The hypothesis that luteinizing hormone (LH) secretion in prepubertal females is responsive to estradiol negative feedback and that decreased feedback occurs as puberty approaches was tested in heifers. In the first experiment, seven heifers were maintained prepubertal by dietary energy restriction until 508 days of age (Day 0). All heifers were placed on a high-energy diet on Day 0 at which time they received no additional treatment (CONT), were ovariectomized (OVX) or were ovariectomized and subcutaneously implanted with estradiol-17 beta (OVX-E2). This feeding regimen was used to synchronize reproductive state in all heifers. A second experiment was performed with 16 prepubertal heifers using the same treatments at 266 days (Day 0) of age (CONT, OVX and OVX-E2) but no dietary intake manipulation. In both experiments, LH secretion increased rapidly following ovariectomy in OVX heifers. In the initial experiment, LH secretion was maintained at a low level in OVX-E2 heifers until a synchronous rapid increase was noted coincidental with puberty in the CONT heifer. In the second experiment, LH secretion increased gradually in OVX-E2 heifers and attained castrate levels coincidental with puberty in CONT heifers. A gradual increase in LH secretion occurred as puberty approached in CONT heifers. These results indicate that: a) LH secretion in prepubertal heifers is responsive to estradiol negative feedback; and b) estradiol negative feedback decreases during the prepubertal period in beef heifers.  相似文献

In practice cattle may be slaughtered at different combinations of age and weight. As each of these factors could affect meat quality traits, the present work aimed to identify which combination can be expected to increase overall meat quality of m. rectus abdominis of Charolais heifers. Totally, 40 heifers were slaughtered either at 26±1 or at 36±1 months of age. Young heifers were sampled at two different carcass weights (349±12 and 394±8 kg). Old heifers were also sampled at two different carcass weights (397±6 and 451±9 kg). The m. rectus abdominis was excised 24 h postmortem to determine metabolic enzyme activities, myosin heavy-chain isoform proportions, lipid contents, collagen content and collagen solubility. Shear force measurements were evaluated on raw and broiled meat after 14 days of ageing. Meat quality traits scored between 0 and 10 by sensory analysis. Increasing slaughter age from 26 to 36 months had no impact on either raw/broiled shear force (0.31⩽P⩽0.47) and/or meat quality traits (0.62⩽P⩽0.91) or on physicochemical properties of heifer’s meat samples. Increasing carcass weight for a similar slaughter age of 26 months had also impact neither on meat quality traits (0.52⩽P⩽0.91) nor on muscular properties. On the contrary, increasing carcass weight for a similar slaughter age of 36 months had induced a decrease of muscular shear force (raw muscle; P=0.009) and a concomitant decrease of total collagen content (P=0.03). Nevertheless, no significant impact on meat quality traits was revealed by the sensorial panel (0.13⩽P⩽0.49). Metabolic enzyme activities (0.13⩽P⩽0.86) and myosin heavy-chain proportions (0.13⩽P⩽0.96) were not significantly impacted by slaughter age and carcass weight. Thus, the impact of increasing carcass weight and/or slaughter age in young Charolais heifers has a limited impact on meat quality traits and associated muscular characteristics. Modulating heifer’s cycles (age and/or carcass weight in the studied range) appears to be a way to answer to the numerous marketing chains, without penalising meat quality traits.  相似文献

The effect of ovariectomy on the quality and quantity of cancellous bone using the young cynomolgus monkey was evaluated after a 2-year period. The bodies of the second lumbar vertebrae were analyzed for changes in bone mineral quality using density fractionation, chemical analysis, and X-ray diffraction techniques. Changes in bone tissue quality and quantity were evaluated using bone histomorphometry and image analysis. The experimental group (n=14) was made surgically menopausal (bilaterally ovariectomized), compared with intact controls (n=16), and then sacrificed after a 2-year period. There was a non-significant shift in the mineralization profile towards less dense bone in the ovariectomized (OVX) vertebrae compared with controls. Physical characteristics of the bone mineral in terms of crystal size or strain were unaffected by OVX. There was a parallel increase in mineral content with fractions of increasing density, however there was no difference in mineral content or the Ca/P ratio in each fraction between treatment groups. Histomorphometric analysis for structural parameters demonstrated no difference in bone volume between control and OVX groups. There was no significant change in trabecular width in the OVX vertebrae compared with controls. There was a significant increase in both osteoid volume and osteoid surface in the OVX vertebrae (P<0.001). Trabecular architecture as measured by image analysis was unchanged. There was a significant increase in eroded surface in the OVX vertebrae (P<0.03) compared with the controls. In conclusion, young, ovariectomized female cynomolgus macaques do not appear to be a useful animal model for the study of postmenopausal bone loss, however they may be a useful model to evaluate skeletal pathology which might be observed after surgical ovariectomy in young human females.  相似文献

Eleven heifers, between 63 and 197 days of age, were exposed to 18 hr light/day (L) or natural photoperiods (N), beginning October 19, 1979. They were ovariectomized 8 weeks later. LH concentrations after ovariectomy were not affected by photoperiod, but the rate of increase of FSH after ovariectomy was greater (P<0.10) for group L than for group N. Three weeks after ovariectomy, heiters were injected, IV, with 0.1 mug/kg estradiol-17beta. LH concentrations initially decreased after injection. This was followed by a series of pulses larger than those prior to injection. FSH concentrations declined after injection and remained low throughout the sampling period. The net response of LH concentrations to estradiol (mean post-injection concentration minus mean pre-injection concentration) was greater (P=0.05) for group L (4.7 +/- 0.49 ng/ml) than for group N (2.9 +/- 0.37 ng/ml). Photoperiod did not affect the net response of FSH concentrations to estradiol. We concluded that exposing prepubertal heifers to 18 hr light/day during the winter resulted in a greater rate of increase of FSH after ovariectomy and greater estrogen-induced LH release. Because the response of LH to estradiol-17beta differed from the response of FSH, these hormones may be regulated differently.  相似文献

Plasma luteinizing hormone (LH) levels were determined at monthly intervals in intact and ovariectomized squirrels maintained in a constant 14L:10D photoperiod at a temperature of 23 +/- 2 degrees C. LH was undetectable (less than 0.9 ng/ml) in plasma of intact females at all times of year. Females ovariectomized (OVX) at 9.5 months of age in March showed substantial increases in plasma LH in May and June but LH was undetectable between July and November. Females ovariectomized at 13 months of age in July first manifested detectable LH levels the following January and February (6-7 months post-ovariectomy). Very few adult females trapped in May and ovariectomized in August had detectable LH levels within 2 months of ovariectomy; however, females ovariectomized the following February had detectable LH titers 1 month later. Long-term studies of individual OVX squirrels indicated peak LH levels between March and June, 1980, relatively low or undetectable titers between August and December and elevated LH levels between January and March, 1981. The results are suggestive of a circannual rhythm of LH secretion which appears restricted to one season of the year and occurs independently of steroid feedback from the ovaries; ovarian steroids only modulate the levels of plasma LH during the brief annual period of hypothalamo-hypophysial activity. We suggest that onset and termination of LH release are mediated by central nervous system circannual clocks.  相似文献

With the use of Helmholtz coils and pulsed electromagnetic field (PEMF) stimulators to generate uniform time varying electromagnetic fields, the effects of extremely low frequency electromagnetic fields on osteoporosis and serum prostaglandin E(2) (PGE(2)) concentration were investigated in bilaterally ovariectomized rats. Thirty-five 3 month old female Sprague-Dawley rats were randomly divided into five different groups: intact (INT), ovariectomy (OVX), aspirin treated (ASP), PEMF stimulation (PEMF + OVX), and PEMF stimulation with aspirin (PEMF + ASP) groups. All rats were subjected to bilateral ovariectomy except those in INT group. Histomorphometric analyses showed that PEMF stimulation augmented and restored proximal tibial metaphyseal trabecular bone mass (increased hard tissue percentage, bone volume percentage, and trabecular number) and architecture (increased trabecular perimeter, trabecular thickness, and decreased trabecular separation) in both PEMF + OVX and PEMF + ASP. Trabecular bone mass of PEMF + OVX rats after PEMF stimulation for 30 days was restored to levels of age matched INT rats. PEMF exposure also attenuated the higher serum PGE(2) concentrations of OVX rats and restored it to levels of INT rats. These experiments demonstrated that extremely low intensity, low frequency, single pulse electromagnetic fields significantly suppressed the trabecular bone loss and restored the trabecular bone structure in bilateral ovariectomized rats. We, therefore, conclude that PEMF may be useful in the prevention of osteoporosis resulting from ovariectomy and that PGE(2) might relate to these preventive effects.  相似文献

We studied molecular mechanism of Cistanches Herba aqueous extract (CHAE) in ovariectomized (OVX) rats, as an experimental model of postmenopausal osteoporosis. Female rats were either sham-operated or bilaterally OVX; and at 60 days postoperatively. The OVX group (n = 8) received an ovariectomy and treatment with normal saline for 90 days commencing from 20th post ovariectomy day. The ovariectomized +CHAE (OVX + CHAE) group (n = 8) received an ovariectomy and were treated with Cistanches Herba aqueous extract of 100 mg/kg body weight daily for 90 days commencing from 22nd post ovariectomy day. The ovariectomy +CHAE (OVX + CHAE) group (n = 8) received an ovariectomy, and were treated with the of 200 mg/kg body weight daily for 90 days commencing from 20th post ovariectomy day. Serum BGP and TRAP, E2, FSH and LH level, bone marrow Smad1, Smad5, TGF-β1 and TIEG1 mRNA expression levels were examined. Results showed that serum BGP and TRAP, FSH and LH levels were significantly increased, whereas E2, Smad1, Smad5, TGF-β1 and TIEG1 mRNA and proteins expression levels were significantly decreased in OVX rats compared to sham rats. 90 days of CHAE treatment could significantly decrease serum BGP and TRAP, FSH and LH levels, and increase E2, Smad1, Smad5, TGF-β1 and TIEG1 mRNA and proteins expression levels in OVX rats. It can be concluded that CHAE play its protective effect against OVX-induced bone degeneration partly by regulating some bone metabolism related genes, e.g. Smad1, Smad5, TGF-β1 and TIEG1.  相似文献

Plasma FSH and LH response to a synthetic GnRH analog was measured in adult ovariectomized pony mares (OVX) and in pony foals (<70 days of age) during late spring (May-June). FSH and LH responded in a similar fashion (200% increase) in the OVX mare, which is different from other reports for intact mares. There was a greater mean response to a comparable dose of GnRH in the prepubertal foal for both FSH (500%) and LH (900%) than in the OVX mare. There was a positive correlation between age and the maximum FSH response to GnRH in male and female foals. The LH response was positively correlated with age in male foals, but not in females. The response to GnRH in the prepubertal foals was consistent with the previously observed patterns of gonadotropin secretion during this age period.  相似文献

This experiment was designed to determine the age at which estradiol-17beta (E(2)) first induces a preovulatory-like surge of luteinizing hormone (LH) in prepubertal heifers. Responses of prepubertal animals 3 to 4 and 5 to 6 months of age were compared with those of postpubertal heifers that received 25 mg prostaglandin F(2)alpha at 0800 hr on day 15 of the estrous cycle. E(2) (500mug) induced surges of LH in 1 5 heifers 3 to 4 months of age, 3 3 heifers 5 to 6 months of age and 5 5 postpubertal heifers. Duration of response and interval between E(2) injection and peak of the response were longer in postpubertal heifers than in those 5 to 6 months old (P<0.10). Peak response and total amount of LH released were greater in animals 5 to 6 months old (P<0.10). Only one prepubertal heifer had elevated concentrations of progesterone following an LH surge. Four of 5 postpubertal heifers receiving E(2) and 3 of 4 postpubertal heifers receiving corn oil had corpora lutea and similar patterns of progesterone concentrations. We conclude that ability to release an LH surge in response to E(2) develops in heifers between 3 and 5 months of age, but that this induced surge does not cause ovulation.  相似文献

Taking into account the sexual dimorphism previously reported regarding mitochondrial function and biogenesis in brown adipose tissue, the aim of the present study was to go further into these differences by investigating the effect of ovariectomy and 17-β estradiol (E2) replacement on brown adipose tissue mitochondrial function. In this study, fourteen-week-old control female and ovariectomized female Wistar rats were used. Rats were ovariectomized at 5 weeks of age and were treated every 2 days with placebo (OVX group) or E2 (10 μg/kg) (OVX+E2 group) for 4 weeks before sacrifice. We studied the levels of oxidative capacity, antioxidant defence and oxidative damage markers in brown adipose tissue. Moreover, the levels of key elements of mitochondrial biogenesis as well as UCP1 protein levels, as an index of mitochondrial thermogenic capacity, were also determined. In response to ovariectomy, mitochondrial proliferation increased, resulting in less functional mitochondria, since oxidative capacity and antioxidant defences decreased. Although E2 supplementation was able to restore the serum levels of E2 shown by control rats, the treatment reverted the effects of the ovariectomy only in part, and oxidative and antioxidant capacities in OVX+E2 rats did not reach the levels shown by control females. Taking these results into account, we suggest that ovarian hormones are responsible, at least in part, for the sexual dimorphism in BAT mitochondrial function. However, other signals produced by ovary, rather than E2, would play an important role in the control of mitochondrial function in BAT.  相似文献

检测间隙连接蛋白Cx43、神经组织蛋白S-100在去卵巢致骨质疏松症(OVX-OP)大鼠腺垂体滤泡星形细胞(FS细胞)中的表达.实验采用10月龄未孕产SD雌性大鼠40只,随机均分为卵巢切除组(OVX组,n=20)和假性手术对照组(Sham组,n=20).于术后6周末用双能X线骨吸收测量法(DEXA)测量大鼠全身及腰椎4-6(L4-6)骨密度(BMD).取两组大鼠垂体,制成连续切片.应用FITC标记的IgG探针,对腺垂体组织中Cx43和S-100进行间接免疫荧光染色,并用激光扫描共聚焦显微镜(LSCM)定位和定量分析腺垂体FS细胞中Cx43、S-100的表达.结果发现,术后6周末OVX组大鼠全身及L4-6BMD均明显低于Sham组值(P<0.01,P<0.01).Cx43阳性荧光反应主要定位于相邻的FS细胞的胞浆中和/或胞膜上.OVX组Cx43阳性表达荧光强度和表达阳性率均显著低于Sham组(P<0.01).S-100蛋白表达定位于FS细胞的胞浆中,两组间S-100阳性表达荧光强度和表达阳性率无显著差异(P>0.05).本研究提示,SD大鼠OVX术后6周出现骨质疏松变化;OVX大鼠腺垂体FS细胞数量无明显变化、而Cx43表达显著下降,后者的变化可能与大鼠OVX-OP发生相关.  相似文献

An experiment was done to determine if month of birth and age influenced patterns of luteinizing hormone (LH) secretion in prepubertal heifers. Mean concentrations of LH increased linearly (P < .05) in March-born heifers between one and seven months of age. This was partially due to an increase in number of LH pulses. The prepubertal pattern of LH concentrations was quadratic (P < .05) for heifers born in September because concentrations were slightly higher (P = .15) than those in March-born heifers at one month of age. There were no differences between groups during the remainder of the prepubertal period (3 to 7 months). There was a tendency (P = .18) for September-born animals to reach puberty at younger ages than those born in March. September-born heifers also had greater (P = .06) average daily gains, but body weights at puberty were similar for the two groups. These results show that season of birth influenced LH concentrations at one month of age, but did not significantly affect the increase between three and seven months of age.  相似文献

This study was performed to evaluate the effect of concomitant supplementation of genistein and silicon on bone mineral density and bone metabolism-related markers in ovariectomized rat. Three-month-old Sprague Dawley female rats were subjected to bilateral ovariectomy (OVX) or sham surgery, and then the OVX rats were randomly divided into four groups: OVX-GEN, OVX-Si, OVX-GEN-Si, and OVX. Genistein and silicon supplementation was started immediately after OVX and continued for 10 weeks. In the OVX-GEN group, 5 mg genistein per gram body weight was injected subcutaneously. The OVX-Si group was given soluble silicon daily in demineralized water (Si 20 mg/kg body weight/day). The OVX-GEN-Si group was given subcutaneous injections of 5 mg genistein per gram body weight, at the same time, given soluble silicon daily (Si 20 mg/kg body weight/day). The results showed that the genistein supplementation in the OVX rats significantly prevented the loss of uterus weight; however, the silicon supplementation showed no effect on the uterus weight loss. The lumbar spine and femur bone mineral density was significantly decreased after OVX surgery; however, this decrease was inhibited by the genistein and/or silicon, and the BMD of the lumbar spine and femur was the highest in the OVX-GEN-Si-treated group. Histomorphometric analyses showed that the supplementation of genistein and/or silicon restored bone volume and trabecular thickness of femoral trabecular bone in the OVX group. Besides, the treatment with genistein and silicon for 10 weeks increased the serum levels of calcium and phosphorus in the OVX rats; serum calcium and serum phosphorus in the OVX-GEN-Si group were higher than those in the OVX-GEN and OVX-Si group (P < 0.05). At the same time, the treatment with genistein and/or silicon decreased serum alkaline phosphatase (ALP) and osteocalcin, which were increased by ovariectomy; serum ALP and osteocalcin in the OVX-GEN-Si group were lower than those in the OVX-GEN and OVX-Si groups (P < 0.05). The results above indicate that genistein and silicon have synergistic effects on bone formation in ovariectomized rats.  相似文献

Our primary objective for this study was to determine whether steady-state amounts of alpha- and LH beta-subunit mRNAs in the anterior pituitary are altered during sexual maturation in the bovine female. A secondary objective was to determine whether 17 beta-estradiol (E2) alters amounts of LH subunit mRNAs before onset of puberty. Heifers (7 mo old) were assigned to one of three treatments: 1) ovariectomized (OVX, n = 16); 2) OVX and administered E2 (OVXE, n = 16); or 3) ovary-intact (INTACT, n = 20). Pituitaries were collected at an estimated 120 days before onset of puberty (prepuberty) or 25 days before onset of puberty (peripuberty). Six INTACT heifers were used to determine time of puberty during the experimental period, and their pituitaries were collected 40 h after administration of prostaglandin F2 alpha (postpubertal INTACT group). Relative amounts of mRNAs for LH subunits in each pituitary were determined by Northern analysis and scanning densitometry. Amounts of alpha- and LH beta-subunit mRNAs were lower in pituitaries of INTACT heifers and OVXE heifers, regardless of stage of sexual maturation, than in those of OVX heifers. Amounts of alpha-subunit mRNA were similar in OVXE and INTACT heifers regardless of stage of sexual maturation. Amounts of LH beta-subunit mRNA did not change during sexual maturation in heifers in the INTACT group. Our working hypotheses for this study were that 1) the profile of intrapituitary LH and FSH isoforms would be shifted toward acidic forms as sexual maturation progresses in the bovine female; and 2) concentration of 17 beta-estradiol (E2) in circulation during sexual maturation would be a major factor modulating the percentage of the more acidic isoforms. In addition, the biological-immunoreactive (B:I) ratios of each isoform of LH were evaluated at selected stages of sexual maturation. Heifers (7 mo of age) were assigned to one of three treatment groups: 1) ovariectomized (OVX; n = 16); 2) OVX and administered E2 (OVXE; n = 16); or 3) ovary-intact (INTACT; n = 14). Pituitaries were collected from heifers in each group at an estimated 120 days (prepubertal) of 25 days before puberty (peripubertal). A fourth group of 6 heifers remained intact (postpubertal INTACT) to determine time of puberty during the experimental period. Pituitaries of heifers assigned to the postpubertal INTACT group were collected during the follicular phase of the first or second estrous cycle postpuberty. Pituitaries were used for determination of relative amounts of gonadotropin isohormones. Tissue extracts of the pituitaries were chromatofocused on pH 10.5-4.0 gradients. The LH of all pituitaries resolved into thirteen isoforms that were designated isoforms A-L, and S, with isoform A the most basic form. Isoforms F and G (basic pH range) were the predominant isoforms of each chromatofocusing profile and comprised 50-60% of the immunoreactive LH. The aim of the present study was to investigate the cardiac biomarker changes in experimental bilateral ovariectomized (OVX) rats in addition to evaluating the role of soybean oil-supplemented diet to attenuate these alterations. Female rats were divided into four groups and treated for 2 months as follows: groups 1 and 2 fed with standard diet with or without 15% soybean oil. Groups 3 and 4 were bilateral OVX and received the standard diet with or without 15% soybean oil. The results revealed that rats subjected to ovariectomy exhibited an inhibition in estrogen and high-density lipoprotein cholesterol levels and marked increase of lipid profile, low-density lipoprotein cholesterol, and VLDL-C accompanied with a marked elevation in atherogenic index, cardiac enzyme activity, tumor necrosis factor-α, and C-reactive protein. Signs of cardiovascular injury which included an increase in cardiac thiobarbituric acid-reactive substances were concomitantly noticed with a reduction in the reduced glutathione, total antioxidant capacity, and superoxide dismutase. However, supplementation of soybean oil resulted in the restoration of the changed lipid profile and improved cardiac biomarkers near to normal values as well as improved inflammatory and antioxidant status. It was concluded that consumption of soybean oil may have a role in retarding atherosclerosis and risk of cardiovascular disorders associated with estrogen deficiency in ovariectomy status.  相似文献

Effect of estrogen and placental lactogen on lactogenesis in pregnant rats   总被引:1,自引:0,他引:1  
The removal of the corpora lutea or ovariectomy on Day 18 of pregnancy induced a rise in serum prolactin 24 h after surgery with a rapid decline to control values 4 h after the surge, only in the ovariectomized group. When hysterectomy was performed in addition to luteectomy or ovariectomy a similar rise in prolactin was obtained. Lactose synthetase activity in mammary tissue was significantly higher in the luteectomized and luteectohysterectomized rats when compared with ovariectomized, ovariohysterectomized rats and the sham-operated group. Estrogen treatment 12 h after ovariectomy increased serum prolactin and lactose synthetase activity to values similar to those measured in luteectomized rats, but this increase was significantly greater when compared with the ovariectomized-nontreated group. Treatment with Tamoxifen did not decrease serum prolactin in the luteectomized rats but lactose synthetase was reduced to values similar to that obtained in ovariectomized rats. Treatment with 2 bromo-alpha-ergocryptine-mesylate (CB-154) prevented the rise in serum prolactin in the ovariectomized, luteectomized and luteectohysterectomized groups, but lactose synthetase activity was lowered to control values (sham-operated rats) only in the luteectohysterectomized rats. According to these findings, rat placental lactogen in the absence of prolactin and progesterone induces lactose synthesis. Estrogen facilitates prolactin but not placental lactogen action on lactose synthetase activity.  相似文献

The influence of chronic administration of 5-HT1A receptor agonist 8-OH-DPAT (0.05 mg/kg, s.c.) and 5-HT1A receptor antagonist NAN-190 (0.1 mg/kg, i.p.) injected for 14 days alone or in combination with 17beta-estradiol (0.5 microg i.m./rat/day) was studied on passive avoidance performance (PAR) and on behavior in the open field test in adult intact and ovariectomized (OVX) female rats. Administration of 5-HT1A receptor antagonist NAN-190 alone significantly improved PAR (p<0.05) in intact females with proestrus and estrus and in OVX females. Administration of 5-HT1A receptor agonist 8-OH-DPAT alone or in combination with 17beta-estradiol significantly (p<0.05) improved PAR in OVX rats and failed to normalize PAR in intact rats with proestrus and estrus. Results of the work specify the involvement of 5-HT1A receptors in the mechanisms of passive avoidance learning in OVX female rats.  相似文献

Beginning 15 days after ovariectomy (OVX), a high mammary tumor strain of SHN virgin mice at 3 months of age received subcutaneous injections of danazol (0.5 mug / 0.1 ml olive oil, once a day), perphenazine (0.05 mg / 0.1 ml saline, twice a day) or ovine prolactin (oPRL: 0.25 mg / 0.05 ml buffer, twice a day) for 3 days to modulate their circulating PRL levels. The serum PRL level was significantly decreased by danazol and increased by perphenazine compared to the intact and OVX-control groups. The expression of both transforming growth factor alpha (TGFalpha) mRNA and epidermal growth factor receptor (EGFR) mRNA in the mammary gland was increased by danazol. However, TGFalpha mRNA expression was decreased by perphenazine. Meanwhile, mammary end-bud formation was inhibited in danazol-treated group. All findings suggest that the manifestation of the effect of TGFalpha on mammary gland is rather suppressed by PRL, while mammary gland growth needs the participation of PRL; in other words, PRL is dominant to TGFalpha on the mammary gland growth. OVX resulted in a significant decrease of TGFalpha mRNA expression in the mammary gland despite of little alteration in serum PRL, confirming the previous observations. The similar trend was observed in ICR mice; however, the response to hormonal modulation is generally less susceptible than SHN mice.  相似文献
