Evidence suggesting that small molecular-weight precursors as well as polymers such as proteins and RNA, are not homogeneously distributed in the "soluble" phase of the cell, and that there may be, on one hand, hitherto unidentified barriers to normal diffusion within the cell and, on the other, channels through which certain molecular species may move in the cell sap much faster than their diffusion coefficients will permit, is discussed. Is green fluorescent protein toxic to the living cells?   总被引:27,自引:0,他引:27  
Green fluorescent protein (GFP) has become more popular to be used as a living marker for positively transfected clones in many studies. To establish stable cell lines constitutively expressing GFP, three GFPs expressed from plasmid pBIEGFP, pSG5GFP, and pRSGFP were introduced into NIH/3T3, BHK-21, Huh-7, and HepG2 cells. All the GFPs we used are the mutant forms of a common wild phenotype. The pBIEGFP expressed enhanced GFP (EGFP). The pRSGFP and pSG5GFP expressed red shift GFP (RSGFP). The RSGFP gene in pSG5GFP was driven by a strong SV40 promoter and showed at least 20-fold higher RSGFP expression by western blot analysis. Despite of the variation in the levels of GFP expression, many GFP expressing cells contracted, rounded-up, and died, which was confirmed by decreasing luciferase activity. CPP32 activity and flow cytometric analyses further demonstrate that cells expressing GFP underwent apoptosis. Our observation is contradictory to other reports that GFP is nontoxic to the cells. Most importantly, this paper shows for the first time the link between expression of GFP and induction of apoptosis. This finding should promote studies of GFP cytotoxicity and attempts to isolate new non-toxic mutants of GFP.  相似文献

The fission/division and fusion of mitochondria are fundamental aspects of mitochondrial biology. The balance of fission and fusion sets the length of mitochondria in cells to serve their physiological requirements. The fission of mitochondria is markedly induced in many disease states and in response to cellular injury, resulting in the fragmentation of mitochondria into dysfunctional units. The mechanism that drives fission is dependent on the dynamin related protein 1 (Drp1) GTPase. mdivi‐1 is a quinazolinone originally described as a selective inhibitor of Drp1, over other dynamin family members, and reported to inhibit mitochondrial fission. A recent study has challenged the activity of mdivi‐1 as an inhibitor of Drp1. This study raises serious issues regarding the interpretation of data addressing the effects of mdivi‐1 as reflective of the inhibition of Drp1 and thus fission. This commentary considers the evidence for and against mdivi‐1 as an inhibitor of Drp1 and presents the following considerations; (1) the activity of mdivi‐1 toward Drp1 GTPase activity requires further biochemical investigation, (2) as there is a large body of literature using mdivi‐1 in vitro with effects as predicted for inhibition of Drp1 and mitochondrial fission, reviewed herein, the evidence is in favor of mdivi‐1's originally described bioactivity, and (3) until the issue is resolved, experimental interpretations for the effects of mdivi‐1 on inhibition of fission in cell and tissue experiments warrants stringent positive controls directly addressing the effects of mdivi‐1 on fission. © 2017 Wiley Periodicals, Inc. In mammals, left-right symmetry is broken by a mechanically driven leftward flow of liquid at the embryonic node (nodal flow). Various models have emerged explaining how this may happen. Work from Tanaka and colleagues has provided a new mechanism by which nodal flow may be breaking symmetry. They describe small membrane-bound particles, which they term nodal vesicular parcels (NVPs), that are carried to the left side of the node. In the paper, they argue how signals carried within these parcels may break L-R symmetry.  相似文献

Purpose

The concept of electro mobility is gaining importance and has become more dynamic in recent years, particularly in developed economies. Besides a significant reduction of mobility-related CO2 emissions, electro mobility is also expected to minimize the current dependence on oil, while maximizing energy conversion efficiency. However, the associated shift in resource requirements towards so-called strategic metals gives reason to suspect that trade-offs could threaten the desired merits of e-mobility with regard to sustainability. This study aims to give a more comprehensive understanding of the challenges—including the issue of uncertainties—which the broad implementation of e-mobility could place on resource availability and especially on a sustainable management of special metals for the high voltage traction batteries forming the heart of the electric powertrain.

Methods

Future metal flows for three possible cathode materials containing the special metals lithium and cobalt are estimated in this paper by means of a Material Flow Analysis. Using two scenarios (dominant and pluralistic) projecting the annual demand for electric vehicles until 2050 and the free software STAN in order to perform the calculation steps to build up the model for the analysis, the MFA considers the resource input requirements based on annual vehicle registrations and the consequent energy requirements.

Results and discussion

The results indicate continuously rising lithium requirements with a wide variation in absolute terms depending on the scenario, which can be considered symptomatic for the uncertainty regarding the development of e-mobility. In the case of cobalt, the projected demand trajectories differ even more drastically between the two scenarios. In comparison to lithium though, for both scenarios cobalt requirements in absolute terms are much less than lithium requirements. With a view to currently known reserves, the cumulative demand for battery technology projected in the dominant scenario will consume 74–248 % (for two different cases) of the lithium reserves and 50 % of the cobalt reserves by 2050.

Conclusions

Despite significant differences between the examined scenarios, it becomes clear that e-mobility will be an increased driver for cobalt and particularly lithium demand in the future. Exact increases in demand for both metals are difficult to predict, especially due to the necessity of numerous assumptions, such as recycling rates, as well as data availability and quality. The results of this study imply a shift from managing primary resources, resource uses, and waste separately, towards managing materials, i.e., resource flows and their implications over the entire life cycle.

Nucleotide diversity of the human Y chromosome is much lower than that in the rest of the genome. A new hypothesis postulates that this invariance may result from mutations in maternally inherited mitochondrial DNA (mtDNA), leading to impaired reproduction among males and lowered male effective population size. If correct, we should expect to see low levels of polymorphism in the male-specific Y chromosome of many organisms but not necessarily in the female-specific W chromosome in organisms with female heterogamety. However, recent observations from birds suggest that the avian W chromosome is very low in nucleotide diversity. This indicates that mtDNA mutations cannot broadly explain the evolution of the sex-limited chromosome. Other work has suggested that sexual selection at loci involved in sex determination or secondary sexual characteristics might reduce levels of genetic variability on Y through hitch-hiking effects. Although the W chromosome does not seen to play a dominant role for sex determination in birds, it cannot be excluded that selective sweeps arising from natural or sexual selection contribute to the low levels of genetic variability seen on this chromosome.  相似文献

The allometric equation, y = axb, is commonly fitted to data indirectly by transforming predictor (x) and response (y) variables to logarithms, fitting a straight line to the transformations, and then back‐transforming (exponentiating) the resulting equation to the original arithmetic scale. Sometimes, however, transformation fails to linearize the observations, thereby giving rise to what has come to be known as non‐loglinear allometry. A smooth curve for observations displayed on a log–log plot is usually interpreted to mean that the scaling exponent in the allometric equation is a continuously changing function of body size, whereas a breakpoint between two (or more) linear segments on a log–log plot is typically taken to mean that the exponent changes abruptly, coincident with some important milestone in development. I applied simple graphical and statistical procedures in re‐analyses of three well‐known examples of non‐loglinear allometry, and showed in every instance that the relationship between predictor and response can be described in the original scale by simple functions with constant values for the exponent b. In no instance does the allometric exponent change during the course of development. Transformation of data to logarithms created new distributions that actually obscured the relationships between predictor and response variables in these investigations, and led to erroneous perceptions of growth. Through simulation, Whitlock showed that when all the alternatives have the same effect size, the weighted z-test is superior to both unweighted z-test and Fisher's method when combining P-values from independent studies. The present view is that the antigen-presenting cell (APC) processes and presents simultaneously on its surface several different antigens that are displayed randomly (with respect to their being Self or Nonself) as peptide-MHC complexes. The naive T-cell interacting with its ligand on the APC is activated by "co-stimulation," the first step on the pathway to effectors. This view ignores the requirement for associative recognition of antigen (ARA) in mediating both the Self-Nonself discrimination and the regulation of effector class. The introduction of ARA as a requirement for these two decision functions highlights a critical role for the effector T-helper (eTh) and necessitates rethinking the contribution of the APC.  相似文献

Encroachment of woody plants into grasslands is a global phenomenon that has substantial impacts on pastoral productivity and ecosystem services. Over the past half century, pastoralists and land management agencies have explored various options to control woody plants in order to improve ecosystem services in shrub‐encroached grasslands. We examined the effectiveness of controlling the encroachment of the shrub Caragana microphylla into grassland in Inner Mongolia, China. We cut and removed all of the aboveground biomass from 450 shrubs, predicting that the effectiveness of this technique to control shrubs would depend on shrub morphology. Specifically, we expected that larger shrubs with more biomass would be more difficult to kill by cutting than smaller shrubs. A year after treatment, we found that cutting killed only 11% of the 450 treated shrubs, and of these, three‐quarters of the locations that they occupied reverted to grasses and one‐quarter to bare soil. Shrubs that survived the cutting treatment produced more stems and leaf biomass, and therefore had a greater leaf to stem ratio. Shrubs that died after cutting had a lower crown area and basal area, and less stem biomass than shrubs that resprouted within 12 months of cutting. There were no effects of shrub height on the fate of treated shrubs. Cutting had no effect on understory plant cover or richness, but reproductive plants were taller under shrubs that were not cut. Overall, our study showed that removing aboveground shrub biomass by cutting is an ineffective technique for “restoring” the original grassland community unless shrubs are very small. Strategic targeting of small shrubs would be a more effective technique for controlling the spread of C. microphylla in the long term.  相似文献

To determine whether phospholipid is required for the activity of mammalian signal peptidase, the enzyme was partially purified from porcine pancreas and then extensively freed of phospholipid by SP-Sephadex C-50 chromatography. The delipidated enzyme showed signal peptidase activity, with a low concentration of detergent. Phospholipid was found to release the enzyme from the inhibition due to excess detergent.  相似文献

The silver nanoparticles (AgNPs) prepared by chemical reduction with sodium hypophosphite as a reducing agent and sodium hexametaphosphate as a stabilising agent were highly cytotoxic against human cells (U-937 and HL-60). The aim of the study was to determine the impact of selected antioxidants: ascorbic acid (AA), gallic acid (GA), scavenger (trolox (TX)) and Ag+ chelator (N-acetylcysteine, NAC) on viability, modulation of inflammatory response and apoptosis index of cells treated by AgNPs. Selected protectants added individually or together affects the viability of cells treated by AgNPs (1?mg/L). The mixtures assuring the most efficient defense against AgNPs were: AgNPs?+?TX?+?AA, AgNPs?+?GA?+?AA, AgNPs?+?TX?+?GA?+?AA and AgNPs?+?TX?+?GA?+?AA?+?NAC which synergistically interact in the mixture. The greatest reduction in IL-6 and TNF-α levels was found for the mixture containing AgNPs?+?TX?+?GA?+?AA. Mixture of this composition exhibited also the strongest anti-apoptotic effect. Highly cytotoxic AgNPs may not damage human cells if cytoprotectants are present.  相似文献

O'Connor, LM and Vozenilek, JA. Is it the athlete or the equipment? An analysis of the top swim performances from 1990 to 2010. J Strength Cond Res 25(12): 3239-3241, 2011-Forty-three world record swims were recorded at the 2009 Fédération Internationale de Natation (FINA) World Championship meet in Rome. Of the 20 FINA recognized long-course (50-m pool) swimming events, men set new world records in 15 of those events, whereas women did the same in 17 events. Each of the men's world records and 14 of the 17 women's records still stand. These performances were unprecedented; never before had these many world records been broken in such a short period of time. There was much speculation that full-body, polyurethane, technical swimsuits were the reason for the conspicuous improvement in world records. Further analysis led the FINA to institute new rules on January 1, 2010, that limited the types of technical swimsuits that could be worn by athletes. No long-course world record has been broken since then. We sought to understand this phenomenon by analyzing publicly available race data and exploring other possible causes including improvements in other sports, improvements in training science, changes in rules and regulations, gender differences, anaerobic vs. aerobic events, unique talent, and membership data.  相似文献
