Resource limitation during the juvenile stages frequently results in developmental delays and reduced size at maturity, and dietary restriction during adulthood can affect longevity and reproductive output. Variation in food intake can also result in alteration to the normal pattern of resource allocation among body parts or life-history stages. My primary aim in this study was to determine how varying juvenile and/or adult feeding regimes affect particular female and male traits in the sexually cannibalistic praying mantid Pseudomantis albofimbriata. Praying mantids are sit-and-wait predators whose resource intake can vary dramatically depending on environmental conditions within and across seasons, making them useful for studying the effects of feeding regime on various facets of reproductive fitness. In this study, there was a significant trend/difference in development and morphology for males and females as a result of juvenile feeding treatment, however, its effect on the fitness components measured for males was much greater than on those measured for females. Food-limited males were less likely to find a female during field enclosure experiments and smaller males were slower at finding a female in field-based experiments, providing some of the first empirical evidence of a large male size advantage for scrambling males. Only adult food limitation affected female fecundity, and the ability of a female to chemically attract males was also most notably affected by adult feeding regime (although juvenile food limitation did play a role). Furthermore, the significant difference/trend in all male traits and the lack of difference in male trait ratios between treatments suggests a proportional distribution of resources and, therefore, no trait conservation by food-limited males. This study provides evidence that males and females are under different selective pressures with respect to resource acquisition and is also one of very few to show an effect of juvenile food quantity on adult reproductive fitness in a hemimetabolous insect. 相似文献

Auxin is a pivotal plant hormone, usually occurring in the form of indole-3-acetic acid (IAA). However, in maturing pea (Pisum
sativum) seeds, the level of the chlorinated auxin, 4-chloroindole-3-acetic acid (4-Cl-IAA), greatly exceeds that of IAA. A key issue is how plants produce halogenated compounds such as 4-Cl-IAA. To better understand this topic, we investigated the distribution of the chlorinated auxin. We show for the first time, to our knowledge, that 4-Cl-IAA is found in the seeds of Medicago truncatula, Melilotus indicus, and three species of Trifolium. Furthermore, we found no evidence that Pinus spp. synthesize 4-Cl-IAA in seeds, contrary to a previous report. The evidence indicates a single evolutionary origin of 4-Cl-IAA synthesis in the Fabaceae, which may provide an ideal model system to further investigate the action and activity of halogenating enzymes in plants.The chlorinated form of auxin, 4-chloroindole-3-acetic acid (4-Cl-IAA), is a highly active hormone that is thought to play a key role in early pericarp growth (Reinecke et al., 1995, 1999; Ozga et al., 2009). Exogenous 4-Cl-IAA, for example, has been shown to promote the pericarp elongation of deseeded pea (Pisum sativum) pods (Reinecke et al., 1999). Johnstone et al. (2005) reported that 4-Cl-IAA and bioactive GA (GA3 or GA1) act synergistically on pericarp growth when applied simultaneously, and a growth regulatory role has been proposed for 4-Cl-IAA through induction of GA biosynthesis and inhibition of ethylene action. In other species, e.g. tomato (Solanum lycopersicum), the nonchlorinated form of auxin, indole-3-acetic acid (IAA), also stimulates fruit growth via GAs (Serrani et al., 2008; Tang et al., 2015). The chlorinated auxin is mainly found in reproductive structures (Katayama et al., 1988), in which its levels often exceed those of the more widespread IAA (Tivendale et al., 2012). The chlorinated form is thought to be restricted to members of the leguminous tribe Fabeae (Reinecke 1999), which includes the genera Vicia, Pisum, Lathyrus, Lens, and Vavilovia (Schaefer et al., 2012). However, there is a curious exception: 4-Cl-IAA has been reported also from Scots pine (Pinus sylvestris; Ernstsen and Sandberg, 1986).We previously published evidence that most 4-Cl-IAA in maturing pea seeds is synthesized from 4-Cl-tryptophan (4-Cl-Trp) via 4-Cl-indole-3-pyruvic acid (Tivendale et al., 2012, 2014). 4-Cl-Trp has been identified in extracts from pea and broad bean (Vicia faba) seeds (Kettner et al., 1992; Manabe et al., 1999), but whether the precursors of Trp can be chlorinated is unknown.Virtually nothing is known about the enzymes that catalyze halogenation reactions in plants. In bacteria, fungi, and marine algae, there are six types of enzymes responsible for the addition of halogen atoms to organic molecules. These include heme haloperoxidases, vanadium-dependent haloperoxidases, mononuclear nonheme iron halogenases, flavin-dependent halogenases, S-adenosyl-l-Met-dependent chlorinases and fluorinases, and methyl halide transferases (Butler and Sandy, 2009; Wagner et al., 2009). However, in the genomes of angiosperms, the only type of halogenating enzyme that has been annotated are haloperoxidases, but very little is known about these enzymes. To further understand the activity and action of halogenating enzymes in plants, a comparative system is required.In this study, we investigated the distribution of 4-Cl-IAA and 4-Cl-Trp in the Fabaceae by monitoring these compounds in the seeds of representative species spanning the phylogeny of this family. Most of these species have not been previously tested for the presence of the chlorinated compounds. In addition, we reexamined the reported occurrence of 4-Cl-IAA outside the Fabaceae, namely in Scots pine; several other Pinus species were investigated here as well. We also examined the endogenous levels of 4-Cl-IAA in both vegetative tissues and seeds of broad bean to address the question of whether 4-Cl-IAA is largely restricted to seeds (Pless et al., 1984; Katayama et al., 1988). 相似文献

Adaptive flexibility in response to environmental variation is often advantageous and occurs in many types of traits in many species. Although the basic designs of the orb webs of a given species are relatively uniform, spiders can adjust their webs to some types of environmental variation. This study of adult female Leucauge argyra tests the extremes to which they can adjust with respect to reduced area in which to build, and documents probably the most pronounced flexibility in orb design ever recorded. These adjustments revealed several behavioral rules that guide orb construction behavior. Spiders adjusted at least seven probably independent aspects of orb design when confined in tiny spaces that spanned about 7% of the maximum distance normally spanned by webs in the field and that had diameters that were only about three times the length of the spider itself. Webs in intermediate sized containers had intermediate designs, and many of the adjustments appear to result from extensions of the behavioral rules guiding orb construction in less severely restricted spaces in the field. 相似文献

Although studies on sperm competition examined a wide range of taxa, little is known about the selection pressures on male traits in systems with simultaneous risk of sperm competition and sexual cannibalism. Here, we experimentally studied how the risk of sperm competition affects male copulatory behavior in the sexually cannibalistic praying mantis Mantis religiosa. We recorded the onset and duration of copulations following the introduction of virgin, adult praying mantises into mating arenas with three different sex ratio treatments: polyandrous, monogamous, and polygynous. We did not detect any female phenotypic trait predicting cannibalism. The chance of male survival was related to his condition, with males in better condition being cannibalized significantly less often. In contrast, we did not identify any male trait that would favor some males to obtain copulations. Our results on copulation duration support sperm competition theory in that the copulations in the male‐biased treatment, where the perceived risk of sperm competition was greatest, were significantly longer than those in single‐male treatments. Importantly, males in better condition copulated significantly longer regardless of sex ratio. Overall, our study suggests that males can adjust their copulation behavior in response to the risk of sperm competition even in a system with frequent sexual cannibalism. 相似文献

Previous observations that aquaporin overexpression increases the freeze tolerance of baker's yeast (Saccharomyces cerevisiae) without negatively affecting the growth or fermentation characteristics held promise for the development of commercial baker's yeast strains used in frozen dough applications. In this study we found that overexpression of the aquaporin-encoding genes AQY1-1 and AQY2-1 improves the freeze tolerance of industrial strain AT25, but only in small doughs under laboratory conditions and not in large doughs under industrial conditions. We found that the difference in the freezing rate is apparently responsible for the difference in the results. We tested six different cooling rates and found that at high cooling rates aquaporin overexpression significantly improved the survival of yeast cells, while at low cooling rates there was no significant effect. Differences in the cultivation conditions and in the thawing rate did not influence the freeze tolerance under the conditions tested. Survival after freezing is determined mainly by two factors, cellular dehydration and intracellular ice crystal formation, which depend in an inverse manner on the cooling velocity. In accordance with this so-called two-factor hypothesis of freezing injury, we suggest that water permeability is limiting, and therefore that aquaporin function is advantageous, only under rapid freezing conditions. If this hypothesis is correct, then aquaporin overexpression is not expected to affect the leavening capacity of yeast cells in large, industrial frozen doughs, which do not freeze rapidly. Our results imply that aquaporin-overexpressing strains have less potential for use in frozen doughs than originally thought. 相似文献

Natural history collections house an enormous amount of plant and animal specimens, which constitute a promising source for molecular analyses. Storage conditions differ among taxa and can have a dramatic effect on the success of DNA work. Here, we analyze the feasibility of DNA extraction from ethanol preserved spiders (Araneae). We tested genotyping success using several hundred specimens of the wasp spider, Argiope bruennichi, deposited in two large German natural history collections. We tested the influence of different factors on the utility of specimens for genotyping. Our results show that not the specimen’s age, but the museum collection is a major predictor of genotyping success. These results indicate that long term storage conditions should be optimized in natural history museums to assure the utility of collections for DNA work. Using historical material, we also traced historical genetic and morphological variation in the course of a poleward range expansion of A. bruennichi by comparing contemporary and historical specimens from a native and an invasive population in Germany. We show that the invasion of A. bruennichi is tightly correlated with an historical increase of genetic and phenotypic variation in the invasive population. 相似文献

Understanding why certain species can successfully colonize new areas while others do not is a central question in ecology. The nine-banded armadillo (Dasypus novemcinctus) is a conspicuous example of a successful invader, having colonized much of the southern United States in the last 200 years. We used 15 years (1992–2006) of capture-mark-recapture data from a population of armadillos in northern Florida in order to estimate, and examine relationships among, various demographic parameters that may have contributed to this ongoing range expansion. Modeling across a range of values for γ, the probability of juveniles surviving in the population until first capture, we found that population growth rates varied from 0.80 for γ = 0.1, to 1.03 for γ = 1.0. Growth rates approached 1.0 only when γ ≥0.80, a situation that might not occur commonly because of the high rate of disappearance of juveniles. Net reproductive rate increased linearly with γ, but life expectancy (estimated at 3 years) was independent of γ. We also found that growth rates were lower during a 3-year period of hardwood removal that removed preferred habitat than in the years preceding or following. Life-table response experiment (LTRE) analysis indicated the decrease in growth rate during logging was primarily due to changes in survival rates of adults. Likewise, elasticity analyses of both deterministic and stochastic population growth rates revealed that survival parameters were more influential on population growth than were those related to reproduction. Collectively, our results are consistent with recent theories regarding biological invasions which posit that populations no longer at the leading edge of range expansion do not exhibit strong positive growth rates, and that high reproductive output is less critical in predicting the likelihood of successful invasion than are life-history strategies that emphasize allocation of resources to future, as opposed to current, reproduction. 相似文献

Across animal species, offspring of closely related mates exhibit lower fitness, a phenomenon called inbreeding depression. Inbreeding depression in humans is less well understood because mating between close relatives is generally rare and stigmatised, confounding investigation of its effect on fitness-relevant traits. Recently, the availability of high-density genotype data has enabled quantification of variation in distant inbreeding in ‘outbred’ human populations, but the low variance of inbreeding detected from genetic data in most outbred populations means large samples are required to test effects, and only a few traits have yet been studied. However, it is likely that isolated populations, or those with a small effective population size, have higher variation in inbreeding and therefore require smaller sample sizes to detect inbreeding effects. With a small effective population size and low immigration, Northern Finland is such a population. We make use of a sample of ∼5,500 ‘unrelated’ individuals in the Northern Finnish Birth Cohort 1966 with known genotypes and measured phenotypes across a range of fitness-relevant physical and psychological traits, including birth length and adult height, body mass index (BMI), waist-to-hip ratio, blood pressure, heart rate, grip strength, educational attainment, income, marital status, handedness, health, and schizotypal features. We find significant associations in the predicted direction between individuals'' inbreeding coefficient (measured by proportion of the genome in runs of homozygosity) and eight of the 18 traits investigated, significantly more than the one or two expected by chance. These results are consistent with inbreeding depression effects on a range of human traits, but further research is needed to replicate and test alternative explanations for these effects. 相似文献

Hexanucleotide (GGGGCC) repeat expansion in C9ORF72 (HRE) causes frontotemporal lobar degeneration, frontotemporal dementia–amyotrophic lateral sclerosis, and amyotrophic lateral sclerosis. Inbreeding is known to have adverse effects on fitness-related traits in a range of insect species. A series of theoretical and experimental studies have suggested that polyandrous insects could avoid the cost of inbreeding via pre-copulatory mate choice and/or post-copulatory mechanisms. We looked for evidence of pre-copulatory inbreeding avoidance using female mate preference trials, in which females were given the choice of mating with either of two males, a sibling and a non-sibling. We also tested for evidence of post-copulatory inbreeding avoidance by conducting double mating experiments, in which four sibling females were mated with two males sequentially, either two siblings, two non-siblings or a sibling and a non-sibling in either order. We identified substantial inbreeding depression: offspring of females mated to full siblings had lower hatching success, slower development time from egg to adult, lower survival of larval and pupal stages, and lower adult body mass than the offspring of females mated to non-sibling males. We also found evidence of pre-copulatory inbreeding avoidance, as females preferred to mate with non-sibling males. However, we did not find any evidence of post-copulatory inbreeding avoidance: egg hatching success of females mating to both sibling and non-sibling males were consistent with sperm being used without bias in relation to mate relatedness. Our results suggest that this cabbage beetle has evolved a pre-copulatory mechanism to avoid matings between close relative, but that polyandry is apparently not an inbreeding avoidance mechanism in C. bowringi. 相似文献

Zebrafish embryonic slow muscle cells, with their superficial localization and clear sarcomere organization, provide a useful model system for genetic analysis of muscle cell differentiation and sarcomere assembly. To develop a quick assay for testing CRISPR-mediated gene editing in slow muscles of zebrafish embryos, we targeted a red fluorescence protein (RFP) reporter gene specifically expressed in slow muscles of myomesin-3-RFP (Myom3-RFP) zebrafish embryos. We demonstrated that microinjection of RFP-sgRNA with Cas9 protein or Cas9 mRNA resulted in a mosaic pattern in loss of RFP expression in slow muscle fibers of the injected zebrafish embryos. To uncover gene functions in sarcomere organization, we targeted two endogenous genes, slow myosin heavy chain-1 (smyhc1) and heat shock protein 90 α1 (hsp90α1), which are specifically expressed in zebrafish muscle cells. We demonstrated that injection of Cas9 protein or mRNA with respective sgRNAs targeted to smyhc1 or hsp90a1 resulted in a mosaic pattern of myosin thick filament disruption in slow myofibers of the injected zebrafish embryos. Moreover, Myom3-RFP expression and M-line localization were also abolished in these defective myofibers. Given that zebrafish embryonic slow muscles are a rapid in vivo system for testing genome editing and uncovering gene functions in muscle cell differentiation, we investigated whether microinjection of Natronobacterium gregoryi Argonaute (NgAgo) system could induce genetic mutations and muscle defects in zebrafish embryos. Single-strand guide DNAs targeted to RFP, Smyhc1, or Hsp90α1 were injected with NgAgo mRNA into Myom3-RFP zebrafish embryos. Myom3-RFP expression was analyzed in the injected embryos. The results showed that, in contrast to the CRISPR/Cas9 system, injection of the NgAgo-gDNA system did not affect Myom3-RFP expression and sarcomere organization in myofibers of the injected embryos. Sequence analysis failed to detect genetic mutations at the target genes. Together, our studies demonstrate that zebrafish embryonic slow muscle is a rapid model for testing gene editing technologies in vivo and uncovering gene functions in muscle cell differentiation. 相似文献

Spatial and temporal variation in ecological parameters selects for plastic responses to prevailing conditions. Males of web spiders have been shown to adjust developmental decisions to cues that relate to the degree of competition from other males. Here, we explore experimentally whether males of the spider Nephila senegalensis base behavioural decisions in the context of mate choice and sperm allocation to the presence of cues of a rival and to their own competitive dominance. Large and small males were offered a simultaneous choice between two penultimate instar females that varied in quality and in whether they were previously visited by a rival male. Large and small males avoided webs visited by a previous male, and this preference overrode differences in female quality. In a second experiment, males were assigned to copulate with a virgin female whose web was either previously visited by a rival male or not. We found substantial variation in male sperm allocation, but this variation was not related to our treatment. Our experiments demonstrate that males notice silk cues left behind by rival males on a female’s web and that they can utilize these cues in relation to decisions over whether to visit a web, but not in relation to sperm allocation. 相似文献

Polysaccharide responsiveness is tested by measuring antibody responses to polysaccharide vaccines to diagnose for humoral immunodeficiency. A common assumption is that this responsiveness is biased by any previous exposure to the polysaccharides in the form of protein-coupled polysaccharide vaccines, such as those used in many childhood vaccination programmes.To examine this assumption, we investigated the effect of protein-coupled polysaccharide vaccination on subsequent polysaccharide responsiveness.HIV-infected adults (n = 47) were vaccinated twice with protein-coupled polysaccharides and six months later with pure polysaccharides. We measured immunoglobulin G responses against three polysaccharides present in only the polysaccharide vaccine (non-memory polysaccharides) and seven recurring polysaccharides (memory polysaccharides). Responsiveness was evaluated according to the consensus guidelines published by the American immunology societies.Impaired responsiveness to non-memory polysaccharides was more frequent than to memory polysaccharides (51% versus 28%, P = 0.015), but the individual polysaccharides did not differ in triggering sufficient responses (74% versus 77%, P = 0.53). Closer analysis revealed important shortcomings of the current evaluation guidelines. The interpreted responseś number and their specificities influenced the likelihood of impaired responsiveness in a complex manor. This influence was propelled by the dichotomous approaches inherent to the American guidelines. We therefore define a novel more robust polysaccharide responsiveness measure, the Z-score, which condenses multiple, uniformly weighted responses into one continuous variable. Using the Z-score, responsiveness to non-memory polysaccharides and memory-polysaccharides were found to correlate (R2 = 0.59, P<0.0001).We found that polysaccharide responsiveness was not biased by prior protein-coupled polysaccharide vaccination in HIV-infected adults. The aim of this study was to determine the role played by vigilance on the anaerobic performance recorded during a Wingate test performed at the bathyphase (nadir) of the circadian rhythmicity. Twenty active male participants performed a 60-s Wingate test at 6 a.m. during 3 test sessions in counter-balanced order the day after either (i) a normal reference night, (ii) a total sleep deprivation night, or (iii) a total sleep deprivation night associated with an extended simulated driving task from 9 p.m. to 5 a.m. During this task, the number of inappropriate line crossings (ILCs) was used to control and quantify the effective decrease in the level of vigilance. The main findings show that (i) vigilance of each participant was significantly altered (i.e., a drastic and progressive increase in ILCs is shown during the 7.5 hours of driving) by the sleep deprivation night associated with an extended driving task; (ii) the subjective evaluation of vigilance performed by self-rated scale revealed an increased impairment of the vigilance level between the normal reference night, the total sleep deprivation night and the total sleep deprivation night associated with an extended driving task; and (iii) the morning following this last condition, during the Wingate test, the recorded cycling biomechanical parameters (peak power, mean power and fatigue index values, power decrease, and cycling kinetic and kinematic patterns) were not significantly different from the two other conditions. Consequently, these results show that anaerobic performances recorded during a Wingate test performed at the bathyphase of the circadian rhythmicity are not altered by a drastic impairment in vigilance. These findings seem to indicate that vigilance is probably not a factor that contributes to circadian variations in anaerobic performance. 相似文献
