Physiological parameters such as viability, gross RNA synthesis,β-galactosidase induction, development of phages T4, T7 andλ have been studied in temperature-sensitiveEscherichia coli strains harbouring fit A76,fit A24 andfit A76fit A24 mutations in rpoB+ andrpoB240 genetic backgrounds. The efficiently of expression of these functions is influenced by thefit A alleles depending upon the medium of growth and/or temperature. Strains harbouring therpoB240 mutation and thefit A76 mutation, either alone or together with thefit A24 mutation, are rifampicin-sensitive even at the perfssive temperature. The wild-type mycelia of the fungusPhycomyces blakesleeanus are yellow because they contain small amounts of β-carotene. Some mutations lead to large increases in β-carotene content. These “deep yellow” mutants carry recessive mutations in either of two genes,carD andcarS, not linked to each other or to other genes related to carotenogenesis. ThecarS mutants contain up to 100 times more β-carotene than the wild type; thecarD mutants, up to 20 times.carS mutants are unable to form zygospores and their carotenogenesis is not activated by retinol; on the other hand,carD mutants complete the sexual cycle and respond to retinol.carS mutations are epistatic overcarD mutations. The product of genecarS mediates the end-product regulation of the pathway; it is suggested that thecarD gene product increases the amount or the activity of thecarS gene product.  相似文献

We have used a biological phenomenon that occurs inNeurospora crassa, termed Repeat-Induced Point mutation (RIP), to create partially functional mutant alleles of thealbino-3 (al-3) gene encoding geranylgeranyl pyrophosphate synthase, an enzyme involved in the biosynthesis of carotenoids and diverse prenylated compounds. A total of 70 RIP-inducedal- 3 mutants were identified by their pale albino phenotype, resulting from inactivation of carotenoid biosynthesis. Nucleotide sequence analysis of theal-3 gene in five of the RIP-induced mutants revealed that in each case RIP had introduced no more than six point mutations. The low frequency of RIP mutants (0.42%) and the isolation of only leaky mutants with very few mutations suggest that ascospores containing a heavily mutatedal-3 gene do not survive. These results are evidence that the RIP phenomenon, used to inactivate and silence duplicated genes inN. crassa, may be exploited in its mild version as a method of sequence-specific in vivo mutagenesis to obtain functional mutant alleles ofNeurospora genes. This mild form of mutagenesis may be particularly advantageous in selecting for leaky mutations in essentialNeurospora genes.  相似文献
