Subcellular compartmentation of pyrophosphate (PP1) was determined by rapid membrane filtration of evaeuolated oat mesophyll protoplasts. By improving both the extraction procedure and its assay via bioluminescence, PP1 recovery from samples was quantitative and linear down to below 200 fmol. Based on the content of the different fractions obtained after membrane filtration and compared to the respective pools of marker metabolites [cytosol, fructose 2,6-bisphosphate (F26BP); chloroplast stroma, ribulose bisphosphate] rather than enzymes, we found ca 2/3 of the total cellular content to be chloroplast-assotiated. Referred to compartmental volumes, cytosolic and stromal concentrations of PP1 were nearly equal (70–100 μ M ). PP1 was higher in evacuolated compared to racuotated protoplasts which indicates a possible role of the tonoplast-located H+ pumping PP1ase in regulating the cellular pool size of PP1. During dark-light-transition the pool sizes of PP1 changed only marginally in both vacuolated and evacuolated protoplasts, while there were pronounced changes in those of F26BP, starch and sucrose. Thus our findings support the notion that the cellular pool size of PP1 is kept rather constant. They are, however, in contrast to the assumption that appreciable PP1 levels only exist in the cytosol.  相似文献

Amylase activity extracted from tulip ( Tulipa gesneriana L. cv. Apeldoorn) bulbs that had been stored for 6 weeks at 4°C was resolved to 3 peaks by anion-exchange chromatography on diethylaminoethyl-Sephacel. These 3 amylases exhibited different relative mobilities during non-denaturing polyacrylamide gel electrophoresis (PAGE). The most abundant amylase form (amylase I) was purified to apparent homogeneity using hydrophobic interaction chromatography, gel filtration and chromatofocusing. The apparent molecular mass of the purified amylase was estimated to be 51 kDa by sodium dodecyl sulfate-PAGE and 45 kDa by gel filtration chromatography. The purified amylase was determined to be an endoamylase (EC 3.2.1.1) based on substrate specificity and end-product analysis. The enzyme had a pH optimum of 6.0 and a temperature optimum of 55°C. The apparent Km value with soluble starch (potato) was 1.28 mg ml−1. The presence of Ca2+ increased the activity and thermal stability of the enzyme. The presence of dithiothreitol enhanced the activity, while β -mercaptoethanol and reduced glutathione had no significant effect. When pre-incubated in the absence of the substrate, N-ethylmaleimide and 5,5'-dithiobis-(2-nitrobenzoic acid) partially inhibited the enzyme. α -cyclodextrins or β -cyclodextrins had no effect on enzyme activity up to 10 m M . In addition to CaCl2, CoCl2 slightly enhanced activity, while MgCl2 and MnCl2 had no significant effect at a concentration of 2 m M . ZnCl2, CuSO4, AgNO3 and EDTA partially inhibited enzyme activity, while AgNO3 and HgCl2 completely inhibited it at 2.0 m M .  相似文献

The intracellular levels of orthophosphate (P1), pyrophosphate (PP1) and short- and long-chain polyphosphate (Poly P) were measured in Leishmania major promastigotes incubated in a phosphate-free medium. In the absence of exogenous substrate, the levels of both P1 and PP1 increased during a 1 h incubation. The increase in both P1 and PP1 was prevented when glucose was present, but glycerol prevented the rise in P1 only. A rise in P1 and PP1 was also seen in cells incubated in the absence of exogenous substrate under anaerobic conditions. This was reversed upon addition of glucose plus oxygen. Polyphosphate, here shown to be present in L. major , was measured by means of a polyphosphate glucokinase assay. Short-chain Poly P content did not differ between cells incubated for 1 h in the absence of exogenous substrate or in the presence of glucose or glycerol. Long-chain Poly P content, however, was lower in cells incubated without glucose than in cells incubated with glucose and was also lower in cells incubated for 1 h with glycerol as compared with freshly washed cells. Up to 61% of the increase in P1 and PP1 that occurred in promastigotes incubated in the absence of exogenous substrate could have arisen from the concomitant decrease in long-chain Poly P.  相似文献

Among 30 plant species examined, the PPi-phosphofructokinase (EC 2.7.1.90) was found in leaves of 21 plants. Some of the plants exhibit no activity of ATP-dependent phosphofructokinase but display only activity of PPi-phosphofructokinase. A partly purified preparation of PPi-phosphofructokinase with specific activity of 8.4 Hmol (mg protein)−1 min−1 was obtained from Sanseviera trifasciata leaves. The enzyme is restricted to the cytoplasm, it exhibits pronounced substrate specifity, requires Mg2+ ions, is inhibited by AMP, PEP, methylenediphosphonate and stabilized by mercaptoethanol. At pH 7.8 with 1.5 m M MgCl2 the following KM values were observed: pyrophosphate, 0.58 m M ; fructose 6-phosphate, 0.8 m M . The KM values for substrates of reverse reaction (pH 7.3; 2 m M MgCl2) are of the same order of magnitude: 0.83 m M for fructose 1,6-diphosphate, and 0.14 m M for orthophosphate. The molecular weight of the studied enzyme is about 125 000 dalton as estimated by gel filtration.  相似文献

Konno, H., Yamasaki, Y. and Katoh, K. 1987. Purification of an α-L-arabinofurano-sidase from carrot cell cultures and its involvement in arabinose-rich polymer degradation.
An α-L-arabinofuranosidase (α-L-arabinofuranoside arabinofuranohydrolase, EC 3.2.1.55) was isolated from a homogenate of cell suspension cultures of carrot ( Daucus carota L. cv. Kintoki). The buffer-soluble enzyme was purified to homogeneity by a procedure involving ammonium sulfate fractionation, chromatography on DEAE-Sephadex A-50, Sephadex G-150, Con A-Sepharose 4B and CM-Sephadex C-50, and preparative polyacrylamide gel electrophoresis. The size of this enzyme as determined by polyacrylamide gel electrophoresis in the presence of sodium laurylsulfate and by Sephadex G-200 gel filtration was 94 and 110 kDa, respectively. The isoelectric point was at pH 4.7. The Km and Vmax values for p-nitrophenyl α-L-arabinofuranoside were 1.33 mM and 20.2 μimol (mg protein)-1 h-1, respectively. The optimal activity occurred at pH 4.2 with Mcllvaine buffer. The enzyme was stimulated by Ca2+ and Zn2+, whereas it was strongly inhibited by Cu2+, Ag2+, Hg2+, p-chloromercuri-benzoate and L-arabono-l,4-lactone. The enzyme acted on beet arabinan in an exo-fashion. Furthermore, the enzyme was partially involved in the hydrolysis of the ara-binogalactan and pectic polymer purified from carrot cell walls.  相似文献

The lactic acid bacterium, Leuconostoc mesenteroides, when grown on an arbutin-containing medium, was found to produce an intracellular β-glucosidase. The enzyme was purified by chromatofocusing, ion-exchange chromatography and gel filtration. The molecular mass of the purified intracellular β-glucosidase, as estimated by gel filtration, was 360 kDa. The tetrameric structure of the β-glucosidase was determined following treatment of the purified enzyme with dodecyl sulphate (SDS). The intracellular β-glucosidase exhibited optimum catalytic activity at 50°C and pH 6 with citrate–phosphate buffer, and 5·5 with phosphate buffer. The enzyme was active against glycosides with (1→4)-β, (1→4)-α and (1→6)-α linkage configuration. From Lineweaver–Burk plots, K m values of 0·07 mmol l−1 and 3·7 mmol l−1 were found for p -nitrophenyl-β- D -glucopyranoside and linamarin, respectively. The β-glucosidase was competitively inhibited by glucose and by D -gluconic acid–lactone and a glucosyl transferase activity was observed in the presence of ethanol. The β-glucosidase of Leuconostoc mesenteroides, with cyanogenic activity, could be of potential interest in cassava detoxification, by hydrolysing the cyanogenic glucosides present in cassava pulp.  相似文献

Abstract β-d-Xylosidase production was maximal for Humicola grisea var. thermoidea grown on xylan as the sole carbon source. The main β-d-xylosidase activity was localised in the periplasm. β-Xylosidase was purified from crude extracts by heat treatment, ammonium sulfate precipitation and chromatography on DEAE-cellulose and Sephadex G-100. The purified enzyme was a monomer of molecular mass estimated to be 43 kDa by SDS-PAGE and gel filtration. Optima of pH and temperature were 6.0 and 50 °C, respectively. The enzyme activity was stimulated by Ca2+, Fe2+, and Mg2+. The purified β-xylosidase did not exhibit xylanase, carboxymethylcelullase, galactosidase, glucosidase, fucosidase or arabinosidase activities. The purified β-xylosidase hydrolysed xylobiose and xylo-oligosaccharides of up to five monosaccharide units. The enzyme had a K m of 0.49 mM for p -nitrophenyl- β -d-xylopyranoside and was not inhibited by its product, xylose.  相似文献

A 1-FEH II (1-fructan exohydrolase, EC 3.2.1.80) was purified from forced chicory roots ( Cichorium intybus L. var. foliosum cv. Flash) by a combination of ammonium sulfate precipitation, concanavalin A (Con A) affinity chromatography and anion and cation exchange chromatography. This protocol produced a 70-fold purification and a specific activity of 52 nkat mg−1 protein. The apparent size of the enzyme was 60 kDa as estimated by gel filtration and 64 kDa on SDS-PAGE. Optimal activity was found between pH 5.0 and 5.5. The temperature optimum was around 35°C. No product other than fructose could be detected with inulin as the substrate. The purified enzyme exhibited hyperbolic saturation kinetics with an apparent Km of 58 m M for 1-kestose (Kes) and 64 m M for 1,1-nystose (Nys). The purified 1-FEH II hydrolyzed the β (2↠1) linkages in inulin, Kes and Nys at rates at least 5 times faster than the β (2↠6) linkages in levan oligosaccharides and levanbiose. Fructose did not affect the 1-FEH II activity but sucrose (Suc) was a strong inhibitor of this 1-FEH II (Ki=5.9 m M ). The enzyme was partially inhibited by Na-EDTA and CaCl2 (1 m M ).  相似文献

Abstract β-xylosidase (EC 3.2.1.37) has been purified from Aspergillus nidulans mycelium grown on oat-spelt xylan as sole carbon source. Its pH optimum for activity was found to be 5.0 and the optimum temperature was 50 °C. Its molecular mass was estimated by gel filtration to be 180000. Using p-nitrophenyl-β-d-xylopyranoside as substrate, the K m and V max values have been found to be 1.1 mM and 25.6 μmol min−1(mg protein)−1, respectively. Enzyme activity was inhibited by Hg2+, Ag2+, and Cu2+ at a concentration of 1 × 10−3 M. The synthesis of β-xylosidase in A. nidulans is strongly induced by arabinose and xylose and is subject to carbon catabolite repression mediated by the cre A gene product.  相似文献

High β- N -acetylhexosaminidase (EC.3.2.1.52) activity was detected during autolysis of Penicillium oxalicum . Purification of the enzyme to homogeneity yielded an enzyme with a molecular weight of 132 000 Da by gel filtration and 71 900 Da by SDS polyacrylamide gel electrophoresis, suggesting a dimeric structure. The enzyme is an acidic protein with a pl of 5.0. Optimal activity was at pH 4.0 and 40°C, with a K m of 0.80 mmol 1-1 for p -nitrophenyl-β- N -acetylglucosaminide and 1.03 mmol 1-1 for p -nitrophenyl-β- N -acetylgalactosaminide. The K i with the competitive inhibitor O-(2-acetamido-2-deoxy-D-glucopyranosylidene) amino- N -phenylcarbamate was 1 μmol 1-1. Hg2+, Ag+ and Fe3+ were effective inhibitors. β- N -acetylhexosaminidase hydrolysed chitobiose, chitotriose, chitotetrose and chitopentose to monomer to an extent of 92, 74, 44 and 17% respectively in 40 min. This enzyme, in conjunction with a purified endochitinase from P. oxalicum , hydrolysed a cell-wall chitin fraction isolated from this fungus, with the production of N -acetylglucosamine.  相似文献

The extracellular amylase activity from Myxococcus coralloides D was purified by Sephacryl S-200 gel filtration and by ion-exchange chromatography on DEAE-Sephadex A-25. The molecular weight was estimated by SDS-PAGE and by gel filtration as 22.5 kDa. The optimum temperature was 45°C. The pH range of high activity was between 6.5 and 8.5, with an optimum at pH 8.0. Activity was strongly inhibited by Hg2+, Zn2+, Cu2+, Ag+, Pb2+, Fe2+ and Fe3+, EDTA and glutardialdehyde, but was less affected by Ni2+ and Cd2+. Li+, Mg2+, Ba2+, Ca2+, N -ethylmaleimide, carbodiimide and phenyl methyl sulphonyl fluoride had almost no affect. The K m (45°C, pH 8) for starch hydrolysis was 2.0 times 10-3 gl-1. Comparison of the blue value-reducing curves with the time of appearance of maltose identified the enzyme produced by M. coralloides D as an α-amylase.  相似文献

Calcium/calmodulin-dependent kinase II (CaMKII) facilitates L-type calcium channel (LTCC) activity physiologically, but may exacerbate LTCC-dependent pathophysiology. We previously showed that CaMKII forms stable complexes with voltage-gated calcium channel (VGCC) β1b or β2a subunits, but not with the β3 or β4 subunits ( Grueter et al. 2008 ). CaMKII-dependent facilitation of CaV1.2 LTCCs requires Thr498 phosphorylation in the β2a subunit ( Grueter et al. 2006 ), but the relationship of this modulation to CaMKII interactions with LTCC subunits is unknown. Here we show that CaMKII co-immunoprecipitates with forebrain LTCCs that contain CaV1.2α1 and β1 or β2 subunits, but is not detected in LTCC complexes containing β4 subunits. CaMKIIα can be specifically tethered to the I/II linker of CaV1.2 α1 subunits in vitro by the β1b or β2a subunits. Efficient targeting of CaMKIIα to the full-length CaV1.2α1 subunit in transfected HEK293 cells requires CaMKII binding to the β2a subunit. Moreover, disruption of CaMKII binding substantially reduced phosphorylation of β2a at Thr498 within the LTCC complex, without altering overall phosphorylation of CaV1.2α1 and β subunits. These findings demonstrate a biochemical mechanism underlying LTCC facilitation by CaMKII.  相似文献

Abstract— Evidence for multiple forms of the α and β subunits of tubulin isolated from rat brain has been obtained by means of SDS polyacrylamide gel electrophoresis, isoelectric focusing, and SDS hydroxylapatite column chromatography. Fourteen distinct bands, localized near pH 5.4, were formed when tubulin was subjected to isoelectric focusing in a gradient established with a very narrow range ampholyte mixture. Three tubulin subunits, a1., α2, and β, were separated by sodium dodecyl sulfate polyacrylamide slab gel electrophoresis in a second dimension. The β subunit was more acidic than the α subunits. Brain sections were incubated in tissue culture medium containing 32P1 and radiolabeled tubulin was subsequently isolated and subjected to electrophoresis. Only the β subunit was labeled. All radioactivity was associated with two or three adjacent bands on isoelectric focusing gels.  相似文献

Borrelia hermsii , a spirochaete responsible for relapsing fever in humans, grows to high density in the bloodstream and causes thrombocytopenia. We show here that B. hermsii binds to human platelets. Extended culture in bacteriological medium resulted in both diminished infectivity in vivo and diminished platelet binding in vitro . Platelet binding was promoted by the platelet integrin αIIbβ3: the bacterium bound to purified integrin αIIbβ3, and bacterial binding to platelets was diminished by αIIbβ3 antagonists or by a genetic defect in this integrin. Integrin αIIbβ3 undergoes a conformational change upon platelet activation, and bacteria bound more efficiently to activated rather than resting platelets. Nevertheless, B. hermsii bound at detectable levels to preparations of resting platelets. The bacterium did not recognize a point mutant of αIIbβ3 that cannot acquire an active conformation. Rather, B. hermsii was capable of triggering platelet and integrin αIIbβ3 activation, as indicated by the expression of the platelet activation marker P-selectin and integrin αIIbβ3 in its active conformation. The degree of platelet activation varied depending upon bacterial strain and growth conditions. Prostacyclin I2, an inhibitor of platelet activation, diminished bacterial attachment, indicating that activation enhanced bacterial binding. Thus, B. hermsii signals the host cell to activate a critical receptor for the bacterium, thereby promoting high-level bacterial attachment.  相似文献

The effects of pretreatment with HCN on the level of fructose 2.6-bisphosphate (F-2,6-P2). on activity of fructose 6-phosphate 2-kinase (EC 2.7.1.150; F-6-P. 2K. enzyme synthesizing F-2,6-P2), as well as on activities of PP1-dependent and ATP-dependent phosphofructokinases (EC 2.7.1.90. PP1-PFK and EC 2.7.1.11, ATP-PFK) were studied in cultured, dormant embryos of apple ( Malus domestica Borb. cv. Antonówka). HCN increased the F-2.6-P2 level and F-6-P, 2K activity in embryonal axes (3-fold), but had no effect in cotyledons. HCN pretreatment of embryos or the addition of F-2.6-P2 to enzyme extract stimulated PP1, -PFK activity, whereas the activity of ATP-PFK was slightly inhibited by HCN in axes and in cotyledons. Glycolysis is one of the first processes in the germination of apple embryos, and the stimulation of glycolysis by HCN may be the result of F-6-P, 2K activation in the axes. This will lead to accumulation of F-2,6-P2, which, in turn, enhances glycolysis by activation of PP1PKF.  相似文献

A mitochondrial serine hydroxymethyltransferase (EC 2.1.2.1) has for the first time been purified close to homogeneity from a photosynthetically active tissue, spinach ( Spinacea oleracea L. cv Viking II) leaves. The specific activity of the enzyme was 7.8 μmol (mg protein)−1 min−1 using L-serine as substrate. The enzyme was stable for at least 8 weeks at 4°C in the presence of folate. The pH optimum was at pH 8.5 where the enzyme had a Km for L-serine of 0.9 m M . Carboxymethoxylamine was a strong competitive inhibitor with a K1 of 1.4 μM. An absorption spectrum taken of the enzyme in the presence of glycine and tetrahydrofolate showed a peak at 492 nm, probably originating from a substrate-enzyme complex. The molecular weight obtained by gel filtration was 209 kDa. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis of the purified enzyme showed that the apparent molecular weight of the subunit was 53 kDa, indicating four subunits.  相似文献

Abstract Adenylate cyclase activity of Alternaria solani bound to the particulate cell fraction was solubilized by 2.5% Ficoll. The apparent K m of the solubilized enzyme for adenosine 5'-triphosphate was 2.5 mM, and it required Mn2+ for maximum activity. M r as determined by gel filtration was approximately 500. Fluoride ions at millimolar concentrations, GTP, adenosine, and N6-phenylisopropyl adenosine at micromolar concentrations did not stimulate adenylate cyclase activity. The enzyme was inhibited 25–55% by millimolar concentrations of 2'-deoxyadenosine, 2'-0 methyl adenosine, 9-β- d -arabinofuranosyl adenosine and 2'-3'-isopropylidene adenosine. Partially purified enzyme obtained after DEAE-BioGel chromatography was very unstable. The amount of extractable enzyme activity varied during the fungal growth cycle.  相似文献

T.M. ALCONADA AND M.J. MARTÍNEZ. 1996. Fusarium oxysporum f. sp. melonis produces cellulase and β-glucosidase activities in a medium with glucose and avicel as carbon source. A β-glucosidase from this crude material was purified by gel filtration and ion exchange chromatography successively. This enzyme is a unique band of protein in SDS-PAGE and isoelectric focussing. It had a molecular weight of 66000 and a pI of 5. Using p -nitrophenyl-β-D-glucopyranoside as substrate β-glucosidase shows a K m of 210 μmol 1-1, an optimum pH of 5.5 and an optimum reaction temperature of 60°C, being stable in a pH range of 5–7 for 48 h at room temperature.  相似文献
