Oxic–anoxic interfaces harbor significant numbers and activity of chemolithoautotrophic microorganisms, known to oxidize reduced sulfur or nitrogen species. However, measurements of in situ distribution of bulk carbon dioxide (CO2) assimilation rates and active autotrophic microorganisms have challenged the common concept that aerobic and denitrifying sulfur oxidizers are the predominant autotrophs in pelagic oxic–anoxic interfaces. Here, we provide a comparative investigation of nutrient, sulfur, and manganese chemistry, microbial biomass distribution, as well as CO2 fixation at the pelagic redoxcline of the eastern Gotland Basin, Baltic Sea. Opposing gradients of oxygen, nitrate, and sulfide approached the detection limits at the chemocline at 204 m water depth. No overlap of oxygen or nitrate with sulfide was observed, whereas particulate manganese was detected down to 220 m. More than 70% of the bulk dark CO2 assimilation, totaling 9.3 mmol C m−2 day−1, was found in the absence of oxygen, nitrite, and nitrate and could not be stimulated by their addition. Maximum fixation rates of up to 1.1 μmol C L−1 day−1 were surprisingly susceptible to altered redox potential or sulfide concentration. These results suggest that novel redox-sensitive pathways of microbial sulfide oxidation could account for a significant fraction of chemolithoautotrophic growth beneath pelagic chemoclines. A mechanism of coupled activity of sulfur-oxidizing and sulfur-reducing microorganisms is proposed.  相似文献

Marine pelagic redoxclines are zones of high dark CO2 fixation rates, which can correspond up to 30% of the surface primary production. However, despite this significant contribution to the pelagic carbon cycle, the identity of most chemolithoautotrophic organisms is still unknown. Therefore, the aim of this study was to directly link the dark CO2 fixation capacity of a pelagic redoxcline in the central Baltic Sea (Landsort Deep) with the identity of the main chemolithoautotrophs involved. Our approach was based on the analysis of natural carbon isotope signatures in fatty acid methyl esters (FAMEs) and on measurements of CO2 incorporation in 13C-bicarbonate pulse experiments. The incorporation of 13C into chemolithoautotrophic cells was investigated by rRNA-based stable isotope probing (RNA-SIP) and FAME analysis after incubation for 24 and 72 h under in situ conditions. Our results demonstrated that fatty acids indicative of Proteobacteria were significantly enriched in 13C slightly below the chemocline. RNA-SIP analyses revealed that two different Gammaproteobacteria and three closely related Epsilonproteobacteria of the Sulfurimonas cluster were active dark CO2-fixing microorganisms, with a time-dependent community shift between these groups. Labelling of Archaea was not detectable, but after 72 h of incubation the 13C-label had been transferred to a potentially bacterivorous ciliate related to Euplotes sp. Thus, RNA-SIP provided direct evidence for the contribution of chemolithoautotrophic production to the microbial food web in this marine pelagic redoxcline, emphasizing the importance of dark CO2-fixing Proteobacteria within this habitat.  相似文献

SYNOPSIS. Heterotrophic (dark) CO2 fixation by Euglena gracilis strain Z varies with phase of batch culture and mode of nutrition. Dark CO2 fixation increased transiently during the growth of cells under photoautotrophic (CO2, light) and heterotrophic (glucose, dark) conditions. Cells grown heterotrophically with acetate or ethanol had no transient increase in fixation. The addition of acetate to a heterotrophically growing culture during the period of increasing dark CO2 fixation resulted in rapid elimination of this fixation. The results suggest that dark CO2 fixation in Euglena functions in anaplerotic feeding of the tricarboxylic acid cycle, drained by biosyntheses during growth. Induction of the glyoxylate cycle by acetate may provide an alternate source of tricarboxylic cycle intermediates, obviating the requirement for dark CO2 fixation as a source of the intermediates.  相似文献

The maximum specific growth rates of Pseudomonas fragi, Bacillus cereus and Streptococcus cremoris were studied over a wide range of carbon dioxide concentrations. The growth rate compared with a control was reduced to 50% in Ps. fragi at 0–5 atm CO2, in B. cereus at 1—3 atm and in Strep, cremoris at 8–6 atm. B. cereus and Strep, cremoris were completely inhibited at 3 and 11 atm CO2, respectively. The growth rate of the aerobic Ps. fragi at 0–99 atm CO2 (0–01 atm oxygen) was reduced to about 20% of that in air. The growth rate of Ps. fragi was decreased at oxygen concentrations lower than 0–01 atm.
When Ps. fragi was grown at oxygen limitation (0.0025 atm oxygen) and exposed to 0.99 atm CO2, the inhibiting effect of the CO2 was added to that of the oxygen limitation. No indications of a synergistic effect between CO2 inhibition and oxygen limitation were noted.

The Nostoc-Gunnera symbiosis: carbon fixation and translocation   总被引:2,自引:0,他引:2  
The in vitro specific activity of ribulose-1,5-bisphosphate carboxylase (Rubisco; EC 4. 1. 1. 39) and the dark and light in vivo CO2 fixation activities were determined in the cyanobiont of Gunnera . Compared to the free-living isolate Nostoc PCC 9231, the in vitro Rubisco activity was high, while the in vivo CO2 fixation was very low. Light did not significantly influence CO2 fixation if the cyanobiont was left in the sliced Gunnera tissues, while a small light stimulation was found for CO2 fixation of the freshly-isolated cyanobiont. The adjacent non-infected Gunnera tissue showed a very low CO2 fixation. A rapid translocation of fixed 14CO2 from leaves towards apical parts of the plant was apparent, in particular to the symbiotic tissue. The 14C label appeared mainly in soluble form in this tissue and was rapidly catabolised as shown by 14C chase experiments. Also, short-term experiments revealed that maximum 14C accumulation occurred in the symbiotic tissue showing the highest rates of nitrogen fixation (Söderbäck et al. 1990), about 10–15 mm from the plant apex. The data were taken to indicate that there is a modification in the photosynthetic light reaction of the cyanobiont and that the cyanobiont lives heterotrophically in the dark on photo-synthate rapidly delivered from nearby leaves of the host plant.  相似文献

During starch degradation in intact isolated chloroplasts from Chlamydomonas reinhardtii gas exchange was studied with a mass spectrometer. Oxygen uptake by intact chloroplasts in the dark never exceeded 1.5% of the starch degradation rate [maximum 15 nmol O2 (mg Chl)−1 h−1 consumed. 1 000 nmol glucose (mg Chl)−1h−1 degraded]. Evolution of CO2 under aerobic conditions [9.8–28 nmol (mg Chl)−1 h−1] was stimulated by addition of 0.1–0.5 m M oxaloacetate [393–425 nmol CO2 (mg Chl)−1 h−1]. Pyridoxal phosphate (5 m M ) inhibited starch degradation by more than 80%, but had no effect on O2 uptake. Starch degradation rates and CO2 evolution did not differ under acrobic and anaerobic conditions. Increasing Pi in the reaction medium from 0.5 m M to 5.0 m M stimulated starch degradation by 230 and 260% under aerobic and anaerobic conditions, respectively. A rapid autooxidation of reduced ferredoxin was observed in a reconstituted system consisting of purified Chlamydomonas ferredoxin, purified Chlamydomonas NADP-ferredoxin oxidoreductase (EC 1.6.7.1) and NADPH. Addition of isolated thylakoids from C. reinhardtii did not affect the rate of O2 uptake. Our results clearly indicate the absence of any oxygen requirement during starch degradation in isolated chloroplasts.  相似文献

Delayed luminescence was measured from samples of a synchronously growing cell culture of the unicellular green alga, Scenedesmus obtusiusculus Chod., every second hour during the 24 h cell cycle under a 15/9 h lighi/dark regime. Both high (air + 2.5% CO2) and low (0.03% CO2) CO2 conditions were used. Under high CO2 conditions, while light excitation induces formation of a late (maximum reached after 10–60 s) transient peak in delayed luminescence in cells sampled after 10–16 h in the cell cycle. During most of the cell cycle low CO2 conditions stimulate a late transient peak formation. Excitation with 700 nm light, but not with 680 nm light, induces a late transient peak in delayed luminescence under high CO2 conditions. The transient peak is more or less pronounced depending on the cell stage. The variations might be due to a changing capacity for light-induced state I/stale II transitions during the cell cycle. It is assumed that the formation of a late transient peak in delayed luminescence is due to ATP hydrolyzation and is thus favoured by a high ATP/NADPH ratio. Hydrolyzation of ATP affects the transthylakoidal ΔpH, which regulates the reverse electron flow to the plastoquinone-pool and QA/QB, thus affecting the decay kinetics of the delayed luminescence.  相似文献

Abstract. A controlled-environment chamber constructed from a standard chest freezer was used to grow and measure the CO2 exchange of small stands of lettuce ( Lactuca sativa L. ev. Ambassador). The chamber, with horizontal air flow, provided good control of air temperature ( c. 6 to 16°C), irradiance (0–300 μmol PAR m 2S1 and CO2 (350–1000 μmol mol−1). The photosynthetic response to changes in these variables was measured using an inexpensive CO2 dosing system which recorded the input rate required to maintain a constant concentration of CO2 (to ± 2.5%). Characteristis of the growth environment and the changes in response to temperature and irradiance are described.  相似文献

Abstract. Cyperus longus L. , which has a widespread but disjunct distribution throughout Europe and extends northwards into Britain, was found to be a C4 species based upon its Kranz leaf anatomy, low CO2 compensation point and the labelling of malate as an early product of 14CO2 fixation. The photosynthetic characteristics of C. longus are similar to many other C4 species with a high maximum rate of photosynthesis (> 1.5 mg CO2 m −2 s −1) and a relatively high temperature optimum (30–35°C), but unlike many C4 species the rate of photosynthesis does not decline rapidly below the optimum temperature and a substantial rate (0.6 mgCO2 m−2s−1)occursat 15°C. Leaf extension is very slow at 15°C and shows a curvilinear response to temperatures between 15 and 25°C. Leaves extend at a rate of almost 4 cm d−1 at 25°C.  相似文献

Sugar-beet plants ( Beta vulgaris L. cv. Monohill) were cultivated for 4 weeks in a complete nutrient solution. Indirect effects of cadmium were studied by adding 5, 10 or 20 μ M CdCl2 to the culture medium while direct effects were determined by adding 1, 5, 20, 50 or 2 000 μ M CdCl2 to the assay media. The photosynthetic properties were characterized by measurement of CO2 fixation in intact plants, fluorescence emission by intact leaves and isolated chloroplasts, photosystem (PS) I and PSII mediated electron transport of isolated chloroplasts, and CO2-dependent O2 evolution by protoplasts. When directly applied to isolated leaves, protoplasts and chloroplasts. Cd2+ impeded CO2 fixation without affecting the rates of electron transport of PSI or PSII or the rate of dark respiration. When Cd2+ was applied through the culture medium the capacity for, and the maximal quantum yield of CO2 assimilation by intact plants both decreased. This was associated with: (1) decreased total as well as effective chlorophyll content (PSII antennae size), (2) decreased coupling of electron transport in isolated chloroplasts, (3) perturbed carbon reduction cycle as indicated by fluorescence measurements. Also, protoplasts isolated from leaves of Cd2+-cultivated plants showed an increased rate of dark respiration.  相似文献

To determine how increased atmospheric CO2 will affect the physiology of coppiced plants, sprouts originating from two hybrid poplar clones ( Populus trichocarpa × P. deltoides - Beaupre and P. deltoides × P. nigra - Robusta) were grown in open-top chambers containing ambient or elevated (ambient + 360 μmol mol−1) CO2 concentration. The effects of elevated CO2 concentration on leaf photosynthesis, stomatal conductance, dark respiration, carbohydrate concentration and nitrogen concentration were measured. Furthermore, dark respiration of leaves was partitioned into growth and maintenance components by regressing specific respiration rate vs specific growth rate. Sprouts of both clones exposed to CO2 enrichment showed no indication of photosynthetic down-regulation. During reciprocal gas exchange measurements, CO2 enrichment significantly increased photosynthesis of all sprouts by approximately 60% ( P < 0.01) on both an early and late season sampling date, decreased stomatal conductance of all sprouts by 10% ( P < 0.04) on the early sampling date and nonsignificantly decreased dark respiration by an average of 11%. Growth under elevated CO2 had no consistent effect on foliar sugar concentration but significantly increased foliar starch by 80%. Respiration rate was highly correlated with both specific growth rate and percent nitrogen. Long-term CO2 enrichment did not significantly affect the maintenance respiration coefficient or the growth respiration coefficient. Carbon dioxide enrichment affected the physiology of the sprouts the same way it affected these plants before they were coppiced.  相似文献

Abstract. Apart from its impact on global warming, the annually increasing atmospheric [CO2] is of interest to plant scientists primarily because of its direct influence on photosynthesis and photorespiration in C3 species. But in addition, 'dark' respiration, another major component of the carbon budget of higher plants, may be affected by a change in [CO2] independent of an increase in temperature. Literature pertaining to an impact of [CO2] on respiration rate is reviewed. With an increase in [CO2], respiration rate is increased in some cases, but decreased in others. The effects of [CO2] on respiration rate may be direct or indirect. Mechanisms responsible for various observations are proposed. These proposed mechanisms relate to changes in: (1) levels of nonstructural carbohydrates, (2) growth rate and structural phytomass accumulation, (3) composition of phytomass, (4) direct chemical interactions between CO2 and respiratory enzymes, (5) direct chemical interactions between CO2 and other cellular components, (6) dark CO2 fixation rate, and (7) ethylene biosynthesis rate. Because a range-of (possibly interactive) effects exist, and present knowledge is limited, the impact of future [CO2] on respiration rate cannot be predicted. Theoretical considerations and types of experiments that can lead to an increase in the understanding of this issue are outlined.  相似文献

1. Inorganic carbon availability influences species composition of phytoplankton in acidic and highly alkaline lakes, whereas the overall influence on community photosynthesis and growth is subject to debate.
2. The influence of total dissolved inorganic carbon (DIC) and free CO2 on community photosynthesis was studied in six Danish lakes during the summer of 1995. The lakes were selected to ensure a wide range of chlorophyll a concentrations (1–120 μg l–1), pH (5.6–9.6) and DIC concentration (0.02–2.5 m m ). Photosynthesis experiments were performed using the 14C technique in CO2-manipulated water samples, either by changing the pH or by adding/removing CO2.
3. Lake waters were naturally CO2 supersaturated during most of the experimental period and inorganic carbon limitation of photosynthetic rates did not occur under ambient conditions. However, photosynthesis by phytoplankton in lakes with low and intermediate DIC concentrations was seriously restricted when CO2 concentrations declined. Similarly, photosynthesis was limited by low CO2 concentrations during phytoplankton blooms in the hardwater alkaline lakes.  相似文献

Methods for measuring the rates of photosynthesis and respiration in plants are reviewed. Closed systems that involve manometric techniques, 14CO2 fixation, O2 electrodes and other methods for measuring dissolved and gas phase O2 are described. These methods typically provide time-integrated rate measurements, and limitations to their use are discussed. Open gas exchange systems that use infra-red CO2 gas analysers and differential O2 analysers for measuring instantaneous rates of CO2 and O2 exchange are described. Important features of the analysers, design features of gas exchange systems, and sources of potential error are considered. The analysis of chlorophyll fluorescence parameters for estimating the quantum yield for O2 evolution and CO2 fixation is described in relation to new fluorescence imaging systems for large scale screening of photosynthetic phenotypes, and the microimaging of individual chloroplasts.  相似文献

1. A new open-top chamber for measuring CO2 efflux from the soil is reported here. The new design enables measurement of the equilibrium CO2 efflux, when there is no detectable pressure difference between the chamber and outside nor leakage of CO2 into or out of the chamber.
2. In previous dynamic-chamber techniques, the measured CO2 efflux is dependent on the pressure difference between the inside and outside of the chamber, and a negative pressure difference of –1Pa may cause an order of magnitude increase in measured CO2 efflux. Although the measured CO2 efflux is less sensitive to a positive pressure difference than to a negative one, a positive pressure difference of even a few tenths of a Pa will lead to a considerable underestimation in soil CO2 evolution.
3. The influence of pressure difference on measured CO2 efflux is negligible in the new design and the estimated CO2 efflux is close to the undisturbed soil respiration rate. Flow rates up to 8lmin–1, or air movement over the soil surface up to 55cmmin–1, will not affect CO2 evolution from the soil. The influence of pressure difference is related to the type of soil being measured and this has also been reported here for the new design.  相似文献

14CO2 evolution of prelabeled Scenedesmus obliquus Kütz, has been followed in the dark and in the light. In the light, no carbon dioxide is evolved. Addition of unlabeled NaHCO, leads to 14CO2 release attaining 20 to 30% of the dark rate. Double-reciprocal plots of NaHCO3 concentrations vs 14CO2 release results in a straight line, indicative of competition between exogenously supplied bicarbonate and endogenously evolved carbon dioxide. With this method, it is possible to measure CO2 evolved by respiration in the light and to show that true photoinhibition of respiration occurs in Scenedesmus . In the light. DCMU substantially increases 14CO2 evolution; in the presence of the uncoupler carbonyl cyanide- m -chlorophenylhydrazone. 14CO2 evolution is comparable to that in the dark. 14CO2 release and oxygen uptake in the dark are only slightly affected by cyanide, indicative of a cyanide-resistant respiration and/or fermentation as the essential CO2-yielding processes in the presence of cyanide. These results, compared with concurrent ATP levels, lead us to assume that energy charge is not the only factor responsible for photoinhibition of respiration.  相似文献

Abstract Denitrification was measured in intact sediment cores and in homogenised slurries using membrane inlet mass spectrometry. Dissolved concentrations of O2, N2, N2O and CO2 were simultaneously monitored. Using a 0.8 mm diameter needle probe, a comparison was made of the gas profiles of intact cores obtained under different conditions, i.e. with air or argon as the headspace gas and after the addition of nitrate and/or a carbon source to the sediment surface. O2 was detectable to a depth of 1 cm under a headspace of air and the depth at which the maxima of denitrification products occurred was 1.5–2 cm. Denitrification products (N2O, N2) occurred in the surface layers where O2 was above the minimum level of detectability (> 0.25 μM): diffusion of N2 and N2O upwards from the anoxic zone, local anaerobic microenvironments or aerobic denitrification are alternative explanations for this observation. The addition of nitrate and/or acetate increased the concentrations of N2, N2O and CO2 in the sediment core. In sediment slurries, the pH, nitrate concentration, carbon source and the depth from which the sample was taken affected the rate of denitrification. Nitrogen was the sole detectable end product. Maximum denitrification occurred at pH 7.5 and at 20 mM nitrate. Denitrification was at a maximum in those slurries prepared from sections of core at 1–2 cm depth.  相似文献

The effect of long-term exposure to different inorganic carbon, nutrient and light regimes on CAM activity and photosynthetic performance in the submerged aquatic plant, Littorella uniflora (L.) Aschers was investigated. The potential CAM activity of Littorella was highly plastic and was reduced upon exposure to low light intensities (43 μmol m−2 s−1), high CO2 concentrations (5.5 mM, pH 6.0) or low levels of inorganic nutrients, which caused a 25–80% decline in the potential maximum CAM activity relative to the activity in the control experiments (light: 450 μmol m−2 s−1; free CO2: 1.5 mM). The CAM activity was regulated more by light than by CO2, while nutrient levels only affected the activity to a minor extent. The minor effect of low nutrient regimes may be due to a general adaptation of isoetid species to low nutrient levels.
The photosynthetic capacity and CO2 affinity was unaffected or increased by exposure to low CO2, irrespective of nutrient levels. High CO2, low nutrient and low light, however, reduced the capacity by 22–40% and the CO2 affinity by 35-45%, relative to control.
The parallel effect of growth conditions on CAM activity and photosynthetic performance of Littorella suggest that light and dark carbon assimilation are interrelated and constitute an integrated part of the carbon assimilation physiology of the plant. The results are consistent with the hypothesis that CAM is a carbon-conserving mechanism in certain aquatic plants. The investment in the CAM enzyme system is beneficial to the plants during growth at high light and low CO2 conditions.  相似文献

Abstract A dense population of the purple sulfur bacterium Amoebobacter purpureus in the chemocline of meromictic Mahoney Lake (British Columbia, Canada) underwent consistent changes in biomass over a two year study period. The integrated amount of bacteriochlorophyll reached maxima in August and declined markedly during early fall. Bacteriochlorophyll was only weakly correlated with the light intensity and water temperature in the chemocline. In the summer, bacterial photosynthesis was limited by sulfide availability. During this period the intracellular sulfur concentration of A. purpureus cells decreased. A minimum concentration was measured at the top of the bacterial layer in August, when specific photosynthetic rates of A. purpureus indicated that only 14% of the cells were photosynthetically active. With the exception of a time period between August and September, the specific growth rates calculated from CO2 fixation rates of A. purpureus were similar to growth rates calculated from actual biomass changes in the bacterial layer. Between August and September 86% of the A. purpureus biomass disappeared from the chemocline and were deposited on the littoral sediment of Mahoney Lake or degraded within the mixolimnion. This rise of cells to the lake surface was not mediated by an increase in the specific gas vesicle content which remained constant between April and November. The upwelling phenomenon was related to the low sulfur content of A. purpureus cells and a low resistance of surface water layers against vertical mixing by wind.  相似文献

When D-[14C-U]-glucose was administered intraperitoneally into goldfish Carassius amatus at 20° C and 12L: 12D (dark period 1800–0600 hours) at 0600, 1200, 1800, 2400 and 0600 hours on the following day, glucose was metabolized to release 14CO2 and then it was incorporated into otoliths as carbonate. The rate of metabolic activity, judging from the ratio of inorganic to organic radiocarbon in plasma, was low during the dark period. Carbon incorporation into otoliths was also minimized during 1800–2400 h. When fish were exposed to ambient water containing NaH14CO3, plasma radioactivity was lowest during 1800–2400 hours, during which time carbon incorporation into otoliths was lowest. Plasma total CO2 levels markedly increased during the dark period. These results clearly indicate that carbonate formation in otoliths has a diel variation with a nadir lasting 6 h from 1800 to 2400 hours under the photoperiod used.  相似文献
