Results of digestibility trials on fresh and ensiled maize (aerial parts) were analysed and compared with laboratory data.

Digestibility of the organic matter of ears was almost constant at 84–85%. Digestibility of organic matter of stover showed great variation. Variation between animals in apparent digestibility of organic matter was greater for maize diets than for diets based on grass products. Most of the differences between animals were caused by differences in the digestion of cell walls. When the true digestibility of cell contents was assumed to be complete, endogenous excretion was, on average, only 7% with respect to organic matter intake, and seemed dependent on digestibility of organic matter.

Prediction of apparent digestibility was more accurate with digestibility in vitro than with crude fibre, although for maize silage, significant relationships were found between crude fibre content and digestibility of organic matter. Standard deviation of organic matter digestibility between lots of fresh and ensiled maize grown on commercial farms amounted to only about 1.2 –2%. As this was only twice the accuracy of digestibility in vivo with four animals, fixed values will be presented to farmers, possibly corrected for year, hybrid selection and cropping practice.  相似文献

In current climate change scenarios, mean air temperatures and summer droughts are expected to increase over the long-term average in large parts of Europe. These changes will strongly affect the growth and health of cultivated plants. In a field experiment in 2009 and 2010 in rain-out shelters, the consequences of plant water availability under three water regimes on the severity of Fusarium ear rot, deoxynivalenol (DON) contamination and yield of maize were investigated. Water was provided exclusively to the plants by a mobile sprinkler system installed in the rain-out shelter. Three maize cultivars were supplied with 50, 75, and 125% of the long-term average monthly precipitation of the experimental site. In 2009, Fusarium graminearum-infected oat kernels were placed on the soil surrounding maize plants to promote ear infection, whereas in 2010, the maize silks were directly inoculated with suspensions of F. graminearum conidia. Heavy drought stress in the 50% water regime was reflected in the average ear yield of the three maize cultivars of 75?dt ha-1 compared with 192?dt ha-1 at 125% water. In this comparison, the DON concentrations in the kernels were fivefold higher at 50% water than at the high water regime: 380 compared with 75???g DON kg-1 DM. In 2010, the drought stress symptoms were less pronounced than in 2009, and a much lower ear yield loss from 128 to 108?dt DM ha-1 was observed with decreasing water supply. The DON contamination of the kernels was at a higher level than in 2009, but a similar upward trend from 330???g?kg -1 DM at 125% water supply to about 3.5-fold higher DON levels at 75 and 50% water supply was observed. These different yield responses presumably resulted from variable climatic conditions at the experimental site in the 2?years. The results of this study suggest that the risk of DON contamination of maize kernels increases when plants are grown under conditions of long-term water deficit.  相似文献

A survey of aflatoxin contamination in selected Colombian foods was conducted over a 12-month period on a total of 248 samples. Samples were collected in supermarkets, retail stores and stock centres and were grouped into five categories: (1) corn and corn products, (2) cereal grains, (3) rice and rice products, (4) legume seeds; and (5) snacks and breakfast cereals. Aflatoxins were identified and quantitated using a liquid chromatographic technique with a limit of detection of 1 ng/g for each aflatoxin. Aflatoxins were detected in 14 of 109 samples of corn and corn products, 4 of 40 samples of rice and rice products, 2 of 30 samples of legume seeds, and 2 of 11 samples of snacks and breakfast cereals. None of the cereal grains samples analysed contained detectable levels of aflatoxins. Twelve of the total of 22 positive samples exceeded the maximum tolerable level of aflatoxin B1 adopted in most countries (5 ng/g); 10 of these 12 samples corresponded to corn and corn products. The results of the present study indicate that aflatoxin B1 contamination in certain foods in Colombia is a major public health concern. Continuous monitoring of aflatoxin B1 levels in Colombian foods is advised.  相似文献

Validation methods for chemometric models are presented, which are a necessity for the evaluation of model performance and prediction ability. Reference methods with known performance can be employed for comparison studies. Other validation methods include test set and cross validation, where some samples are set aside for testing purposes. The choice of the testing method mainly depends on the size of the original dataset. Test set validation is suitable for large datasets (>50), whereas cross validation is the best method for medium to small datasets (<50). In this study the K-nearest neighbour algorithm (KNN) was used as a reference method for the classification of contaminated and blank corn samples. A Partial least squares (PLS) regression model was evaluated using full cross validation. Mid-Infrared spectra were collected using the attenuated total reflection (ATR) technique and the fingerprint range (800–1800 cm−1) of 21 maize samples that were contaminated with 300 – 2600 μg/kg deoxynivalenol (DON) was investigated. Separation efficiency after principal component analysis/cluster analysis (PCA/CA) classification was 100%. Cross validation of the PLS model revealed a correlation coefficient of r=0.9926 with a root mean square error of calibration (RMSEC) of 95.01. Validation results gave an r=0.8111 and a root mean square error of cross validation (RMSECV) of 494.5 was calculated. No outliers were reported. Distribution of expansins in graviresponding maize roots   总被引:5,自引:0,他引:5  
To test if expansins, wall loosening proteins that disrupt binding between microfibrils and cell wall matrix, participate in the differential elongation of graviresponding roots, Zea mays L. cv. Merit roots were gravistimulated and used for immunolocalization with anti-expansin. Western blots showed cross-reaction with two proteins of maize, one of the same mass as cucumber expansin (29 kDa), the second slightly larger (32 kDa). Maize roots contained mainly the larger protein, but both were found in coleoptiles. The expansin distribution in cucumber roots and hypocotyls was similar to the distribution in maize. Roots showed stronger expansin signals on the expanding convex side than the concave flank as early as 30 min after gravistimulation. Treatment with brefeldin A, a vesicle transport inhibitor, or the auxin transport inhibitor, naphthylphthalamic acid, showed delayed graviresponse and the appearance of differential staining. Our results indicate that expansins may be transported and secreted to cell walls via vesicles and function in wall expansion.  相似文献

The relationship between deoxynivalenol (DON) intake and first morning urinary DON was examined in UK adults to validate the latter as a biomarker of human exposure. DON was assessed in first morning samples collected during a period of normal diet, a wheat-restriction intervention diet, and partial wheat-restriction intervention in which bread was allowed. During the partial intervention duplicate bread portions were collected for DON analysis. During the normal diet, partial intervention and full intervention, urinary DON was detected in 198/210 (geometric mean 10.1?ng DON mg?1 creatinine, 95% confidence interval (CI) 8.6–11.6?ng mg?1; range nd–70.7?ng mg?1), in 94/98 (5.9?ng mg?1, 95% CI 4.8–7.0?ng mg?1; range nd–28.4?ng mg?1), and 17/40 (0.5?ng mg?1, 95% CI 0.3–0.7?ng mg?1; range nd–3.3?ng mg?1) volunteers, respectively. A strong correlation between DON intake and the urinary biomarker was observed (p <0.001, adjusted r2?=?0.83) in models adjusting for age, sex and body mass index. These data demonstrate a quantitative correlation between DON exposure and urinary DON, and serve to validate the use of urinary DON as an exposure biomarker.  相似文献

Fluorescent in situ hybridization was used to examine the distribution of six abundant long terminal repeat (LTR) retroelements, Opie, Huck, Cinful-1, Prem-2/Ji, Grande, and Tekay/Prem-1 on maize pachytene chromosomes. Retroelement staining in euchromatin was remarkably uniform, even when we included the structurally polymorphic abnormal chromosome 10 (Ab10) in our analysis. This uniformity made it possible to use euchromatin as a control for quantitative staining intensity measurements in other regions of the genome. The data show that knobs, known to function as facultative neocentromeres when Ab10 is present, tend to exclude retroelements. A notable exception is Cinful-1, which accumulates in TR-1 knob arrays. Staining for each of the six retroelements was also substantially reduced in centromeric satellite arrays to an average of 30% of the staining in euchromatin. This contrasted with two previously described centromere-specific retrotransposable (CR) elements that were readily detected in centromeres. We suggest that retroelements are relatively rare in centromeres because they interrupt the long satellite arrays thought to be required for efficient centromere function. CR elements may have evolved mutualistic relationships with their plant hosts: they are known to interact with the kinetochore protein CENH3 and appear to accumulate in clusters, leaving long satellite arrays intact.  相似文献

Summary Three-way cross means were predicted with formulae involving linear functions of general (GCA) and specific combining ability (SCA) effects estimated from single-cross factorials between genetically divergent populations. Data from an experiment with 66 single-cross and 66 three-way cross forage maize (Zea mays L.) hybrids was used for comparing the prediction formulae. The genotypic correlation (r) between observed and predicted three-way crosses increased with increasing , the weighting factor of SCA effects, for plant height and ear dry matter (DM) content. It displayed slightly convex curves for total and stover DM yield, ear percentage, and metabolizable energy content of stover. For Jenkins' method B, r was considerably less than 1.0 for all traits, indicating the presence of epistasis. The square root of heritability (h) of the predicted means decreased with increasing , the reduction being small with a greater number of test environments. Using the product r·h as a criterion of efficiency, none of the prediction methods was consistently superior and the differences among them were rather small (< 7.5%) for all traits, irrespective of the number of test environments. We recommend evaluating the GCA of a greater number of lines from each parent population in testcrosses with a small number of elite lines from the opposite population. All possible three-way or double crosses between both sets of lines should be predicted by Jenkins's method C. The presence of mycotoxins in forage maize (zearalenone, fumonisin B1, T-2 toxin and diacetoxyscirpenol) and green coffee (ochratoxin A) from Nayarit State (Mexico) has been studied. All maize samples analyzed showed fumonisin B1 contamination, with an average concentration of 2,541 microg/kg. Fifteen percent of the samples contained zearalenone, with an average concentration of 1,610 microg/kg. Only one sample showed T-2 toxin contamination (7 microg/kg), and no diacetoxyscirpenol was detected. Sixty-seven per cent of green coffee samples were contaminated with ochratoxin A, with an average concentration of 30.1 microg/kg. This is the first study about mycotoxins developed in Nayarit and it has shown that mycotoxin contamination is a real problem in both foodstuffs studied.  相似文献

The objective of the performed investigations was to isolate pathogenic fungi from contaminated maize cobs, to assess the appearance of maize cob fusariosis and to determine grain contamination with deoxynivalenol in the cultivation of genetically modified maize containing a gene resistance against European corn borer (Ostrinia nubilalis Hbn) as well as selected non-modified cultivars. The plant material comprised the following genetically modified maize cultivar: DKC 3421 YG (MON 810) and non-modified cultivars obtained from Smolice Plant Breeding Ltd., IHAR Group: Junak (FAO 210-220), Prosna (FAO 220), SMH (FAO 230), Baca (FAO 220). Prior to harvesting, the occurrence of maize cob fusariosis was determined in the 89 (BBCH) developmental ripening stage. Microbiological assessment was carried out on grains selected from cobs characterized by various pathological symptoms. In 2008, a total of 133 isolates was obtained from the examined samples of infected maize plants, of which 51 isolates were species-identified, while in 2009, the total of 123 isolates were determined, of which 63 were species-identified. In both experimental years, the majority of isolates contained fungi from the Fusarium genus. The performed analysis of mean levels of cob contamination by fusarioses revealed that DKC 3421 YG (MON 810) and SMH (FAO 230) cultivars showed the smallest levels of contamination as well as the lowest percent of cob contamination per plant, while Junak (FAO 210-220) and Baca (FAO 220) cultivars were characterized by the highest degree of contamination. The lowest deoxynivalenol concentrations were determined in years 2008 and 2009 in the case of the DKC 3421 YG (MON 810) cultivar, whereas Prosna (FAO 220) cultivar was characterized by the highest deoxynivalenol concentration.  相似文献

The rapid decline in bumblebee populations within Europe has been linked to habitat loss through agricultural intensification, and a consequential reduction in the availability of preferred forage plants. The successful introduction of four European Bombus species to the South Island of New Zealand from England (in 1885 and 1906) provides an opportunity to determine how important different forage plants (also introduced from the U.K.) are to two severely threatened European bumblebee species (Bombus ruderatus and B. subterraneus). In January 2003 we conducted a survey of bumblebee populations across 70 sites in the central and southern South Island, recording which plant species were being used as pollen and nectar sources for each Bombus species. All four bumblebee species showed a clear preference for plants of European origin. Only B. terrestris, the most polylectic species, was recorded feeding on native plant species. The longer-tongued bumblebees, B. hortorum, B. ruderatus, and B. subterraneus, foraged predominantly on just two plant species; Trifolium pratense for both nectar and pollen, and Echium vulgare for nectar. These plant species are now declining in abundance in the U.K. Our results provide support for the hypothesis that the loss of flower-rich meadows, particularly those containing populations of Fabaceae species with long corollae, is responsible for the decline of bumblebee species across Europe. Comparison with earlier bumblebee surveys suggests that long-tongued bumblebees may also be in decline in New Zealand, particularly B. subterraneus which is now very localised and scarce.  相似文献

A selective analytical method based on high-performance liquid chromatography (HPLC), combined with atmospheric pressure chemical ionisation (APCI-) mass spectrometry (MS), has been developed for simultaneous determination of B-trichothecenes and the major metabolites of deoxynivalenol. The method allows simultaneous analysis of nivalenol (NIV), deoxynivalenol (DON), 15-acetyldeoxynivalenol (15-AcDON), 3-acetyldeoxynivalenol (3-AcDON), fusarenon X (Fus-X) and de-epoxydeoxynivalenol (DOM-1). The method is based on one-step sample clean-up using a multifunctional MycoSep column. A linear gradient mobile phase system, consisting of water:acetonitrile:methanol (H2O:ACN:MeOH) at a flow-rate of 1 ml/min, and a Polar-RP C18 column, were utilised to obtain the best resolution of all tested compounds along with column and equilibrating within 30 min. Dexamethasone (Dex) was used as internal standard. The developed method shows good repeatability for inter- and intra-day precisions as well as good linearity of calibration curves (r2 ranged from 0.9936 to 0.9998). Average recoveries for tested compounds in both matrices have been determined ranging from 63.7 to 102.3% and limit of quantification (LOQ) ranged from 25 to 150 ng/g. The utility and practical impact of the method is demonstrated using contaminated pig urine and maize samples.  相似文献

The analysis of deoxynivalenol (DON) in naturally infected wheat samples, after having been separated into four fractions through laboratory sieves, showed very low levels of DON in the fraction of largest kernels >2.8 mm (0 up to 1 mg/kg). The highest concentration of DON was found in fractions 2.2 to 2.5 mm and <2.2mm with up to 14mg/kg and 15mg/kg DON, respectively. In two samples (fractions <2.2mm) nivalenol was detected in concentrations up to 1,4mg/kg.  相似文献

The effects of aldicarb and benomyl on plant-parasitic nematodes, vesicular arbuscular mycorrhiza and the growth of forage maize were measured in 1980—1982 in two field experiments at Woburn, Bedfordshire and in a pot experiment using loamy sand soil from the field site. The most numerous migratory nematode, Tylenchorhynchus dubius increased three to four-fold during each season in untreated soil and was effectively controlled by aldicarb. Pratylenchus species were fewer but equally well controlled. The cereal cyst-nematode (Heterodera avenae), a serious maize pathogen in Northern France, was relatively scarce in untreated roots and was further decreased by aldicarb treatment; post-harvest H. avenae egg numbers were not affected by treatments; they declined equally because maize is such a poor host. Significant yield benefits (up to 37%) followed aldicarb treatment and were ascribed to nematode control in the absence of attribution to insect or other pests. Benomyl did not increase yields nor did it significantly affect the incidence of mycorrhiza. The results confirm that considerable losses of forage maize can be caused by nematodes in light soil and that aldicarb is an effective nematicide even at the rate of 1·7 kg a.i./ha.  相似文献

In leaves of maize (Zea mays) the activity of pyrophosphate:fructose 6-phosphate 1-phosphotransferase (PFP) is much less than that of ATP:fructose 6-phosphate 1-phosphotransferase. A sequential extraction technique was used to study the location of PFP in this tissue. When compared with enzymes known to be restricted to specific locations in maize, the distribution of PFP activity in the sequential extracts indicated that PFP is located predominantly, if not exclusively, in the mesophyll cytoplasm. Although confined to the same site as sucrose synthesis, the level of PFP activity is inadequate to contribute significantly to the gluconeogenic flux from fructose 1,6-bisphosphate to fructose 6-phosphate. The absence of PFP activity from the bundle-sheath demonstrates that this activity is not essential for glycolysis in higher plants.  相似文献

Biolayer interferometry allows for the real time monitoring of the interactions between molecules without the need for reagents with enzymatic, fluorescent, or radioactive labels. The technology is based upon the changes in interference pattern of light reflected from the surface of an optical fiber when materials bind to the tip of the fiber. The technique represents an alternative to technologies such as surface plasmon resonance, with an advantage in that the flow of extracts through small capillaries is not required. In this report, a deoxynivalenol-bovine serum albumin (DON-BSA) conjugate was non-covalently immobilized to the surface of aminopropylsilane sensors and the change in interference pattern resulting from the binding of DON-specific antibodies was measured. The basis for the assay was the competition between DON and the immobilized DON-BSA for binding to limited amounts of antibody. The technique was used to measure DON in extracts of spiked whole wheat flour, with a limit of detection of 0.10 mg DON/kg. Matrix interferences were an issue, and adequate quantification required using matrix-matched standards. When samples were tested with sensors that had not been conditioned to remove loosely attached DON-BSA, the recoveries at five spiking levels over the range from 0.2 to 5 mg/kg averaged 108.8% [relative standard deviation (RSD) 16.0%]. Using sensors that had been conditioned lowered the average recovery (101.4%) and improved the RSD (13.2%). This suggests that conditioning the sensors helped reduce a bias in the assay towards overestimation. These results, and the ease with which assays can be conducted, suggest further exploration of this technology for detection of mycotoxins is warranted.  相似文献

The objective of the present study was to demonstrate the efficiency of the decontamination process applied to deoxynivalenol (DON)-contaminated maize by sodium sulphite (Na2SO3) treatment in vivo. Additionally, in vitro characterisation of the toxicity of the DON sulphonates (DONS 1, 2 and 3 denote structurally different forms), the resulting DON metabolites, on peripheral blood mononuclear cells (PBMC) should substantiate the inactivation of DON. In a piglet experiment, both DON-contaminated maize and -uncontaminated control maize either untreated (DON?, CON?) or Na2SO3-treated (DON+, CON+) were mixed into feed and fed for 42 d starting from weaning. The results showed that feed intake and daily weight gain of animals fed DON? were significantly lower compared to animals fed CON? and CON+, whereas group DON+ reached the control level or even exceeded it. The feed-to-gain ratio was unaffected (p = 0.45). Furthermore, DON concentrations in plasma markedly reflected the diets’ DON concentrations. These were < 0.1, < 0.1, 5.4 and 0.8 mg/kg feed for CON?, CON+, DON? and DON+, and amounted to 0.3, 0.4, 33.0 and 9.3 ng/ml in plasma, respectively. Whereas DONS 2 and 3 were detected in the DON+ diet, only DONS 2 was recovered in plasma. Regarding the toxicity of DONS, no or much lower toxicity was found compared to DON. DONS 1 and Na2SO3 did not affect the viability of PBMC. At 32.71μM DONS2 the viability was reduced by 50% and thus this compound was less toxic than DON by a factor of 73. Consequently, wet preservation of maize with Na2SO3 was an effective tool to avoid the adverse effects of DON on performance of piglets.  相似文献
