Calcium-dependent protein kinases (CDPKs) constitute a unique family of enzymes in plants that are characterized by a C-terminal calmodulin (CaM)-like domain. Through protein kinase assays, we have examined the levels of cucumber calcium-dependent kinase (CsCDPK) activity in various organs of cucumber seedlings and plants. The activity of CsCDPK was highest in cucumber plant leaves followed by seedling roots and hypocotyls; however, cucumber plant flowers, seedling cotyledons, and hooks had levels that were barely detectable. The CsCDPKs were immunolocalized using polyclonal antibodies that are highly specific against a part of the kinase domain of a calcium-dependent protein kinase (CsCDPKS) in the phloem sieve elements (SEs) in various organs of cucumber. In addition, this study indicates the presence of CsCDPKs in organelle-like bodies associated with the plasma membrane of sieve elements in mature stems and roots as well as in the storage bodies of immature seeds. These findings are discussed in terms of the likely roles played by CDPKs in the signal transduction pathways for Ca2+-regulated phloem transport of assimilates from leaves to various organs during growth and development of cucumber seedlings and plants.  相似文献

The effect of synthetic analogs of phytohormones and red light absorbed by phytochrome on the phospholipase D activity (PLD) was studied in oat (Avena sativa L.) seedlings. ABA manifested a short-term stimulating effect on PLD activity in the green seedlings and inhibited phospholipase activity in the etiolated plants. Kinetin inhibited enzyme activity in the etiolated seedlings and did not affect its activity in light. GA did not markedly affect PLD activity in the etiolated plants and activated this enzyme in the green seedlings. Finally, IAA did not affect the enzyme activity. The relationship of the regulatory effects of phytohormones and light on PLD activity is discussed.  相似文献

Benzyladenine (BA), zeatin and kinetin caused the unfolding of cucumber (Cucumis sativus L. Var. Jing-ye No. 4) cotyledons. But the other plant hormones e.g. gibberellic acid (GA3), indoleacetic acid(IAA), ethylene (ethrel), abscisic acid (ABA) and other related compound e.g. KCl, adenine were not so. This response was caused by even lower concentration (0.01 ppm). Therefore, it could be used as a bioassay for cytokinins. KCl affaeted synergistically on cytokinin-induced unfolding. Basally applied BA (10 ppm) stimulated the opening of the intact hypocotylar hook. The light affacted synergistically on BA-induced opening. However, basally applied BA prevented the opening of decapitated hooks, especially in the light. The cotyledons were found to play a part of effects in maintaining the hook.  相似文献

The triazole growth retardant BAS 111‥W delayed senescence in cotyledons of pumpkin ( Cucurbita maxima L. cv. Gelbe genetzte Riesenmelone) and stimulated chlorophyll synthesis in greening cotyledons of oilseed rape ( Brassica napus L. cv. Petranova) seedlings. In both cases, changes of phytohormone-like substances in the cotyledons were analyzed on a fresh weight basis by immunoassay.
After soil treatment with increasing retardant concentrations, a close correlation was observed in senescing cotyledons of pumpkin between a reduced loss in total chlorophyll and increasing levels of dihydrozeatin riboside (DZR) and trans -zeatin riboside (ZR)-type cytokinins. In contrast, the levels of isopentenyladenosine (IPA)-type cytokinins, 3-indoleacetic acid (IAA) and gibberellin (GA) did not change significantly. The levels of abscisic acid (ABA) were slightly elevated at low retardant concentrations but dropped considerably below those of controls at higher doses. Consequently, the molar ratio of total cytokinin to ABA content changed from approximately 1:40 in controls (50% of initial chlorophyll) to 1:3 in cotyledons treated with 3 mg BAS 111‥W plant−1 (85% of initial chlorophyll). These changes, together with the known reduction of ethylene production by plants treated with nitrogen-heterocyclic retardants, can explain the delayed senescence in pumpkin cotyledons. Likewise, when etiolated, BAS 111‥W-treated seedlings of oilseed rape were exposed to light, the stimulation of chlorophyll synthesis in the cotyledons was accompanied by an accumulation of DZR- and, particularly, ZR-type cytokinins and IAA. In contrast, GA and ABA contents decreased slightly. We conclude that the influence of BAS 111‥W on cytokinin levels might be involved in the stimulation of greening.  相似文献

The level of indol-3yl-acetic acid (IAA) in the cotyledons of cucumber seedlings increases in the period 4 to 11 days after germination. In hypocotyls and roots the IAA level decreases. IAA-oxidase activity of homogenates of cotyledons, hypocotyls and roots decreases with age. The soluble fraction of all three organs contains an IAA-oxidase, which may be allosteric. It is suggested that this IAA oxidase limits the upper level of IAA in the cell. The cell wall fraction of the three organs contains IAA-oxidases with conventional kinetics. The function of this IAA oxidase is probably to degrade exogenous IAA.  相似文献

1 ppm表油菜素内酯(epiBR)能显著地促进光下生长的黄瓜下胚轴伸长。其促进伸长的效应需约5h滞后期,明显超过IAA而与GA_3的滞后期接近。GA_3对epiBR促进黄瓜下胚轴的伸长具有增效作用。epiBR能提高内源GA_3,ABA的水平,处理24h后,GA_3/ABA值约为对照的两倍。经epiBR处理后的黄瓜下胚轴内淀粉含量较低,并维持在同一水平,这与GA_3的作用颇为一致。  相似文献

We studied the effect of 24-epibrassinolide (EB) on the levels of endogenous hormones and photomorphogenesis of Arabidopsis thaliana (L.) Heynh wild-type (Ler) and mutant (hy4) seedlings. This mutant is deficient in the cryptochrome 1 (CRY1) synthesis. CRY1, which is a product of the HY4 gene, is a blue light photoreceptor in wild-type plants, but is sensitive to green light as well. In dark-grown seven-day-old mutant seedlings, the ABA/zeatin ratio differed from this ratio in wild-type seedlings. Thehy4 mutant exhibited a lower zeatin and higher free-ABA contents, which could retard its hypocotyl growth in darkness. EB retarded the growth of hypocotyls in etiolated hy4 seedlings and enlarged their cotyledons more efficiently than in wild-type seedlings. Green light (GL) did not affect the growth of hypocotyls but enlarged cotyledons of hy4 seedlings, which might be associated with some increase in the level of free IAA and a considerable decrease in free ABA and also with a decrease in the cytokinin level in seedlings. The hy4 cotyledon response to GL depended evidently on photoreceptors other than CRY1. GL enhanced the effects of EB on the morphogenesis of both Ler and hy4 seedlings, which was coupled with changes in the balance of endogenous IAA, ABA, and cytokinins. We may suppose that EB is involved in the control of photomorphogenesis by interaction with endogenous hormones, which are involved in the transduction of a light signal absorbed by the GL photoreceptors.  相似文献

The endogenous contents of indoleacetic acid (IAA) and abscisicacid (ABA) were assayed simultaneously in etiolated squash hypocotylsthat had been subjected to water stress applied to squash rootsby immersing them in 60 mM polyethylene glycol solution. Thegrowth rate and suction force of the hypocotyl under this water-stresscondition were determined for comparison with the two hormonecontents. Water stress retarded hypocotyl growth and increased the suctionforce of hypocotyl cells, as the ABA content increased, by 5-foldof the initial content one day after treatment with polyethyleneglycol. The ABA contents thereafter remained high up to day4. The increase in ABA content was detected in stressed hypocotylas early as 6 h after treatment, when water stress had not affectedhypocotyl growth. When the water stress was terminated one dayafter treatment began, hypocotyl growth immediately recovered,and the ABA content decreased to the value for unstressed hypocotylsin one day. The IAA content gradually decreased in unstressedhypocotyls, but was maintained at a high value in stressed hypocotyls. Logarithmic concentrations of endogenous ABA (ng/g fr wt) werecorrelated significantly with the suction force of the hypocotyls(r=0.92) and with the growth rates (r=–0.78). IAA contentwas correlated neither with the growth rate nor the suctionforce. These results suggest that the endogenous ABA contentwas associated with the stunted growth of the etiolated squashhypocotyls produced by water stress but that the endogenousIAA content did not have an important role in growth regulationunder water stress conditions. 黄瓜幼苗用0.1~1 ppm表油菜素内酯(epiBR)处理1~3d后,下胚轴中过氧化物酶活性明显低于对照;随着处理浓度的增加和处理时间的延长,与对照之间的差别愈趋增大。当浓度高于1ppm时,过氧化物酶的活性不再继续降低。表油菜素内醋对过氧化物酶活性的这种抑制作用需经约1O h的滞后期。IAA氧化酶的活性变化与过氧化物酶相似,epiBR处理时间愈长酶活性增加愈趋缓慢。 经IAA处理的下胚轴,过氧化物酶和IAA氧化酶的活性变化与对照无明显差异。这提示油菜素内酯与IAA促进生长可能是通过不同的作用方式。  相似文献

Phytochrome changes in hypocotyls of light grown mustard seedlings (Sinapis alba L.) and the phytochrome destruction in cotyledons of etiolated plants have been analysed as influenced by exogenous gibberellic acid (GA3). The rate of phytochrome appearance in hypocotyls was decreased after GA3 treatment. Destruction of phytochrome in cotyledons of etiolated seedlings was significantly accelerated by the same hormone treatment. This effect was also observed in the hypocotyls of light grown seedlings.  相似文献

Results of studies on the interaction of blue light (B) and exogenous applied plant hormones (IAA, GA3 and ABA) as well as inhibitors of their synthesis on the control of hypocotyl elongation in etiolated seedlings of Lactuca sativa (L.) cv. Grand Rapids are presented, and compared with endogenous GA3, IAA and ABA levels measured by capillary gas chromatography-mass spectrometry-selected ion monitoring (GC-MS-SIM). Hypocotyl elongation of etiolated seedlings was linearly inhibited by increasing the B fluence rate between 0 and 18.3 mol m–2 s–1. Both GA3 and IAA added to the incubation medium at different concentrations were able to eliminate partially the inhibition of growth caused by 7.2 mol m–2 s–1 B. When dark (D)-grown seedlings were treated with Ca-prohexadione, a specific inhibitor of gibberellin 3-hydroxylation, they showed a growth inhibition similar to under B. Also, a suppression of growth as in B was obtained when D-grown seedlings were treated with ABA 380 M. By lowering ABA levels with fluridone (an inhibitor of ABA synthesis) a partial reversion of hypocotyl growth inhibition was obtained in B-grown seedlings. While none of the growth promoters used were able to reverse completely the growth inhibition caused by B, a proper combination of GA3, IAA and (eventually) fluridone, abolished the B effects. Correspondingly, lower levels of GA3 and IAA and a higher concentration of ABA were measured by GC-MS-SIM in B-grown hypocotyls than in D-grown ones. These results support the hypothesis that hormones are implicated in mediation of B light-dependent inhibition of hypocotyl elongation, which seems to be the result of a balance among endogenous levels of growth promoting and growth inhibiting hormones.  相似文献

During induction of somatic embryogenesis from immature embryos of soybean, a smooth-shiny and a rough callus were obtained. The smooth-shiny type developed from callus derived from cotyledonary tissue and cultured on media containing 10 mg/l 2,4-D. The rough type was derived from immature embryos, cotyledons, hypocotyls and hypocotyl segments from germinated seedlings on a medium containing various growth regulators. Plants were obtained from the smooth-shiny type but rough callus did not differentiate into plants. This study examined the influence of three inoculant strains of Bradyrhizobium japonicum (Thal-8, Tal 620, Dulawala) on the ability of chickpea (Cicer arietinum (L.) to adapt to drought-stress. Strain Thal-8 was most effective in the root-nodule symbiosis and also partially alleviated decreased growth and yield imposed by drought stress. Strain Thal-8, in pure culture, also produced higher amounts of gibberellic acid (GA) and indole-3-acetic acid (IAA) and lower amounts of abscisic acid (ABA) than the other two test strains. Thal-8 increased the root biomass, GA and IAA contents of leaves of chickpea plants, including ICC 4948NN, a non-nodulating line. These results are consistent with the hypothesis that GA and IAA is produced by the Thal-8 strain and/or elevates levels of these phytohormones in chickpeas. This contributes to its high performance as a nitrogen-fixing microsymbiont. The growth-promoting response evoked by different strains of Bradyrhizobium correlated with higher ratios of GA and IAA relative to ABA phytohormones in the plants.  相似文献
