Prosopis strombulifera, a common legume in high-salinity soils of Argentina, is a useful model for elucidation of salt tolerance mechanisms and specific biochemical pathways in halophytes, since its NaCl tolerance exceeds the limit described for most halophytic plants. We analyzed the effects of the increasing concentration of two main soil salts, Na2SO4 and NaCl, on growth parameters of P. strombulifera, chlorophyll levels, and content of jasmonates (JAs) and polyamines (PAs), which are key molecules involved in stress responses. P. strombulifera showed a halophytic response (growth promotion) to NaCl, but strong growth inhibition by iso-osmotic solutions of Na2SO4. Chlorophyll levels, number of leaves and leaf area were also differentially affected. An important finding was the partial alleviation of SO42− toxicity by treatment with two-salt mixture. JAs are not directly involved in salt tolerance in this species since its levels decrease under all salt treatments. Beneficial effects of Putrescine (Put) accumulation in NaCl treated plants maybe inferred probably associated with the antioxidative defense system. Another novel finding is the accumulation of the uncommon PA cadaverine in roots under high Na2SO4, which may be related to SO42− toxicity.

Abstract

The role of abscisic acid (ABA) was analyzed in roots and leaves of the halophyte Prosopis strombulifera in response to low osmotic potential (Ψo: ?1.0, ?1.9, and ?2.6?MPa) induced by sodium chloride (NaCl), sodium sulfate (Na2SO4), and the iso-osmotic combination of both compounds (NaCl?+?Na2SO4). P. strombulifera plants were sprayed with ABA, as well as with inhibitors of ABA biosynthesis (sodium tungstate and fluridone). Different parameters were measured, including total plant height, leaf number, root length, root and shoot biomass, water content, transpiration rate, and total soluble carbohydrates, specific carbohydrates and ABA concentrations. Results showed that sodium salts affected growth parameters in varying ways, depending on the type of salts used as well as the osmotic potentials. ABA-sprayed plants displayed the lowest transpiration values. These plants had a higher content of total soluble carbohydrates in roots, greater root biomass and length and increased root/shoot rate. This study shows that ABA triggers different biochemical and physiological responses after the perception of a stressful condition, and that the interaction between different concentrations and types of salts, and the addition of ABA or its inhibitors generates responses that affect development and growth in the halophyte P. strombulifera.

Kochia sieversiana (Pall.) C. A. M., a naturally alkali-resistant halophyte, was chosen as the test organism for our research. The seedlings of K. sieversiana were treated with varying (0–400 mM) salt stress (1:1 molar ratio of NaCl to Na2SO4) and alkali stress (1:1 molar ratio of NaHCO3 to Na2CO3). The concentrations of various solutes in fresh shoots, including Na+, K+, Ca2+, Mg2+, Cl, SO42−, NO3, H2PO3, betaine, proline, soluble sugar (SS), and organic acid (OA), were determined. The water content (WC) of the shoots was calculated and the OA components were analyzed. Finally, the osmotic adjustment and ion balance traits in the shoots of K. sieversiana were explored. The results showed that the WC of K. sieversiana remained higher than 6 [g g−1 Dry weight (DW)] even under the highest salt or alkali stress. At salinity levels >240 mM, proline concentrations increased dramatically, with rising salinity. We proposed that this was not a simple response to osmotic stress. The concentrations of Na+ and K+ all increased with increasing salinity, which implies that there was no competitive inhibition for absorption of either in K. sieversiana. Based on our results, the osmotic adjustment feature of salt stress was similar to that of alkali stress in the shoots of K. sieversiana. The shared essential features were that the shoots maintained a state of high WC, OA, Na+, K+ and other inorganic ions, accumulated largely in the vacuoles, and betaine, accumulated in cytoplasm. On the other hand, the ionic balance mechanisms under both stresses were different. Under salt stress, K. sieversiana accumulated OA and inorganic ions to maintain the intracellular ionic equilibrium, with close to equal contributions of OA and inorganic ions to anion. However, under alkali stress, OA was the dominant factor in maintaining ionic equilibrium. The contribution of OA to anion was as high as 84.2%, and the contribution of inorganic anions to anion was only 15.8%. We found that the physiological responses of K. sieversiana to salt and alkali stresses were unique, and that mechanisms existed in it that were different from other naturally alkali-resistant gramineous plants, such as Aneurolepidium chinense, Puccinellia tenuiflora. Physiological and biochemical responses of Hordeum maritimum and H. vulgare to salt stress were studied over a 60‐h period. Growth at increasing salinity levels (0, 100, 200 and 300 mM NaCl) was assessed in hydroponic culture. H. maritimum was shown to be a true halophyte via its typical behaviour at high salinity. Shoot growth of cultivated barley was gradually reduced with increasing salinity, whereas that of wild barley was enhanced at 100 and 200 mm NaCl then slightly reduced at 300 mM NaCl. The higher salt tolerance of H. maritimum as compared to H. vulgare was due to its higher capacity to maintain cell turgor under severe salinity. Furthermore, H. maritimum exhibited fine regulation of Na+ transport from roots to shoots and, unlike H. vulgare, it accumulated less Na+ in shoots than in roots. In addition, H. maritimum can accumulate more Na+ than K+ in both roots and shoots without the appearance of toxicity symptoms, indicating that Na+ was well compartmentalized within cells and substituted K+ in osmotic adjustment. The higher degree of salt tolerance of H. maritimum is further demonstrated by its economic strategy: at moderate salt treatment (100 mm NaCl), it used inorganic solutes (such as Na+) for osmotic adjustment and kept organic solutes and a large part of the K+ for metabolic activities. Indeed, K+ use efficiency in H. maritimum was about twofold that in H. vulgare; the former started to use organic solutes as osmotica only at high salinity (200 and 300 mm NaCl). These results suggest that the differences in salt tolerance between H. maritimum and H. vulgare are partly due to (i) differences in control of Na+ transport from roots to shoots, and (ii) H. maritimum uses Na+ as an osmoticum instead of K+ and organic solutes. These factors are differently reflected in growth.

In order to identify physiological components that contribute to salinity tolerance, we compared the effects of Na+, Mg2+ and K+ salts (NaCl, Na2SO4, MgCl2, MgSO4, KCl and K2SO4), Ca2+ (CaSO4), mannitol and melibiose on the wild type and the single-gene NaCl-tolerant mutants stl1 and stl2 of Ceratopteris richardii. Compared with gametophytic growth of the wild type, stl2 showed a low level of tolerance that was restricted to Na+ salts and osmotic stress. stl2 exhibited high tolerance to both Na+ and Mg2+ salts, as well as to osmotic stress. In response to short-term exposure (3 d) to NaCl, accumulation of K+ and Na+ was similar in the wild type and stl1. In contrast, stl2 accumulated higher levels of K+ and lower levels of Na+. Ca2+ supplementation (1.0 mol m?3) ameliorated growth inhibition by Na+ and Mg2+ stress in wild type and stll, but not in stl2. In addition, under Na+ stress (175 mol m?3) wild-type, stll and stl2 gametopbytes maintained higher tissue levels of K+ and lower levels of Na+ when supplemented with Ca2+ (1.0 mol m?3). stl2 gametophytes were extremely sensitive to K+ supplementation. Growth of stl2 was greater than or equal to that of the wild type at trace concentrations of K+ but decreased substantially with increasing K+ concentration. Supplementation with K+ from 0 to 1.85 mol m?3 alleviated some of the inhibition by 75 mol m?3 NaCl in the wild type and in stl1. In stl2, growth at 75 mol m?3 NaCl was similar at 0 and 1.85 mol m?3 K+ supplementation. Although K+ supplementation above 1.85 mol m?3 did not alleviate inhibition of growth by Na+ in any genotype, stl2 maintained greater relative tolerance to NaCl at all K+ concentrations tested.

Salt‐affected farmlands are increasingly burdened by chlorides, carbonates, and sulfates of sodium, calcium, and magnesium. Intriguingly, the underlying physiological processes are studied almost always under NaCl stress. Two faba bean cultivars were subjected to low‐ and high‐salt treatments of NaCl, Na2SO4, and KCl. Assimilation rate and leaf water vapor conductance were reduced to approximately 25–30% without biomass reduction after 7 days salt stress, but this did not cause severe carbon shortage. The equimolar treatments of Na+, K+, and Cl? showed comparable accumulation patterns in leaves and roots, except for SO42? which did not accumulate. To gain a detailed understanding of the effects caused by the tested ion combinations, we performed nontargeted gas chromatography–mass spectrometry‐based metabolite profiling. Metabolic responses to various salts were in part highly linearly correlated, but only a few metabolite responses were common to all salts and in both cultivars. At high salt concentrations, only myo‐inositol, allantoin, and glycerophosphoglycerol were highly significantly increased in roots under all tested conditions. We discovered several metabolic responses that were preferentially associated with the presence of Na+, K+, or Cl?. For example, increases of leaf proline and decreases of leaf fumaric acid and malic acid were apparently associated with Cl? accumulation.

Plants of two wheat (Triticum aestivum L.) cultivars differing in salt tolerance were grown in sand with nutrient solutions. 35-d-old plants were subjected to 5 levels of salinity created by adding NaCl, CaCl2 and Na2SO4. Growth reduction caused by salinity was accompanied by increased Na+ and Cl- concentrations, Na+/K+ ratio, and decreased concentration of K+. The salt tolerant cv. Kharchia 65 showed better ionic regulation. Salinity up to 15.7 dS m-1 induced increased uptake of Na+ and Cl- but higher levels of salinity were not accompanied by further increase in uptake of these ions. Observed increases in Na+ and Cl- concentrations at higher salinities seemed to be the consequence of reduction in growth. Uptake of K+ was decreased; more in salt sensitive cultivar. This was also accompanied by differences in its distribution.

Using confocal microscopy, X‐ray microanalysis and the scanning ion‐selective electrode technique, we investigated the signalling of H2O2, cytosolic Ca2+ ([Ca2+]cyt) and the PM H+‐coupled transport system in K+/Na+ homeostasis control in NaCl‐stressed calluses of Populus euphratica. An obvious Na+/H+ antiport was seen in salinized cells; however, NaCl stress caused a net K+ efflux, because of the salt‐induced membrane depolarization. H2O2 levels, regulated upwards by salinity, contributed to ionic homeostasis, because H2O2 restrictions by DPI or DMTU caused enhanced K+ efflux and decreased Na+/H+ antiport activity. NaCl induced a net Ca2+ influx and a subsequent rise of [Ca2+]cyt, which is involved in H2O2‐mediated K+/Na+ homeostasis in salinized P. euphratica cells. When callus cells were pretreated with inhibitors of the Na+/H+ antiport system, the NaCl‐induced elevation of H2O2 and [Ca2+]cyt was correspondingly restricted, leading to a greater K+ efflux and a more pronounced reduction in Na+/H+ antiport activity. Results suggest that the PM H+‐coupled transport system mediates H+ translocation and triggers the stress signalling of H2O2 and Ca2+, which results in a K+/Na+ homeostasis via mediations of K+ channels and the Na+/H+ antiport system in the PM of NaCl‐stressed cells. Accordingly, a salt stress signalling pathway of P. euphratica cells is proposed.

Sorghum bicolor L. Moench, RS 610, was grown in liquid media salinized with NaCl, KCl, Na2SO4, K2SO4 or with variable mixtures of either NaCl/KCl or Na2SO4/K2SO4 at osmotic potentials ranging from 0 to -0.8 MPa. The purpose was to study the effects of different types and degrees of salinity in growth media on growth and solute accumulation. In 14-day-old plants the severity of leaf growth inhibition at any one level of osmotic potential in the medium increased according to the following order: NaCl < Na2SO4 < KCl = K2SO4. Inhibition of growth by mixtures of Na+ and K+ salts was the same as by K+ salts alone. Roots responded differently. Root growth was not affected by Na+ salts in the range of 0 to -0.2 MPa while it was stimulated by K+ salts. The major cation of leaves was K+ because S. bicolor is a Na+-excluder, while Na+ was the major cation in roots except at low Na+/K+ ratios in media. Anions increased in tissues linearly in relation to total monovalent cation, but not with a constant anion/cation ratio. This ratio increased as the cation concentrations in tissues increased. Sucrose in leaf tissue increased 75 fold in Chloride-plants (plants growing in media in which the only anion of the salinizing salts was Cl?) and 50 fold in Sulphate-plants (the only anion of the salinizing salts was SO42-). Proline increased 60 and 18 fold in Chloride- and Sulphate-plants, respectively, as growth media potentials decreased from 0 to -0.8 MPa. The concentrations of both sucrose and proline were directly proportional to the amount of total monovalent cation in the tissue. Sucrose concentrations began increasing when total monovalent cations exceeded 100 μmol (g fresh weight)?1 (the monovalent cation level in non-stressed plants), but proline did not start accumulating until monovalent cation concentrations exceeded 200 μmol (g fresh weight)?1. Therefore, sucrose seemed to be the solute used for osmotic adjustment under mild conditions of saline stress while proline was involved in osmotic adjustment under more severe conditions of stress. Concentrations of inorganic phosphate, glucose, fructose, total amino acids and malic acid fluctuated in both roots and leaves in patterns that could be somewhat correlated with saline stress and, sometimes, with particular salts in growth media. However, the changes measured were too small (at most a 2–3 fold increase) to be of importance in osmotic adjustment.

Summary Growth and physiological responses of date palm. Phoenix dactylifera L. cv. Barhee, callus to salinity stress were examined. Callus induced from shoot tips of offshoots was cultured on Murashige and Skoog medium supplemented with NaCl at concentrations ranging from 0 to 225 mM, in consective increments of 25 mM. Data obtained after 6 wk of exposure to salt have shown a significant increase in callus proliferation in response to 25 mM NaCl the lowest level tested, beyond which callus weight decreased. At 125 mM NaCl and higher, callus growth was nearly completely inhibited. Physiological studies on callus exposed to salt stress have shown an increase in proline accumulation in response to increased salinity. Proline accumulation was correlated to callus growth inhibition. Furthermore, increasing the concentration of NaCl in the culture medium generally resulted in a steady increase in Na+ and reduction in K+ concentrations. However, at 25 mM NaCl, the only level at which callus growth was significantly enhanced, an increase in K+ content was noted, in comparison to the NaCl free control. In response to increasing external NaCl level, the Na+/K+ ratio increased The Na+/K+ ratio was positively correlated to proline accumulation and hence callus growth inhibition. This study provides, an understanding of the response of date palm callus to salinity, which is important for future studies aimed at developing strategies for selecting and characterizing somaclonal variants tolerant to salt stress.

A group of sunflower lines that exhibit a range of leaf Na + concentrations under high salinity was used to explore whether the responses to the osmotic and ionic components of salinity can be distinguished in leaf expansion kinetics analysis. It was expected that at the initial stages of the salt treatment, leaf expansion kinetics changes would be dominated by responses to the osmotic component of salinity, and that later on, ion inclusion would impose further kinetics changes. It was also expected that differential leaf Na + accumulation would be reflected in specific changes in cell division and expansion rates. Plants of four sunflower lines were gradually treated with a relatively high (130 mm NaCl) salt treatment. Leaf expansion kinetics curves were compared in leaves that were formed before, during and after the initiation of the salt treatment. Leaf areas were smaller in salt‐treated plants, but the analysis of growth curves did not reveal differences that could be attributed to differential Na+ accumulation, since similar changes in leaf expansion kinetics were observed in lines with different magnitudes of salt accumulation. Nevertheless, in a high leaf Na+‐including line, cell divisions were affected earlier, resulting in leaves with proportionally fewer cells than in a Na+‐excluding line. A distinct change in leaf epidermal pavement shape caused by salinity is reported for the first time. Mature pavement cells in leaves of control plants exhibited typical lobed, jigsaw‐puzzle shape, whereas in treated plants, they tended to retain closer‐to‐circular shapes and a lower number of lobes.

The impact of salinity and water stress was analyzed in the xero-halophyte Atriplex hortensis using two varieties: green orach (A. hortensis var. purpurea) and red orach (A. hortensis var. rubra). A. hortensis L. is a C3 species well adapted to salt and drought conditions. To collect information on the physiological impact of different salt and water deficit levels on their water stress resistance, plants were exposed for 3 months to solution containing four levels of NaCl or to water stress regimes including four levels of field capacity. Osmotic potential at zero turgor Ψs0, osmotic potential at full turgor (Ψs100), relative water content (RWC), ion concentration (Na+, K+, Ca2+, Mg2+, and Cl), and malondialdehyde (MDA) were determined at the end of the treatment. The salinity and water stress induced a decrease in Ψs100, Ψs0, and RWC in both varieties, recorded changes being higher in plants of red variety than those of green variety. Both varieties specifically accumulated Na+ in response to drought and salt stress, suggesting that this element could play a physiological role in the stress response of this xero-halophyte species. In contrast, the presence of NaCl and water stress induced a decrease in K+, Ca2+, and Mg2+ concentration in both varieties. Salinity clearly induced an increase in Cl concentration in all tissues, but water stress had no impact on this parameter. MDA concentration increased in response to water stress and exogenous NaCl. Based on these findings the more drought-tolerant red orach may be grown in water-limiting soils.

Shoots of Thellungiella derived by micropropagation were used to estimate the plants'' salt tolerance and ability to regulate Na+ uptake. Two species with differing salt tolerances were studied: Thellungiella salsuginea (halophilla), which is less tolerant, and Thellungiella botschantzevii, which is more tolerant. Although the shoots of neither ecotype survived at 700 mM NaCl or 200 mM Na2SO4, micropropagated shoots of T. botschantzevii were more tolerant to Na2SO4 (10–100 mM) and NaCl (100–300 mM). In the absence of roots, Na2SO4 salinity reduced shoot growth more dramatically than NaCl salinity. Plantlets of both species were able to adapt to salt stress even when they did not form roots. First, there was no significant correlation between Na+ accumulation in shoots and Na+ concentration in the growth media. Second, K+ concentrations in the shoots exposed to different salt concentrations were maintained at equivalent levels to control plants grown in medium without NaCl or Na2SO4. These results suggest that isolated shoots of Thellungiella possess their own mechanisms for enabling salt tolerance, which contribute to salt tolerance in intact plants.Key words: Thellungiella salsuginea, Thellungiella botschantzevii, salt tolerance, isolated shoots, growth, rhizogenesis, ion accumulation  相似文献

Salinity remarkably inhibits NO3 - uptake but the mechanisms are not well understood. This study was addressed to elucidate the role of ionic and osmotic components of salinity on NO3 - influx and efflux employing classic kinetics involving a low affinity transport system (LATS) and a high affinity transport system (HATS). In the presence of KCl, NaCl, and Na2SO4 at 100 mM concentrations, in both LATS and HATS, Michaelis constant (Km) was similar for the three salts and maximum rate (Vmax) decreased as follows: KCl > NaCl > Na2SO4, compared to control indicating a non-competitive interaction with NO3 -. Unexpectedly, iso-osmotic solutions (osmotic potential Ψπ = -0.450) of polyethylene glycol (PEG, 17.84 %, v/v) and mannitol (100 mM) remarkably increased Km in both the LATS and the HATS, but Vmax did not change indicating a competitive inhibition. Under the PEG and mannitol treatments, Km and Vmax were higher than under the salt treatments. The salts increased slightly NO3 - efflux in the following order KCl > NaCl > Na2SO4. In contrast, mannitol strongly stimulated and the PEG inhibited NO3 - efflux. The obtained data reveal that salinity effects were not dependent on the anion type (Cl- versus SO4 2-) indicating a non-competitive inhibition mechanism between Cl- and NO3 -. In contrast, the cation types (K+ versus Na+) had a pronounced effect. The osmotic component is important to net NO3 - uptake affecting remarkably the influx in both LATS and HATS components of cowpea roots.

Glucose‐6‐phosphate dehydrogenase (G6PDH) is important for the activation of plant resistance to environmental stresses, and ion homeostasis is the physiological foundation for living cells. In this study, we investigated G6PDH roles in modulating ion homeostasis under salt stress in Carex moorcroftii callus. G6PDH activity increased to its maximum in 100 mM NaCl treatment and decreased with further increased NaCl concentrations. K+/Na+ ratio in 100 mM NaCl treatment did not exhibit significant difference compared with the control; however, in 300 mM NaCl treatment, it decreased. Low‐concentration NaCl (100 mM) stimulated plasma membrane (PM) H+‐ATPase and NADPH oxidase activities as well as Na+/H+ antiporter protein expression, whereas high‐concentration NaCl (300 mM) decreased their activity and expression. When G6PDH activity and expression were reduced by glycerol treatments, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein level and K+/Na+ ratio dramatically decreased. Simultaneously, NaCl‐induced hydrogen peroxide (H2O2) accumulation was abolished. Exogenous application of H2O2 increased G6PDH, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein expression and K+/Na+ ratio in the control and glycerol treatments. Diphenylene iodonium (DPI), the NADPH oxidase inhibitor, which counteracted NaCl‐induced H2O2 accumulation, decreased G6PDH, PM H+‐ATPase and NADPH oxidase activities, Na+/H+ antiporter protein level and K+/Na+ ratio. Western blot result showed that G6PDH expression was stimulated by NaCl and H2O2, and blocked by DPI. Taken together, G6PDH is involved in H2O2 accumulation under salt stress. H2O2, as a signal, upregulated PM H+‐ATPase activity and Na+/H+ antiporter protein level, which subsequently resulted in the enhanced K+/Na+ ratio. G6PDH played a central role in the process.

Basil (Ocimum basilicum L., cultivar Genovese) plants were grown in Hoagland solution with or without 50 mM NaCl or 25 mM Na2SO4. After 15 days of treatment, Na2SO4 slowed growth of plants as indicated by root, stem and leaf dry weight, root length, shoot height and leaf area, and the effects were major of those induced by NaCl. Photosynthetic response was decreased more by chloride salinity than by sulphate. No effects in both treatments on leaf chlorophyll content, maximal efficiency of PSII photochemistry (F v/F m) and electron transport rate (ETR) were recorded. Therefore, an excess of energy following the limitation to CO2 photoassimilation and a down regulation of PSII photochemistry was monitored under NaCl, which displays mechanisms that play a role in avoiding PSII photodamage able to dissipate this excess energy. Ionic composition (Na+, K+, Ca2+, and Mg2+) was affected to the same extent under both types of salinity, thus together with an increase in leaves Cl, and roots SO4 2− in NaCl and Na2SO4-treated plants, respectively, may have resulted in the observed growth retardation (for Na2SO4 treatment) and photosynthesis activity inhibition (for NaCl treatment), suggesting that those effects seem to have been due to the anionic component of the salts.

Salt Tolerance in Aquatic Macrophytes: Ionic Relation and Interaction
Effects of seawater salinity (SWS) and pure NaCl on the intracellular contents of Na+, K+, Mg2+, Ca2+, chlorophylls (Chl) and carotenoids (Car) were studied in three submerged aquatic macrophytes, Hydrilla verticillata, Najas indica and Najas gramenia, which differed in their tolerance to salinity. NaCl resulted in significant increase in Chl/Car ratio in the salt-sensitive H. verticillata and moderately salt-tolerant N. indica, but not in the salt-tolerant N. gramenia. SWS treatment did not result in any significant change in the ratio. The intracellular content of Na+ increased significantly in all the test plants upon exposure to both NaCl and SWS. The content of K+ decreased significantly in these plants upon salinity treatment, except in N. gramenia. The contents of Ca2+ and Mg2+ decreased significantly upon NaCl treatment and remained unchanged or increased upon SWS treatment. No relationship between salt tolerance and K+/Na+ ratio was observed. The maintenance of a minimal level of K+ was observed to be the most probable requirement of salt tolerance in aquatic macrophytes.
