Abstract

Flowering phenology, sexual polymorphism and spatial segregation were studied over two consecutive years in a population of Thymelaea hirsuta (L.) Endl. (Thymelaeaceae) growing on coastal cliffs in north-western Italy. The study population was geographically segregated and, together with Croatian sites, it constituted the northernmost margin of the species' distribution range. Very frequent observations highlighted a sexual tendency towards dioecy already shown by T. hirsuta in other parts of the Mediterranean coast. The flowering period lasted 10?–?11 months, more or less. A high proportion of female flowers and a low number of hermaphrodite flowers were recorded, in contrast with the abundance borne by the species elsewhere; gender segregation into two different subgroups was noted, one consisting mainly of male and protogynous plants, the second consisting basically of female and protandrous specimens.  相似文献

Fibroblast growth factor 2 ( FGF2 ) plays an important role in fertility and early embryo development. The objectives of this study were to test the association of FGF2 polymorphisms with fertilization success in cattle using an in vitro fertilization experimental system and to investigate the mechanisms leading to the presence of rare alleles of FGF2 in the Holstein population. A total of 7502 fertilizations were performed and a total of 5049 embryos were produced to collect fertilization and embryo survival records. A total of 444 ovaries, from which oocytes were aspirated and fertilized, were genotyped for two single nucleotide polymorphisms (SNPs) previously identified in FGF2 (g.23G>T and g.11646A>G). Frequency of the TT genotype of the g.23G>T SNP was low in the ovary population (5.4%) and in a different Holstein cattle population (6.6%) examined in this study. Single SNP analysis showed that both SNPs were associated with early embryonic survival rate. Two-way interaction analysis revealed significant association of epistatic interaction between the SNPs with fertilization rate. To test whether or not low frequency of allele T for the g.23G>T SNP in the population is a result of a fertilization failure of T oocytes, semen from six GG bulls was used to fertilize a total of 458 oocytes obtained from 19 GT ovaries. A significant segregation distortion was observed for 169 embryos genotyped for the g.23G>T SNP. We conclude that oocytes carrying the T allele show a reduced fertilization rate and that segregation distortion leads to rarity of the TT genotype in the population.  相似文献

Rutilus alburnoides complex is a common and widely distributed Iberian cyprinid, whose natural populations include mainly diploid and triploid forms. The Guadiana populations of R. alburnoides were studied to determine whether habitat segregation and morphological differences exist between these forms. The ploidy level of each specimen was determined by measuring erythrocyte DNA content using flow cytometry. Evidence of spatial segregation between diploid males and the two female forms was found. Diploid males were best represented in the River Degebe, which was shallow, with higher temperatures (especially during the spring and summer), and silt and sandy substrate. Diploid females were found in deeper water, on steeper gradients and coarse substrata, while triploid females preferred higher current velocity and a high proportion of instream cover, especially during the spring. The ecological differences may reduce competitive interactions, and should promote a stable coexistence of the different forms. Morphological distinction between fish of different ploidy levels was not established, but differences were found between the males and females. Discriminant analysis allowed, with a 10% error, the separation of both sexes through six morphological characteristics that could be recorded in the field.  相似文献

While the essential role of episomal par loci in plasmid DNA partitioning has long been appreciated, the function of chromosomally encoded par loci is less clear. The chromosomal parA-parB genes are conserved throughout the bacterial kingdom and encode proteins homologous to those of the plasmidic Type I active partitioning systems. The third conserved element, the centromere-like sequence called parS, occurs in several copies in the chromosome. Recent studies show that the ParA-ParB-parS system is a key player of a mitosis-like process ensuring proper intracellular localization of certain chromosomal regions such as oriC domain and their active and directed segregation. Moreover, the chromosomal par systems link chromosome segregation with initiation of DNA replication and the cell cycle.  相似文献

Non-essential extra-chromosomal DNA elements such as plasmids are responsible for their own propagation in dividing host cells, and one means to ensure this is to carry a miniature active segregation system reminiscent of the mitotic spindle. Plasmids that are maintained at low numbers in prokaryotic cells have developed a range of such active partitioning systems, which are characterized by an impressive simplicity and efficiency and which are united by the use of dynamic, nucleotide-driven filaments to separate and position DNA molecules. A comparison of different plasmid segregation systems reveals (i) how unrelated filament-forming and DNA-binding proteins have been adopted and modified to create a range of simple DNA segregating complexes and (ii) how subtle changes in the few components of these DNA segregation machines has led to a remarkable diversity in the molecular mechanisms of closely related segregation systems. Here, our current understanding of plasmid segregation systems is reviewed and compared with other DNA segregation systems, and this is extended by a discussion of basic principles of plasmid segregation systems, evolutionary implications and the relationship between an autonomous DNA element and its host cell.  相似文献

The influence of host and parasite genetic background on infection outcome is a topic of great interest because of its pertinence to theoretical issues in evolutionary biology. In the present study, we use a classical genetics approach to examine the mode of inheritance of infection outcome in the crustacean Daphnia magna when exposed to the bacterial parasite Pasteuria ramosa. In contrast to previous studies in this system, we use a clone of P. ramosa, not field isolates, which allows for a more definitive interpretation of results. We test parental, F1, F2, backcross and selfed parental clones (total 284 genotypes) for susceptibility against a clone of P. ramosa using two different methods, infection trials and the recently developed attachment test. We find that D. magna clones reliably exhibit either complete resistance or complete susceptibility to P. ramosa clone C1 and that resistance is dominant, and inherited in a pattern consistent with Mendelian segregation of a single-locus with two alleles. The finding of a single host locus controlling susceptibility to P. ramosa suggests that the previously observed genotype-genotype interactions in this system have a simple genetic basis. This has important implications for the outcome of host-parasite co-evolution. Our results add to the growing body of evidence that resistance to parasites in invertebrates is mostly coded by one or few loci with dominance.  相似文献

The influence of moisture on the survival, movement anddegradation activity of a 2,4-D degrading bacterium,Burkholderia cepacia strain BRI6001L, geneticallyengineered to contain bioluminescent and lactoseutilization genes, was studied in unsaturated soil columns.The distance traveled by BRI6001L was dependent on theclay content of the soil, higher clay contents beingresponsible for higher filtration coefficients. Long termsurvival, in excess of one year, was attributed to strainBRI6001L's ability to survive dry conditions. Changes inthe 2,4-D biodegradation rate showed a better correlationwith the BRI6001L population density than with the totalviable bacterial population. At moisture levels betweenfield capacity and 40% moisture (– 33 kPa to –100 kPa)2,4-D degradation was attributed mainly to BRI6001L. Atmoisture levels between 6 and 15%, 2,4-D disappearancewas attributed to the indigenous microbial population,with no degradation occurring at moisture levels below6%. Returning the moisture to above 40% led to anincrease of 4 orders of magnitude in the BRI6001Lpopulation density and to a 10-fold increase in the 2,4-Ddegradation rate. The ability to monitor a specificmicrobial population using reporter genes hasdemonstrated the importance of controlling moisturelevels for maximizing biodegradation rates in unsaturatedsoil environments.  相似文献

Climate alteration is one of the most cited ecological consequences of urbanization. However, the magnitude of this impact is likely to vary with spatial scale. We investigated how this alteration affects the biological fitness of insects, which are especially sensitive to ambient conditions and well‐suited organisms to study urbanization‐related changes in phenotypic traits. We monitored temperature and relative air humidity in wooded sites characterized by different levels of urbanization in the surroundings. Using a split‐brood design experiment, we investigated the effect of urbanization at the local (i.e., 200 × 200 m) and landscape (i.e., 3 × 3 km) scale on two key traits of biological fitness in two closely related butterfly species that differ in thermal sensitivity. In line with the Urban Heat Island concept, urbanization led to a 1°C increase in daytime temperature and an 8% decrease in daytime relative humidity at the local scale. The thermophilous species Lasiommata megera responded at the local scale: larval survival increased twofold in urban compared to rural sites. Urbanized sites tended to produce bigger adults, although this was the case for males only. In the woodland species Pararge aegeria, which has recently expanded its ecological niche, we did not observe such a response, neither at the local, nor at the landscape scale. These results demonstrate interspecific differences in urbanization‐related phenotypic plasticity and larval survival. We discuss larval pre‐adaptations in species of different ecological profiles to urban conditions. Our results also highlight the significance of considering fine‐grained spatial scales in urban ecology.  相似文献

Summary Infection of Mu-sensitive bacteria with a recombinant phage that carries the EcoRI·C fragment from the immunity end of wild type Mu DNA causes filamentous growth. Transmission electron microscopy revealed that the cell-division cycle was inhibited at, or prior to, the initiation of septation. The filamentation does not occur after infection of Mu-immune bacteria or after infection with a phage carrying the same EcoRI·C fragment, but with an IS1 insertion in gene B of Mu, showing that either gpB and/or some non-essential functions (e.g. kil) mapping downstream from the insertion are required for the inhibition of cell division. These data and previously published evidence suggest that in the killing of E. coli K12 by early Mu functions expressed from the cloned EcoRI·C fragment, two components have to be distinguished: one, a highly efficient elimination of plasmid DNA carrying the early Mu genes, and second, a series of interactions with host functions conducent to an inhibition of cell division. It is suggested that functions normally involved in the SOS reaction participtate in the inhibition of cell division by early Mu functions. Infected bacteria synthesize the replication protein B (MR 33000) of Mu, which was found by cell fractionation experiments to be associated with the inner cell membrane. The role of this association for filamentous growth and for the integrative replication of the phage is discussed. The recombinant phage might be useful as a tool for the study of the E. coli cell division cycle.  相似文献

土壤微生物群落与植物的生长发育密切相关, 入侵植物可以改变入侵地土壤微生物类群, 使土壤理化性质发生变化, 从而促进其入侵过程。该文通过比较高丹草(Sorghum bicolor × S. sudanense)、向日葵(Helianthus annuus)、紫花苜蓿(Medi- cago sativa)和多年生黑麦草(Lolium perenne)4种替代植物与黄顶菊(Flaveria bidentis)混合种植(以下简称混种)后不同时期的土壤细菌多样性的变化, 揭示土壤细菌群落对黄顶菊入侵及替代管理措施的响应规律。结果表明, 单独种植(以下简称单种)黄顶菊的土壤细菌多样性下降, 并且在整个生长期多样性指数多数情况下低于高丹草、向日葵、紫花苜蓿和多年生黑麦草单种或与黄顶菊混种的土壤。当4种替代植物与黄顶菊混种后, 土壤细菌16S rRNA的变性梯度凝胶电泳(DGGE)图谱与它们分别种植时存在明显差异, 且不同生长期各个混种土壤都有特征细菌群落。4种替代植物单种或与黄顶菊混种的土壤细菌Shannon多样性指数变化规律与植物生长发育趋势相同, 7月份达到高峰, 8月份开始降低。总之, 黄顶菊入侵降低了土壤细菌群落多样性, 4种替代植物与黄顶菊混种后, 又可提高土壤细菌群落多样性, 这种变化对黄顶菊成功入侵和替代防控具有重要作用。  相似文献

Marine megafauna, including seabirds, are critically affected by fisheries bycatch. However, bycatch risk may differ on temporal and spatial scales due to the uneven distribution and effort of fleets operating different fishing gear, and to focal species distribution and foraging behavior. Scopoli's shearwater Calonectris diomedea is a long‐lived seabird that experiences high bycatch rates in longline fisheries and strong population‐level impacts due to this type of anthropogenic mortality. Analyzing a long‐term dataset on individual monitoring, we compared adult survival (by means of multi‐event capture–recapture models) among three close predator‐free Mediterranean colonies of the species. Unexpectedly for a long‐lived organism, adult survival varied among colonies. We explored potential causes of this differential survival by (1) measuring egg volume as a proxy of food availability and parental condition; (2) building a specific longline bycatch risk map for the species; and (3) assessing the distribution patterns of breeding birds from the three study colonies via GPS tracking. Egg volume was very similar between colonies over time, suggesting that environmental variability related to habitat foraging suitability was not the main cause of differential survival. On the other hand, differences in foraging movements among individuals from the three colonies expose them to differential mortality risk, which likely influenced the observed differences in adult survival. The overlap of information obtained by the generation of specific bycatch risk maps, the quantification of population demographic parameters, and the foraging spatial analysis should inform managers about differential sensitivity to the anthropogenic impact at mesoscale level and guide decisions depending on the spatial configuration of local populations. The approach would apply and should be considered in any species where foraging distribution is colony‐specific and mortality risk varies spatially.  相似文献

Coordination of chromosome segregation and cytokinesis is crucial for efficient cell proliferation. In Bacillus subtilis, the nucleoid occlusion protein Noc protects the chromosomes by associating with the chromosome and preventing cell division in its vicinity. Using protein localization, ChAP‐on‐Chip and bioinformatics, we have identified a consensus Noc‐binding DNA sequence (NBS), and have shown that Noc is targeted to about 70 discrete regions scattered around the chromosome, though absent from a large region around the replication terminus. Purified Noc bound specifically to an NBS in vitro. NBSs inserted near the replication terminus bound Noc–YFP and caused a delay in cell division. An autonomous plasmid carrying an NBS array recruited Noc–YFP and conferred a severe Noc‐dependent inhibition of cell division. This shows that Noc is a potent inhibitor of division, but that its activity is strictly localized by the interaction with NBS sites in vivo. We propose that Noc serves not only as a spatial regulator of cell division to protect the nucleoid, but also as a timing device with an important role in the coordination of chromosome segregation and cell division.  相似文献

用含有GFP基因标记的内生细菌菌株BS-2-gfp和TB2-gfp,采用喷雾接种的方法,研究其在荔枝叶片、花、幼果及成熟果实果皮内的定殖动态及其与防病保鲜之间的关系.结果表明:内生细菌BS-2-gfp和TB2-gfp能在荔枝叶片、花、幼果及采后果皮上定殖,能在各组织内繁殖并可在花和幼果间传导.内生细菌在荔枝叶片上的定殖因季节和荔枝生长期的不同而异,与秋季相比,春季定殖期限长,定殖量大.内生细菌在荔枝不同部位的定殖时间和定殖量也不同,在叶片上接种37d后还可分离回收到接种的2种目标细菌,在花上接种10d后就回收不到BS-2-gfp,而成熟荔枝果皮上2种菌的定殖量最大.防病试验表明,当荔枝霜疫病的病情指数急剧上升时,TB2-gfp在荔枝果皮的定殖量达到最大,为1.90×106CFU.g-1FM;保鲜试验发现,TB2-gfp菌株的保鲜效果优于BS-2-gfp,菌株在荔枝果皮内的定殖量也高于菌株BS-2-gfp,表明供试内生细菌在荔枝果皮的定殖量与其防病及保鲜效果呈正相关.  相似文献

以浙江省天目山国家级自然保护区为例,采用多尺度分析思想,利用多重分形分析方法,研究了不同尺度下物种多样性的变化、空间分布格局以及多样性与空间格局之间的关系。研究主要得到3方面的结论:(1)物种多样性具有尺度依赖性,随着空间尺度的增大,Shannon-Wiener多样性指数Ⅳ增大,Margalef多样性指数足和均匀度指数E减小;(2)多重分形参数αmin。多重分形谱的变化范围SR等能够定量反映物种的空间分布特征,空间大尺度越大,物种越聚集,空间分布越不均匀;(3)物种多样性与空间格局存在线性或幂函数关系。研究表明多重分形分析定量描述物种空间格局是有效性的,多重分形参数与生物多样性之间的定量关系为研究物种空间格局、生态属性与尺度之间的关系奠定了基础。因此,分形结合传统方法,在生物多样性方面的研究将有很大的潜在价值。  相似文献

The efficacy of Pseudomonas fluorescens EPS62e in the biocontrol of Erwinia amylovora, the causal agent of fire blight of apple and pear, depends on the colonization of plant surfaces after spray application. A procedure to increase cell survival in the phyllosphere was developed consisting of saline stress and osmolyte amendment to the growth medium during inoculum preparation. Hyperosmotic stress induced the synthesis of the osmolytes trehalose, N-acetylglutaminylglutamine amide and glucosyl-glycerol, but decreasing growth rate. Amendment of the growth medium with glycine betaine increased growth rate and cell yield and promoted its intracellular accumulation. Under controlled environment conditions, osmoadaptation increased by 10- to 100-fold cell survival to desiccation and to low relative humidity conditions on plant surfaces, in comparison with the nonosmoadapted controls. In the field, cell survival increased 100-1000 times in immature fruit upon osmoadaptation but was not significantly affected in flowers where active colonization occurs. The efficacy in the control of fire blight infections was increased 30-50% upon osmoadaptation on immature fruits but was not affected in blossoms. The method of osmoadaptation may be useful for improving the fitness and efficacy of biological control agents of phyllosphere pathogens under limiting humidity conditions.  相似文献
