Abstract: Feeding bioassays were conducted on several Pinus sylvestris clones to establish if there were any differences in suitability for two pine defoliators: the sawfly Diprion pini L. (Hym., Diprionidae), which causes considerable damage in Europe, and the rare and protected moth Graellsia isabellae galliaegloria Oberthür (Lep., Attacidae). There were significant differences in survival, weight, sex-ratio and female fecundity of D. pini on Scots pine clones. However, sawfly survival appeared to be the most stable variable in time and three clone categories were revealed by conducting feeding bioassays on 16 clones. The survival of G. isabellae galliaegloria until the formation of the chrysalis was significantly different among clones. It varied from 35 to 75% but no clear clone category was distinguished . The two clone classifications according to insect survival showed differences. In particular, one unsuitable clone for D. pini development appeared to be favourable to moth development. The 3-carene richness of foliage was significantly linked with sawfly survival and the monoterpene composition of foliage was not linked with G. isabellae galliaegloria survival.  相似文献

Abstract: Based on haploid males, random amplified polymorphic DNA (RAPD) markers were used to study genetic variation within and among four French populations and one Finnish outgroup population of the common pine sawfly, Diprion pini (L.), representing a severe European forest pest associated with mass outbreaks. Taking into account that all multilocus haplotypes were detected, a total of 140 individuals were completely discriminated by means of 17 polymorphic markers (present or absent), which were amplified from nine selected random-decamer primers. All populations shared the same genetic types, but pronounced population-specific frequency distributions were found, indicating that on average 84% of the present genetic variation exists within populations and the remaining smaller part counts for interpopulational variation. The haplotype differentiation registered was able to distinguish between the class of lowland populations and the class of highland populations. Reproductive isolation and therefore limited altitudinal gene flow, indicated in case of a French highland population by an enlarged number of fixed markers, and hitchhiking effects with respect to selective processes at certain loci following local adaptation and speciation are discussed to explain the population structures found. A possible selection is indicated by five markers, showing significantly different frequency distributions between the class of highland populations and the class of lowland populations.  相似文献

Abstract. Face flies overwinter as adults in reproductive diapause. Administration of 20-hyroxyecdysone and/or methoprene induced reproductive development in diapausing flies which were maintained in a diapause-inducing environment. Hormone effects were additive and female flies were more sensitive than males. Release of vitellogenin from cultured fat body was stimulated by 20-hydroxyecdysone or methoprene. Transfer of flies from diapause to diapause-breaking environments induced some to break diapause, but this decreased with the time flies had been in a diapause-inducing environment. In contrast, topical application of methoprene to diapausing flies induced reproductive development irrespective of their ages even when they were kept in the diapause-inducing environment for 80 day degrees above a 12°C base temperature (14.5 days). Therefore diapause induction must depend on hormone levels less than some threshold level. The putative threshold varied according to diapause propensities of different genetic lines. Lines showing high frequencies of diapause required greater amounts of methoprene for reproductive development in diapause conditions than did lines showing low frequencies of diapause.  相似文献

Abstract: Species composition and impact of larval parasitoids during an endemic density phase of a pine forest pest, the common pine sawfly Diprion pini (L.), were investigated by exposure of laboratory-reared larvae of the sawfly to parasitoid attack in natural habitats. Colony-associated host larvae of different instars were exposed continuously from early June until end of September of the years 1994 and 1995 in two pine stands with a different history of pine sawfly outbreaks. Host larvae were exchanged after a defined time interval and those collected up were examined in the laboratory for parasitism. Parasitoid assemblages were shown to be very species poor at both pine stands. The dominant species were the ichneumonid Olesicampe macellator and the tachinid Drino gilva , which reached attack rates on the collected host larvae of 60 and 94%, respectively, during peak activity. The patterns of temporal attack, of multiparasitism, and of superparasitism of these dominant species were studied. In an additional experiment, the response of these species to different host-patch sizes (number of host larvae per colony) were investigated. No aggregative response to higher host-patch densities could be detected. The role and function of these larval parasitoids in the population dynamics of D. pini are discussed.  相似文献

Ecdysone 20-hydroxylase activity has been detected in pupal wing discs of Pieris brassicae. This activity is due to an enzyme system located in microsomal fractions. Its apparent Km is 58 nM for ecdysone. The enzyme is inhibited by the reaction product 20-hydroxyecdysone with an apparent Ki of 2.6 μM. Its activity varied during pupal-adult development with a maximum on day 4, when ecdysone levels are the highest in the animal. Although low, the peak activity is sufficient to assure 25% of the conversion of endogenous ecdysone into 20-hydroxyecdysone in pupae. Ecdysone and 20-hydroxyecdysone levels were measured in hemolymph and whole animals; ecdysone appears to be mainly located in hemolymph, whereas 20-hydroxyecdysone seems to be equally distributed between hemolymph and tissues. All these findings are discussed in relation to the roles of ecdysone and 20-hydroxyecdysone during pupal-adult development.  相似文献

The radiolabeled lectins, concanavalin A* and wheat germ agglutinin, were used to study surface properties of two insect cell lines. We also looked at the effects of tunicamycin and 20-hydroxyecdysone on the binding of these lectins to one of the cell lines. Both UMBGE-2 and CH-MRRL cells bound both lectins, specifically. The CH-MRRL cells showed an overall higher binding for the lectins than the UMBGE-2 cells. This difference may account for some of the striking morphological difference seen between these cells. Tunicamycin and 20-hydroxyecdysone decreased the binding of both [125I]-Con A and [125I]-WGA to CH-MRRL cells. These results suggest that cell surface glycoproteins play a role in the modification of cellular morphology and in other hormone-mediated physiological functions.  相似文献

Studies of the effects of a novel insect growth regulator (tebufenozide, RH 5992) in pest lepidopterans are extended to its influence on pre- and post-diapause larvae of the pine-tree lappet, Dendrolimus pini (L.) (Lep., Lasiocampidae). Larval diapause was induced under experimental conditions. The relation between the width of head capsule and each larval instar of the experimental populations is presented. The toxicity of tebufenozide applied topically and per os (orally), and its effects on feeding activity were examined. High doses and concentrations (≥100 ng/larva and ≥0.001%) of tebufenozide were toxic and reduced the feeding intensity in larvae of D. pini , however, post-diapause larvae were less sensitive to oral treatment with tebufenozide then pre-diapause insects. In pre-diapausing insects of the fourth instar a greater than four-fold fall in mortality was observed among the insects treated with low doses/ concentrations (0.1–10 ng/larva and 0.00001%–0.001%, respectively). A remarkable stimulatory effect on feeding behaviour was observed in pre-diapausing IV instar larvae when treated with tebufenozide topically, in a dose of 10 ng/larva. A possible ecdysteroid mimicking effect of tebufenozide onto feeding and mortality among diapause-destined D. pini is discussed.  相似文献

The effects of 20-hydroxyecdysone (5 μg/g body weight), cycloheximide (5 μg/g body weight), puromycin (30 μg/g body weight) and actinomycin D (30 μg/g body weight) were investigated on the larval fat body cells of 48–70-hours-old last instar larvae of Pieris brassicae.20-Hydroxyecdysone was found to induce the formation of autophagic vacuoles 3 hr after its administration, but this was, however, prevented by simultaneous cycloheximide treatment in a parallel experiment. On the contrary, puromycin proved to induce autophagocytosis. These diverse effects of the two translational inhibitors on hormone-induced autophagocytosis may be explained by differences in their modes of action.Actinomycin D, when administered 21 hr prior to the hormone, exerted a preventive effect on induced autophagocytosis, but was ineffective when applied 3 hr before the injection of 20-hydroxyecdysone.It was concluded that the presence of RNA synthesized several hours prior to the hormone treatment was a prerequisite for induction of autophagocytosis by 20-hydroxyecdysone.  相似文献

Transmission of a nuclear polyhedrosis virus (NtNPV) of the loblolly pine sawfly,Neodiprion taedae linearis Ross was examined in larval populations on loblolly pine. NtNPV infection of larvae in one colony resulted in transmission to other colonies in the tree. Mortality in untreated colonies was highest when the treated and untreated colonies were located in the upper canopy and when small larvae were treated with virus (42.6–61.9%). Levels of mortality from NtNPV in the untreated colonies were low (2.8–25.0%) when the treated colony was located low in the canopy. Larvae from untreated colonies in the tree were usually large at death. Mortality in untreated larval colonies collected from nearby trees was much less (0.0–27.7%) than that in the trees containing the treated colony. Injection of the hormone 20-hydroxyecdysone (20-E) into partially fed (virgin) female adults of the American dog tick, Dermacentor variabilis, while they are attached and feeding on the rabbit host, initiated the expression of the vitellogenin (Vg) gene, and Vg protein secretion and uptake by the ovary. The induction of egg production by 20-E in this bioassay was dose dependent in the range of 1-50 times the concentration normally found in a replete, vitellogenic female. Ticks examined 4 d after the 50 x treatment were still attached to the host, had numerous enlarged vitellin-filled (brown) oocytes in their ovaries, but had not engorged to repletion. The ovaries reached weights similar to those found in untreated, replete (mated) females (pre-oviposition) while solvent-injected controls demonstrated no increase in oocyte size or increase in ovary weight. An increase in the levels of a putative Vg protein was observed in hemolymph samples collected 1, 2 and 3d post-20-E injection but was not observed in the corresponding solvent controls as determined by native PAGE. Analysis of the ecdysteroid-induced protein by tryptic digestion-mass fingerprinting and BLASTP found that the putative Vg had the strongest match to GP80 (U49934), the partial sequence for the vitellogenin protein from Boophilus microplus. A partial Vg cDNA was cloned and sequenced from replete females of D. variabilis with a high similarity to GP80. Using this message as a probe, Northern blots conducted with RNA collected from partially fed, virgin females 1, 2 and 3d post-20-E injection showed upregulation of the Vg mRNA on all 3 days. Controls injected with solvent only showed no Vg mRNA. Injections with juvenile hormone III did not stimulate Vg expression, oocyte growth or full engorgement. These studies indicate that ecdysteroids and not JH can initiate expression of the Vg gene, Vg protein synthesis and release into hemolymph, and Vg uptake into developing oocytes under bioassay conditions mimicking normal feeding on the host.  相似文献

When the titre of juvenile hormone III in female Leptinotarsa decemlineata was elevated by the implantation of supernumerary corpora allata or by the injection of the hormone, the rate of endogenous hormone production by the host glands was significantly restrained, as determined by the short-term in vitro radiochemical assay. From denervation studies, it is suggested that during phases of elevated juvenile hormone titre, the corpus allatum activity is regulated via humoral as well as neural factors requiring intact nerve connections. Restrainment of gland activity appears to be mainly via the neural pathway. Isolated corpora allata were not influenced by 10?5 M juvenile hormone III added to the incubation medium in vitro.Studies with farnesenic acid revealed that the final two enzymatic steps in the biosynthetic pathway of juvenile hormone are also diminished during prolonged neural inhibition of the corpora allata.20-Hydroxyecdysone and precocene II had no apparent effect on the corpus allatum activity of Leptinotarsa decemlineata.  相似文献

It is known that the low temperature is the most important factor inducing the pre-pupal diapause in Trichogramma species. The position of the thermosensitive period over the life cycle and temporal variation of the degree of responsiveness were investigated in T. embryophagum Htg. by transferring pre-imaginal stages between 'neutral' temperature of 15°C and 'diapause-inducing' temperature of 10°C. Our experiments showed that 6 days long exposure at 10°C significantly increased the percentage of diapausing pre-pupae when started during rather large part of development: from embryo up to early pre-pupa. The highest thermosensitivity was recorded during the embryo and the larval stages, with some decrease during the hatching period. Treatments with shorter cold exposures (2–3 days) gave similar results. Even 24 h long exposure at 10°C increased the percentage of diapausing pre-pupae when applied during egg or early larval stage. Being started at the same stage of development, longer cold exposures caused stronger increase in the percentage of diapausing individuals. The experiments did not reveal any significant daily changes in thermosensitivity: at 12 : 12 h light : dark, larvae subjected to the low temperature during six photophases showed practically the same percentage of diapausing individuals as those subjected to the low temperature during six scotophases, and as those subjected to the 3 days long uninterrupted cold exposure. Hence, in natural conditions even occasional short-term cold periods could be accumulated.  相似文献

Abstract Bruchidius atrolineatus (Pic) is a common pest infesting cowpea seeds ( Vigna unguiculata Walp) in Africa. In the Sahelian zone, the adults are in reproductive diapause during the dry season. Diapause induction depends on the climatic conditions during embryonic and post-embryonic development. A strain of B. atrolineatus originating from the Niamey (13oN) was reared in different thermoperiodic and photoperiodic conditions. In the thermoperiodic regime 40:25oC, in continuous darkness, induction of diapause was dependent on the duration of the thermophase. Photoperiod could also influence diapause induction but the response depended on the temperature. When the beetles were reared in conditions of LD 12:12 h, 40:25oC, for seven generations, the proportion of diapausing adults decreased and their sensitivity to photoperiod increased. In LD 14:10 h, 40:25CC, the proportion of diapausing adults remained high regardless of the photoperiod. In these conditions, the first emerging adults were sexually active and were used for selection of a strain with a low incidence of diapause. After twelve generations, the proportion of diapausing beetles was lower than 5%. The responses of this selected strain to photo- and thermoperiods were reduced. The sensitivity of B. atrolineatus to photoperiod and thermoperiod could be controlled by genetic systems as observed in other insect species.  相似文献

In this study the effects of 23 carbohydrates belonging to various groups upon the survival egg production and egg of female adult Pimpla turionellae L. were investigated. The best results among the carbohydrates tested was obtained with sucrose which was also employed as control. Glucose, maltose, trehalose and melezitose on the other hand showed no significant effect. The egg production was observed to be unaffected by glucose and maltose although it showed significant increase with trehalose and significant decrease with melezitose. Fructose and sorbitol caused a significant decrease in the survival of the insect. Fructose and sorbitol did not have any significant effect upon egg production whereas galactose caused a significant decrease. With the exception of galactose, no carbohydrate caused any significant effect upon egg hatching. Although they did not produce any eggs the female insects survived for 13.17, 15.13, 11.58 and 15.83 days in mannose, melibiose, raffinose and mannitol, respectively. The shortest life span was observed in arabinose followed by α-methyl- D -glucoside, dulcitol, rhamnose, cellobiose, xylose, starch, lactose, sarbose, ribose and glycogen.  相似文献

为从生理生化水平上探讨二化螟滞育幼虫应对温度胁迫的生理机制,分别对系列温度胁迫(STS)和梯度温度胁迫(GTS)处理后的幼虫水、脂质、总糖、小分子碳水化合物含量及超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性进行了测定.结果表明:随着温度的降低,两种处理二化螟滞育幼虫虫体含水量趋于减少,且0℃以下时GTS处理下降幅度较STS处理显著;两种处理脂质含量均逐步下降且二者间无显著差异;两种处理总糖含量分别先减后增和持续下降,均检测出4种小分子碳水化合物,其中STS处理葡萄糖、甘油和果糖含量先增后减,海藻糖含量变化与此相反,而GTS处理海藻糖含量先减后增,葡萄糖和甘油含量呈相反变化,果糖含量无变化;14~-14℃范围内STS处理SOD和POD活性较GTS处理低,CAT活性相反.二化螟滞育幼虫生理指标的变化反映了其应对不同温度胁迫的生理响应.  相似文献
