of root extension encroached to within 4 mm of the apex (cf. 10 mm in unimpeded roots), while radial swelling extended 10 mm behind the apex in impeded roots. Oxygen demand by segments of these short, thick, impeded roots was significantly different from segments of unimpeded roots when the zones of elongation in each treatment were compared. Specifically, impeded roots consumed O2 faster and O2 consumption was more likely to be O2-limited over a substantial proportion of the elongation zone, making these roots more susceptible to O2 deficit. Impeded roots used more O2 per unit growth (measured as either unit of elongation or unit of volumetric expansion) than unimpeded roots. Extension of impeded roots in situ was O2-limited at sub-atmospheric O2 levels (21% O2), while unimpeded roots were only limited below 11% O2. CONCLUSIONS: The shift in the zone of extension towards the apex in impeded roots coincided with greater vulnerability to hypoxia even after soil was removed. Roots still encased in impeded soil are likely to suffer from marked O2 deficits.  相似文献

经嫁接黄瓜接穗的南瓜根系分泌物对黄瓜和南瓜的发芽率和胚根、胚轴的伸长均具有明显的抑制作用.分析表明:嫁接黄瓜根系分泌物可以促进黄瓜和南瓜体内吲哚乙酸氧化酶的活性,抑制淀粉酶的活性,从而降低其吲哚乙酸(IAA)水平,影响子叶中贮藏物质的转化和利用,抑制其萌发和生长.  相似文献

Two experiments were performed in simplified soil-less systems to study how roots respond to changes in mechanical impedance. In the first the increases in root force and diameter that occur when a pea root was impeded mechanically inside a hole with rigid conical walls were determined. The experiment was performed at 8°C and at 25°C, and the root growth pressures generated were calculated during periods of 12, 24, 48 and 72 hours. The maximum growth pressures generated were approximately the same at both temperatures, although the maximum pressure was achieved approximately twice as quickly at 25°C than at 8°C, being reached within 15–20 hours. In the second set of experiments a new technique was developed to measure simultaneously the elongation rate and the force exerted by the roots of seedlings grown in moist air. A constant force was exerted by a force transducer on a pea radicle using a system of pulleys, and the elongation rate of the pea root was monitored using a linear variable differential transformer (LVDT). The changes in root elongation rate were recorded that occurred in response to increases and decreases in the applied force. Root elongation rate decreased by more than 50% within 30 min of increasing the applied force by 100 mN. A similarly fast, but smaller increase in growth rate occurred when the force was removed. The interpretation of results from both studies will be discussed in terms of a modified form of the Lockhart model of growth.  相似文献

采用PEG模拟水分胁迫的方法,研究在5%PEG-6000胁迫下,不同浓度(0.025、0.05、0.1 mmol·L-1)茉莉酸甲酯(MeJA)对油菜种子萌发、幼苗生长、叶片氧化损伤和保护酶活性等生理生化特性的影响.结果显示:0.1mmol·L-1MeJA处理能显著缓解5%PEG-6000的胁迫伤害,使油菜种子发芽率、发芽势、发芽指数和活力指数分别显著提高62.3%、11.2%、55.2%、126.3%,主根长度降低45.0%,不定根数提高59.4%,但对株高无显著影响;同时叶片MDA含量降低20.6%,SOD、CAT、POD和APX活性分别提高100.0%、109.1%、452.6%和134.8%.研究表明,MeJA在一定程度上能够缓解渗透胁迫的抑制作用,提高SOD等保护酶的活性,缓解渗透胁迫造成的氧化损伤,有效促进渗透胁迫下油菜种子的萌发和幼苗生长.  相似文献

采用室内生物测定的方法,研究了不同砧木、不同浓度和不同生育期嫁接茄子根系分泌物的化感效应.结果表明,嫁接茄根系分泌物与自根茄相比,促进了茄子种子的萌发和幼苗生长.各砧木品种嫁接茄根系分泌物的化感效应显著或极显著高于对照,其中发芽率和苗高最高,增加29.1%和37.1%;不同浓度试验结果呈现“低促高抑”的规律,当浓度为0.04g·ml-1时发芽率最高,比对照增加50%,当浓度增加到0.24g·ml-1时,根长比对照降低了30.3%;在生长后期嫁接茄的根系分泌物对种子萌发的促进作用小于前中期,而自根茄抑制作用增强.因此,生产中使用嫁接技术是缓解由自毒作用引起的连作障碍的有效方法之一.  相似文献

Pea seedlings ( Pisum sativum L. cv. Weibull's Marma) were used to investigate the possible role of ethylene in light-induced inhibition of root elongation. Illumination of the roots with white light inhibited root elongation by 40–50% and increased ethylene production by the roots about 4-fold. Our main approach was to use exogenous 1-aminocyclopropane-1-carboxylic acid (ACC), supplied in the growth solution, to monitor ethylene production of the roots independent of light treatment. Ethylene production of excised root tips increased with increasing ACC concentrations. The rate of ethylene production in dark-grown roots treated with 0.1 μ M ACC was similar to that caused by illumination. Low ACC concentrations (0.01–0.1 μ M ) decreased the rate of root elongation, especially in seedlings grown in the dark, and 0.1 μ M ACC inhibited elongation to about the same extent as light. In light the roots curved and grew partly plagiogravitropically. This effect was also simulated by the 0.1 μ M ACC treatment. At 1 μ M and higher concentrations, ACC inhibited root growth almost completely and caused conspicuous curvatures of the root tips both in light and darkness. Inhibitors of ethylene synthesis and action partially counteracted the inhibition of root elongation caused by light. These observations suggest that the increase in ethylene production caused by light is at least partly responsible for the decreased growth of light-exposed roots.  相似文献

以发芽率、苗长、根长、苗干重、根干重变化为种子萌发和幼苗生长参数,研究苗期紫花苜蓿(Medicago sativa)植株浸提液对不同地区垂穗披碱草(Elymus nutans)种子萌发生长的化感作用。结果表明:地上部浸提液对LS、DX垂穗披碱草种子发芽率具有明显的促进作用,而对GS、KMX、LKZ、LZ地区垂穗披碱草种子发芽率均表现为抑制作用,其中对GS垂穗披碱草种子发芽率的抑制作用最强,在14.5%和5.5%浓度处理时抑制率分别为57.89%、55.26%;苗长方面,浸提液5.5%浓度对垂穗披碱草苗长的抑制率顺序为:LZNQDXLSKMXQHGSLKZ,14.5%处理时抑制率顺序为:LZKMXLKZNQGSQHDXLS,其中抑制率最高的为LZ垂穗披碱草,在14.5%和5.5%浓度处理时抑制率分别为33.03%、28.97%;根长方面,5.5%处理对垂穗披碱草根长的抑制率顺序为:QHNQLZKMX、GSLKZDX、LS,14.5%处理时抑制率顺序为:GSQH、NQLZLSKMXDXLKZ,其中在高浓度下抑制率最高的为GS垂穗披碱草,抑制率为57.69%;地上部浸提液对LKZ、LZ垂穗披碱草苗干重均具有促进作用,高浓度浸提液对NQ垂穗披碱草苗干重产生促进作用(RI0),而对KMX、DX垂穗披碱草苗干重均表现为抑制作用;根干重方面,浸提液对LS、QH、GS、NQ、LKZ垂穗披碱草根干重均有明显的抑制作用,而对KMX、DX垂穗披碱草根干重产生促进作用,高浓度浸提液对LZ垂穗披碱草的根干重产生促进作用。从根浸提液的作用来看,根浸提液除对LS、DX垂穗披碱草种子发芽率和根干重、GS垂穗披碱草种子发芽率和苗干重及NQ垂穗披碱草根干重具有促进作用外(P 0.05),对其余地区垂穗披碱草的各项指标均有明显的抑制作用(RI0)。所有以上结果表明,紫花苜蓿植株浸提液对垂穗披碱草种子萌发生长的作用具有一定的浓度效应。不同地区垂穗披碱草对紫花苜蓿地上部浸提液的敏感性趋势总体为:GSQHLSKMXNQLKZLZ,最不敏感或有促进作用的是DX垂穗披碱草;对根浸提液的敏感性趋势总体为:QHNQLZKMX,根浸提液对LS、GS、LKZ、DX垂穗披碱草种子萌发生长具有促进作用。紫花苜蓿植株不同部位浸提液对垂穗披碱草种子萌发生长的化感效应顺序为:地上部根。  相似文献

Effects of exogenous polyamines and difluoromethylornithine (DFMO) on seed germination and seedling root growth of Arabidopsis thaliana were investigated. Root growth was stimulated by low concentrations of putrescine but was increasingly inhibited by high concentrations of putrscine. DFMO inhibited root growth and this inhibition was reversed by applying putrescine. In contrast, both spermidine and spermine had no effect on root growth but inhibited seed germination. 金钟藤种子在室内萌发率很低,为进一步探讨金钟藤种子的特性,阐明其种子萌发率低的主要原因,对金钟藤种皮的透水性、种子解剖结构、种子活力和种子内源抑制物的生物测定进行了研究。结果表明:金钟藤种皮透水性较差,完整种子比破皮种子吸水达到最高水平慢38h;种子空瘪粒多,占所有种子的30%;种子活力较低,平均活力仅为35%;金钟藤种子甲醇粗提液对白菜种子萌发率、根长和芽长均有较强的抑制作用,其浸提液浓度在25mg/mL时,严重抑制白菜种子萌发和生长,即金钟藤种子内部含有较高的内源抑制物质。金钟藤种子萌发率低,表明其近年来突发性蔓延成灾主要不是由种子生成新个体造成的,导致其蔓延成灾的关键因素还需要进一步深入研究。  相似文献

Hoot elongation of rooted cuttings of aspen, Populus tremula L., was followed on moist filter paper. The time course of root growth during treatments which decrease or interrupt the supply of carbohydrates to the roots was studied. At light intensities below 4000 lux the rate of root elongation was found to berelated to the light intensity over the leaves. In the dark, root growth decreased within 24 hours and the roots stopped growing completely during the second or third day. Excision of the leaves or steam-killing of the tissue in a stem section below the leaves was followed by root growth stoppage within 24 hours. These results imply that the rate of root growth under certain conditions is determined by the supply of carbohydrates from the leaves.  相似文献
