Monokaryotic mycelia of the homobasidiomycete Coprinus cinereus form asexual spores (oidia) constitutively in abundant numbers. Mycelia with mutations in both mating type loci (Amut Bmut homokaryons) also produce copious oidia but only when exposed to blue light. We used such an Amut Bmut homokaryon to define environmental and inherent factors that influence the light-induced oidiation process. We show that the Amut function causes repression of oidiation in the dark and that light overrides this effect. Similarly, compatible genes from different haplotypes of the A mating type locus repress sporulation in the dark and not in the light. Compatible products of the B mating type locus reduce the outcome of light on A-mediated repression but the mutated B function present in the Amut Bmut homokaryons is not effective. In dikaryons, the coordinated regulation of asexual sporulation by compatible A and B mating type genes results in moderate oidia production in light. Copyright 1998 Academic Press.  相似文献

Four recessive primordiumless mutants from anAmut Bmut strain ofCoprinus cinereus were crossed with wild-type strain. Summary Twenty-four sporeless mutants were isolated from an Amut Bmut strain (mutant in the incompatibility factors) of the basidiomycete Coprinus cinereus. All the sporeless mutations were recessive to the wild type. These mutants and a previously isolated recessive sporeless strain, N2-7 (Kanda and Ishikawa 1986) were crossed with a wildtype strain. An F1 random spore analysis indicated that sporulation deficiencies in these mutants were caused by single nuclear gene mutations. These mutations were all complementary to each other, thus twenty-five sporulation genes were identified. Five of them were linked to the A incompatibility factor. Cytological observations classified these mutants into the following four types according to the stage of the blockage: (1) meiosis stopped at meta-anaphase I; (2) meiosis was completed, but further basidial development did not occur; (3) basidial development stopped at the sterigma stage; (4) basidial development stopped at the prespore stage.  相似文献

Polak, E., Hermann, R., Kues, U., and Aebi, M. 1997. Asexual sporulation in Coprinus cinereus: Structure and development of oidiophores and oidia in an Amut Bmut homokaryon. 22, 112-126. Coprinus cinereus strain AmutBmut is a homokaryon with mutations in both mating type loci. It produces asexual spores (oidia) in sticky liquid droplets on specialized aerial structures (oidiophores). These oidiophores have uninucleate cells and are organized as those of the monokaryon 5026 from which the strain derived. However, unlike in the monokaryon, oidiophores in strain AmutBmut are induced by light. Young oidiophores are easily detected upon light induction and the process of oidiophore development is readily followed in this strain. Fully grown oidiophores consecutively give rise to short branches (oidial hyphae) that break up into two or occasionally three uninucleate oidia (arthroconidia) until up to 200 oidia are collected at the tip of the oidiophore. Mature spores are enclosed by a mucilage and a double-layered primary cell wall with hair-like structures except for the sides of former cell attachments. In a summary of our microscopic observations on developing oidiophores and nuclear stainings we present a model showing the successive steps of oidiophore and spore development.  相似文献

A predominantly autogamous breeding system is described forP. occidentale Scribn.,P. pacificum Hitchc. & Chase, andP. thermale Boland. Five synthetic hybrids and three generations of progeny are also described for crosses among these three species. All parents haven = 9 chromosomes and normal meiosis. Hybrids exhibit various irregularities during meiosis, but nine bivalents are usually formed. One population ofP. occidentale and one ofP. thermale were found to have chromosomes with interchanges larger than those of other populations. The F2 and F3 generations had markedly increased fertility, as determined by pollen stainability. It is proposed that autogamy and hybridization are a common means of maintaining and stabilizing genetic variability in these grasses and that these means account for much of the taxonomic difficulty posed by the subgenus. The possibility of a low rate of introgression between populations is also proposed.  相似文献

Cytogenetic investigations have been made in the fourPetrorhagia species and hybrids of the sectionKohlrauschia. The three diploid species show close similarities in chromosome number, size and morphology, with the exception ofP. velutina, where one pair of metacentric chromosomes is represented by a pair of telocentrics. Meiotic studies in hybrids indicate close genomic homology between the diploid species and also between the two floral forms ofP. prolifera. The tetraploidP. nanteuilii behaves as an allotetraploid forming only bivalents at meiosis and results suggest thatP. velutina andP. prolifera are the diploid progenitors of this species. Since meiosis in diploid and triploid hybrids results in extensive intergenomic pairing it is concluded that the natural tetraploid has a bivalent promoting mechanism that prevents pairing between the genomes of its diploid progenitors.  相似文献

The C-banding patterns in the chromosomes ofMicrotus oeconomus, M. arvalis andM. ochrogaster demonstrate differences in the amount and distribution of heterochromatin. Autosomal centromeric heterochromatin appears as conspicuous blocks or as small dots, and in several chromosomes no heterochromatin was detected; interstitial heterochromatin was observed in one autosome pair ofM. ochrogaster. The sex chromosomes also demonstrate differences in the C-banding pattern. InM. oeconomus, the X chromosome exhibits a block of centromeric heterochromatin which is larger than that of the autosomes; this characteristic helps to recognize the X chromosomes in the karyotype. InM. arvalis no heterochromatin was appreciated in the sex chromosomes. The Y chromosomes ofM. ochrogaster andM. oeconomus are entirely heterochromatic. During male meiosis heterochromatin shows condensation, association and chiasma prevention; the sex chromosomes pair end to end in the three species. At pairing, the Y chromosome ofM. arvalis is despiralized, but it appears condensed again shortly before separation of the bivalent.  相似文献

Populations ofLuzula multiflora s.l. in Ireland were examined karyologically. Plants from 14 populations were invariably tetraploid with 2n=24. Chromosomes of the tetraploid are of AL type (true tetraploidy). Meiosis of the tetraploids is of the same type as described for otherLuzula taxa in the literature. In meiosis, 12 bivalents are regularly formed. A hypothesis based on the morphological and allozyme data, that the tetraploids are of alloploid origin, is supported by the present results. Meiosis in an artificial hybrid between the presumed parental taxa,L. campestris andL. pallidula, was studied; a certain tendency towards chromosome doubling was observed. The geographical distribution of theL. multiflora cytotypes is also discussed.  相似文献

A fully normal meiosis was found inAllium sativum L. by means of PMC analysis. Quadrivalents were formed only in several cultivars. The cultivation of inflorescence stem (after the exstirpation of bulblets) in a tetracycline solution of 900 i.u. induced a formation of fertile flowers. The first pollen mitosis was studied and the pollen parameters compared with those ofAllium cepa L. Evidence for the pollen sterility was given by crossings on sterile types. Natural and synthetic hybrids are reported betweenPanicum, scribnerianum Nash and two other species,P. pacificum Hitchc. & Chase andP. occidentale Scribn. & Merr. The hybrids are morphologically intermediate between the parents, and those in whichP. pacificum is one parent strongly resemble the rareP. shastense Scribn. & Merr. All the parent plants studied cytologically had regular meiosis and a gametic number ofn = 9 chromosomes. The hybrids usually showed good pairing of chromosomes in meiosis, but irregularities such as univalents and lagging chromosomes were common. In the hybrids generally less than 7% of the pollen grains stained with cotton blue in lactophenol, and the plants set no seed, whereas the parent plants generally had over 90% pollen stainability and a high percentage of seed set. Pollen fromP. shastense, like that of the hybrids, was 3–4% stainable.Panicum shastense is believed to be based on a first generation sterile hybrid and the nameP. xshastense is proposed.  相似文献

The mean chiasma frequency at first metaphase of meiosis in pollen mother cells of a maize variety, Black Mexican Sweet Corn, increases with increasing number ofB-chromosomes. The increased chiasma frequency is also reflected by a decrease in the number of rod bivalents at first metaphase. There is some indication that theB-chromosomes also influence the distribution of chiasmata between pollen mother cells. The increase in chiasma frequency due toBs is closely analogous to the increase in crossing-over carlier attributed toBs between marker genes located on chromosome III. Together the evidence confirm the role ofBs in controlling genetic recombination at meiosis.  相似文献

We have isolated 37 radiation-sensitive mutants of the basidiomyceteCoprinus cinereus. Each mutation is recessive, and the collection defines at least ten complementation groups for survival of gamma irradiation. Four complementation groups define the genesrad3, rad9, rad11 andrad12, which are required both for survival of gamma irradiation and for meiosis. Mutants in each of these four groups fail to complete meiosis and produce mushrooms with greatly reduced numbers of viable spores. Propidium iodide staining of meiotic nuclei showed a characteristic terminal appearance for each mutant: few cells of any of the meiotic mutants progress beyond prophase I, and both condensation and fragmentation or dispersal of meiotic chromatin are frequently observed. Scanning electron micrographs showed that the meiotic mutants make varying numbers (0–6) of basidiospore initials and that few of these initials develop into mature spores. When initials are present they are always symmetrically arrayed on the basidium, regardless of initial number. In quantitative measurements of gamma ray sensitivity, double mutants of every tested combination ofrad3, rad9, rad11 andrad12 consistently showed the same gamma ray sensitivity as the more sensitive single mutant parent of the cross. Therefore, these four genes are in the same pathway for the repair of gamma radiation damage, and this pathway also represents one or more functions essential for meiosis.  相似文献

Meiosis is a process unique to the differentiation of germ cells and exhibits sex-specific in timing. Previous studies showed that retinoic acid (RA) as the vitamin A metabolite is crucial for controlling Stra8 (Stimulated by retinoic acid gene 8) expression in the gonad and to initiate meiosis; however, the mechanism by which retinoid-signaling acts has remained unclear. In the present study, we investigated the role of the enzyme retinaldehyde dehydrogenase 2 (RALDH2) which catalyzes RA synthesizes by initiating meiosis in chicken ovarian germ cells. Meiotic germ cells were first detected at day 15.5 in chicken embryo ovary when the expression of synaptonemal complex protein 3 (Scp3) and disrupted meiotic cDNA 1 homologue (Dmc1) became elevated, while Stra8 expression was specifically up-regulated at day 12.5 before meiosis onset. It was observed from the increase in Raldh2 mRNA expression levels and decreases in Cyp26b1 (the enzyme for RA catabolism) expression levels during meiosis that requirement for RA accumulation is essential to sustain meiosis. This was also revealed by RA stimulation of the cultured ovaries with the initiation of meiosis response, and the knocking down of the Raldh2 expression during meiosis, leading to abolishment of RA-dependent action. Altogether, these studies indicate that RA synthesis by the enzyme RALDH2 and signaling through its receptor is crucial for meiosis initiation in chicken embryonic ovary.  相似文献

In megasporogenesis ofTorreya nucifera (Taxaceae) more than one product of meiosis can start to germinate, a process previously observed inTaxus. Ultrastructural analysis ofT. nucifera revealed that this behaviour is cytologically determined by the presence of a transfer cell type wall-membrane apparatus and by the presence of uncommonly complex plasmalemmasomes in the megasporocyte. These features may determine a uniform supply of nutrients to the entire megasporocyte cytoplasm, so that after meiosis the germination potential is not restricted to the chalazal megaspore. Even if more than one product of meiosis can originate the female gametophyte in bothTaxus andTorreya, this process involves different ultrastructural aspects in the two species. In conclusion, the extension of the germination potential is a common characteristic to both species but it is most probably the result of some evolutionary convergence.  相似文献

In their natural habitats, some representatives of Prasiolales are regularly, or often, washed by waves; others are sprayed with sea-water only occasionally, or not at all. In correspondance to this ecological grouping, species require marine or freshwater media respectively for optimal growth in cultures. The halophilic group consists ofPrasiola stipitata andRosenvingiella constricta. Though not analysed in detail, morphological characters and behaviour in cultures suggest that several different species are united under the namePrasiola stipitata. Rosenvingiella constricta, new for Helgoland and thus far reported only twice in Europe, is a dioecious haplont; both meiosis and genotypic sex-determination occur in the germinating zygote. The parenchymatous “Gayella-stage” ofRosenvingiella polyrhiza proved to be male and female gametangia, producing spermatozoids and non-motile eggs respectively. Fertile threads were extremely rare in natural material and — contrary toRosenvingiella constricta — did not occur in cultures, so the life cycle could not be elucidated.Prasiola calophylla is frequently associated withRosenvingiella polyrhiza. The life cycle includes heteromorphic asexual generations, the spores of the leafy thallus giving rise to unicellular aplanosporangia.Prasiola furfuracea reproduces only by spores. Samples ofRosenvingiella polyrhiza frequently containPrasiolopsis in addition to a number of other Chlorophyceae. Consequently, it is easy to appreciate that previous investigators came to erroneous conclusions about life histories and biological relations in studies based only on field material.  相似文献

Forty-four chromosome counts from the North American genusCallirhoe are reported, including first-recorded counts ofC. alcaeoides, C. digitata, C. bushii, C. triangulata, andC. involucrata var.tenuissima. Information concerning chromosome numbers from present and previous reports is summarized and evaluated. Observations are presented on meiosis for the genus in general, for male sterile individuals of gynodioecious species, and for selected F1 hybrids. Pollen fertility data are recorded for the latter. Chromosomally three species groups are recognized: an exinvolucellate, diploid species group withn=14; an involucellate, tetraploid and octoploid species pair,n=28 and 56; and an involucellate, diploid and tetraploid species group withn=15 and 30. Intrageneric relationships and those within the tribe Malveae subtribe Malvinae are discussed.  相似文献
