The hemocytes of selected stages of Hyalophora cecropia from first instar larvae to four-day-old adults were examined and compared with those of Samia cynthia and Antheraea polyphemus. Five classes and two subclasses of hemocytes are described in these moths: (1) prohemocytes, (2) plasmatocytes (of several morphological types), (3) spherule cells, (4) adipohemocytes (two subclasses), and (5) oenocytoids. All types except oenocytoids and subclass II adipohemocytes, are found in all stages examined. Mitotic figures were common among prohemocytes of most stages, but were seen only rarely among plasmatocytes and adipohemocytes, and were not seen among spherule cells or oenocytoids. Prohemocytes and plasmatocytes often contain lipid but rarely PAS positive material. Spherules of spherule cells are PAS positive, as are occasional cytoplasmic inclusions of oenocytoids. Adipohemocytes of both subclasses contain lipid and PAS positive materials in all stages examined. Adipohemocytes and plasmatocytes proved to be most active in phagocytizing ink. Relationships between hemocytes of these and other insects, and some possible functions of hemocytes are discussed.  相似文献

The morphological features of the hemocytes of the crustacean Ligia exotica are similar to hemocytes of insects and millipedes. Jones system of hemocyte classification is extended to crustacean hemocytes. As in insects, seven classes of hemocytes, identified as prohemocytes, plasmatocytes, granular hemocytes, cystocytes, oenocytoids, spherule cells and adipohemocytes, occur. The prohemocytes can be subdivided into five categories that probably represent the precursor of major cell types. The structural and chemical features of other major cell classes are distinct and support the concept of Jones ('62) that these types might have different lineages and might not be capable of transforming into one another. Some of the prohemocytes, plasmatocytes and granular hemocytes are amoeboid. Cystocytes do not bring about any visible plasma coagulation similar to their counterpart in millipedes. Oneocytoids and adipohemocytes are rare. Plasmatocytes, cystocytes and oenocytoids occur in conglomerates, the significance of which is discussed. The cell types are compared with those of the hemocytes of other crustaceans. It is suggested that the nomenclature based on morphological characters is more suited for crustacean hemocytes than a nomenclature based on behavioural and physiological characters.  相似文献

【目的】蝗虫血细胞的研究以及不同血细胞类型间的演化关系,是蝗虫细胞免疫学的依据,也是防蝗治蝗的重要依据。【方法】本研究通过Wright-Giemsa对黑翅痂蝗Bryodema nigroptera Zheng血细胞进行了混合染色,并使用光学显微镜进行了观察。【结果】发现黑翅痂蝗血细胞包括原血胞、浆血胞、粒血胞、类绛色血细胞、囊血胞、脂血胞、蠕形细胞和巨核血细胞8种类型。【结论】在成虫体内,原血胞占细胞总数的1%,浆血胞占32.2%,粒血胞占57.3%,类绛色血细胞占1%,囊血胞占6%,脂血胞占1%,蠕形细胞占1%,巨核细胞占细胞总数的0.5%,粒血胞数量最多,巨核细胞最少。  相似文献

ABSTRACT. Adults of five species of Triatominae were found to stridulate when handled: Triatoma protracta, T. lenti, T. infestans, Panstrongylus megistus and Dipetalogaster maximus. All five larval stages of D. maximus were also found to stridulate. Stridulation is caused by the tip of the reflexed proboscis rubbing along the transversely ridged prosternal groove. Each syllable of the resulting song is produced by a single antero-posterior movement of the proboscis and contains 80–150 intrasyllabic spikes, each corresponding to the striking of a single ridge in the prosternal groove. Analysis of tape recordings of the songs showed that all species have a broad frequency band without well-defined maxima, extending into the ultrasonic with frequencies of up to 50–100 kHz. The lowest frequency produced by each species is consistent with the repetition rate of the striking of prosternal ridges by the proboscis. No behavioural response to the replayed song was detected, but when the song was replayed to female P. megistus the frequency of action potentials within the antennal nerve correlated well with the intrasyllabic spike frequency of the replayed song. Possible microphonic artefacts were not, however, excluded. Possible functions of the song are discussed.  相似文献

Characterization of hemocytes from the yellow fever mosquito,Aedes aegypti   总被引:3,自引:0,他引:3  
Mosquitoes are the most important arthropod disease vectors, transmitting a broad range of pathogens that cause diseases such as malaria, lymphatic filariasis, and yellow fever. Mosquitoes and other insects are able to mount powerful cellular and humoral immune responses against invading pathogens. To date, most studies have concentrated on the humoral response. In the current study we describe the hemocytes (blood cells) of the yellow fever mosquito, Aedes aegypti, by means of morphology, lectin binding, and enzyme activity and immunocytochemistry. Our light and electron microscopic studies suggest the presence of four distinct hemocyte types: granulocytes, oenocytoids, adipohemocytes, and thrombocytoids. We believe granulocytes and oenocytoids are true circulating hemocytes, but adipohemocytes and thrombocytoids are likely adhered to fixed tissues. Granulocytes, the most abundant cell type, have acid phosphatase and alpha-naphthyl acetate esterase activity, and bind the exogenous lectins WGA, HPA, and GNL. Phenoloxidase, an essential enzyme in the melanotic encapsulation immune response, was detected inside oenocytoids. This is, to our knowledge, the first report that has detected phenoloxidase inside mosquito hemocytes at the ultrastructural level. These results have begun to form a knowledge base for our ongoing studies on the function of Ae. aegypti hemocytes, and their involvement in controlling infections.  相似文献

Hemozoin (Hz) is a heme crystal produced by several blood-feeding organisms in order to detoxify free heme released upon hemoglobin (Hb) digestion. Here we show that heme crystallization also occurs in three species of triatomine insects. Ultraviolet-visible and infrared light absorption spectra of insoluble pigments isolated from the midgut of three triatomine species Triatoma infestans, Dipetalogaster maximus and Panstrongylus megistus indicated that all produce Hz. Morphological analysis of T. infestans and D. maximus midguts revealed the close association of Hz crystals to perimicrovillar membranes and also as multicrystalline assemblies, forming nearly spherical structures. Heme crystallization was promoted by isolated perimicrovillar membranes from all three species of triatomine bugs in vitro in heat-sensitive reactions. In conclusion, the data presented here indicate that Hz formation is an ancestral adaptation of Triatominae to a blood-sucking habit and that the presence of perimicrovillar membranes plays a central role in this process.  相似文献

A comparative study of holocentric chromosomes in the triatomine species Panstrongylus megistus, Rhodnius pallescens and Triatoma infestans was carried out in order to characterize heterochromatin, rDNA active sites and nucleolar proteins. Cytological preparations of seminiferous tubules were stained by silver impregnation, C banding, fluorochromes cma3/da and dapi/da, and fluorescent in situ hybridization (FISH) with Drosophila melanogaster 28S rDNA probe. Our results showed interesting aspects of the organization of chromatin and chromosomes in the meiotic cells of these insects. In R. pallescens, sex chromosomes (X, Y) were distinct from autosomes, when submitted to silver impregnation, C banding, CMA3 staining, and FISH, confirming that these chromosomes bear nucleolar organizer regions (NORs). In P. megistus, two of the three sex chromosomes were CMA3/DAPI-; at early meiotic prophase and at diakinesis, silver impregnation corresponded with FISH signals, indicating that in this species, two chromosomes (probably a sex chromosome and an autosome) bear NORs. In T. infestans, silver nitrate and FISH also stained corresponding areas on meiotic chromosomes. Our data suggest that in triatomines, in general, the number and location of NORs are species-specific. These regions may be considered important chromosome markers for comparative studies to improve the understanding of evolutionary mechanisms in these hematophagous insects.  相似文献

Hemocytes are key players in the immune response against pathogens in insects. However, the hemocyte types and their functions in the white-spotted flower chafers, Protaetia brevitarsis seulensis (Kolbe), are not known. In this study, we used various microscopes, molecular probes, and flow cytometric analyses to characterize the hemocytes in P. brevitarsis seulensis. The circulating hemocytes were classified based on their size, morphology, and dye-staining properties into six types, including granulocytes, plasmatocytes, oenocytoids, spherulocytes, prohemocytes, and adipohemocytes. The percentages of circulating hemocyte types were as follows: 13% granulocytes, 20% plasmatocytes, 1% oenocytoids, 5% spherulocytes, 17% prohemocytes, and 44% adipohemocytes. Next, we identified the professional phagocytes, granulocytes, which mediate encapsulation and phagocytosis of pathogens. The granulocytes were immunologically or morphologically activated and phagocytosed potentially hazardous substances in vivo. In addition, we showed that the phagocytosis by granulocytes is associated with autophagy, and that the activation of autophagy could be an efficient way to eliminate pathogens in this system. We also observed a high accumulation of autophagic vacuoles in activated granulocytes, which altered their shape and led to autophagic cell death. Finally, the granulocytes underwent mitotic division thus maintaining their number in vivo.  相似文献

The aim of this work is to study, characterize, and compare different morphological types of hemocytes of Glossina austeni, G. morsitans, Calliphora erythrocephala, Stomoxys calcitrans, Lucilia sericata, Aedes aegypti, and Culex quinquefasciatus. This information is intended to provide a basis for future studies of the cellular defense mechanisms of these dipterans. Seven morphological types of hemocytes were identified by phase-contrast optics: prohemocytes, plasmatocytes, thrombocytoids, granulocytes, adipohemocytes, oenocytoids, and spindle cells of various sizes. Adipohemocytes are difficult to distinguish from both fat body cells and granulocytes. All seven cell types are not present in every species. For example, thrombocytoids and spindle cells were not found in A. aegypti or C. quinquefasciatus, and oenocytoids were observed only in A. aegypti, C. quinquefasciatus, and in C. erythrocephala. In addition to the hemocytes, fat body cells and nephrocytes are also freely present in the hemolymph of some species but may have gained access to the blood during the bleeding process. With electron microscopy and with thick plastic sections of G. austeni hemolymph, only nephrocytes, plasmatocytes, granulocytes, and spindle cells were identified with any certainty, and the large spindle cells are morphologically very different from those found in the other dipterans. They are rigid cells supported by microtubules running throughout their entire length. These hemocytes are present in large numbers only in newly emerged flies, they are absent in larvae and young pupae, and are rare in old adults. Their disappearance from the hemolymph of newly emerged Glossina appears to be a result of phagocytosis by the plasmatocytes.  相似文献

The response to intra- and interspecific faecal assembling signals was tested in Rhodnius prolixus. Papers impregnated with excrement of R. prolixus induced the aggregation of larvae of this species, but also of those of Triatoma infestans. However, faeces belonging to T. infestans were not able to assemble larvae of R. prolixus. On the other hand, there was no response of R. prolixus to putative chemical factors from their cuticle (footprints), in contrast to T. infestans. Results are discussed as related to the ecology of both species.  相似文献

Six types of hemocytes were found in Agallia constricta leafhoppers: plasmatocytes, spherule cells, granular hemocytes, adipohemocytes, oenocytoids, and prohemocytes. Plasmatocytes, spherule cells, and granular hemocytes accounted for 90–95% of all hemocytes in numphs and adult leafhoppers. As the insect aged from second- and third-instar nymphs to 7- and 8-week-old adults, there was a significant decrease in plasmatocytes in healthy leafhoppers compared to wound tumor virus-infected insects. In contradistinction, there were more granular and spherule hemocytes in healthy leafhoppers than in virus-infected ones as the insects aged. In general, there were more prohemocytes in infected than in healthy leafhoppers. Plasmatocytes from 4- to 8-week-old, infected leafhoppers contained large irregularly shaped, cytoplasmic inclusions. Electron microscopy of these cells showed that the inclusions were either large accumulations of wound tumor virus particles or virus-free electron dense bodies.  相似文献

The aim of the work was to investigate the pattern of chemoreceptor sensilla in adults and fifth stage nymphs of Rhodnius prolixus, R. neglectus, Triatoma infestans and T. sordida in order to study differences and similarities between genera and species. Three types of sensilla were analyzed by light microscopy: thin-walled trichoidea, thick-walled trichoidea and basiconica. The number of sensilla of each three types were counted. The length of the antennal segments were also used as a variable for the analysis. The statistical analysis showed that the number of these antennal chemoreceptors had significant differences between species and between adults and nymphs of each species. Discriminant analysis separates incompletely the fifth stage nymphs of the four species and showed similarity between them. Discriminant analysis performed with 12 variables of the antennae, allowed a complete separation of the adults of the four species.  相似文献

Brazil has just been certificated by Pan American Health Organization as 'free of Chagas disease transmission due to Triatoma infestans'. During the early 1980s, this species of blood-sucking bug alone was considered responsible for approximately 80% of Chagas disease transmission. But it was not always so. The species originally abundant in houses of central and eastern Brazil was Panstrongylus megistus, which seems to have been progressively displaced from houses by T. infestans during the past century. Indeed, T. infestans seems able to displace other Triatominae in artificial environments. Recent studies suggest that it might simply be because T. infestans feeds more efficiently than its Triatominae competitors.  相似文献

The subfamily Triatominae (Hemiptera: Reduviidae) comprises hematophagous insects, most of which are actual or potential vectors of Trypanosoma cruzi, the protozoan agent of Chagas' disease (American trypanosomiasis). DNA sequence comparisons of mitochondrial DNA (mtDNA) genes were used to infer phylogenetic relationships among 32 species of the subfamily Triatominae, 26 belonging to the genus Triatoma and six species of different genera. We analyzed mtDNA fragments of the 12S and 16S ribosomal RNA genes (totaling 848-851 bp) from each of the 32 species, as well as of the cytochrome oxidase I (COI, 1447 bp) gene from nine. The phylogenetic analyses unambiguously supported several clusters within the genus Triatoma. In the morphological classification, T. costalimai was placed tentatively within the infestans complex while T. guazu was not included in any Triatoma complex. The placement of these species in the molecular phylogeny indicated that both belong to the infestans complex. We confirmed with a strong support the inclusion of T. circummaculata, a member of a different complex based on morphology, within the infestans complex. On the other hand, the present phylogenetics analysis did not support the monophyly of the infestans complex species as it was suggested in our previous studies. While no strong inference of polyphyly of the genus Triatoma was provided by the bootstrap analyses, the other species belonging to Triatomini analyzed could not be distinguished from the species of Triatoma.  相似文献

Fifty nine chronic chagasic patients were simultaneously submitted to xenodiagnosis and hemoculture for Trypanosoma cruzi samples isolations. The xenodiagnosis was done with 40 Panstrongylus megistus, Triatoma infestans and Dipetalogaster maximus nymphs, performing 120 triatomines. Groups of 10 insects per specie were dissecated and the intestinal content pooled and examined, after previous trituration and homogenization. The microscopically negative material was seed into LIT medium and examined after 20 days. Twenty nine patients were parasitologically proved, being 15 only by xenodiagnosis, 4 only by hemoculture and 10 by both methods. It was discussed the parasitological comprovation difficulties in chronic chagasic patients, the value of the simultaneous utilization of different triatomine species in xenodiagnosis and the hemoculture, in a favorable positive association to the sensitivity increase in the diagnosis' disease. The 49.2% of positivity obtained in this group, visualize approaches like clinic-therapeutic assay and or epidemiological (case-control) with the purpose to investigate a possible association with T. cruzi samples and different clinic forms in Chagas' disease.  相似文献

Insect hemocytes have historically been identified on the basis of morphology, ultrastructure and hypothesized function. Among insects in the order Lepidoptera, five hemocyte classes are usually recognized: granular cells, plasmatocytes, spherule cells, oenocytoids and prohemocytes. We have generated a panel of monoclonal antibodies (mAbs) against hemocytes of the moth Pseudoplusia includens. In this study, hemocyte identification using 16 different mAbs was compared to identification methods using morphological characters. Three main categories of mAb binding activity were identified: (1) mAbs that specifically labeled only one morphological class of hemocytes, (2) mAbs that labeled granular cells and spherule cells, and (3) mAbs that labeled plasmatocytes and oenocytoids. With one exception, none of the antibodies bound to other tissues in P. includens. However, certain mAbs that specifically labeled granular cells and/or spherule cells in separated hemocyte populations also labeled plasmatocytes co-cultured with granular cells or cultured in granular cell conditioned medium. Overall, our results suggest that granular cells are antigenically related to spherule cells, and that plasmatocytes are antigenically related to oenocytoids. The use of mAbs as hemocyte markers are discussed.  相似文献

Mosquitoes are important vectors of disease. These insects respond to invading organisms with strong cellular and humoral immune responses that share many similarities with vertebrate immune systems. The strength and specificity of these responses are directly correlated to a mosquito's ability to transmit disease. In the current study, we characterized the hemocytes (blood cells) of Armigeres subalbatus by morphology (ultrastructure), lectin binding, enzyme activity, immunocytochemistry, and function. We found four hemocyte types: granulocytes, oenocytoids, adipohemocytes, and thrombocytoids. Granulocytes contained acid phosphatase activity and bound the exogenous lectins Helix pomatia agglutinin, Galanthus nivalis lectin, and wheat germ agglutinin. Following bacteria inoculation, granulocytes mounted a strong phagocytic response as early as 5 min postexposure. Bacteria also elicited a hemocyte-mediated melanization response. Phenoloxidase, the rate-limiting enzyme in the melanization pathway, was present exclusively in oenocytoids and in many of the melanotic capsules enveloping bacteria. The immune responses mounted against different bacteria were not identical; gram(–) Escherichia coli were predominantly phagocytosed and gram(+) Micrococcus luteus were melanized. Transovarial transmission was not detectable among Blastocrithidia triatomae-infected Triatoma infestans. Rather, B. triatomae was transmitted directly between triatomines by cannibalism and coprophagy. Cannibalism conditions that excluded coprophagy always resulted in an infection of Dipetalogaster maxima. The efficiency of transmission was not influenced by the blood source--mice or chickens--fed to the infected donor bugs although chicken blood lyses the epimastigotes of the stomach population. Triatoma infestans was infected by coprophagy only if fed, not if unfed. Blastocrithidia triatomae in dry feces was taken up only if the feces were redissolved in fresh feces. Infections also appeared in groups of bugs fed on chickens previously used for feeding infected bugs.  相似文献

Hemocytes are an essential component of the mosquito immune system but current knowledge of the types of hemocytes mosquitoes produce, their relative abundance, and their functions is limited. Addressing these issues requires improved methods for collecting and maintaining mosquito hemocytes in vitro, and comparative data that address whether important vector species produce similar or different hemocyte types. Toward this end, we conducted a comparative study with Anopheles gambiae and Aedes aegypti. Collection method greatly affected the number of hemocytes and contaminants obtained from adult females of each species. Using a collection method called high injection/recovery, we concluded that hemolymph from An. gambiae and Ae. aegypti adult females contains three hemocyte types (granulocytes, oenocytoids and prohemocytes) that were distinguished from one another by a combination of morphological and functional markers. Significantly more hemocytes were recovered from An. gambiae females than Ae. aegypti. However, granulocytes were the most abundant cell type in both species while oenocytoids and prohemocytes comprised less than 10% of the total hemocyte population. The same hemocyte types were collected from larvae, pupae and adult males albeit the absolute number and proportion of each hemocyte type differed from adult females. The number of hemocytes recovered from sugar fed females declined with age but blood feeding transiently increased hemocyte abundance. Two antibodies tested as potential hemocyte markers (anti-PP06 and anti-Dox-A2) also exhibited alterations in staining patterns following immune challenge with the bacterium Escherichia coli.  相似文献
