Thirteen microsatellites with dinucleotide repeats were isolated from genomic DNA in solution by hybridization capture. Eleven were evaluated against Acarus siro using a multiple‐tube approach to assess polymorphism and reliability of the results. The multiple‐tube method demonstrated the presence of null alleles due to low levels of DNA. Seven of the 11 microsatellite loci were polymorphic and six produced reliable amplification products, if results only from individuals that scored at all six loci were used in the analysis. These were evaluated in eight strains of A. siro. Two of the samples showed some genetic divergence using FST values.  相似文献

The response to pirimiphos-methyl, in one strain of Acarus farris and two strains of Acarus siro, was assessed using an impregnated filter paper bioassay and by the selection of adults following exposure to pirimiphos-methyl. It was concluded that one of the strains of A. siro was resistant to pirimiphos-methyl and that a major resistance mechanism was involved. The second strain of A. siro gave a response similar to that of a laboratory strain unexposed to organophosphates and was considered to be susceptible. The A. farris strain responded to selection at the ED50 but not at the ED99, and it was concluded that a minor resistance mechanism is present in this strain. Assays of esterase activity were used to attempt to identify the biochemical mechanisms involved in the resistance detected by the bioassays. The A. farris and susceptible A. siro strains showed similar levels of esterase activity but the esterase activity of the resistant A. siro strain was significantly greater. An increase in esterase activity followed selection of both the A. farris strain and the resistant A. siro strain. An acetylcholinesterase assay showed no significant difference between the susceptible and pirimiphos-methyl selected strains of A. siro. The results suggest that esterases are involved in the resistance to pirimiphos-methyl found in A. siro and A. farris but that in A. siro, at least, other mechanisms may also be present.  相似文献

Immature individuals ofAcarus siro L. ingest a total of 0.1408 J of food energy, 58.5% of which is consumed by the deutonymph, 27.1% by the protonymph and only 14.1% by the larva. The total ingestion is much greater (2.8169 J±0.0981se). Much energy, 0.7031 J±0.0231se, is used for egg production, which is concentrated in the first 20 days of adult life. Assimilation efficiency during this period varies from 63 to 71%. Most energy ingested between the 20th and 40th days of adult life is used in biomass production, and biomass increases significantly up to th 30th day. The peritrophic membrane in Acarus siro L. (Acari: Acaridae) is produced by distinct cells located in the ventriculus. In this study, the chitin inside the peritrophic membrane was detected using wheat germ-lectin conjugated with colloidal gold (10 nm). The chitin fibrils of the peritrophic membrane were a target for chitin effectors, including 1) chitinase, which hydrolyzes chitin fibers inside the peritrophic membrane; 2) calcofluor, which binds to chitin and destroys the peritrophic membrane mesh structure; and 3) diflubenzuron, which inhibits chitin synthesis. In addition, soybean trypsin protease inhibitor (STI) and cocktails of chitinase/calcofluor, diflubenzuron/calcofluor and chitinase/STI were tested. These compounds were supplemented in diets and an increase of population initiated from 50 individuals was observed after 21 d of cultivation. Final A. siro densities on experimental and control diets were compared. The chitin in the peritrophic membrane was determined to be a suitable target for novel acaricidal compounds for suppressing the population growth of A. siro. The most effective compounds were calcofluor and diflubenzuron, whereas the suppressive effects of chitinase and STI were low. The failure of chitinase could be due to its degradation by endogenous proteases. The combination of chitinase and STI suppressed A. siro population growth more effectively than when they were tested in oral admission separately. The combinations of calcofluor/chitinase or calcofluor/difluorbenzuron showed no additive effects on final A. siro density. The presence of chitin in peritrophic membrane provides a target for novel acaricidal compounds, which disrupt peritrophic membrane structure. The suitability of chitin effectors and their practical application in the management of stored product mites is discussed.  相似文献

Acarus siro L. 1758 (Acari: Acaridida: Acarididae) is an important pest of stored grain because it contaminates the grain by allergens and transfers pathogenous microorganisms. Rapid detection of contamination enables to intercept an early grain infestation by the pest. In this study, we compared the usability and efficiency of various detection approaches. Under laboratory conditions, grain samples of various sizes were infested by different levels of the following contaminants: eggs, adults, and feces of A. siro. The samples were analyzed by enzyme-linked immunosorbent assay (ELISA) by using anti-A. siro polyclonal antibody (immunochemical method), extracted in Berlese-Tullgren funnels, sieved, and processed by filth-flotation (conventional methods). The adults or juveniles of A. siro could be detected by all the three tested conventional methods and ELISA with detection limits in the range from 221 to 1,157 mites/kg grain. Eggs were detected by filth-flotation only; the detection limit was 1,950 eggs/kg grain. The feces of A. siro were detectable by ELISA test, only. ELISA enabled the detection of the feces with the minimal threshold level of 1.04 microg feces/g grain; it means the assay allowed to trace less than one metabolically active mite per gram of grain. The study thus demonstrated that reliable A. siro detection in grain can only be achieved by combining different detection methods. European Union and U.S. administratives dictate zero or near-zero tolerance level for mite infestation in stored products. This demand is difficult to fulfill, because every detection method is limited by its detection limit; thus, it is hard to reliably detect infestation levels lower than obtained detection limits. This methodical limitation is discussed in context with the determined detection limits of the tested methods.  相似文献

The gut of the mite Acarus siro is characterized on the ultrastructural level. It consists of the foregut (pharynx, esophagus), midgut (ventriculus, caeca, colon, intercolon, postcolonic diverticula, postcolon), and hindgut (anal atrium). The gut wall is formed by a single-layered epithelium; only regenerative cells are located basally and these have no contact with the lumen. Eight cell types form the whole gut: (i) simple epithelial cells forming fore- and hindgut; (ii) cells that probably produce the peritrophic membrane; (iii) regenerative cells occurring in the ventriculus, caeca, colon, and intercolon; (iv) spherite cells and (v) digestive cells forming the ventriculus and caeca; (vi) colonic cells and (vii) intercolonic cells; and (viii) cells forming the walls of postcolonic diverticula and postcolon. Spherite and digestive cells change in structure during secretory cycles, which are described and discussed. The cycle of spherite, colonic, and intercolonic cells is terminated by apoptosis. Ingested food is packed into a food bolus surrounded by a single homogeneous peritrophic membrane formed by addition of lamellae that subsequently fuse together. The postcolonic diverticula serve as a shelter for filamentous bacteria, which also are abundant in the intercolon.  相似文献

Comerinia chuetolamina gen. & sp. nov. was collected from a cargo of dried Daphnia imported to Britain from Mexico. Females can be distinguished from all others described in the family Acaridae in that setae 14, 15 and d5 are flattened dorsoventrally over most of the basal half. This region appears blade-like, bearing numerous strong pectinations along the ventral side. Males may be recognized by a series of characters, some of which reflect a close relationship with Forcellinia Ouds., 1924 and others with Tyrophagus Ouds., 1924. The position of the new genus in a commonly used key to adults of the family Acaridae is indicated.  相似文献

Stored product mites can often infest stored products, but currently there is little information regarding the efficacy of pesticides that can be used for control. In this study we evaluated several common pesticides formulated from single active ingredients (a.i.) or commercially available mixtures (chlorpyrifos, deltamethrin, beta-cyfluthrin, and a combination of deltamethrin and S-bioallethrin), plus an acaricide composed of permethrin, pyriproxyfen and benzyl benzolate, for efficacy against Acarus siro, Tyrophagus putrescentiae, and Aleuroglyphus ovatus. The pesticides were incorporated into the mite diets in a dose range of 10–1000 μg a.i. g−1 diet. Concentrations for suppression of 50 and 90% population growth and eradication (rC0) of mites were fit to linear regression models. None of the tested pesticides gave complete eradication of A. siro, which was the most tolerant of the three mite species tested. The most effective pesticide Allergoff 175 CS was a combination product (a nano-capsule suspension of permethrin, pyriproxyfen and benzyl benzolate) labeled for dust mites, with rC0 range of 463–2453 μg a.i. (permethrin) g−1 diet depending on the species. Least effective were chlorpyrifos and deltamethrin.  相似文献

Laboratory experiments were conducted to establish the best combination of relative humidity (r.h.) and time of exposure to be applied on Cabrales cheeses infested with Acarus farris. Laboratory assays revealed that less than 30 h were required to obtain 90% mortality of mites at 50% and 60% r.h. Males were more susceptible than females to low relative humidity, since their lethal period values (LP) were lower than those obtained for females at the same relative humidity. Moreover, the response within sexes to low moisture treatments changed as exposure time increased, since the LP50 obtained for each sex at 50 and 60% r.h. were statistically different whereas the LP90 showed no significant differences within sexes. Accordingly, two modifications of the traditional maturing process were established to assess the efficacy of low moisture treatments to control A. farris on infested cheeses. The first modification consisted of one single exposure of 48 h at 50% r.h. and the second one consisted of two exposures of 48 h at 50% r.h. separated by a time interval of 15 days. No significant differences in final population density were observed for both low moisture treatments compared to control cheeses. Therefore, low humidity treatments are not effective to control A. farris in Cabrales cheese, despite the good results obtained in laboratory assays.  相似文献

Phytoseiulus persimilis Athias-Henriot (Acari Phytoseiidae) is a major predator of Tetranychus urticae (Acari Tetranychidae). The performance of P. persimilis in controlling T. urticae may be altered by pesticides used to manage other pests. Therefore, knowledge of the side-effects of pesticides is essential for IPM. A number of laboratory methods were suggested to evaluate pesticide side-effects on predatory mites. Most methods assess residual effects only, and a number of them are characterised by high predator escape rates from experimental units. A method aimed at evaluating the topical and residual effects of pesticides on P. persimilis is herein described. Mites were treated by microimmersion and then reared in holding cells, on bean leaves previously dipped in a pesticide solution. Three insecticides (pyrethrins, spinosad and thiamethoxam), an insecticide-acaricide (abamectin), and two fungicides (azoxystrobin and tolylfluanide) were evaluated. The strain of P. persimilis used for evaluation was collected from unsprayed vegetable plants. All the pesticides affected the survival and fecundity of P. persimilis. Pesticides did not affect the egg-hatching of P. persimilis females exposed to pesticides. Pyrethrins and abamectin proved to be more toxic than other pesticides, and thiamethoxam was more toxic than spinosad, azoxystrobin and tolylfluanide. The escape rate from experimental units was lower than 5% in all trials. Additional experiments were performed on P. persimilis eggs by dipping leaves with eggs in the pesticide solution. None of the pesticides affected egg survival. Semi-field trials conducted on potted bean plants obtained results similar to those reported in laboratory trials.  相似文献

Mites are an important group of arthropod pests affecting crops, animals and humans. Despite this, detailed physiological studies on these organisms remain sparse due largely to their small size. Unifying models are required to draw together the diverse information from studies on different groups and species. This paper describes a model for digestion in the parasitic mite, Psoroptes ovis, the causative agent of psoroptic mange or sheep scab disease. The limited information about this species is supplemented with data from other acarines, especially house dust mites and ticks. We review the range of enzymes and allergens found in mites and consider their possible roles in digestion in mites, generally and in particular, P. ovis. Histological studies, enzyme biochemistry and molecular biology and experimental evidence suggest that P. ovis utilises a digestive system reliant upon acid peptidases functioning in a largely intracellular environment. The actions of the digestive enzymes are supplemented by the involvement of bacteria as potential direct and indirect sources of nutrition. It is possible that some extra-corporeal digestion also takes place. The interaction of bacteria and digestive enzymes on the skin surface of the sheep may be responsible for the excessive pathological reactions evident in clinical sheep scab.  相似文献

Spin down in response to pesticides is a known phenomenon. This study aimed at investigating the effects of varying acaricide concentrations and acaricidal combinations on the rate of spin down and thread length, the fate of mites after spinning down and, thus, the implications of spin down in the context of mite control and the relationship between webbing and the efficacy of ovicides. In response to cyhexatin at a concentration of up to 100 mg dm-3 the rate of spin down did not change, but in response to higher concentrations it occurred earlier and ceased before the end of the experiment. The mean thread length did not vary with the concentration of acaricide and, whilst the proportion of mites spinning down increased with a cyhexatin concentration of up to 10 mg dm-3, it did not increase further with higher concentrations thereafter. It is therefore postulated that at any given time only a certain proportion of mites have a propensity to spin down. In combination with cyhexatin, only amitraz (at low concentrations) increased the proportion of mites spinning down, but amitraz (at higher concentrations), flubenzimine and permethrin all inhibited spin down, which may be a useful attribute in pesticide formulations. With regard to the fate of mites after spinning down, they were able to climb back up threads, but the proportion doing so was too small to be of practical importance. Mites which spin down and remain at the end of a thread die, but those which reached another leaf were able to spin down again and were more likely to do so if the leaf bore a cyhexatin residue. Mites reaching another leaf are also able to lay eggs, the number being greater if the leaf which they reach is untreated. Spin down is therefore likely to be of significant advantage to Tetranychus urticae in avoidance of the effects of acaricides. Finally, some eggs were observed to be laid on webbing and not to come into direct contact with clofentezine residues. The use of a fine water spray to simulate dew increased the ovitoxicity of clofentezine, probably by resuspension of the pesticide, but not of amitraz, which inhibits webbing production. The avoidance of acaricides should be added to the several known advantages of webbing production for spider mites. It is concluded that, all other factors being equal, pesticide formulations which inhibit spin down and webbing production are likely to be more effective in control programmes than those which stimulate such behaviour.  相似文献

Abstract  This paper tests the hypothesis that habitat differences affect the migratory ability of the Chilean predatory mite, Phytoseiulus persimilis , an introduced biological control agent of the spider mite, Tetranychus urticae . It is suggested that habitat resistance accounts for the species' inability to invade rainforests in south-eastern Queensland, Australia. Like its prey, P. persimilis migrates to distant plants on air currents. To test our hypothesis, populations of the Chilean predatory mite were established on potted bean plants in both remnant rainforest and adjacent open fields, and their migration monitored using sticky traps. Overall it was found that prey populations on leaves were similar in both habitats, but those of predators were about 20% lower in rainforest. However, the numbers of both predators and prey caught on sticky traps in rainforest were about 6% and 25%, respectively, of those caught in open fields, indicating a strongly reduced rate of aerial migration in the forest. The number of P. persimilis caught on the sticky traps increased with increasing populations of predators on foliage. Thus, dense vegetation inhibits the movement of air currents and inhibits colonisation by both predators and, to a lesser extent, spider mites. These results suggest that the inhibition of aerial migration is one reason for lower numbers of P. persimilis in forest habitats, both because its own vagility is restricted, and because its prey is less able to disperse.  相似文献

Social insect colonies possess a range of defences which protect them against highly virulent parasites and colony collapse. The host–parasite interaction between honey bees (Apis mellifera) and the mite Varroa destructor is unusual, as honey bee colonies are relatively poorly defended against this parasite. The interaction has existed since the mid‐20th Century, when Varroa switched host to parasitize A. mellifera. The combination of a virulent parasite and relatively naïve host means that, without acaricides, honey bee colonies typically die within 3 years of Varroa infestation. A consequence of acaricide use has been a reduced selective pressure for the evolution of Varroa resistance in honey bee colonies. However, in the past 20 years, several natural‐selection‐based breeding programmes have resulted in the evolution of Varroa‐resistant populations. In these populations, the inhibition of Varroa's reproduction is a common trait. Using a high‐density genome‐wide association analysis in a Varroa‐resistant honey bee population, we identify an ecdysone‐induced gene significantly linked to resistance. Ecdysone both initiates metamorphosis in insects and reproduction in Varroa. Previously, using a less dense genetic map and a quantitative trait loci analysis, we have identified Ecdysone‐related genes at resistance loci in an independently evolved resistant population. Varroa cannot biosynthesize ecdysone but can acquire it from its diet. Using qPCR, we are able to link the expression of ecdysone‐linked resistance genes to Varroa's meals and reproduction. If Varroa co‐opts pupal compounds to initiate and time its own reproduction, mutations in the host's ecdysone pathway may represent a key selection tool for honey bee resistance and breeding.  相似文献

Methomyl was 15 and 31.3 times more toxic than bendiocarb to bulb mites at the LC50 and LC90 values respectively. However, methomyl (pI50 3.0) was at least 126 times less active than bendiocarb (pI50 5.1) as an inhibitor of bulb mite cholinesterase in vitro. The disparity between the high toxicity of methomyl and its extremely low activity as an inhibitor of mite cholinesterase in vitro indicated that another mechanism was likely involved in its toxic action. Pharmacokinetic studies of methomyl and bendiocarb showed that penetration and metabolism were rapid and that there were no substantial differences in the internal levels of the respective parent carbamates during the 24 h test period. However, volatile radioactive material(s), some of which was carbon dioxide, was produced in appreciably greater amounts from methomyl than from bendiocarb. We speculate that the production of volatiles, such as carbon dioxide, acetonitrile and/or methylamine, may contribute to the toxicity of methomyl to bulb mites. © Rapid Science Ltd. 1998  相似文献

Two rows (anterior and posterior) of long fine setae inserted on the 3rd, 4th and 5th segments of the last two pairs of legs of L. americana form functional swimming blades. Each swimming setal is mounted in a mobile basal socket, and the structure of the setal base and socket configuration ensure that the blades will be passively erected during the leg's power stroke to provide increased thrust and collapsed during the recovery stroke to reduce water resistance. The erect swimming blades increase the effective area for thrust by approximately 500%, making it possible for the mite to lift itself off the bottom. The IVth legs contribute the greater proportion of the thrust developed during paddling, and the 4th segment (genu) bears the largest swimming blades on both legs.  相似文献

Mites are one of the serious pests of turfgrass. Our survey of turfgrass fields from 2013 to 2015 in Korea showed that the occurrence of leaf chlorotic symptom has gradually extended to at least 60% of the examined golf courses. We identified the zoysiae mite Aceria zoysiae in most damaged zoysiagrasses. Artificial infestation of A. zoysiae into zoysiagrasses in pots resulted in symptoms of chlorosis and marginal rolling of the leaves within 3 weeks. We firstly determined the nucleotide sequence of mitochondrial cytochrome c oxidase subunit I (COI) and internal transcribed spacer 2 (ITS2) of the nuclear ribosomal DNA region of A. zoysiae. The variations in COI and ITS2 between A. zoysiae and other species of the genus were 20.9%–43.0% and 7.5%–67.3%, respectively, suggesting significant genetic divergence within the genus. Our study provides valuable information for the rapid diagnosis and infestation monitoring of A. zoysiae in turfgrass fields.  相似文献

The response of the predatory mite Amblyseius longispinosus (Acari: Phytoseiidae) to the webnest of the spider mite nanjingensis (Acari: Tetranychidae) was examined using two-choice tests in the laboratory. A. longispinosus females were found significantly more often on leaves with webnests than on leaves without webnests and were often observed searching under the webbing. Because spider mites and their eggs were removed from the webnests before experiments, predators responded to stimuli associated with webbing, mite feeding damage and other residues in the webnests.  相似文献

The interaction of plants with certain types of parasites leads to the formation of galls, organised structures that create the habitat of the parasite, caused by an abnormal proliferation of host plant's cells under the influence of growth regulators, secreted by the parasite, or by the plant itself under the influence of the parasite. Arthropods, mites in particular, are the largest group of gall‐inducing phytoparasites, but the mechanisms of their interaction with plants remain virtually unexplored. The interaction of the gall‐inducing eriophyoid mite Fragariocoptes setiger with Fragaria viridis plants was used as a model gall–mite system where data were obtained on the changes in the histological structure of F. viridis leaf blades under the influence of the mites as well as F. viridis gene expression during gall formation. For histological purposes, gall formation was split into four stages with each corresponding to the age of the gall as well as to specific changes that occur during that period. A dramatic change of adaxial–abaxial polarity of the lamina throughout the four stages was observed. Moreover, qRT‐PCR analysis of F. viridis gene expression in the developing gall revealed changes in the expression levels of certain meristem‐specific genes, as well as the genes that determine adaxial–abaxial polarity and signalling of phytohormones.  相似文献
