Despite its long history, biogeography has received relatively little attention within the field of microbial ecology. Consequently, a fierce debate rages whether protists inhabit restricted geographic areas (endemism hypothesis) or are globally dispersed (ubiquitous dispersal hypothesis). The data presented in this article support the endemism hypothesis. We succeeded in isolating an oligohymenophorean ciliate from a microbial mat in a meromictic anoxic alpine lake (Alatsee) in Germany. The ciliary pattern and the morphometry of this isolate are remarkably similar to Urocentrum turbo (Mueller, 1786) Nitzsch, 1827. However, the organism does not possess trichocysts, a conspicuous and characteristic feature of U. turbo. Instead, the U. turbo-like isolate from lake Alatsee displays merely trichocyst anlagen (“ghosts”) in the cytoplasm that are only visible after protargol impregnation and which become never attached to the cell’s cortex. Despite the distinctness of this difference, such a morphospecies has not been described from any other environment. Thus, we suggest that the U. turbo-like isolate from lake Alatsee is a local endemic ecotype, although the sequences of the 18S rRNA, ITS1, 5.8S rRNA, and ITS2 genes are nearly identical to those of U. turbo (Mueller, 1786) Nitzsch, 1827. This indicates that neither 18S rDNA nor ITS1, ITS2, and 5.8S rDNA sequences are reliable means to conclusively resolve different morphospecies or ecotypes of ciliates. As a consequence, we argue that protist species richness can only be reliably accounted for by considering both molecular and morphological data.  相似文献

Background

During much of the Late Cretaceous, a shallow, epeiric sea divided North America into eastern and western landmasses. The western landmass, known as Laramidia, although diminutive in size, witnessed a major evolutionary radiation of dinosaurs. Other than hadrosaurs (duck-billed dinosaurs), the most common dinosaurs were ceratopsids (large-bodied horned dinosaurs), currently known only from Laramidia and Asia. Remarkably, previous studies have postulated the occurrence of latitudinally arrayed dinosaur “provinces,” or “biomes,” on Laramidia. Yet this hypothesis has been challenged on multiple fronts and has remained poorly tested.

Methodology/Principal Findings

Here we describe two new, co-occurring ceratopsids from the Upper Cretaceous Kaiparowits Formation of Utah that provide the strongest support to date for the dinosaur provincialism hypothesis. Both pertain to the clade of ceratopsids known as Chasmosaurinae, dramatically increasing representation of this group from the southern portion of the Western Interior Basin of North America. Utahceratops gettyi gen. et sp. nov.—characterized by short, rounded, laterally projecting supraorbital horncores and an elongate frill with a deep median embayment—is recovered as the sister taxon to Pentaceratops sternbergii from the late Campanian of New Mexico. Kosmoceratops richardsoni gen. et sp. nov.—characterized by elongate, laterally projecting supraorbital horncores and a short, broad frill adorned with ten well developed hooks—has the most ornate skull of any known dinosaur and is closely allied to Chasmosaurus irvinensis from the late Campanian of Alberta.

Conclusions/Significance

Considered in unison, the phylogenetic, stratigraphic, and biogeographic evidence documents distinct, co-occurring chasmosaurine taxa north and south on the diminutive landmass of Laramidia. The famous Triceratops and all other, more nested chasmosaurines are postulated as descendants of forms previously restricted to the southern portion of Laramidia. Results further suggest the presence of latitudinally arrayed evolutionary centers of endemism within chasmosaurine ceratopsids during the late Campanian, the first documented occurrence of intracontinental endemism within dinosaurs.  相似文献

Abstract The growth rate of 31 stocks of Euplotes, a cosmopolitan, marine, unicellular protist, on six food species representing two different food types, microalgae and bacteria, has been determined. The 31 stocks represented nine reproductively isolated groups (biological species?) based on breeding relationships. Three morphospecies, E. vannus, E. crassus, and E. minuta, each with both autogamous and cross-breeding breeding groups, were included. The mean number of fissions completed in 5 days of a breeding group growing on one of the six food species varied from zero to 17.06. There is a strong interaction between morphospecies and food type. The largest morphospecies, E. vannus, translates the nutritional content of algae into growth better than it translates that of bacteria, while the reverse is true for E. minuta, the smallest morphospecies. Autogamous breeding groups grow more rapidly on algae than on bacteria when compared to cross-breeding groups in the same morphospecies. Two breeding groups cannot grow on Escherichia coli. ANOVA of fissions completed in 5 days revealed significant main effects and interactions between many hierarchical levels of stocks and food species. These significant interactions indicate that genetically determined ecologically important information is present at all taxonomic levels—morphospecies, breeding system, breeding group, and stock. As all these levels are biologically meaningful, measuring biodiversity in the E. vannus–crassus–minuta complex solely on morphospecies will inadequately represent the ecological diversity present in the organisms and their environment. The possible existence of endemism among microorganisms resulting from and preserved by geographic isolation is one of the most controversial topics in microbial ecology. We isolated 31 strains of "Spumella-like" flagellates from remote sampling sites from all continents, including Antarctica. These and another 23 isolates from a former study were characterized morphologically and by small-subunit rRNA gene sequence analysis and tested for the maximum temperature tolerance. Only a minority of the Spumella morpho- and phylotypes from the geographically isolated Antarctic continent follow the worldwide trend of a linear correlation between ambient (air) temperature during strain isolation and heat tolerance of the isolates. A high percentage of the Antarctic isolates, but none of the isolates from locations on all other continents, were obligate psychrophilic, although some of the latter were isolated at low ambient temperatures. The drastic deviation of Antarctic representatives of Spumella from the global trend of temperature adaptation of this morphospecies provides strong evidence for geographic transport restriction of a microorganism; i.e., Antarctic protistan communities are less influenced by transport of protists to and from the Antarctic continent than by local adaptation, a subtle form of endemism.  相似文献

Platelets achieve bleeding arrest at sites of vascular injury via secretion of secretory proteins from their storage granules, termed alpha-granules. We have recently analyzed granule targeting of platelet factor 4 (PF4), a secretory alpha-granule chemokine, and demonstrated that PF4 alpha-granule storage relied upon determinants within PF4 mature sequence. To define these determinants, PF4 mutants fused to the fluorescent reporter protein green fluorescent protein were generated by progressive deletions and site-directed mutagenesis. They were then transfected in AtT20 cells and assessed for granule targeting by colocalization with ACTH-containing granules, using laser scanning confocal microscopy. This strategy identified the amino acid 41-50 (LIATLKNGRK) sequence as most critical for PF4 granule targeting and/or storage; its deletion from PF4 induced a marked decrease in granule storage (from 81 +/- 2% to 17 +/- 3%, p < or = 0.0001). Ala-scanning mutagenesis of LIATLKNGRK narrowed down the targeting motif to LKNG. A direct role for LKNG in alpha-granule targeting was confirmed in the thrombopoietin-induced human megakaryocytic Dami cells, in which the LKNG-green fluorescent protein chimera exhibited an 82.5 +/- 1.8% colocalization with the alpha-granule proteins von Willebrand factor and P-selectin. LKNG is poorly conserved within the chemokine family. However three-dimensional alignments of the human alpha-granule chemokines Nap-2 (neutrophil-activating peptide) and RANTES (Regulated upon Activation Normal T Cell Expressed and Secreted) with PF4 revealed that LKNG, a surface-exposed hydrophilic turn/loop, matched Nap-2 (LKDG) and RANTES (TRKN) peptides with similar features. Moreover Nap-2 and RANTES peptides exhibited the same alpha-granule targeting efficiency than LKNG. We therefore postulate that the three-dimensional and physicochemical characteristics of PF4 LKNG are of general relevance to alpha-granule targeting of chemokines and possibly of other alpha-granule proteins.  相似文献

This study examines key elements of glutamatergic transmission within sensory ganglia of the rat. We show that the soma of primary sensory neurons release glutamate when depolarized. Using acute dissociated mixed neuronal/glia cultures of dorsal root ganglia (DRG) or trigeminal ganglia and a colorimetric assay, we show that when glutamate uptake by satellite glial cells (SGCs) is inhibited, KCl stimulation leads to simultaneous increase of glutamate in the culture medium. With calcium imaging we see that the soma of primary sensory neurons and SGCs respond to AMPA, NMDA, kainate and mGluR agonists, and selective antagonists block this response. Using whole cell patch-clamp technique, inward currents were recorded from small diameter (<30 µm) DRG neurons from intact DRGs (ex-vivo whole ganglion preparation) in response to local application of the above glutamate receptor agonists. Following a chronic constriction injury (CCI) of either the inferior orbital nerve or the sciatic nerve, glutamate expression increases in the trigeminal ganglia and DRG respectively. This increase occurs in neurons of all diameters and is present in the somata of neurons with injured axons as well as in somata of neighboring uninjured neurons. These data provides additional evidence that glutamate can be released within the sensory ganglion, and that the somata of primary sensory neurons as well as SGCs express functional glutamate receptors at their surface. These findings, together with our previous gene knockdown data, suggest that glutamatergic transmission within the ganglion could impact nociceptive threshold.  相似文献

In a recent paper (Bellwood & Meyer, Journal of Biogeography , 2009, 36 , 569–576), we critically evaluated the utility of marine endemics for marking the geographical origins of species. In reply, Briggs (2009) identified two issues that needed clarification: (1) whether endemics are assumed to mark the geographical origins of species or areas of exceptionally high rates of origination, and (2) whether our evaluation of the role of endemics disproves the centre of origin hypothesis. Of these two issues, the first can be clearly resolved by recourse to the original literature that explicitly states that it has been assumed that endemics do indeed mark the probable sites of origin of species. The second is equally clear: our evidence does not and can not disprove the centre of origin theory. We suggest, however, that the current data, and endemics in particular, provide limited support for centre of origin theories and that they are more consistent with some centre of accumulation theories. The Indo-Australian Archipelago (IAA; Coral Triangle) therefore appears to be an area where species persist, a centre of survival, regardless of the site of origin of species.  相似文献

The bacterium Helicobacter pylori colonizes the human stomach, with individual infections persisting for decades. The spread of the bacterium has been shown to reflect both ancient and recent human migrations. We have sequenced housekeeping genes from H. pylori isolated from 147 Iranians with well-characterized geographical and ethnic origins sampled throughout Iran and compared them with sequences from strains from other locations. H. pylori from Iran are similar to others isolated from Western Eurasia and can be placed in the previously described HpEurope population. Despite the location of Iran at the crossroads of Eurasia, we found no evidence that the region been a major source of ancestry for strains across the continent. On a smaller scale, we found genetic affinities between the H. pylori isolated from particular Iranian populations and strains from Turks, Uzbeks, Palestinians and Israelis, reflecting documented historical contacts over the past two thousand years.  相似文献

Environmental factors restrict the distribution of microbial eukaryotes but the exact boundaries for eukaryotic life are not known. Here, we examine protistan communities at the extremes of salinity and osmotic pressure, and report rich assemblages inhabiting Bannock and Discovery, two deep-sea superhaline anoxic basins in the Mediterranean. Using a rRNA-based approach, we detected 1,538 protistan rRNA gene sequences from water samples with total salinity ranging from 39 to 280 g/Kg, and obtained evidence that this DNA was endogenous to the extreme habitat sampled. Statistical analyses indicate that the discovered phylotypes represent only a fraction of species actually inhabiting both the brine and the brine-seawater interface, with as much as 82% of the actual richness missed by our survey. Jaccard indices (e.g., for a comparison of community membership) suggest that the brine/interface protistan communities are unique to Bannock and Discovery basins, and share little (0.8–2.8%) in species composition with overlying waters with typical marine salinity and oxygen tension. The protistan communities from the basins’ brine and brine/seawater interface appear to be particularly enriched with dinoflagellates, ciliates and other alveolates, as well as fungi, and are conspicuously poor in stramenopiles. The uniqueness and diversity of brine and brine-interface protistan communities make them promising targets for protistan discovery.  相似文献

Protists are a diverse group of eukaryotes that possess a unicellular level of organization. As unicellular organisms, the differentiation of cells into tissues does not occur, although when cell differentiation does occur, it is limited to sexual reproduction, alternate vegetative morphologies or quiescent life history stages. Protistan parasites may possess simple or complex life histories that are important factors to consider when investigating protistan diseases of decapods. Unfortunately, the life histories of many protistan parasites of decapods are insufficiently described, resulting in the fact that modes of infection and transmission are often unidentified. This is surprising considering the economic importance of many marine decapods and the ability of protistan parasites to produce significant, but generally transient and area limited mortalities. However, the marine disease landscape is changing and will continue to change as climate change and ocean acidification will play important roles in disease occurrence and distribution. As a result, the following discussion attempts to summarize current knowledge on several crab diseases, their protistan etiological agents, the impact of disease on economically important crab populations and draw attention to areas of needed research. The discussion is not complete as only selected diseases are addressed, or perfect as the Microsporidia are included in the discussion (a traditional error continued in this summary) despite the recent, but controversial placement of the taxon with the fungi.  相似文献

Endemicity is important for the delimitation of conservation areas. Endemic areas are those that contain two or more taxa with their distribution restricted to the area. The aim of this study was to detect endemic areas for palms in the Amazon region and to determine whether the species that define these endemic areas are protected within conservation units. Records of occurrence were extracted from the global biodiversity information facility (GBIF). The final dataset consisted of 17,310 records, for 177 species of Amazonian palms. For analysis we used parsimony analysis of endemicity (PAE) and NDM-VNDM program, and grid square size of 1° and 3° as operational geographic units (OGUs). The distribution of endemic species was superimposed on occurrence of the conservation units (CUs). PAE did not show endemic areas in grid squares of 1°, but found 10 palm endemic areas in grid squares of 3° in the western Amazon and Andean sub-region. However, the NDM-VNDM program identified an endemic area in grid squares of 1° located at the eastern Guiana with endemicity score = 2.9, and in grid squares of 3° it identified seven consensus areas with endemicity score > 6.0, all in the western Amazon. The combination of PAE and NDM-VNDM analyses resulted in eight endemic palm areas in the combined western Amazon and Andean sub-region. Of the species that define the endemic areas, five are threatened with extinction in one of three IUCN categories (EN, VU, NT), and they are not protected in any conservation units. The western Amazon, besides having high palm richness, also has palm endemic areas, especially, near the Andean sub-region and the Peruvian Amazon.  相似文献

We describe the monoxenous trypanosomatids parasitizing true bugs and flies on the island of Curaçao. Out of 248 examined true bugs belonging to 17 species, 93 individuals were found to be infected (overall 38% prevalence) by at least one trypanosomatid species (referred to as typing units; TUs). Out of 80 flies, six were infected. All detected trypanosomatids were compared based on their 18S rRNA sequences with TUs parasitizing bugs and flies described from mainland South America, allowing us to assess their diversity and distribution. Besides Leptomonas pyrrhocoris and Leptomonas seymouri, two known species of the subfamily Leishmaniinae, our analysis revealed six new TUs falling into the groups ‘jaculum’, Blastocrithidia and Herpetomonas. Moreover, two new members of the genus Phytomonas and three new TUs belonging to the monophyletic group designated as ‘new clade II’ sensu Mol. Phylogenet. Evol, 69, 255 (2013) were isolated. The detected trypanosomatids were characterized by moderate diversity (13 TUs) species richness. Out of nine and four TUs from the heteropteran and dipteran hosts, respectively, 11 TUs have not been encountered before. Although a sampling bias may partially affect the comparison between trypanosomatid communities on Curaçao and the mainland, the high proportion of unique TUs from the former location suggests that the prominent role of islands in increasing the global diversity of macroscopic organisms may also extend to their protistan parasites.  相似文献

Approximately 5% of the human genome consists of segmental duplications that can cause genomic mutations and may play a role in gene innovation. Reticulate evolutionary processes, such as unequal crossing-over and gene conversion, are known to occur within specific duplicon families, but the broader contribution of these processes to the evolution of human duplications remains poorly characterized. Here, we use phylogenetic profiling to analyze multiple alignments of 24 human duplicon families that span >8 Mb of DNA. Our results indicate that none of them are evolving independently, with all alignments showing sharp discontinuities in phylogenetic signal consistent with reticulation. To analyze these results in more detail, we have developed a quartet method that estimates the relative contribution of nucleotide substitution and reticulate processes to sequence evolution. Our data indicate that most of the duplications show a highly significant excess of sites consistent with reticulate evolution, compared with the number expected by nucleotide substitution alone, with 15 of 30 alignments showing a >20-fold excess over that expected. Using permutation tests, we also show that at least 5% of the total sequence shares 100% sequence identity because of reticulation, a figure that includes 74 independent tracts of perfect identity >2 kb in length. Furthermore, analysis of a subset of alignments indicates that the density of reticulation events is as high as 1 every 4 kb. These results indicate that phylogenetic relationships within recently duplicated human DNA can be rapidly disrupted by reticulate evolution. This finding has important implications for efforts to finish the human genome sequence, complicates comparative sequence analysis of duplicon families, and could profoundly influence the tempo of gene-family evolution.  相似文献

Sexually dimorphic characteristics in men may act as cues, advertising long-term health, dominance, and reproductive potential to prospective mates. Evolution has accordingly adapted human cognition so that women perceive sexually dimorphic facial features as important when judging the attractiveness and suitability of potential mates. Here we provide evidence showing, for the first time, that women's memory for details encountered in recently experienced episodes is also systematically biased by the presence of men's facial cues signaling enhanced or reduced sexual dimorphism. Importantly, the direction and strength of this bias are predicted by individual differences in women's preferences for masculine versus feminine facial features in men and are triggered specifically while viewing images of male but not female faces. No analogous effects were observed in male participants viewing images of feminized and masculinized women's faces despite the fact that male participants showed strong preferences for feminized facial features. These findings reveal a preference-dependent memory enhancement in women that would promote retention of information from encounters with preferred potential mates. We propose that women's memory for recently experienced episodes may therefore be functionally specialized for mate choice and in particular for the comparative evaluation of alternative potential mates. This also raises the possibility that similar specialization may be present in other species where it has been established that precursor, ‘episodic-like’ forms of memory exist.  相似文献
