The effects of prostaglandin F on human blood platelet function were investigated. PGF at 15 μM completely blocked platelet aggregation induced by 500 μM arachidonic acid or 3 μM U46619 but had no effect on aggregatin induced by 7.5 μM ADP. A similar specificity of action was not obtained with either PGI2 or PGE2. Thus concentrations of PGI2 (3 nM) or PGE2 (20 μ M) which inhibited U46619-induced aggregation by 100% also blocked ADP-stimulated aggregation.The inhibitory properties of PGF were not related to increases in platelet cAMP, since direct measurement of intracellular cAMP revealed that 15 μ M PGF produced no substantial change in cAMP levels. This finding was in direct contrast to results obtained using either PGI2 or PGE2. Both PGI2 (3 nM) and PGE2 (20 μ M) induced significant increases in platelet cAMP levels.The possibility that PGF directly interacts at the platelet TXA2/PGH2 receptor was investigated by measuring [3H]PGF binding to isolated platelet membranes. It was found that [3H] PGF binding reached equilibrium within 30 min at room temperature and could be 90% displaced by addition of 1000 fold excess of unlabelled PGF. Furthermore, when 1000 fold excess of either the TXA2/PGH2 “mimetic” U46619 or the TXA2/PGH2 antagonist 13-azaprostanoic acid was added, specific [3H] PGF binding was displaced by 95% and 85% respectively. In contrast, the same molar excess of 6-keto-PGF, azo analog 1, or TXB2, caused displacement of only 15%, 20% or 25% of the [3H] PGF binding. Scatchard analysis indicated that [3H] PGF has two binding sites; i.e., a high affinity binding site with an apparent Kd of 50 nM and a low affinity binding site with apparent Kd of 320 nM. These results suggest that the selective inhibition by PGF of AA or U46619-induced aggregation may be mediated through interaction at the platelet TXA2/PGH2 receptor.  相似文献

The response of endothelial adenylate cyclase (AC) to prostaglandins (PGE1, PGE2, PGF, PGF, PGD2 and PGI2) and the relationship of PGE2 to adrenergic systems were investigated in cerebrovascular endothelial cultures. E-type prostaglandins and PGI2 were more effective in stimulating endothelial AC (EC50 = 3 × 10?7M, and 3 × 10?7M, respectively) than prostaglandins of the F-series and PGD2 which activated AC at high doses only. A modulation of endothelial AC response to either PGE2 or norepinephrine (NE) was observed in the presence of both agents in the system. It was manifested by a dose-dependent NE inhibition of the PGE2-stimulated formation of cAMP, which was partially restored by phentolamine. Alpha and β-adrenergic agonists (α, clonidine and 6-fluoronorepinephrine; β, isoproterenol) also partly blocked while forskolin and PGE2 synergistically stimulated the production of cAMP in the endothelial cultures. These findings strongly suggest that the interaction of prostaglandins and α- and β-adrenergic agonists with the AC system in cerebrovascular endothelium may play a role in the regulation of the cerebral microcirculation and/or blood pressure.  相似文献

Prostanoids comprising prostaglandins (PGs) and thromboxanes (TXs) have been shown to play physiological and pathological roles in zebrafish. However, the molecular basis of zebrafish prostanoid receptors has not been established. Here, we demonstrate that there exist at least five ‘contractile’ (Ca2+-mobilizing) and one ‘inhibitory’ (Gi-coupled) prostanoid receptors in zebrafish; five ‘contractile’ receptors consisting of two PGE2 receptors (EP1a and EP1b), two PGF receptors (FP1 and FP2), and one TXA2 receptor TP, and one ‘inhibitory’ receptor, the PGE2 receptor EP3. [3H]PGE2 specifically bound to the membranes of cells expressing zebrafish EP1a, EP1b and EP3 with a Kd of 4.8, 1.8 and 13.6 nM, respectively, and [3H]PGF specifically bound to the membranes of cells expressing zebrafish FP1 and FP2, with a Kd of 6.5 and 1.6 nM, respectively. U-46619, a stable agonist for human and mouse TP receptors, significantly increased the specific binding of [35S]GTPγS to membranes expressing the zebrafish TP receptor. Upon agonist stimulation, all six receptors showed an increase in intracellular Ca2+ levels, although the increase was very weak in EP1b, and pertussis toxin abolished only the EP3-mediated response. Zebrafish EP3 receptor also suppressed forskolin-induced cAMP formation in a pertussis toxin-sensitive manner. In association with the low structural conservation with mammalian receptors, most agonists and antagonists specific for mammalian EP1, EP3 and TP failed to work on each corresponding zebrafish receptor. This work provides further insights into the diverse prostanoid actions mediated by their receptors in zebrafish.  相似文献

Effect of various prostaglandins on the release of arachidonic acid from [14C]arachidonic acid labeled fibroblasts was studied. Prostaglandin(PG) F was found to enhance the release of radioactive arachidonic acid from the cells. The stimulatory effect was dose dependent, and was greater than that of bradykinin. The active compounds can be ranked in potency for the release of arachidonic acid from the pre-labeled cells per cent of control: PGF(200.1%)>PGF (141.8%)>PGD2 (137.1%)>thromboxane B2 (113.7%)>PGE2 (109.4%). On the other hand, PGI2 showed a strong inhibitory effect on the arachidonic acid release from the pre-labeled cells (the value was only 69% of the control), while 6-ketoPGF, an end metabolite of PGI2, had no effect.  相似文献

Suspensions of dispersed bovine luteal cells prepared by collagenase digestion of luteal tissue specifically bound [3H]Prostaglandin (PG) E1 and [3H]PGF. While the number of sites per cell (~ 1.8 × 105) were about the same for both [3H]PGs, the apparent Kds were different: [3H]PGE1 ? 2.4 nM; [3H]PGF ? 11 nM. The [3H]PGs binding was inhibited in a dose-dependent manner in the presence of increasing concentrations of unlabeled PGs. Potency order for inhibition of [3H]PGE1 binding was: PGE2 > PGE1 > PGF > PGF. The corresponding data for [3H]PGF was: PGF > PGF > PGE2 > PGE1. While [3H]PGE1 and [3H]PGF bind to their own receptors with high affinity, their affinities for each other's binding were extremely low. Thus, these results demonstrate that luteal cells, like plasma membranes isolated from luteal tissue, contain receptors for PGEs and PGF which are discrete with respect to specificity and affinity.  相似文献

Prostaglandins (PGs) are signaling lipids derived from arachidonic acid (AA), which is metabolized by cyclooxygenase (COX)-1 or 2 and class-specific synthases to generate PGD2, PGE2, PGF, PGI2 (prostacyclin), and thromboxane A2. PGs signal through G-protein coupled receptors (GPCRs) and are important modulators of an array of physiological functions, including systemic inflammation and insulin secretion from pancreatic islets. The role of PGs in β-cell function has been an active area of interest, beginning in the 1970s. Early studies demonstrated that PGE2 inhibits glucose-stimulated insulin secretion (GSIS), although more recent studies have questioned this inhibitory action of PGE2. The PGE2 receptor EP3 and one of the G-proteins that couples to EP3, GαZ, have been identified as negative regulators of β-cell proliferation and survival. Conversely, PGI2 and its receptor, IP, play a positive role in the β-cell by enhancing GSIS and preserving β-cell mass in response to the β-cell toxin streptozotocin (STZ). In comparison to PGE2 and PGI2, little is known about the function of the remaining PGs within islets. In this review, we discuss the roles of PGs, particularly PGE2 and PGI2, PG receptors, and downstream signaling events that alter β-cell function and regulation of β-cell mass.  相似文献

While many observations indicate that prostaglandins may act as positive regulators of hepatocyte proliferation, the underlying mechanisms are not known. We have examined some of the signal pathways in the growth response induced by prostaglandins in hepatocytes, with particular focus on adenylyl cyclase and phosphoinositide-specific phospholipase C. Adult rat hepatocytes were cultured as primary monolayers in serum-free medium in the presence of EGF and insulin. PGE2 or PGF (added 0-3 h after plating) enhanced the incorporation of [3H]-thymidine into DNA (measured at 50 h); at 100 γM the stimulation was about threefold. PGI2 and PGD2 also showed significant but smaller stimulatory effects. No significant increase in the level of cyclic AMP (cAMP) was detected in response to any of the prostaglandins. Low concentrations of glucagon (0.1-10 nM), a potent activator of hepatic adenylyl cyclase, or 8-bromo-cAMP (0.1-10 γM) enhanced the DNA synthesis. When 8-bromo-cAMP was used in maximally effective concentrations, no further stimulation was obtained by combining it with glucagon, whereas the effects of PGE2 and 8-bromo-cAMP were completely additive. All the prostaglandins also showed additivity with the effect of glucagon on the DNA synthesis. PGE2, PGF, PGI2, and PGD2 increased intracellular inositol-1,4,5-trisphosphate (InsP3), with a relative order of efficacy roughly corresponding to their activity as stimulators of DNA synthesis. Increases in cytosolic free Ca2+, as measured in single cells, were elicited in a majority of the hepatocytes by all these prostaglandins at 1 γM. Supramaximal concentrations of vasopressin, a strong activator of phospholipase C in hepatocytes, acted additively with PGE2 on the DNA synthesis. Pretreatment of the hepatocytes with a concentration of pertussis toxin that prevented the inhibitory effect of PGE2 on glucagon-induced cAMP accumulation did not abolish the ability of PGE2 to stimulate the DNA synthesis. The results do not support a role for adenylyl cyclase activation in the stimulatory effect of prostaglandins on hepatocyte growth. While the data are compatible with an involvement of phosphoinositide-specific phospholipase C in the growth-promoting effect of prostaglandins in cultured rat hepatocytes, they suggest this may not be the sole mechanism. © 1995 Wiley-Liss, Inc.  相似文献

The effect of exposing platelets to prostaglandin D2 (PGD2) on hormone binding was studied. Incubation of platelets with PGD2 for 2 hr resulted in a decrease in [3H]PGD2 binding that was dose dependent. Inhibition of binding was 14% after incubation with 10?8M PGD2, 19% after incubation with 10?7M PGD2, and 40% after exposure to 10?6M PGD2. This decreased binding (desensitization) was specific for [3H]PGD2 as binding to platelets by [3H]PGE1 and the α-adrenergic antagonist [3H] dihydroergocryptine (DHEC) was comparable to control platelets. Saturation of [3]PGD2 binding to desensitization platelets was at 27 fmole ligand/108 platelets compared to 43 fmoles/108 platelets in control platelets. Half-maximal saturation occured at 20 nM PGD2 both for desensitized and control platelets, suggesting that decreased binding sites rather than altered affinity between ligand and receptor accounted for these results. These platelets had a partial increase in [3H]PGD2 binding a few hours after plasma was washed free of PGD2 with complete resensitization after 24 hr. Since prostaglandins such as PGI2, PGD2, and PGE1 are potent inhibitors of platelet aggregation, decreased binding of platelets to these hormones after prostaglandin exposure may provide a mechanism for altered responsiveness of platelets to aggregating stimuli.  相似文献

The vasoconstrictor effects of PGE2 and PGF are less pronounced on retinal vessels of the newborn than of the adult pig. We tested the hypothesis that the decreased vasomotor response to these prostaglandins might be due to relatively fewer receptors and/or different receptor subtypes (in the case of PGE2) on retinal vessels of the newborn animal. Binding studies using [3H]PGE2 and [3H]PGF revealed that PGE2 (EP) and PGF (FP) receptor densities in retinal microvessel membrane preparations from newborn animals were approximately 25% of those found in vessels from the adult. The Kd for PGF did not differ; however, the Kd for PGE2 was less in newborn than in adult vessels. Competition binding studies using AH 6809 (EP1 antagonist), butaprost (EP2 agonist), M&B 28,767 (EP3 agonist), and AH 23848B (EP4 antagonist) suggested that the retinal vessels of the newborn contained approximately equal number of EP1 and EP2 receptor subtypes whereas the main receptor subtype in the adult vessels was EP1. In addition, PGE2 and butaprost produced comparable increases in adenosine 3′,5′-cyclic monophosphate synthesis in newborn and adult vessels. PGE2, 17-phenyl trinor PGE2 (EP1agonist) and PGF caused a 2.5 to 3-fold greater increase in inositol1,4,5-triphosphate (IP3) formation in adult than in newborn preparations. It is concluded that fewer PGF receptors and an associated decrease in receptor-coupled IP3 formation in the retinal vessels of the newborn could lead to weaker vasoconstrictor effects of PGF on retinal vessels of the newborn than of adult pigs; fewer EP1 receptors (associated with vasoconstriction) and a relatively greater proportion of EP2 receptors (associated with vasodilation) might be responsible for the reduced retinal vasoconstrictor effects of PGE2 in the newborn.  相似文献

Kidney membrane fractions metabolized [1-14C]PGH2 to TXB2, PGE2, PGF, PGD2, 6-keto PGF, and HHT. TXA2, as measured by TXB2, was enzymatically formed in cortex microsomes and was identified by thin layer chromatography and gas chromatography - mass spectrometry. PGH2 caused a labile inhibition of cortical PGE2-stimulated adenylate cyclase. PGE2, PGF, and PGD2 are stimulators of cortical adenylate cyclase. The inability of two thromboxane synthetase inhibitors, imidazole and 9,11-azoprosta-5,13 dienoic acid, to block PGH2 inhibition suggested that TXA2 was not an obligatory intermediate in this process. Therefore, a potential function of cortical PGH2 is inhibition of adenylate cyclase.  相似文献

To determine the influence of prostaglandins on cAMP metabolism in renal papillary collecting tubule (RPCT) cells, intracellular cAMP levels were measured after incubating cells with prostaglandins (PGs) alone or in combination with arginine vasopressin (AVP). PGE1, PGE2 and PGI2, but not PGD2 or PGF, increased intracellular cAMP concentrations. At maximal concentrations (10−5 tthe effects of PGE2 plus PGI2 (or PGE1), but not of PGI2 plus PGE1, were additive suggesting that at least two different PG receptors may be present in RPCT cell populations. Bradykinin treatment of RPCT cells caused an accumulation of intracellular cAMP which was blocked by aspirin and was quantitatively similar to that observed with 10−5 PGE2. PGs, when tested at concentrations (e.g. 10−9 ) which had no independent effect on intracellular cAMP levels, did not inhibit the AVP-induced accumulation of intracellular cAMP in RPCT cells. These results indicate that PGs do not block AVP-induced accumulation of intracellular cAMP in RPCT cells at concentrations of PGs which have been shown to inhibit the hydroosmatic effect of AVP on perfused collecting tubule segments. However, at higher concentrations of PGs (e.g. 10−5 ), the effects of AVP plus PGE1, PGE2, PGI2 or bradykinin on intracellular cAMP levels were not additive. Thus, under certain conditions, there is an interaction between PGs and AVP at the level of cAMP metabolism in RPCT cells.  相似文献

Intact viable bovine luteal cell suspensions prepared by collagenase digestion of luteal tissue were used in studying the selected properties of [3H] prostaglandin (PG) F binding and compared with those observed in plasma membranes. [3H]PGF specific binding to luteal cells was a rapid (K1 = 8.4 × 104M?12αS?1), reversible (K?1 = 1.8 × 10?4S?1) and saturable process at 24°. There was a single class of receptors with an apparent dissociation constant of 10.6 nM and 1.8 × 105 receptors per cell. The presence of increasing amounts of unlabeled PGs inhibited [3H]PGF binding in a dose-dependent manner. The potency order for this inhibition was: (15S) 15-methyl-PGF methyl ester > ICI-80,996 > PGF > ICI-81,008 > PGF > PGE2, (15S) 15-methyl-PGE2 methyl ester > PGF metabolites > other PGs, PGF metabolites and PGE metabolites. Other than the homegeneous nature of binding and a greater association rate in cells, the rest of the [3H]PGF binding properties in cells were in good agreement with those observed in plasma membranes.  相似文献

The biological effects of leukotriene (LT)B4 were compared, on a molar basis, with those LTC4, LTD4, LTE4, 5-hydroxyeicosatetraenoic acid (5-HETE), PGD2, PGE1, PGF, PGI2, 6-oxo-PGF, bradykinin (BK) and angiotensin II (Ang II) on isolated strips of guinea-pig lung parenchyma (GPP) and ileum smooth muscle (GPISM) superfused in series.LTB4 similar to LTC4 and LTD4 on GPP, in relation to potency and contractions induced, but differed from LTE4 in being ten times more active and causing contractions of a much shorter duration of action on this tissue. However, unlike the other LTs, LTB4 produced contractions which were resistant to FPL 55712 (1.9μM) and, when given repeatedly, caused tachyphylaxis in GPP,LTB4 was considerable more active on GPP than the other substances investigated. Further, PGD2, PGF and PGI2 contracted GPP, the order of potency being PGD2 > PGF2α ? PGI2 whereas PGE1 and PGE2 relaxed this tissue. In contrast to all other agonists tested which contracted GPISM, LTD4 displaying the highest activity, LTB4 was inactive on this tissue. 5-HETE and 6-oxo-PGF were inactive on both GPP and GPISM.On the basis of differential effects of LTB4 on GPP and GPISM, this assay repressents a simple and selective means to distinguish LTB4-like materials from other naturally-occuring substances likely to be generated in inflammatory fluids.  相似文献

Influences of prostaglandin(PG)s on electrophoretic mobilities and aggregation of rabbit platelets were studied. The PGs studied (PGI2, PGE1, PGD2, PGE2, PGF, PGA2 and PGA1) had no effect on platelet electrophoretic mobility. However, both PGE1 and PGI2 in 0.3 and 3.0 μM inhibited ADP-induced aggregation and ADP-induced decrease in the mobility. PGD2 in 0.3 and 3.0, and PGE2 in 30 μM inhibited the aggregation but did not depress the ADP-induced decrease in the mobility. PGF, PGA2 and PGA1 had no effect on the decrease in electrophoretic mobility and on the aggregation caused by ADP.  相似文献

Stimulation of cultured rabbit endometrial cells by one of the rabbit endometrial cell culture proliferation factors, prostaglandin F (PGF), resulted in a very rapid increase in the intracellular levels of [3H]-inositol triphosphate (IP3), [3H]-inositol biphosphate (IP2), and [3H]-inositol monophosphate (IP1) in cells prelabeled with [3H]-inositol. These increases in inositol phosphate levels were detected in periods of stimulation as short as 30 seconds, reached a maximum by 1 1/2?2 min and declined to control levels by 6–10 min. The stimulation was dose-dependent with maximal increases observed near 10?6 M PGF. The cholinergic agent, carbachol, also led to time and dose-independent increases in IP3. Lithium, cadmium, silver, copper, and zinc ions had no effect either on the breakdown of IP3 or on the accumulation of IP1. In contrast, vanadate at 10?6 or 10?5 M did lead to a decrease in the breakdown of IP1 and a concomitant increase in IP1, IP2, and IP3. PGF was found previously to induce an increase in rabbit endometrial cell DNA synthesis which was inhibited by concomitant or prior addition of prostaglandin E1 (PGE1). PGE1, in a dose-dependent manner, was found to inhibit the observed IP3 increase by PGF at 1 1/2 min of stimulation. PGF treated and control cultures did not differ in cAMP or cGMP levels, cellular 45Ca uptake, nor cellular 22Na uptake. We propose that IP3 may be one of the intracellular messenger(s) synthesized following the treatment of rabbit endometrial cell cultures with the proliferation agent PGF and that it may play a crucial role with cAMP in growth regulation.  相似文献

The exogenous and endogenous syntheses of prostaglandins (PG's) by the cochlea of adult mongolian gerbils were studied . After incubation of the whole membraneous cochlea with [3H]-arachidonic acid (AA), syntheses of PGF, 6-keto PGF, PGE2, thromboxane (TX) B2 and PGD2 were evidenced in this order. The synthesis of radioactive PG's was almost completely inhibited by incubation with 10−5 M indomethacin. No significant amounts of those PG's were detected by radioimmunoassay (RIA) in the cochlea obtained from animals killed by microwave irradiation at 5.0 kw for 0.8 sec. However, when the homogenate of the whole membraneous cochlea obtained from animals without microwave irradiation was incubated at 37°C for 0–15 min, PGD2, PGE2, PGF2α and 6-keto PGF1α were found to be formed from endogenous AA in the cochlea by RIA. PG's were formed already at 0 time to considerable level (PGD2, PGF2α and 6-keto PGF1α, 90–120 pg/cochlea; PGE2, 370 pg/cochlea), reached to the maximum level (PGD2, PGF2α and 6-keto PGF1α, 170–200 pg/cochlea; PGE2, 500 pg/cochlea) at a 5-min incubation, and then gradually decreased. On the other hand, the amount of TXB2 was lower than the detection limit by RIA (<50 pg/cochlea) even after the incubation. The cochlea was dissected into three parts: organ of Corti + modiolus (OC + M), lateral wall (LW), and cochlear nerve (CN), and then PG's formed by these tissues were determined after a 5-min incubation of the homogenates. In the CN and OC + M, PGE2 was the major PG (100 and 160 pg/tissue, respectively), and the amounts of PGD2, PGF2α and 6-keto PGF1α were about of those of PGE2. In the LW, the amounts of PGD2, PGE2, PGF2α and 6-keto PGF1α were about the same level (70–100 pg/LW).  相似文献

New bone formation is associated with an increase in blood flow by the invasion of capillaries. Endothelial cells that line the capillaries can produce paracrine factors that affect bone growth and development, and in turn, could be affected by products produced by bone cells, in particular the osteoblasts. Since osteoblasts produce prostaglandins E2 and F (PGE2, PGF), it was investigated if these PGs were agonists to bone-derived endothelial cells (BBE) by assessing changes in cAMP and free cytosolic calcium concentration ([Ca2+]i) second messenger generation. We found that confluent cultures of BBE cells, a clonal endothelial cell line derived from bovine sternal bone, responded to 1 μM PGE2 by an increase in cAMP. PGF at the same concentration was less potent in stimulating an increase in cAMP production in confluent BBE cells. Subconfluent cells with a morphology similar to that of fibroblastic cells were not as sensitive to PGE2-stimulated cAMP generation. PGF failed to elicit any cAMP production in subconfluent cultures. PGE2 and PGF both stimulated an increase in [Ca2+]i concentration in a dose-dependent manner. The potency of PGE2 was similar to that of PGF in stimulating an increase in [Ca2+]i. The Ca2+ response was mostly independent of extracellular Ca+, was unchanged even with prior indomethacin treatment, was unaffected by caffeine pretreatment, but was abolished subsequent to thapsigargin pretreatment. The PG-induced increase in [Ca2+]i was also dependent on the confluency of the cells. In a subconfluent state, the responses to PGE2 or PGF were either negligible, or only small increases in [Ca2+]i were noted with high concentrations of these two PGs. Consistent, dose-dependent increases in [Ca2+]i were stimulated by these PGs only when the cells were confluent and had a cobblestoned appearance. Since it was previously demonstrated that BBE cells respond to parathyroid hormone (PTH) by the production of cAMP, we tested if bovine PTH(1-34) amide bPTH(1—34) also increased [Ca2+]i in these cells. No change in [Ca2+]i was found in response to bPTH (1—34), although bPTH (1—34) stimulated a nine to tenfold increase in cAMP. We conclude that BBE cells respond to PGE2 and PGF but not to bPTH(1—34) by an increase in [Ca2+]i probably secondary to stimulation of phospholipase C and that the cAMP and [Ca2+]i second messenger responses in BBE cells are dependent on the state of confluency of the cells. © 1994 Wiley-Liss, Inc.  相似文献

Infusion of [14C] arachidonic acid (AA) into the isolated, Tyrode perfused rabbit spleen resulted in the release of a substance into the venous effluent with the musculotropic activity and chromatographic properties of prostaglandin (PG)E2. Smaller amounts of radioactive materials with the chromatographic properties of PGF, 6-keto-PGF, and PGD2 were also released. The radiolabeled material released in largest amounts from the spleen was identified as PGE2 on the basis of: 1) Co-chromatography with PGE2 in three solvent systems, 2) Conversion of the radioactive material and of authentic [3H] PGE2 to similar products by treatment with sodium borohydride and with potassium hydroxide, and 3) Stability of the musculotropic activity in Tyrode solution at 37°C. The effect of 7-fluoro proscyclilin (PGI2-F), a chemically stable analogue of prostacyclin, on cAMP accumulation in and [3H]PGE binding to mastocytoma P-815 cells was compared with those of the Na salt and methyl ester of prostacyclin (PGI2Na or PGI2Me), which are rapidly inactivated in aqueous solution or metabolized in the tissue.PGIF was as effective as PGI2Me, and slightly less effective than PGI2Na in stimulating cAMP accumulation in mastocytoma cells and rabbit platelets. PGI2F was also more stable than PGI2Me or PGI2Na, and retained its original cAMP elevating activity even after incubation with or without cells for 4 h at 37°C. Cells which had been exposed to PGI2F and then washed free of unbound reagent continued to produced cAMP for more than 3 h. PGI2F was also as effective as PGE1 or PGE2 in displacing [3H]PGE2 bound to the cells. Non-competitive inhibition by PGI2F or PGI2Me of [3H]PGE2 binding to the cells, with apparent Kis of 1.29 μM and 1.13 μM, respectively, indicates the presence of different receptors for PGE2 and for PGI2F or PGI2Me in mastocytoma P-815 cells.  相似文献

The actions of prostacyclin (PGI2) and its stable metabolite 6-OXO-PGF were investigated in strips of normal human uterus and in fallopian tubes.Both compounds were also compared with natural prostaglandins (PGE2, PGF and PGD2).PGI2 showed biphasic response both in uterus and fallopian tubes qualitatively and quantitatively similar to that induced by PGE2 and PGD2; prostacyclin was also able to inhibit the spasmus induced by PGF but not that induced by BaCl2 and vasopressin.6-OXO-PGF on the other hand induced only small contractions on both tissues investigated.The authors discusse the possible implication of these findings in the physiology of the reproductive system.  相似文献
