The position of the centre of resistance (Cre) as well as the centre of rotation (Cro) of a tooth under a force-system is still an open question. This paper presents a reliable and efficient three-dimensional rigid-body finite element technique to accurately estimate these centres. The influence of not only the root length but also the root diameter, the thickness of the periodontal ligament, as well as its material properties on the position of the Cre and Cro is investigated. Additionally, an explanation is given for the meaning of the coefficient (0.068 h(2) ) involved in Burstone's theoretical formula which is generalised and is expressed as the ratio of the flexibilities of tooth support in translation and pure moment rotation, respectively. The former ratio determines the position of the centres of rotation as a function of the applied moment-to-force ratio (M/F) and the relevant curve remains an isosceles hyperbola for any arbitrary-shaped tooth. The present study focuses on single-rooted teeth, such as maxillary canines and maxillary incisors, but the proposed methodology is generally applicable to any tooth.  相似文献

The objectives of this study were to develop a numerically controlled experimental set-up to predict the movement caused by the force systems of orthodontic devices and to experimentally verify this system. The presented experimental set-up incorporated an artificial tooth fixed via a 3D force/moment sensor to a parallel kinematics robot. An algorithm determining the initial movement of the tooth in its elastic embedding controlled the set-up. The initial tooth movement was described by constant compliances. The constants were obtained prior to the experiment in a parameterised finite element (FE) study on the basis of a validated FE model of a human molar. The long-term tooth movement was assembled by adding up a multiple of incremental steps of initial tooth movements. A pure translational movement of the tooth of about 8 mm resulted for a moment to force ratio of ? 8.85 mm, corresponding to the distance between the bracket and the centre of resistance. The correct behaviour of this linear elastic model in its symmetry plane allows for simulating single tooth movement induced by orthodontic devices.  相似文献

In a recent paper by D. R. Bellwood and C. P. Meyer ('Searching for heat in a marine biodiversity hotspot', Journal of Biogeography , 2009, 36 , 569–576), the authors had two evident objectives: (1) to disprove the theory that the geographical origins of reef organisms could be determined by locating concentrations of endemic species, and (2) to emphasize that the high diversity of the Coral Triangle was due to an accumulation of species from outside that area. With regard to the first point, no such theory had previously been proposed to my knowledge. Second, the accumulation theory was promoted without consideration of the facts supporting the centre of origin hypothesis, except to dismiss it by saying that it had its origin in pre-continental drift ideas. This short response outlines the properties and evidence for the operation of centres of origin in this region.  相似文献

The (bacterio)chlorophylls of photosynthetic antenna and reaction centre complexes are bound to the protein via a fifth, axial ligand to the central magnesium atom. A number of the amino acids identified as providing such ligands are conserved between the large antenna of the cyanobacterial Type-I reaction centre and smaller antennas of the Type-I reaction centres of green sulphur bacteria and heliobacteria, and these numbers match closely the estimated number of antenna bacteriochlorophylls in the latter. The possible organisation of the antenna in the latter reaction centres is discussed, as is the mechanism by which the more pigment-rich antenna of the cyanobacterial reaction centre evolved. The homology modelling approach is also extended to the six-helix antenna proteins CP47 and CP43 associated with the Photosystem II reaction centre.  相似文献

In a recent paper (Bellwood & Meyer, Journal of Biogeography , 2009, 36 , 569–576), we critically evaluated the utility of marine endemics for marking the geographical origins of species. In reply, Briggs (2009) identified two issues that needed clarification: (1) whether endemics are assumed to mark the geographical origins of species or areas of exceptionally high rates of origination, and (2) whether our evaluation of the role of endemics disproves the centre of origin hypothesis. Of these two issues, the first can be clearly resolved by recourse to the original literature that explicitly states that it has been assumed that endemics do indeed mark the probable sites of origin of species. The second is equally clear: our evidence does not and can not disprove the centre of origin theory. We suggest, however, that the current data, and endemics in particular, provide limited support for centre of origin theories and that they are more consistent with some centre of accumulation theories. The Indo-Australian Archipelago (IAA; Coral Triangle) therefore appears to be an area where species persist, a centre of survival, regardless of the site of origin of species.  相似文献

1. The excitation spectrum for the bacteriochlorophyll P890 fluorescence in reaction centre preparations was determined at wavelengths ranging from 360 to 890 nm.2. A fluorescence excitation spectrum corresponding to the absorbance spectrum of bacteriopheophytin was also obtained. This spectrum was used in an analysis of the absorbance spectrum of a reaction centre preparation. Based on this spectrum and on literature data, we estimated that the bacteriopheophytin: bacteriochlorophyll ratio in reaction centre particles is at least 1 : 2.3. On the basis of literature data, it is shown that bacteriopheophytin occurs probably as such in reaction centres in vivo.  相似文献

The quiescent centre of primary roots of Zea mays L. (cvs LG 11 and Golden Bantam) consists of a population of slowly cycling diploid cells. These metabolically inactive cells may be triggered to synthesise DNA under specific conditions and constitute a good model for studying the regulation of the cell cycle. An excision and squash technique is described for the quiescent centre which, when coupled with Feulgen and fluorochrome staining, allows nuclear DNA contents to be determined by microdensitometry in less than a day. This technique was coupled with experiments in which excised quiescent centres were placed on solid culture medium into which hormones and radioactive DNA precursors were incorporated. In complementary and control experiments [methyl-3H]thymidine was supplied to intact roots (with or without root caps) by means of fibre-glass cubes as donors.
Progression of the cell cycle was followed by microdensitometry and autoradiography. Distribution of DNA content was similar in excised and squashed quiescent centres and in histological sections. Labelling experiments showed that the quiescent centre is made up of cells that differ in their cycle time. While some labelled cells had reached mitosis after 8 h, others were still in G2 after 16 h of continuous labelling. Excision and culture of the quiescent centre resulted in a dramatic activation of the cell cycle as shown by the labelling index that increased from 15% in intact roots fed during 16 h with [methyl-3H]thymidine to 31% in excised quiescent centres to which radioactive precursor was supplied during the same time. Supplying hormones (50 μ M abscisic acid [ABA], 0. 1 μ M zeatin, 1 μ M zeatin riboside) to quiescent centres via the culture medium restored their inactivity (labelling indices dropped to 1% after ABA. and to 11% after zeatin and zeatin riboside treatments. respectively).  相似文献

Recent studies of sprinters and distance runners have suggested that variations in human foot proportions and plantarflexor muscle moment arm correspond to the level of sprint performance or running economy. Less clear, however, is whether differences in muscle moment arm are mediated by altered tendon paths or by variation in the centre of ankle joint rotation. Previous measurements of these differences have relied upon assumed joint centres and measurements of bone geometry made externally, such that they would be affected by the thickness of the overlying soft tissue. Using magnetic resonance imaging, we found that trained sprinters have shorter plantarflexor moment arms (p = 0.011) and longer forefoot bones (p = 0.019) than non-sprinters. The shorter moment arms of sprinters are attributable to differences in the location of the centre of rotation (p < 0.001) rather than to differences in the path of the Achilles tendon. A simple computer model suggests that increasing the ratio of forefoot to rearfoot length permits more plantarflexor muscle work during plantarflexion that occurs at rates expected during the acceleration phase following the sprint start.  相似文献

The minimal photounit of the photosynthetic membranes of the purple non-sulphur bacterium Rhodospirillum rubrum, comprising the reaction centre and the light-harvesting complex has been purified and crystallised in two dimensions in the presence of added phospholipids, and subsequently visualised by electron microscopy after negatively-staining. The position of the reaction centres within the light-harvesting ring has been determined at low resolution by the application of a new analysis for rotationally disordered identical units (here the reaction centres) within a two-dimensional crystalline lattice comprised of perfectly aligned unit cells (here the light-harvesting complexes). The reaction centre was found to preferentially occupy one of four orientations within the light-harvesting complex. The light-harvesting complex appears to be distorted to C4 symmetry, thus assuming a squarish shape when visualised by negative staining. A tentative structural model of the reaction centre-light-harvesting complex photounit which fits the experimental data is proposed.  相似文献

The oxidation-reduction potential of the iron-quinone electron acceptors in the reaction centre of Rhodopseudomonas viridis has been reinvestigated. In chromatophores treated with o-phenanthroline to remove the secondary acceptor Qb, two steps were observed in the reduction of the primary electron acceptor Qa with Em ≈ − 100 and ≈ − 330 mV. In isolated reaction centres only one step was observed in the reduction of Qa with E ≈ −150 mV. Reconstitution of the reaction centres with additional menaquinone resulted in an increase in the Qa EPR signal and reconstitution of the low-potential step in the oxidation-reduction titration. Reconstitution with ubiquinone resulted in the recovery of the secondary quinone Qb. The addition of ubiquinone did not reconstitute the low-potential step of Qa reduction, or affect the reconstitution of this step by menaquinone. It is concluded that menaquinone can bind to two sites on the reaction centre. Both have properties of the Qa site but with different pK values. It is unlikely that either is the same as the Qb site.  相似文献

The purple photosynthetic bacterium Thermochromatium tepidum is a moderate thermophile, with a growth optimum of 48–50 °C. The X-ray crystal structure of the reaction centre from this organism has been determined, and compared with that from mesophilic bacteria such as Blastochloris viridis and Rhodobacter sphaeroides (Nogi T et al. (2000) Proc Natl Acad Sci USA 97: 13561–13566). Structural features that could contribute to the enhanced thermal stability of the Thermochromatium tepidum reaction centre were discussed, including three arginine residues exposed at the periplasmic side of the membrane that are not present in reaction centres from mesophilic organisms, and potentially could increase the affinity of the complex for the surrounding membrane. In the present report these arginine residues, plus a histidine identified from an extensive sequence alignment, were engineered into structurally homologous positions in the Rhodobacter sphaeroides reaction centre, and the effect on the thermal stability of the Rhodobacter sphaeroides complex was examined. We find that these residues do not enhance the thermal stability of the reaction centre, as assessed by absorbance spectroscopy of the bacteriochlorin cofactors in membrane-bound reaction centres. Possible roles of these residues in the Thermochromatium tepidum reaction centre are discussed, and it is proposed that they facilitate stronger binding of the reaction centre to the encircling LH1 antenna complex, through ionic interactions with acidic residues at the C-terminal end of the LH1 α-polypeptide. Such an interaction could enhance the stability of the so-called ‘RC–LH1 core’ complex that is formed between the reaction centre and the LH1 antenna, and which represents the minimal functional photosynthetic unit in all known purple photosynthetic bacteria. Stronger bonding interactions between the two complexes could also contribute to an increase in the rigidity of the photosynthetic membrane in Thermochromatium tepidum, in accord with the general finding that the cytoplasmic membrane from thermophilic eubacteria is less fluid than its counterpart in mesophilic bacteria.  相似文献

This paper introduces a ‘hypothesis about the growth pattern of the secondary ossification centre (SOC)’, whereby two phases are assumed. First, the formation of cartilage canals as an event essential for the development of the SOC. Second, once the canals are merged in the central zone of the epiphysis, molecular factors are released (primarily Runx2 and MMP9) spreading and causing hypertrophy of adjacent cells. In addition, there are two important molecular factors in the epiphysis: PTHrP and Ihh. The first one inhibits chondrocyte hypertrophy and the second helps the cell proliferation. Between these factors, there is negative feedback, which generates a highly localised and stable pattern over time. From a mathematical point of view, this pattern is similar to the patterns of Turing. The spread of Runx2 hypertrophies the cells from the centre to the periphery of the epiphysis until found with high levels of PTHrP to inhibit hypertrophy. This mechanism produces the epiphyseal bone-plate. Moreover, the hypertrophy is inhibited when the cells sense low shear stress and high pressure levels that maintain the articular cartilage structure. To test this hypothesis, we solve a system of coupled partial differential equations using the finite element method and we have obtained spatio-temporal patterns of the growth process of the SOC. The model is in qualitative agreement with experimental results previously reported by other authors. Thus, we conclude that this model can be used as a methodological basis to present a complete mathematical model of the whole epiphyseal development.  相似文献

In addition to the energy dissipation of excess light occurring in PSII antenna via the xanthophyll cycle, there is mounting evidence of a zeaxanthin-independent pathway for non-photochemical quenching based within the PSII reaction centre (reaction centre quenching) that may also play a significant role in photoprotection. It has been demonstrated that acclimation of higher plants, green algae and cyanobacteria to low temperature or high light conditions which potentially induce an imbalance between energy supply and energy utilization is accompanied by the development of higher reduction state of QA and higher resistance to photoinhibition (Huner et al., 1998). Although this is a fundamental feature of all photoautotrophs, and the acquisition of increased tolerance to photoinhibition has been ascribed to growth and development under high PSII excitation pressure, the precise mechanism controlling the redox state of QA and its physiological significance in developing higher resistance to photoinhibition has not been fully elucidated. In this review we summarize recent data indicating that the increased resistance to high light in a broad spectrum of photosynthetic organisms acclimated to high excitation pressure conditions is associated with an increase probability for alternative non-radiative P680+QA - radical pair recombination pathway for energy dissipation within the reaction centre of PSII. The various molecular mechanisms that could account for non-photochemical quenching through PSII reaction centre are also discussed.  相似文献

The role of cartilage canals is to transport nutrients and biological factors that cause the appearance of the secondary ossification centre (SOC). The SOC appears in the centre of the epiphysis of long bones. The canal development is a complex interaction between mechanical and biological factors that guide its expansion into the centre of the epiphysis. This article introduces the ‘Hypothesis on the growth of cartilage canals’. Here, we have considered that the development of these canals is an essential event for the appearance of SOC. Moreover, it is also considered to be important for the transport of molecular factors (RUNX2 and MMP9) at the ends of such canals. Once the canals are merged in the centre of the epiphysis, these factors are released causing hypertrophy of adjacent cells. This RUNX2 and MMP9 release occurs due to the action of mechanical loads that supports the epiphysis. In order to test this hypothesis, we use a hybrid approach using the finite element method to simulate the mechanical stresses present in the epiphysis and the cellular automata to simulate the expansion of the canals and the hypertrophy factors pathway. By using this hybrid approach, we have obtained as a result the spatial–temporal patterns for the growth of cartilage canals and hypertrophy factors within the epiphysis. The model is in qualitative agreement with experimental results previously reported by other authors. Thus, we conclude that this model may be used as a methodological basis to present a complete mathematical model of the processes involved in epiphyseal development.  相似文献

Honeybee subspecies have been affected by human activities in Europe over the past few decades. One such example is the importation of nonlocal subspecies of bees which has had an adverse impact on the geographical repartition and subsequently on the genetic diversity of the black honeybee Apis mellifera mellifera. To restore the original diversity of this local honeybee subspecies, different conservation centres were set up in Europe. In this study, we established a black honeybee conservation centre Conservatoire de l'Abeille Noire d'Ile de France (CANIF) in the region of Ile‐de‐France, France. CANIF's honeybee colonies were intensively studied over a 3‐year period. This study included a drone congregation area (DCA) located in the conservation centre. MtDNA COI‐COII marker was used to evaluate the genetic diversity of CANIF's honeybee populations and the drones found and collected from the DCA. The same marker (mtDNA) was used to estimate the interactions and the haplotype frequency between CANIF's honeybee populations and 10 surrounding honeybee apiaries located outside of the CANIF. Our results indicate that the colonies of the conservation centre and the drones of the DCA show similar stable profiles compared to the surrounding populations with lower level of introgression. The mtDNA marker used on both DCA and colonies of the conservation centre seems to be an efficient approach to monitor and maintain the genetic diversity of the protected honeybee populations.  相似文献

Aim To provide evidence suggesting the existence of a dynamic system of extinction and replacement. Location The Indo-West Pacific Ocean. Methods Utilization of species distribution patterns produced by detailed systematic works. Results The distribution patterns appear to suggest a sequence of events that is consistent with the centre of origin hypothesis. The East Indies centre is considered to operate according to the centrifugal speciation model. Main conclusions Disjunct patterns appear to have originated in the East Indies and spread out from there. In cases where the vacated area is occupied by a sibling species, competitive (sympatric) speciation may have occurred. Over time the extinction pattern will gradually extend outward until it is played out among small populations far from their centre of origin  相似文献

The properties of Photosystem I iron-sulphur centres A and B from spinach and barley chloroplasts were investigated by electron paramagnetic resonance spectroscopy (EPR). Barley chloroplasts were shown to photoreduce significant amounts of centre B at cryogenic temperatures unlike those from spinach which only photoreduced centre A. Centre B in barley chloroplasts was also reduced by dithionite before centre A and the EPR spectrum of reduced centre B was obtained. Illumination of barley chloroplasts at 15 K where centre B was chemically reduced resulted in the reduction of centre A and the appearance of spectral features indicating interaction between the two reduced centres. The variation of behaviours of iron-sulphur centres A and B between species favours a scheme of electron flow for Photosystem I where either centre A or centre B act as parallel electron acceptors from the earlier acceptor X.  相似文献
