Pollen monitoring is an important and widely used tool in allergy research and creation of awareness in pollen‐allergic patients. Current pollen monitoring methods are microscope‐based, labour intensive and cannot identify pollen to the genus level in some relevant allergenic plant groups. Therefore, a more efficient, cost‐effective and sensitive method is needed. Here, we present a method for identification and quantification of airborne pollen using DNA sequencing. Pollen is collected from ambient air using standard techniques. DNA is extracted from the collected pollen, and a fragment of the chloroplast gene trnL is amplified using PCR. The PCR product is subsequently sequenced on a next‐generation sequencing platform (Ion Torrent). Amplicon molecules are sequenced individually, allowing identification of different sequences from a mixed sample. We show that this method provides an accurate qualitative and quantitative view of the species composition of samples of airborne pollen grains. We also show that it correctly identifies the individual grass genera present in a mixed sample of grass pollen, which cannot be achieved using microscopic pollen identification. We conclude that our method is more efficient and sensitive than current pollen monitoring techniques and therefore has the potential to increase the throughput of pollen monitoring.  相似文献

植物雄性不育既是研究植物生殖生物学重要的植物学性状也是研究作物杂种优势利用重要的农艺性状,在遗传和分子生物学中具有重要地位。以模式植物拟南芥和水稻为主,对植物雄性不育的控制基因和相关分子机理已有众多进展,按照花药发育时期和雄性败育的表现形式可以归纳为减数分裂异常、胼胝质代谢异常、绒毡层发育异常、花粉壁发育异常、花药开裂异常,以及其它类型的雄性不育。在不育相关基因中,导致胼胝质代谢异常、绒毡层发育异常和花粉壁发育异常的基因往往表现一因多效,一个相关基因的突变会产生复合表型。关于植物雄性不育相关基因的研究表明,雄性器官和小孢子形成过程中的任何相关基因的改变,均可导致雄性不育的产生。本文总结了植物核基因雄性不育的研究进展,以期促进不同物种间雄性不育基因的比较分析,使植物雄性不育研究更加深入。  相似文献

Summary Several unrelated compounds are known to selectively inhibit the development of the male gametophyte. When applied at suitable dosages to plants at the appropriate stages of anther development, these substances block the formation of fertile pollen. The affected stage of pollen development is characteristic of the specific chemical structure of the compound, ranging from effects on microspore meiosis to the formation of pollen defective in the ability to germinate or fertilize. The range of effects mediated by these substances, and by known male-sterile mutants, indicates that microspore development has several critical phases that are particularly sensitive to fatal inhibition. We propose that chemical inhibitors of pollen development deserve attention as tools for elucidating the regulation of pollen development.  相似文献

In order to conduct studies concerning genetic variability of pollen beetles (Meligethes aeneus), a genotyping protocol was established. No genome information is available for pollen beetles so the amplified fragment length polymorphism (AFLP) technique was chosen since it does not depend on any prior sequence information of the samples and also is a sensitive and robust technique. However, several modifications were needed in order to adapt the method for analysis of pollen beetles. Basic modifications included (i) alterations of DNA purification, (ii) use of two six-cutter restriction enzymes, (iii) and modified PCR conditions. This protocol resulted in a favourable number of fragments of an appropriate size range for standard gel analysis by a DNA sequencer applicable to a single insect and even body parts enabling different assays to be conducted on a single specimen. Pollen beetles from different areas of Sweden were analysed to verify the reproducibility and efficacy of the protocol as well as for phenetic analysis. The high reproducibility of the modified AFLP protocol allows it to be used as a reliable tool for genotype analysis of pollen beetles.  相似文献

Most recent studies describing pollination networks are based on observed flower visits, and few have explicitly tested if the floral visitors actually carry pollen. Since floral visitors can vary in their ability to remove and transfer pollen, it is important to show that visitation patterns reflect effective pollination. Given the difficulty of measuring per-visit pollen deposition at the community scale, a first step is to examine the amount of conspecific pollen carried by insect visitors. Here I compared the plant–animal visitation network with the pollen-transport network, estimated from insect pollen loads, for a montane meadow community from southern California, USA. Visitation and pollen-transport networks were positively associated with each other in both 2001 and 2002. However, the exclusion of visitors that do not carry any conspecific pollen reveals that pollen-transport networks are more specialized from the plants' perspective and that species are involved in fewer mutualistic interactions compared with estimates derived from visitation frequencies. Although conspecific pollen loads were smaller in 2002, bees tended to carry the largest conspecific loads in both years and were responsible for transporting the most pollen. These results suggest that, although visitation networks are suitable first-order approximations of pollination networks, information on which visitors carry conspecific pollen, and in what amounts, is crucial for distinguishing between antagonistic and mutualistic interactions.  相似文献

第四纪孢粉学利用现代和化石孢粉重建第四纪古植被、古气候和古环境,属于可持续发展的前沿学科领域。本文基于CiteSpace软件数据可视化和计量分析模块,以CNKI (中国知网)数据库与Web of Science核心数据库中1990–2020年间的共计6081篇第四纪孢粉学相关文献为基础,梳理了近30年来第四纪孢粉学的过去和现在,并探讨了未来发展趋势。研究结果表明,近30年第四纪孢粉学领域发文量呈增加趋势,中国的总发文量和国际影响力整体呈增长趋势;第四纪孢粉学研究主要集中于各大高校和科研院所,研究成果有明显的聚集性;中国科学院是外文发文量最多的国际机构。第四纪孢粉学近30年的研究主要涉及古植物学、古地理学、古生态学和古气候学等领域,古植被、古气候、古环境的定性和定量重建一直是国内外学者关注和研究的重点。表土花粉研究、花粉与人类活动、亚洲季风演变和孢粉分类学是近几年发刊的热点。虽然一些关键词如花粉产量、代表性以及现代花粉与植被和气候的定量关系等出现频次低,但对第四纪孢粉学十分重要。未来还需要进一步加强湖泊表层沉积物的花粉研究,探讨花粉与植被、气候的定量关系,为准确解译化石花粉提供坚实基础。...  相似文献

Despite much effort, a robust protocol for in vitro germination of Arabidopsis thaliana pollen has been elusive. Here we show that controlled temperatures, a largely disregarded factor in previous studies, and a simple optimized medium, solidified or liquid, yielded pollen germination rates above 80% and pollen tube lengths of hundreds of microns, with both Columbia and Landsberg erecta (Ler) ecotypes. We found that pollen germination and tube growth were dependent on pollen density in both liquid and solid medium. Pollen germination rates were not substantially affected by flower or plant age. The quartet1 mutation negatively affected pollen germination, especially in the Ler ecotype. This protocol will facilitate functional analyses of insertional mutants affecting male gametophyte function, and should allow detailed gene expression analyses during pollen tube growth. Arabidopsis thaliana can now be included on the list of plant species that are suitable models for physiological studies of pollen tube elongation and tip growth.  相似文献

Previous research on the Black Beauty bush cv. of zucchini has documented a strong positive relationship between the size of the pollen load and the vigor (performance) of the progeny. Here we report the results of three studies designed to test the hypothesis that the previously observed differences in progeny vigor are heritable. Two studies examined the transmission of the pollen load effect to subsequent generations through the ovules (female role). The third study determined if there is genetic variation for pollen performance and if the pollen load effect could be transmitted to a subsequent generation through the pollen (male role). In each of these studies the vigor of the progeny from the subsequent generation was evaluated in the greenhouse and/or the field. The results of these studies reveal (1) that the ability to sire seeds does respond to selection imposed by high pollen loads, (2) that only 23 of the 35 total traits that we measured in the three studies of transmission to subsequent generations changed in the direction predicted by the pollen competition hypothesis, (3) that only 5 of the 35 traits were significantly affected by the size of the pollen load that produced the previous generation (but all 5 were in the direction predicted by the pollen competition hypothesis), and (4) that only one study produced an overall significant difference (MANOVA) attributable to the size of the pollen load that produced the previous generation (but it too was in the direction predicted by the pollen competition hypothesis). From these experiments we conclude that pollen competition appears to play a real but minor role in the production of differences in vigor between progeny arising from low versus high pollen loads. In Black Beauty bush cv. of zucchini, maternal effects, pollen-pistil interactions, or nonrandom patterns of seed abortion must play important roles as well.  相似文献

Susceptibility of pollen to UV-B radiation: an assay of 34 taxa   总被引:1,自引:0,他引:1  
Much of the ultraviolet-B radiation (UV-B) research on plants has concentrated on vegetative plant parts, and only a small fraction has dealt with the reproductive system. The present study analyzed pollen grains of 34 taxa germinated and grown under two levels of UV-B radiation (187 and 460 mW/m2) or no UV-B (control group). Visible radiation at 260 mmol/m/s was present in all treatments. Taxa included those with binucleate and trinucleate pollen types. We detected differences among species. A significant reduction in pollen germination occurred in only five species. Pollen tubes of >50% of the species showed significant reduction in length. Trinucleate pollen types were more likely to exhibit tube length reduction than the binucleate types. Proportionately more monocotyledonous species were sensitive to UV-B treatment than dicotyledonous species, and proportionately more wild species were sensitive than cultivated species and pollen collected from plants growing in the field were somewhat more sensitive than pollen collected from plants grown in the greenhouse. Species in which pollination occurred earlier in the season were more likely to be susceptible to UV-B radiation than those for which anthesis took place later in the season, suggesting a possible adaptation to UV-B radiation.  相似文献

Many of the diverse animals that consume floral rewards act as efficient pollinators; however, others 'steal' rewards without 'paying' for them by pollinating. In contrast to the extensive studies of the ecological and evolutionary consequences of nectar theft, pollen theft and its implications remain largely neglected, even though it affects plant reproduction more directly. Here we review existing studies of pollen theft and find that: (1) most pollen thieves pollinate other plant species, suggesting that theft generally arises from a mismatch between the flower and thief that precludes pollen deposition, (2) bees are the most commonly documented pollen thieves, and (3) the floral traits that typically facilitate pollen theft involve either spatial or temporal separation of sex function within flowers (herkogamy and dichogamy, respectively). Given that herkogamy and dichogamy occur commonly and that bees are globally the most important floral visitors, pollen theft is likely a greatly under-appreciated component of floral ecology and influence on floral evolution. We identify the mechanisms by which pollen theft can affect plant fitness, and review the evidence for theft-induced ecological effects, including pollen limitation. We then explore the consequences of pollen theft for the evolution of floral traits and sexual systems, and conclude by identifying key directions for future research.  相似文献

Despite theoretical assumptions, estimates of dispersal distance in natural plant populations have only recently been undertaken. Since plants are non-motile, gene flow depends on the dispersal of pollen and seed. Methods of estimating pollen and seed dispersal are reviewed and their implications for studies of evolutionary processes discussed. Airborne pollen records are a suitable indicator for the study of climate change. The present work focuses on the role of annual pollen indices for the detection of bioclimatic trends through the analysis of the aerobiological spectra of 11 taxa of great biogeographical relevance in Catalonia over an 18-year period (1994–2011), by means of different parametric and non-parametric statistical methods. Among others, two non-parametric rank-based statistical tests were performed for detecting monotonic trends in time series data of the selected airborne pollen types and we have observed that they have similar power in detecting trends. Except for those cases in which the pollen data can be well-modeled by a normal distribution, it is better to apply non-parametric statistical methods to aerobiological studies. Our results provide a reliable representation of the pollen trends in the region and suggest that greater pollen quantities are being liberated to the atmosphere in the last years, specially by Mediterranean taxa such as Pinus, Total Quercus and Evergreen Quercus, although the trends may differ geographically. Longer aerobiological monitoring periods are required to corroborate these results and survey the increasing levels of certain pollen types that could exert an impact in terms of public health.  相似文献

The actin cytoskeleton is absolutely required for pollen germination and tube growth, but little is known about the regulation of actin polymer concentrations or dynamics in pollen. Here, we report that latrunculin B (LATB), a potent inhibitor of actin polymerization, had effects on pollen that were distinct from those of cytochalasin D. The equilibrium dissociation constant measured for LATB binding to maize pollen actin was determined to be 74 nM. This high affinity for pollen actin suggested that treatment of pollen with LATB would have marked effects on actin function. Indeed, LATB inhibited maize pollen germination half-maximally at 50 nM, yet it blocked pollen tube growth at one-tenth of that concentration. Low concentrations of LATB also caused partial disruption of the actin cytoskeleton in germinated maize pollen, as visualized by light microscopy and fluorescent-phalloidin staining. The amounts of filamentous actin (F-actin) in pollen were quantified by measuring phalloidin binding sites, a sensitive assay that had not been used previously for plant cells. The amount of F-actin in maize pollen increased slightly upon germination, whereas the total actin protein level did not change. LATB treatment caused a dose-dependent depolymerization of F-actin in populations of maize pollen grains and tubes. Moreover, the same concentrations of LATB caused similar depolymerization in pollen grains before germination and in pollen tubes. These data indicate that the increased sensitivity of pollen tube growth to LATB was not due to general destabilization of the actin cytoskeleton or to decreases in F-actin amounts after germination. We postulate that germination is less sensitive to LATB than tube extension because the presence of a small population of LATB-sensitive actin filaments is critical for maintenance of tip growth but not for germination of pollen, or because germination is less sensitive to partial depolymerization of the actin cytoskeleton.  相似文献

Evolutionary transitions from animal to wind pollination have occurred repeatedly during the history of the angiosperms, but the selective mechanisms remain elusive. Here, we propose that knowledge of pollen release biomechanics is critical for understanding the ecological and evolutionary processes underpinning this shift in pollination mode. Pollen release is the critical first stage of wind pollination (anemophily) and stamen properties are therefore likely to be under strong selection early in the transition. We describe current understanding of pollen release biomechanics to provide insights on the phenotypic and ecological drivers of wind pollination. Pollen release occurs when detachment forces dominate resistive forces retaining pollen within anthers. Detachment forces can be active or passive depending on whether they require energy input from the environment. Passive release is more widespread in anemophilous species and involves processes driven by steady or unsteady aerodynamic forces or turbulence-induced vibrations that shake pollen from anthers. We review empirical and theoretical studies suggesting that stamen vibration is likely to be a key mechanism of pollen release. The vibration response is governed by morphological and biomechanical properties of stamens, which may undergo divergent selection in the presence or absence of pollinators. Resistive forces have rarely been investigated for pollen within anthers, but are probably sensitive to environmental conditions and depend on flower age, varying systematically between animal- and wind-pollinated species. Animal and wind pollination are traditionally viewed as dichotomous alternatives because they are usually associated with strikingly different pollination syndromes. But this perspective has diverted attention from subtler, continuously varying traits which mediate the fluid dynamic process of pollen release. Reinterpreting the flower as a biomechanical entity that responds to fluctuating environmental forces may provide a promising way forward. We conclude by identifying several profitable areas for future research to obtain deeper insight into the evolution of wind pollination.  相似文献

Grain set failure in wheat, caused by boron (B) deficiency, is associated with poorly developed pollen and anthers. This paper presents results of a study of the effect of B on pollen viability when it was supplied "internally" through the roots and externally in an agar medium for in vitro germination.There was no major effect of B supply to wheat plants on the number of pollen anther-1 or the percentage of pollen with positive reaction to iodine. Pollen germination in the medium was, however, responsive to both internal and external B supply. When B was not added to the medium, germination was poor, regardless of the level of B supplied to the plant, in both a B deficiency sensitive (SW41) and a B deficiency tolerant (Sonora 64) genotypes. The percentage of germinated pollen and length of the pollen tube increased with increasing medium B. With 20–100 mg H3BO3 L-1 in the medium, the percentage of germinated pollen and length of the pollen tube responded positively to increasing B supply to the plant.No difference was found between sensitive and tolerant genotypes in the effect of B on their pollen viability. On the other hand, without added B in the nutrient solution applied to the plant, grain set was depressed in the B deficiency sensitive SW41 and not in the B deficiency tolerant Sonora 64. A difference in B supply to the germinating pollen in the stigma and style is one possible explanation for this variation in the response to B among wheat genotypes.  相似文献
