The effects of the cytokinin benzyladenine (BA) and the auxin naphthalene acetic acid (NAA) on Camptotheca acuminata Decaisne growth and camptothecin (CPT) accumulation (leaf CPT concentration and total leaf CPT yield) were studied in a hydroponic culture system for three weeks. Increasing BA concentrations from 0 to 3 mg l–1 in growth medium decreased plant height, stem weight, and leaf weight but increased root weight. High BA levels (1 and 3 mg l–1) increased leaf CPT concentration (% of dry weight), whereas BA applications had no effect on total leaf CPT yield, the product of leaf CPT concentration and total leaf dry weight per seedling. There was a positive correlation between root weight and leaf CPT concentration under BA treatments. NAA supplementations (from 0.5 to 4 mg l–1) to growth medium reduced plant height, leaf number, leaf length, specific leaf weight, plant weight, stem weight, and leaf weight compared with the NAA control. Meanwhile, there were no differences in plant height, leaf length, and specific leaf weight among the NAA supplementations. NAA applications had no effect on leaf CPT concentration and NAA applications decreased total leaf CPT yield. There were negative correlations between leaf number and leaf CPT concentration, leaf length and leaf CPT concentration under NAA treatments. Our results suggest that BA applications from 0.3 to 3 mg l–1 are not helpful for achieving high total leaf CPT yield and NAA applications from 0.5 to 4 mg l–1 decrease total leaf CPT yield.  相似文献

Pyrethrins and flower yield of pyrethrum (Chrysanthemum cinerariaefolium Viz.) plants were determined after application of ethrel, chlormequat chloride and paclobutrazol. Ethrel at 50, 100, 250 and 500 mg l−l produced a significant positive effect on pyrethrins level, decreased plant height, while 50 and 100 mg l−l significantly increased fresh and dry flower yield. Chlormequat chloride at 1000 and 2000 mg l−l and paclobutrazol (80 and 160 mg l−l) increased pyrethrins level, single flower weight and decreased plant height and flower yield. 14C-acetate incorporation studies further substantiated positive effect of growth retardants on pyrethrins biosynthesis. The effect of growth retardants on pyrethrins seems to be mediated through its effect on biosynthesis.  相似文献

The effects of waterlogging and salinity on morphological and physiological responses in the marsh grass Sporobolus virginicus (L.) Kunth were investigated in a 4×2 factorial experiment. Plants were subjected to four salinity levels (0, 100, 200 and 400 mol m–3 NaCl) and two soil inundation conditions (drained and flooded) for 42 days. Flooding at 0 mol m–3 NaCl caused initiation of adventitious surface roots, increased internal acration and plant height, induced alcohol dehydrogenase activity (ADH), and decreased belowground biomass and the number of culms per plant. Salinity increase from 0 to 400 mol m–3 NaCl under drained conditions increased leaf and root proline concentrations and decreased photosynthesis, aboveground biomass, number of culms per plant and number of internodes per culm. Concurrent waterlogging and salinity induced ADH activity and adventitious surface roots but decreased plant height and aboveground biomass. Internal air space increased with waterlogging from 0 to 100 mol m–3 NaCl but further increases in salinity to 400 mol m–3 reduced air space. Combined waterlogging and salinity stresses, however, had no effect on photosynthesis or on the concentrations of proline in leaves or roots. These results are discussed in relation to the widespread colonization by S. virginicus of a wide range of coastal environments varying in soil salinity and in the frequency and intensity of waterlogging.  相似文献

Summary Few studies have focused on the effect of a broad range of phytohormones on growth and secondary metabolism of a single hairy root species. We measured growth, development, and production of the antimalarial drug, artemisinin, in Artemisia annua hairy roots in response to the five main hormones: auxins, cytokinins, ethylene, gibberellins (GA), and abscisic acid (ABA). Single roots grown in six-well plates in medium B5 with 0.01 mgl−1 (0.029 μM) GA3 produced the highest values overall in terms of the number of lateral roots, length of the primary root, lateral root tip density, total lateral root length, and total root length. When the total root lengths are compared, the best conditions for stimulating elongation appear to be: GA 0.01 mgl−1 (0.029μM)> ABA 1.0 mgl−1 (3.78μM)=GA 0.02 mgl−1 (0.058μM). Bulk yields of biomass were inversely proportional to the concentration of each hormone tested. All cultures provided with ABA yielded the highest amount of biomass. Both 6-benzylaminopurine and 2-isopentenyladenine inhibited root growth, however, only 2-isopentenyladenine stimulated artemisinin production, more than twice that of the B5 controls, and more than any other hormone studied. These results will prove useful in increasing hairy root growth and artemisinin production.  相似文献

The growth of mustard was increased significantly when treated with up to 80 kg N ha–1 (N80). Spraying with (2-chloroethyl) trimethylammoniumchloride (chlormequat chloride) increased seed yield and seed protein content. Spraying nitrogen fertilized plots with chlormequat chloride, increased leaf area, leaf area ratio, leaf area duration, number of siliquae plant–1, number of seeds siliqua–1 and length of siliqua. Reducing, non-reducing and total sugars in the leaves at 80 days after sowing were also affected significantly. Chlorophyll a, b and total chlorophyll were little affected. The number of siliquae plant–1 was highly correlated with seed yield in both the seasons of experimentation. The correlation coefficient value () was 0.586 in 1982/83 and 0.912 in 1983/84.The total accumulation of nutrients, i.e. nitrogen, phosphorus and potassium in seed and straw was significantly affected by N80 × chlormequat chloride interaction.  相似文献

Leaf pigments, such as chlorophyll and carotenoids, are essential plant molecules. They provide carbohydrates and energy during all plant life. Leaf pigments are also important parameters of decorative plants, such as floriculture items, cut foliage and flowers. Leaf yellowing is a form of senescence caused by an internal hormone imbalance, such as a lack of cytokinins. The aim of this study was to investigate the changes in total carotenoids and endogenous ABA in cut flower stock leaves during post-harvest life. The effect of pulse treatment with 5 or 10 M thidiazuron (TDZ), 150 mg l–1 8-hydroxyquinoline sulphate (8-HQS) and combinations of TDZ with 8-HQS on total carotenoids and ABA concentration was also investigated. Results showed that total carotenoids drastically decreased from 1548 g cm–2, until reaching 565 g cm–2 at the end of vase life. Endogenous ABA strongly increased at the same time, going from 167 ng g–1 DW at the beginning of the experiment to 1322 ng g–1 DW at the end of vase life. The TDZ inhibited carotenoid degradation, but did not affect the ABA concentration, while the 8-HQS did not prevent carotenoid degradation and the ABA concentration was only slightly affected. ABA seems to be a secondary senescence bio-product that may have a synergic effect with other senescence inducers dramatically accelerating leaf senescence.  相似文献

Narrow-leafed lupin (Lupinus angustifolius L.) is usually grown in sandy, acidic and phosphorus (P) deficient soil with low yield and variable harvest index. This study aimed to examine the effects of varying P supply on lupin growth, seed yield and harvest index. Non-abscission plants (cv. Danja) were grown in Lancelin sand at seven rates of P supply (5, 10, 15, 20, 25, 30 or 40 mg kg–1) in a naturally-lit glasshouse. The rate of leaf emergence, flowering time and flower number were decreased or delayed by low P supply (5, 10 or 15 mg kg–1), with no differences at P rates higher than 20 mg kg–1. High P supply (25, 30 or 40 mg kg–1) increased plant seed yield and harvest index largely by increasing the number of pods and consequently yield on the lateral branches, but had less effect on the number of seeds per pod and seed size. Seed yield and seed P concentration continued to increase up to 40 mg P kg–1but harvest index plateaued at 25 mg P kg–1, indicating that low P supply decreased reproductive growth more than vegetative growth in narrow-leafed lupin.  相似文献

Effect of abscisic acid on the transport of assimilates in barley   总被引:15,自引:0,他引:15  
The effect of abscisic acid (ABA) on assimilate transport in barley was investigated in two parallel experiments. First, the effect upon [14C]sucrose transport from the flag leaf to the ear of a single ABA application made at different stages of growth of the fruits was investigated; the effect was measured 24 h after treatment. Second, the effect of a single application of ABA made at the same stages of growth as above on grain weight of the mature plant was investigated. In both types of experiments ABA was applied once to the ear of different plants as an aqueous solution (10-3–10-5 M), one to five weeks after anthesis. [14C] sucrose was applied by means of agar blocks. Parallel to these experiments, the endogenous content of ABA was investigated in the developing grains. When ears were treated with ABA two or four weeks after anthesis, an increase of up to 70% in the 14C-transport from the flag leaf to the ear was observed within a 24-h period after treatment (short duration experiments). At these growth stages the endogenous concentrations of ABA were low. In sharp contrast, ABA, especially in a concentration of 10-3 M, decreased 14C-import from the flag leaf when applied three weeks after anthesis. At this stage the endogenous ABA content had reached its maximum. Long duration experiments with a single application of ABA to the car two weeks after anthesis resulted in a marked increase of weight per thousand kernels. ABA applications made earlier or later than two weeks after anthesis either reduced the grain weight or had no effect. It is concluded that ABA is involved in the regulation of assimilate transport from the leaves to the grains, possibly by influencing the unloading of sieve tubes in the ears. Promotion or inhibition of assimilate import by exogenously applied ABA may depend on the developmental stage of the grains and on the endogenous ABA level.Abbreviations ABA abscisic acid - TKW weight per thousand kernels  相似文献

Protoplasts of Cyamopsis tetragonoloba were isolated from leaves of in vitro grown plants. The yield of the protoplasts, their viability and subsequent divisions were greatly influenced by the pH of the media used for isolation and culture of protoplasts. Sustained divisions of the cultured protoplasts were best supported by a modified Kao and Michayluk (1975) nutrient medium containing glucose (0.4 M), NAA (4 mgl–1), 2,4-D (1 mgl–1) and KIN (2 mgl–1 ). The protoplast derived cells developed calli on transfer to Murashige and Skoog (1962) medium supplemented with 1 mgl–1 each of 2,4-D, NAA and KIN.  相似文献

Responses of Cymbopogon martinii and C. winterianus to drought stress and chlormequat chloride and IAA application are compared. These two species are important source of essential oil production in drought regions. For both species and their cultivars relative water content (RWC), herbage yield and oil amount decreased under drought, while oil biosynthesis increased. Oil concentration increased significantly under drought in C. winterianus while peroxidase activity increased in C. martinii. Amount of geraniol increased under drought stress in C. martinii while citronellal and geraniol accumulation decreased in C. winterianus. Ameliorative effects of chlormequat chloride and IAA were observed in drought stressed plants of both species. Herbage yield increased significantly in chlormequat chloride and IAA treated stressed plants of C. winterianus, while oil concentration increased in C. martinii. Ameliorative effect of IAA in increasing oil yield was significant in drought stressed plants of both the species. Changes in various morpho-physiological traits indicated that chlormequat chloride and IAA can partially alleviate the detrimental effect of drought in these aromatic grasses.  相似文献

In field experiments with Japanese mint using different growth retardants, chlormequat chloride (2-chloroethyltrimethylammonium chloride) increased the oil content significantly and inhibited growth to some extent only, whilst ethephon (2-chloroethyl phosphonic acid) at 0.06% concentration significantly decreased growth but had no significant effect on oil content. Among the two oil components studied, menthone content only was significantly increased by 0.06% ethephon whereas the other growth retardants were ineffective. Correlation studies indicated a strong negative relationship of leaf/stem ratio with plant height and herb yield while a strong positive relationship was obtained between the latter two characters. The oil content of the plant was negatively correlated with herb yield and plant height but it was related positively with leaf/stem ratio.CIMAP Communication No. 688.  相似文献

A collection of bacterial strains obtained from a wide-range origin was screened for ability to promote growth in two types of Prunus rootstocks in a commercial nursery. Only few strains promoted growth significantly and consistently, and a strong specificity for the rootstock cultivar was observed. Irrigation of plants with Pseudomonas fluorescens EPS282 and Pantoea agglomerans EPS427 significantly increased plant height and root weight of the plum Marianna 2624 and the peach–almond hybrid GF-677, respectively. Plant height showed a higher rate of growth in early stages of development (2.6–3.5 times the non-treated controls), but the effect decreased with plant age. However, in aged plants growth promotion was more significant on root weight (1.9 times the non-treated controls) than on plant height. The efficacy of growth promotion and the persistence of strains in the root environment were dependent on the bacterial inoculum concentration applied. Increases in root development were maximum at inoculum concentrations of up to 8 log10 CFU ml–1 (ca 10 log10 CFU L–1 of potting mix). Population levels at the optimum inoculum concentration were around 7 log10 CFU g f.w.–1 root material at early stages of development and decreased to 4 log10 CFU g f.w.–1 after several months of development. The best plant growth-promoting strains were very diverse in secondary metabolite production and antagonistic ability against several plant pathogens.  相似文献

Boron is essential to growth at low concentrations and limits growth and yield when in excess. Little is known regarding plant response to excess boron (B) and salinity occurring simultaneously. The influences of B and salinity on tomatoes (Lycopersicon esculentum Mill. Cv `5656') were investigated in lysimeters. Salinity levels were 1, 3, 6 and 9 dSm–1 and B levels were 0.028, 0.185, 0.37, 0.74, 1.11, 1.48 mol m–3. Excess boron was found to decrease yield and transpiration of tomatoes. This effect was inhibited when plants were exposed to simultaneous B and salinity stresses. Both irrigation water salinity and boron concentration influenced water use of the plants in the same manner as they influenced yield. While yield was found to decrease with increased boron concentration in leaf tissue, increased salinity led to decreased boron accumulation. Yield response was found to correlate better to B concentration in irrigation water and soil solution than to plant tissue B content. A dominant-stress-factor model was assumed and validated. The model applies the principle that when a plant is submitted to conditions of stress caused by B in conjunction with salinity, the more severe stress determines yield. The results of this study have significance in modeling and management of high salinity high boron conditions. Under saline conditions, differences in crop yield and in water use may not be experienced over a significant range of boron concentrations.  相似文献

Plant nitrogen (N)deficiency often limits crop productivity. Early detection of plant N deficiency is important for improving fertilizer N-use efficiency and crop yield. An experiment was conducted in sunlit, controlled environment chambers in the 2001 growing season to determine responses of corn (Zea mays L. cv. 33A14) growth and leaf hyperspectral reflectance properties to varying N supply. Four N treatments were: (1) half-strength Hoagland's nutrient solution applied throughout the experiment (control); (2) 20% of control N starting 15 days after emergence (DAE); (3) 0% N starting 15 DAE; and (4) 0% N starting 23 DAE (0% NL). Plant height, the number of leaves, and leaf lengths were examined for nine plants per treatment every 3–4 days. Leaf hyperspectral reflectance, concentrations of chlorophyll a, chlorophyll b,and carotenoids, leaf and canopy photosynthesis, leaf area, and leaf N concentration were also determined during the experiment. The various N treatments led to a wide range of N concentrations (11 – 48 g kg–1 DW) in uppermost fully expanded leaves. Nitrogen deficiency suppressed plant growth rate and leaf photosynthesis. At final harvest (42 DAE), plant height, leaf area and shoot biomass were 64–66% of control values for the 20% N treatment, and 46-56% of control values for the 0% N treatment. Nitrogen deficit treatments of 20% N and 0% N (Treatment 3) could be distinguished by changes in leaf spectral reflectance in wavelengths of 552 and 710 nm 7 days after treatment. Leaf reflectance at these two wavebands was negatively correlated with either leaf N (r = –0.72 and –0.75**) or chlorophyll (r = –0.60 and –0.72**) concentrations. In addition, higher correlations were found between leaf N concentration and reflectance ratios. The identified N-specific spectral algorithms may be used for image interpretation and diagnosis of corn N status for site-specific N management.  相似文献

The effect of abscisic acid (ABA) on plant regeneration in anther-derived calli of rice Oryza sativa L. varieties Taipei 177, Taipei 309 and Fujisaka 5 was studied.ABA at concentrations up to 4×10–6 M stimulated fresh weight increase in Taipei 309 and Fujisaka 5 while higher concentrations effected corresponding weight decreases in the three varieties tested. Relatively high ABA concentrations resulted in decreased callus size and production of more compact and whitish calli. Pseudomonas putida and Pseudomonas fluorescens present as a coculture were studied for their abilities to degrade benzene, toluene, ethylbenzene, and xylenes (collectively known as BTEX) under various growth conditions. The coculture effectively degraded various concentrations of BTEX as sole carbon sources. However, all BTEX compounds showed substrate inhibition to the bacteria, in terms of specific growth, degradation rate, and cell net yield. Cell growth was completely inhibited at 500mgl–1 of benzene, 600mgl–1 of o-xylene, and 1000mgl–1 of toluene. Without aeration, aerobic biodegradation of BTEX required additional oxygen provided as hydrogen peroxide in the medium. Under hypoxic conditions, however, nitrate could be used as an alternative electron acceptor for BTEX biodegradation when oxygen was limited and denitrification took place in the culture. The carbon mass balance study confirmed that benzene and toluene were completely mineralized to CO2 and H2O without producing any identifiable intermediate metabolites.  相似文献

Background and Aims

The resurgence of malaria, particularly in the developing world, is considerable and exacerbated by the development of single-gene multi-drug resistances to chemicals such as chloroquinone. Drug therapies, as recommended by the World Health Organization, now include the use of antimalarial compounds derived from Artemisia annua – in particular, the use of artemisinin-based ingredients. Despite our limited knowledge of its mode of action or biosynthesis there is a need to secure a supply and enhance yields of artemisinin. The present study aims to determine how plant biomass can be enhanced while maximizing artemisinin concentration by understanding the plant''s nutritional requirements for nitrogen and potassium.

Methods

Experiments were carried out, the first with differing concentrations of nitrogen, at 6, 31, 56, 106, 206 or 306 mg L−1 being applied, while the other differing in potassium concentration (51, 153 or 301 mg L−1). Nutrients were supplied in irrigation water to plants in pots and after a growth period biomass production and leaf artemisinin concentration were measured. These data were used to determine optimal nutrient requirements for artemisinin yield.

Key Results

Nitrogen nutrition enhanced plant nitrogen concentration and biomass production successively up to 106 mg N L−1 for biomass and 206 mg N L−1 for leaf nitrogen; further increases in nitrogen had no influence. Artemisinin concentration in dried leaf material, measured by HPLC mass spectroscopy, was maximal at a nitrogen application of 106 mg L−1, but declined at higher concentrations. Increasing potassium application from 51 to 153 mg L−1 increased total plant biomass, but not at higher applications. Potassium application enhanced leaf potassium concentration, but there was no effect on leaf artemisinin concentration or leaf artemisinin yield.

Conclusions

Artemisinin concentration declined beyond an optimal point with increasing plant nitrogen concentration. Maximization of artemisinin yield (amount per plant) requires optimization of plant biomass via control of nitrogen nutrition.Key words: Artemisia, fertigation, malaria, nitrogen, nutrition, potassium  相似文献

Ultrastructural alterations in mesophyll cells as well as variations in bulk leaf endogenous ABA and IAA concentrations were studied in water-stressed field-grown plants of Fatsia japonica. Under water deficit cellular membranes were modified and an increase in vesicles was observed. The main damage to the chloroplasts included thylakoid swelling and disruption of the chloroplast envelope. Concomitant variations in abscisic acid and indole-3-acetic acid were observed. Despite the expected increased in endogenous ABA concentration in relation to water stress, after the highest concentration of ABA, observed at predawn in severely stressed plants (29-1), there was a sharp decline from 2768 pmol g fw–1 to 145 pmol g fw–1; thus in severely stressed plants ABA levels were not related to changes in bulk leaf ABA contents. Water stress did not influence the concentrations of indole-3-acetic acid, although the increase in the endogenous abscisic acid concentration could be related with the ultrastructural changes.Abbreviations ABA abscisic acid - IAA indole-3-acetic acid - leaf water potential  相似文献

In West-Europe, intensive cereal management uses plant growth regulators (PGRs) especially for wheat. A green-house experiment compared the effects of two PGRs on flag leaf characteristics and yield of winter wheat. Chlormequat chloride + choline chloride (CCC) and chlormequat chloride + choline chloride + imazaquin (CCC+I) were applied to winter wheat at growth stage 5 (Feekes Large scale). CCC and CCC+I significantly increased flag leaf surface area at anthesis. Both treatments also enhanced chlorophyll content of the main stem flag leaf. The grain filling period was extended with PGR application by 2 days. CCC and CCC+I significantly increased net CO2 assimilation rates during the flag leaf life. No effects of PGR spraying were observed on the pattern of 14C labelled assimilate distribution. Increased grain yield was due to the increase in average grain weight. The results indicate that PGR treatments increased flag leaf contribution to grain filling. The addition of imazaquin (I) to chlormequat (CCC) improved the effects of CCC.  相似文献

Procedures have been developed that increase the rate of shoot regeneration of hybrid seed geranium from month-old primary callus cultures. Hybrid geranium callus tissue covered with green nodular structures was initiated by placing shoot tip explants on solidified Murashige & Skoog medium (MS) supplemented with 2.0 mgl-1 zeatin and 1.9 mgl-1 indoleacetic acid. Hybrids Red Orbit, White Orbit and Scarlet Orbit were shown to produce 5–50 shoot primordia per explant when callus was initiated on this medium. Regal geranium callus was initiated by placing leaf explants on MS medium supplemented with 2.0 mgl-1 6-benzylaminopurine and 2.0 mgl-1 naphthaleneacetic acid. Regal geranium cultivars Tiny Tot and Lavender Grand Slam were shown to produce between 2–50 shoot primordia per explant when initiated on the same medium.  相似文献
