Resting membrane potential (RMP) and whole cell currents wererecorded in human THP-1 monocytes adherent to polystyrene, unstimulated human umbilical vein endothelial cells (HUVECs),lipopolysaccharide (LPS)-treated HUVECs, immobilizedE-selectin, or vascular cell adhesion molecule 1 (VCAM-1)using the patch-clamp technique. RMP after 5 h on polystyrene was24.3 ± 1.7 mV (n = 42) with delayed rectifier K+(Idr) andCl currents(ICl) presentin >75% of the cells. Inwardly rectifying K+ currents(Iir) werepresent in only 14% of THP-1 cells. Adherence to unstimulated HUVECsor E-selectin for 5 h had no effect on Iir orICl but decreasedIdr. Five hoursafter adherence to LPS-treated HUVECs, outward currents were unchanged,but Iir waspresent in 81% of THP-1 cells. A twofold increase inIir and ahyperpolarization (41.3 ± 3.7 mV,n = 16) were abolished by pretreatmentof THP-1 cells with cycloheximide, a protein synthesis inhibitor, orherbimycin A, a tyrosine kinase inhibitor, or by pretreatment of theLPS-treated HUVECs with anti-VCAM-1. Only a brief (15-min) interactionbetween THP-1 cells and LPS-treated HUVECs was required toinduce Iir expression 5 h later. THP-1 cells adherent to VCAM-1 exhibited similarconductances to cells adherent to LPS-treated HUVECs. Thus engagementof specific integrins results in selective modulation of differentK+ conductances.

We investigatedthe role of intracellular calcium concentration([Ca2+]i) in endothelin-1 (ET-1) production,the effects of potential vasospastic agents on[Ca2+]i, and the presence of L-typevoltage-dependent Ca2+ channels in cerebral microvascularendothelial cells. Primary cultures of endothelial cells isolated frompiglet cerebral microvessels were used. Confluent cells were exposed toeither the thromboxane receptor agonist U-46619 (1 µM),5-hydroxytryptamine (5-HT; 0.1 mM), or lysophosphatidic acid (LPA; 1 µM) alone or after pretreatment with the Ca2+-chelatingagent EDTA (100 mM), the L-type Ca2+ channel blockerverapamil (10 µM), or the antagonist of receptor-operated Ca2+ channel SKF-96365 HCl (10 µM) for 15 min. ET-1production increased from 1.2 (control) to 8.2 (U-46619), 4.9 (5-HT),or 3.9 (LPA) fmol/µg protein, respectively. Such elevated ET-1biosynthesis was attenuated by verapamil, EDTA, or SKF-96365 HCl. Toinvestigate the presence of L-type voltage-dependent Ca2+channels in endothelial cells, the [Ca2+]isignal was determined fluorometrically by using fura 2-AM. Superfusionof confluent endothelial cells with U-46619, 5-HT, or LPA significantlyincreased [Ca2+]i. Pretreatment ofendothelial cells with high K+ (60 mM) or nifedipine (4 µM) diminished increases in [Ca2+]i inducedby the vasoactive agents. These results indicate that 1)elevated [Ca2+]i signals are involved in ET-1biosynthesis induced by specific spasmogenic agents, 2) theincreases in [Ca2+]i induced by thevasoactive agents tested involve receptor as well as L-typevoltage-dependent Ca2+ channels, and 3) primarycultures of cerebral microvascular endothelial cells express L-typevoltage-dependent Ca2+ channels.

Hypoxia of endothelial cells leads to MMP-2-dependent survival and death   总被引:2,自引:0,他引:2  
Exposure of endothelial cells (ECs) to hypoxia has separately been shown to induce their angiogenesis or death. Matrix metalloproteinase (MMP)-2 is associated with EC angiogenesis, although recent studies also implicate this molecule in EC death. We studied the effect of hypoxia in the absence or presence of TNF- (characteristic of the inflammatory microenvironment accompanying hypoxia) on MMP-2 expression and its role in angiogenesis (proliferation, migration, and tube formation) and in the death of primary human umbilical vein endothelial cells (HUVECs). Hypoxia alone (24–48 h in 0.3% O2 in the hypoxic chamber) and furthermore, when combined with TNF-, significantly enhanced MMP-2 expression and activity. Hypoxia also led to a reduction in membrane type 1 MMP (MT1-MMP) and tissue inhibitor of metalloproteinase-2 mRNA and protein while enhancing the expression of v3 integrin and the cytoskeletal protein phosphopaxillin. Moreover, hypoxia led to colocalization of v3 and MMP-2, but not MT1-MMP, with phosphopaxillin in ECs. These results suggest MT1-MMP-independent activation of MMP-2 during hypoxia and support interactions between the ECM, integrins, and the cytoskeleton in hypoxia-induced MMP-2-related functions. Hypoxia enhanced EC migration in an MMP-2-dependent manner while leading to a reduction of cell number via their apoptosis, which was also dependent on MMP-2. In addition, hypoxia caused an aberrant tubelike formation on Matrigel that appeared to be unaffected by MMP-2. The hypoxia-induced, MMP-2-dependent migration of ECs is in accordance with the proangiogenic role ascribed to MMP-2, while the involvement of this protease in the hypoxia-related death of ECs supports an additional apoptotic role for this protease. The effect ofinositol 1,4,5-trisphosphate(IP3) receptor blockade onplatelet-derived growth factor (PDGF), fibroblast growth factor (FGF),endothelin-1 (ET-1), or -thrombin receptor-mediated intracellularCa2+(Ca2+i) release was examined using fura 2 microspectrofluorometry in single Chinese hamster ovary cells andmyoblasts. Blockade of the IP3receptor was achieved by microinjection of heparin or monoclonalantibody (MAb) 18A10 into the IP3type 1 receptor. Heparin completely inhibitedCa2+i release after flash photolysis withcaged IP3 and after exposure toPDGF and FGF. In contrast, heparin failed to blockCa2+i release after -thrombin andET-1. After application of ligand, IP3 levels were five- to sevenfoldhigher for -thrombin than for ET-1 or PDGF.IP3 levels after PDGF and ET-1were comparable. Similar to heparin, MAb 18A10 blockedCa2+i release after PDGF but failed toblock Ca2+i release after ET-1 or-thrombin. These data suggest that the mechanisms of Ca2+i release by tyrosine kinase andcertain 7-transmembrane receptors may differ. Although both receptortypes use the IP3-signaling system, the ET-1 and -thrombin receptors may have a second,alternative mechanism for activatingCa2+i release.

System-specific O2 sensitivity of the tandem pore domain K+ channel TASK-1   总被引:1,自引:0,他引:1  
Hypoxic inhibition of TASK-1, a tandem pore domain background K+ channel, provides a critical link between reduced O2 levels and physiological responses in various cell types. Here, we examined the expression and O2 sensitivity of TASK-1 in immortalized adrenomedullary chromaffin (MAH) cells. In physiological (asymmetrical) K+ solutions, 3 µM anandamide or 300 µM Zn2+ inhibited a strongly pH-sensitive current. Under symmetrical K+ conditions, the anandamide- and Zn2+-sensitive K+ currents were voltage independent. These data demonstrate the functional expression of TASK-1, and cellular expression of this channel was confirmed by RT-PCR and Western blotting. At concentrations that selectively inhibit TASK-1, anandamide and Zn2+ were without effect on the magnitude of the O2-sensitive current or the hypoxic depolarization. Thus TASK-1 does not contribute to O2 sensing in MAH cells, demonstrating the failure of a known O2-sensitive K+ channel to respond to hypoxia in an O2-sensing cell. These data demonstrate that, ultimately, the sensitivity of a particular K+ channel to hypoxia is determined by the cell, and we propose that this is achieved by coupling distinct hypoxia signaling systems to individual channels. Hyperbaric oxygen (HBO) is beingstudied as a therapeutic intervention for ischemia/reperfusion(I/R) injury. We have developed an in vitro endothelial cell model ofI/R injury to study the impact of HBO on the expression ofintercellular adhesion molecule-1 (ICAM-1) and polymorphonuclearleukocyte (PMN) adhesion. Human umbilical vein endothelial cell (HUVEC)and bovine aortic endothelial cell (BAEC) induction of ICAM-1 requiredsimultaneous exposure to both hypoxia and hypoglycemia as determined byconfocal laser scanning microscopy, ELISA, and Western blot. HBOtreatment reduced the expression of ICAM-1 to control levels. Adhesionof PMNs to BAECs was increased following hypoxia/hypoglycemia exposure(3.4-fold, P < 0.01) and was reduced to control levels withexposure to HBO (P = 0.67). Exposure of HUVECs and BAECs to HBOinduced the synthesis of endothelial cell nitric oxide synthase (eNOS).The NOS inhibitor nitro-L-arginine methyl ester attenuatedHBO-mediated inhibition of ICAM-1 expression. Our findings suggest thatthe beneficial effects of HBO in treating I/R injury may be mediated inpart by inhibition of ICAM-1 expression through the induction of eNOS.

This study was undertaken toinvestigate the molecular constituents mediating LS174T colonadenocarcinoma cell adhesion to 4-h TNF--stimulated human umbilicalvein endothelial cells (HUVECs) under flow. At 1 dyn/cm2,~57% of cells rolled and then became firmly adherent, whereas otherscontinuously rolled on endothelium. Initial cell binding was primarilymediated by endothelial E-selectin. By using neuraminidase, glycolipidbiosynthesis inhibitord,l-threo-1-phenyl-2-hexadecanoylamino-3-pyrrolidino-1-propanol · HCl,trypsin, and flow cytometry, LS174T cells were shown to express sialylLewisx (sLex)- anddi-sLex-decorated, but not sLea-decorated,glycolipid and glycoprotein ligands for E-selectin. The cellspreferentially employed sialylated glycoproteins over glycolipids inadhesion as measured by conversion of rolling to firm adhesion,resistance to detachment by increased shear stress, and rollingvelocity. However, a nonsialylated E-selectin counterreceptor alsoexists. Furthermore, LS174T 2, 6, and1 integrins support a minor pathway in adhesion toHUVECs. Finally, tumor cell attachment specifically increases HUVECendocytosis of E-selectin. Altogether, the data indicate the complexityof carcinoma cell-endothelium adhesion via sialylated glycoconjugates,integrins, and their respective counterreceptors.

Muchevidence supports the view that hypoxic/ischemic injury is largely dueto increased intracellular Ca concentration([Ca]i) resulting from 1) decreasedintracellular pH (pHi), 2) stimulated Na/H exchangethat increases Na uptake and thus intracellular Na (Nai),and 3) decreased Na gradient that decreases or reverses net Catransport via Na/Ca exchange. The Na/H exchanger (NHE) is alsostimulated by hypertonic solutions; however, hypertonic media mayinhibit NHE's response to changes in pHi (Cala PM and Maldonado HM. J Gen Physiol 103: 1035-1054, 1994). Thus wetested the hypothesis that hypertonic perfusion attenuates acid-induced increases in Nai in myocardium and, thereby, decreasesCai accumulation during hypoxia. Rabbit hearts wereLangendorff perfused with HEPES-buffered Krebs-Henseleit solutionequilibrated with 100% O2 or 100% N2. Hypertonic perfusion began 5 min before hypoxia or normoxicacidification (NH4Cl washout). Nai,[Ca]i, pHi, and high-energyphosphates were measured by NMR. Control solutions were 295 mosM, andhypertonic solutions were adjusted to 305, 325, or 345 mosM by additionof NaCl or sucrose. During 60 min of hypoxia (295 mosM),Nai rose from 22 ± 1 to 100 ± 10 meq/kg dry wt while[Ca]i rose from 347 ± 11 to 1,306 ± 89 nM.During hypertonic hypoxic perfusion (325 mosM), increases inNai and [Ca]i were reduced by 65 and 60%, respectively (P < 0.05). Hypertonicperfusion also diminished Na uptake after normoxic acidification by87% (P < 0.05). The data are consistent with the hypothesisthat mild hypertonic perfusion diminishes acid-induced Na accumulationand, thereby, decreases Na/Ca exchange-mediated Caiaccumulation during hypoxia.

We studied dynamics of cell surface expression ofproteolytically activated thrombin receptor (PAR-1) in human pulmonaryartery endothelial cells (HPAEC). PAR-1 activation was measured bychanges in cytosolic calcium concentration([Ca2+]i)and HPAEC retraction response (determined by real-time transendothelial monolayer electrical resistance).[Ca2+]iincrease in response to thrombin was abolished by preexposure to 25 nMthrombin for >60 min, indicating PAR-1 desensitization, butpreexposure to 25 nM thrombin for only 30 min or to 10 nM thrombin forup to 2 h did not desensitize PAR-1. Exposure to 10 or 25 nM thrombindecreased monolayer electrical resistance 40-60%. Cellspreexposed to 10 nM thrombin, but not those preexposed to 25 nMthrombin, remained responsive to thrombin 3 h later. Loss of cellretractility was coupled to decreased cell surface PAR-1 expression asdetermined by immunofluorescence. Cell surface PAR-1 disappeared uponshort-term (30 min) thrombin exposure but reappeared within 90 minafter incubation in thrombin-free medium. Exposure to 25 nM thrombinfor >60 min prevented rapid cycloheximide-insensitive PAR-1reappearance. Cycloheximide-sensitive recovery of cell surface PAR-1expression required 18 h. Therefore, both duration and concentration ofthrombin exposure regulate the time course of recovery of HPAEC surfacePAR-1 expression. The results support the hypothesis that initialrecovery of PAR-1 surface expression in endothelial cells results froma rapidly mobilizable PAR-1 pool, whereas delayed recovery results fromde novo PAR-1 synthesis. We conclude that thrombin itself regulatesendothelial cell surface PAR-1 expression and that decreased surfaceexpression interferes with thrombin-induced endothelial cell activation responses.

Recently, we demonstrated that the peroxisome proliferator-activated receptor- (PPAR-) ligands, either 15-deoxy-12,14-prostaglandin J2 (15d-PGJ2) or ciglitazone, increased endothelial nitric oxide (·NO) release without altering endothelial nitric oxide synthase (eNOS) expression (4). However, the precise molecular mechanisms of PPAR--stimulated endothelial·NO release remain to be defined. Superoxide anion radical (O2·) combines with ·NO to decrease·NO bioavailability. NADPH oxidase, which produces O2·, and Cu/Zn-superoxide dismutase (Cu/Zn-SOD), which degrades O2·, thereby contribute to regulation of endothelial cell·NO metabolism. Therefore, we examined the ability of PPAR- ligands to modulate endothelial O2· metabolism through alterations in the expression and activity of NADPH oxidase or Cu/Zn-SOD. Treatment with 10 µM 15d-PGJ2 or ciglitazone for 24 h decreased human umbilical vein endothelial cell (HUVEC) membrane NADPH-dependent O2· production detected with electron spin resonance spectroscopy. Treatment with 15d-PGJ2 or ciglitazone also reduced relative mRNA levels of the NADPH oxidase subunits, nox-1, gp91phox (nox-2), and nox-4, as measured using real-time PCR analysis. Concordantly, Western blot analysis demonstrated that 15d-PGJ2 or ciglitazone decreased nox-2 and nox-4 protein expression. PPAR- ligands also stimulated both activity and expression of Cu/Zn-SOD in HUVEC. These data suggest that in addition to any direct effects on endothelial·NO production, PPAR- ligands enhance endothelial·NO bioavailability, in part by altering endothelial O2· metabolism through suppression of NADPH oxidase and induction of Cu/Zn-SOD. Thrombinreceptor is activated by thrombin-mediated cleavage of the receptor'sNH2 terminus between Arg-41 andSer-42, generating a new NH2terminus that functions as a "tethered ligand" by binding tosites on the receptor. We prepared antibodies (Abs) directed againstspecific receptor domains to study the tethered ligand-receptor interactions required for signaling the increase in endothelial permeability to albumin. We used polyclonal Abs directed against thepeptide sequences corresponding to the extracellularNH2 terminus [residues70-99 (AbDD) and 1-160 (AbEE)] and extracellular loops 1 and 2 [residues 161-178 (AbL1) and 244-265(AbL2)] of the seven-transmembrane thrombin receptor. Receptoractivation was determined by measuring changes in cytosolicCa2+ concentration([Ca2+]i)in human dermal microvascular endothelial cells (HMEC) loaded withCa2+-sensitive fura2-acetoxymethyl ester dye. The transendothelial 125I-labeled albumin clearancerate (a measure of endothelial permeability) was determined across theconfluent HMEC monolayers. AbEE (300 µg/ml), directed against theentire extracellular NH2-terminal extension, inhibited the thrombin-induced increases in[Ca2+]iand the endothelial 125I-albuminclearance rate (>90% reduction in both responses). AbDD (300 µg/ml), directed against a sequence within theNH2-terminal extension, inhibited70% of the thrombin-induced increase in[Ca2+]iand 60% of the increased125I-albumin clearance rate. AbL2(300 µg/ml) inhibited these responses by 70 and 80%, respectively.However, AbL1 (300 µg/ml) had no effect on either response. Weconclude that NH2-terminalextension and loop 2 are critical sites for thrombin receptoractivation in endothelial cells and thus lead to increased[Ca2+]iand transendothelial permeability to albumin.

Effects of 5-day hypoxia on cardiac adrenergic neurotransmission in rats   总被引:1,自引:0,他引:1  
Chronic hypoxiainduces an overall sympathetic hyperactivation associated with amyocardial -receptor desensitization. The mechanismsinvolved in this desensitization were evaluated in 32 male Wistar ratskept in a hypobaric pressure chamber(PO2 = 40 Torr, atmospheric pressure = 450 Torr) for 5 days. In hypoxic compared with normoxic conditions,plasma norepinephrine (NE) levels were higher (2.1 ± 0.7 vs. 0.6 ± 0.2 ng/ml) with no difference in the plasma epinephrine levels(2.2 ± 0.7 vs. 1.8 ± 0.3 ng/ml). In hypoxia neuronal NE uptakemeasured by [3H]NE wasdecreased by 32% in the right ventricle (RV) and by 35% in the leftventricle (LV), and[3H]mazindol in vitrobinding showed a decrease in uptake-1 carrier protein density by 38%in the RV and by 41% in the LV. In vitro binding assays with[3H]CGP-12177 indicate-adrenoceptor density reduced by 40% in the RV and by 32% in theLV, and this was due to reduced1-subtype fraction (competitionbinding experiments with practolol). Hypoxia reduced the production ofcAMP induced by isoproterenol (36% decrease in the RV and 41%decrease in the LV), 5'-guanylylimododiphosphate (40% decreasein the RV and 42% decrease in the LV), and forskolin (39% decrease inthe RV and 41% decrease in the LV) but did not alter the effect ofMnCl2 and NaF. Quantitation ofinhibitory G-protein -subunit by immunochemical analysis showed a46% increase in the cardiac-specific isoformGi2 inhypoxic hearts. The present data demonstrate that in rats 5-day hypoxialeads to changes in pre- and postsynaptic myocardial adrenergicfunction. The myocardial desensitization associated with both areduction in externalized 1-adrenoceptor and an increasein inhibitory G-protein subunit may be caused by increased synaptic NElevels due to impaired uptake-1 system.

Failure toautoresuscitate by hypoxic gasping during prolonged sleep apnea hasbeen suggested to play a role in sudden infant death. Furthermore,maternal smoking has been repeatedly shown to be a risk factor forsudden infant death. The present experiments were carried out onnewborn rat pups to investigate the influence of perinatal exposure tonicotine (the primary pharmacological and addictive agent in tobacco)on their time to last gasp during a single hypoxic exposure and ontheir ability to autoresuscitate during repeated exposure to hypoxia.Pregnant rats received either nicotine (6 mg · kg1 · 24 h1) or vehiclecontinuously from day 6 of gestationto days 5 or 6 postpartum via an osmotic minipump.On days 5 or6 postpartum, pups were exposed eitherto a single period of hypoxia (97%N2-3% CO2) and their time to last gaspwas determined, or they were exposed repeatedly to hypoxia and theirability to autoresuscitate from primary apnea was determined. Perinatalexposure to nicotine did not alter the time to last gasp, but it didimpair the ability of pups to autoresuscitate from primary apnea. Aftervehicle, the pups were able to autoresuscitate from 18 ± 1 (SD)periods of hypoxia, whereas, after nicotine, the pups were able toautoresuscitate from only 12 ± 2 periods(P < 0.001) of hypoxia. Thus ourdata provide evidence that perinatal exposure to nicotine impairs the ability of newborn rats to autoresuscitate from primary apnea duringrepeated exposure to hypoxia, such as may occur during episodes ofprolonged sleep apnea.

We investigated theeffects of epidermal growth factor (EGF) on activeNa+ absorption by alveolarepithelium. Rat alveolar epithelial cells (AEC) were isolated andcultivated in serum-free medium on tissue culture-treated polycarbonatefilters. mRNA for rat epithelial Na+ channel (rENaC) -, -,and -subunits and Na+ pump1- and1-subunits were detected inday 4 monolayers by Northern analysisand were unchanged in abundance in day5 monolayers in the absence of EGF. Monolayerscultivated in the presence of EGF (20 ng/ml) for 24 h fromday 4 to day5 showed an increase in both1 and1Na+ pump subunit mRNA but noincrease in rENaC subunit mRNA. EGF-treated monolayers showed parallelincreases in Na+ pump1- and1-subunit protein by immunoblotrelative to untreated monolayers. Fixed AEC monolayers demonstratedpredominantly membrane-associated immunofluorescent labeling withanti-Na+ pump1- and1-subunit antibodies, withincreased intensity of cell labeling for both subunits seen at 24 hfollowing exposure to EGF. These changes inNa+ pump mRNA and protein precededa delayed (>12 h) increase in short-current circuit (measure ofactive transepithelial Na+transport) across monolayers treated with EGF compared with untreated monolayers. We conclude that EGF increases activeNa+ resorption across AECmonolayers primarily via direct effects onNa+ pump subunit mRNA expressionand protein synthesis, leading to increased numbers of functionalNa+ pumps in the basolateralmembranes.

Soluble form of IL-6 receptor alpha (sIL-6R) is known to serve as an agonist, without exogenous IL-6, on endothelial cells which do not express IL-6R but have only IL-6 receptor beta chain, gp130. We investigated the effect of sIL-6R on fractalkine expression in human umbilical vein endothelial cells (HUVECs) in culture. sIL-6R markedly inhibited HUVEC fractalkine/CX3CL1 expression induced by interleukin (IL)-1alpha, tumor necrosis factor (TNF)-alpha, or interferon (IFN)-gamma. IL-1alpha-induced fractalkine expression was inhibited by sIL-6R in time- and concentration-dependent manners. The experiment using actinomycin D indicated that sIL-6R lowered the stability of fractalkine mRNA. The inhibitory effect of sIL-6R was reversed by anti-gp130 neutralizing antibody. sIL-6R inhibited adhesion of mononuclear cells (MNCs) to HUVEC monolayers stimulated with IFN-gamma, but it did not inhibit the adhesion to monolayers stimulated with IL-1alpha. MNC chemotactic activity of conditioned medium of HUVEC stimulated with IL-1alpha or IFN-gamma was inhibited by co-treatment with sIL-6R. sIL-6R may play a regulatory role in immune responses by modulating the interaction between leukocytes and the vascular endothelium.  相似文献

Vascular endothelial growth factor (VEGF), an angiogenic factor induced by hypoxia, also exerts direct effects on neural tissues. VEGF up‐regulation after hypoxia coincides with expression of its two tyrosine kinase receptors Flt‐1(VEGFR‐1) and Flk‐1 (KDR/VEGFR‐2), which are the key mediators of physiological angiogenesis. We have recently shown that hypoxic‐preconditioning (PC) leading to tolerance to hypoxia–ischemia in neonatal piglet brain resulted in increased expression of VEGF. In this study, we used a hypoxic‐preconditioning model of ischemic tolerance to analyze the expression and cellular distribution of VEGF receptors and phosphorylation of cAMP‐response element‐binding protein (CREB) in newborn piglet brain. The response of Flt‐1 and Flk‐1 mRNA to PC alone was biphasic with peaks early (6 h) and late (1 week) after PC. The mRNA expression of Flt‐1 and Flk‐1 in piglets preconditioned 24 h prior to hypoxia–ischemia was significantly higher than non‐preconditioned piglets and remained up‐regulated up to 7 days. Furthermore, PC prior to hypoxia–ischemia significantly increased the protein levels of Flt‐1 and Flk‐1 compared with hypoxia–ischemia in a time‐dependent manner. Double‐immunolabeling indicated that both Flt‐1 and Flk‐1 are expressed in neurons and endothelial cells with a similar time course of expression following PC and that PC leads to the growth of new vessels. Finally, our data demonstrate that PC significantly phosphorylated and activated cAMP‐response element‐binding protein in nucleus. These results suggest that mechanism(s) initiated by PC can induce VEGF receptor up‐regulation in newborn brain and that VEGF–VEGF receptor‐coupled signal transduction pathways could contribute to the establishment of tolerance following hypoxia–ischemia.  相似文献

HumanNa+-K+-ATPase11,21, and 31heterodimers were expressed individually in yeast, and ouabainbinding and ATP hydrolysis were measured in membrane fractions. Theouabain equilibrium dissociation constant was 13-17 nM for11 and 31at 37°C and 32 nM for 21, indicatingthat the human -subunit isoforms have a similar high affinity forcardiac glycosides. K0.5 values for antagonism of ouabain binding by K+ were ranked in order as follows:2 (6.3 ± 2.4 mM) > 3(1.6 ± 0.5 mM)  1 (0.9 ± 0.6 mM),and K0.5 values for Na+ antagonismof ouabain binding to all heterodimers were 9.5-13.8 mM. Themolecular turnover for ATP hydrolysis by11 (6,652 min1) was abouttwice as high as that by 31 (3,145 min1). These properties of the human heterodimersexpressed in yeast are in good agreement with properties of the humanNa+-K+-ATPase expressed in Xenopusoocytes (G Crambert, U Hasler, AT Beggah, C Yu, NN Modyanov, J-DHorisberger, L Lelievie, and K Geering. J Biol Chem275: 1976-1986, 2000). In contrast to Na+ pumpsexpressed in Xenopus oocytes, the21 complex in yeast membranes wassignificantly less stable than 11 or31, resulting in a lower functionalexpression level. The 21 complex was also more easily denatured by SDS than was the11 or the31 complex.

Background: To evaluate the influence of hypoxia and molecular events in endothelial and embryonic stem cells.Materials and Methods: Human umbilical vein endothelial cells (HUVECs) and mouse embryoid body (EB) cells were subjected to hypoxic conditions for different time courses. DNA fragmentation assay, quantification of apoptotic cells by TUNEL assay measured by flowcytometry, and Western blot analysis for the molecular events of apoptosis were performed.Results: DNA fragmentation could be identified under hypoxic conditions in HUVECs and mouse EBs. The DNA fragmentation increased when the hypoxic interval was extended.In situ internucleosomal DNA fragmentation-TUNEL assay also found that the percentages of apoptotic cells increased gradually in HUVECs and mouse EBs when the hypoxic interval was extended. Furthermore, the levels of expression of p53 and Bax both increased in hypoxic conditions.Conclusions: Hypoxia increases both HUVEC and mouse EB apoptosis, which is associated with increase in p53/Bax expression.  相似文献
