When the model polynucleotide poly(dG-dC)?poly(dG-dC) [polyGC] is titrated with a strong acid (HCl) in unbuffered aqueous solutions containing the chlorides of the alkali metals in the concentration range 0.010?M-0.600?M, two transitions in the absorbance vs. pH plots are evidenced, characterized by the constants pK(a(?)) and pK(a(?)). The limiting values at infinite saline concentrations of these two constants, namely pK(∞)(a(?)) and pK(∞)(a(?)) obtained making use of the "one site saturation constant" equation or, in turn, of the double logarithmic plot: pK(a) vs. log([salt]?1), exhibit a clear dependence on the nature of the cations. The effects of the different alkali cations on the pK(∞)(a) values follow the Hofmeister series. In fact, the pK(∞)(a(?)) and the pK(∞)(a(?)) values are smaller for Li+ and Na+ than for Rb+ and Cs+, with K+ at the border between the two, showing that the transitions require higher concentrations of protons to occur in the presence of high concentrations of the cosmotropic ions.  相似文献

In three publications in 1883 and 1884 Carl Düsing of Jena gave a mathematical account of the influence of natural selection on the sex ratio based on the same argument that Darwin had advanced in The Descent of Man (1871). The argument thus became quite well known, being included in the principal books on the subject around the turn of the century, as well as in the Encyclopaedia Britannica, references to Düsing being given. By 1930, when Fisher gave a verbal account of the argument in The Genetical Theory of Natural Selection, he saw no need to give references, and no other book of the period treated the subject, as a result of which Düsing's contribution became lost to view. The dependence of the efficacy of the influence of a kava-pyrone (±)-kavain (330 µM) on the frequency of activation of Na+ channels and voltage dependence of the effects of (±)-kavain on the rate of inactivation of these channels were studied in experiments on isolated neurons from the rat hippocampus. In all series of experiments, the holding potential equalled –100 mV. The efficacy of (±)-kavain-induced blockade of Na+ channels was independent of the frequency of stimulation within the range up to 10/sec. In the control experiments, the rate of inactivation increased with the rise of depolarization from –40 mV to +30 mV, and then the saturation effect was observed. At the membrane potential of –40 mV, the rate of (±)-kavain-evoked inactivation increased approximately by a factor of 2.5. At the more positive shifts of the membrane potential, the efficacy of the effects of (±)-kavain on the rate of inactivation became noticeably reduced, and at +30 mV (±)-kavain exerted no distinct influence on this parameter.Neirofiziologiya/Neurophysiology, Vol. 28, No. 6, pp. 312–315, November–December, 1996.  相似文献

The article is devoted to the 100th anniversary of the outstanding world-renown Russian scientist Vladimir Alexandrovich Strunnikov, Academician, Professor, Head of Group of Developmental Cytology and Sex Regulation at Koltzov Institute of Developmental Biology and Head of genetic studies on silkworms in a number of sericulture institutes in CIS. Laureate of the State Prize of the USSR (1981), Hero of Socialist Labor (1990), awarded the I.I. Mechnikov Gold Medal, Academy of Sciences of the USSR (1981), founder of the theory explaining the origin of heterosis. One of his most significant achievements is the “Methods of Fixing the Heterosis Effect,” which makes it possible to abandon the production of hybrid seed and increase the yield of many crops by 20–50%. Fixing the gene complexes that determine the heterosis effect will become the “springboard” that will allow obtaining even more productive heterotic hybrids on the basis of new-generation varieties. The efficiency of this method in plant objects at the organismal and molecular levels was shown in the All-Russia Research Institute of Rice. A modification of this method reducing its laboriousness and increasing its efficiency was developed.  相似文献

Summary The follicle cells, nurse cells and germinal epithelia, which are closely associated with the oocyte of Agriolimax reticulatus (Müller) during its development in the ovotestis, have been studied using light and electron microscopy. The various secretory, digestive and phagocytic activities of these cells have also been investigated using electron cytochemical tests for oxidisable polysaccharide, acid phosphatase and electron-opaque tracer molecules. The oocyte lies initially between the germinal epithelia and a layer of nurse cells but, as oocyte vitellogenesis proceeds, it becomes encapsulated by a layer of follicle cells. Both the follicle and the nurse cells are active in secretion and digestion and contain Golgi apparatus, granular endoplasmic reticulum and acid phosphatase-rich digestive vacuoles. The significance of these activities is discussed in relation to oocyte vitellogenesis, secondary envelope formation and the digestion and recycling of cellular material.  相似文献

Summary The ovarian oocytes of Agriolimax reticulatus (Müller) have been studied by light and electron microscopy and electron cytochemistry. The development of the oocyte in the ovotestis may be divided into three stages.During Stage I the oocyte cytoplasm contains mainly ribosomes and also strands of endoplasmic reticulum, scattered mitochondria and Golgi systems. The nucleus contains both a paranucleolus and an eunucleolus. By Stage II the oocyte has enlarged, especially in a plane parallel to the basement membrane. In addition to the above mentioned organelles, the cytoplasm contains lipid, glycogen and early yolk platelets. During Stage III, the oocyte continues to enlarge, but mainly in a plane perpendicular to the basement membrane. A considerable degree of cytoplasmic differentiation has also taken place. The plasma membrane of the oocyte has become specialized with the appearance of a polysaccharide-rich glycocalyx, microvilli and pinocytotic tubules. Elsewhere, much of the background cytoplasm, containing Golgi-derived, polysaccharide and acid phosphatase-rich multivesiculate bodies, lipid and glycogen, is sequestered by smooth membranes and ultimately fuses with the growing yolk platelets. The nucleus contains an amphinucleolus, characteristic of many gastropods.The findings of this study are discussed in relation to results from other studies on oogenesis.  相似文献

The biological activity of human immune interferon depends on its carboxy-terminal structure. New genes coding for mature immune interferon molecules lacking 4, 5, 6, 8, 9, 10 and 11 amino acid residues were constructed, expressed in Escherichia coli and purified to homogeneity. Deletions with 14 and 18 carboxy-terminal amino acids were obtained by limited proteolysis of full-length immune interferon with mouse submaxillary gland ‘Arg-C’ protease and human plasmin, respectively. If a limited number of carboxy-terminal residues are removed, the antiviral and antiproliferative activities and the potential to activate macrophages is drastically enhanced. Maximal enhancement is found with the deletion of 9 or 10 residues. However, removal of 14 or more carboxy-terminal residues results in a sharp decrease of activity. We suggest that human immune interferon is synthesized as a preprotein and that its activity is modulated by proteolytic digestion.  相似文献

Myrmecia dispar (Clark) is found in the drier inland areas of New South Wales and southern Queensland in Australia. Its nests, which are situated adjacent to trees, have a distinctive structure of simple architecture. The composition of the colonies varies seasonally with little or no brood production during the colder months. Colony founding probably takes place in March, April, and/or May. Several myrmecophiles are found in nests ofM. dispar, and some of these are present in relatively large numbers.  相似文献

We isolated a temperature-sensitive mutant which did not exhibit derepression of acid phosphatase and invertase at the restrictive temperature. The mutation was mapped in the cryl gene, the structural gene for the catalytic subunit of adenylate cyclase. On electron microscopic observation of the mutant cells at the restrictive temperature, it was observed that they accumulated carbohydrate particles in the cytoplasm. Isolated particles had a rosette-like structure of 40 to 60 nm in diameter, and were identified as glycogen. After 2 hr incubation at the restrictive temperature, glycogen particles occupied most of the cytoplasmic space and the vacuoles were observed to be fragmented.  相似文献

The antimutagenic activity of protein-constituting amino acids except histidine on the mutagenicity of N-methyl-N′-nitro-N-nitrosoguanidine (MNNG) was investigated in vitro using Salmonella typhinurium TA-100 as an indicator bacterium (Ames test), and concentrations (IC50) of amino acids that inhibit 50% of the mutagenecity were measured. Cysteine was found to be most active and glycine, tryptophan, lysine, and arginine were strong antimutagenic amino acids. Other amino acids showed moderate or weak antimutagenic activities, depending on the amino acids. The results indicate that amino acids play a substantial role in chemoprevention of N-nitroso amine-induced mutagenicity.  相似文献

(3) one subsection, one series, eight species and one variety are described as new. Summary Acclimation to low salinity induces changes in the ultrastructure of the gill cells of the marine euryhaline amphipod, Gammarus oceanicus. The gills are composed of a single cell type. In 100% artificial sea water, these cells contain moderate numbers of mitochondria which are randomly distributed in the cytoplasm. The plasma membrane is extensively invaginated at the apical, lateral, and basal surfaces. Acclimation to 20% artificial sea water induces a further invagination at the apical cell membrane to form an elaborate apical labyrinth. The extracellular spaces between the folds in the basal cell membrane dilate to 1500 Å or more. Mitochondria are more abundant and in many cells they undergo a change in conformation. The mitochondria are crowded into thin leaflets of cytoplasm between the dilated basal invaginations or into the narrow space between apical and basal cell membranes. The effects of fenvalerate exposure on the net-spinning behaviour of Hydropsyche siltalai were examined in a laboratory study. The larvae were exposed to nominal pulse-doses of 0.25 and 0.5 μg fenvalerate l-1. Nets were collected and examined for anomalies after four days of exposure to fenvalerate. Additional nets were collected after another four day of exposure. The fenvalerate dissipated rapidly from the water column, and since only two doses of fenvalerate were given, the larvae were exposed to two pulse-doses of fenvalerate rather than to a constant concentration. In the 0.5 μg l-1-treatment the net-spinning behaviour was significantly affected, expressed as an increased mesh-opening and a decreased symmetry of the nets. No significant effects of fenvalerate exposure on the net-spinning behaviour were detected in the 0.25 μg l-1-treatment. Thus, with the conditions given in this experiment, exposure to fenvalerate starts to affect the net-spinning behaviour of Hydropsyche siltalai at a concentration between 0.25 and 0.5 μg l-1. The use of net-anomalies and Hydropsyche as bioindicators for monitoring pollutants in stream ecosystems are discussed.
