在豚鼠肠系膜下神经节(IMG)及其支配的结肠段联合标本上,对IMG细胞内电位与肠段纵肌或环肌舒缩活动进行了同步记录。实验结果表明:(1)肠段预置张力为零时,约50%IMG细胞有自发的快兴奋性突触后电位(EPSP)活动,切断结肠神经或以筒箭毒(50μmol/L)灌流IMG后消失;(2)筒箭毒或低钙高镁溶液阻断神经节传递时,环肌节律性收缩幅度增大,节律变慢,但对纵肌节律性收缩无明显影响,(3)串刺激节前神经,在IMG细胞引起一串快EPSP或动作电位并常跟随迟慢的EPSP,同时,纵肌在0.1-0.2s潜伏期后出现迅速的、时程基本与动作电位串一致的舒张波,后者在筒箭毒灌流IMG后消失,而环肌运动可见舒张、舒张波延长或收缩波增大。结果提示:IMG不仅中继经典的胆碱能传出功能,还参与以胆碱能传递为中介的肠-肠反射,该反射活动的传出效应主要在于抑制环肌收缩。  相似文献

The background activity of the guinea pig caudal mesenteric ganglion (CMG) neurons and their reflex reactions to colonic distension were studied on isolated combined preparations including the CMG and a colon segment connected with the lumbar colonic nerves. In the control, 62% of the neurons under study generated background activity, which consisted of irregular or regular “fast” excitatory postsynaptic potentials (fEPSP) and action potentials (AP). In 27% of the CMG neurons called “pacemaker-like neurons” (PLN), the background activity was represented by highly regular AP never observed in the CMG completely isolated from the distal colon. Reflex responses evoked by colonic distension were recorded from 76% of the units studied. The distension evoked fEPSP and AP in “silent” neurons and increased the background activity. Both the background activity and reflex responses were shown to be due to nicotinic cholinergic transmission. In some neurons, reflex responses (regular AP) were generated as superimposed on a slow depolarization; the latter was insensitive to nicotinic antagonists and either sensitive or insensitive to muscarinic antagonists. It has been concluded that CMG neurons receive nicotinic, muscarinic, and, probably, peptidergic afferent inputs from the distal colon. Although there are no true pacemaker neurons in CMG, some neurons generate pacemaker-like activity of a synaptic origin.  相似文献

A two-compartment, flat-sheet preparation of rat colon was devised, which enabled exclusive measurement of longitudinal muscle activity during the ascending and descending phases of the peristaltic reflex. A previous study using longitudinal muscle strips revealed the operation of an integrated neuronal circuit consisting of somatostatin, opioid, and VIP/pituitary adenylate cyclase-activating peptide (PACAP)/nitric oxide synthase (NOS) interneurons coupled to cholinergic/tachykinin motor neurons innervating longitudinal muscle strips that could lead to descending contraction and ascending relaxation of this muscle layer. Previous studies in peristaltic preparations have also shown that an increase in somatostatin release during the descending phase causes a decrease in Met-enkephalin release and suppression of the inhibitory effect of Met-enkephalin on VIP/PACAP/NOS motor neurons innervating circular muscle and a distinct set of VIP/PACAP/NOS interneurons. The present study showed that in contrast to circular muscle, longitudinal muscle contracted during the descending phase and relaxed during the ascending phase. Somatostatin antiserum inhibited descending contraction and augmented ascending relaxation of longitudinal muscle, whereas naloxone had the opposite effect. VIP and PACAP antagonists inhibited descending contraction of longitudinal muscle and augmented ascending relaxation. Atropine and tachykinin antagonists inhibited descending contraction of longitudinal muscle. As shown in earlier studies, the same antagonists and antisera produced opposite effects on circular muscle. We conclude that longitudinal muscle contracts and relaxes in reverse fashion to circular muscle during the peristaltic reflex. Longitudinal muscle activity is regulated by excitatory VIP/PACAP/NOS interneurons coupled to cholinergic/tachykinin motor neurons innervating longitudinal muscle.  相似文献

Metabolism and tension were examined in single fibers of the semitendinosus muscle of Rana pipiens at 15 degree C after excitation- contraction uncoupling by stretch and hypertonicity. Interrupted tetanic stimulation at 20 HZ for 150 s, of control fibers in isotonic Ringer at a rest sarcomere length (SL) of 2.3 micrometers, resulted in a steadily declining tension, stimulated glycolysis, and significantly reduced fiber phosphocreatine (PCr) and ATP concentrations. Stretching resting muscle fibers to an SL of 4.7 micrometers did not alter metabolite concentrations, but glucose-6-phosphate rose and PCr fell markedly when the stretched fibers were stimulated tetanically, although tension was absent. Immersion of untetanized fibers in 2.5 X isotonic Ringer produced a transient rise in resting tension, an increase in glucose-6-phosphate, and a significant reduction in PCr. During the transient rise in resting tension, PCr consumption per unit of tension-time integral was the same as that in fibers stimulated tetanically in isotonic Ringer. Tetanization of fibers in hypertonic solution did not further alter metabolite concentrations or produce tension. The results indicate that exposure to hypertonicity induces an increase in both tension and consumption of high-energy phosphate bonds (approximately P) in resting fibers, but stretch does not. during tetanic stimulation, stretch interferes with contraction but does not prevent activation, whereas hypertonicity inhibits activation as well as contraction.  相似文献

Little is known about the effects of aging on synapses in the mammalian nervous system. We examined the innervation of individual mouse submandibular ganglion (SMG) neurons for evidence of age-related changes in synapse efficacy and number. For approximately 85% of adult life expectancy (30 months) the efficacy of synaptic transmission, as determined by excitatory postsynaptic potential (EPSP) amplitudes, remains constant. Similarly, the number of synapses contacting individual SMG neurons is also unchanged. After 30 months of age, however, some neurons (23%) dramatically lose synaptic input exhibiting both smaller EPSP amplitude and fewer synaptic boutons. Attenuation of both the amplitude and frequency of miniature EPSPs was also observed in neurons from aged animals. Electron micrographs revealed that, although there were many vesicle-laden preganglionic axonal processes in the vicinity of the postsynaptic membrane, the number of synaptic contacts was significantly lower in old animals. These results demonstrate primary, age-associated synapse elimination with functional consequences that cannot be explained by pre- or postsynaptic cell death.  相似文献

Stretching arteries from resting length to 1.7 times the resting length increased myosin light chain phosphorylation from 40 to 70% in a graded fashion, reaching a plateau at 1.6 times the resting length. When the fully stretched arteries were released, active tension developed without any exogenous stimulating agent. This stretch-release-induced tension approached the same magnitude as that of the control K+-induced tension. Stretch-induced phosphorylation and the subsequent tension development upon release of stretch were prevented by incubating the arteries in physiological salt solutions containing ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) or chlorpromazine. The inhibition produced by EGTA was reversible. Stretch-induced phosphorylation decreased as a function of time, regardless of whether stretch was maintained, or slackened slowly, or released quickly. While tension developed upon release of stretch, light chain phosphorylation simultaneously decreased. As tension reached and maintained its maximal value, phosphorylation continued to decrease. Thus, light chain phosphorylation is necessary for activation of arterial muscle contraction, but it need not be maintained during tension development or maintenance.  相似文献

As an initial step in constructing a quantitative biomechanical model of the medicinal leech (Hirudo medicinalis), we determined the passive properties of its body wall over the physiological range of dimensions. The major results of this study were:
  1. The ellipsoidal cross section of resting leeches is maintained by tonic muscle activation as well as forces inherent in the structure of the body wall (i.e., residual stress).
  2. The forces required for longitudinal and circumferential stretch to maximum physiological dimensions were similar in magnitude. Cutting out pieces of body wall did not affect the passive longitudinal or circumferential properties of body wall away from the edges of the cut.
  3. The strain (i.e., the percentage change in dimension) of different body segments when subject to the same force was identical, despite differences in muscle crosssections.
  4. Serotonin, a known modulator of tension in leech muscles, affected passive forces at all physiological muscle lengths. This suggests that the longitudinal muscle is responsible for at least part of the passive tension of the body wall.
  5. We propose a simple viscoelastic model of the body wall. This model captures the dynamics of the passive responses of the leech body wall to imposed step changes in length. Using steady-state passive tensions predicted by the viscoelastic model we estimate the forces required to maintain the leech at any given length over the physiological range.

The serotonergic metacerebral cell (MCC) of the mollusk Aplysia produces slow synaptic potentials in motor neurons of the buccal muscle, and increases the rate of ongoing rhythmic burst output of the buccal ganglion. In addition, the MCC acts peripherally to enhance the strength of buccal muscle contractions that are produced by firing of motor neurons. The potentiation of contraction is not associated with any detectable changes of resting membrane potential of muscle cells. Although MCC activity produces a small enhancement of excitatory junctional potentials, several experiments clearly indicate that the MCC has a direct potentiating effect on excitation-contraction coupling. The data suggest that potentiation of contraction might be mediated by cAMP. For example, activity of the MCC enchances the rate of accumulation of cAMP in buccal muscle, application of phosphodiesterase resistant analogs of cAMP potentiates muscle contraction, and a phosphodiesterase inhibitor enhances the effect of MCC stimulation. Recordings from free-moving animals indicate that the MCC becomes activated by exposure of the animal to food stimuli, and that the activation parallels the presence of a food-arousal state. Food-arousal is characterized by enhanced strength and increased frequency of biting responses. Both these effects can result from activity of the MCC. Thus, in this system, modulatory synaptic actions function to provide the substrate for a type behavioral modulation.  相似文献

Oxytocin (OT) has been reported to have a potential protective effect on stress-induced functional gastrointestinal disorders. This study determined whether colonic contraction in adults was affected by antenatal maternal hypoxia, and whether OT is involved in antenatal maternal hypoxia induced colonic contraction disorder. Isometric spontaneous contractions were recorded in colonic longitudinal muscle strips in order to investigate colonic contractions and the effects of exogenous OT on the contraction in antenatal maternal hypoxia and control mice. Both high potassium and carbachol-induced contractions of proximal colon but not distal colon were reduced in antenatal maternal hypoxia mice. Exogenous OT decreased the contractions of proximal colonic smooth muscle strips in control mice, while it increased contractions in antenatal maternal hypoxia mice. OT increased the contractions of distal colonic smooth muscle strips in both antenatal maternal hypoxia and control mice. Hexamethonium blocked the OT-induced potentiation of proximal colon but not distal colon in antenatal maternal hypoxia mice. These results suggest that exogenous oxytocin reverses the decrease of proximal colonic smooth muscle contraction in antenatal maternal hypoxia mice via ganglia.  相似文献

The intensity of light scattered by chemically skinned rabbit psoas fibers in relaxed, rigor, and activated states was monitored at 90 degrees to the incident beam. In the relaxed state, scattering varied in proportion to the volume of muscle in the beam. Scattering increased to 2.3 times the resting value when rigor was induced by withdrawal of MgATP or when the myofibrils were activated by the caffeine-induced release of Ca from the sarcoplasmic reticulum. The rigor-induced increase in scattering decreased monotonically when MgATP was reintroduced stepwise (0-100 microM). This decrease in scattering was accompanied by an increase in tension up to an optimum MgATP level of approximately 10 microM, and then tension decreased at higher concentrations (10-100 microM). The increase in scattering during both rigor and activation was dependent upon fiber length. At lengths when thick-thin filament overlap was near zero, the light signal due to rigor and activation fell to within 10% of the signal for the relaxed fiber at that length. The signal during rigor increased only minimally (approximately 10%) when stretch (approximately 1%) was applied. This increase in signal was small despite a measured 5- to 10-fold increase in tension and an estimated twofold increase in stiffness. Thus, the increased light scattering caused by rigor and activation depends on filament overlap and not tension, stiffness, or substrate binding.  相似文献

The fine structure of the longitudinal muscle fibres of the cockroach proctodeum was investigated by electron microscopy. The fibre is separated incompletely into fibrils, the resting sarcomere length is variable: about 5·8 to 7·3 μm, and the A- and I-bandings are not always clear in longitudinal sections. The ratio of thin and thick filaments at the overlapped region is about 4·1:1 when relaxed, and about 9·8:1 when fully contracted. The myofilament array is not well organized.The previously observed prolonged time course of muscle contraction seems to correlate with the present observations on the poorly developed sarcoplasmic reticulum and irregular distribution of transverse tubules. The Z-bands are irregularly aligned and discontinuous in longitudinal sections. The Z-band structure was studied in relation to the supercontractility. It was found that at maximal isotonic contraction (about 25 per cent rest length) the myofilaments pass through the expanded Z-regions.  相似文献

The role of extraocular muscle (EOM) proprioceptors on eye motility has been investigated in lambs on "encéphale isolé", by evaluating the tension of EOMs at various lengths and velocities of stretch before and after proprioceptive blocks. The EOM tension, in the absence of proprioceptive input, was higher than in normal conditions. Such an effect occurred at lengthening values greater than 3 mm of stretch from resting muscle length, corresponding to 18 degrees of eye deviation and was dependent on the velocity of the stretch, being more effective at high velocity. The muscle receptors responsible for this effect was determined by comparing the sensitivity to vibratory stimulation of spindles and tendon organs to the amount of inhibition provoked by the same stimulation on an EOM electromyographic activity. The tension inhibition appeared to be correlated to muscle spindle activation. Thus, the presence of muscle spindles can determine a reduction of the tension within the stretched muscles. This result suggests that the EOM length and velocity signals operate moment to moment reduction on the stiffness of the muscle which antagonizes eye displacement, thus facilitating the ocular movements.  相似文献

Diabetes affects many aspects of gastrointestinal motility, in part due to changes in interstitial cells of Cajal (ICC). The effect of diabetes on the colon, however, is not well characterized, and the aim of the present study was to investigate possible relationships between altered colonic motility as a consequence of streptozotocin-induced diabetes and injury to ICC. Physiological, immunohistochemical, and ultrastructural techniques were employed. The motor pattern of the rat colon was dominated by rhythmic high-amplitude, low-frequency contractions that were primarily myogenic in origin. These rhythmic contractions were induced by stretch associated with increased tension; the amplitude of the superimposed rhythmic contractions increased with increasing applied tension. In diabetic rats, the stretch-induced rhythmic contractile activity remained robust and of similar frequency but was significantly higher in amplitude compared with that in control rats. At 700 mg of applied tension, the force of contraction in circular colonic muscle strips of the diabetic rats was 370% of control values. This robust presence of low-frequency contractions is consistent with the unaffected pacemaker, the ICC associated with Auerbach's plexus, and the increased amplitude correlates with loss of and injury to ICC of the submuscular plexus and intramuscular ICC. Loss of inhibitory nitrergic nerves does not appear to be a factor based on unaltered nNOS immunoreactivity.  相似文献

The patterns of motor activity that exist in isolated full-length human colon have not been described. Our aim was to characterize the spontaneous motor patterns in isolated human colon and determine whether these patterns are different in whole colons obtained from patients with slow-transit constipation (STC). The entire colon (excluding the anus), was removed from patients with confirmed STC and mounted longitudinally in an organ bath ~120 cm in length, containing oxygenated Krebs' solution at 36°C. Changes in circular muscle tension were recorded from multiple sites simultaneously along the length of colon, by use of isometric force transducers. Recordings from isolated colons from non-STC patients revealed cyclical colonic motor complexes (CMCs) in 11 of 17 colons, with a mean interval and half-duration of contractions of 4.0 ± 0.6 min and 51.5 ± 15 s, respectively. In the remaining six colons, spontaneous irregular phasic contractions occurred without CMCs. Interestingly, in STC patients robust CMCs were still recorded, although their CMC pacemaker frequencies were slower. Intraluminal balloon distension of the ascending or descending colon evoked an ascending excitatory reflex contraction, or evoked CMC, in 8 of 30 trials from non-STC (control) colons, but not from colons obtained from STC patients. In many control segments of descending colon, spontaneous CMCs consisted of simultaneous ascending excitatory and descending inhibitory phases. In summary, CMCs can be recorded from isolated human colon, in vitro, but their intrinsic pacemaker frequency is considerably faster in vitro compared with previous human recordings of CMCs in vivo. The observation that CMCs occur in whole colons removed from STC patients suggests that the intrinsic pacemaker mechanisms underlying their generation and propagation are preserved in vitro, despite impaired transit along these same regions in vivo.  相似文献

Responses of neurons in the antennal lobe (AL) of the moth Manduca sexta to stimulation of the ipsilateral antenna by odors consist of excitatory and inhibitory synaptic potentials. Stimulation of primary afferent fibers by electrical shock of the antennal nerve causes a characteristic IPSP-EPSP synaptic response in AL projection neurons. The IPSP in projection neurons reverses below the resting potential, is sensitive to changes in external and internal chloride concentration, and thus is apparently mediated by an increase in chloride conductance. The IPSP is reversibly blocked by 100 microM picrotoxin or bicuculline. Many AL neurons respond to application of GABA with a strong hyperpolarization and an inhibition of spontaneous spiking activity. GABA responses are associated with an increase in neuronal input conductance and a reversal potential below the resting potential. Application of GABA blocks inhibitory synaptic inputs and reduces or blocks excitatory inputs. EPSPs can be protected from depression by application of GABA. Muscimol, a GABA analog that mimics GABA responses at GABAA receptors but not at GABAB receptors in the vertebrate CNS, inhibits many AL neurons in the moth.  相似文献

Passive stretch, isometric contraction, and shortening were studied in electron micrographs of striated, non-glycerinated frog muscle fibers. The artifacts due to the different steps of preparation were evaluated by comparing sarcomere length and fiber diameter before, during, and after fixation and after sectioning. Tension and length were recorded in the resting and contracted fiber before and during fixation. The I filaments could be traced to enter the A band between the A filaments on both sides of the I band, creating a zone of overlap which decreased linearly with stretch and increased with shortening. This is consistent with a sliding filament model. The decrease in the length of the A and I filaments during isometric contraction and the finding that fibers stretched to a sarcomere length of 3.7 µ still developed 30 per cent of the maximum tetanic tension could not be explained in terms of the sliding filament model. Shortening of the sarcomeres near the myotendinous junctions which still have overlap could account for only one-sixth of this tension, indicating that even those sarcomeres stretched to such a degree that there is a gap between A and I filaments are activated during isometric contraction (increase in stiffness). Shortening, too, was associated with changes in filament length. The diameter of A filaments remained unaltered with stretch and with isometric contraction. Shortening of 50 per cent was associated with a 13 per cent increase in A filament diameter. The area occupied by the fibrils and by the interfibrillar space increased with shortening, indicating a 20 per cent reduction in the volume of the fibrils when shortening amounted to 40 per cent.  相似文献

The role of the longitudinal muscle (LM) layer during the peristaltic reflex in the small and large intestine is unclear. In this study, we have made double and quadruple simultaneous intracellular recordings from LM and circular muscle (CM) cells of guinea pig distal colon to correlate the electrical activities in the two different muscle layers during circumferential stretch. Simultaneous recordings from LM and CM cells (<200 microm apart) at the oral region of the colon showed that excitatory junction potentials (EJPs) discharged synchronously in both muscle layers for periods of up to 6 h. Similarly, at the anal region of the colon, inhibitory junction potentials (IJPs) discharged synchronously in the two muscle layers. Quadruple recordings from LM and CM orally at the same time as from the LM and CM anally revealed that IJPs occurred synchronously in the LM and CM anally at the same time as EJPs in LM and CM located 20 mm orally. Oral EJPs and anal IJPs were linearly related in amplitude between the two muscle layers. Spatiotemporal maps generated from simultaneous video imaging of the movements of the colon, combined with intracellular recordings, revealed that some LM contractions orally could be correlated in time with IJPs in CM cells anally. N(omega)-nitro-L-arginine (L-NA; 100 microM) abolished the IJP in LM, whereas a prominent L-NA-resistant "fast" IJP was always observed in CM. In summary, in stretched preparations, synchronized EJPs in both LM and CM orally are generated by synchronized firing of many ascending interneurons, which simultaneously activate excitatory motor neurons to both muscle layers. Similarly, synchronized IJPs in both LM and CM anally are generated by synchronized firing of many descending interneurons, which simultaneously activate inhibitory motor neurons to both muscle layers. This synchronized motor activity ensures that both muscles around the entire circumference are excited orally at the same time as inhibited anally, thus producing net aboral propulsion.  相似文献

The mandibular muscle receptor organ (Mand. M.R.O.) of Homarus gammarus (L) exhibits increased activity to receptor muscle (R.M.) contraction (decrease in length) and to stretch (increase in length).

The sensory units of this receptor differ in their threshold to R.M. tension and in the frequency change they exhibit for a given increment in tension. The units fire tonically during maintained tension once their threshold has been reached.

Both the number of active units and their frequency increase with R.M. tension. The Mand. M.R.O. is velocity sensitive and exhibits a higher degree of activity during rapid stretch. This dynamic response increases with rate of R.M. stretch.

The activity of the Mand. M.R.O. sensory neurones is compared with that of other M.R.O.’s and the authenticity of some aberrant units is discussed.  相似文献

The aim of this study was to identify myenteric pathways to the circular and longitudinal muscle of the guinea pig proximal colon. To identify excitatory and inhibitory muscle motoneurones, we applied the neuronal retrograde tracer DiI onto the circular or longitudinal muscle layer and performed additional immunohistochemistry for nitric oxide synthase (NOS) and choline acetyltransferase (ChAT). On average 166 +/- 81 circular muscle motoneurones (CMMN) and 100 +/- 74 longitudinal muscle motoneurones (LMMN) were labelled by DiI tracing. Myenteric pathways innervating the muscle were either ascending (DiI-labelled neurones with oral projections) or descending (DiI-labelled neurones with anal projections). The circular muscle was preferentially innervated by ascending pathways (66.0 +/- 9.1%). Most ascending CMMN were ChAT-positive (87.2 +/- 8.5%), whereas descending CMMN were mainly NOS-positive (82.3 +/- 14.6%). Most ascending (62.2 +/- 11.1%) and descending (82.0 +/- 12.5%) CMMN had circumferential projection preferences (circumferential projections were longer than projections along the longitudinal gut axis). In contrast to the polarised projections to the circular muscle, the longitudinal muscle was equally innervated by ascending (46.2 +/- 15.1%) and descending (53.9 +/- 15.1%) neurones. Ascending and descending pathways to the longitudinal muscle consisted predominantly of ChAT-positive neurones (98.1 +/- 1.9% and 68.0 +/- 8.5%, respectively), and both pathways had prominent longitudinal projection preferences. Only 25.5% of the descending LMMN were NOS-positive. In conclusion, the circular muscle in the proximal colon is innervated by descending inhibitory (NOS-positive neurones) and ascending excitatory (ChAT-positive neurones) pathways. In contrast, the longitudinal muscle is primarily innervated by ascending and descending excitatory motoneurones, and only a small proportion of the descending pathway consisted of inhibitory motoneurones.  相似文献
