In autotetraploids, chromosome pairing may be in the form of quadrivalents or bivalent pairs. Whether or not the quadrivalents are maintained until first meiotic metaphase depends on the formation of chiasmata. The relative frequencies of M I configurations thus contain information both on pairing and on chiasma formation. With distal chiasma localisation six configurations can be recognised and their relative frequencies determined: ring quadrivalents, chain quadrivalents, trivalents (with univalent), ring bivalents, open (rod) bivalents, univalent pairs. These represent five degrees of freedom permitting five parameters to be estimated: the frequency (f) of quadrivalent pairing; the frequencies of chiasmate association of the two ends (arms in metacentrics), a′, b′, after quadrivalent pairing, and a, b after bivalent pairing. — The appropriate formulae have been derived and applied to observations on Tradescantia virginiana (4n=24) which has pronounced distal chiasma localisation. Slight modifications make the model applicable to autotetraploids with interstitial in addition to distal chiasmata. In T. virginiana, chromosome pairing appeared to be random between homologues (65.8% quadrivalent pairing; 55.4% observed at M I). After quadrivalent pairing chiasmate association is frequent in the “average long” arm (95.0%) and much less so in the other arm (60.5%). This is attributed to partner exchange. After bivalent pairing chiasma frequencies are still different for the two arms (93.8% and 83.5% association respectively) but much less pronounced. Various complications are discussed.  相似文献

Chiasmata constitute one of the cornerstones of sexual reproduction in most eukaryotes. They mediate the reciprocal genetic exchange between homologues and are essential to the proper orientation of the homologous centromeres in meiosis I. As markers of recombination, they offer a cytological means of mapping. Rather than trying to accurately count individual chiasmata, we have examined properties of the mathematical relationship between frequencies of nonadorned disomic configurations in meiosis (ring, rods, and univalents) and the probabilities at which arms of the respective chromosomes are chiasmate (one or more chiasma per arm). Numerical analyses indicated that conventionally analyzed bivalents with nonidentified arms yield statistically biased estimates of chiasma probabilities under a broad range of circumstances. We subsequently analyzed estimators derived from adorned configurations with ISH-marked arms, which were found to be statistically far superior, and with no assumptions concerning interference across the centromere. We applied this methodology in the study of chromosomes 16 and 23 of cotton (Gossypium hirsutum), and estimated their arm lengths in centimorgans. The results for chromosome 23, the only one of the two chromosomes with a documented RFLP map, were consistent with the literature. Similar molecular-meiotic configuration analyses can be used for a wide variety of eukaryotic organisms and purposes: for example, providing far more powerful meiotic comparisons of genomes of chromosomes, and a rapid means of evaluating effects on recombination. The hypothesis of a connection between rod bivalent formation and incomplete meiotic association at NORs of SAT-chromosomes of H. marinum is supported. PMCs of H. marinum ssp. gussoneanum (2x), two diploid ssp. marinum × ssp. gussoneanum (2x) hybrids and two ssp. gussoneanum (4x) × Secale cereale hybrids at metaphase I (M-I) were analyzed by in situ hybridization. The probe pTa71 labelled rDNA sites at NORs of a single pair of homologous or near-homologous SAT-chromosomes of H. marinum in each material. In the three diploids, M-I was regular with ring bivalents and one or a few rods (av.13.52 bound arms cell(-1) ). More rod bivalents than the expected one out of seven, i.e. 30, 67 and 89% included rDNA-carrying chromosomes. Corresponding bound short arm frequencies were 0.89, 0.72 and 0.52, while long arms and arms of other chromosomes presented complete or near- complete association. The two heterogenomic hybrids had a less regular M-I (av. 8.04 bound arms cell(-1) ) including 20% rDNA-carrying rods with bound arm frequencies of 0.29 in short and 0.87 in long arms. Positions of chromosome associations were established in all 150 rDNA-carrying bivalents. In 77 bivalents with short arm associations, 4% of these occurred proximally to, none at, and 96% distally to rDNA sites, i.e. in satellites. In 143 bivalents with long arm associations, 83% occurred at interstitial and 17% at terminal positions. The observations combine increased frequency of rDNA-carrying rods with decreased frequency of association at NOR regions of SAT-chromosomes. The basis for the relationship is discussed.  相似文献

Information concerning natural variation either in chiasma frequency or in the genetic basis of any such variation is a valuable tool to characterize phenotypic traits and their genetic control. Here meiotic recombination frequencies are analysed in nine geographically and ecologically diverse accessions of Arabidopsis thaliana, and a comparative study was carried out incorporating previous data from another eight accessions. Chiasma frequencies, estimated by counting rod and ring bivalents at metaphase I, varied up to 22% among accessions. However, no differences were found among plants of the same accession. There was a relationship, which does not necessarily imply direct proportionality, between the size of the chromosomes and their mean chiasma frequency. Chiasma frequency and distribution between arms and among chromosomes were not consistent over accessions. These findings indicate the existence of genetic factors controlling meiotic recombination both throughout the whole genome and at the whole chromosome level. The reliability of chiasma scoring as an indicator of reciprocal recombination events is also discussed.  相似文献

Eight different sized supernumerary segments located at distal ends of the long arms of chromosomes M4, M5, M6, and S8 of the grasshopper Stenobothrus festivus were studied in males with regard to the synaptic process and chiasma distribution in the bivalents that carry them. The M4, M5, and M6 bivalents heterozygous for extra segments were always monochiasmate, in contrast to their bichiasmate condition observed in basic homozygotes. Furthermore, the presence of any of these extra segments led to chiasma redistribution in the carrier bivalents, so that such chiasmata were formed preferentially further away from the extra segment. The intensity of this effect is dependent on the size of the segment. Not all heteromorphic bivalents exhibited synaptonemal complexes with equalized axes at pachytene, but there was always a variable proportion of heterosynapsis around the distal ends of the long arms that was dependent on both the size of the segment and the size of the carrier chromosome. It is proposed that the absence of chiasmata in nonhomologous synapsed regions is responsible for the results obtained. Length measurements of the different extra segments and their carrier chromosomes between pachytene and diplotene indicated that synaptonemal complex is underrepresented in supernumerary heterochromatin.  相似文献

Summary A translocation heterozygote in tomato (Lycopersicon esculentum) is shown to have a cyclical type of interchange between the long arms of chromosomes 1, 2 (nucleolar) and 3. A study of chromosome association in this plant at metaphase I has indicated that in 21% of the cells a ring of six chromosomes is present. Since an open ring hexavalent can occur only if there is chiasma formation in all the translocated segments and in all the short arms of the three chromosomes, it is concluded that there is considerable frequency of chiasma formation in the short arm of the nucleolar chromosome. This conclusion contradicts the previous observations that chiasma formation is either absent or very rare in the entirely dark staining chromatic, sometimes referred to as heterochromatic, short arm of the nucleolar chromosome.Part of this investigation was carried out at the Department of Genetics, Agricultural University, Wageningen, when the author was serving a contract between the EURATOM-I.T.A.L. and the Agricultural University.  相似文献

Chiasma frequency,distribution and interference maps of mouse autosomes   总被引:11,自引:0,他引:11  
Chiasma frequencies were analysed and chiasma positions measured in diakinesis/metaphase I autosomal bivalents from oocytes and spermatocytes of F1 hybrid C3H/HeH×101/H mice. Twenty chromosome size ranks, including the presumptive X bivalent, could be distinguished in oocytes, and nineteen autosomal ranks plus the XY pair spermatocytes. Overall, mean cell chiasma frequencies of the two sexes did not differ significantly once the contribution of the presumptive X bivalent and the XY pair were taken into account. Sex related differences in chiasma distribution patterns were evident, however. In monochiasmate bivalents, the chiasma was most commonly located interstitially in oocytes while in spermatocytes it could be either interstitial or distal. In dichiasmate bivalents, the chiasmata tended to be more centrally located in oocytes than in spermatocytes. Minimum inter-chiasma distances did not appear to show any great variation in chromosome pairs of different sizes, however, mean inter-chiasma distances did increase with the bivalent length. The minimum-inter chiasma distance data suggest that chiasma interference is complete over a chromosomal segment equating to approximately 60 Mb. Measurement of the positions of chiasmata along chromosome arms open up the possibility of producing chiasma-based genetic maps for all the autosomes of the mouse.  相似文献

Summary Several studies have indicated a noncorrespondence between genetic and physical distances in wheat chromosomes. To study the physical distribution of recombination, polymorphism for C-banding patterns was used to monitor recombination in 67 segments in 11 B-genome chromosome arms of Triticum turgidum. Recombination was absent in proximal regions of all chromosome arms; its frequency increased exponentially with distance from the centromere. A significant difference was observed between the distribution of recombination in physically short and physically long arms. In physically short arms, recombination was almost exclusively concentrated in distal segments and only those regions were represented in their genetic maps. In physically long arms, while a majority of the genetic distance was again based upon recombination in distal chromosome segments, some interstitial recombination was observed. Consequently, these regions also contributed to the genetic maps. Such a pattern of recombination, skewed toward terminal segments of chromosomes, is probably a result of telomeric pairing initiation and strong positive chiasma interference. Interference averaged 0.81 in 35 pairs of adjacent segments and 0.57 across the entire recombining portions of chromosome arms. The total genetic map lengths of the arms corresponded closely to those expected on the basis of their metaphase-I chiasma frequencies. As a consequence of this uneven distribution of recombination there can be a 153-fold difference (or more) in the number of DNA base pairs per unit (centiMorgan) of genetic length.  相似文献

Using a Giemsa C-banding procedure it has been possible to identify at meiosis three chromosome pairs of a local Spanish rye cultivar. Two of these chromosomes (3 and 5) were heterozygous for an interstitial C-band in the long arm and the other (chromosome 7) was heterozygous for a telomeric C-band, also in the long arm. From the frequency of being bound at metaphase I and the frequency of recombined chromatids at anaphase I in the arms considered, estimates of actual chiasma frequencies have been derived. The results have been compared with those obtained in a Fl between two inbred lines. It is concluded that: (i) Although the frequency of bound arms analyzed was similar in all cases, the chiasma frequency was higher in the cultivar than in the Fl plants. Cultivar plants showed a variation in chiasma frequency for the bivalent arms studied which was correlated with the frequency of bound arms per cell, indicating that the estimation for chiasma frequency by means of bound arm frequency has an error that increases with increasing number of bound arms per cell, (ii) Evidence of chiasma terminalization has not been found, (iii) It is suggested that the different rye chromosomes have different chiasma localization patterns, which, in turn, are related with the chiasma frequency.  相似文献

This work describes the first report about the occurrence of recombination nodules (RNs) in spread pachytene cells of two species of Coleoptera: Palembus dermestoides (Tenebrionidae) and Epicauta atomaria (Meloidae). The RNs were observed in preparations contrasted with phosphotungstic acid. Considering RN morphology and its occurrence in pachytene bivalents (one per autosome bivalent) these structures were interpreted to be late RNs. P. dermestoides and E. atomaria have 2n = 20 chromosomes including an Xy(p) sex determination system. In spite of most frequently subtelocentric morphology observed in the autosomes of both species, the occurrence of RNs is limited only to the synaptonemal complex (SC) structure of the long arms. These findings are in agreement with those obtained using light microscopy analysis in which only one chiasma or terminalization event is observed per autosomal bivalent in early or late metaphase I cells. The RNs have the same average width of the SC of each analyzed species, a circular shape, strong electron density, and are observed mainly between the lateral elements of the SC. The RNs of P. dermestoides and E. atomaria have approximately the same average size (width), 180 +/- 20 nm and 160 +/- 80 nm, respectively. The absence of RNs in the short arms and its occurrence in the long arms are discussed considering the short arm pericentromeric and pro-centric heterochromatin.  相似文献

C-banding and silver staining of the somatic and germ cells of the male domestic dog. Canis familiaris, have shown that: (1) the amount of C-banding is small compared to most other mammalian species, (2) three pairs of autosomes have nucleolus organizer regions (NORs) at the terminal ends of their long arms, whereas the Y chromosome has an NOR on the terminal end of the short arm, (3) the organization of the synaptonemal complex (SC) is similar to that of other mammalian species, (4) a distinct SC is formed between the long arm of the Y chromosome and probably the short arm of the X chromosome, and (5) the differential axes of both sex chromosomes do not demonstrate fusiform thickenings nor do they stain darkly with silver as do the XY bivalents in many other mammalian species.  相似文献

Summary The two arms of a bivalent can be bound (contain at least one chiasma) or not-bound (contain no chiasma). Formulae are presented to test the randomness of distribution of the bound arms over the chromosome complement, under the assumption that the probability of being bound is related to the arm length.Testing meiotic configurations in Secale cereale (rye), Crotalaria intermedia and several species of Coffea, non-randomness was found in all cases, resulting in an excess of bivalents with only one arm bound. The effect is slightly different in the different plants. The basis is considered to be localization of chiasmata.In rye plants, homozygous for a rather large translocation, an excess of univalents was observed to more than compensate the localization effect. This is attributed to the limited probability of the newly formed short arm of the small translocation chromosome to be bound.The section on rye was started at the Laboratory of Genetics, director Prof. Dr. R. The complete process of meiosis was investigated in Pinus wallichiana, P. strobus and their artificial hybrid (F1) using microsporocytes. It is revealed that there were slightly lower chiasma frequency, lower ring bivalent frequency, lower meiotic index and distinctly higher frequency of aberrance (chromosomal bridges, fragments or micronuclei) in pollen mother cells (PMCs) of the hybrid (F1) than those of the parental species, which showed a certain degree of differentiation between homologous chromosomes of the two parents. However, relatively higher frequency of ring bivalents and higher meiotic index in all the three entities indicate the great stability of genomes of parental species, and the differentiation of genomes between the two parents must have been slight. Total nineteen signal loci of 18S rDNA were observed in nine bivalents of the hybrid (F1), among which one bivalent bears two loci, while the others have only one. It is suggested that distinct differentiation at genetic level existed in homologous chromosomes of the two parental species, whereas only slight differentiation at karyotypic and genomic levels take place between the parent species.  相似文献

Summary Chromosome pairing and chiasma frequency were studied in bread wheat euhaploids (2n = 3x = 21; ABD genomes) with and without the major pairing regulatorPh1. This constitutes the first report of chromosome pairing relationships among the A, B, and D genomes of wheat without the influence of an alien genome. AllPh1 euhaploids had very little pairing, with 0.62–1.05 rod bivalents per cell; ring bivalents were virtually absent and mean arm-binding frequency (c) values ranged from 0.050 to 0.086. In contrast, theph1b euhaploids had extensive homoeologous pairing, with chiasma frequency 7.5–11.6 times higher than that in thePh1 euhaploids. They had 0.53–1.16 trivalents, 1.53–1.74 ring bivalents, and 2.90–3.57 rod bivalents, withc from 0.580 to 0.629. N-banding of meiotic chromosomes showed strongly preferential pairing between chromosomes of the A and D genomes; 80% of the pairing was between these genomes, especially in the presence of theph1b allele. The application of mathematical models to unmarked chromosomes also supported a 21 genomic structure of theph1b euhaploids. Numerical modeling suggested that about 80% of the metaphase I association was between the two most related genomes in the presence ofph1b, but that pairing under Ph1 was considerably more random. The data demonstrate that the A and D genomes are much more closely related to each other than either is to B. Dichroplus pratensis has a complex system of Robertsonian rearrangements with central-marginal distribution; marginal populations are standard telocentric. Standard bivalents show a proximal-distal chiasma pattern in both sexes. In Robertsonian individuals a redistribution of chiasmata occurs: proximal chiasmata are suppressed in fusion trivalents and bivalents which usually display a single distal chiasma per chromosome arm. In this paper we studied the synaptic patterns of homologous chromosomes at prophase I of different Robertsonian status in order to find a mechanistic explanation for the observed phenomenon of redistribution of chiasmata. Synaptonemal complexes of males with different karyotypes were analysed by transmission electron microscopy in surface-spread preparations. The study of zygotene and early pachytene nuclei revealed that in the former, pericentromeric regions are the last to synapse in Robertsonian trivalents and bivalents and normally remain asynaptic at pachytene in the case of trivalents, but complete pairing in bivalents. Telocentric (standard) bivalents usually show complete synapsis at pachytene, but different degrees of interstitial asynapsis during zygotene, suggesting that synapsis starts in opposite (centromeric and distal) ends. The sequential nature of synapsis in the three types of configuration is directly related to their patterns of chiasma localisation at diplotene-metaphase I, and strongly supports our previous idea that Rb fusions instantly produce a redistribution of chiasmata towards chromosome ends by reducing the early pairing regions (which pair first, remain paired longer and thus would have a higher probability of forming chiasmata) from four to two (independently of the heterozygous or homozygous status of the fusion). Pericentromeric regions would pair the last, thus chiasma formation is strongly reduced in these areas contrary to what occurs in telocentric bivalents.  相似文献

矮牡丹(Paeonia suffruticosa subsp. spontanea)(永济居群)存在多种结构杂合现象,减数分裂存在一些异常:如单价体、异形二价体、互锁四价体、六价体、后期I倒位桥、落后单价体、不均等分离、后期Ⅱ桥和微核等。统计了这些异常现象出现的频率,并对其形成的机制和对正常小孢子形成的影响进行了讨论。从细胞学水平上探讨了矮牡丹可能的濒危机制。同时结合前人的研究,对芍药属内3个组的结构杂合程度进行了比较。  相似文献

A cytological study was carried out, using male Dictyocaulus filaria, that revealed the diploid number of chromosomes was 2n = 11 and the sex determining mechanism was XO. The behaviour of the chromosomes in the different stages of meiosis was also investigated. Cross, open ring and rod bivalents were observed in diakinesis. The chromosomes appeared to be acrocentric since they acquired a radial disposition in Metaphase-II. The chiasma frequency was 1 and the nucleolus-organizing region was located at the ends of the chromosomes.  相似文献

以小麦(Triticum aestivum L.)双端体3DL为细胞学标记,用具有phKL基因的小麦地方品种"开县罗汉麦"为受体连续回交,将促进小麦外源部分同源染色体配对的phKL基因和Ph2基因缺失重组在一起获得了重组体phKL+Ph2.这种重组体有正常的育性.与只有phKL基因的小麦材料相比,重组体与外源物种AegilopsvariabilisEig.或黑麦(Secale cereale L.)杂种的部分同源染色体配对水平显著增加,表明Ph2基因的缺失体与phKL基因可能存在加性效应.部分同源染色体配对水平的增加表现在棒状二价体、环状二价体和三价体的数量变多而单价体数量减少.单价体的减少主要是由于棒状二价体的增加所造成的.在小麦外源遗传转移中,运用重组体pHKL +Ph2可能比单纯应用Ph2缺失或phKL基因材料更理想.当与具有ph1b基因的材料比较时发现,重组体phKL+Ph2与 Ae.variabilis(或黑麦)杂种的部分同源染色体配对水平显著降低,这主要是由环状二价体和多价体的减少造成的,但是棒状二价体数量表现增加(与Ae.variabilis杂种)或达到类似水平(与黑麦杂种),这一有趣的发现从表现型上证明了Ph1基因与Ph2或phKL基因在诱导部分同源染色体配对时的遗传作用机制存在差异.  相似文献
