cDNA of human gene Surf-6 (hSutf-6) was amplified and cloned into vector pGEX-2T for the expression in the bacterial system of protein hSURF-6 translationally fused to glutathione S-transferase. The resulting vector is named as pGEX-2T-GST-hSurf-6. Superproducer of chimeric protein GST-hSURF-6 was obtained on the basis of Escherichia coli strain BL21-CodonPlus(DE3)-RIL. Its purification was performed by the affinity chromatography on L-glatathione-sepharose. The proportion of recombinant protein GST-hSURF-6 in the optimized conditions was not less than 15% of the total bacterial protein, and up to 7 mg of the protein was isolated from 1 liter of culture of the producer strain. The final fraction of eluate contained approximately 80% of GST-hSURF-6. The amount and the purity of the isolated protein were sufficient to immunize animals and obtain antibodies. Protein GST-hSURF-6 can also be used as an affinity ligand for revealing protein partners of hSURF-6 in human cells.  相似文献

Metallothioneins are rather ubiquitous metal-binding proteins induced by stressing or physiological stimuli. Two major metallothionein isoforms have been identified in mussel: MT10 and MT20. Nevertheless the high sequence homology, the two isoforms exhibit different expression and inducibility in vivo. We cloned and produced in Escherichia coli the MT20 isoform from Mytilus galloprovincilis. cDNA was subcloned into pGEX-6P.1 vector, in frame with a sequence encoding a glutathione-S-transferase (GST) tail. Recombinant protein was purified to electrophoretic homogeneity by affinity chromatography. After enzymatic cleavage of the GST tail the MT moiety was recovered with a final yield of about 5 mg of protein per litre of bacterial culture. The metal-binding ability of MT20 was assessed by absorption spectroscopy upon addition of cadmium equivalents and the metal release was checked as a function of the environment pH. Moreover the protein was analysed for the propensity to polymerization, typical of this class of protein, before and after exposure to reducing and alkylating agents.  相似文献

Mature human interleukin-11 (HuIL-11) is a cytokine consisting of 178 amino acid residues that results from scission of the N-terminal signal peptide, consisting of 21 amino acid residaues, from the corresponding nascent polypeptide. A DNA fragment encoding a truncated HuIL-11 (trHuIL-11), with an additional 5 amino acid residues removed from the N-terminus, was cloned into vector pGEX-2T between the BamHI site and the EcoRI site. Upon transformation with Escherichia coli BL21, the construct over-produced a glutathione S-transferase (GST)-fused protein in a soluble form after IPTG induction. The fusion protein was initially fractionated with butyl-Sepharose 4 fast flow column and by affinity chromatography using a GSH-Sepharose 4B column. On-site enzymatic release with thrombin gave the target protein at 96% purity as judged by SDS-PAGE and HPLC. Expression of the interleukin as a GST-fused protein thus greatly improved downstream processing. Subsequent biological activity assay suggested that trHuIL-11 had similar activity profile to the naturally produced sample and may be a promising candidate for further development as biopharmaceutical.  相似文献

Ascorbate peroxidase (APX) is an important enzyme to scavenge the reactive oxygen species (ROS), which are often caused by the salt stress. Here, APX cDNA from Brassica napus was amplified by RT-PCR and cloned into the prokaryotic expression vector pGEX-6p-1 to express BnAPX as a glutathione S-transferase (GST) fusion protein. The recombinant expression plasmid was then transformed into Escherichia coli BL21 (DE3) and induced with 0.2 mM IPTG at 28°C. The enzyme activity analysis of the induced protein showed the GST-APX protein had the similar enzyme activity with the other found APXs, which decompose H2O2. Moreover, the GST-APX fusion protein was purified by affinity chromatography using the glutathione-Sepharose 4B column. The purified GST-APX protein was then used to immunize rabbits to obtain the anti-BnAPX serum, which was suitable to recognize both the recombinant exogenous BnAPX and the endogenous BnAPX in vivo by western blotting and the immunohistochemical experiment. Furthermore, the immuno-fluorescent microscopy observation revealed that BnAPX was expressed in the chloroplasts. Finally, the bacteria expressing BnAPX grew much faster in the presence of 3% NaCl than the control cells, indicating that the transformant expressing BnAPX acquired resistance to salt stress.  相似文献

The high yield expression of the human LAT1 transporter has been obtained for the first time using E. coli. The hLAT1 cDNA was amplified from HEK293 cells and cloned in pH6EX3 vector. The construct pH6EX3-6His-hLAT1 was used to express the 6His-hLAT1 protein in the Rosetta(DE3)pLysS strain of E. coli. The highest level of expression was detected 8 h after induction by IPTG at 28 °C. The expressed protein was collected in the insoluble fraction of cell lysate. On SDS-PAGE the apparent molecular mass of the polypeptide was 40 kDa. After solubilization with sarkosyl and denaturation with urea the protein carrying a 6His N-terminal tag was purified by Ni2+-chelating affinity chromatography and identified by anti-His antibody. The yield of the over-expressed protein after purification was 3.5 mg/L (cell culture). The human CD98 cDNA amplified from Imagene plasmid was cloned in pGEX-4T1. The construct pGEX-4T1-hCD98 was used to express the GST-hCD98 protein in the Rosetta(DE3)pLysS strain of E. coli. The highest level of expression was detected in this case 4 h after induction by IPTG at 28 °C. The expressed protein was accumulated in the soluble fraction of cell lysate. The molecular mass was determined on the basis of marker proteins on SDS-PAGE; it was about 110 kDa. GST was cleaved from the protein construct by incubation with thrombin for 12 h and the hCD98 was separated by Sephadex G-200 chromatography (size exclusion). hCD98 showed a 62 kDa apparent molecular mass, as determined on the basis of molecular mass markers using SDS-PAGE. The yield of CD98 was 2 mg/L of cell culture.  相似文献

为了在原核细胞中表达青岛文昌鱼Branchiostoma belcheri tsingtaunese S-腺苷高半胱氨酸水解酶(S-adeno-sylhomocysteine hydrolase,SAHH),采取构建文昌鱼SAHH基因的原核表达重组质粒pGEX-6P-1-SAHH的方法,转化入大肠杆菌JMl09感受态细胞中,IPTG诱导蛋白表达,并进行分离纯化.结果经SDS-PAGE分析,重组质粒在JM109中表达并纯化得到的融合蛋白大约为70 kDa,成功构建了文昌鱼SAHH基因原核表达载体,且重组载体表达出融合蛋白,分离纯化得到目的蛋白.  相似文献

A ClpS homologue from Celosia cristata was expressed as maltose-binding fusion protein under the control of strong inducible tac promoter of pMALc2X vector in TB1 strain of Escherichia coli. SDS-PAGE analysis showed that fused ClpS is produced as about 63 kDa protein in recombinant bacteria. Expressed product was purified to homogeneity with a yield of about 31 mg/l of bacterial culture. The results indicated that heterologous expression of Celosia ClpS does not affect bacterial growth under different induced conditions. Total cellular antioxidant assessment results revealed that the induction of ClpS activates the bacterial antioxidative system. Since, the purified ClpS did not exhibit antioxidant activity in vitro, we speculated a functional corelation between bacterial proteolytic apparatus and its anti-oxidative system. This prediction may contribute to our better understanding of functional relationship between proteolytic and antioxidative systems in biological worlds in the future investigations.  相似文献

Interleukin-2 (IL-2) is a vital cytokine secreted by activated T lymphocytes, and plays an important role in the regulation of cellular and immunity of animals. In this study, a gene encoding duck IL-2 was cloned and the soluble recombinant duck IL-2 (rDuIL-2) was expressed in Escherichia coli via fusion with glutathione S-transferase (GST). The results indicated that the GST-rDuIL-2 fusion protein expressed in E. coli Origami (DE3) was confirmed to be of about 40 kDa molecular mass by SDS-PAGE and western blotting. In order to produce soluble rDuIL-2 in a low-cost, nontoxic and high-level expression process, lactose was used as a substitute for Isopropyl-β-D-thiogalactopyranoside (IPTG) to induce the above recombinant strain Origami (pGEX-DuIL-2). By optimizing the expression conditions, the production of soluble GST-rDuIL-2 fusion protein was about 29% of total cellular soluble proteins, which was similar with IPTG used as inducer. The soluble GST-rDuIL-2 fusion protein was purified by one-step affinity chromatography, and GST was removed by thrombin. Then rDuIL-2 was purified by a second affinity chromatography. As a result, the 95% pure rduIL-2 was obtained, and the yield of rDuIL-2 was about 10.6 mg/l bacterial culture. The bioactivity of rduIL-2 was determined by lymphocyte proliferation assay in vitro. Our study provided a feasible and convenient approach to produce soluble and biologically active rDuIL-2, which would be used as an immunoadjuvants for enhancing vaccine efficacy.  相似文献

Riemerella anatipestifer (RA) infections cause major economic losses in the duck industry. In this study, an immunogenic protein, chaperonin GroEL (GroEL), was identified from the outer membrane of RA strain WJ4 by immunoproteomic assay based on matrix-assisted laser desorption/ionization time of flight mass spectrometry. The complete sequence of the encoding gene, chaperonin groEL (groEL) was amplified and determined to be 1,629 base pairs in length. groEL was then cloned into expression vector pGEX-6P-1, and the expression of the recombinant GroEL (rGroEL) in Escherichia coli strain BL21 was confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis and Western blotting analysis. Immunization assay showed that ducklings or rabbits immunized with purified rGroEL generated 53- or 160-fold more anti-GroEL antibodies than those with no immunization. Importantly, bactericidal assay showed that rabbit anti-GroEL serum killed 30.0–57.3% of bacteria representing different serotypes, while rabbit anti-bacterin serum killing activity exhibits large serotype-dependent variations between 0.2% and 63.6%. Animal challenge experiment showed that ducklings immunized with rGroEL were 50%, 37.5%, and 37.5% protected from the challenge with RA strains WJ4 (serotype 1), Th4 (serotype 2), and YXb-2 (serotype 10), respectively. In addition, groEL from 34 additional RA strains was amplified by polymerase chain reaction (PCR), and products from nine were sequenced. groEL is highly conserved among RA strains, as the DNA sequence identity was over 97.5% between WJ4 and the nine additional strains. Our results suggest that GroEL may be a good candidate for new RA vaccine development.  相似文献

为获得重组人MBD4蛋白,将编码MBD4的开放式阅读框(ORF)插入原核表达载体pGEX6P1 GST基因下游的多克隆位点(MCS).将获得的表达质粒转化入大肠杆菌BL21(DE3) 菌株扩大培养并用IPTG诱导融合蛋白的表达.用谷胱甘肽琼脂糖凝胶 4B亲和介质从菌体裂解液中纯化了GST-MBD4融合蛋白.经过Prescision protease专一性裂解成功去除了融合蛋白上的GST标签.通过Mono Q阴离子交换层析获得了纯度达94%以上的MBD4蛋白,该蛋白具有甲基化DNA结合和糖苷酶生物活性.  相似文献

根据GenBank已发表的猪轮状病毒vp4基因保守序列,设计一对引物扩增猪轮状病毒地方株JL94株vp4全基因;将扩增产物与载体pMD-18T连接进行序列测定并将测序结果同国外分离株进行序列比较。用限制性内切酶BamHI和SalI将vp4基因从pMD-18T-vp4切下;同样用BamHI和SalI双酶切表达载体pGEX-6P-1;将这2个双酶切产物连接并转化,通过酶切鉴定和PCR鉴定证明完成了vp4原核表达载体的构建。测序结果表明:vp4全基因长2362bp,JL94株同国外分离株BEN307株、Gottfried株vp4全基因片段核苷酸同源性分别为93.44%和69.43%;氨基酸同源性分别为96.06%和71.88%。说明JL94株同BEN-307株属同一VP4血清型,而同Gottfried株属于不同血清型。重组原核表达载体pGEX-6P1-vp4的构建为表达VP4蛋白,研制诊断性抗原和病毒重组活载体疫苗奠定了基础。  相似文献

A procedure has been developed for the overexpression and purification of milligram quantities of theKlebsiellaK-36 arylsulfate sulfotransferase (ASST). The structural gene was amplified by means of a polymerase chain reaction (PCR) technique and inserted into the plasmid vector pGEX-3X. The plasmid pGEX-100, carrying theKlebsiellaK-36astAstructural gene under the control of theEscherichia coli tacpromoter, was transformed into theE. colistrain BL21 (DE3). The ASST was produced inE. colias a fusion with glutathioneS-transferase. Conditions for protein production, isolation on glutathione Sepharose 4B, and Xa cleavage to generate active ASST were developed. The purification yielded approximately 0.7 mg of pure enzyme per liter of bacterial culture. Kinetic analysis of the overexpressed enzyme indicated that it had kinetic properties almost the same as those of the enzyme purified fromKlebsiellaK-36 cells. The purification procedure was very rapid and is suitable for obtaining considerable amounts of enzyme at a relatively high yield compared with its purifying method from the culture of theKlebsiellaK-36 strain.  相似文献

A 1.4-kb gene encoding the “small” sialidase isoenzyme ofClostridium perfringensA99, including its own promoter, was previously cloned in and expressed byEscherichia coliJM 101. Since all attempts to purify this enzyme to homogeneity were unsuccessful, a new strategy was developed. The structural gene was amplified by means of a PCR technique and inserted into the plasmid vector pQE-10, transferring a six-histidine affinity tag (His6) to the N-terminus of the protein. In order to minimize proteolytic degradation of the sialidase protein, the gene was subcloned into theEscherichia colistrain BL21(DE3)pLys S with reduced protease activity. The sialidase production was increased about 2.5-fold when compared with that of the original clone. The enzyme, released by lysozyme treatment of the bacterial cells, was purified by metal chelate chromatography on Ni–nitrilo-triacetic acid agarose to apparent homogeneity in SDS–PAGE. The 42-kDa protein was enriched 62-fold with a yield of 82% and a specific activity of 280 U mg−1. A total amount of 1 mg sialidase was obtained from 1 liter of bacterial culture. For future studies, including crystallization experiments, the histidine affinity tag was removed from the sialidase enzyme by aminopeptidase K. The sialidase was then separated from aminopeptidase K by ion-exchange chromatography, resulting in an overall yield of 83% and a specific activity of 305 U mg−1using 4-methylumbelliferyl-α- -N-acetylneuraminic acid under standard conditions. The two forms (with or without the histidine tag) of sialidase exhibited similar kinetic properties when compared to the wild-type enzyme.  相似文献

To develop a vaccine, which can simultaneously prevent the diseases caused by various pathogenic bacteria in fish, we try to find a conserved outer membrane protein (OMP) antigen from different bacterial pathogens. In this study, an OMP fragment of 747 bp (named as Omp-G), which was highly conserved in seven Aeromonas OMP sequences from the NCBI database, was amplified by PCR from one Aeromonas sobria strain (B10) and two Aeromonas hydrophila strains (B27 and B33) with the designed specific primers. The sequence was cloned into pGEX-2T (6 × His-tag) vector, expressed in Escherichia coli system, and then the recombinant protein (named as rOmp-G) was purified with nickel chelating affinity chromatography. The purified rOmp-G showed a good immunogenicity in rabbits and well-conserved characteristics in these three pathogens by enzyme-linked immunosorbed assay. Furthermore, the rOmp-G also showed good immunogenicity in eels (Anguilla anguilla) for eliciting significantly increased specific antibodies (P < 0.01), and providing higher protection efficiencies (P < 0.05) after the pathogens challenge. The values of the relative percent survival in eels were 70% and 50% for two A. hydrophila strain challenge, and 75% for A. sobria strain challenge. This is the first report of a potential vaccination in eels that simultaneously provide protectiveness against different Aeromonas pathogens with a conserved partial OMP.  相似文献

以人胎脑RNA为模板, 采用RT-PCR技术扩增人Artemin cDNA。序列分析表明, 扩增的人Artemin cDNA核苷酸序列与已发表序列(GenBank登录号:AF115765)同源性为99.7%, 氨基酸序列同源性为100%。将经过序列分析确定的Artemin cDNA插入原核表达载体pGEX-6p-1中, 构建重组表达载体pGEX-6p-1-hART。通过SDS-PAGE分析重组人Artemin融合蛋白在大肠杆菌中的表达情况。结果表明, 重组人Artemin融合蛋白表达量约占宿主菌总蛋白的18.32%, 主要以包涵体形式存在。对表达的重组人Artemin融合蛋白包涵体进行溶解和复性, 并进行Western blotting分析。说明体外成功扩增人Artemin cDNA, 并在原核表达系统中高效表达了重组人Artemin融合蛋白。  相似文献

Bacillus thuringiensis (Bt) cyt genes coding hemolytic and cytolytic toxins constitute a gene family, which are divided into two groups: cyt1 and cyt2. A novel cyt2 gene was detected from a soil-isolated Bt strain T301, which was highly homologous to cyt2Ba1 and finally designated cyt2Ba7. Until now, Cyt2Ba has not been expressed alone in Bt or other hosts. In this study, the cyt2Ba7 gene was cloned into the vector pQE30 and expressed as a fusion protein with 6×Histidine residues in Escherichia coli. Unlike cyt1A, cyt2Ba7 was freely expressed and formed cytoplasmic inclusions without the need for a “helper” protein. The 6×His-tagged Cyt2Ba7 was purified in one step by Ni-NTA affinity chromatography, examined cytolytic activity on Sf9 cells, and developed as an antigen to obtain the antiserum against Cyt2Ba by subcutaneous injection into rabbits. This gene was also cloned into the Bt–E. coli shuttle vector pHT3101 and expressed in Bt strain 4Q7.
