Monthly observations (October 1972–October 1974) of a natural population of spined loach, Cobitis taenia (L.), in the River Great Ouse at Newport Pagnell indicated a patchy microdistribution, which varied seasonally. The distribution was clearly linked to the areas of fine substrate, which altered seasonally in position.Flow rates measured at monthly intervals in areas where Cobitis taenia (L.) were present were found to be approximately half those of areas in which Cobitis taenia L. were not found (0.148 m/sec and 0.293 m/sec). Substrate samples from areas where Cobitis taenia L. were present had a fine organic component, in contrast to the remainder of the river bed, which consisted of hard-packed gravel.In the laboratory, choice chamber experiments demonstrated a positive selection for fine organic deposits.  相似文献

The suitability of acrylic colours for marking stone loach (Noemacheilus barbatulus L.) Acrylic colours were tested in the stone loach Noemacheilus barbatulus and were found to be useful for long term marking of small benthic fishes. No negative influences on condition or growth of the fishes were observed. The colours green and white produced best results in durability, still being visible in 90 % of the specimens after one year.  相似文献

The present work provides new data on the banding pattern of diploid Cobitis taenia and its triploid hybrid females, which belong to the diploid–polyploid complex in the Vistula River tributary. C-banding, silver-staining (Ag), and fluorescent staining with chromomycin A3 techniques were used to describe the diploid and triploid karyotype. The karyotype of Cobitis taenia of 2n=48 was characterised by one pair of NOR-bearing subtelocentric chromosomes and at least four chromosomes with CMA3-positive sites. The C-positive heterochromatin was present in the centromeres of almost all chromosomes and the pericentromeric regions of several metacentric and submetacentric chromosomes. The triploid females of 3n=74 had two pairs of chromosomes with active NORs. The NORs-sites were located terminally on two biarmed and two uniarmed chromosomes. The CMA3-staining revealed at least six A3-positive sites. The C-banded and A3-stained triploid karyotype was composed of haploid set of Cobitis taenia and diploid set of unidentified species, so heterochromatin pattern confirmed the possibility of their hybrid origin. The characteristics of banded diploid and triploid karyotype, and the hypothetical karyotype of an unknown species of 2n=50 is discussed. The chromosomal complement of Cobitis taenia was analysed by replication banding techniques to determine whether there were specific patterns that could allow distinction of the different chromosomes. The diploid chromosome number of 2n = 48 is diagnostic of this species. In vivo 5-bromodeoxyuridine (5-BrdU) incorporation induced highly reproducible replication bands. Most of the chromosome pairs were distinguishable on the base of their banding patterns. The karyotype, consisting of five pairs of metacentrics, nine pairs of submetacentrics and 10 pairs of subtelocentrics and acrocentrics, was confirmed. C-banding and replication banding patterns were compared, and heterochromatin was both early and later replicating. C-positive heterochromatin in centromeric regions was mainly early replicating, but that located in pericentromeric regions was late replicating. Most of the late-replicating regions found interstitially were C-band negative. The results obtained so far for combined chromosomal staining methods of C. taenia and other Cobitis fish species are discussed.  相似文献

The spined loach Cobitis taenia L. creates exclusively diploid and mixed diploid–polyploid populations. Allotriploid females, which co-exist with C. taenia or C. elongatoides and a few tetraploid males and females dominate in most Cobitis mixed populations. They reproduce gynogenetically and produce triploid eggs that are stimulated to development by sperm from Cobitis males. Some of these eggs are fertilized, which leads to the production of bisexual tetraploids. Males of C. taenia (2n = 48) from a diploid population in Lake Klawój, Northern Poland (46 individuals) and from a mixed Cobitis population in the Bug River, Eastern Poland (7 individuals), and three tetraploid males (4n = 98) from the same mixed population were examined. All the fish were analyzed karyologically and histologically. Tubules with cysts of the testes of C. taenia from both populations were filled with germ cells at various developmental stages. Among fishes from Lake Klawój sperm maturation in batches simultaneous with the batch spawning of C. taenia females was found. The testes of the loach C. taenia, from a mixed population in the Bug River, were filled with spermatozoa over the entire reproductive season. Sperm maturation in batches was not observed. Sperm maturation in batches seems to be only connected with a few diploid males in this population. So, a continuous process of spermatogenesis in their testes is required. Only in the testes of all tetraploid Cobitis males were cells characteristic of the early stages of spermatogenesis observed, i.e. without spermatids and spermatozoa. Furthermore, the histological sections of the testis of a male captured in August, revealed fragments with connective tissue between the germ cells. However the participation of tetraploid, infertile Cobitis males in the process of reproduction in the investigated mixed population remains controversial. The results obtained so far, reveal that even the infertile sperm of tetraploid males may induce gynogenesis in Cobitis triploid females.  相似文献

Unlike mulberry (Morus alba, M.a.), paper mulberry (Broussonetia papyrifera, B.p.) can acclimate to Karst soil and incline to alien invasion. The photosynthetic parameters, diurnal changes of carbonic anhydrase, and chlorophyll fluorescence induction, and water potential were measured on sunny days (SD) and cloudy days (CD). Photosynthetic midday depression occurred in B.p. but not in M.a. The irradiance-and CO2-saturated photosynthetic rates of B.p. were significantly higher than those of M.a. There was no significant difference in water use efficiency between the two species on a SD. The maximum fluorescence, maximum quantum yield, photochemical quenching, and relative electron transport rate in the leaves of B.p. were much higher than those in M.a. The activity of carbonic anhydrase (CA) of B.p., on either an SD or a CD, was much greater than that of M.a. Higher transpiration rate (E) and net photosynthetic rate (P N) of B.p. resulted in the lack of water in mesophyll cells. Although a higher CA activity of B.p. supplied both water and CO2 for the photosynthesis of mesophyll cells, water in mesophyll cells was the factor limiting photosynthesis, and the intercellular CO2 concentration of B.p. was high and stable.  相似文献

751 specimens of spined loach, Cobitis taenia (L.), were collected and studied over the period October 1972–October 1974. The majority of the males were found to die at the end of the 2+ season and the majority of the females at the end of the 3+ season. Back calculations for length at age were carried out from otolith measurements and found to correspond closely to actual measurements. Males and females grew similarly up until the end of their 1+ year of life, with most rapid growth occurring in the first year. Females continued to grow rapidly up to 2+, the rate then being reduced; the males showed an increasingly reduced rate of growth after the 1+ year. Ford Walford plots gave L∞ results which corresponded closely in most cases with the largest observed specimens. Gonad development and maturation of males and females followed different patterns. Males matured during the 1 + year at a length of approximately 40 mm from which point their dry gonad weight remained at a steady 0.02–0.03% of the body weight. Females appeared to mature a year later and their ovary weight and oocyte diameter increased with fish age. A seasonal cycle in gonad development was evident in females. There was a greater proportion of females to males, especially in the older age group. Spawning time was estimated to be between June and July.  相似文献

The sulphide-tolerant brackish water isopod Saduria entomon has haemocyanin with a high affinity for oxygen. Sulphide is oxidized in the hepatopancreas to thiosulphate. Oxygen needed for this oxidation is transported bound to haemocyanin. Thiosulphate was found to improve haemocyanin oxygen affinity in S. entomon. The modulating effect was time and concentration dependent. The effect was greater at higher pH, i.e. the Bohr effect was greater (−1.82) compared to that seen without thiosulphate (−1.36). S. entomon exposed to simultaneous severe hypoxia and sulphide had a low haemolymph pH and the partial pressure of oxygen required to reach 50% saturation was even lower than that measured in vitro. This is ascribed to the combined effects of thiosulphate and lactate as well as unknown co-factors. Thiosulphate had no effect at all in the sulphide and hypoxia non-tolerant natantian Crangon crangon. It is suggested that thiosulphate can be important as a haemocyanin modulator for crustaceans digging in sulphide-rich sediments or constantly exposed to sulphide. Accepted: 11 August 1999  相似文献

Biochemical genetic typing and cytometry showed that polyploid females account for 87% of the spined loachCobitis taeniapopulation from the middle Dnieper basin. The polyploidy series included triploids, tetraploids, and, possibly, a few pentaploids. A characteristic feature of the genetic structure of polyploids was that their genetic variation was due to the clonal variation in the haploid portion of the genome originating from Cobitis sp. and to polymorphism of the diploid portion originating fromC. taenia. The results are discussed with regard to comparative evolution of alloploid complexes in fish and terrestrial vertebrates.  相似文献

The contents of eight trace elements (Ti, Mn, Ni, Cu, Zn, Sr, Ba, and Pb) in muscles of syntopic sexual and clonal spined loaches (the golden loach Sabanejewia baltica, diploid males and females of Cobitis taenia, and congeneric triploid clonal females) from the upper Dnieper River and in the syntopic spined loaches (males and females of C. melanoleuca and males of C. taenia) from the upper Volga River basin were determined using the X-ray fluorescence spectroscopy technique. The contents of Cu in the clonal triploid Cobitis females and diploid C. taenia females from the Dnieper are different. The intersexual differences in the contents of Pb and Zn between C. taenia males and females, as well as the interspecific differences in the content of Ti between the spined loaches from the Dnieper and Volga basins were revealed. The concentrations of Cu and Pb correlated with the individual fish size. The potential for the use of the revealed differences in the trace element contents as an indicator of the divergence of ecological niches in the syntopic spined loaches is discussed.  相似文献

Transpiration and water absorption rates, stomatal and cuticular resistances to water vapour diffusion, were measured onPlantago maritima (halophyte) andPlantago lanceolata (glycophyte) grown in the presence of NaCl or Na2SO4. Water absorption was reduced in the presence of Na2SO4 and transpiration rate was increased when NaCl was added to the nutrient solution. This study was conducted to describe the major and the minor rDNA chromosome distribution in the spined loach Cobitis taenia (2n = 48) and the Danubian loach Cobitis elongatoides (2n = 50), and their laboratory-produced diploid reciprocal F1 hybrid progeny. It was tested by fluorescence in situ hybridisation (FISH) whether the number of 28s and 5s rDNA sites in the karyotypes of diploid hybrids corresponds to the expectations resulting from Mendelian ratio and if nucleolar organiser regions (NOR)were inherited from both parents or nucleolar dominance can be observed in the induced F1 hybrid progeny. Ten (females) or twelve (males) 28s rDNA loci were located in nine uniarm chromosomes of C. taenia. Two of such loci terminally bounded on one acrocentric chromosome were unique and indicated as specific for this species. Large 5s rDNA clusters were located on two acrocentric chromosomes. In C. elongatoides of both sexes, six NOR sites in terminal regions on six meta-submetacentric chromosomes and two 5s rDNA sites on large submetacentrics were detected. The F1 hybrid progeny (2n = 49) was characterised by the intermediate karyotype with the sites of ribosome synthesis on chromosomes inherited from both parents without showing nucleolar dominance. 5s rDNA sites were detected on large submetacentric and two acrocentric chromosomes. The observed number of both 28s and 5s rDNAs signals in F1 diploid Cobitis hybrids was disproportionally inherited from the two parental species, showing inconsistency with the Mendelian ratios. The presented rDNA patterns indicate some marker chromosomes that allow the species of the parental male and female to be recognised in hybrid progeny. The 5s rDNA was found to be a particularly effective diagnostic marker of C. elongatoides to partially discern genomic composition of diploid Cobitis hybrids and presumably allopolyploids resulting from their backcrossing with one of the parental species. Thus, the current study provides insight into the extent of rDNA heredity in Cobitis chromosomes and their cytotaxonomic character.  相似文献

Hyperhydricity is a physiological disorder frequently affecting shoots propagated in vitro. Since it negatively affects shoot multiplication vigor, and impedes the successful transfer of micropropagated plants to in vivo conditions, hyperhydricity is a major problem in plant tissue culture. In commercial plant micropropagation, there are reports of up to 60% of cultured shoots or plantlets which demonstrate hyperhydricity, which reflects the pervasiveness of this problem. The phenomenon has been correlated to water availability, microelements, and/or hormonal imbalance in the tissue culture. In this study, the ultrastructure and the characteristics of reactive oxygen species between hyperhydric and normal shoots of garlic were studied. We observed that in some cells of hyperhydric tissues, the intranuclear inclusion was separated, the mitochondrion was swollen and its intracristae had splits, the organelles were compressed against the cell wall, and the chloroplasts and intergranal thylakoids were also compressed. Additionally, the content of chlorophyll and soluble protein in hyperhydric shoots decreased significantly. For instance, chlorophyll a decreased 43.61%, chlorophyll b decreased 49.29%, chlorophyll a+b decreased 48.10%, and soluble protein dropped 47.36%. In contrast, the O2 generation rate and H2O2 level increased 45.36% and 63.98%, respectively, obviously higher than the normal shoots. Lipoxygenase activity and malondialdehyde content in the hyperhydric shoots increased significantly, while the electrolyte leakage rose, indicating a serious membrane lipid peroxidatic reaction. Superoxide dismutase, peroxidase, catalase, glutathione peroxidase, and ascorbate peroxidase activities in hyperhydric tissue were all significantly higher than in normal leaf tissue. The antioxidant metabolism demostrated a close connection between hyperhydricity and reactivated oxygen species.  相似文献

A single incomplete specimen of Teleost fish has been discovered in lignitic shales of Pliocene age croppingout West of Rochefort-Montagne (Puy-de-Dôme, France). The specimen is referred to the recent species Cobitis taenia L., to which it appears identical in every respect. The presence of Cobitis cf. taenia L. in the Pliocene of Rochefort-Montagne allows the environment of deposition of the lignitic shales, which are rich in fossil plants, to be more precisely defined.  相似文献

Sudden events of seagrass die‐off have been suggested to be induced by invasion of the phytotoxin sulphide under environmental stress generating low oxygen supply in seagrass tissues. Laboratory experiments were conducted with eelgrass (Zostera marina L.) to measure intra‐plant changes in oxygen and sulphide by means of microelectrodes at different oxygen concentrations in the water column. The objectives were to examine whether sulphide intrusion into seagrass tissues can be induced, to determine the role of plant oxygen status for sulphide intrusion and to determine how fast internal sulphide pools are depleted after internal oxygen supplies have been restored. Under conditions with oxygen partial pressures (pO2) above 7.4 kPa (> 35% of air saturation) within eelgrass rhizomes or meristematic tissues no intrusion of sulphide occurred in spite of high sediment concentrations of gaseous sulphide (> 1000 µm ). Lack of sulphide intrusion at high internal pO2 suggested that oxygen release from the roots ensured complete re‐oxidation of sulphide in the rhizosphere. Under oxygen stress, however, the experiments clearly demonstrated intrusion of sulphide in eelgrass rhizomes and meristematic tissues. Rates of sulphide intrusion were controlled by internal pO2, which in turn was controlled by water column oxygen concentrations. Maximum internal sulphide content reached 325 µm which by far exceeded the 1–10 µm known to inhibit mitochondrial activity in eukaryotic cells. Sulphide and low levels of oxygen could coexist in the eelgrass tissues reflecting fast internal transport of sulphide and slow rates of sulphide re‐oxidation. Upon re‐establishment of high internal oxygen concentrations the depletion of the sulphide pool was slow (half‐life = 20–30 min) indicating, that sulphide re‐oxidation within the eelgrass tissue was not bacterially or enzymatically facilitated but occurred by simple chemical oxidation. The results of this study are consistent with the proposed detrimental role of sulphide intrusion in events of sudden seagrass die‐off.  相似文献

Nodulated soybean (Glycine max L. Merr. cv White Eye inoculated with Bradyrhizobium japonicum strain CB 1809) plants were cultured in the absence of combined N from 8 to 28 days with their root systems maintained continuously in 1, 2.5, 5, 10, 20, 40, 60, or 80% O2 (volume/volume) in N2. Plant dry matter yield was unaffected by partial pressure of oxygen (pO2) and N2 fixation showed a broad plateau of maximum activity from 2.5 to 40 or 60% O2. Slight inhibition of nitrogenase activity occurred at 1% O2 and as much as 50% inhibition occurred at 80% O2. Low pO2 (less than 10%) decreased nodule mass on plants, but this was compensated for by those nodules having higher specific nitrogenase activities. Synthesis and export of ureides in xylem was maintained at a high level (70-95% of total soluble N in exudate) over the range of pO2 used. Measurements of nitrogenase (EC 1.7.99.2) activity by acetylene reduction indicated that adaptation of nodules to low pO2 was largely due to changes in ventilation characteristics and involved increased permeability to gases in those grown in subambient pO2 and decreased permeability in those from plants cultured with their roots in pO2 greater than ambient. A range of structural alterations in nodules resulting from low pO2 were identified. These included increased frequency of lenticels, decreased nodule size, increased volume of cortex relative to the infected central tissue of the nodule, as well as changes in the size and frequency of extracellular voids in all tissues. In nodules grown in air, the inner cortex differentiated a layer of four or five cells which formed a band, 40 to 50 micrometers thick, lacking extracellular voids. This was reduced in nodules grown in low pO2 comprising one or two cell layers and being 10 to 20 micrometers thick in those from 1% O2. Long-term adaptation to different external pO2 involved changes which modify diffusive resistance and are additional to adjustments in the variable diffusion barrier.  相似文献

Oxygen consumption by Thais varied seasonally with higher values in summer than in winter. This seasonal difference was due in part to the effects of temperature and in part to those of feeding. During feeding, rates of oxygen consumption were high, but declined in the period between meals. There was little evidence of acclimation of oxygen consumption to changes in temperature; low (winter) rates of consumption were more sensitive to increases in temperature than were high (summer) rates. Net photosynthetic rates (P n) of easy (EK 16-3) and difficult-to-acclimatize (EK 11-1) sea oats genotypes were examined under the following culture conditions: (1) photoautotrophic [sugar-free medium, high photosynthetic photon flux (PPF), high vessel ventilation rates and CO2 enrichment, (PA)]; (2) modified photomixotrophic [sugar-containing medium diluted with sugar-free medium over time, high PPF, and high vessel ventilation rates (PM)]; (3) modified photomixotrophic enriched [same as PM with CO2 enrichment, (PME)]; or (4) conventional photomixotrophic [sugar-containing medium, low PPF, and low vessel ventilation rates (control)]. Regardless of genotype, plantlets cultured under PA conditions died within 2 wk, whereas under PM and PME conditions, plantlets increased their P n. After 6 wk, P n per gram dry weight was 1.7 times greater in EK 16-3 than EK 11-1 plantlets cultured under PME conditions. In vitro-produced leaves of EK 16-3 plantlets were elongated with expanded blades, whereas EK 11-1 produced short leaves without expanded blades, especially under control conditions. After in vitro culture, EK 16-3 PME plantlets exhibited the highest dry weights among treatments. EK 16-3 PME and EK 16-3 PM had similarly high survivability, shoot and root dry weights and leaf lengths ex vitro compared to EK 16-3 control and EK 11-1 PM and PME plantlets. Ex vitro growth, survivability and P n per leaf area of either genotype were not affected by CO2 enrichment under modified photomixotrophic conditions. These results suggest that growth and survivability of sea oats genotypes with different acclimatization capacities can be enhanced by optimizing culture conditions.  相似文献

Nitrogenous excretion in two snails, Littorina saxatilis (high intertidal) and L. obtusata (low intertidal) was studied in relation to temperature acclimation (at 4° and 21°C), including total N excretion rates, the fraction of urea in N excretion, corresponding O:N ratios and the partitioning of deaminated protein between catabolic and anabolic processes at 4°, 11° and 21°C. Aggregate N excretion rates in both species showed no significant compensatory adjustments following acclimation. Total weight specific N excretion rates at 21°C were higher in standard 3 mg L. saxatilis (739 ng N mg−1 h−1) than standard 5 mg L. obtusata (257 ng N mg−1 h−1) for snails acclimated to 21°C. Comparisons of Q10 values of total weight specific N excretion to Q10 values for weight specific oxygen consumption ({xxV}O2) between 4° to 11 °C and 11° to 21°C indicated that, while total rates of catabolic metabolism ({xxV}O2) and protein deamination in L. obtusata were essentially parallel, the relationship between N excretion and {xxV}O2 in L. saxatilis revealed the partitioning of a larger share of deaminated protein carbon into anabolism at 4° and 21°C than at 11°C. Urea N accounted for a larger share of aggregate N excreted in L. saxatilis than in L. obtusata, but in both species urea N is a greater proportion of total N excreted when acclimated at 4°C (urea N: ammonia N ratio range: 1 to 2.15) than in snails acclimated to 21°C (urea N: ammonia N ratio range: 0.46 to 1.39). Molar O:N ratios indicate that the proportion of metabolism supported by protein catabolism is greater in L. saxatilis (O:N range: 2.5–8.4) than in L. obtusata (O:N range: 7.3–13.0). In both species, regardless of acclimation temperature, the O:N ratios are generally lowest (high protein catabolism) at 4°C and highest at 21°C.  相似文献

For the systematic study of the gonostomatid fishes of theDiplophos taenia species complex, a total of 698 specimens was obtained from the three oceans. Four valid species were recognized:D. taenia Günther,D. proximus Parr,D. orientalis Matsubara, andD. australis sp. nov. The diagnostic characters are 90–100 total vertebrae (TV) (37–41 abdominal (AV) +52–60 caudal vertebrae (CV)) and 103–115 IC photophores (IC) for D.taenia, 85–90 TV (36–39 AV + 48–52 CV) and 98–104 IC forD. proximus, 83–86 TV (33–35 AV + 49–52 CV) and 92–100 IC for D.orientalis, and 84–91 TV (33–37 AV+ 50–54 CV) and 99–105 IC forD. australis. In addition to the above,D. proximus has larger orbital diameter (the proportion to head length 21–28%) than the other three species (15–23%) beyond 70mm in standard length. Due to wide distribution,D. taenia shows meristic variations: the numbers of TV, IC and anal fin rays (A) are smaller at lower latitudes and larger at higher ones in all oceans, and the number of A is smaller in the Atlantic (56–71) than in the other oceans (59–72) of the same latitude. Because of these variations, identification to species level is possible only area by area. The distribution of each of the four species is also distinct:D. taenia is a cosmopolitan between about 40° N and 30° S exclusive of the eastern tropical Pacific;D. proximus is endemic to the eastern tropical Pacific;D. orientalis is limited to the North Pacific transitional zone between about 30° and 40° N; andD. australis in a transitional zone of the Southern Ocean south of 20° S.  相似文献
