人工孵育的鲎胚,以光镜和电镜观察其血细胞发生过程。血细胞包括变形细胞和蓝细胞二系列,均可分为原始、幼稚和成熟三阶段。血细胞生成开始于胚胎的附肢原基形成阶段,变形细胞最早见于胚区,蓝细胞先出现于胚外部分。鲎胚中未发现具备组织结构的造血器官,造血系在结缔组织和血腔中进行。  相似文献

We have developed a simple and reliable method of preserving antigen immunoreactivity with concomitant excellent retention of the cell ultrastructure. Using this method, we have been able to follow the origin and developmental stages of nuage accumulations within the nurse cell/oocyte syncytium in the ovary of the fruit fly, Drosophila melanogaster, at the ultrastructural level. We have found two morphologically and biochemically distinct forms of nuage material in the nurse cell cytoplasm: translocating accumulations of nuage containing the Vasa protein, termed sponge bodies and stationary polymorphic accumulations of nuage enriched in Argonaute and Survival of motor neuron proteins. Immunogold labeling combined with confocal fluorescent and ultrastructural analyses have revealed that the Vasa-containing nuage accumulations remain closely associated with the cisternae of the endoplasmic reticulum throughout their lifetimes. The migration mechanism of the Vasa-positive nuage appears distinct from the microtubule-dependent translocation of oskar ribonucleoprotein complexes. We postulate that these two distinct nuage translocation pathways converge in the formation of the polar granules within the polar/germ plasm of the oocyte posterior pole. We also provide morphological and immunocytochemical evidence that these polymorphic nuage accumulations correspond to the recently described cytoplasmic domains termed U body-P body complexes.  相似文献

This study investigated the correlations between the system treatment efficiencies and total nitrogen (TN) and total phosphorus (TP) accumulations of aboveground tissues of the wetland macrophytes of Hsin-Hai Bridge phase II constructed wetland. Among 19 emergent macrophytes studied, the optimal TN contents, 3.82% and 3.52% (w/w) were found for water spinach (Ipomoea aquatica) and Ludwigia x taiwanensis; while the optimal TP contents were found for the above two macrophytes at 0.64% and 0.83% (w/w). The accumulations of total plant TN and TP uptakes increased from 213 to 403 kg and 41 to 75 kg from March 2007 to the peak at September 2007, respectively. The TN ratios between plant tissue accumulations and the removals from the influents were 1.57%, 2.76%, 1.51% and 3.2% from March 2007 to March 2008. In the same period, the TP ratios between plant tissue accumulations and the removals from influents were 1.71%, 8.0%, 0.58% and 10.1%. The roles of the uptakes by aboveground portions of emergent macrophytes in system nutrient removals from the influents were more significant during growth seasons.  相似文献

Gaucher disease results from GBA1 mutations that lead to defective acid β-glucosidase (GCase) mediated cleavage of glucosylceramide (GC) and glucosylsphingosine as well as heterogeneous manifestations in the viscera and CNS. The mutation, tissue, and age-dependent accumulations of different GC species were characterized in mice with Gba1 missense mutations alone or in combination with isolated saposin C deficiency (C*). Gba1 heteroallelism for D409V and null alleles (9V/null) led to GC excesses primarily in the visceral tissues with preferential accumulations of lung GC24∶0, but not in liver, spleen, or brain. Age-dependent increases of different GC species were observed. The combined saposin C deficiency (C*) with V394L homozygosity (4L;C*) showed major GC18∶0 degradation defects in the brain, whereas the analogous mice with D409H homozygosity and C* (9H;C*) led to all GC species accumulating in visceral tissues. Glucosylsphingosine was poorly degraded in brain by V394L and D409H GCases and in visceral tissues by D409V GCase. The neonatal lethal N370S/N370S genotype had insignificant substrate accumulations in any tissue. These results demonstrate age, organ, and mutation-specific quantitative differences in GC species and glucosylsphingosine accumulations that can have influence in the tissue/regional expression of Gaucher disease phenotypes.  相似文献

Norepinephrine and serotonin augment by about 2-fold the accumulation of cyclic [3H]AMP elicited by 2-chloroadenosine in [3H]adenine-labeled guinea-pig cerebral cortical slices. Histamine causes a 3-fold augmentation. The first two agents have no effect on cyclic AMP alone, while histamine has only a small effect alone. The augmentation of the 2-chloroadenosine response appears to be mediated by α1-adrenergic, 5HT2-serotonergic and H2-histaminergic receptors. VIP-elicited accumulations of cyclic AMP are also augmented through stimulation of α1-adrenergic, 5HT2-serotonergic and H1-histaminergic receptors. Activation of these amine receptors also increases the turnover of phosphatidylinositols in [3H]inositol-labeled guinea pig cerebral cortical slices. Norepinephrine causes a 5-fold, serotonin a 1.2-fold, and histamine a 2.5-fold increase in accumulations of [3H]inositol phosphates. 2-Chloroadenosine, vasoactive intestinal peptide, baclofen, and somatostatin have no effect on phosphatidylinositol turnover, nor do the last two agents augment accumulations of cyclic AMP elicited by 2-chloroadenosine. The data suggest a possible relationship between turnover of phosphatidylinositol and the augmentations of the cyclic AMP accumulations elicited by biogenic amines in brain slices.  相似文献

Aminooxyacetate, a known inhibitor of transaminase reactions and glycine decarboxylase, promotes rapid depletion of the free pools of serine and aspartate in nitrate grown Lemna minor L. This compound markedly inhibits the methionine sulfoximine-induced accumulation of free ammonium ions and greatly restricts the methionine sulfoximine-induced depletion of amino acids such as glutamate, alanine, and asparagine. These results suggest that glutamate, alanine, and asparagine are normally catabolized to ammonia by transaminase-dependent pathways rather than via dehydrogenase or amidohydrolase reactions. Aminooxyacetate does not inhibit the methionine sulfoximine-induced irreversible deactivation of glutamine synthetase in vivo, indicating that these effects cannot be simply ascribed to inhibition of methionine sulfoximine uptake by amino-oxyacetate. This transaminase inhibitor promotes extensive accumulation of several amino acids including valine, leucine, isoleucine, alanine, glycine, threonine, proline, phenylalanine, lysine, and tyrosine. Since the aminooxyacetate induced accumulations of valine, leucine, and isoleucine are not inhibited by the branched-chain amino acid biosynthesis inhibitor, chlorsulfuron, these amino acid accumulations most probably involve protein turnover. Depletions of soluble protein bound amino acids are shown to be approximately stoichiometric with the free amino acid pool accumulations induced by aminooxyacetate. Aminooxyacetate is demonstrated to inhibit the chlorsulfuron-induced accumulation of α-amino-n-butyrate in L. minor, supporting the notion that this amino acid is derived from transamination of 2-oxobutyrate.  相似文献

Effect of penconazole (PEN) treatment on drought-stressed Mentha pulegium L. plants was investigated. Six weeks after sowing, seedlings were grown under soil moisture corresponding to 100, 75, 50 and 25 % field capacity (FC) with or without PEN (15 mg l−1) for 4 weeks. Results showed that the seedlings at 75 % FC showed maximum growth and water supply lower than 75 % FC was the threshold of drought-initiated negative effects on seedling growth. Drought stress significantly induced proline and carbohydrate contents and the decreased chlorophyll, photosynthesis parameters, soluble proteins and ion accumulations. Exogenous PEN increased the growth parameters, pigments, photosynthesis and ion accumulations in drought stressed and unstressed plants, but the effects of PEN were more significant under water deficit conditions. PEN also reduced the negative effects of drought by osmotic balance and protein accumulations. Electrophoretic patterns indicated that PEN treatment increased the intensity of some protein bands with the molecular weights of 30 kDa in shoot and 31 kDa in roots, and several new protein bands with the molecular masses between 116 and 14 kDa appeared in leaves, shoots and roots. These results suggest that the PEN application can be a useful tool in alleviation of effects of drought stress in M. pulegium plants.  相似文献

That infective juveniles of the nematode Neoaplectana carpocapsae accumulated specifically around certain host insect larvae was previously reported by us. In this work, the nematode's behavior was tested on defined chemical and bacterial gradients to determine whether these stimuli could cause the phenomenon. Nematode accumulations occurred around the peaks of certain gradients and, with NaCl, the initial accumulation rate was directly proportional to concentration up to 15 mM. Since the nematode did not respond to K-acetate, K and acetate salts were used to analyze responses to different ions. Maximal accumulations were observed with 60 mM Na+, 6 mM Mg2+, 0.75–7.5 mM Ca2+, and 6 mM CO32?. Accumulations to concentrations of Cl?, basic pH, and gram-negative bacteria were also observed. Acidic pH, 2.5, and 7.5 mM NH4+, repelled nematodes.  相似文献

Resveratrol production in Saccharomyces cerevisiae was compared to that in Escherichia coli. In both systems, 4-coumarate:coenzyme A ligase from tobacco and stilbene synthase from grapes were expressed. When p-coumaric acid was used as the precursor, resveratrol accumulations in the culture medium were observed to be comparable in E. coli (16 mg/liter) and yeast (6 mg/liter).  相似文献

To investigate the accumulation patterns of Cd and/or Pb in various body parts, organs and tissues of the Asian gypsy moth (Lymantria dispar) larvae under Cd and/or Pb stress, Cd and Pb treated artificial diets were used to feed the larvae in the current study. These larval body parts/organs/tissues included the heads, integuments (body walls), alimentary canals, fat bodies and hemolymphs. Our results showed that under Cd and/or Pb stress, their accumulations in larval body parts/organs/tissues were significantly higher than those in the control, with the amounts of tested metals in the fat bodies and hemolymphs and alimentary canals being significantly higher than those in the heads and integuments. Under the single Cd (0, 7.5, 10.5 mg/kg) or Pb stress (0, 55, 90, 125 mg/kg), the accumulations of these heavy metals were positively correlated with their concentrations in diets. Under the combined stress (Cd × Pb), the Cd accumulation at the lower Pb concentrations was higher than that at higher Pb concentrations for the body parts/organs/tissues, similar effects of Cd concentrations on Pb accumulations were also recorded. These results clearly showed that Cd and/or Pb were accumulated in various body parts/organs/tissues at different levels under the heavy metal stress. And accumulations of Pb/Cd were positively correlated with their concentrations in the diets under the single stress. Under the combined stresses, Cd and Pb had synergistic effects at low concentrations whereas antagonistic effects at high concentrations. The accumulations of Cd and/or Pb in the gypsy moth larvae affects normal physiological and biochemical functions, and thus affects their growth and development.  相似文献

The frontal ganglion of the tobacco hornworm, Manduca sexta (L.), was found to contain two neurosecretory (NS) cells (max dia = 40–45 μm). The cytoplasmic inclusions of the NS cells were stained purple with paraldehyde fuchsin, and marked fluctuations in amounts of NS material in the perikarya were observed, depending upon the developmental status of the insect. The perikarya of NS cells in the frontal ganglia of starved larvae and diapause pupae contained large accumulations of NS material, whereas feeding larvae and developing pharate adults showed relatively low amounts of neurosecretion. Electron microscopy revealed large accumulations of NS granules (dia = 80–240 nm) in the frontal ganglia of diapause pupae, but only slight accumulations of granules were seen in the NS cells of developing larvae and pharate adults.It was concluded that axonal transport and release but not synthesis is shut down during starvation and diapause, leading to accumulation of NS material in the perikarya. It is also suggested that the failure of many investigators to differentiate NS cells in the frontal ganglion of various insects may have been due to the selection of very active stages when the amount of available NS material was too low to be visualized by conventional staining techniques.  相似文献

A degree-day model was derived to predict egg hatch for Criconemella xenoplax. Eggs collected from gravid females were incubated in distilled water at constant temperatures of 10-35 C. Sixty-six percent of all eggs hatched between 13 and 32 C, and 42% hatched at 10 C. All eggs aborted above 32.5 C. Between 25 and 32 C, 8.5 ± 0.5 days were required for egg hatch. Degree-day requirement for egg hatch at 10-30 C was estimated to be 154 ± 5 with a base of 9.03 ± 0.04 C. This base of 9 C was adopted in studies of the relationship between degree-days and nematode population increase on Prunus seedlings grown 9-11 weeks in a greenhouse. Degree-day accumulations were based upon daily averages from maximum and minimum air temperatures. Ratios of final to initial population densities exhibited an exponential pattern in relation to degree-day accumulations with proportionate doubling increment of 0.100 ± 0.049 every 139 ± 8 degree-days. These results provide a means of predicting nematode population increase under greenhouse conditions and a basis for choosing sampling intervals when evaluating nematode multiplication.  相似文献

Northern blot hybridization analyzes revealed that poly(A+) RNAs homologous to eight heat shock (HS)-specific cDNA clones were induced by arsenite (As) or Cd treatments. The mRNAs accumulated slower, and maximum accumulations were consistently lower than HS-induced levels. Prolonged treatment with low concentrations (50-100 micromolar) of As for 6 hours, or Cd for 12 hours, resulted in decreased accumulations of HS-specific mRNAs. This response resembled the `autoregulation' observed during continuous 40°C HS. However, no autoregulation was evident when soybean seedlings were exposed to high concentrations of As (250 micromolar) or Cd (1 millimolar) for 12 hours. The cDNA probe pCE54 detected a second higher molecular weight poly(A+) RNA following As or Cd treatments which accumulated concomitantly with the lower molecular weight HS-specific poly(A+) RNA. The patterns of low molecular weight HS polypeptides from in vitro translations induced by HS, As, and Cd, and analyzed by one-dimensional and two-dimensional SDS-PAGE, were similar but temporal differences were apparent. In addition to HS proteins, many control proteins were also detected in both in vitro and in vivo labeling patterns from As and, to a lesser extent, Cd treatments. The chemical agents used in this study apparently induced the accumulation and translation of HS messages in vivo but not in the selective manner as observed during HS treatment.  相似文献

Fumigation of soybean leaves (Glycine max [L.] Merr. with ozone caused stippling and silvering at the same time that large accumulations of the isoflavonoid compounds daidzein, coumestrol, and sojagol occurred. Nitrogen dioxide and sulfur dioxide caused lesser accumulation of the isoflavonoids, and peroxyacetyl nitrate did not result in significant accumulation. Visible toxicity and chemical changes in ozone-fumigated leaves were similar to the hypersensitive disease defense reaction of soybean leaves to the pathogen Pseudomonas glycinea, except that the phytoalexin hydroxyphaseollin was not produced in the ozone-treated leaves.  相似文献

Quantitative estimates of time-averaging (age mixing) in gastropod shell accumulations from Quaternary (the late Pleistocene and Holocene) eolian deposits of Canary Islands were obtained by direct dating of individual gastropods obtained from exceptionally well-preserved dune and paleosol shell assemblages. A total of 203 shells of the gastropods Theba geminata and T. arinagae, representing 44 samples (= stratigraphic horizons) from 14 sections, were dated using amino acid (isoleucine) epimerization ratios calibrated with 12 radiocarbon dates. Most samples reveal a substantial variation in shell age that exceeds the error that could be generated by dating imprecision, with the mean within-sample shell age range of 6670 years and the mean standard deviation of 2920 years. Even the most conservative approach (Monte Carlo simulations with a non-sequential Bonferroni correction) indicates that at least 25% of samples must have undergone substantial time-averaging (e.g., age variations within those samples cannot be explained by dating imprecision alone). Samples vary in shell age structure, including both left-skewed (17 out of 44) and right-skewed distributions (26 out of 44) as well as age distributions with a highly variable kurtosis. Dispersion and shape of age distributions of samples do not show any notable correlation with the stratigraphic age of samples, suggesting that the structure and scale of temporal mixing is time invariant. The statistically significant multi-millennial time-averaging observed here is consistent with previous studies of shell accumulations from various depositional settings and reinforces the importance of dating numerous specimens per horizon in geochronological studies. Unlike in the case of marine samples, typified by right-skewed age distributions (attributed to an exponential-like shell loss from older age classes), many of the samples analyzed here displayed left-skewed distributions, suggestive of different dynamics of age mixing in marine versus terrestrial shell accumulations.  相似文献

The interpretation of accumulations of mammoth bones has been a long-term subject of archaeozoological studies. As the issue is typically approached from a taphonomic perspective, the broad spectrum of represented bones, especially the presence of large, non-meat bones, is taken as evidence of the local death of animals – either through natural causes or as a consequence of hunting. The number of mandibles and lower molars are used to calculate the number of hunted individuals, as they are the most frequently represented element. However, this approach faces numerous pitfalls. Interpretations, even contradictory, always neglect some important fact, especially associations with artefacts and the living floor. In our opinion, the use of ethnohistorical knowledge could mitigate, alleviate or eliminate some of the discrepancies. The fact that these enormous bone accumulations are typical only for the Gravettian (s. lato) underscores their culturally conditioned character. A traditional source of controversial interpretations is the well-known mammoth bone heap at Kraków – St. Bronislawa Hill (Spadzista Street B+B1). Until recently, all that was known from the classic settlement agglomeration of “mammoth hunters” near Dolní Věstonice was a “large accumulation of mammoth bones” studied by B. Klíma. However, based on unpublished terrain documentation, it has recently been possible to reconstruct several other accumulations directly in the base camp in the upper part of the Dolní Věstonice I site. The various phenomena with which the bone depositions there are associated provide new impulses for additional discussions.  相似文献

Microneedle perforations of the plasma membrane of Amoeba proteus were studied on the ultrastructural level. In each individual cell one hole was produced, which subsequently was marked with an eyelash left in place. Cells were quickly fixed, and sections cut parallel to the longitudinal axis of the eyelash. It was clear that the eyelash penetrated the plasma membrane, and that its free tip was located in the interior of the cell. A gap remained between the plasma membrane and the eyelash which may correspond to the electrical leak sometimes found by microelectrode punctures. The edges of the broken plasma membrane curled back into the cytoplasm. Here, a great redundancy of the plasma membrane was observed. Within these membrane accumulations, a large quantity of dense droplets was apposed at the inner leaflet of the plasma membrane. Their involvement in the formation and expansion of the plasma membrane in Amoeba proteus and Xenopus laevis has been suggested previously [1, 18]. Present studies offer more supportive evidence to that effect. Therefore, the interpretation seems to be plausible, that these membrane accumulations are the result of membrane expansion to minimize the hole produced during injury. This is in agreement with Holtfreter's [8] and Bluemink's [1] concept that the wound closure may occur by proliferation of the plasma membrane.  相似文献

Chemical analyses for the elementary compositions of the ashes of the fruit peels and trunks of the tropical plantain Musa paradisiaca have been undertaken. The elements, categorized as trace elements, generally are found to have higher mean concentrations in the fruit peels than in the trunks (except in the case of Zn). Their peel-trunk uptake ratios have been calculated and range between 1 and 4, showing normal levels of accumulations in the fruit peels over the trunks.  相似文献
