Cold temperature acclimation in strawberry (Fragaria virginiana) leaves apparently involves the alteration of cellular osmotic properties. Alterations in leaf osmotic potential were closely correlated with alterations in soluble carbohydrate content of the leaf tissue and changing temperatures. Leaf starch content was inversely related to soluble carbohydrate levels, suggesting that starch is a partial source of osmoticum during osmotic adjustment associated with cold temperature stress. Free amino acid changes were more closely linked to senescence and growth processes while changes in ion content suggested a rapid mobilization of solutes at the onset of freezing temperatures. This was supported by changes in whole plant gradients in leaf osmotic potential before and after exposure to freezing temperatures. In terms of freezing resistance and the role of osmotic adjustment in the development of resistance, it was found that of all leaves undergoing osmotic adjustment only the younger leaves survived, suggesting an age-dependent component to freezing resistance in leaves. Freezing resistance appears to involve alterations in several cellular properties that act in concert to confer a hardy state of the tissue. Although osmotic adjustment may be an important component of the final combination of cellular properties, this study indicates that solute accumulation does not function alone to confer freezing resistance.  相似文献

Primary roots of maize (Zea mays L. cv WF9 × Mo17) seedlings growing in vermiculite at various water potentials exhibited substantial osmotic adjustment in the growing region. We have assessed quantitatively whether the osmotic adjustment was attributable to increased net solute deposition rates or to slower rates of water deposition associated with reduced volume expansion. Spatial distributions of total osmotica, soluble carbohydrates, potassium, and water were combined with published growth velocity distributions to calculate deposition rate profiles using the continuity equation. Low water potentials had no effect on the rate of total osmoticum deposition per unit length close to the apex, and caused decreased deposition rates in basal regions. However, rates of water deposition decreased more than osmoticum deposition. Consequently, osmoticum deposition rates per unit water volume were increased near the apex and osmotic potentials were lower throughout the growing region. Because the stressed roots were thinner, osmotic adjustment occurred without osmoticum accumulation per unit length. The effects of low water potential on hexose deposition were similar to those for total osmotica, and hexose made a major contribution to the osmotic adjustment in middle and basal regions. In contrast, potassium deposition decreased at low water potentials in close parallel with water deposition, and increases in potassium concentration were small. The results show that growth of the maize primary root at low water potentials involves a complex pattern of morphogenic and metabolic events. Although osmotic adjustment is largely the result of a greater inhibition of volume expansion and water deposition than solute deposition, the contrasting behavior of hexose and potassium deposition indicates that the adjustment is a highly regulated process.  相似文献

This study aimed to assess the accumulation of organic and inorganic solutes and their relative contribution to osmotic adjustment in roots and leaves of Jatropha curcas subjected to different water deficit intensity. Plants were grown in vermiculite 50% (control), 40%, 30%, 20% and 10% expressed in gravimetric water content. The water potential, osmotic potential and turgor potential of leaves decreased progressively in parallel to CO2 photosynthetic assimilation, transpiration and stomatal conductance, as the water deficit increased. However, the relative water content, succulence and water content in the leaves did not show differences between the control and stressed plants, indicating osmotic adjustment associated with an efficient mechanisms to prevent water loss by transpiration through stomatal closure. The K+ ions had greater quantitative participation in the osmotic adjustment in both leaves and roots followed by Na+ and Cl, while the NO3 ion only showed minor involvement. Of the organic solutes studied, the total soluble sugars showed the highest relative contribution to the osmotic adjustment in both organs and its concentration positively increased with more severe water deficit. The free amino acids and glycinebetaine also effectively contributed to the osmotic potential reduction of both the root and leaves. The role of proline was quantitatively insignificant in terms of osmotic adjustment, in both the control and stressed roots and leaves. Our data reveal that roots and leaves of J. curcas young plants display osmotic adjustment in response to drought stress linked with mechanisms to prevent water loss by transpiration by means of the participation of inorganic and organic solutes and stomatal closure. Of all the solutes studied, soluble sugars uniquely display a prominent drought-induced synthesis and/or accumulation in both roots and leaves.  相似文献

Eight cultivars Sorghum bicolor (L.) Moench were grown as callus cultures under induced, prolonged water stress (8 weeks), with polyethylene glycol in the medium. Concentrations of soluble carbohydrates and organic acids in callus were measured at the end of the growth period to determine differences in response to prolonged water stress. Sucrose, glucose, fructose, and malate were the predominant solutes detected in all callus at all water potentials. All cultivars had high levels of solutes in the absence of water stress and low levels in the presence of prolonged water stress. However, at low water potentials, low levels of solutes were observed in drought-tolerant cultivar callus and high solute levels were observed in drought-susceptible cultivar callus. Estimated sucrose concentrations were significantly higher in water-stressed, susceptible cultivar callus. Large solute concentrations in susceptible cultivar callus were attributed to osmotic adjustment and/or reduced growth during water stress.  相似文献

Marine hyperthermophiles accumulate small organic compounds, known as compatible solutes, in response to supraoptimal temperatures or salinities. Pyrococcus furiosus is a hyperthermophilic archaeon that grows optimally at temperatures near 100°C. This organism accumulates mannosylglycerate (MG) and di-myo-inositol phosphate (DIP) in response to osmotic and heat stress, respectively. It has been assumed that MG and DIP are involved in cell protection; however, firm evidence for the roles of these solutes in stress adaptation is still missing, largely due to the lack of genetic tools to produce suitable mutants of hyperthermophiles. Recently, such tools were developed for P. furiosus, making this organism an ideal target for that purpose. In this work, genes coding for the synthases in the biosynthetic pathways of MG and DIP were deleted by double-crossover homologous recombination. The growth profiles and solute patterns of the two mutants and the parent strain were investigated under optimal growth conditions and also at supraoptimal temperatures and NaCl concentrations. DIP was a suitable replacement for MG during heat stress, but substitution of MG for DIP and aspartate led to less efficient growth under conditions of osmotic stress. The results suggest that the cascade of molecular events leading to MG synthesis is tuned for osmotic adjustment, while the machinery for induction of DIP synthesis responds to either stress agent. MG protects cells against heat as effectively as DIP, despite the finding that the amount of DIP consistently increases in response to heat stress in the nine (hyper)thermophiles examined thus far.  相似文献

The effect of a short period of saline stress was studied in two phenotypically different cultivars, one of normal fruit-size (L. esculentum cv. New Yorker) and one of cherry fruit-size (L. esculentum var.cerasiforme cv. PE-62). In both cultivars the relative growth rate (RGR) and the leaf area ratio (LAR) decreased following salinisation. The leaf turgor potential (p) and the osmotic potential at full turgor (os) decreased to the same extent in both cultivars. However, the contributions of organic and inorganic solutes to the osmotic adjustment was different between cultivars. New Yorker achieved the osmotic adjustment by means of the Cl and Na+ uptake from the substrate, and by synthesis of organic solutes. In the cherry cultivar organic solutes did not contribute to the osmotic adjustment, instead, their contribution decreased after salinisation. After the salt stress was removed, the water stress disappeared, the content of organic solutes decreased in plants of both cultivars and, therefore, their growth was not retarded by the diversion of resources for the synthesis of organic solutes. However, the toxic effects of the Cl and Na+ did not disappear after removal of the salt stress, and the net assimilation rate (NAR) and the rate of growth (RGR) did not recover.  相似文献

The absolute pressure in conducting xylem vessels of roots of 2-week-old, slowly transpiring intact maize plants (bathed in nutrition medium) was determined to be +0·024 ± 0·044 MPa using the xylem pressure probe. When the roots were subjected to osmotic stress (NaCI, KCI or sucrose), the xylem pressure decreased immediately and became more negative. However, the response of xylem pressure to osmotic stress was considerably attenuated, indicating that the radial reflection coefficients, σ13 of the maize root for these solutes were rather low (between 0·2 and 0·4 depending on the concentration of the osmoticum). The low values of a, may be caused (partly) by unstirred layer effects. In repeated osmoticum/nutrition regimes a complex pattern of changes in xylem pressure was observed which was apparently linked to the interplay between transpiration and (passive and/or active) solute loading of the xylem. These processes were not observed when the roots were subjected to osmotic stress after excision. In this case, a biphasic response was observed comparable to that found for excised roots using the root pressure probe.  相似文献

Cells of Nicotiana tabacum L. var Wisconsin 38 adapted to NaCl (up to 428 millimolar) which have undergone extensive osmotic adjustment accumulated Na+ and Cl as principal solutes for this adjustment. Although the intracellular concentrations of Na+ and Cl correlated well with the level of adaptation, these ions apparently did not contribute to the osmotic adjustment which occurred during a culture growth cycle, because the concentrations of Na+ and Cl did not increase during the period of most active osmotic adjustment. The average intracellular concentrations of soluble sugars and total free amino acids increased as a function of the level of adaptation; however, the levels of these solutes did not approach those observed for Na+ and Cl. The concentration of proline was positively correlated with cell osmotic potential, accumulating to an average concentration of 129 millimolar in cells adapted to 428 millimolar NaCl and representing about 80% of the total free amino acid pool as compared to an average of 0.29 millimolar and about 4% of the pool in unadapted cells. These results indicate that although Na+ and Cl are principal components of osmotic adjustment, organic solutes also may make significant contributions.  相似文献

Salt-tolerant selected cells of Shamouti orange (Citrus sinensis) and Sour orange (Citrus aurantium) grew considerably better than nonselected cells at any NaCl concentration tested up to 200 millimolar. Also, the growth response of each treatment was identical in the two species. However, the performance of cells of the two species under osmotic stress induced by polyethylene glycol (PEG), which is presumably a nonabsorbed osmoticum, was significantly different. The nonselected Shamouti cell lines were significantly more sensitive to osmotic stress than the selected cells. The salt adapted Shamouti cells were apparently also adapted to osmotic stress induced by PEG. In Sour orange, however, the selected lines had no advantage over the nonselected line in response to osmotic stress induced by PEG. This response was also similar quantitatively to the response of the selected salt-tolerant Shamouti cell line. It seems that the tolerance to salt in Shamouti, a partial salt excluder, involves an osmotic adaptation, whereas in Sour orange, a salt accumulator, such an adaptation apparently does not occur. PEG-induced osmotic stress causes an increase in the percent dry weight of salt-sensitive and salt-tolerant cells of both species. No such increase was found under salt stress. The size of control and stressed cells is not significantly different.  相似文献

Abstract. The effect of accumulation of 3- O -methylglucose (MG) on growth and steady-stale concentrations of the endogenous osmotic solutes proline and sucrose was studied in Chlorella emersonii grown at external osmotic pressure (II) of 0.08-1.64 MPa. NaCL was used as osmoticum. The total solute content of the cells was manipulated by supplying 2 mol m−3 MG for 4 and 48 h. MG accumulated to 50–230 mol m−3 within 4h and was not metabolized. Uptake of MG resulted in decreases in concentrations of proline and sucrose, the two solutes mainly responsible for osmotic adaptation of C. emersonii to high II. After 4 or 48 h growth in the presence of MG, the decreases in concentration of proline and sucrose were as predicted from the contribution of MG to the total solute content of the cell.  相似文献

Understanding trees adaptation to arid, saline conditions is a major challenge for catchment revegetation in Australia. The accumulation of low molecular weight solutes is an established response of trees to the effects of salt and/or drought stress. Recent studies have shown that quercitol – a cyclitol – contributes significantly towards the adjustment of osmotic potential in some species of Eucalyptus. The present study investigated the role of quercitol in leaf tissues of Eucalyptus leptophylla (F. Muell) under fluctuating environmental stresses. Analysis of leaf tissues from trees growing at distances between 0 and 125 m from hyper-saline lakes suggests that quercitol contributes significantly to the adjustment of osmotic potential induced by drought in E. leptophylla. The presence of substantial concentrations of quercitol in xylem sap suggests that quercitol plays additional roles in signalling amelioration of osmotic stress in myrtaceous species. Quercitol concentrations fluctuate in both xylem and leaf tissues on a seasonal basis, suggesting a form of environmental regulation of solutes. The capacity of soil profiles to store rainwater, rather than proximity to hyper-saline groundwater largely determined osmotic stress in studied trees. Understanding such avoidance/tolerance mechanisms will be crucial to advance tree selection and breeding for stress tolerance.  相似文献

Drought stress is one of the most important factors limiting the survival and growth of plants in the harsh karst habitats of southwestern China. Detailed knowledge about the ecophysiological responses of native plants with different growth forms to drought stress could contribute to the success of re-vegetation programs. Two shrubs, Pyracantha fortuneana and Rosa cymosa, and four trees, Broussonetia papyrifera, Cinnamomum bodinieri, Platycarya longipes and Pteroceltis tatarinowii, were randomly assigned to four drought treatments, i.e. well-watered, mild drought stress, moderate drought stress, and severe drought stress. Midday water potential, the maximum quantum efficiency of PSII photochemistry (Fv/Fm), pigments, osmotic solutes (soluble sugars and proline), cellular damages, and antioxidant enzymes (superoxide dismutase, catalase and peroxidase) were investigated. Drought stress significantly decreased pigments content, but increased the ratio of carotenoids to total chlorophylls in the studied species. After prolonged severe drought stress, the two shrubs exhibited higher Fv/Fm, less reductions of midday water potential, and lower increases of malondialdehyde content and ion leakage than the four trees. Prolonged severe drought stress largely decreased accumulations of osmotic solutes and activities of antioxidant enzymes in the four trees, but significantly increased proline content and superoxide dismutase activity in the two shrubs and peroxidase activity in P. fortuneana. The positive relationships were observed among activities of antioxidant enzymes, and between contents of osmotic solutes and activities of antioxidant enzymes. These findings suggested that the two shrubs had higher tolerance to severe drought stress than the four trees due to higher capacities of osmotic adjustment and antioxidant protection.  相似文献

In this study, the effect of irrigation intervals (drought stress) on growth, predawn xylem water potential (Ψ w), the osmotic potential at full turgor (Ψπ 100), the osmotic potential at the turgor loss point (Ψπ TLP), osmotic adjustment and osmotic solutes (soluble sugars and proline) of Pinus pinea L. seedlings were examined. An experiment was carried out under greenhouse conditions using four watering treatments (control, 7-, 14- and 21-day irrigation intervals) in the first growth season; from mid-July to early November. Results showed that irrigation interval had significant effect on growth characteristics, Ψ w, water relation parameters, and osmotic solutes. The increasing irrigation interval significantly decreased the seedling height, root collar diameter, root, stem and needle dry weight, number of lateral branches, root percentage, root:shoot ratio and diameter:height ratio. Ψ w and total soluble sugars decreased while proline content increased with the increase of drought stress. The Ψπ 100 and Ψπ TLP significantly decreased in drought-stressed seedlings compared to control (no stress) seedlings. The results suggest that the impact of drought stress increased with the increase of irrigation interval. Therefore, in the drought-stressed P. pinea seedlings were indicated osmotic adjustment by increasing the proline content and decreasing Ψπ 100 and Ψπ TLP during drought stress. Growth decreased under drought stress conditions in P. pinea seedlings.  相似文献

Water relations of growing segments of maize (Zea mays L.) coleoptiles were investigated with osmotic methods using either mannitol (MAN) or polyethylene glycol 6000 (PEG) as external osmotica. Segments were incubated in MAN or PEG solutions at 0 to - 15 bar water potential (Ψo) and the effects were compared on elongation growth, osmotic shrinkage, cell sap osmolality (OC), and osmotic pressure (πi). The nonpenetrating osmoticum PEG affects πi in agreement with Boyle-Mariotte's law, i.e. the segments behave in principle as ideal osmometers. There is no osmotic adjustment in the Ψo range permitting growth (0 to −5 bar) nor in the Ψo range inducing osmotic shrinkage (−5 to −10 bar). Promoting growth by auxin (IAA) has no effect on the osmotic behavior of the tissue toward PEG. In contrast to PEG, MAN produces an apparent increase in πi accompanied by anomalous effects on segment elongation and shrinkage leading to a lower value for Ψo which establishes a growth rate of zero and to an apparent recovery from osmotic shrinkage after 2 hours of incubation. These effects can be quantitatively attributed to uptake of MAN into the tissue. MAN is taken up into the apoplastic space and the symplast as revealed by a large temperature-dependent component of MAN uptake. It is concluded that MAN, in contrast to PEG, is unsuitable as an extemal osmoticum for the quantitative determination of water relations of growing maize coleoptiles.  相似文献

As a response to hyperosmotic stress bacterial cells accumulate compatible solutes by synthesis or by uptake. Beside the instant activation of uptake systems after an osmotic upshift, transport systems show also a second, equally important type of regulation. In order to adapt the pool size of compatible solutes in the cytoplasm to the actual extent of osmotic stress, cells down-regulate solute uptake when the initial osmotic stress is compensated. Here we describe the role of the betaine transporter BetP, the major uptake carrier for compatible solutes in Corynebacterium glutamicum, in this adaptation process. For this purpose, betP was expressed in cells (C. glutamicum and Escherichia coli), which lack all known uptake systems for compatible solutes. Betaine uptake mediated by BetP as well as by a truncated form of BetP, which is deregulated in its response to hyperosmotic stress, was dissected into the individual substrate fluxes of unidirectional uptake, unidirectional efflux and net uptake. We determined a strong decrease of unidirectional betaine uptake by BetP in the adaptation phase. The observed decrease in net uptake was thus mainly due to a decrease of Vmax of BetP and not a consequence of the presence of separate efflux system(s). These results indicate that adaptation of BetP to osmotic compensation is different from activation by osmotic stress and also different from previously described adaptation mechanisms in other organisms. Cytoplasmic K+, which was shown to be responsible for activation of BetP upon osmotic stress, as well as a number of other factors was ruled out as triggers for the adaptation process. Our results thus indicate the presence of a second type of signal input in the adaptive regulation of osmoregulated carrier proteins.  相似文献

When intact roots of lentil (Lens culinaris Med.) are subjected to severe osmotic stress by treatment with a solution of low water potential, they immediately begin to shrink. Within 10 to 15 minutes, shrinkage ceases, and within 20 minutes, the roots resume growth. The time lag between application of osmoticum and resumption of growth varies from about 10 to 30 minutes over the range of external water potentials of −2 to −12.4 bars. For external water potentials as low as −8.7 bars the new steady rate of growth in the presence of osmoticum is approximately equal to that prevailing before application of osmoticum. For external water potentials between −8.7 and −13 bars growth resumes, but the new rate is less than that prior to addition of osmoticum. Measurements of changes in the internal solute content during adaptation show that the solute content of the root increases but that the magnitude of the increase is, by itself, insufficient to account for the resumption of rapid growth.  相似文献

The influence of water potential on linear mycelial growth, secretion, and the in vitro activities of enzymes β-glucosidase, cellobiohydrolase, β-xylosidase, exochitinase, and chymotrypsin of Trichoderma harzianum strain T66 was studied at different temperatures. Nearly linear correlation was found between water potential and colony growth rate at both 25°C and 10°C, with higher growth rates at the higher temperature and higher water potentials. The amounts of enzyme secretion depended on the water potential and not on the quality of salt (NaCl or KCl) used as osmoticum. Enzyme activities were significantly affected by water potential. Significant enzyme activities were measured for most of the enzymes even at −14.800 megapascal (MPa), which is below the water potential where mycelial growth ceased. These results suggest the possibility of using mutants with improved xerotolerance for biocontrol purposes in soils with lower water potentials. 本文用PEG模拟水分亏缺对春小麦红芒麦和绵阳11号胚芽伸长过程中生长、膨压、渗透势、水势和渗透调节能力与ATP含量、能荷变化及能量代谢间的关系进行了研究。结果表明,通过降低能荷,改变分解代谢与合成代谢的比率,使渗透调节物质积累,增加了幼苗的吸水能力,从而使其在一定的ATP能量水平上维持缓慢生长;抗旱品种红芒麦在水分亏缺下成苗速率较快,能保持一定的ATP能量水平和能荷值,渗透调节和吸水能力都比较强。  相似文献

Drought stress is one of the abiotic stresses and it may alter plant growth, metabolism and yield. The present study aims to analyze the growth, chlorophyll pigments, osmotic adjustment and antioxidative enzymes activity in Panicum sumatrense under drought stress. The control was irrigated daily and treated plants were irrigated at 4-, 7-, 10-, 13-day intervals. Later, they were irrigated at 3-day interval up to 70 DAS. The root and leaf samples were collected on 30 DAS, 50 DAS and 70 DAS and used for analysis. The root length increased gradually in all drought treatments at all growth stages of P. sumatrense. The chlorophyll pigments and plant height showed a reduction in 13 DID treatment when compared to all treatment. Compatible solutes like proline, glycine betaine and free amino acid increased in all drought treatment when compared to control at 70 DAS. Furthermore, stress treatment caused an increase in activity of antioxidant enzymes like superoxide dismutase, catalase and peroxidase. Panicum sumatrense possess many growth and physiological drought tolerance characters which can be used in future breeding program.  相似文献
