Relatively large amounts of cyclic AMP are produced by the prothoracic glands (source of the insect moulting hormone or moulting hormone percursor) of the tobacco hornworm, Manduca sexta. Pharate pupal glands produce more cyclic AMP than early fifth instar larval glands, and the addition of aminophylline enhances cyclic AMP accumulation. The much lower cyclic AMP level in the absence of aminophylline indicates the presence of potent cyclic AMP phosphodiesterase activity. Brains (sources of the prothoracicotropic hormone) also produce cyclic AMP but at a lower rate. Brains efficiently produce adenosine from ATP while β-ecdysone inhibits adenosine formation in early fifth instar larval brains. β-Ecdysone stimulates adenyl cyclase in brains of both stages when aminophylline and fluoride are present but has no effect on cyclic AMP accumulation in prothoracic glands. The absence of fluoride greatly reduces the amount of cyclic AMP produced by prothoracic glands when aminophylline is present. No cyclic AMP is accumulated in prothoracic glands when both fluoride and aminophylline are absent or in brains when fluoride is absent, notwithstanding the presence of aminophylline. Other insect tissues were also analysed for cyclic AMP production and none showed levels nearly as high as the prothoracic glands, suggesting a close relationship between cyclic AMP production and the function of the gland.  相似文献

【目的】上颚腺(mandibular gland)是昆虫重要的外分泌腺,它产生的化学物质在昆虫的种内信息交流中起重要的作用。本研究目的在于了解中华蜜蜂Apis cerana cerana上颚腺的组织结构以及胚后发育特点。【方法】本研究通过组织形态学、BrdU免疫组织化学等技术,对中华蜜蜂上颚腺的结构和发育过程进行了比较研究。【结果】中华蜜蜂的上颚腺在不同级型间差异显著,蜂王的面积最大,工蜂较小,而雄蜂退化。上颚腺出现在末龄幼虫到预蛹阶段,细胞分裂活动的高峰期发生在蛹发育的第1天,随后分裂细胞数减少,并一直持续到蛹发育的第6天结束。在上颚腺发育早期,由分泌细胞分化的内膜就已经出现,并一直保持到成虫。【结论】本研究为昆虫上颚腺的发育和功能研究提供了理论基础。  相似文献

Breakdown of the moulting glands in Oncopeltus can be completely inhibited by topical application of a juvenile hormone analogue prior to day 2 of the fifth instar, and partially inhibited by application prior to day 4. The analogue-sensitive period for the inhibition of external metamorphosis is very similar to that for the inhibition of cell death in the moulting glands. A decline in response to the analogue between days 2 and 4 is correlated with rising ecdysteroid levels in the haemolymph. That this rise in ecdysteroids may be responsible for termination of the juvenile hormone-sensitive period is suggested by premature loss of sensitivity to the analogue by the moulting glands following premature exposure to 20-hydroxyecdysone.  相似文献

The degeneration of the prothoracic glands of the male cricket, Gryllus bimaculatus, was analyzed by using an in vitro assay for ecdysteroid release from the moulting glands in last instar nymphs as well as in adult animals, and correlated with light and transmission electron microscopy. Apoptosis was examined by the TUNEL-reaction. The ability to synthesize ecdysteroids reached a peak at the 8th day of the last larval instar, identified as the moulting peak. After adult ecdysis it decreased to barely measurable values. Prothoracic gland degeneration was initiated at the time of the moulting peak, characterized by TUNEL positive reactions, nuclear and cytoplasmatic condensation, a striking abundance of residual basal laminae; besides a great amount of autophagic vacuoles are observed. The results reveal that apoptosis and autophagy are the basic mechanisms for programmed cell death in the prothoracic gland of Gryllus bimaculatus.  相似文献

The prothoracotropic action of ecdysone analogues was examined, using the brainless, diapausing pupae of Bombyx mori with or without inclusion of prothoracic glands. The most effective of those hormones to stimulate prothoracic glands to secrete the moulting hormone was found to be cyasterone. The other analogues such as ponasterone A, ecdysterone, and inokosterone showed a lower activity with regard to prothoracotropic action. The female prothoracic glands were found to be more sensitive to the ecdysones than the male ones. The time lag from hormone injection to emergence indicated the dual actions of the injected ecdysones, directly on the target organs and indirectly on the prothoracic glands subsequent to secreting the moulting hormone.  相似文献

Summary The secretion of the mandibular glands of a honey bee queen enables the worker bees to react to the presence of their queen. Extirpating the mandibular glands of the queen does not prevent that she is accepted by her colony. Hitherto this was attributed to contamination of the queen's body by mandibular gland substances during or preceding the extirpation. When, however, these glands are extirpated before they have secreted any material and the queens are inseminated artificially, the colonies still accept these queens. A normal-sized retinue, the absence of emergency cell building and the absence of activation of the worker's ovaries indicate that such a queen is still able to maintain her social position. This supports Verheijen-Voogd's (1959) conclusion that the queen's influence on her workers has a behavioural basis (chemoreception) rather than a biochemical one.Laboratory experiments reveal that apart from the mandibular gland substances other queen pheromones are produced in glands on the abdomen, most probably in the glands described by Renner and Baumann (1964).  相似文献

Abstract. Attack by the ectoparasitoid Eulophus pennicornis Nees (Hymenoptera: Eulophidae) prevents larvae of Lacanobia oleracea L. (Lepidoptera: Noctuidae) from moulting. Prothoracic glands (PGs) excised from parasitized or artificially envenomated hosts show a reduced basal level of ecdysteroid release at a time when non-parasitized caterpillars produce an ecdysteroid surge (48 h post moult to 5th stadium = penultimate stadium in non-venomated hosts). By contrast, PGs from similarly parasitized or envenomated caterpillars release comparatively high levels of ecdysteroid at 120 h post-moult. Temporary inactivation of PGs cannot be attributed solely to a parasitoid-induced reduction in cell viability, and incubation in E. pennicornis venom in vitro does not exert any direct effect on either PG cell viability or ecdysteroid release. However, inactivated PGs are not stimulated by forskolin, which may indicate that the absence of the required pre-moult ecdysteroid surge in developmentally arrested L. oleracea is due to insensitivity to a prothoracicotropic hormone. Even though parasitized caterpillars never moult, reversed-phase HPLC separations and radioimmunoassay confirm that they produce active moulting hormone (20-hydroxyecdysone) at 120 h post-moult. These results suggest that E. pennicornis arrests host development through the indirect effects on their hosts' PGs. This effect is not achieved through the destruction of gland cells, but more likely reflects the interruption of an innate cycle in PG activity, such that they lose their ability to respond to a normal cue to produce an essential hormone peak at a crucial point in development.  相似文献

Honey bee workers normally produce brood food at an age of 5 to 15 days. However, natural events like swarming or brood diseases may lead to the occurrence of over-aged nurses. Here we investigated the physiological consequences of prolonged nursing for both the nurses and the brood they rear, and tried to separate the effects of chronological age and of task affiliation on some important physiological parameters. Brood was reared in groups of colonies with either a normal age structure or with moderately over-aged workers.The haemolymph concentrations of total protein and vitellogenin, the development of mandibular and hypopharyngeal glands, and the activity of α-glucosidase in the hypopharyngeal glands of nurses from these groups of colonies were compared. Moreover, we used the fertility of young workers reared by normal- and overaged nurses as a bioindicator for the quality of the brood care they had received. It showed that parameters linked to the production of brood food proteins remained stable in over-aged nurses, whereas the development of mandibular glands regressed. Workers reared by over-aged nurses had more ovarioles and showed stronger ovary development under queenless conditions. Our results indicate that while over-aged nurses remain capable of producing brood food, they are not functionally equivalent to young nurses. The partial degeneration of the mandibular glands normally occurring at the end of the nursing period cannot be prevented by prolonged nursing. The distinct phenotype of workers reared by old nurses raises the question of possible age-related specialisations among nurses in colonies with a normal age structure. The moulting glands of the milkweed bug, Oncopeltus fasciatus, normally degenerate just before the time of ecdysis to an adult (day 7 of the fifth instar). Morphologically normal cell death can be prematurely stimulated in vitro by 20-hydroxyecdysone. Breakdown is triggered by a 24-hr period of exposure to 20-hydroxyecdysone, but an additional incubation period is required before clear signs of degeneration are manifested. Glands removed after the onset of endogenous ecdysteroid secretion degenerate in vitro in the absence of added hormones. Thus, in the moulting glands of Oncopeltus, ecdysteroids appear to act as an important trigger for metamorphic cell death.  相似文献

Ecdysteroid titers were determined in tissues (gut plus Malpighian tubules, carcass tissue, fat body, muscles, haemolymph, accessory reproductive glands, and testes) of male adult crickets, Gryllus bimaculatus, during the first 20 days of adult life as well as in spermatophores. In all tissues, except testes, total ecdysteroid titers are high on the day of imaginal moulting and then drop more or less continuously until day 8 after moulting. Distinctly higher levels are found on day 12 and 18 as well. Freshly moulted males contain high quantities of polar ecdysteroid conjugates in the digestive tract, testes, accessory reproductive glands, and haemolymph. Apolar ecdysteroid conjugates are mainly detectable in carcass tissue and fat body, but also in the haemolymph during entire adulthood. Free ecdysteroids represent the domineering class of moulting hormones in the gut during all stages of adult life. The significance of cycling ecdysteroid concentrations during adulthood is discussed in relation to spermatophore production and development of male accessory reproductive glands.  相似文献

Summary The content of moulting hormones has been determined in homogenates of isolated prothoracic glands and oenocytes during the 5th instar of the silkworm,Bombyx mori by means of the Calliphora bioassay.Prothoracic glands show variable activity in the production of moulting hormones, reaching a maximum near the end of the larval period. Comparable activities, but at higher levels, could be demonstrated in oenocytes. Controls with doubled quantities of tissue produced in a proportionate reaction in the bioassay. Fat bodies were inactive.Prothoracic glands and oenocytes incubated together resulted in a slower pupation index than would be expected from the sum of single determinations of oenocytes and prothoracic glands. The rôle of the moulting hormone in regeneration was studied in Porcellio dilatatus by destroying the Y-organs (moulting glands) and by injecting synthetic ecdysterone. The destruction of the Y-organs prevents the formation of the regeneration bud of the limb. Several successive injections of a very weak solution of ecdysterone allow the formation of a regeneration bud in animals without Y-organs. A single injection of ecdysterone at a dose which induces apolysis blocks regeneration by making all the epidermal cells secrete cuticle, including those of the regeneration bud.  相似文献

The kinetics of secretion of ecdysone by the prothoracic glands of Locusta migratoria were studied during the last larval instar. Three stages of intense production of ecdysone (α-ecdysone) were monitored during this developmental period: they correspond to three peaks of moulting hormone concentration in the blood, which indicates that the main regulation of the moulting hormone titre is achieved through variations in prothoracic gland activity. In the haemolymph the ratio of ecdysone to ecdysterone (20-hydroxy-ecdysone) is in favour of ecdysone during the two first moulting hormone peaks ecdysterone being by far predominant over ecdysone at the time of the third (major) peak; these results support previous studies on the metabolic fate of injected labelled ecdysone during the same developmental period in Locusta migratoria.  相似文献

Males of a solitary digger wasp, the European beewolf, Philanthus triangulum, possess large mandibular glands that have been reported to produce a scent marking pheromone. We analysed the morphology and ultrastructure of these glands using light microscopy as well as scanning and transmission electron microscopy. The paired glands are located laterally in the head and each side consists of a larger and a smaller part. Both parts possess a collecting duct each with distinct openings at the mandible base. However, the collecting duct of the larger part is additionally connected to the pharynx through a lateral extension. The collecting ducts are bordered by a monolayered epithelium lined with cuticle that exhibits conspicuous ramified protuberances. About 1400 acini consisting of class 3 gland cells surround the ducts and are connected to them through conducting canals. The main components in the cytoplasm of these gland cells are mitochondria, well-developed smooth endoplasmatic reticulum, and electron lucent vesicles suggesting a high secretory activity. The connection between the large gland parts and the pharynx suggests that the secretion of the mandibular glands might not only be delivered directly onto the mandibles but might also be transported to and stored in the postpharyngeal gland.  相似文献

The mandibular glands of workers of M. lobicornis produce a mixture of 3-alkanones and 3-alkanols in the C6–C10 carbon chain length range, in addition to nanogram amounts of acetone, ethanal and 2-methylpropanal. Ethological studies have shown that the three major constituents, 3-octanol, 3-octanone and 3-decanol, are also the major pheromonally active components. When presented together they stimulate an alarm response in the workers similar to that induced by a worker's crushed head.In M. sulcinodis the mandibular gland secretion is composed of a mixture of the same ketones and alcohols, but in different proportions.  相似文献

Summary Processes occurring during moulting in Tetranychus urticae (Acari, Tetranychidae) are described by means of electron microscopy.Moulting is characterized by a pre-ecdysial phase which is initiated by the detachment of cuticle and epidermis. Epicuticular material is deposited as plaques but fuses to form a continuous layer. The epidermis folds up and ridges become determined. Procuticular material is synthesized inside the epidermis and packed into granules which accumulate below the epicuticular portions already deposited. Prior to ecdysis, portions of the old cuticle are dissolved. Ecdysis is achieved by moulting glands which effect bursting of the old cuticle. During the post-ecdysial phase, the endocuticle is synthesized during which a lamellation becomes obvious.Processes occuring during moulting are compared to published information on the tick cuticle.  相似文献

Summary

Several evidences have been obtained in various insect species demonstrating that, besides prothoracic glands and ovaries, other tissues could be alternative sites of moulting hormone production. After a detailed review on the various methods of investigation and criteria required to validate such observations, the nature of these sites, namely oenocytes, epidermis and testes, is discussed. Their possible involvement in moulting and/or reproduction is analyzed, giving the opportunity to put forward several new hypotheses. In particular, autocrine and paracrine secretions of ecdysteroids could play a role in localized developmental events, more difficult to control from endocrine glands possibly mitosis, meiosis, reprogramming, regeneration or early steps of embryogenesis).  相似文献

Experiments were conducted to determine the role of drone mandibular gland secretions in attracting flying drones and the effect of age on the secretory activity. Extracts of mandibular glands and of cephalic tissues were applied to cotton lures which were attached below balloons tethered at 8–12 m above the ground. Most flying drones were attracted to extracts of mandibular glands but a few were drawn to other cephalic tissues or to solvent controls. Histological and electron microscope studies showed that the structure of the tiny (0.12 mm long) mandibular gland varied according to age. Its secretory activity in 0–3-day old drones was evident from the abundant rough endoplasmic reticulum. At 7-days the glands were fully developed. After 9 days the glands were no longer active and showed an autolytic process; the product was stored in the gland lumen for further emission during drone mating flights.  相似文献

ABSTRACT. Larvae of Plodia interpunctella deposit droplets of mandibular gland secretion onto silk filaments connecting particles within the substrate they inhabit, and on to the substrate itself. Active participation of the mouthparts is necessary for both the formation and deposition of these droplets along the filaments and occurs as the silk is spun. Deposition of secretion directly on the substratum does not involve the mouthparts and appears to result from contact between the lower surface of the head and the substratum. Larvae from which the mandibular glands have been removed by excision neither produce droplets on silk nor deposit them on the substratum. Those which are incapable of spinning silk due to cauterization of the spinneret are still able to deposit secretion on the substratum, however. Deposition of secretion on silk webbing does not occur when the setae which regulate formation of droplets on filaments are removed by shaving. Droplets are not characteristic of a particular larval instar but are produced by larvae of all instars as a normal function which does not depend upon intraspecific encounters. Four other Lepidopteran pests, Ephestia elutella, Ephestia cautella, Anagasta kuehniella and Antigasta catolaunalis , were also found to produce droplets of mandibular gland secretion in a similar manner to larvae of P. interpunctella.  相似文献
