The development of resistant crop varieties depends on the reliability of the method of screening for resistance. One of the techniques widely used in evaluating field resistance is the field screening method. However, most cowpea varieties regarded as having field resistance are often found to be susceptible in farmers’ fields, where the inoculum density can be higher. This study rescreened 96 cowpea varieties, evaluated earlier in breeders’ fields as resistant to Xanthomonas campestris pv. vignicola, in field and greenhouse under high inoculum pressure. There were significant differences (P ≤ 0.05) in the reactions of cowpea varieties to bacterial blight in terms of disease incidence and severity. Results from the field screening showed that there were 69 susceptible, 25 moderately susceptible, and 2 resistant varieties. In artificial inoculation in the greenhouse IT81D‐1228‐14, IT82E‐16, IT93K‐2271‐2‐2, TVu 1235, and TVu 4630, which were moderately susceptible in the field showed a susceptible reaction. Tvu 12349 and Tvu 15549 gave consistent reactions in the field and in the greenhouse and are therefore good sources of stable resistance to bacterial blight pathogen. Stem canker incidence also varied significantly (P ≤ 0.05) among the cowpea varieties. Seventeen varieties did not manifest canker symptoms.  相似文献

Genetic analysis of 38 rice varieties released by the Bangladesh Rice Research Institute (BRRI) identified 34 as indica, 2 as admixed between indica and aus, and 4 as belonging to the aromatic/Group V subpopulation. Indica varieties developed for the two major rice-growing seasons, the wet monsoon (aman) and the dry winter (boro), were not genetically differentiated. The Additive Main Effect and Multiplicative Interaction (AMMI) model was used to assess the effect of genotype (G), environment (E) and genotype-environment interaction (GEI) on grain arsenic (As) concentration when these rice varieties were grown at ten BRRI research stations located across diverse agro-ecological zones in Bangladesh. G, E and GEI, significantly influenced grain As concentration in both seasons. Overall, E accounted for 69%–80%, G 9%–10% and GEI 10%–21% of the observed variability in grain As. One site, Satkhira had the highest mean grain As concentration and the largest interaction principle component analysis (IPCA) scores in both seasons, indicating maximum interaction with genotypes. Site effects were more pronounced in the boro than in the aman season. The soil level of poorly crystalline Fe-oxide (AOFe), the ratio of AOFe to associated As, soil phosphate extractable As and soil pH were important sub-components of E controlling rice grain As concentration. Irrespective of environment, the mean grain As concentration was significantly higher in the boro (0.290 mg As kg?1) than in the aman (0.154 mg As kg?1) season (p?<?0.0001), though the reasons for this are unclear. Based on mean grain As concentration and stability across environments, the variety BR3 is currently the best choice for the boro season, while BR 23 and BRRI dhan 38 are the best choices for the aman season. Popular varieties BR 11 (aman) and BRRI dhan 28 and 29 (boro) had grain As concentrations close to the mean value and were fairly stable across environments, while high-yielding, short-duration aman season varieties (BRRI dhan 32, 33 and 39) developed for intensified cropping had relatively high grain As concentrations. Results suggest that genetic approaches to reducing As in rice grain will require the introduction of novel genetic variation and must be accompanied by appropriate management strategies to reduce As availability and uptake by rice.  相似文献

A series of 2,5-substituted-1,3,4-oxadiazole/thiadiazole sulfone derivatives were synthesized and evaluated for their antibacterial activities against rice bacterial leaf blight and leaf streak caused by Xanthomonas oryzae pv. oryzae and Xanthomonas oryzae pv. oryzicolaby via the turbidimeter test in vitro. Antibacterial bioassay results indicated that most compounds demonstrated good inhibitory effect antibacterial bioactivities against rice bacterial leaf blight and leaf streak. Among the title compounds, compound 6c demonstrated the best inhibitory effect against rice bacterial leaf blight and leaf streak with half-maximal effective concentration (EC50) values of 1.07 and 7.14 μg/mL, respectively, which were even better than those of commercial agents such as Bismerthiazol and Thiediazole Copper. In vivo antibacterial activities tests at greenhouse conditions demonstrated that the controlling effect of compounds 6c (43.5%) and 6g (42.4%) against rice bacterial leaf blight were better than those of Bismerthiazol (25.5%) and Thiediazole Copper (37.5%).  相似文献

In order to assess resistance to common bean blight, populations of two isolates of Xanthomonas campestris pv. phaseoli were monitored in leaves of two Phaseolus vulgaris breeding lines, BLT87-2 (susceptible) and OAC88-1 (partially resistant) and a resistant tepary bean accession, P. acutifolius P. I. 440795. The breeding line OAC88-1 possesses resistance to common bacterial blight which was incorporated from P. acutifolius by an interspecific cross. In susceptible, leaves, bacterial populations increased to 108 CFU/g leaf at 3 wk after inoculation whereas, in resistant leaves, bacterial populations declined to 101 - 103 CFU/g leaf. In partially resistant leaves the population first declined similar to that in resistant P. acutifolius but later increased, and typical bacterial blight symptoms appeared. Cellulase, protease and amylase activities were monitored in culture and intercellular leaf spaces. Only cellulase activity was, clearly related to bacterial growth in the susceptible host; other enzyme activities were variable in their relationship to host resistance and bacterial growth. Differences between strains in cellulase activity inside partially resistant leaves corresponded to their ability to secrete cellulase in culture. Measuring cellulase activity in intercellular wash fluids may be a simple and sensitive method for determining X. c. phaseoli populations in leaves.  相似文献

A study was carried out to evaluate the chemical composition, in vitro apparently and truly degraded dry matter (DM), utilizable crude protein at the duodenum (uCP) (total CP at the duodenum minus endogenous CP), methane production, and short chain fatty acid production of haulms of six cowpea varieties. The study was arranged in a 2 × 2 × 2 factorial design, with three replicates. Three improved (ITA2, ITA6 and ITA8) and three commercial (Oloyin, Peu and Sokoto) cowpea varieties harvested during wet and dry seasons were used for the study. After an initial gas test to evaluate 96 h gas production profiles of haulms with and without polyethylene glycol (PEG), the time to half maximal gas production was calculated and a second incubation conducted with fermentation stopped at substrate specific half time (t1/2) and 24 h for each substrate. True DM degradability was measured from incubated residues and combined with gas volume to estimate the partitioning factor. Crude protein flow to the duodenum was estimated by combining gas volume with the measured ammonia nitrogen in the incubated fluid. Addition of PEG did not have any effect (P>0.05) on all the variables determined. Interaction between group (improved vs. commercial) and season was observed for CP (P=0.002), lignin (P=0.003) and hemicellulose (P=0.030) contents of the haulms. A group × season interaction was also observed for some of the variables at both substrate specific t1/2 and 24 h. Commercial cowpea haulms had greater (P=0.002) microbial mass and produced less (P<0.05) methane than the improved cowpea haulms. The improved cowpea haulms were less (P<0.001) degraded in the rumen and as a result ensured greater (P<0.001) amount of uCP. The results validated that cowpea haulm is an important agro-based by-product that is adequate in protein and energy to sustain ruminant animal production in Nigeria and other Sub-Saharan African countries during the extended dry season.  相似文献

Ranbir Basmati is one of the traditional Basmati varieties of India and of the most popular traditional Basmati variety grown in Jammu's region (State of Jammu & Kashmir). It is a tall and short-duration variety with strong aroma and excellent cooking quality. However, it is susceptible to bacterial blight (BB) disease caused by Xanthomonas oryzae pv oryzae (Xoo) and prone to lodging. In this study, semi-dwarf (sd1) and BB resistance genes (Xa21 and xa13) were introgressed into Ranbir Basmati using marker-assisted backcross breeding (MABB) scheme. A high-yielding PAU148 carrying Xa21, xa13 and sd1 genes was used as a donor parent. On each generation target, genes were selected, while polymorphic SSR markers were used to select plants having maximum recovery of the recurrent genome. The maximum genome recovery of Ranbir Basmati in BC2F2 was 86.9% in introgressed line SBTIL121. The genotypes carrying resistant genes exhibited very high levels of tolerance against BB disease along with good Basmati rice grain quality traits. The agronomic traits of introgressed lines evaluated in the field and the laboratory showed that most of the agro-morphological traits were similar or superior to Ranbir Basmati. The identified lines can be further evaluated and released as Improved Ranbir Basmati variety.  相似文献

Seeds from cotton plants infected with Xanthomonas campestris pv. malvacearum were collected in different parts of Nicaragua in 1986. When the seeds were homogenized the pathogen could not always be identified by dilution plating. Therefore, an enrichment of bacteria on the natural host was induced before isolation. The cotton seeds were shaken with water and sand for 2 days and then sown in sand for germination. Rather often the developing cotyledons showed typical water-soaked spots, from which the pathogen could be isolated easily. This new method needed more time but made it possible to detect a low level of bacterial infestation. Altogether, 42 bacterial isolates were obtained. For inoculation experiments suspensions with 5x105 CFU - ml?1 were infiltrated into cotton leaves. Incubations of inoculated plants in growth chambers, but not in greenhouses, resulted in typical and uniform disease symptoms (water-soaked leaf spots). Nine of the ten cotton differentials tested were highly susceptible to all the 42 bacterial isolates. Since only line 101-102B proved to be resistant, the Nicaraguan isolates of bacterial blight of cotton were characterized as race 18, of the pathogen. The main cotton cultivars grown in Nicaragua (H-373 and G-286) were strongly affected by the isolated bacterial strains. In order to reduce the disease incidence in Nicaragua, the cultivation of resistant cotton varieties is suggested.  相似文献

Partial resistance to powdery mildew (Erysiphe polygoni) in twenty one cowpea cultivars was assessed over two to three seasons under field conditions. Mildew incidence was recorded as number of leaflets affected/plant and severity as AUDPC or infection rate using Gompertz transformation. Irrespective of the parameter used to measure mildew resistance or the season, mean response of cvs V-105, V-269, V-276, V-282 and V-385 showed clear slow-mildewing, while RC-48, S-488, TVX-944-02E, V-27, V-36 and V-118 were rated as susceptible. Mildew resistance in some cultivars was negatively correlated with that for the leaf rust (Uromyces vignae). AUDPC was a more robust disease parameter than infection rate and showed a good correlation (r2= 0.79) with incidence.  相似文献

Bacterial canker and die-back of poplars in Britain is caused by Aplano-bacterium populi Ridé. Pseudomonas syringae (van Hall), which has been suggested as the causal agent, plays at most only a minor role in canker lesions. It may cause a shoot blight in spring, a distinct pathological condition frequently associated with bacterial canker. Marked variation in infectivity of bacterial slime between seasons and at different dates of collection within one year appears to be the main cause for situations in which varieties, resistant in trials to inoculation with slime, have proved susceptible in the field. Leaf scars afford the main avenues for infection but their infectibility declines rapidly during October. The implications of the temporal separation of availability of natural inoculum and available infection sites are discussed.  相似文献

The changes in some biochemical parameters due to Phytophthora leaf blight infection were assessed in leaf tissues of one resistant (DP‐25), two moderately resistant (Duradim and Jhankri) and one susceptible (N‐118) genotypes of taro [Colocasia esculenta (L.) Schott]. Phytophthora spore suspension (15 000 spore/ml water) was sprayed onto the in vitro raised taro plantlets at 30 days after establishment in pots to induce disease. In comparison with the uninoculated leaves, blight infected leaves showed reduction in protein content and activity of nitrate reductase and increase in total soluble sugar, reducing sugar content and activities of acid phosphatase and alkaline phosphatase among the studied genotypes. Changes in biochemical parameters under induced blight stress as compared with uninoculated control were less in resistant genotypes than that in susceptible genotype. The deviations in biochemical contents were highest in susceptible genotype N‐118. Based on the variations of above parameters under stress and non‐stress control among the four tested genotypes, the overall pattern of changes was N‐118 > Duradim > Jhankri > DP‐25, which is in accordance with the pattern of increasing resistance. The resistant genotypes could be used for commercial cultivation and genetic improvement programme to develop resistant varieties to Phytophthora leaf blight disease.  相似文献

A Soil bacterium, Bacillus subtilis, isolated from the rhizosphere of rice, showed high biocontrol activity against blast, sheath blight and bacterial blight. In the present study, four B. subtilis strains isolated from paddy soil were studied under laboratory, greenhouse and field conditions. Among the four strains assayed, UASP17 gave maximum inhibition of paddy pathogens and was validated under field trials. B. subtilis (UASP17) under different doses and methods of applications was evaluated for two seasons at Agriculture Research Station (UAS, Raichur). UASP17 was effective in reducing the severity of blast (9.00% and 15.57%), bacterial leaf blight (BLB) (5.00% and 6.11%) and sheath blight (11.93% and 4.17%) diseases for two seasons. The application of bioagent also increased the paddy grain yield (61.00 and 64.30?Q/ha) in the two seasons, respectively. Taken together, these results indicate that B. subtilis UASP17 as seedling dip for 30?min (10?mL/L of water) prior to transplanting and 2.50?L/ha foliar application was effective in managing the diseases of paddy.  相似文献

Anthracnose disease is a major constraint for the production of cowpea, accounting for significant yield losses in Nigeria. Trials were conducted in 2016 and 2017 to evaluate an integrated approach in the management of anthracnose disease on cowpea. The experiment was conducted in a screenhouse with ten treatments and three replications in a completely randomised design using a susceptible cowpea variety IT07K-298-9. Ten varieties of cowpea were evaluated for seed-to-plant transmission of pathogen with respect to disease incidence, severity, and yield. The highest yield of 12–14 pods per plant was observed in treatment of Trichoderma asperellum and poultry manure combined. Seed treatments using T. asperellum and soil amendment with poultry manure had a significantly (P?=?0.05) lower disease incidence of 8.4–10.5%. This study showed the potential of combining naturally occurring organic products in the ecosystem that compete favourably with synthetic fungicides in the management of anthracnose disease of cowpea.  相似文献

Data were obtained from the research done in the Guinea Savanna (Zaria 11°11′N; 070 38′E) and Sudan Savanna (Maiduguri ?11°51′N; 13°15′E) regions of Nigeria, respectively on different cultural methods of Striga control and management. In the Guinea Savanna, trials on the effects of nitrogen on the response of resistant and susceptible upland rice varieties to Striga hermonthica infestation and the effect of resistant and susceptible varieties of maize and crop rotation on Striga infestation was carried out, while the effect of inter-cropping trap crop (Bambaranut) with resistant sorghum varieties on S. hermonthica was studied in the Sudan Savanna Zone of Nigeria. In the Guinea Savanna, it was observed that a combination of upland rice variety, Faro 40 and an application of 90 kg N/ha in the wet season and WAB 56-50 upland rice variety and 120 kg N/ha in the dry season, respectively reduced Striga infestation and produced maximum grain yield. Also, the growing of resistant variety of maize (Across 97ITZ comp. I-W) after 1 or 2 years' rotation with cowpea or soybean was observed not only to be effective in Striga control, but resulted in higher grain yield of maize. In the Sudan Savanna, the use of resistant varieties of sorghum, 1CSV1002 and 1CSV1007 intercropped with bambaranut significantly reduced Striga infestation, but the grain yield of the resistant varieties was low. From these studies, Faro 40 with 90 kg N/ha application rate and WAB56–50 with 120 kg N/ha were suitable for the management of Striga and for higher grain yield of upland rice in both wet and dry seasons, respectively, while Across 97ITZ comp. I-W, resistant maize variety and 1 or 2 years rotation with cowpea or soybean were also the best for the management of S. hermonthica and for higher maize yield in the Guinea Savanna zone. Further research needs to be carried out in the Sudan Savanna to select a high yielding resistant variety of sorghum which when intercropped with bambaranut will not only control Striga infestation but will also give high grain yield.  相似文献

Laboratory studies were carried out to determine the functional response of the parasitoid, Pteromalus cerealellae (Boucek) to cowpea weevil, Callosobruchus maculatus (F.) hosts, and the relationship between parasitism and cowpea variety susceptibility to the weevil. The attack response of P. cerealella to weevil larvae was best described by a type III functional response using an egg limitation model. The mean attack rate (a) of the parasitoid was estimated as 1.7 and the upper limit of the response was 24 weevil larvae per individual parasitoid within a 48-h period. The results also showed that cowpea varieties resistant to weevil infestation supported fewer weevil progeny and facilitated a greater suppression of the weevil by the parasitoid. The parasitoid progeny index, a measure that compares the number of the parasitoid progeny to the number of the beetle progeny was higher in cowpea weevil resistant varieties than in susceptible varieties. Additionally, the presence of P. cerealellae resulted in reduced weight losses of cowpea caused by the weevil infestation in both susceptible and resistant varieties.  相似文献

Cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis (Xam), is the most important bacterial disease affecting this crop. A continuous surveillance of the pathogen population dynamics is required to develop an efficient disease management program. During the 1990s, Xam populations showed high levels of genetic variation and relevant migratory processes that were important determinants of the distribution of the pathogen diversity in Colombia. Aiming to characterize the current population structure of the pathogen and the evolutionary forces that shape these populations, sampling collections were carried out from September 2008 until November 2010 in the Colombian Caribbean Region. One hundred and sixty bacterial isolates were characterized using amplified fragment length polymorphism (AFLP) markers. Additionally, a subset of effector genes were sequenced in some isolates to determine their usefulness in Xam population studies and to provide additional information to that obtained with AFLPs. Virulence patterns of ten isolates were determined in nine cassava accessions. Our results show a complex architecture of population and confirm migratory process previously reported in the Caribbean Region. Chinú, one of the locations sampled, presented remarkable features in population dynamics such as longer genetic distances, higher diversity indices, and a genetically differentiated population when it was compared with other locations. Virulence tests showed that MCOL2215, one of the most cultivated cassava varieties in the Caribbean coast, was susceptible to the majority of Xam isolates tested. This study shows the current condition of populations of Xam in the Caribbean Region of Colombia, and it contributes to improve the existing bacterial blight control practices.  相似文献

Cowpea varieties show a wide range of characteristics in their composition that can determine Callosobruchus maculatus (Fabricius) response and survival. Consequently, this work investigated the influence of sublethal doses of Piper guineense on C. maculatus raised on four cowpea varieties: ART-98–12, Niger White, Oloyin, and Ife Brown. Our results demonstrated that C. maculatus from different varieties, despite being reared under the same conditions, responded differently to sublethal doses of P. guineense. Cowpea beetles from ART-98–12 were the most susceptible while those reared on Ife Brown were the least susceptible. Across varieties, survival and reproductive parameters of C. maculatus differed. Furthermore, the ranking for the percentage reduction in the number of eggs laid by C. maculatus was ART-98–12 > Niger White > Oloyin > Ife Brown. According to the log-rank statistic test, the difference observed in C. maculatus’ survival can be explained by variety (log-rank statistic: χ2 [df = 3, N = 16] = 15, p = 0.004). Conversely, the survivability of the chrysomelid beetles reared on Niger white was significantly lower (p = 0.012) compared to the other varieties. This study highlights the role of cowpea variety in dictating the efficacy of a given botanical against C. maculatus.  相似文献

A globulin fraction prepared from the meal of Callosobruchus maculatus-resistant cowpea (Vigna wiguiculata) seeds was shown to be detrimental to this bruchid when incorporated in artificial seeds.2. The performance of C. maculatus was also shown to be strongly hindered by vicilins from resistant seeds when these storage proteins were incorporated in artificial seeds at the level of 2%.3. The purified vicilins from seeds of both resistant and susceptible cowpea varieties were shown to have the same SDS-PAGE pattern but different mobilities in non-denaturing polyacrylamide gel electrophoresis.4. These results and previous ones obtained by us (Silva and Xavier-Filho, 1991; Sales et al., 1992) strongly suggest that the resistance of cowpea seeds from the cultivar TVu 2027 and from others bred from it is associated with the presence of vicilin molecules which are refractory to digestion by bruchid midgut proteinases.  相似文献

A study of life-history traits was made to determine factors associated with the fitness of Meloidogyne incognita isolates virulent to resistance gene Rk in cowpea. Egg hatch, root penetration, egg mass production, and fecundity (eggs per egg mass) of avirulent and virulent phenotypes were compared among M. incognita isolates, isofemale lines, and single descent lines over multiple generations on resistant and susceptible cowpea. Variation (P ≤ 0.05) in both hatch and root penetration rates was found among isolates at a given generation. However, this variation was not consistent within nematode lines among generations, and there was no correlation with level of virulence, except for penetration and virulence on resistant cowpea at generation 20. Resistant and susceptible cowpea roots were penetrated at similar levels. Differences in reproductive factors on resistant plants were correlated with levels of virulence expression. In some isofemale lines, single descent lines, and isolates, lower (P ≤ 0.05) rates of egg mass production and fecundity on susceptible cowpea were associated with virulence to Rk, indicating a trade-off between reproductive fitness and virulence. Other virulent nematode lines from the same isolates did not have reduced reproductive ability on susceptible cowpea over 27 generations. Thus, virulent lineages varied in reproductive ability on susceptible cowpea, contributing to adaptation and maintenance of virulence within M. incognita populations under stabilizing selection.  相似文献

This study investigated effects of wet and dry seasons on the bacterial community structure of the Three Gorges Reservoir by using denaturing gradient gel electrophoresis analysis of the PCR-amplified bacterial 16S rRNA gene. Bacterial diversity, as determined by the Shannon index, the Simpson’s index, and the Richness, dramatically changed in between the dry and wet seasons. The changes in the diversity and relative abundance of microbial populations among the five sites during the wet season have become more marked than those observed during the dry season. Furthermore, cluster analysis also showed these changes. The phylogenetic analysis indicated that Betaproteobacteria is the dominant population, followed by Actinobacterium, in both the wet season and dry season. The water quality parameters were quite stable at all five sites during the same season but noticeably varied from season to season. Canonical correspondence analysis also indicated that the changes in the bacterial community composition were primarily correlated with the variations in temperature, transparency, and the concentrations of NH4 +-N. Slight changes in bacterial community composition among the five sites during the dry season were correlated with different environments. However, during the wet season, major changes were correlated not only with environments, but also it may be associated with the bacterial populations from the surrounding areas and tributaries of the Three Gorges Reservoir.  相似文献
