The influence of different sporulation temperatures (30, 37, 44 and 52°C) upon heat resistance of Bacillus subtilis was investigated.
Heat resistance was greater after higher sporulation temperatures. Relation of heat resistance and temperature of sporulation was not linear over all the range of temperatures tested. Heat resistance increased about tenfold in the range of 30–44°C. Sporulation at 52°C did not show any further increase in heat resistance.
This effect was constant over all the range of heating temperatures tested (100–120°C). z value remained constant ( z = 9°C).
Greater heat resistances at higher temperatures of sporulation were not due to selection of more heat resistant cells by a higher sporulation temperature. Spores obtained from cells incubated at 32 or 52°C always possessed heat resistances that corresponded to the sporulation temperature regardless of the incubation temperature of their vegetative cells.  相似文献

The effect of temperature on germination of a South African isolate of Peronospora parasitica , and infection of Brassica oleracea was studied. The optimum condition for germination was 20° C at 100% relative humidity. The percentage germination obtained was 80–98% and 70–80% between 15 and 25° C at 100% relative humidity, after a 12 and 6h incubation period, respectively. Optimum temperature for germ tube growth was also 20° C. The temperature range for maximum infection of seedlings of a highly susceptible cabbage cultivar and subsequent disease development in vitro was 15–25° C and 90–100% infection was achieved after 48 h of incubation. At<15°C and 26–30° C infection percentage was decreased to 40–50% and 35–40%, respectively. No disease incidence was recorded at temperatures above 35° C. A scanning electron microscope study of the infection process showed that penetration of cotyledons by germ tubes was mostly via stomata and occasionally directly through the cuticle. Results are discussed in relation to the need for future studies of P. parasitica in South Africa.  相似文献

Spores of Bacillus subtilis SA22 harvested after 22 d incubation on nutrient agar at 30°C were more resistant to 0–04% peracetic acid at 20°C than spores harvested following 2 d incubation. Similarly, spores of B. subtilis globigii B17, harvested after 7 d incubation on a sporulation agar were up to 10 times less resistant to 0.04% peracetic acid at 20°C than spores harvested after 35 d incubation. An increase in resistance to heating at 100°C and to exposure to 17.7% hydrogen peroxide at 20°C occurred as the age of B. subtilis SA22 spores prior to harvesting increased, whereas differences in resistance were not observed with spores of B. subtilis globigii B17.  相似文献

The effects of different soil water potentials, temperature and NaCl concentration on seed germination of Zygophyllum dumosum Boiss., a common shrub in Israeli deserts, was investigated. Seeds had to be exposed to constant field capacity conditions (–0.0316 MPa) for a minimal period of two days before germination could start. Maximal germination under such conditions occurred after four days or more. Under simulated conditions of gradual dehydration of the soil, seeds were inhibited either at low soil water potentials (–0.10 to –10.00 MPa) or at high ones (–0.002 to –0.0398 MPa). Germination of Z. dumosum was independent of temperature in the range of 10–25°C, but strongly inhibited at 30 and 35°C. At 20°C germination was inhibited by salinity of the medium but still occurred (0.5%) even at a concentration of 0.5 M NaCl.  相似文献

Growth of Listeria monocytogenes at refrigeration temperatures   总被引:1,自引:1,他引:0  
The growth of three strains of Listeria monocytogenes at refrigeration temperatures (-0.5 to 9.3°C) in chicken broth and/or UHT milk was determined using a rocking temperature gradient incubator. Minimum growth temperatures ranged from -0.1 to -0.4°C for the three strains. Lag times of 1–3 d and 3 to >34 d were observed with incubation at 5 and 0°C respectively. Corresponding generation times ranged from 13–24 h at 5°C and 62–131 h at 0°C. The type of culture medium had an influence on both the rate and extent of growth. Incubation of cultures at 4°C before inoculation caused a marked reduction in the lag time when compared with cultures which had been previously incubated at 30°C.  相似文献

Bacillus sp. A-001, which produced large amounts of amylase, was isolated from fermenting tef ( Eragrostis tef ) on tryptone soya agar supplemented with 1% starch. The organism grew between pH 4.5 and 10.5 with an optimum at 7–7.5. Growth occurred between 20 and 55°C but the optimum was about 35–40°C. At optimum medium pH (7.5) and a temperature of 35°C the organism entered the stationary phase after about 72 h and amylase production was at its highest (9.6 units ml-1) at this time. Enzyme activity was optimal at pH 5.5 and 40°C and showed good thermal stability; it required 110 min to lose 50% of its activity at 70°C. The enzyme hydrolysed native starch (flour from tef, corn and kocho) to various oligosaccharides, including maltotriose, maltose and glucose.  相似文献

Growth of a heat resistant, food poisoning strain of Clostridium welchii was followed in raw, minced breast and leg muscle of the chicken. Within the range 22–50° growth was slightly more rapid in the leg (pH 6·5–6·7) than in the breast (pH 5·6–5·7) and was fastest in leg muscle at 50°. No growth occurred at 15 or 52°.
In a comparison between chicken and turkey, inoculated breast and leg muscle were cooked for 1 h at 85° and held at 37°. Multiplication of surviving organisms was initiated much more rapidly in chicken than in turkey meat, though the growth rates were comparable in each case.
Sporulation of several strains of CI. welchii , including other heat resistant, food poisoning types, was generally 10–100 times greater in leg than in breast muscle of the chicken. Differences in sporulation could be attributed both to differences in pH and type of meat.  相似文献

The control of postharvest decay in table grapes using acetaldehyde vapours   总被引:3,自引:0,他引:3  
Grapes ( Vitis vinifera L. cv. 'Sultanina') harvested at the end of the 1985–1988 seasons, received postharvest application of acetaldehyde (AA) vapours for 24–40 h. Treatment with AA vapour at 20 °C or 0 °C reduced significantly the decay caused by several fungi: Botrytis cinerea, Rhizopus stolonifer, Aspergillus niger and Alternaria alternata . In grapes treated with 0.5% AA for 24 h, no R. stolonifer was found after 8 days of storage at 20 °C. Treatment with 0.25% AA vapour for 40 h of grapes cv. 'Perlette' inoculated with R. stolonifer reduced the decay by 89%.  相似文献

Abstract
No immature stages of Culex annulirostris were found during field sampling in 1979–1980 when the average water temperature was < 17 °C; they reappeared when the average water temperature was 19 °C and reached the peak density (mean 107 immatures/cylinder) at 26.5 °C.
The effect of 6 temperatures (15–40°C) on egg hatching, development and survival of the immature stages of Cx annulirostris in the laboratory showed that at 15 and 40°C, eggs failed to hatch and larvae died in the first instars. The optimum temperatures for egg hatching and the survival of immature stages were 25 and 30°C. At these temperatures, 85 and 82% respectively of egg rafts hatched, the mean number of larvae per raft was 258 ± 9.8 and 260 ± 11.4 with immature survival of 83.5 and 79.0% respectively. Mean time to hatch at 20–35°C ranged from 1.2 d (35°C) to 2.9 d (20 °C). Developmental times from first instar to adult ranged from 7.1 d (35 °C) to 25.2 d (20 °C). The threshold for development of the immatures was 15.6 ± 2.5°C and the thermal constant was 142.9 ± 26.5 day—degrees (incubation temperatures 20–35°C). At less suitable temperatures of 20 and 35 °C, hatching (57.5 and 45%), number larvae per raft (mean 139.8 ± 9.8 and 102.6 ± 14.2) and survival were low.  相似文献

Abstract Netropsin stimulated the rate of synthesis of an extracellular metalloproteinase in Bacillus megaterium incubated in a sporulation medium. The antibiotic delayed but did not suppress the decrease in the ability to synthesize the proteinase occurring at later sporulation stages. Netropsin also stimulated the synthesis of the proteinase when added to a growing culture; it inhibited the increase of protein turnover which was switched on between the 2nd and 3rd hour in the sporulating population. No refractile spores were developed during 6 h at 35°C in the antibiotic-treated culture. In the control 60% of sporulating cells were observed under similar conditions.  相似文献

Spores of Bacillus cereus ATCC 7004, 4342 and 9818 were obtained in nutrient agar at several pH from 5·9 to 8·3. The optimum pH for sporulation was around 7, but good production of spores was obtained in the range 6·5–8·3. With all three strains, D -values clearly dropped with sporulation pH, decreasing by about 65% per pH unit. z -Values were not significantly modified ( P > 0·05) by this factor. Mean z -values of 7·13 °C ± 0·16 for strain 7004, 7·67 °C ± 0·04 for 4342 and 8·80 °C ± 0·64 for 9818 were obtained.  相似文献

Abstract. Germination responses of redroot pigweed ( Amaranthus retroflexus L.) seeds to ethylene were determined at 25, 30, 35, or 40° C after preincubation at various temperatures (15–35° C) for different periods (0.5–32 d). After 7 d preincubation, seeds showed a log-linear germination response to ethylene concentration in most of the temperature treatments. Sensitivity to ethylene increased with longer preincubation; response thresholds of 0.03−0.09 cm3 m−3 were observed after 32 d, compared to 0.18−1.6 cm3 m−3 after 7 d of preincubation. Preincubation at 15 or 20° C generally enhanced germinability, whereas 25 or 30° C produced secondary dormancy, which was readily broken with ethylene. Temperature during preincubation also significantly influenced the slope of the dose-response curve. The responses of preincubated redroot pigweed seeds to ethylene suggested that, in the field, seeds would probably not lose their sensitivity to this gas during prolonged burial in soil.  相似文献

Abstract Ten different isolates of Thiobacillus ferrooxidans were characterized with respect to temperature in the range 2–35°C. Four of the ten strains oxidized ferrous iron exponentially over the entire range of incubation temperatures, including the lowest temperature tested (2°C), and were therefore characterized as psychrotrophic. Jarosite production was substantially reduced at temperatures less than 10°C and was not observed at 2°C. Energy of activation values were in the range 75.2–96.6 kJ/mol°C and indicated that iron oxidation at low temperatures was governed by both a chemical and a physical control.  相似文献

Abstract.  Although laboratory studies demonstrate that cooling to ecologically relevant temperatures and/or at ecologically relevant rates induces rapid cold-hardening (RCH) in a variety of insects, little is known of the induction of RCH in nature. In the present study, caged Drosophila melanogaster (1–2 days posteclosion) from a colony established with flies collected locally are placed in a field setting (i.e. in approximately 4-cm deep leaf litter beneath an apple tree in Mount Pleasant, Michigan) during late afternoon (18.00 h EST; 05.00 h GMT). As the cage temperature falls from 22.1 ± 0.8 to 10.1 ± 0.1 °C between 18.00 and 06.00 h, the proportion of flies surviving a transfer to –6 °C for 1 h increases from 10.0 ± 6.2% to 68.1 ± 7.2%. When obtained from field cages, and then cooled from 23 °C at approximately 0.33 °C min−1, more female flies remain behaviourally responsive (clinging to surfaces, exhibiting an active righting response, and/or climbing) at temperatures of 8–12 °C (24.00 h samples) or 7–12 °C (06.00 h samples), than do those sampled from cages kept in an incubator (23 °C). Field cooling reduces chill coma temperature from 8.7 ± 0.2 °C at 06.00 h to 7.1 ± 0.2 °C at 24.00 h, and to 6.6 ± 0.2 °C at 06.00 h. These data demonstrate that, in a recently collected culture of D. melanogaster , natural changes in microenvironmental temperature induce RCH that can benefit the organism at temperatures encountered in nature.  相似文献

About 34% of the strains of Yersinia enterocolitica isolated from raw milk were found to produce lecithinase. A selected strain produced phospholipase C at 22°C and 37°C; production was optimum at 37°C in the stationary phase (14–16 h). A decrease in phospholipase C activity at various storage temperatures (—5°C, 4°C, 37°C) was also observed, although the enzyme was active over a wide range of temperature (5–65°C) and pH (3mD5–7mD5). The phospholipase C was partially purified by ammonium sulphate precipitation and Sephadex column chromatography, and characterized.  相似文献

Abstract. Alternating temperatures stimulate the germination of Rumex crispus L. and Rumex obtusifolius L. The optimum period spent at the lower temperature in a diurnal cycle is greater than that spent at the higher temperature. Under most conditions the optimum period at the upper temperatures is about 8 h but, as the upper temperature of a cycle is increased, the optimum period at the upper temperature becomes shorter and more critical. Thus when it is 35°C the optimum period is 2.5–4 h in the light, or about 1 h in the dark. The effect of alternating temperatures is much less in the dark than in the light and in general only extreme alternations with short periods at the higher temperature are effective in the dark. In the light any temperature alternation within the range 1–35°C is effective to at least some extent, providing the temperature difference is 5°C or more and providing the alternation includes one temperature which is above approximately 15°C and one which is below approximately 25°C. The optimum temperature difference is about 15°C. In the light, 4 to 10 cycles saturate the response, but in the dark, where the effect is much less, the response may not be saturated even by 16 cycles. KNO3 at 10−3 M has little effect on the response to alternating temperatures either in the light or the dark. The response to alternating temperature regimes does not appear to vary in quality, i.e., in terms of which particular treatments are best, but it varies in magnitude with site and year of seed collection; and it increases slowly during dry storage, even when stored at a temperature as low as 1.5°C.  相似文献

SUMMARY. Eggs collected from Hexagenia bilineata females were successfully reared in the laboratory at temperatures of 15, 20, 25 and 30°C. Eggs did not hatch at 10°C and although hatching was successful at 35°C, all nymphs at this temperature died while in early instars.
Survival of nymphs between the approximate size interval of 4–14 mm showed a significant decrease with increased temperatures. Nymphs at 15°C, however, generally did not survive transformation to the subaduit stage.
The growth pattern of individual nymphs was well described by a logistic curve at most temperatures. Furthermore, growth pattern was significantly affected by both temperature and sex.
Rate of development from oviposition to first emergence increased with increasing temperatures in a linear fashion between 15 and 30°C. The relationship was equally well described by a hyperbolic equation and a power-law equation. By extrapolation from the hyperbolic equation, the lower threshold temperature for development was estimated to be 10.1°C3.1°C. The degree (°C)-days required for development from oviposition to first emergence was calculated to be 2337 days with 95% confidence limits of 2045–2727 days under laboratory conditions.  相似文献

The development of muscle innervation pattern was investigated in larvae of the Amazonian fish, the tambaqui Colossoma macropomum. The time to hatching decreased from 28–29 h at 23.5° C to 11–12 h at 31° C. The larvae hatched after the completion of somitogenesis (38-somite stage) at 23.5° C but only at the 33-somite stage at 28–31° C. Embryos were stained for acetylcholinesterase activity and with an acetylated tubulin antibody in order to visualize neural processes. All muscle fibre types were initially innervated at their myoseptal ends. The development of motor innervation to the trunk muscle was delayed with respect to hatching at higher temperatures. At hatching, muscle fibres were innervated only to somites 16–17 at 28–31° C and somite 23–26 at 23.5–25° C (counting from the head), although the larvae swam vigorously to avoid sinking. In contrast, in newly hatched larvae myofibrils were present right along the trunk at all temperatures in both the superficial and inner muscle fibres. At hatching numerous multi-layered membrane contacts with the ultrastructural characteristics of gap junctions, were found between muscle fibres and at the inter-somite junctions, suggesting the somites were initially electrically coupled. These structures disappeared concomitant with the development of muscle endplates right down the trunk. The larvae started feeding 5 days post-hatch at 28° C. First feeding was associated with a dramatic decrease in the volume density of mitochondria and an increase in the volume density of myofibrils in the inner muscle fibres. The polyneuronal and multi-terminal pattern of innervation characteristic of adult slow-muscle fibres also developed around the time of first feeding.  相似文献

Bacillus sphaericus 9602 was grown in batch culture at various temperatures. At 10°C and 12°C the maximum sporulation yield was <10%, while at 15°C, 20°C and 30°C, a sporulation yield of >95% was achieved. However at 40°C B. sphaericus grew only vegetatively. The heat resistances (D values at 90°C) of spores grown at 15°C and 20°C were significantly higher than those grown at 30°C.  相似文献

The effects of temperature on the length of the incubation and latent periods of clematis powdery mildew, caused by Erysiphe polygoni , were studied. At constant temperatures over the range of 10–29°C, the length of the incubation period ranged from 4 to 11 days, and the length of the latent period ranged from 4 to 13 days; no visible colonies developed at 30°C after 19 days. The relationship between temperature and the rates of fungal development within the incubation and latent periods (expressed as the reciprocal of the lengths of the incubation and latent periods) under constant temperature were described well by a non-linear model. The resulting curves were not the usual form of an asymmetrically bell-shaped type; instead, they were of exponential type with the development rate increasing with increasing temperatures. The relationship between temperature and the development rate during the post-incubation (before sporulation) period was non-linear with an optimum temperature of approximately 20°C.  相似文献
