Summary This study describes the fine structure of the hamster seminal vesicle. Intact, sexually mature young and aged animals were used to obtain the results presented. The epithelium of the hamster seminal vesicle contains numerous large electron-dense structures, with a single limiting membrane, with many internal granules and, in the aged animals, myelin-like figures. Based on histochemical results presented in this study, and those of others, it is suggested that this material is a lipofuscin pigment. This substance is of special interest inasmuch as in the hamster seminal vesicle epithelium, pigment is a more sensitive indicator of circulating androgen than is, as in the case of the rat seminal vesicle, regression of cell height. It is possible that this pigment takes origin as breakdown products of mitochondria, the endoplasmic reticulum and the Golgi apparatus. The relation of pro-pigment granules and lysosomes is discussed. Results indicate that pigment is formed at sites of increased catabolism in response to a biological insult to the cell.Supported by U.S.P.H.S. grant RG 6583 from the National Institutes of Health, National Institute of General Medical Sciences.  相似文献

Summary A freeze-etch replica method combined with biochemical analyses was used to investigate the ultrastructural organization of the bovine Descemet's membrane.The freeze-etch replica observations revealed that the intact Descemet's membranes were composed of stacks of two-dimensionally arranged hexagonal lattices, in which four components were resolved; (1) round densities as nodes, (2) rod-like structures connecting the densities, (3) randomly oriented fine filaments within the lattices, and (4) amorphous materials covering the lattices.When the membranes were treated with sodium dodecyl sulfate (SDS) and mercaptoethanol, only the amorphous materials were solubilized. However, both the amorphous materials and rod-like structures disappeared in SDS-mercaptoethanol-urea-treated membranes. When the membranes were treated with a very low concentration (0.0005%) of collagenase, rod-like structures and round densities remained insoluble. If the concentration was raised to 0.01%, only the round densities persisted.Comparing these data with the amino acid analysis of each fraction, the following conclusions may be drawn: rod-like structures and fine filaments contain collagenous proteins of different solubility, while round densities and amorphous materials are non-collagenous in nature.  相似文献

The influx of several monosaccharides into Chara corallina wasstudied under varying conditions of temperature, in the presenceof inhibitors, and by the use of competition experiments. Itwas found that the mechanism of transport was stereospecific,and the complex interactions between the sugars were characteristicof a carrier-mediated system. There is reason to believe thatthe carrier involved in this system has a narrower range ofaffinities than is usually attributed to a sugar carrier, forglucose and fructose appear to have different sites of entry,though each can influence the uptake of the other. Summary A new isolate of C. strobilus Parke and Manton has been investigated with light and electron microscopy to add significant details not previously studied. With the aid of a ciné record, to be described more fully elsewhere, some of the more important haptonema movements have been accurately timed. With the aid of improved technical methods of electron microscopy, new observations have been made on the haptonema base, and on the structure and mode of origin of the scales. The presence of 6 microtubules (instead of the more usual 7) in the very long haptonema of this species has been confirmed but the elaborate changes in tubule number and arrangement which take place in the haptonema base are shown to be essentially like those of other species. The morphology and arrangement of the two types of scale present on the cell are as previously described though information on several new and taxonomically valuable surface features have been added. The cup-shaped scales in particular are shown to be more different from those of other species now known in the genus than might previously have been recognised. This difference extends to the position of nascent scales within the Golgi system which is described in a preliminary way. Further discussion is deferred until some other new isolates representing related taxa have been described and named.  相似文献

Summary The fine structure of the testicular interstitium was studied in normal adult golden hamsters sacrificed in the reproductive season (spring and summer) and in the winter. The Leydig cells in the reproductively active testes contain abundant endoplasmic reticulum (ER) and numerous mitochondria. The ER occurs in the form of flattened cisternae and tubules, the former prevailing. The cisternae are extremely extensive and are partly granular and partly agranular, their ends being continuous with the tubular reticulum. Mitochondria intervening between the cisternae are closely associated with the agranular portions of the latter. Adjacent to the Golgi complex and continuous with the centrosome a unique filamentous body with a dense laminar core is often observed. In the regressive testes, the Leydig cells show a great reduction of cytoplasmic volume and a remarkable decline of the organelles, especially agranular tubules. The possible functional significance of the tubular and cisternal ER with the associated mitochondria is discussed in relation to the biosynthesis of androgens. Immunolocalisation of the cytoskeleton to plasmodesmata of Chara corallina   总被引:2,自引:0,他引:2  
The macromolecular structure of plasmodesmata in the giant celled freshwater alga, Chara corallina, was examined using antibodies against cytoskeletal elements. The large internodal cells of Chara are separated by a nodal complex of smaller cells which are interconnected by plasmodesmata. Putative plasmodesmata-associated proteins can be identified by a comparison of proteins extracted from preparations of clean walls of nodal complexes and those extracted from the external walls of internodal cells which have no plasmodesmata. Actin and tubulin were identified in the protein extracts of nodal walls and the cytoplasm of nodes and internodes but not in the extracts of internodal external walls. Immunogold labelling confirmed the localisation of actin and myosin to plasmodesmata of Chara.  相似文献

Summary In the albino rabbit the sclera is composed of (a) collagen fibrils arranged in fibers, (b) interposed elastic fibers and (c) fibroblasts. The diameter of collagen fibrils and fibers increases from the internal toward the external surface of the sclera. In the same direction the number of elastic fibers and fibroblasts decreases. A peculiar structure is found in the rough surfaced endoplasmic reticulum of the fibroblasts in the innermost portions of the sclera. Summary The ultrastructure of Cyclidium, including the cilia, kinetosomes, pellicle, microtubules and kinetodesmal fibers is similar to that recorded for other ciliates. Of special interest is the attachment of rod-shaped bacteria within the longitudinally directed shallow surface folds of the protozoan. Both the bacteria and the surface of Cyclidium seem to possess an outer coating of a sticky substance which upon contact holds the bacteria to the protozoan. The bacteria appear to be attached by only a relatively small area of their surfaces. A dense substance appears within the alveolus of the pellicle at the regions of the attachment of the bacteria. The association of the organisms is probably a temporary one, and it is unknown whether either organism is benefited or harmed by the association. The position of the mitochondria in Cyclidium is unusual in that they all lie flattened against the inner membrane of the pellicle, usually in a position directly opposite to that of the attachment of the bacteria to the surface, thus being separated from the bacteria by only the outer cell membrane and the pellicle. Summary As in other trypanosomatids, the cell membrane of Crithidia fasciculata overlies a single layer of microtubules. Each microtubule possesses a large number of periodically arranged drumstick-like appendages and adjacent microtubules are joined by fibrillar connectives. Anteriorly, the microtubules gradually taper to terminate just before or just after entering the reservoir. An attempt is made to correlate microtubule tapering with maintenance of form of the truncated anterior end of the cell. Smooth and coated vesicles are proliferated from the Golgi saccules and the prominent contractile vacuole lies nearby. The single mitochondrion is extensive and expanded at one point to form a capsule for the kinetoplast. The cristae are predominantly plate-like but other configurations do occur. The cytostome, a shallow invagination of the reservoir membrane, is found between two constrictions in the reservoir wall. Supporting the cytostome are several microtubules which penetrate deeply into the cytoplasm. Ingestion of ferritin occurs by pinocytosis from the cytostome and by coated vesicle formation from the reservoir membrane. Digestion probably occurs in multivesicular bodies which contain acid phosphatase activity.  相似文献
