Twelve graft copolymers have been evaluated for their ability to prevent the adhesion of bacteria to substrata. The copolymers had polyethylene glycol (PEG) side-chains (‘teeth’) and a backbone that was either uncharged, acidic, basic or amphoteric. The copolymers were adsorbed onto glass, stainless steel and hydroxyapatite substrata, and 2-hpetri-dish adhesion experiments performed with bacteria isolated from marine (Pseudomonas sp. NCMB 2021), paper mill (S. marcescens NCIB 12211) and oral (S. mutans NCTC 10449) environments. The copolymers containing the most charged groups in the backbone had the most significant effect on bacterial adhesion levels, with anti-adhesive effects up to 99% achieved. An amphoteric copolymer (Compound 12) on glass, and acidic copolymer (Compound 11) on stainless steel and hydroxyapatite gave the most impressive anti-adhesive effects. These copolymers had non-specific bacterial anti-adhesive properties.It is proposed that the graft copolymers adsorbed onto hydrophilic surfaces via their charged backbone in such a way that the PEG side-chains were pointing out into the aqueous phase, and it was this orientation that was responsible for the observed anti-adhesive effect.  相似文献

Aims:  To investigate the effect of the biosurfactants surfactin and rhamnolipids on the adhesion of the food pathogens Listeria monocytogenes , Enterobacter sakazakii and Salmonella Enteritidis to stainless steel and polypropylene surfaces.
Methods and Results:  Quantification of bacterial adhesion was performed using the crystal violet staining technique. Preconditioning of surfaces with surfactin caused a reduction on the number of adhered cells of Ent. sakazakii and L. monocytogenes on stainless steel. The most significant result was obtained with L. monocytogenes where number of adhered cells was reduced by 102 CFU cm−2. On polypropylene, surfactin showed a significant decrease on the adhesion of all strains. The adsorption of surfactin on polystyrene also reduces the adhesion of L. monocytogenes and Salm. Enteritidis growing cells. For short contact periods using nongrowing cells or longer contact periods with growing cells, surfactin was able to delay bacterial adhesion.
Conclusions:  The prior adsorption of surfactin to solid surfaces contributes on reducing colonization of the pathogenic bacteria.
Significance and Impact of the Study:  This is the first work investigating the effect of surfactin on the adhesion of the food pathogens L. monocytogenes , Ent. sakazakii and Salm. Enteritidis to polypropylene and stainless steel surfaces.  相似文献

The attachment of three anaerobic microorganisms, Desulfomonile tiedjei, Syntrophomonas wolfei, and Desulfovibrio sp. strain G11, was investigated to determine if the presence of one species could influence the adhesion of another species to glass surfaces. The results indicated that the numbers and distribution of attached cells of one species could be influenced considerably by the presence of another species and the order in which the test species were exposed to the surface. D. tiedjei was found to detach readily from surfaces when it was not the primary colonizer. The attachment of Desulfovibrio G11 as the primary colonizer appeared to be stabilized by exposure to another test species. Under certain experimental conditions the test organisms formed close associations with each other on the surfaces. These findings demonstrate that the characteristics of anaerobic community biofilms can be determined by both the adhesion characteristics of the individual species and the interactions among those microorganisms.  相似文献

Block co-polymer surfactants have been used for cleaning hydrogel medical devices that contact the body (eg contact lenses) because of their biocompatibility. This work examined the relationship between concentration and detergency of two non-ionic polymeric surfactants (Pluronic F127 and Triton X-100) for cleaning protein soil, with anionic surfactants (sodium dodecyl sulfate and sodium laureth sulfate) as positive controls. Surface plasmon resonance was used to quantify removal of simulated tear soil from self-assembled monolayer surfaces, and a microplate format was used to study the removal of fluorescently labeled soil proteins from contact lenses. While detergency increased as a function of concentration for anionic surfactants, it decreased with concentration for the two polymeric surfactants. The fact that the protein detergency of some non-ionic polymeric surfactants did not increase with concentration above the critical micelle concentration could have implications for optimizing the tradeoff between detergency and biocompatibility.  相似文献

目的 研究2株乳酸菌(L1来自牙鲆肠道,L2为干酪乳杆菌)在牙鲆体表和消化道粘液的粘附及其对4种致病菌的粘附抑制情况。方法 异硫氰酸荧光素(FITC)标记法。结果 2种乳酸菌在牙鲆体表和消化道粘液中均有粘附性。在胃的粘附效果最好,粘附百分率分别是48.18%和63.0%,L2在牙鲆体表、盲囊和肠的粘附能力强于L1。L1对白色念珠菌(Candida albicans)在各部位均无粘附抑制作用,对荧光假单胞菌(P.Fluorescens)粘附抑制作用最强.使其在牙鲆消化道粘液的粘附百分率下降了19.01%,其余依次为非01型霍乱弧菌(V.cholerae non-Оl strains)、嗜水气单胞菌(Aeromonas hydrophila),分别降低了5.22%、3.22%;L2对非Оl型霍乱弧菌粘附抑制效果最强,使其粘附百分率降低了22.31%。使荧光假单胞菌、嗜水气单胞菌和白色念珠菌分别下降了17.50%、12.69%、5.15%。结论 对4种常见致病菌的粘附抑制在消化道粘液中好于体表。  相似文献

Human intestinal glycoproteins extracted from faeces were used as a model for intestinal mucus to investigate adhesion of pathogenic Escherichia coli and Salmonella strains, and the effect of probiotics on this adhesion. S-fimbriated E. coli expressed relatively high adhesion in the mucus model, but the other tested pathogens adhered less effectively. Probiotic strains Lactobacillus GG and L. rhamnosus LC-705 as well as a L. rhamnosus isolated from human faeces were able to slightly reduce S-fimbria-mediated adhesion. Adhesion of S. typhimurium was significantly inhibited by probiotic L. johnsonii LJ1 and L. casei Shirota. Lactobacillus GG and L. rhamnosus (human isolate) increased the adhesion of S. typhimurium suggesting that the pathogen interacts with the probiotic.  相似文献

Zoospores of Enteromorpha compressa (L.) Grev. secrete an adhesive cell coat which is involved in their attachment to various substrata. Two monoclonal antibodies (mAbs), designated Ent 1 and Ent 6, were raised against settled zoospores displaying secreted adhesive. Both antibodies labelled specifically the anterior region of the cell containing putative adhesive vesicles. During settlement the antigens recognised by both mAbs were secreted but whereas Ent 6 recognised a fibrillar material released within a few minutes of settlement, Ent 1 recognised components which were associated predominantly with the developing cell wall at later time points. Both mAbs also labelled a Golgi-rich region of settled spores, suggesting that these antigens are also synthesised after settlement. Both mAbs labelled the cell walls of vegetative tissue. Competitive enzyme-linked immunosorbent assay indicated that the two antibodies recognise separate, but overlapping epitopes. In spore settlement assays the Ent 6 immunoglobulin strongly reduced initial adhesion at low concentration whereas the inhibitory effects of Ent 1 occurred at later time points. On analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, (SDS-PAGE) both MAbs recognised a major buffer- and SDS-soluble, polydisperse 110-kDa antigen. The 110-kDa component was present in extracts of zoospores and sporulating tissue, but absent, in soluble form, from vegetative tissue. Deglycosylation of zoospore extract with anhydrous HF and peptide N-glycosidase digestion, showed that the major 110-kDa antigen is an N-linked glycan, and that the epitope is borne by the protein component. Time-course experiments showed that the Ent 6 antigen became progressively insoluble after zoospore attachment. Taken together, the data indicate that the two antibodies recognise separate but closely related antigens which have distinctive roles in adhesion and cell wall development. 目的 研究混合乳酸菌粘附及抑菌机制。方法 测定混合菌株粘附相关能力及抑菌方式,初步研究菌株粘附相关的表面活性成分。结果 混合菌株的表面疏水性、自聚合能力较高,对ICE-6细胞的粘附性亦较高。可抑制G+菌(金黄色葡萄球菌、枯草杆菌、艰难梭菌)和G‒菌(大肠埃希菌、沙门菌、假单胞菌)粘附ICE-6细胞,抑制方式效果为排除>竞争>替代;热处理对菌株粘附性影响明显高于胃蛋白酶、胰蛋白酶、LiCl和NaIO4等处理,表面蛋白(80~140 kDa)可使LiCl处理菌株的粘附率提升45%。结论 混合乳酸菌主要通过排除方式抑制外源菌的粘附,表面蛋白影响混合乳酸菌的粘附能力。  相似文献

Adhesion of probiotic microorganisms to the intestinal mucosa is considered important for many of the reported health effects. The influence of the endogenous microflora on the adhesion of four probiotic lactobacilli to immobilised intestinal mucus was investigated. It was observed that pre-treatment of the immobilised mucus with faecal extract slightly increased the adhesion of Lactobacillus GG. Pre-treatment of the immobilised mucus with faecal bacteria did not affect the adhesion of the tested strains. These results suggest that the normal microflora may not greatly affect the initial adhesion of the probiotic bacteria. This validates the results of earlier reports where the influence of the normal microflora was not taken into account.  相似文献

Pretreatments of wool fabrics with cationic, anionic or non-ionic surfactants were investigated to reduce surface tension and improve the wettability of the fibres in order to promote protease activity on the fibres in subsequent processes. Results showed that an ethoxylated alkyl phosphate (specific anionic surfactant) as well as the widely used non-ionic surfactant was compatible with proteases in the enzymatic treatment of wool. There is therefore a potential for using specific anionic surfactants to achieve efficient enzymatic scouring processes.  相似文献

The ability of Flexibacter BH3 to adhere to solid surfaces and to overcome the horizontal drag involved in gliding across the surfaces was considered in terms of the Stefan adhesion principle. The extracellular slime produced by Flexibacter BH3 was suitable as a Stefan adhesive because it exhibited viscous properties characteristic of a linear colloid, increasing the adhesiveness of the bacterium but allowing translational motion across the surface. The water-soluble slime was a glycoprotein, containing glucose, fucose, galactose and some uronic acid. Vesicles and tubules on the outer surface of Flexibacter BH3 possessed trilaminar membranes, contained 2-keto-3-deoxyoctonate (KDO), and showed identity with phenol-extracted lipopolysaccharide (LPS) in gel-diffusion tests.Sections of Flexibacter BH3 gliding on a gold film overlaying an agar medium reveraled a highly convuluted cell envelope outer membrane, portions of which closely conformed to the microcontours of the gold surface. Possible mechanisms of gliding are discussed in relation to this close association with solid surface features, to the finding that flexibility and spiral motion are not essential for gliding, and to evidence revealing the extrusion of slime in advance of pathfinder bacteria.Abbreviations used KDO 2-keto-3-deoxyoctonate - LPS lipopolysaccharide  相似文献

Bacterial initial adhesion to inert surfaces in aquatic environments is highly dependent on the surface properties of the substratum, which can be altered significantly by the formation of conditioning films. In this study, the impact of conditioning films formed with extracellular polymeric substances (EPS) on bacterial adhesion was investigated. Adhesion of wild type Pseudomonas aeruginosa PAO1 to slides coated with model EPS components (alginate, humic substances, and bovine serum albumin (BSA)) was examined. Surface roughness of conditioning film coated slides was evaluated by atomic force microscopy (AFM), and its effect on the bacterial initial adhesion was not significant. X-ray photoelectron spectroscopy (XPS) studies were performed to determine the elemental surface compositions of bacterial cells and substrates. Results showed that bacterial adhesion to bare slides and slides coated with alginate and humic substances increased as ionic strength increased. Conversely, BSA coating enhanced bacterial adhesion at low ionic strength but hindered adhesion at higher ionic strength. It was concluded that forces other than hydrophobic and electrostatic interactions were involved in controlling bacterial adhesion to BSA coated surfaces. A steric model for polymer brushes that considers the combined influence of steric effects and DLVO interaction forces was shown to adequately describe the observed bacterial adhesion behaviors.  相似文献

The effect of relative humidity (r.h.) (0%, 34% and 75%) and temperature (4, 15 and 25°C) on the desiccation survival of Pseudomonas sp., Acinetobacter calcoaceticus , a coryneform, Staphylococcus sp., and Staphylococcus aureus attached to and grown on glass surfaces was studied. The survival of Gram-negative and Gram-positive species was similar. Bacterial survival was shortest at 25°C for all species studied, and in most cases longest at 4°C. The effect of r.h. on survival varied with species. The number of microcolonies and the number of cells within microcolonies for Pseudomonas sp. increased on the dry surfaces after 7 d incubation under all desiccation conditions.  相似文献

 Laser scanning confocal microscopy (LSCM), light microscopy (LM) and epifluorescence microscopy (FM) were used to observe the extramatrical hyphae, mantle patterns and associated bacteria on mycorrhizal tips of Pinus strobus L. seedlings grown in pot cultures. Laccaria sp. and Tuber sp. formed ectomycorrhizas with Pinus strobus, while Phialophora finlandia Wang & Wilcox and E-strain (sensu Danielson 1982) formed ectendomycorrhizas. Distinct mantle patterns and cystidia were observed with greater resolution using LSCM, and intracellular hyphae were visualized in three dimensions. Trypan blue penetrated fresh whole mounts to 20 μm and was an excellent stain for visualizing fungal hyphae and bacteria with LSCM. Fluorescein isothiocyanate and acridine orange were used in conjunction with LSCM and FM to localize bacteria on ectomycorrhizal tips. With LSCM, bacteria were visible in the surface mucigel, and optical sectioning through the root tip showed that bacteria were also present within the mantle. LSCM is a non-intrusive and fast method for visualizing mycorrhizal structures and their associated bacteria on fresh, whole root tips. Accepted: 17 July 1996  相似文献

The marine Vibrio DW1 exhibited a positive response in heat output to a dialysis membrane surface in the presence of substrate (100 mM sodium glutamate) and, more particularly, in the absence of exogenous substrate (starvation conditions). The latter result paralleled the previously reported decrease in cell volume and increase in oxygen consumption by starving bacteria at a similar surface. Modified Morita's salts (MMS) did not extract nutrients from the dialysis membrane, but an artificial seawater containing tris buffer (ASW-tris) did extract surface active and nutrient materials from the membrane. The ASW-tris membrane extract and a commercial surfactant, Tween 85, were found to mimic the effects of the dialysis membrane surface by inducing a decrease in cell volume, and an increasing oxygen consumption and heat output of Vibrio DW1 even in the bulk liquid. Sulfate-reducing bacteria (SRB), which cause microbiologically influenced material corrosion under anoxic conditions, form one of the major groups of microorganisms responsible for the generation of hydrogen sulfide. In this study, which is aimed at reducing the presence of SRB, a novel alternative approach involving the addition of magnesium peroxide (MgO2) compounds involving the use of reagent-grade MgO2 and a commercial product (ORC™) was evaluated as a means of inhibiting SRB in laboratory batch columns. Different concentrations of MgO2 were added in the columns when black sulfide sediment had appeared in the columns. The experimental results showed that MgO2 is able to inhibit biogenic sulfide. The number of SRB, the sulfide concentration and the sulfate reducing rate (SRR) were decreased. ORC™ as an additive was able to decrease more effectively the concentration of sulfide in water and the SRB-control effect was maintained over a longer time period when ORC™ was used. The level of oxidation–reduction potential (ORP), which has a linear relationship to the sulfide/sulfate ratio, is a good indicator of SRB activity. As determined by fluorescence in-situ hybridization (FISH), most SRB growth was inhibited under increasing amounts of added MgO2. The concentration of sulfide reflected the abundance of the SRB. Utilization of organic matter greater than the theoretical SRB utilization rate indicated that facultative heterotrophs became dominant after MgO2 was added. The results of this study could supply the useful information for further study on evaluating the solution to biocorrosion problems in practical situations.  相似文献

We have examined as a function of temperature the effect of changes in the composition of the fatty acid chains of membrane phospholipids on the rate of cell to cell adhesion in the neuronal cell line B103. The rate of cell to cell adhesion in this cell line is highly temperature dependent but is not influenced by changes in the fatty acid composition of the plasma membrane generated by growing the cells either in the presence of oleic acid or elaidic acid. In contrast the temperature dependence of the rate of cell to cell adhesion, measured in a monolayer adhesion assay, is highly dependent on the shear force used during the assay. A two-step model of cell to cell adhesion involving multiple adhesion ligands is presented which can be used to explain these observations.  相似文献

Abstract

The solid surface tension of titanium was varied by using organosilane monolayers of various terminations, minimising differences in other material properties. Both the quantity of Pseudomonas fluorescens biofilms grown on the modified surfaces, and the percentage of biofilm remaining after exposure to hydrodynamic shear stress, varied significantly as a function of solid surface tension. The quantity of biofilm was less on chloropropyl-terminated surfaces than on an alkyl-terminated surfaces. However, the percentage of biofilm remaining after exposure to hydrodynamic shear stress (which depends on the adhesion and cohesion strengths of the biofilm) was less for the alkyl-terminated surface than for the chloropropyl-terminated surface, for one of the two sample sets analysed. These results demonstrate the importance of differentiating between the quantity of biofilm on a surface and the adhesion and cohesion strength of the biofilm, and may help explain discrepancies in the existing literature regarding the effect of solid surface tension on the propensity of a surface for microfouling.  相似文献
