应用超薄切片和免疫金标记电镜技术,结合体视学分析研究了受蚕豆萎焉病毒2(BBWV 2) 中国分离物B935侵染的豌豆(Pisum sativum)叶细胞中线粒体的异常变化。结果表明,感病细胞线粒体增生并聚集于细胞质的膜增生区周围,体积增大,形状畸变,一些线粒体内含有类结晶包涵体。病叶细胞与健康对照之间线粒体的体积密度(VV)、表面积密度(SV)、数密度(NV)等参数存在显著差异(P<0.01),而形状因子(PE)、周长指数(CI)、比表面积(RSV)等参数随不同病变阶段而有变化。在线粒体周围及线粒体之间的网格结构可被BBWV 2金标记抗体特异性标记.推断为正在组装的病毒粒子。子代病毒形成结晶体和管状体,有高密度的免疫金颗粒标记。上述研究结果提示BBWV 2 引起的细胞线粒体异常变化与病毒复制组装有关,聚集线粒体的外膜粘连面可能是病毒粒子组装部位,一些线粒体内的类结晶包涵体可能代表了某种蛋白质异常积累。  相似文献

电镜超微结构观察和免疫金标记显示:受蚕豆萎蔫病毒2号(Broad bean wilt virus 2,BBWV 2)中国分离物B935侵染的豌豆(Pisum sativum)和蚕豆(Vicia faba)叶细胞中膜结构增生,形成膜结构增生区,病毒以结晶体和管状体形式存在于细胞质中。在病变早期,叶肉细胞的胞间连丝处连接有小管结构,病毒样颗粒呈纵列排在小管中,穿越胞间连丝的小管能被BBWV 2的金标记抗体特异性标记。维管束组织的薄壁细胞、伴胞及转移细胞内存在膜增生区及病毒管状体,在筛管壁附近存在的病毒样颗粒能被BBWV 2金标记抗体特异性标记。实验结果表明BBWV 2胞间运动形式与豇豆花叶病毒(CPMV)相似,以完整粒子通过在胞间连丝处形成的小管结构穿越胞间连丝;细胞质中存在的直径160nm管状体只是一种病毒聚集体,与胞间运动无直接关系;该病毒在筛管中可能也是以完整粒子形式进行长距离转运的。  相似文献

SARS患者病理尸检肺组织样品分离病毒出现细胞病变的Hep2 培养细胞,按常规制作超薄切片,透射电镜下观察。电镜下,检出在感染细胞内复制、组装的呼肠孤病毒及其包涵体。病毒粒子衣壳立体对称、无包膜、直径在60~80nm。成熟病毒粒子核心致密常排列呈晶格状,不成熟病毒粒子核心空亮。数目不等的上述两种病毒粒子、长短不等的微管样结构和病毒浆常在核旁胞质内组成大小不等、无定形的病毒包涵体。此发现进一步提供了呼肠孤病毒感染有可能与SARS相关的形态学依据。  相似文献

在自然情况下, 番茄环纹斑点病毒(TZSV)和马铃薯Y病毒(PVY)通常复合侵染同一植株。该文首次报道了TZSV和PVY复合侵染烟草(Nicotiana sp.)植株的细胞病理特征, 并与单独侵染进行了比较分析。复合侵染的烟草植株细胞中, TZSV病毒颗粒明显增多, 并聚集于囊膜内形成包涵体, 与PVY的风轮状及片层状内含体和病毒颗粒聚集体交叉分布于细胞质(内含线粒体明显增多)中, 线粒体、叶绿体和细胞核结构较完整; 两种病毒的颗粒、包涵体和内含体数量均较单一侵染增多, 且对寄主亚细胞结构的破坏较单一侵染为轻, TZSV和PVY及其与寄主的互作可能存在协生作用。  相似文献

本文研究丁真菌传杆状病毒组(Furovirus)四个病毒成员,即甜菜坏死黄脉病毒(BNYVV)、甜菜土传病毒(BSBV)、花生丛生病毒印度株系(PCA-I)以及非洲株系(PCA-A)和小麦土传花叶病毒(SBWMV)所引起的细胞病理变化。这四种病毒在病毒粒子聚集的结构,是否引起细胞膜聚集及聚集的结构,过氧化物酶体泡状物的形成与否及结构均有差异。另外,同一种病毒的不同株系或分离物所引起的细胞病理变化也不相同。  相似文献

Nonoccluded, icosahedral picornavirus-like (PVL) particles, 23 nm in diameter, forming paracrystalline arrays were seen in the cytoplasm of various cells in Cotylogaster occidentalis. Viral inclusions were visible in live specimens and in sections prepared for light and electron microscopy. All worms examined over a 2-year period were found to be infected. Infections were naturally acquired and susceptibility was not associated with any particular developmental stages. Development of viral inclusions involved an increase in the inclusion volume, progressive accumulation and condensation of materials into the interior of the inclusions, and formation of multilamellar membrane networks. Virus particles were observed in the stroma of the inclusions in association with multilamellar spherical bodies. Mature PVL particles aggregated into polygonally shaped paracrystalline arrays. When such arrays occurred in the surface tegument, local disruption of the tegumentary membrane may liberate these particles into the environment. PVL particle production did not exhaust glycogen content of infected cells and did not appear to affect short-term survival of the parasite outside the molluscan host.  相似文献

通过对犬传染性肝炎病毒(ICHV)在犬肾传代细胞内形态发生及其抗原定位的电镜和免疫胶体金电镜研究,发现ICHV除了在宿主细胞核内发生外,还有一条细胞质内的发生途径。在细胞质内病毒核壳体的装配是以均质致密包涵体和副晶格包涵体为“基地”,这与人们熟知的细胞核内形态发生方式相似。免疫胶体金标记显示,细胞质包涵体中含有大量的ICHV抗原成分,显核壳体在细胞质内装配病毒的结构蛋白来源。此外,在感染的细胞质内还观察到与核内相同的病毒核心样结构。  相似文献

Using light and electron microscopy, as well as stereological analysis, a study was made of structural changes in mitochondria of cultured L-cells acted upon with 1 mM cyanide. After 23 hours of incubation, in cells cultured in the presence of cyanide the volume density of mitochondria, the volume of the average mitochondrion and the surface area of its membranes increased nearly twice as much. Concurrently, the number of mitochondria decrease also practically two-fold without any reliable changes in the surface area of membranes per unit of mitochondrion volume. A shorter (7 hours) incubation of L-cells in the presence of cyanide results in a decreased volume density of mitochondria in the cytoplasm, decreased volume of the average mitochondrion, without any significant changes in other above mentioned parameters. It is supposed that the changes in mitochondria under a prolonged cyanide treatment of cells are conditioned by the fusion of mitochondria as well as by a compensatory increase in the total surface of mitochondrial membranes.  相似文献

SYNOPSIS Intramitochondrial inclusions of a paracrystalline nature were observed in the peritrichous ciliate Carchesium polypinum This ciliate was found growing in the effluent of a sewage treatment plant. When paracrystalline inclusions were present, there was one per mitochondrion. A variety of profiles was encountered, all presumably of the same structure. Cross-sections revealed packed, spherical masses of 100-150 A diameter with dense walls 30-40 A thick and a variety of core densities. In more longitudinal sections the paracrystalline array appeared as long. electron-dense, finely filamentous elements oriented in parallel arrays and regularly spaced with a diameter of 60-100 A. Mitochondria tended to cluster, and between closely apposed mitochondria a dense, amorphous material was present. Numerous bud-like processes projected from mitochondria. Structural evidence, particularly the arrangement and core densities of these arrays, suggests a filamentous type of viral infection.  相似文献

In adult regenerating cardiomyocytes in culture, in contrast to fetal cells, mitochondrial creatine kinase (Mi-CK) was expressed. In the same cell, two populations of mitochondria, differing in shape, in distribution within the cell and in content of Mi-CK, could be distinguished. Immunofluorescence studies using antibodies against Mi-CK revealed a characteristic staining pattern for the two types of mitochondria: giant, mostly cylindrically shaped, and, as shown by confocal laser light microscopy, randomly distributed mitochondria exhibited a strong signal for Mi-CK, whereas small, "normal" mitochondria, localized in rows between myofibrils, gave a much weaker signal. Transmission EM of the giant mitochondria demonstrated paracrystalline inclusions located between cristae membranes. Immunogold labeling with anti-Mi-CK antibodies revealed a specific decoration of these inclusions for Mi-CK. Addition of 20 mM creatine, the substrate of Mi-CK, to the essentially creatine-free culture medium caused the disappearance of the giant cylindrically shaped mitochondria as well as of the paracrystalline inclusions, accompanied by an increase of the intracellular level of total creatine. Replacement of creatine in the medium by the creatine analogue and competitor beta-guanidinopropionic acid caused the reappearance of the enlarged mitochondria. It is believed that the accumulation of Mi-CK within the paracrystalline inclusions, similar to those observed in certain myopathies, represents a compensatory effect of the cardiomyocytes to cope with a metabolic stress situation caused by low intracellular total creatine levels.  相似文献

Electron microscopy and immunolabelling with antiserum specific to cucumber mosaic virus coat protein were used to examine tobacco leaf cells infected by cucumber mosaic virus isolated from Catharanthus roseus (CMV-Cr). Crystalline and amorphous inclusions in the vacuoles were the most obvious cytological modifications seen. Immunogold labelling indicated that the crystalline inclusion was made up of virus particles and amorphous inclusions contained coat protein. Rows of CMV-Cr particles were found between membranes of dictyosomes, but membranous bodies and tonoplast-associated vesicles were not evident. Virus particles and/or free coat protein were easily detected in the cytoplasm by immunolabelling. No gold labelling was found within nuclei, chloroplasts and mitochondria.  相似文献

Summary. Electronmicroscopic immunogold cytochemistry was used to investigate the cellular and subcellular distribution of glutathione in root and leaf cells of Styrian pumpkin (Cucurbita pepo L. subsp. pepo var. styriaca Greb.) plants. Gold particles bound to glutathione were found in various cell structures. Statistical evaluation of the gold particle density was made for different cell compartments including nuclei, mitochondria, plastids, peroxisomes, and the cytosol. In each cell type the highest level of glutathione immunoreactivity occurred in mitochondria, for which the labeling density was found to be higher in mesophyll cells of the youngest fully developed leaves (younger leaves) than in the 5th leaves (older leaves) or in root tip cells. Additionally, a statistically significant increase of gold particles bound to glutathione was observed in nuclei (22%) and the cytosol (14%) of the root cells in comparison with mesophyll cells of older (17% and 9%, respectively) and younger leaves (11% and 6%, respectively). The relevance and specificity of glutathione labeling is discussed with respect to difficulties of immunolocalization of low-molecular-weight compounds.  相似文献

Summary Growing rats (4 weeks old) were kept for 3 weeks at 11° C and 24° C respectively. The cold-adapted animals showed a significantly higher oxygen consumption (64%). Volume density of subsarcolemmal and interfibrillar mitochondria as well as volume density of fat droplets were estimated in M. soleus and the diaphragm of both groups. In cold-adapted animals, the total volume of mitochondria was significantly increased by 24% in diaphragm and 37% in M. soleus. The volume of subsarcolemmal mitochondria was almost doubled in each muscle, but the volume of interfibrillar mitochondria did not change significantly. The surface of the inner mitochondrial membranes per unit volume of mitochondrion in M. soleus was significantly increased both in interfibrillar and subsarcolemmal mitochondria, whereas the surface of the outer mitochondrial membranes per unit volume of mitochondrion was increased only in the subsarcolemmal mitochondria. The volume of fat droplets in the diaphragm and M. soleus of cold adapted animals increased significantly by 62% and 150% respectively.  相似文献
