The senescence-promoting activities of methyl jasmonate and its related compounds were compared with respect to structure-activity relationships. The activities were assayed by using oat ( A vena saliva L. cv. Victor) leaf segments in the presence of 2 μg/ml kinetin. Dextrorotatory methyl jasmonate prepared from an authentic sample of the racemate mixture was less active than the naturally occurring levorotatory form especially at its low concentrations (0.1 to 2.5 μg/ml). The activity of jasmonic acid, the free acid form of methyl jasmonate, was much less than the methyl ester, and this relationship was true for the other compounds tested. The reduction of the unsaturated bond in the substituent at the C-2 position and the keto group at the C-3 position greatly reduced the activity. The length of the n -alkyl substituents at the C-2 position had also a significant effect on the activity. From these results, it is concluded that the important functional groups for the high senescence-promoting activity of the methyl jasmonate related compounds are the methyl acetate substituent at the C-l position, the 2' cis -pentenyl or n -pentyl group at the C-2, position and the keto group at the C-3 position in methyl jasmonate.  相似文献

The effects of different growth regulators on induction and growth of callus ofAsparagus densiflorus cv. Sprengeri were studied. Calluses grew more rapidly on Murashige and Skoog basal medium supplemented with 5.4 μM p-chlorophenoxyacetic acid (pCPA) and 4.4 μM 6-benzylaminopurine (BA) (medium 1) as compared to the same medium with 11.3 μM 2,4-dichlorophenoxyacetic acid (2,4-d) and 4.6 μM kinetin (medium 2). Calluses on medium 1 were soft and friable, whereas, compact, hard calluses originated on medium 2. Different concentrations and combinations of BA and/or kinetin were also used to study their effects on shoot regeneration. Kinetin was found to be less effective than BA in the initiation of shoots (1.8 shoots/callus). High numbers of shoots were produced in the presence of 0.4 μM BA alone (3.3 shoots/callus). The addition of ancymidol (5 μM) in MS with 0.4 μM BA enhanced multiplication of shoots (9.8 shoots/explant) and also produced well-developed crowns.  相似文献

Summary The cv Ringo Rose of hybrid seed geranium (Pelargonium x hortorum Bailey), previously shown to be recalcitrant in culture, produced somatic embryos when cotyledonary explants were cultured on regeneration medium containing thidiazuron (TDZ), forchlorfenuron (CPPU), or a combination of indole-3-acetic acid and N6 benzylaminopurine (IAA+BAP). Amendment of the basal medium with TDZ (0.5 M) was the most effective treatment. Addition of amino acids to the medium promoted the growth of somatic embryos. Retention of the proximal region of the cotyledon was crucial for regeneration, but the removal of the distal 1/3 to 1/2 cotyledon had no significant effect on somatic embryogenesis. Cotyledonary explants formed somatic embryos in higher frequency and much earlier than hypocotyl explants cultured on the same medium. The somatic embryos induced on cotyledonary explants were germinated on basal medium. A novel, genotype dependent system for rapid high frequency somatic embryogenesis in Medicago sativa L. was developed in which the first embryos are visible as early as 15 days after the explant (hypocotyl, petiole, leaf) is put into culture. The simplest method involves culture of the explants on a single Murashige and Skoog (MS) medium supplemented with 2 g l−1 casein hydrolysate, 9 μ M 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.2 μ M kinetin. An efficient two-step, two-medium system was developed to allow separation of the induction and differentiation phases. The explants are cultured on MS with 22.6 μ M 2,4-D and 4.7 μ M kinetin (induction medium) for 10 days and then on basal MS for 20 days. Embryo yields and embryo conversion to plantlets were strongly dependent on the 2,4-D and kinetin concentrations in the induction medium. Both petiole and leaf explants were highly embryogenic and very little callus proliferation occurred when this method was used. Selected clones from three ssp. falcata -based M. sativa cultivars showed a response very similar to the highly regenerable falcata clone F1.1, but it was not possible to produce large numbers of somatic embryos in tissue cultures of cv. Regen S, which is used in most M. sativa tissue culture research, with this procedure. These results suggest that there are two distinct developmental pathways for somatic embryogenesis in M. sativa , with Regen S cultures requiring extensive dedifferentiation during a prolonged callus phase, while the genotypes described in this report have no such requirement.  相似文献

The effects of methyl jasmonate (MeJA) in relation to abscisic acid (ABA) on different phases of somatic embryogenesis were studied in Medicago sativa L. Different concentrations of both the growth inhibitors (0.0, 0.5, 5.0, 50.0 and 500.0 μM) were tested in five distinct phases of somatic embryogenesis, viz., induction, proliferation, differentiation, maturation and regeneration. Like ABA, MeJA also inhibited callus induction, callus growth, proliferation of embryogenic suspension as well as germination and conversion of somatic embryos. However, its inhibitory effects on various phases of somatic embryogenesis were less pronounced as compared to that due to ABA. In contrast to ABA, MeJA did not have any significant influence on the development of somatic embryos when applied in the differentiation phase. The study showed that ABA used routinely as an inducer of somatic embryo maturation in M. sativa could not be replaced by MeJA.  相似文献

Growth, K+ content, and alkaloid production were compared in nonorganogenetic callus cultures ofNicotiana tabacum cv. Burley 21 grown at 25°C in the dark on two different media: a basal medium with 1 M -naphthaleneacetic acid and 1 M kinetin, and one with 1 M -naphthaleneacetic acid and 1 M 4PU-30 (N-(2-chloro-4-pyridyl)-N-phenylurea). These callus tissues behaved differently not only in growth and K+ content but also in alkaloid production. In comparison to cultures grown with kinetin, those grown with 4PU-30 showed a significantly higher fresh weight and dry weight and K+ content during the growth period studied. The data clearly indicate a positive correlation between K+ uptake rate stimulated by 4PU-30 and cell enlargement rate. However, the alkaloid biosynthesis in the callus tissues was activated by the supply of kinetin and diminished by that of 4PU-30. It thus appears that cellular enlargement of meristematic tissue stimulated by 4PU-30 limited alkaloid production.  相似文献

In the cucumber cotyledon greening system, abscisic acid (ABA)is more potent inhibitor of growth and chlorophyll productionand/or destruction than methyl jasmonate (MJ). The inhibitoryeffect of ABA is apparent within 5 h of exposure to light whereasMJ is ineffective at all concentrations tested (10–6 to10–3 M). With longer exposure of 24 h to light and inthe presence of 40 mM KC1, the inhibition of growth and chlorophyllproduction by ABA is more pronounced whereas MJ does not inhibitgrowth and inhibits chlorophyll levels only at the higher concentrations.Both benzyladenine and KC1 stimulate chlorophyll productionand increase the fresh weights of the cucumber cotyledons andeither one of these compounds reverse the inhibitory effectsof ABA. Inhibition of chlorophyll production by ABA is valuableas a simple and rapid bioassay for abscisic acid. Under similarconditions cytokinins increase chlorophyll production and hencethe cucumber cotyledon greening system is ideal for detectingboth ABA which inhibits and cytokinins which stimulate chlorophyllproduction. A tissue culture method was developed to investigate the production of cotton (Gossypium hirsutum L. cv. Texas Marker-1) fibers in vitro. Ovules were excised from 3, 5, 7 and 9 days preanthesis ovaries and placed on an agar-solidified, modified Murashige and Skoog medium containing 2.3 μ M kinetin and 0.45 μ M –2,4–dichlo-rophenoxyacetic acid or 2.3 μ M kinetin and 10.7 μ M naphthaleneacetic acid. Ovules formed fibers and callus tissue. Fibers formed in vitro were up to 10 mm long, 10–22 μ wide and the cell wall was 1–3 μ M thick. Callus tissue cells were subcultured for over 25 weeks and their degree of elongation was monitored. The ability of ovule-derived cells to direct expansion in a longitudinal direction diminished, while lateral expansion increased with time in culture.  相似文献

The effects of methyl jasmonate (MJ) and abscisic acid (ABA) on some physiological processes of rice were compared. MJ exhibited ABA-like effects by promoting senescence of detached leaves, by inducing acid phosphatase activity of detached leaves, by inhibiting ethylene production and shoot growth of seedlings, as well as inhibiting callus formation from anthers. However, MJ and ABA had opposite effects on 1-aminocyclopropane-1-carboxylic acid-dependent ethylene production in detached leaves. The regeneration ability of anther-derived callus was inhibited by MJ but not by ABA. MJ but not ABA markedly induced peroxidase activity in senescing detached leaves. We investigated the effect of 24-epibrassinolide (BR) on the cytokinin-bioassay based on growth of isolated radish ( Raphanus sativus L. cv. Tondo Rosso Quarantino) cotyledons. BR stimulated growth of the cotyledons by about 50% when applied at saturating concentrations (3 μ M ). This effect was much lower than that of saturating concentrations of benzyladenine (BA) which was about 150% at 10 μ M BA. The effects of saturating concentrations of BR and BA were additive. BR, but not BA, stimulated H+-secretion by the cotyledons (measured as acidification of the incubation medium) slightly but reproducibly. These results indicate that the modes of action of the two phytoregulators are at least partially different.
BR-induced stimulation of H+-secretion was of similar amplitude to that induced by a concentration of fusicoccin (30 n M ) such as to stimulate growth to the same extent as BR. These results suggest that BR-induced stimulation of radish cotyledon growth might depend, at least in part, on BR-induced acidification of the wall space, similar to that observed on Azuki bean epicotyls and maize roots.  相似文献

The effects of methyl jasmonate and jasmonic acid on uptake of abscisic acid (ABA) by suspension-cultured runner-bean cells and subapical runner-bean root segments have been investigated. Increasing concentrations of methyl jasmonate inhibit ABA uptake by the cultured cells with a K i of 22±3 M. This is not due to cytoplasmic acidification or to effects on metabolism of ABA, and is not additive with inhibition of radioactive ABA uptake by nonradioactive ABA. Uptake of indol-3-yl acetic acid (IAA) is unaffected by methyl jasmonate. The maximum effect of nonradioactive ABA in inhibiting uptake of radioactive ABA, previously shown to reflect saturation of an ABA carrier, is generally greater than the effect of maximally inhibitory concentrations of methyl jasmonate. Similar results were obtained with root segments, but longer incubation times were necessary to observe inhibitory effects of methyl jasmonate. Demethylation of methyl jasmonate to jasmonic acid does not appear to be required since similar concentrations of jasmonic acid had no observable direct effect on ABA uptake other than that attributable to cytoplasmic acidification. Histidine reagents, a proton ionophore and acidic external pH all affect in parallel the inhibition by methyl jasmonate and nonradioactive ABA of uptake of radioactive ABA by the cultured cells. There is no effect of ABA or nonradioactive methyl jasmonate on uptake of radioactive methyl jasmonate by the cultured cells. It is proposed that methyl jasmonate interacts with the ABA carrier. Callus cultures of Zanthoxylum stenophyllum were initiated in vitro and the effect of growth regulators and elicitors was tested both upon callus growth and secondary metabolite production. On a medium containing naphthaleneacetic acid, kinetin, and 2,4-dichlorophenoxyacetic acid, a yellowish and friable callus was obtained from 90% of cotyledon explants. Callus growth and secondary metabolite accumulation was followed after sub-culturing the established callus culture on different media containing various hormonal combinations. Results indicate that medium containing naphthaleneacetic acid and a higher concentration of 2,4-dichlorophenoxyacetic acid gave the highest stimulation of growth. Addition of an organic nitrogen source also had a positive effect on growth. Rapid HPLC screening of methanol extractable secondary metabolites from calluses showed that phytohormones and nutrients were able to modify the chromatographic pattern of compounds. Calluses grown on the medium giving the highest stimulation of growth show a reduced accumulation of some secondary products, but not all. In response to elicitation by methyl jasmonate, metabolite production was different for the different classes of compounds, and hormonal composition of the culture medium influenced the response. Thus, results confirm the importance of the reciprocal interactions between hormones, nutrients, and elicitors when attempts are made to enhance secondary metabolite accumulation in in vitro cultures.  相似文献

Increase in fruit set of grape was observed when vines were sprayed with zeanic acid before blossom, but when sprayed after blossom the effect was not clear.

Promotion of growth of radish cotyledons by zeanic acid was observed in the presence and absence of kinetin. Zeanic acid also promoted growth of dwarf mutant of rice seedlings (cv. Tanginbozu), but inhibited that of normal seedlings (cv. Tonewase). Zeanic acid showed promotive effect on proliferation of rice callus tissue in the absence of kinetin and auxin.

Zeanic acid showed low toxic activity against mice, earthworm and killifish, and no antimicrobial activity.  相似文献

Cotyledons of sunflower seedlings ( Helianthus annuus L. cv. Giant gray stripe) expand and their protein content first rises then begins to decrease during the first three days of growth. Storage protein structures, which are visible with scanning electron microscopy, undergo modification that leads to storage protein disappearance by day 4 post-imbibition. Expansion of cotyledons detached from seeds prior to imbibition is greatly reduced, total protein levels remain high, and storage protein structures remain visible in cells of these cotyledons. Incubation of excised cotyledons in 1.0 μM benzyladenine or kinetin increases the rates of cotyledon expansion and storage protein loss to levels higher than in intact seedling cotyledons, Incubation in 10 μM indole-3-acetic acid inhibits cotyledon expansion and protein mobilization. More rapid hydrolysis of storage proteins in cotyledons of intact seedlings or detached cotyledons treated with cytokinin is further indicated in day 2 specimens by SDS-polyacrylamide gel electrophoresis. These results suggest a possible mechanism for regulation of cotyledon development by interactions of the promotive effects of cytokinin and inhibitory effects of auxin.  相似文献

Somatic embryo formation occurred on leaf callus of grape (Vitis vinifera cv. Koshusanjaku). An embryogenic callus was induced from somatic embryo clusters cultured on vitamin-, inositol- and glycine-free Nitsch and Nitsch (1969) medium supplemented with 1.0M 2,4-D. This callus has retained a high embryogenic activity after repeated subculture on the same medium for over two years, and has produced numerous embryos after transfer to a hormone-free medium. The effect of cytokinin treatment on somatic embryogenesis from leaf callus was also examined. N-(2-chloro-4-pyridyl)-N-phenylurea (KT-30) and N-(1,2,3-thiadiazol-5-yl)-N-phenylurea (TAG), both synthetic cytokinins, were found to be effective for the induction of somatic embryogenesis. Osmotic stress and endogenous hormone levels may have a role in shoot organogenesis, but a systematic study has not yet to investigate the links. We evaluated the changes of the endogenous indole-3-acetic acid (IAA) and abscisic acid (ABA) levels in rice (Oryza sativa L. cv. Tainan 5) callus during shoot organogenesis induced by exogenous plant growth regulator treatments or under osmotic stress. Non-regenerable callus showed low levels of endogenous ABA and IAA, with no fluctuation in level during the period evaluated. The addition of 100 μM ABA or 2 mM anthranilic acid (IAA precursor) into Murashige and Skoog basal induction medium containing 10 μM 2,4-D enhanced the regeneration frequency slightly, to 5 and 35%, respectively, and their total cellular ABA or IAA levels were increased significantly, correspondingly to the treatments. However, the regeneration frequency was greatly increased to 80% after treatment with 0.6 M sorbitol or 100 μM ABA and 2 mM anthranilic acid combined. Both treatments produced high levels of total cellular ABA and IAA at the callus stage, which was quickly decreased on the first day after transfer to regeneration medium. Thus, osmotic stress-induced simultaneous accumulation of endogenous ABA and IAA is involved in shoot regeneration in rice callus.  相似文献

Summary Jasmonic acid (JA) and its derivatives are a novel group of plant endogenous growth regulators. In our experiments some new data about the physiological effects of JA were obtained using electron paramagnetic resonance spectroscopy. Experiments were performed on potato plants (Solanum tuberosum L. cv. Vesna) grown in vitro. Different quantities of JA (0.1–10 M) were added to the growth medium. Root samples of plants grown on media supplemented with JA showed a more rapid spin probe N-oxyl-2,2,6,6-tetramethyl piperidine reduction than the control plants, which is a possible indicator of altered root permeability. Samples of leaf protoplasts were probed with methyl ester of 5-doxyl-haxadecanoic acid. Senescence of rice ( Oryza sativa L. cv. Jaya) leaves was regulated with kinetin and abscisic acid (ABA) sprays at the reproductive stage. The effect of such sprays on grain-filling and yield was analyzed. Spraying 100-day-old plants with kinetin solution (100 μg ml-1) significantly delayed senescence as indicated by higher total chlorophyll and protein levels in the three uppermost leaves compared with the controls. In contrast, spraying with ABA (15 μg ml-1) significantly promoted foliar senescence. The number of spikelets per panicle, number of panicles, percentage filled grains, panicle weight and grain yield per plant and the mobilization and harvest indices were significantly increased by kinetin treatment, while ABA decreased most of them. The possibility of increased grain-filling and thus, yield due to delayed foliar senescence by kinetin treatment and decreased grain-filling due to hastening of senescence by ABA is discussed.  相似文献

The roles of phytohormones and fusicoccin in H+ extrusion by isolated protoplasts from rape leaves ( Brassica napus L. cv. Belinda) were investigated and compared to results obtained with leaf segments of the same plants. Net H+ release by protoplasts, which was at least partly due to ATPase activity, was enhanced by 10 μ M indole-3-acetic acid and reduced by 20 μ M abscisic acid, whereas fusicoccin (10 μ M ), brassinosteroid (3 μ M ), kinetin (20 μ M ) and gibberellic acid (10 μ M ) had no effect. Hormone effects and H+ release were not detectable with leaf segments from the same plants. However, using field-grown plants, indole-3-acetic acid and especially fusicoccin stimulated the acidification of the external medium by leaf segments. Hormonecontrolled H+ release by leaf cells is interpreted as the first step in acid-triggered and turgor-regulated cell growth.  相似文献
