Minced salivary glands from seven white-lipped marmosets (Saguinus fuscicollis and Saguinus nigricollis) and one cotton-topped marmoset (Saguinus oedipus) were cocultivated with marmoset cell cultures. A viral agent, designated SSG, was isolated from two Saguinus fuscicollis. Slowly progressing foci of rounded, vacuolated, refractile cells were first observed at 40-43 days incubation. Electron microscopy revealed intranuclear herpesvirus nucleocapsids and intracytoplasmic and extracellular enveloped particles. Infected cells stained with hematoxylin and eosin contained eosinophilic intranuclear and cytoplasmic inclusion bodies. SSG could be passaged in cell cultures only using viable whole cells; infectious cell-free virus was not detected in either culture supernatants or cell lysates. SSG replicated in marmoset fibroblastic but not in marmoset epithelioid or human fibroblastic cell cultures. Plasma antibodies to SSG were detected by indirect immunofluorescence assays in 16 of 56 (28.6%) adult wild-caught marmosets but were absent in 40 colony-born, hand-reared marmosets. Antigenic cross-reactivity of SSG with a rhesus monkey (Macaca mulatta) cytomegalovirus (bidirectional) and with a human cytomegalovirus (unidirectional) was also demonstrated by indirect immunofluorescence assays. SSG was identified as a herpesvirus by morphology and was classified as a cytomegalovirus by its site of isolation, biologic properties in vitro, and antigenic characteristics.  相似文献

Development of an isoimmune serum capable of identifying a specific leukocyte antigen in the marmoset, Saguinus fuscicollis illigeri, permitted detection of lymphoid cell chimerism in this species by the cytotoxic test. This reagent was then used to identify the cell population in the chimera responsible for antibody production against a test antigen, sheep red blood cells. Primary in vitro antibody formation as measured by plaque-forming cells with blood leukocytes or splenic lymphocytes of six animals chimeric for the leukocyte antigen, MLA-1, revealed an immune response by both cell types of the chimeric population from three animals and a response by only one cell type in the other three.  相似文献

A lymphocyte population of common marmosets (Callithrix jacchus) was identified by rosette formation with African green monkey erythrocytes; the rosette-forming cells appeared to be T lymphocytes, as approximately 62% of circulating lymphocytes and 85% of thymus cells formed rosettes with African green monkey erythrocytes. In addition, common marmoset lymphoid cells carrying T-lymphotropic Herpesvirus saimiri or Herpesvirus ateles formed rosettes with African green monkey erythrocytes and treatment of common marmoset circulating lymphocytes with an anti-T cell serum and complement (C′) eliminated rosette-forming cells. Common marmoset T lymphocytes apparently carry a surface receptor for African green monkey erythrocytes, but unlike humans and other closely related nonhuman primates, T lymphocytes of common marmosets fail to form rosettes with sheep erythrocytes.  相似文献

Non-human primates (NHPs) offer valuable animal models for basic research into human diseases and for the preclinical validation of new therapeutics. Detailed in situ examination of the involved cell types using immunohistochemistry is often hampered by the lack of cross-reactive antibodies (Abs). In the current study, we have tested a large panel of monoclonal antibodies raised against human leukocyte differentiation and activation markers for cross-reactivity on cryosections of lymphoid tissue from six NHP species. In total, we have tested 130 Abs against 69 antigens expressed in tissues from one great ape species (chimpanzee/Pan troglodytes), two Old World species (rhesus macaque/Macaca mulatta and cynomolgus macaque/Macaca fascicularis), and three New World species (common marmoset/Callithrix jacchus, cotton-top tamarin/Saguinus oedipus, and owl monkey/Aotus triviogatus). We have found a large panel of cross-reactive Abs: 93 of 102 (91%) in chimpanzee, 97 of 125 (78%) in rhesus macaque, 70 of 109 (64%) in cynomolgus macaque, 69 of 116 (60%) in common marmoset, 40 of 81 (49%) in cotton-top tamarin, and 35 of 80 (44%) in owl monkey. The availability of a reliable panel of cross-reactive markers is important to gaining further insight into immunological processes in disease-affected tissues from NHP species. The cleared and alizarin-stained feet of eight marmoset (Saguinus oedipus) fetuses revealed a sequence of initial ossification of the 19 bones of the foot that is almost totally different from that of man. The most striking difference is in the order of ossification of the distal phalanx of the big toe; in man it is the first bone to commence ossification, in the marmoset it is the fourteenth. It is suggested that the sequence of initial ossification in the foot may be species specific among the primates and may serve as a useful marker of teratogenic action.  相似文献

We describe Rondon’s marmoset (Mico rondoni sp. n.) from the Rio Jamari in the Brazilian state of Rondônia and differentiate the species from other Amazonian marmosets on the basis of morphological, genetic, and zoogeographic characteristics. Mico rondoni sp. n. is a typical bare-eared marmoset of predominantly grayish coloration and unpigmented face and cheiridia, which contrasts clearly with its more brownish and pigmented parapatric congeners Mico melanurus and M. nigriceps. Genetically, the new species is unquestionably distinct from Mico emiliae, with which it had been synonimized previously. The geographic range of the new species is defined as the area delimited by the Mamoré, Madeira, and Jiparaná rivers to the west, north, and east, respectively, and the Serra dos Pacaás Novos, to the south, where it is substituted by Mico melanurus. Depending on its exact limits, this range may cover an area of little more than 100,000 km2. A characteristic of the ecology of Mico rondoni sp. n. is its range-wide sympatry with the similarly sized tamarin Saguinus fuscicollis weddelli, which may be a key factor determining the rarity or absence of the new species from many areas within its geographic range. Its apparent scarcity may make the new species exceptionally vulnerable to the effects of the intense human colonization occurring throughout much of its known range, and provoke cause for concern for its long-term survival.  相似文献

A monkey-adapted strain of Plasmodium vivax (Achiote) was transferred by trophozoites through 11 serial passages in Saguinus geoffroyi (Titi marmoset). Patent infections developed in both normal (23 of 24) and splenectomized (8 of 9) marmosets. The infections in the altered animals were of greater severity than in the intact subjects as indicated by patent periods (x? = 87 vs 61 days) and maximum parasitemias (x? = 35,018 vs 16,218 per mm3).Relapses were recorded in 13 of 14 unaltered and 4 of 4 splenectomized animals that survived the primary attack. As evidence for acquired immunity, the mean maximum parasitemias during relapse in the normal animals were 18 of the values during the primary attack; patent periods were shorter than those in the initial infection. There was also some indication of an acquired immunity in the splenectomized group of animals. However, one splenectomized marmoset experienced a patent period of 325 days during the first relapse.There was considerable variation of infection parameters among the animals in each group.  相似文献

The distribution of the human endogenous retrovirus (HERV)-K genome was investigated by Southern-blot analyses using a HERV-K-env DNA probe. With the exception of one DNA-sample, obtained from a Chinese individual in whom an amplification of HERV-K was detected, Southern-blot analyses yielded identical hybridization patterns with DNA from peripheral blood lymphocytes of 37 normal healthy blood donors, with DNA from six tumor cell lines, or with 23 DNA samples prepared from various carcinoma tissues. To elucidate whether the integration of HERV-K genomes into the primate lineage occurred as a single event or as an integration with later expansion, we further examined the evolutionary history of HERV-K by Southern blot analyses with DNA samples from different primate species. We detected HERV-K genomes in Macaca mulatta and Macaca silenus, which represent Old World monkeys, but not in prosimians (Galago demidovii) and New World monkeys, represented by Saguinus fuscicollis, Saguinus oedipus, and Calliihrix iacchus. Thus, we assume that the infection of the primate lineage with HERV-K had occurred after the divergence of New World and Old World monkeys, but before the evolutionary expansion of large hominoids. In contrast to the apparent lack of HERV-Kenv sequences in DNA from tissue of the New World monkey Saguinus oedipus (cotton-top marmoset), we found HERV-K-DNA in the B95-8 cell-line, which is a Saguinus oedipus leukocyte cell-line, immortalized in vitro by Epstein-Barr virus (EBV) and cultivated in human cells. It may be speculated that HERV-K-DNA or HERV-K-particles were introduced into these cells during in vitro transformation with EBV.  相似文献

The common marmoset (Callithrix jacchus) is a small New World primate that has been used as a non-human primate model for various biomedical studies. We previously demonstrated that transplantation of neural stem/progenitor cells (NS/PCs) derived from mouse and human embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) promote functional locomotor recovery of mouse spinal cord injury models. However, for the clinical application of such a therapeutic approach, we need to evaluate the efficacy and safety of pluripotent stem cell-derived NS/PCs not only by xenotransplantation, but also allotransplantation using non-human primate models to assess immunological rejection and tumorigenicity. In the present study, we established a culture method to efficiently derive NS/PCs as neurospheres from common marmoset ESCs. Marmoset ESC-derived neurospheres could be passaged repeatedly and showed sequential generation of neurons and astrocytes, similar to that of mouse ESC-derived NS/PCs, and gave rise to functional neurons as indicated by calcium imaging. Although marmoset ESC-derived NS/PCs could not differentiate into oligodendrocytes under default culture conditions, these cells could abundantly generate oligodendrocytes by incorporating additional signals that recapitulate in vivo neural development. Moreover, principal component analysis of microarray data demonstrated that marmoset ESC-derived NS/PCs acquired similar gene expression profiles to those of fetal brain-derived NS/PCs by repeated passaging. Therefore, marmoset ESC-derived NS/PCs may be useful not only for accurate evaluation by allotransplantation of NS/PCs into non-human primate models, but are also applicable to analysis of iPSCs established from transgenic disease model marmosets.  相似文献

Saddleback tamarins (Saguinus fuscicollis, S. melanoleucus, and S. tripartitus) occur in the upper Amazon, west of the rios Madeira and Mamoré–Guaporé to the Andes. They currently comprise 13 species and subspecies in Bolivia, Brazil, Colombia, Ecuador, and Peru. Here we report on a previously undescribed subspecies of Saguinus fuscicollis from the interfluvium of the lower rios Madeira and Purus in the central Amazon of Brazil. It is a distinct form with a very pronounced mottling on the back giving the appearance of a saddle, a characteristic shared by the 3 species, and the reason for their collective common name. The lack of a white superciliary chevron and its highly mottled ochraceous-dark brown to black saddle that extends from the scapular region to the base of the tail distinguish the new taxon from Saguinus fuscicollis weddelli. The latter characteristic also distinguishes it from another saddleback tamarin (Saguinus fuscicollis ssp.) that has been reported immediately to the south of its range in the same interfluvium. We compare pelage coloration, external characters and craniometric measurements with its geographically closest relatives, Saguinus fuscicollis weddelli and S. f. avilapiresi, and indicate its probable geographic distribution in a small area between the rios Madeira and Purus, just south of the Rio Amazonas.  相似文献

Whole blood cultures from humans and from the New World primate, Saguinus fuscicollis, were irradiated with various doses of 250 kV X-rays. The resulting centric ring plus dicentric aberration yields were fitted to the three models, Y = a+bD, Y = a+bD+cD2, and Y = a+cD2, by least squares regression. In both instances the best fit was to the model Y = a+bD+cD2, with coefficients of the one- and two-track components for human and marmoset being: b = (0.78 ± 0.09)·10−3, c = (5.92 ± 0.31)·10−6, and b = (1.11 ± 0.36)·−3, c = (7.7 ± 1.7)·10−6, respectively.  相似文献

The genus Fusobacterium includes not only strict anaerobic Gram-negative rods found in the normal oral and gastrointestinal microbiota but also those involved in a variety of clinical infections. This amphibiontic relationship with the host may be related to an adaptive capacity of aerotolerance permitting their passage from an extremely reduced (digestive ecosystem) to a less reduced (host tissue) environment. In the present study, 74 oral stock or recently isolated strains of Fusobacterium recovered from human beings (58) and marmosets (16) were evaluated for their aerotolerant capability. The marmoset strains were also evaluated after a handling period of 2 months in an anaerobic chamber. StockFusobacterium strains from both healthy human (mean tolerance time: 23.71±8.67 h) and marmoset (21.25±9.36 h) oral cavities were more aerotolerant (P<0.05) than recent isolates from healthy human (10.67±10.61 h) and marmoset (8.33±8.46 h) oral cavities. After a two month handling period, the same fresh isolates became more aerotolerant (P<0.05) than soon after isolation (30.00±0.00 h and 16.17±9.36 h for human and marmoset strains, respectively). There was no difference (P>0.05) between stock strains from healthy human oral cavities (23.71±8.67 h) or human oral cavities presenting periodontal disease (17.91±10.10 h) or between healthy human and marmoset strains either after a long time storage or a recent isolation. But after a two-month handling period, the recently isolated strains from marmosets were less aerotolerant than those of human origin (P<0.05). During growth in culture, all strains tested were more aerotolerant during the exponential phase and drastically lost this characteristic when they reached the stationary stage. We conclude that the Fusobacterium strains used in these experiments present a wide and varying aerotolerance capability, probably important for the adaptation of these bacteria to the environment.  相似文献

Ultrasonography was used in six saddle back tamarin females (Saguinus fuscicollis) to diagnose pregnancy, monitor the patterns of uterine growth and embryonic/foetal development and examine the incidence loss of single embryos/foetuses. Pregnancy was reliably diagnosed 17 days after conception, 10 days earlier than by plasma progesterone measurement. The patterns of uterine and embryonic/foetal growth paralleled those reported for the common marmoset, including a delay in embryonic development. The results support the hypothesis of retardation of organogenesis in most callitrichid species. Individual embryos could be reliably identified from day 50 of pregnancy; a loss of single embryos/foetuses after this stage did not occur. All pregnancies were carried to term, resulting in five times twins and one singleton. The smaller litter size compared to the common marmoset may be due to loss of single embryos at earlier stages of pregnancy or to a lower ovulation rate.  相似文献

To assess the important aspects of a large area for environmental enrichment, the behavior of families of common marmosets Callithrix jacchus and cotton-top tamarins Saguinus oedipus when in laboratory cages and when in much larger outside areas was compared. All animals avoided areas with little cover. The two tamarin families preferred an indoor cage to a large open area with little cover and consequently only showed a limited range of behavior when outside. When dense cover was provided to the marmoset family, they used this area extensively, showing a wider range of behavior than when in cages and shifting to less active behavior patterns. By feeding marmosets in a central area, it was possible to allow unlimited access to the outside while effectively containing them.  相似文献

The mammalian retina has more diversity of neurons than scientists had once believed in order to establish complicated vision processing. In the monkey retina, morphological diversity of retinal ganglion cells (RGCs) besides dominant midget and parasol cells has been suggested. However, characteristic subtypes of RGCs in other species such as bistratified direction-selective ganglion cells (DSGC) have not yet been identified. Increasing interest has been shown in the common marmoset (Callithrix jacchus) monkey as a “super-model” of neuroscientific research. Here, we established organotypic tissue culture of the adult marmoset monkey retina with particle-mediated gene transfer of GFP to survey the morphological diversity of RGCs. We successfully incubated adult marmoset monkey retinas for 2 to 4 days ex vivo for transient expression of GFP. We morphologically examined 121 RGCs out of more than 3240 GFP-transfected cells in 5 retinas. Among them, we identified monostratified or broadly stratified ganglion cells (midget, parasol, sparse, recursive, thorny, and broad thorny ganglion cells), and bistratified ganglion cells (recursive, large, and small bistratified ganglion cells [blue-ON/yellow-OFF-like]). By this survey, we also found a candidate for bistratified DSGC whose dendrites were well cofasciculated with ChAT-positive starburst dendrites, costratified with ON and OFF ChAT bands, and had honeycomb-shaped dendritic arbors morphologically similar to those in rabbits. Our genetic engineering method provides a new approach to future investigation for morphological and functional diversity of RGCs in the monkey retina.  相似文献

The amino acid sequence of the myoglobin of the South American Night Monkey, Aotes trivirgatus, is identical to that of the marmoset (Callithrix jacchus [1]) except for residue 21 which is isoleucine in the marmoset, like in all other anthropoids, but valine in Aotes. Analysis of a possible pathway of the evolution of Aotes myoglobin using 18 known primate myoglobin sequences [2–5] supports the classification of the Night Monkey within Anthropoidea and Platyrrhini but it indicates that this species might be more closely related to the marmoset (family Callitrichidae) than to the family Cebidae as a member of which it is commonly classified.  相似文献

Paternal behaviour is critical for the survival of offspring in many monogamous species. Common marmoset (Callithrix jacchus) and cotton-top tamarin (Saguinus oedipus) fathers spend as much or more time caring for infants than mothers. Expectant males of both species showed significant increases in weight across the pregnancy whereas control males did not (five consecutive months for marmoset males and six months for cotton-top tamarin males). Expectant fathers might be preparing for the energetic cost of fatherhood by gaining weight during their mate's pregnancy.  相似文献

The behavioral patterns and social interaction of a marmoset (Saguinus fuscicollis) group in a semi-naturalistic environment were observed for 14 months. The analysis showed that, of the 32 behavior patterns observed, the 10 most frequent accounted for over 97 per cent of the total behavior. One pattern, sit and look, accounted for 44 per cent of the total behavior. The two most frequent social behavior patterns, grooming and social play, were concentrated in different parts of the group. The focal point of grooming was the adult female; social play was characteristic of younger animals 4–20 months of age. These results were compared with other studies of marmosets and with primate studies on grooming and social play.  相似文献
