Changes in the antioxidant defense system and proline accumulation were examined at different growth stages (vegetative, boot and reproductive) in plants of two hexaploid spring wheat cultivars (S-24, salt tolerant; MH-97, salt sensitive), grown in hydroponics and salinity-affected with 0, 50, 100 and 150 mM of NaCl. Salt stress provoked a marked decline in plant dry mass and ascorbic acid contents, and increased proline, total soluble proteins and H2O2 contents in both wheat cultivars at all growth stages. However, higher accumulation of proline and H2O2 contents was recorded at the vegetative and boot stages, respectively, in both wheat cultivars. Salt stress caused a consistent rise in the activities of some key antioxidant enzymes (CAT, SOD, POD, and APX) at all growth stages only in the salt tolerant cultivar S-24, whereas such pattern of enhanced activities of enzymatic antioxidants in cv. MH-97 was found only at the vegetative stage under saline regimes. Maximum activities of various enzymatic antioxidants were observed at the vegetative stage in both wheat cultivars under varying external salt treatments. The results showed that high salinity tolerance of cv. S-24, as manifested by lower decrease in its dry matter under salt stress, was associated with higher activities of antioxidant enzymes, increased accumulation of proline, and lower levels of H2O2, as compared with cv. MH-97 at all growth stages under saline regimes.  相似文献

The study investigated the reduction in metalloid uptake at equimolar concentrations (~53.3 μM) of As(III) and As(V) in contrasting pair of rice seedlings by pretreating with H2O2 (1.0 μM) and SA (1.0 mM). Results obtained from the contrasting pair (arsenic tolerant vs. sensitive) of rice seedlings (cv. Pant Dhan 11 and MTU 7029, respectively) shows that pretreatment of H2O2 and H2O2 + SA reduces As(V) uptake significantly in both the cultivars, while no reduction in the As(III) uptake. The higher growth inhibition, higher H2O2 and TBARS content in sensitive cultivar against As(III) and As(V) treatments along with higher As accumulation (~1.2 mg g−1 dw) than in cv. P11, unravels the fundamental difference in the response between the sensitive and tolerant cultivar. In the H2O2 pretreated plants, the translocation of As increased in tolerant cultivar against AsIII, whereas, it decreased in sensitive cultivar both against AsIII and AsV. In both the cultivars translocation of Mn increased in the H2O2 pretreated plants against As(III), whereas, the translocation of Cu increased against As(V). In tolerant cultivar the translocation of Fe increased against As(V) with H2O2 pretreatment whereas, it decreased in the sensitive cultivar. In both the cultivars, Zn translocation increased against As(III) and decreased against As(V). The higher level of H2O2 and SOD (EC 1.15.1.1) activity in sensitive cultivar whereas, higher, APX (EC 1.11.1.11), GR (EC 1.6.4.2) and GST (EC 1.6.4.2) activity in tolerant cultivar, also demonstrated the differential anti-oxidative defence responses between the contrasting rice cultivars.

Salinity, as a serious and prevalent abiotic stress, causes widespread crop losses by restricting plant growth and production throughout the world. In this study, the biochemical and molecular responses of the pistachio (Pistacia vera L.) plant were studied under NaCl and salicylic acid (SA) treatments using hydroponically grown salt tolerant (Ghazvini) and salt sensitive (Sarakhs) pistachio cultivars. NaCl treatment (250 mM) increased the production of hydrogen peroxide (H2O2) and malondialdehyde (MDA) and the activity of antioxidant enzymes in both cultivars. In the sensitive cultivar, the H2O2 content was higher than the tolerant cultivar, especially in the roots. SA application to both salt-stress-treated cultivars resulted in an increase in photosynthetic pigment contents and antioxidant enzyme activity and a decrease in the H2O2 and MDA contents. After NaCl treatment, the isochorismate synthase (ICS) gene was upregulated in Ghazvini which leads to an increase in the SA content of the salt tolerant pistachio cultivar. In contrast, the salt treatment downregulated the expression of the ICS gene in Sarakhs. The ICS gene expression was positively regulated by SA treatment under the salt stress condition. Our results suggest that Ghazvini has higher salinity tolerance than Sarakhs due to its higher antioxidant capacity, photosynthetic pigment content, and the cultivar-specific expression pattern of the ICS gene. In this study, the potential alleviative effects of SA on the adverse effect of salt stress in P. vera (Pistacia vera) were also identified and highlighted.

Lime-induced iron chlorosis is a major nutritional disorder causing severe plant growth and yield reduction in the calcareous soils of Tunisia. The understanding the behavior of key metabolic functions of peas on calcareous soils, the identification of useful traits of tolerance, and the exploration of the genotypic differences in response to this constraint remain the most efficient approaches due to their coast, environmental benefits, and sustainability. For this purpose, a greenhouse experiment was conducted on three pea genotypes (Alexandra: Alex, Douce de provence: DP, and Merveille de Kelvedon: MK) cultivated on calcareous soil (Fe-deficient) and fertile soil (control). Plant growth, SPAD index, iron nutrition and distribution, photosynthesis, and antioxidant enzymes were deeply analyzed to discriminate genotypic differences. Calcareous-induced iron deficiency reduced SPAD index, plant growth, net photosynthesis, and tissue Fe content against a significant stimulation of the oxidative stress indicators, H2O2 and Malondialdehyde (MDA). Moreover, we reported a significant induction of SOD and CAT activity in shoots and roots of the Alexandra genotype. Fe use efficiency increased on calcareous soil and clearly discriminated the studied genotypes. Alexandra genotype was found to be the most tolerant to lime-induced iron chlorosis. This genotype protects its tissues against oxidative stress by stimulating enzyme activities (SOD and CAT) and develops significant efficiency of Fe uptake, translocation to shoots and use when cultivated on calcareous soil.  相似文献

  • Zinc (Zn) is an essential micronutrient for the growth and development of plants. However, Zn deficiency is a common abiotic stress causing yield loss in crop plants. This study elucidates the mechanisms of Zn deficiency tolerance in maize through physiological and molecular techniques.
  • Maize lines tolerant (PAC) and sensitive (DAC) to Zn deficiency were examined physiologically and by atomic absorption spectrometry (AAS). Proteins, H2O2, SOD, POD, membrane permeability and gene expression (using real‐time PCR) of roots and shoots of both maize lines were assessed.
  • Zn deficiency had no significant effect on root parameters compared with control plants in PAC and DAC but showed a substantial reduction in shoot parameters in DAC. AAS showed a significant decrease in Zn concentrations in both roots and shoots of DAC but not PAC under Zn deficiency, implying that Zn deficiency tolerance mechanisms exist in PAC. Consistently, total protein and membrane permeability were significantly reduced in DAC but not PAC in both roots and shoots under Zn deficiency in comparison with Zn‐sufficient plants. Real‐time PCR showed that expression of ZmZIP1, ZmZIP4 and ZmIRT1 transporter genes significantly increased in roots of PAC, but not in DAC due to Zn deficiency compared with controls. The H2O2 concentration dramatically increased in roots of DAC but not PAC. Moreover, tolerant PAC showed a significant increase in POD and SOD activity due to Zn deficiency, suggesting that POD‐ and SOD‐mediated antioxidant defence might provide tolerance, at least in part, under Zn deficiency in PAC.

The effects of Zn deficiency on antioxidant responses of two pea (Pisum sativum L.) genotypes, a Zn-efficient IPFD-99-13 and Zn-inefficient KPMR-500, grown in sand culture were studied. In the pea genotype KPMR-500, Zn deficiency decreased dry matter yield, tissue Zn concentration, and antioxidant enzyme activities istronger than in the genotype IPFD-99-13. Genotype IPFD-99-13 developed more efficient antioxidant system to scavenge ROS than genotype KPMR-500. Zinc deficiency produced oxidative damage to pea genotypes due to enhanced accumulation of TBARS and H2O2 and decreased activities of antioxidant enzymes (Cu/Zn superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX)). In the leaves of IPFD-99-13 genotype, the higher activity of ROS-scavenging enzyme, e.g., SOD, CAT, POD, and glutathione reductase, and antioxidants, such as ascorbate and non-protein thiols, led to the lower accumulation of H2O2 and lipid peroxides. These results suggest that, by maintaining an efficient antioxidant defense system, the IPFD-99-13 genotype shows a lower sensivity to Zn deficiency than the KPMR-500 genotype.  相似文献

Salicylic acid (SA) acts as an endogenous signal molecule responsible for inducing abiotic stress tolerance in plants. In this study, the role of SA in improving drought tolerance in two maize cultivars (Zea mays L.) differing in their tolerance to drought was evaluated. The plants were regularly watered per pot and grown until the grain filling stage (R2) under a rainout shelter. At stage R2, parts of the plants were treated with SA, after which drought stress was applied. Leaf samples were harvested on the 10th and 17th days of the drought. Some antioxidant enzyme activity, such as the superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), hydrogen peroxide (H2O2) and malondialdehyde (MDA) content, was measured during the drought period. Exogenous SA prevented water loss and delayed leaf rolling in comparison with control leaves in both cultivars. As a consequence of drought stress, lipid peroxidation, measured in terms of malondialdehyde content, was prevented by SA. SA pretreatment induced all antioxidant enzyme activities, and to a greater extent than the control leaves, during drought. SA also caused a reduction in the ascorbate (ASC) and glutathione (GSH) content in two maize cultivars. The H2O2 level was higher in SA pretreated plants than the controls in both cultivars. Pretreatment with SA further enhanced the activities of antioxidant enzymes and the concentrations of non-enzymatic antioxidants in the tolerant cultivar compared with the sensitive cultivar. Results suggested that exogenous SA could help reduce the adverse effects of drought stress and might have a key role in providing tolerance to stress by decreasing water loss and inducing the antioxidant system in plants with leaf rolling, an alternative drought protection mechanism.  相似文献

The effects of thiamin (Thi) applied as seed soaking or foliar spray on some key physiological parameters were investigated in two differentially salt-responsive maize (Zea mays L.) cultivars, DK 5783 and Apex 836 F1, exposed to saline stress in two different experiments. An initial experiment (germination experiment) was designed to identify appropriate doses of Thi which could lessen the deleterious effects of salt on plants and screen all available maize cultivars for their differential tolerance to salt stress (100 mM NaCl). The seeds of nine maize cultivars were soaked for 24 h in solutions containing six levels of Thi (25, 50, 75, 100, 125 and 150 mg l?1). Based on the results obtained from the germination experiment, maize cultivar DK 5783 was found to be the most salt tolerant and Apex 836 as the most sensitive cultivar. Also, of six Thi levels used, two levels (100 and 125 mg l?1) were chosen for subsequent studies. In the second experiment (glasshouse experiment), two maize cultivars, DK 5783 (salt tolerant) and Apex 836 (salt sensitive) were subjected to saline regime (100 mM NaCl) and two levels of Thi (100 and 125 mg l?1) applied as foliar spray. Salt stress markedly suppressed shoot and root dry mass, total chlorophylls (“a” + “b”), leaf water potential and maximum fluorescence yield (Fv/Fm) in the plants of both maize cultivars, but it increased proline accumulation, leaf osmotic pressure, malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations, electrolyte leakage (EL) as well as activities of some key antioxidant enzymes, superoxide dismutase (SOD; EC. 1.15.1.1), peroxidase (POD; EC. 1.11.1.7) and catalase (CAT; EC. 1.11.1.6). Salt-induced reduction in plant growth parameters was higher in the salt-sensitive cultivar, Apex 836, which was found to be associated with relatively increased EL, and MDA and H2O2 levels, and decreased activities of the key antioxidant enzymes. Application of Thi as seed soaking or foliar spray partly mitigated the deleterious effects of salinity on plants of both maize cultivars. The most promising effect of Thi on alleviation of adverse effects of salt stress on maize plants was found when it was applied as foliar spray at 100 mg l?1. Thiamin application considerably reduced tissue Na+ concentration, but improved those of N, P, Ca2+ and K+ in the salt-stressed maize plants. Exogenously applied thiamin-induced growth improvement in maize plants was found to be associated with reduced membrane permeability, MDA and H2O2 levels, and altered activities of some key antioxidant enzymes such as CAT, SOD and POD as well as increased photosynthetic pigment concentration under saline regime.  相似文献

Water deficit for rice is a worldwide concern, and to produce drought-tolerant varieties, it is essential to elucidate molecular mechanisms associated with water deficit tolerance. In the present study, we investigated the differential responses of nonenzymatic antioxidants ascorbate (AsA), glutathione (GSH), and their redox pool as well as activity levels of enzymes of ascorbate–glutathione cycle in seedlings of drought-sensitive rice (Oryza sativa L.) cv. Malviya-36 and drought-tolerant cv. Brown Gora subjected to water deficit treatment of ?1.0 and ?2.1 MPa for 24–72 h using PEG-6000 in sand cultures. Water deficit caused increased production of reactive oxygen species such as O2??, H2O2, and HO? in the tissues, and the level of production was higher in the sensitive than the tolerant cultivar. Water deficit caused reduction in AsA and GSH and decline in their redox ratios (AsA/DHA and GSH/GSSG) with lesser decline in tolerant than the sensitive seedlings. With progressive level of water deficit, the activities of monodehydroascorbate reductase, dehydroascorbate reductase, ascorbate peroxidase (APX), and glutathione transferase increased in the seedlings of both rice cultivars, but the increased activity levels were higher in the seedlings of drought-tolerant cv. Brown Gora compared to the sensitive cv. Malviya-36. Greater accumulation of proline was observed in stressed seedlings of tolerant than the sensitive cultivar. In-gel activity staining of APX revealed varying numbers of their isoforms and their differential expression in sensitive and tolerant seedlings under water deficit. Results suggest that an enhanced oxidative stress tolerance by a well-coordinated cellular redox state of ascorbate and glutathione in reduced forms and induction of antioxidant defense system by elevated activity levels of enzymes of ascorbate–glutathione cycle is associated with water deficit tolerance in rice.  相似文献

The purpose of this study was to examine the impact of low pH and Al stress on the apoplastic production of H2O2 and POD activity against guaiacol, ferulic acid, coniferyl alcohol, NADH and chlorogenic acid in the root tip (RT) of two cultivars of Lotus corniculatus and the model Lotus japonicus Gifu, with the goal to determine the possible role of POD activity in proton and Al tolerance. Our results suggest that Lotus corniculatus cv. UFRGS is more tolerant to low pH and Al than cv. Draco due to the high POD activities involved in CW strengthening. The enzymatic response of Lotus japonicus Gifu is similar rather to the sensitive cultivar. On the other hand, in cvs Draco and Gifu low pH induced the activation of CW-modifying PODs, which is probably a component of the defence response of roots to the presence of toxic protons. Aluminium did not activate further these activities suggesting that defence response and acclimation to low pH confers also effective defence against Al toxicity in Lotus species. The activity of NADH-POD and CGA-POD were not affected significantly by exposure to low pH or Al. Al and pH induced drop in H2O2 production, which was more relevant in cv. Gifu and Draco than in cv. UFRGS is probably associated with enhanced activity of peroxidases involved in secondary CW metabolism utilizing H2O2 as an electron acceptor.  相似文献

In order to better understand the role of cold acclimation in alleviating freezing injury, two barley cultivars with different cold tolerance, i.e. a sensitive cv. Chumai 1 and a tolerant cv. Mo 103, were used. The freezing treatment increased leaf soluble protein content more in the tolerant cultivar than in the sensitive one. Cold acclimation increased H2O2 content of the two cultivars during freezing treatment, especially in Mo 103. Glutathione and ascorbate contents during freezing and recovery were significantly higher in cold-acclimated plants than in non-acclimated ones. Activities of peroxidase, ascorbate peroxidase and glutathione reductase were also higher in cold-acclimated plants than non-acclimated plants during freezing treatment. However, there was no significant difference between cold-acclimated plants and the control plants in catalase activity. It may be assumed that cold acclimation induced H2O2 production, which in turn enhanced activities of antioxidative enzymes and synthesis of antioxidants, resulting in alleviation of oxidative stress caused by freezing.  相似文献

The aim of this work was to investigate whether Fe reduction and antioxidant mechanisms were expressed differently in five Prunus rootstocks (‘Peach seedling,’ ‘Barrier,’ ‘Cadaman,’ ‘Saint Julien 655/2’ and ‘GF-677’). These rootstocks differ in their tolerance to Fe deficiency when grown in the absence of Fe (−Fe) or in presence of bicarbonate (supplied as 5 or 10 mM NaHCO3). Fe deficiency conditions, especially bicarbonate, were shown to decrease Fe and total chlorophyll (CHL) concentration. In the (−Fe)-treated roots of all rootstocks and in the 5 mM NaHCO3-treated ones of the tolerant ‘GF-677’ the Fe(III)-chelate reductase (FCR) activity was stimulated. On the contrary, apart from the ‘GF-677,’ FCR activity was greatly inhibited by the 10 mM NaHCO3. From the results obtained with decapitated rootstocks, it is not entirely clear whether or not the presence of shoot apex was a prerequisite to induce FCR function in all rootstocks tested. In the leaves of rootstocks exposed to the (−Fe) treatment, superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities were enhanced whereas the levels of the non-enzymatic antioxidants (FRAP values) were increased in the Fe-deprived leaves, irrespective of the rootstock. Except for ‘Peach seedling,’ foliar SOD activity was stimulated by the presence of NaHCO3. Furthermore, POD activity was increased in the ‘Saint Julien 655/2’ and ‘GF-677,’ but was depressed in the ‘Barrier’ rootstocks exposed to 10 mM NaHCO3. As a result of 10 mM NaHCO3, the expression of a Cu/Zn-SOD and a POD isoform was diminished in the leaves of ‘Peach seedling’ and ‘Barrier,’ respectively. By contrast, an additional isoform of both POD and Mn–SOD were expressed in the leaves of ‘GF-677’ exposed to 10 mM NaHCO3 suggesting that the tolerance of rootstocks to Fe deficiency is associated with induction of an antioxidant defense mechanism. Although CAT activity was increased in the 5 mM NaHCO3-treated leaves of ‘GF-677,’ specifically the 10 mM NaHCO3 treatment resulted in a decrease of CAT activity and an accumulation of H2O2, indicating that bicarbonate-induced Fe deficiency may cause more severe oxidative stress in the rootstocks, than the absence of Fe. A general link between Fe deficiency-induced oxidative stress and Fe reduction-sensing mechanism is also discussed.  相似文献

Salinity is the second most prevalent abiotic stress faced by plants, and rice is not an exception. Through this study, it has been tried upon, to study the relative salinity tolerance of eight local varieties of North East India. Preliminary screening was based on their dose- and time-dependent physiological responses to salinity stress. Among the cultivars, Tampha was found to be relatively more tolerant, whereas MSE9 the most sensitive. To further ascertain their tolerance capacity, MDA and H2O2 content was determined, which also confirmed the tolerance level of the two cultivars. Histochemical assays for root plasma membrane integrity and leaf and root H2O2 and O2 ? content also showed more damage in Tampha in comparison to MSE9. Finally, gene expression analysis for Na+/K+ co-transporters, OsHKT2;1, OsHKT2;3 and OsHKT2;4, was performed to observe how the expression level of these transporters varies with the tolerance capacity of these two cultivars in leaves and roots under different time frames. The study reveals Tampha to be the most tolerant and MSE9 the most sensitive when compared to the other six screened cultivars for salinity stress.  相似文献

Salt sensitive (pea) and salt tolerant (barley) species were used to understand the physiological basis of differential salinity tolerance in crops. Pea plants were much more efficient in restoring otherwise depolarized membrane potential thereby effectively decreasing K+ efflux through depolarization‐activated outward rectifying potassium channels. At the same time, pea root apex was 10‐fold more sensitive to physiologically relevant H2O2 concentration and accumulated larger amounts of H2O2 under saline conditions. This resulted in a rapid loss of cell viability in the pea root apex. Barley plants rapidly loaded Na+ into the xylem; this increase was only transient, and xylem and leaf Na+ concentration remained at a steady level for weeks. On the contrary, pea plants restricted xylem Na+ loading during the first few days of treatment but failed to prevent shoot Na+ elevation in the long term. It is concluded that superior salinity tolerance of barley plants compared with pea is conferred by at least three different mechanisms: (1) efficient control of xylem Na+ loading; (2) efficient control of H2O2 accumulation and reduced sensitivity of non‐selective cation channels to H2O2 in the root apex; and (3) higher energy saving efficiency, with less ATP spent to maintain membrane potential under saline conditions.  相似文献

Forty-four genotypes from the rice germplasm were identified under photoinhibition/photooxidation and shade conditions and divided into four basic types: (1) cultivars tolerant to both photooxidation and shading, (2) cultivars tolerant to photooxidation but sensitive to shading, (3) cultivars tolerant to shading but sensitive to photooxidation, and (4) cultivars sensitive to both photooxidation and shading. Photosynthetic characteristics of a cultivar tolerant (cv. Wuyugeng 3) and a cultivar sensitive (cv. Xiangxian) to photooxidation and shading were compared. The photochemical efficiency (Fv/Fm) of photosystem 2 (PS2) and the content of PS2-D1 protein in the tolerant cultivar Wuyugeng 3 decreased less under photooxidative conditions as compared with Xiangxian. Under similar conditions, superoxide dismutase was induced rapidly to a higher activity and the active oxygen (O) built up to a lower level in Wuyugeng 3 than in Xiangxian. Net photosynthetic rate (P N) decreased by 23 % in Wuyugeng 3 vs. 64 % in Xiangxian. Shading (80 %) during the booting stage caused only small decreases (7–13 %) in ribulose-1,5-bisphosphate carboxylase activity and P N in Wuyugeng 3 but severe decreases (57–64 %) were observed in Xiangxian which corresponded to the decreases in grain yield of the two cultivars (38 and 73 %, respectively). We described a simple and effective screening method and physiological basis for breeding crops for enhanced tolerance to both high and low irradiance.  相似文献

Indices of oxidative stress viz., superoxide radical and H2O2 content increased in leaves of all the cultivars with the rise in salinity level, the increase was more pronounced and significant in salt-sensitive varieties and non-significant in resistant cultivars. Except for glutathione reductase (GR), basal activities of all other antioxidative enzymes viz. superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione reductase (GR) were significantly higher in leaves of all the resistant cultivars as compared to the sensitive ones. A differential response of salinity was observed on various enzymatic and non-enzymatic components of antioxidant system in leaves of salt-tolerant and salt-sensitive cultivars of rice (Oryza sativa L.). Activities of superoxide dismutase and glutathione reductase enhanced in all the tolerant cultivar while declined in the sensitive cultivars with increasing salinity from 0 to 100 mM. Salt-stress induced the activities of catalase and peroxidase in all the cultivars but the magnitude of increase was more pronounced in the sensitive cultivars than in the tolerant cultivars. Contrarily, APX activity increased in the salt-sensitive cultivars but showed no significant change in the salt-tolerant cultivars. The amount of ascorbic acid content, reduced glutathione (GSH), reduced/oxidized glutathione (GSSG) ratio was higher in leaves of the tolerant cultivars than that of the sensitive cultivars under saline conditions. It is inferred that leaves of salt-tolerant cultivars tend to attain greater capacity to perform reactions of antioxidative pathway under saline conditions to combat salinity-induced oxidative stress.  相似文献

Aluminium (Al) is the main factor that limits crop production in acidic soils. There is evidence that antioxidant enzymes such as superoxide dismutase (SOD) play a key role against Al‐induced oxidative stress in several plant species. Rye is one of the most Al‐tolerant cereals and exudes both citrate and malate from the roots in response to Al. The role of SOD against Al‐induced oxidative stress has not been studied in rye. Al accumulation, lipid peroxidation, H2O2 production and cell death were significantly higher in sensitive than in tolerant rye cultivars. Also, we characterised two genes for rye SOD: ScCu/ZnSOD and ScMnSOD. These genes were located on the chromosome arms of 2RS and 3RL, respectively, and their corresponding hypothetical proteins were putatively classified as cytosolic and mitochondrial, respectively. The phylogenetic relationships indicate that the two rye genes are orthologous to the corresponding genes of other Poaceae species. In addition, we studied Al‐induced changes in the expression profiles of mRNAs from ScCu/ZnSOD and ScMnSOD in the roots and leaves of tolerant Petkus and sensitive Riodeva rye. These genes are mainly expressed in roots in both ryes, their repression being induced by Al. The tolerant cultivar has more of both mRNAs than the sensitive line, indicating that they are probably involved in Al tolerance.  相似文献

Low temperature (LT) is one of the major factors that limit crop production and reduce yield. To better understand the cold-tolerance mechanism in the plantains, a sensitive cultivar Williams (Musa acuminata AAA cv. Williams) and a tolerant cultivar Cachaco (Musa paradisiaca ABB cv. Dajiao) were used. LT resulted in increased malondialdehyde (MDA) content, elevated contents of hydrogen peroxide (H2O2) and superoxide radical (O2·−), and decreased photochemical efficiency (Fv/Fm) and net photosynthetic rate (P N), but cv. Cachaco showed better LT tolerance than cv. Williams. After LT treatment for 120 h, total scavenging capability (DPPH· scavenging capability) in Williams showed a significant decrease but no significant alternations was found in Cachaco. Ascorbate peroxidase (APX) and peroxidase (POD) displayed a significant increase but superoxide dismutase (SOD) showed no significant alternations and catalase (CAT) showed a significant decrease in Cachaco after 120 h of LT treatment. All the four antioxidant enzymes above showed a significant decrease in Williams after 120 h of LT treatment. Our results suggest that higher activities of APX, POD, SOD, and DPPH· scavenging capability to a certain extent can be used to explain the higher cold tolerance in the plantain, which would provide a theoretical guidance for bananas production and screening cold-resistant variety.  相似文献

Salt-induced changes in the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), and lipid peroxidation in terms of malondialdehyde (MDA), level of H2O2, and some key metabolites such as soluble proteins, free proline and phenolics in the leaves of six radish cultivars (Radish Red Neck, Radish Lal Pari, Radish Mino Japani, Radish 40 Days, Mannu Early and Desi) were investigated. Varying levels of NaCl (0, 80 and 160 mM) applied for 40 days adversely affected the shoot fresh weight, chlorophyll contents and soluble proteins, while increased the levels of proline, and the activities of SOD, POD and CAT. However, leaf H2O2 and total phenolic contents were not affected by salt stress. Cultivars Mannu Early, Radish 40 Days and Desi were relatively higher in shoot fresh weight (percent of control) while cvs. Radish Mino Japani and Mannu Early in proline, and cvs. Radish 40 Days and Desi in total soluble proteins at 160 mM of NaCl. However, levels of H2O2 and phenolics were higher in cvs. Desi, Radish Lal Pari and Mannu Early and SOD, POD and CAT activities only in Radish Lal Pari and Mannu Early than the other cultivars under saline conditions. Overall, the differential salt tolerance of radish cultivars observed in the present study was not found to be associated with higher antioxidant enzyme activities and other key metabolites analyzed, so these attributes cannot be considered as selection criteria for salt tolerance in radish.  相似文献

Heavy metal (HM) contamination of the environment is a serious threat to sustainable crop production. Among the HMs, chromium (Cr) is one of the most toxic HMs that is known to negatively affect growth and metabolic activities of diverse crop plants. The present study was designed to investigate the ameliorative role of 5-aminolevulinic acid (ALA) under Cr stress in two maize (Zea mays L.) cultivars showing differential sensitivity to Cr tolerance. ALA is a biosynthesis precursor and it has a dominant regulatory effect related to physiological, respiratory, and photosynthesis processes in various plant species. Three concentrations of Cr (0, 5, and 10 mg kg−1) were tested under the graded levels of ALA application (0, 12.5, and 25 mg L−1). The results indicated that Cr stress differentially reduced plant growth attributes, gas exchange characteristics, photosynthetic pigments, and biomass in both the cultivars. Oxidative stress increased as evidenced in the form of electrolyte leakage, malondialdehyde, and hydrogen peroxide (H2O2) accumulation in plants. The anti-oxidative enzyme activities, that is, catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD) both in the leaves and roots of maize cultivars decreased due to Cr stress. The concentration of Cr increased in roots and shoots of maize under Cr levels without ALA. Under Cr stress, ALA exogenous application markedly enhanced plant growth, photosynthetic pigments, gas exchange capacity, and biomass. Furthermore, ALA application decreased the Cr-induced oxidative stress in maize cultivars by improving the activities of CAT, POD, and SOD in plants. After ALA application, the Cr concentrations and total Cr uptake by plants differently decreased in both cultivars. The 6103 cultivar of maize was found to be a tolerant cultivar against Cr stress due to its strong defensive system with a higher rate of antioxidant enzyme activities. On the other hand, the other maize cultivar (9108) was found to be a sensitive cultivar against Cr stress due to its weak defense system with higher contents of reactive oxygen species. These findings suggest that ALA can play a regulatory role in maintaining optimum plant growth and efficient photosynthetic processes under Cr-challenged habitats in maize. Thus, ALA application may be used as a sustainable remedial strategy to alleviate Cr-induced stress in maize cultivars.
